Glycation regulation between dual-natural milk-derived proteins and fucoidan by dielectric barrier discharge-cold plasma and its potential for delivery of astaxanthin

Yangcheng Ge,Yingcong Yang,Zhen Xu,Gongshuai Song,Mengna Zhang,Jinyan Gong

Milk proteins are attractive building blocks for food-grade delivery systems due to their biocompatibility and versatile functionality. However, their structural instability and limited protection capacity for lipophilic bioactives restrict broader applications. In this study, β-lactoglobulin fibrils (β-LGF) and β-casein (β-CN) were self-assembled to construct a dual-natural milk-protein complex, followed by dielectric barrier discharge cold plasma (DBD-CP)-assisted glycation with fucoidan (FD) to develop a novel protein-polysaccharide conjugate for astaxanthin (AST) delivery. The structural characteristic, physicochemical property, antioxidant activity, and emulsifying performance of the conjugate were systematically investigated. The β-LGF/β-CN complex at a mass ratio of 1:1 exhibited optimal properties, including reduced particle size, enhanced thermal stability, increased apparent viscosity, and the highest DPPH radical scavenging activity (21.05%). CP-assisted glycation significantly increased the degree of glycation from 9.83% to 18.07%, accompanied by the increased β-sheet content, strengthened intermolecular interactions, and formation of high-molecular-weight conjugates. The LGF-CN-FD conjugate showed superior antioxidant capacity and structural stability. AST-loaded emulsions stabilized by the conjugate achieved the highest encapsulation efficiency (80.57%) and loading capacity (10.73%). In vitro digestion demonstrated enhanced gastric protection (63.25% AST retention) and improved bioaccessibility (51.07%). This work demonstrates that CP-assisted glycation effectively tailors the structure and functionality of dual milk-protein assemblies, providing a simple and efficient strategy to design robust colloidal delivery systems for lipophilic bioactives in functional foods.

Fucoidan from Saccharina japonica alleviates OVA-induced allergy in mice: A role for the AMPK/SIRT1/FOXO1 pathway in enhancing intestinal barrier function

Qing Liu,Zengying Sui,Shiyi Li,Miaomiao Wan,Meilan Xue,Xia Qiu,Hui Liang,Lirui Sun

Fucoidan is a sulfated polysaccharide derived from brown algae ( Saccharina japonica ), and exhibits immunomodulatory properties. Emerging evidence suggests a strong correlation between the development of food allergy and intestinal dysfunction. The present study aimed to investigate the potential anti-food allergy activity of fucoidan and to elucidate its mechanism at the intestinal barrier function. The ovalbumin (OVA)-induced allergy mouse model was established, exhibiting increased OVA-specific IgE, histamine, Th2 inflammatory response, and intestinal damage. Oral administration of fucoidan markedly alleviated allergic symptoms by inhibiting mast cell activation and modulating intestinal immune responses. Furthermore, fucoidan significantly ameliorated intestinal immunological dysregulation, maintained barrier integrity, modulated gut microbiota composition, and enhanced short-chain fatty acid (SCFA) production. Mechanistically, the protective effects of fucoidan were closely associated with activation of the AMPK/SIRT1/FOXO1 pathway, which correlated with improved intestinal barrier function. Collectively, these results demonstrate that fucoidan may protect against food allergy by enhancing intestinal barrier function, highlighting the potential of natural biological substances in addressing food allergy.

Synergistic protective effects of pine pollen extract and fucoidan on adriamycin-induced acute kidney injury via regulation of oxidative stress and inflammatory pathways

Xiaomin Xi,Hailong Zheng,Chunping Luo,Micun Zhong,Lei Yin,Ruting Chen,Jiahui Qian,Zhaoxi Liu,Yiping Zhang

PPAE and FUC synergistically attenuated ADR-induced acute kidney injury both in vitro and in vivo. • Integrated RNA-seq and network pharmacology revealed dual anti-oxidative and anti-inflammatory mechanisms. • The synergy was closely associated with Nrf2-Keap1-GPX4 and NF-κB signaling pathways. • PPAE and FUC remodeled the ADR-induced dysbiosis of gut microbiota and improved renal histopathological morphology. Adriamycin (ADR)-induced nephrotoxicity remains a significant clinical challenge that limits its chemotherapeutic utility, necessitating the development of effective multi-target renoprotective strategies. Although natural polysaccharides show great promise, the synergistic potential of combining pine pollen aqueous extract (PPAE) and fucoidan (FUC) has been unexplored. This study aimed to investigate the synergistic protective effects of PPAE and FUC against ADR-induced kidney injury and to elucidate the underlying molecular mechanisms. In ADR-treated Human Kidney-2 (HK−2) cells, PPAE+FUC exhibited strong synergy (CI = 0.3–0.7), resulting in improved cell viability, reduced apoptosis, decreased LDH and ATP leakage, and attenuated oxidative stress. RNA-seq profiling confirmed a clear convergence on pathways governing oxidative stress, apoptosis, and inflammation. In vivo results corroborated these findings, as the combination treatment significantly attenuated ADR-induced renal dysfunction, histological damage, and systemic inflammation while restoring redox homeostasis. Mechanistically, the combination was highly associated with the modulation of Nrf2-Keap1-GPX4 and NF-κB signaling axes, which simultaneously bolstered antioxidant defenses and suppressed inflammatory cascades via inhibiting p65 and IκBα phosphorylation. Furthermore, 16S rDNA sequencing revealed that PPAE+FUC potentially reshaped the gut microbiota by reconstituting the probiotic genus Akkermansia and normalizing the Firmicutes / Bacteroidota ratio. Ultimately, our study confirms that the PPAE and FUC combination exerts a multi-target synergistic effect through the attenuation of oxidative stress and inflammation, with concomitant alterations in gut microbiota composition. These findings provide a robust evidence-based rationale for this natural polysaccharide combination as a safe and innovative nutritional strategy to mitigate chemotherapy-associated renal injury. Synergistic protection by pine pollen extract and fucoidan against adriamycin-induced renal injury. This study systematically evaluated the synergistic renoprotective effects of pine pollen extract (PPAE) and fucoidan (FUC) against adriamycin (ADR)-induced kidney injury in vitro and in vivo. Integrated analyses combining network pharmacology, transcriptomics, and 16S rDNA sequencing demonstrated that the protective effects of PPAE and FUC were associated with enhanced antioxidant capacity, attenuated inflammatory responses, and reshaped gut microbiota. These beneficial effects were accompanied by alterations in Nrf2-Keap1-GPX4 and NF-κB signaling-related molecules, suggesting their potential involvement in the attenuation of ADR-induced renal injury. Fucoidan-based smart micelles enable P-selectin-targeted and ROS-responsive delivery of mitochonic acid 5 for mitochondrial functional improvement in acute lung injury

Ye-Fei Ruan,Jin-Hui Wang,Wen-Da Zhong,Lin Qiu

Targeting mitochondrial dysfunction in inflamed lung vessels addresses a key challenge in acute lung injury treatment, where current therapies lack precision and efficacy. • We develop smart nanoparticles that actively seek out injured blood vessels and release medicine only where oxidative stress is high, enabling enhanced mitochondrial repair. • This strategy markedly improves lung function and survival in mice with acute lung injury, demonstrating a promising approach for treating severe pulmonary inflammation. Acute lung injury (ALI) is a life-threatening condition with complex pathogenesis. Mitochondrial dysfunction in pulmonary vascular endothelial cells serves as a central hub, driving oxidative stress, inflammation, and barrier disruption. Existing treatments are limited by poor targeting and insufficient efficacy. To address this, we engineered a novel dual-targeting nanoplatform, P-selectin-directed and ROS-responsive fucoidan micelles loaded with the mitochondrial modulator Mitochonic Acid 5 (Fuc-TP@MA-5). Here we show that this system actively homes to inflamed lung endothelium via fucoidan-P-selectin binding and selectively releases MA-5 within the high-ROS microenvironment via thioketal linker cleavage. This enhanced delivery potently restores mitochondrial membrane potential and ATP production, scavenges excess ROS, and inhibits endothelial apoptosis. Consequently, Fuc-TP@MA-5 significantly attenuates pulmonary edema, vascular hyperpermeability, and inflammatory cytokine storm in a murine model of LPS-induced ALI, outperforming free MA-5. Our work establishes a targeted mitochondrial functional improvement strategy, offering a promising and intelligent nanotherapeutic paradigm for treating ALI and other vascular inflammatory disorders. Encapsulated black rice anthocyanins by mildly heat-induced egg yolk immunoglobulin and fucoidan with electrostatic interaction to enhance stability and delay the oxidation of yak meat

Xuepeng Li,Fuyao Li,Yu Zeng,Hong Zhang,Xuepeng Lv,Yiwei Tang

This study enhanced the stability of black rice anthocyanins (ANS) by gently inducing the formation of electrostatic encapsulation particles through the synergistic interaction of egg yolk immunoglobulin (IGY) and fucoidan. Results showed that after heat induction at 40 °C, the increasing α-helices content in the protein facilitated its binding with anthocyanins, and effectively improved thermal stability by 20.16 %. Additionally, fucoidan effectively reduced anthocyanin degradation caused by illumination (15.01 %) and vitamin C (46.87 %) by forming of a dense surrounding layer on the complex's surface through electrostatic interactions with IGY, which promoted the aggregation of protein and anthocyanin, while maintaining a high anthocyanin retention rate and total reducing capacity during in vitro digestion. Moreover, the results of yak meat storage demonstrated that the stable complex structure of fucoidan/egg yolk immunoglobulin complex facilitated the slow release of anthocyanins, thereby effectively delaying lipid oxidation during the preservation of yak meat. This study provides a theoretical basis for enhancing the stability of anthocyanins.

Exploration of an efficient and universal extraction strategy for fucoidan with glycolipid-lowering activity from typical brown algae: The importance of ball milling combined with acid treatment

Qianni Chen,Lianzhu Lin,Mouming Zhao

Fucoidan possesses a variety of biological activities beneficial for human wellness. It is a good candidate for ameliorating obesity and diabetes. However, high-purity fucoidan products are limited due to the co-extraction of impurities i.e. alginate, protein and small molecular substances. In this study, the optimal preparation strategy for high-purity fucoidan from a template brown alga ( Saccharina japonica ) was screened out by the exploration of key and auxiliary steps. The key step was confirmed as moderate-temperature acid extraction and the auxiliary steps were fixed on ethanol refluxing and ball milling pretreatment. The fucose content of S. japonica fucoidan increased by 7.15 times and the sulfate group content increased by 4.87 times compared with those of fucoidans prepared by classic high-temperature acid extraction. Ball milling-assisted moderate-temperature acid extraction performed great effectiveness and universality in the extraction of high-purity fucoidans derived from four brown algae including S. japonica , Undaria pinnatifida, Sargassum fusiforme, and Fucus vesiculosus . Glycolipid-lowering activities of four fucoidans were confirmed by their interactions with intestinal bile acid, cholesterol micellar, glucose and probiotics including butyrate-producing bacteria, Bacteroides , Bifidobacterium , and Lactobacillus . Our study provided an efficient and universal extraction process for high-purity fucoidan with glycolipid-lowering activity from typical brown algae.
