A curcumin-infused dual-network hydrogel for management of radiation skin injuries

Li Jia,Pang Guobao,Liu Lizhen,Zhang Liwei,Wu Lihua,Xu Yannan

Radiation skin injury (RSI) is a common side effect of radiation therapy for cancer patients, and bacterial infection at the RSI site usually hinders the healing process of the radiation skin injury, thus affecting the subsequent treatment regimen of the patient. To address this issue, we developed a dual network hydrogel containing the natural phenolic antioxidant curcumin. The hydrogel possesses good mechanical and swelling properties to effectively isolate radioactive skin wounds from the external environment, while curcumin released from the hydrogel can effectively inhibit bacterial infections in RSI. In addition, curcumin is able to regulate intracellular redox balance by affecting GSH transferase homologs in cell membranes, thereby reducing the inflammatory response at the wound site. The synergistic effect of antibacterial/anti-inflammatory action produced by the hydrogel effectively accelerates the rate of healing of RSI, providing valuable insights into the clinical management of radiation dermatitis.

Development of tea polysaccharides-xanthan gum complex oleogel: Characterization and curcumin delivery

Yue Chen,Shengqi Li,Na Zhang,Ke Zhang,Xiaoqiang Chen

Oleogels have significant potential for delivering lipid-soluble bioactive compounds. We here prepared a tea polysaccharide conjugate (TPC)-Xanthan gum (XG) complex using the emulsion template method and investigated its curcumin (Cur) loading capacity and in vitro digestion mechanisms. Results unveiled that XG interacts non-covalently with TPC, primarily through electrostatic interactions and hydrogen bonding, enhancing emulsion template formation and stabilization. In addition, XG improved the oleogel’s oil-binding capacity, gel strength, hardness, apparent viscosity, and other gel properties. Meanwhile, the complex oleogel-loaded Cur exhibited a high Cur encapsulation efficiency (>80%), increased antioxidant activity, an augmented free fatty acid release rate, and improved Cur bioaccessibility (>50%) and digestive stability. These findings provide a novel approach to designing oleogels for the effective delivery of lipid-soluble bioactive compounds.

Characterization and application of in situ curcumin/ZNP hydrogels for periodontitis treatment

Liu Chengcheng,Chen Ying,Bai Huimin,Niu Yulong,Wu Yafei

Background Periodontitis is a chronic inflammatory disease that occurs in tooth-supporting tissues. Controlling inflammation and alleviating periodontal tissue destruction are key factors in periodontal therapy. This study aimed to develop an in situ curcumin/zinc oxide (Cur/ZNP) hydrogel and investigate its characteristics and effectiveness in the treatment of periodontitis. Methods Antibacterial activity and cytotoxicity assays were performed in vitro. To evaluate the effect of the in situ Cur/ZNP hydrogel on periodontitis in vivo, an experimental periodontitis model was established in Sprague‒Dawley rats via silk ligature and inoculation of the maxillary first molar with Porphyromonas gingivalis . After one month of in situ treatment with the hydrogel, we examined the transcriptional responses of the gingiva to the Cur/ZNP hydrogel treatment and detected the alveolar bone level as well as the expression of osteocalcin (OCN) and osteoprotegerin (OPG) in the periodontal tissues of the rats. Results Cur/ZNPs had synergistic inhibitory effects on P. gingivalis and good biocompatibility. RNA sequencing of the gingiva showed that immune effector process-related genes were significantly induced by experimental periodontitis. Carcinoembryonic antigen-related cell adhesion molecule 1 (Ceacam1), which is involved in the negative regulation of bone resorption, was differentially regulated by the Cur/ZNP hydrogel but not by the Cur hydrogel or ZNP hydrogel. The Cur/ZNP hydrogel also had a stronger protective effect on alveolar bone resorption than both the Cur hydrogel and the ZNP hydrogel. Conclusion The Cur/ZNP hydrogel effectively inhibited periodontal pathogenic bacteria and alleviated alveolar bone destruction while exhibiting favorable biocompatibility.

Folate-Modified Drug-Carrying Micelles With pH-Responsiveness for Curcumin-Targeted Delivery

Xiuhong Duan,Zimeng Wang,Handa Liu,Hua Ma,Xuepeng Zhang,Lihua Liu,Hongzhou Shang,Ning Qiao

A predrug delivery system combining tumor-targeting and pH-responsive features was synthesized using baicalin (BAI), polyethylene glycol (PEG), and folic acid (FA). BAI and PEG were linked via esterification to create pH-sensitive ester bonds (  COO  ) that facilitate drug release in acidic environments, while FA was attached to PEG through substitution. This system, named FA-PEG-COO-BAI (FPB), encapsulated curcumin (CUR) to form drug-loaded micelles. The impact of drug-loading conditions, such as sonication duration and initial drug dosage, on CUR/FA-PEG-COO-BAI encapsulation efficiency and drug release was investigated. In vitro studies demonstrated a maximum encapsulation rate of (73.09 ± 3.31)% and a drug-loading rate of (11.90 ± 0.69)%. Drug release from CUR/FA-PEG-COO-BAI micelles accelerated as pH decreased, confirming pH-responsive behavior. Cellular uptake and antitumor assays suggested effective tumor targeting and inhibition of proliferation. Hemolysis and cell viability assays indicated low toxicity. These findings underscore the potential of CUR/FA-PEG-COO-BAI micelles for controlled drug release and targeted therapy.

Valine-curcumin complex improves bioavailability and disease resistance in juvenile largemouth bass

Jing Ni,Mao Lin,Ruifang Wang,Ying Ma,Hejian Xiong

Curcumin (Cur) is a polyphenolic compound with pharmacological properties, including anti-inflammatory, antioxidant, and anticancer activities. However, its low solubility and poor stability limit its absorption in vivo. Our laboratory previously synthesized a highly water-soluble valine-curcumin (Val-Cur), increasing its solubility by approximately 9000 times. In this study, based on the evaluation of the antibacterial ability of Val-Cur against three common aquatic pathogens ( Aeromonas hydrophila , Edwardsiella tarda , and Aeromonas veronii ), Val-Cur was added to the feed of juvenile largemouth bass, and for the first time, HPLC was used for Val-Cur detection in vivo; additionally, its pharmacokinetics and effects on disease resistance in fish were investigated. Following a single oral dose (10 mg/kg) of Cur or Val-Cur to largemouth bass, the drug concentration in blood exceeded that in the liver and muscle within 48 h. Compared with Cur, Val-Cur exhibited a delayed peak time in blood, a higher peak concentration (C max = 4.404 μg/mL), a 149 % increase in retention time, and a total drug clearance of 0.005 L·(h·kg) −1 (a 20.4-fold decrease). In the liver and muscle, Val-Cur showed a peak concentration (C max ) 9–10 times higher than Cur, with a 74 %-103 % increase in retention duration and a 15- to 16-fold decrease in clearance. Fecal analysis revealed that Cur excretion was more than five times that of Val-Cur excretion. Compared to those that fed the basal diet (CK) or the diet supplemented with 60 mg/kg curcumin (Cur), the survival rate of largemouth bass infected with Aeromonas hydrophila was significantly higher in the group that fed a diet supplemented with 60 mg/kg Val-Cur. Additionally, the Val-Cur-supplemented diet better preserved the integrity of the intestinal tissue structure. This study provides a valuable theoretical foundation for elucidating the mechanisms of Val-Cur and supports the application of water-soluble curcumin in aquaculture.

Redox-Sensitive Curcumin Prodrug Mediated by Glycyrrhetinic Acid for Liver Cancer Therapy

Wenlan Wu,Mengyi Zhang,Yang Lin,Xuening Li,Ying Liang,Ju Liang

Despite the development of numerous prodrugs for tumor therapy, their clinical application remains limited due to issues such as non-targeted delivery, uncontrolled release, and excessively rapid drug release rates. In this study, we synthesized a dual-functional prodrug, glycyrrhizic acid-polyethylene glycol-disulfide curcumin (GA-PEG-SS-CUR), by conjugating glycyrrhizic acid (GA), which possesses liver-targeting capabilities, with the anticancer agent curcumin (CUR). GA-PEG-SS-CUR can self-assemble in aqueous solutions to form micelles with an approximate diameter of ∼140 nm. In vitro drug release studies demonstrated that GA-PEG-SS-CUR exhibits a sensitive glutathione (GSH) response and sustained release characteristics; after 72 hours, the drug release rate was 30.5% at low GSH concentrations and 46.7% at high GSH concentrations. Due to GA receptor-mediated targeting, the uptake of GA-PEG-SS-CUR in HepG2 cells was significantly higher than that in L02 cells, and the internalization of GA-PEG-SS-CUR in HepG2 cells was markedly greater compared to unmodified PEG-SS-CUR. Cytotoxicity assays indicated that GA-PEG-SS-CUR enhanced the cytotoxicity against tumor cells compared to free CUR, while exhibiting lower toxicity towards normal L02 cells. Pharmacokinetic studies revealed that the half-life of CUR in GA-PEG-SS-CUR was 8.3 times longer than that of free CUR, extending the in vivo circulation time, with CUR accumulation at the tumor site being 6 times greater than that of free CUR, indicating significant tumor-homing ability. Therefore, the developed GA-PEG-SS-CUR represents an ideal delivery system for curcumin in tumor therapy.

Double-Network Bilayer Hydrogel Loaded with Puerarin and Curcumin for Osteochondral Repair

Wenhui Chen,Huan Deng,Yuehan Dong,Xiangjie Wu,Zinuo Xia,Yang Zhou,Lin Yang,Zhijun Huang,Wenjin Xu,Peihu Xu,Haixing Xu

Osteochondral injuries, which could result in cartilage defects and damage to the subchondral bone, pose a significant challenge in the design of scaffolds for bone repair due to the different characteristics of bone and cartilage. In this study, we developed a double-network bilayer hydrogel scaffold composed of an upper layer and a lower layer reinforced with a ZnO nanoparticle. The double-network hydrogel consisted of carboxymethyl chitosan (CMCS), oxidized sodium alginate (OSA), and poly(vinyl alcohol) (PVA) networks. The upper and lower layers were, respectively, loaded with puerarin (Pue) and curcumin (Cur) to facilitate simultaneous repair of cartilage and bone. The scaffold exhibited better water absorption, mechanical properties (tensile strength: 0.8 MPa), self-healing capacity (healing efficiency ≥ 90%), and adhesion (adhesive stress: 0.1 MPa). Approximately 70% of Pue was rapidly released within the first 5 days, while Cur, encapsulated in ZnO, exhibited sustained release over a period of 20 d. In vitro cell experiments revealed that soluble extracts from the scaffolds were noncytotoxic and promoted elevated alkaline phosphatase (ALP) expression and calcium deposition in MC3T3-E1 subclone 14 (MC3T3-14) cells, thereby enhancing bone repair. Meanwhile, extracts fostered the proliferation of ATDC5 cells and glycosaminoglycan (GAG) production, contributing to cartilage repair. In conclusion, the bilayer hydrogel has good properties and is beneficial for the proliferation and differentiation of osteoblasts and chondrocytes.

Antibacterial composite films fabricated from curcumin-loaded lignin micro-nanoparticles and cellulose

Rui Tian,Chaoyuan Zheng,Xingxiang Ji,Srinivas Janaswamy,Guihua Yang,Gaojin Lyu

Degradable regenerated cellulose (RC) composite films have prospects to replace plastic packaging. However, limitations such as lack of functionality, poor mechanical properties, inherent color, and the absence of antibacterial activity restrict their practical application. Herein, butyric anhydride esterified alkali lignin (BAL) encapsulated curcumin micro-nanoparticles were synthesized to develop high-antibacterial composite films. Uniform dispersion of the B-CMPs within the cellulose matrix is critical for enhancing the film's properties. Furthermore, the esterified lignin contributes to the photodynamic bactericidal properties of curcumin by reducing the light absorption of lignin's native chromophoric groups. Results suggest that in the presence of B-CMPs, the RC composite film exhibits a high visible light transmittance of 80.4 % and a tensile strength of 68 MPa. Under blue light excitation, an inhibition rate of 91 % and 92 % against Escherichia coli and Staphylococcus aureus , respectively, was obtained. In addition, the composite films exhibited improved barrier properties with water vapor permeability of 1.12 × 10 −12  g·cm·cm −2 ·s −1 ·Pa −1 and enhanced hydrophobicity (contact angle 82.3° vs. 46.2° for pure RC). These findings demonstrate that reducing reactive groups in lignin via esterification significantly enhances the functional integration of curcumin, yielding composite films with a superior balance of properties. This work offers novel insights into the high-value utilization of lignin toward developing packaging films to meet the growing demand for sustainable packaging materials.

Effects of preparation method on the release kinetics of curcumin-loaded composites

Meixue Cao,Fei Zhou,Shuan Cheng,Xinyue Wang,Ziyu Yuan,Xiuwen Huang,Zuju Shu

To explore the effects of preparation methods on the release kinetics of curcumin (Cur)-loaded sustained-release composites, three fabrication techniques—solvent casting, uniaxial electrospinning, and coaxial electrospinning—were employed to develop Cur-loaded composites using Cur as the model drug, soluble starch (St) and polyvinyl alcohol (PVA) as the matrices. The microstructure and chemical composition of three composite groups were characterized, and their functional properties were evaluated, including swelling capacity, degradation behavior, Cur loading capacity (LC), Cur encapsulation efficiency (EE), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity. Additionally, the in vitro Cur release behavior of the composites was analyzed, and release kinetics were fitted using mathematical models. Results indicated that the preparation methods had no significant impact on the chemical composition of the Cur-loaded composites. However, the nanofibers fabricated by the coaxial electrospinning technique exhibited a unique core-shell microstructure, which distinguished them from the cast films and uniaxial electrospun membranes. Compared with the other two groups, the coaxial membrane showed higher Cur LC and EE, a higher swelling ratio, a lower degradation rate, and a stronger DPPH radical scavenging ability. Kinetic fitting revealed that the Cur release profiles of all three composite groups conformed to the First-order model, but their release performances differed significantly. The coaxial electrospun membrane outperformed the cast film and uniaxial electrospun membrane, with a Cur cumulative release rate of 51.73 % at 96 h and retained significant potential for sustained release. In conclusion, the preparation method directly regulated the antioxidant activity and sustained-release behavior of Cur in the composites. This study provides theoretical basis and technical reference for the design and development of novel drug-loaded sustained-release composite materials.

Curcumin mitigates high glucose-induced cardiac oxidative stress via Notch1 pathway activation

Wu Xia,Qu ZhuangYin,Liu PeiYan

This study aims to investigate the protective effects of curcumin (CUR) in high glucose (HG)-induced oxidative stress and apoptosis of primary cardiomyocytes by activating the Notch1 signaling pathway. CUR is a natural polyphenol isolated from turmeric rhizomes and is known for its antioxidant, anti-apoptotic, and anti-inflammatory effects, particularly relevant in diabetes. Therefore, we used neonatal rat cardiomyocytes exposed to HG conditions, followed by treatment with CUR and DAPT, respectively. We detected and assessed myocardial cells viability and antioxidant enzyme activity by CCK-8 reagent and antioxidant enzyme kit. Apoptosis was detected by flow cytometry. The production of reactive oxygen species was detected by fluorescence labeling, and the expression of related genes and proteins was detected by qRT-PCR and Western blot. HG-induced primary rat cardiomyocytes not only increased apoptosis and ROS production, but also decreased the activity of antioxidant enzymes and the expression of Notch1 and Hes1 proteins. After pre-treatment by CUR, surprisingly, we found that CUR markedly improved viability of HG-treated cardiomyocytes. The results showed that CUR could inhibit the apoptosis of rat cardiomyocytes, inhibit the production of intracellular ROS, and increase the activity of antioxidant enzymes. Further, we found that CUR can upregulate the expression of Notch1 and Hes1 proteins and related genes, suggesting that the protective effect of CUR on HG-induced damage involves the Notch1/Hes1 signaling. These results suggest that CUR protects cardiomyocytes from HG-induced oxidative stress by activating Notch1 and its downstream target genes.

Rapid coordination-driven Fe-DNA nanonetwork loaded with curcumin for fruit preservation

Xiangyang Li,Min Yang,Yushi Xie,Longjiao Zhu,Wentao Xu

Every year, a significant amount of fruit and vegetable spoilage caused by microbial contamination leads to substantial food waste and economic losses in the agricultural and food sectors. There is an urgent need to develop highly efficient and safe multifunctional preservation materials. Curcumin (Cur) is known for its excellent antioxidant and antibacterial properties; however, its poor water solubility and low bioavailability severely limit its practical applications. In this study, we synthesized highly stable and water-dispersible metal-biomolecule network nanoparticles by self-assembly of ferrous ions (Fe 2+ ) with DNA in an aqueous solution at room temperature. Furthermore, by leveraging the strong coordination interaction between Iron(Fe) and Cur, we prepared Cur-modified nanoparticles (Fe-DNA-Cur) with dual antioxidant and antibacterial functions. Fe-DNA-Cur not only preserves the biological activity of curcumin, but also effectively addresses its inherent poor water solubility. After further compounding with chitosan (CS), the as-prepared Fe-DNA-Cur@CS composite demonstrates breakthrough antimicrobial performance, thereby providing an innovative solution for developing novel, highly effective and safe fruit/vegetable preservatives. When used as a sustainable food packaging coating for perishable fruits at room temperature, the Fe-DNA-Cur@CS coating can reduce dehydration, prevent nutrient loss, inhibit microbial growth, increase nutraceutical value, maintain strawberry quality, and extend shelf life during storage. This study demonstrates the potential of Fe-DNA-Cur@CS as a multifunctional coating material to reduce food waste and agricultural economic losses caused by microbial contamination.

Chitosan/squid ring teeth protein hydrogels for the controlled release of curcumin

Shiyu Chen,Xiaoe Chen,Gaofeng Yuan

Biocompatible and degradable hydrogels are extensively utilized for the delivery and controlled release of bioactive agents. Chitosan/squid ring teeth protein (SRT) hydrogels (CH/SRTs) cross-linked by genipin were fabricated, and their gel properties and structural characteristics were analyzed across varying SRT contents. Additionally, the curcumin-release behavior of curcumin-loaded CH/SRTs (Cur-CH/SRTs) was evaluated. Results indicated that increasing SRT content enhanced the elasticity, viscosity, gel strength, hardness, and thermal stability of CH/SRTs. Specifically, the hardness and gel strength of CH/SRT3 (Chitosan: SRT, 1:1) were 207.8 % and 220.9 % higher, respectively, than those of the pure chitosan hydrogel. Higher SRT content significantly increased the curcumin encapsulation efficiency of Cur-CH/SRTs while substantially reducing curcumin release in simulated gastrointestinal fluid. Compared to the curcumin-loaded pure chitosan hydrogel, the curcumin release rate of Cur-CH/SRT3 decreased by 56.12 % after 480 min of simulated gastrointestinal digestion. These findings were attributed to reduced swelling and improved gel strength of Cur-CH/SRTs with SRT addition. Consequently, CH/SRTs demonstrate potential for applications in the controlled release of drugs and bioactive agents.
