Characterization and Quantification of Luteolin-Metal Complexes in Aqueous Extract of Lonicerae Japonicae Flos and Huangshan Wild Chrysanthemum

Weilan Cai,Yunhao Xiong,Manman Han,Zhimin Li,Liang Peng,Hongyi Zhang,Qin Zou,Lin Wu,Qingling Ye,Linfeng Liao

Luteolin is a flavonoid compound widely found in vegetables, fruits, and medicinal plants. In this study, the reaction conditions for luteolin and five metal ions (Ca2+, Mg2+, Zn2+, Fe3+, and Cu2+) to form complexes in hot water were optimized, which was at a molar ratio of 1 : 1 for luteolin and metal ions at 90°C in a volume of 20 mL for 2 h, and the ability of luteolin to form complexes with Cu2+ was the strongest. The DPPH scavenging test showed that luteolin exerted a dose-dependent effect on the clearance of free radicals; luteolin-Cu2+ complexes and luteolin-Fe3+ complexes accentuated the clearance of free radicals. Furthermore, we used high performance liquid chromatography (HPLC) to analyze luteolin in samples from two medicinal plants, obtained from the dissolution of aqueous extracts in two different solvents. The results showed that the peak areas for luteolin in the samples dissolved in 20% formic acid-methanol were significantly larger than those from the samples dissolved in methanol alone, with increases in the peak area being 135.6% (Lonicerae Japonicae Flos), and 161.16% (Huangshan wild chrysanthemum). The aforementioned results indicate that complexes formed from organic compounds and metal ions are present in the decoction of a plant.

Layer-by-layer assembled magnetic molecularly imprinted nanoparticles for the highly specific recovery of luteolin from honeysuckle leaves

Xuemeng Tian,Ruixia Gao,Yue Wang,Yulian He,Sameer Hussain,Jake Heinlein,Jiahao Tian,Lisa D. Pfefferle,Xiaoshuang Tang,Yuhai Tang

Nowadays, the recovery of added-value compounds by pre-treatment and extraction has become imperative in the recycling and transformation of bio-waste. Hence, to achieve efficient bio-waste recycling and avoid the drawbacks of solvent and time consumption in traditional pre-treatment, a green and facile method of efficient separation and enrichment of luteolin (LTL) from honeysuckle leaves has been developed via the green synthesis of novel magnetic molecularly imprinted polymers (Fe3O4-LTL-MIPs). Fe3O4-LTL-MIPs were prepared in aqueous solution using polyethyleneimine and dopamine as eco-friendly bifunctional monomers via layer-by-layer assembly onto the amino-modified Fe3O4 carriers. The thin sandwich-biscuit-like imprinting layer in Fe3O4-LTL-MIPs provides sufficient binding sites with enhanced uniformity and accessibility for the template molecules, thus facilitating excellent recognition ability and rapid mass transfer. The obtained nanomaterials show homogeneous morphology, good crystallinity, satisfactory superparamagnetism, ultra-fast adsorption kinetics, high recognition ability, and excellent reusability. Moreover, Fe3O4-LTL-MIPs as solid-phase extraction adsorbents exhibit high efficiency for the separation and enrichment of LTL from the honeysuckle leaves combined with HPLC analyses. Therefore, the Fe3O4-LTL-MIPs could be potentially exploited as a feasible and efficient way to meet the goals of green chemistry for selective recycling and enriching target compounds from complicated bio-waste matrices.

Boronate affinity imprinted hydrogel sorbent from biphasic synergistic high internal phase emulsions reactor for specific enrichment of Luteolin

Shucheng Liu,Gaohui Lu,Hongxiang Ou,Ruonan Shi,Jianming Pan

The dynamic coexistence of heterostructures is crucial for the synergistic function of molecularly imprinted polymers (MIPs) derived from high internal phase emulsions (HIPEs). In this work, hydrophilic boronate affinity imprinted hydrogel sorbents (H-UIO-66-NH 2 -IHIPEs) were prepared by biphasic synergistic HIPEs droplet reactors filled with reactive microencapsulation system, and used to capture and separate cis -diol containing luteolin (LTL) from complex extraction samples with high selectivity. As the main solid emulsifier, UiO-66-NH 2 , prototype zirconium-based metal–organic frameworks (MOFs) greatly improves the mechanical performance of the hydrogel, whilst preventing overuse of surfactants. Space-confined formation of imprinted sites in the external phase is realized in the presence of hydrophilic acrylamide phenylboric acid monomer (H-BA), which endows the specific affinity with pH responsiveness to LTL . In addition, the filled microinclusion compound containing elastic monomer octadecyl methacrylate (SMA) and functional monomer glycidyl methacrylate (GMA) simultaneously added interfacial cross-linking reaction to provide stable pore volume and pore shape. Combined with these excellent properties, H-UIO-66-NH 2 -IHIPEs showed fast capture kinetics (75 min) and large uptake amount (39.77 mg g −1 ) at 298 K, and confirmed the existence of a uniform chemisorption monolayer. Moreover, excellent recyclability of 6.24% loss in adsorption amount after five adsorption–desorption cycles was observed. Finally, the LTL content of the purified product (about 97.38%) was higher than that of the crude extract (about 85.0%). This study sheds a new light for the design of novel imprinted hydrogel sorbents combined with binary synergistic components.

Electrochemical sensor for facile detection of trace luteolin based on thio-β-cyclodextrin functionalized graphene/gold nanoparticles hybrids

Tingxuan Wu,Zhiguang Liu,Yujing Guo,Chuan Dong

In this work, a simple and highly sensitive electrochemical method was developed to detect luteolin based on thio-β-cyclodextrin functionalized graphene/gold nanoparticles modified glassy carbon electrode (SH-β-CD-GNs/AuNPs/GCE). The peak currents of luteolin on the SH-β-CD-GNs/AuNPs/GCE can be greatly enhanced compared to the SH-β-CD-GNs/GCE and the bare GCE. It suggests that the nanocomposite not only shows the excellent electrical properties of GNs and AuNPs but also exhibits higher enrichment ability of cyclodextrins via the host-guest interaction between cyclodextrin and luteolin . At the SH-β-CD-GNs/AuNPs/GCE, the peak currents increased linearly with the concentration of luteolin in the range of 10.0   pM to 10.0   μM. The detection limit was estimated to be 3.3   pM (S/N   =   3), which was more sensitive or comparable with the previous electrochemical sensors for luteolin detection. The analytical performance of this sensor has been evaluated for the detection of luteolin in human serum with satisfactory results. Furthermore, the method might open up a new possibility by using the nanohybrids to monitor other flavonoids which can form host–guest complexes with cyclodextrins.

Green and integrated recovery of luteolin from peanut shells using ionic liquid-microwave extraction and macroporous resin purification

Xiaoyu Si,Siwen Zhang,Lulu Li,Zunlai Sheng

Ionic liquids enable green and efficient luteolin extraction from peanut shells. • Molecular dynamics reveals hydrogen bonding extraction mechanism. • NKA-9 macroporous resin efficiently enriches and purifies luteolin. • Recycled ionic liquid retains over 90% extraction efficiency. • Integrated process supports circular bioeconomy development. Peanut shells, an abundant agricultural by-product, represent a promising source of the bioactive flavonoid luteolin. This study developed a green and efficient strategy for luteolin recovery by integrating ionic liquid-based microwave-assisted extraction (IL-MAE) with macroporous resin purification. The IL-MAE process was optimized using response surface methodology (RSM), achieving a maximum yield of 3.36 mg/g under optimal conditions: [BMIM][BF₄] concentration of 2 mol/L, liquid-to-solid ratio of 27 mL/g, microwave power of 800 W, and irradiation time of 120 s. Molecular dynamics simulations revealed that hydrogen bonding between luteolin and the ionic liquid governs the extraction mechanism. The crude extract was further purified using NKA-9 macroporous resin, which exhibited high adsorption capacity (2.10 mg/g) and desorption efficiency (93.57%). Dynamic adsorption studies established optimal conditions: sample pH 3.0, loading volume of 1.95 bed volumes (BV), flow rate of 2 BV/h, and temperature of 25 °C. Efficient desorption was achieved by sequential elution with water, 50% ethanol, and 70% ethanol (5 BV each), yielding a final luteolin purity of 84.1%. Notably, the ionic liquid was successfully recycled with over 90% of its initial extraction efficiency retained after multiple cycles. This integrated approach offers a sustainable and scalable pathway for valorizing agricultural waste into high-value bioactive compounds within a circular bioeconomy framework.

Ultrasensitive electrochemical sensor for luteolin based on MOFs-derived metal oxide/alloy-doped nitrogen-containing porous carbon composites

Jiejun Li,Liang Wang,Caiyu Ge,Zuwen Zheng,Yijing Wang,Pengcheng Zhao,Junjie Fei,Yixi Xie

As a natural flavonoid with diverse pharmacological activities, luteolin has shown attractive potential in various applications such as drug discovery, food preservation, health care products, and cosmetics. The precise quantification of luteolin is not only crucial for optimizing its therapeutic efficacy, but also for ensuring compliance with safety standards, as its biological activity largely depends on concentration thresholds and excessive intake may lead to adverse reactions. Here, we have successfully developed an electrochemical sensing platform for luteolin using a simple synthesis strategy. The core of this platform lies in the composite of metal oxides/alloys and nitrogen-doped porous carbon obtained after pyrolysis, whose unique porous architecture and rich pyrrolic nitrogen structure provide efficient adsorption sites for luteolin, while the diverse active centers embedded in the composite significantly enhance its electrocatalytic efficacy. The carefully constructed FeCo-NC-800/GCE electrochemical sensing platform exhibits excellent performance in detecting luteolin. More importantly, we have successfully applied this sensing platform to the detection of luteolin in actual samples such as peanut shells, honeysuckle, and chrysanthemum, which verified the accurate identification of luteolin by FeCo-NC-800/GCE in complex environmental backgrounds and demonstrated its great potential in practical applications.

Luteolin mitigates renal ischemia-reperfusion injury via anti-inflammatory, anti-apoptotic, and Nrf2/HO-1-mediated antioxidant effects

Ji-Ping Wei,Bo Zhao,Zi-Jian Jiang,Peng-Yu Wang,Yan Xu,Ning Ding,Yuan-Yuan Hu,Wei Wang,Bo-Tao Jiang

Renal ischemia/reperfusion (I/R) injury is the most common cause of acute kidney injury (AKI). It can progress to chronic injury and subsequently to chronic kidney disease (CKD) via the renal fibrosis pathway. Luteolin is one of the most commonly occurring flavonoids and exhibits potential therapeutic activity against various pathophysiological processes. In this study, we investigated the protective role of luteolin in counteracting renal I/R injury and its potential mechanisms through systematic network pharmacology, molecular docking, and in vivo experimental studies. Using network pharmacology, we constructed and analyzed a luteolin-renal I/R injury target network. We assessed the relationship between luteolin and renal I/R injury targets using molecular docking analysis. Subsequently, we established a rat model of AKI to CKD transition using unilateral ischemia/reperfusion injury (UIRI), and detected changes in the expression of related proteins using biochemical indices. Network pharmacological analysis and molecular docking showed that luteolin affected renal I/R injury through multiple targets and pathways. As demonstrated by in vivo experiments, luteolin significantly attenuated renal I/R-induced oxidative injury by inhibiting renal lipid peroxidation in rats through the modulation of the nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) pathway. Luteolin attenuated the levels of relevant inflammatory markers, significantly upregulated the synthesis of apoptosis-related proteins, and downregulated the expression of anti-apoptotic proteins. Our results suggest that luteolin effectively inhibited oxidative damage, inflammation, apoptosis, and fibrosis caused by renal I/R injury, thus exerting a nephroprotective effect. The antioxidant effects of luteolin may be related to the regulation of Nrf2/HO-1 signaling.

Interaction between hyaluronic acid and phospholipid bilayer and its influence on stability and bioavailability of luteolin-loaded liposomes

Le Cheng,Ming Zhang,Lin Dong,Yifei Wang,Jianguo Dong,Fang Wu,Chenyan Zheng,Yumeng Ma,Zihan Wang,Ran Wang,Yixuan Li,Bing Fang

The poor solubility of luteolin limits its food applications, and while liposomes offer a potential delivery solution, their phospholipid bilayer membranes are unstable. To overcome these dual challenges, this study developed hyaluronic acid (HA)-coated luteolin liposomes (HA-LUT-lips). HA-LUT-lips exhibited a core-shell structure, high encapsulation efficiency, and good dispersibility. Fourier transform infrared spectroscopy (FTIR) confirmed HA interacted with liposomes via hydrogen bonds, while Raman spectroscopy (RS) showed HA reduced membrane fluidity and enhanced lipid lateral packing. Thermogravimetric (TG) analysis and differential scanning calorimetry (DSC) revealed that HA-LUT-lips were more thermally stable than LUT-lips. HA coating also enhanced ionic strength and storage stability, slowing luteolin release during simulated digestion. HA coating can resist the oxidation of liposomes and delay the leakage of luteolin. These findings suggest HA-coated liposomes were a promising delivery system for luteolin in functional foods and nutritional enhancers.

Protective Effects of Luteolin on Glaesserella parasuis-Induced Injury: An In Vitro Study with Porcine Vascular Endothelial Cells

Pu Guo,Xuwen Liu,Xiaoyi Li,Awais Ihsan,Zhongyuan Wu,Shulin Fu,Chun Ye,Yinsheng Qiu,Xu Wang,Qirong Lu,Yu Liu

Background:Glaesserella parasuis(GPS) is a conditional pathogen that colonizes the upper respiratory tract in pigs and causes Glässer’s disease, resulting in high morbidity and mortality in piglets. GPS infection increases the vascular endothelial permeability, but the mechanism has not been fully elucidated. Luteolin (Lut) is a naturally occurring flavonoid found in plants such as vegetables, herbs, and fruits, but its potential to treat the increased vascular endothelial permeability caused by GPS infection has not been evaluated. Results:This study revealed that GPS infection induces increased vascular endothelial permeability in porcine iliac artery endothelial cells (PIECs) by increasing the gene expressions of tumor necrosis factor (TNF), interleukin 6 (IL-6), IL-8, and IL-1β, and by regulating F-actin cytoskeleton reorganization. Mechanistically, GPS infection or Cluster of differentiation 44 (CD44) overexpression significantly increased the expressions of vascular-endothelial-permeability-related proteins (CD44; vascular endothelial growth factor (VEGFA); matrixmetalloProteinase-3 (MMP-3); MMP-9; and SRC proto-oncogene, non-receptor tyrosine kinase (c-Src)) and increased the vascular endothelial permeability; these changes were alleviated by a Lut treatment or CD44 silencing in the PIECs.Conclusions:This study comprehensively illustrates the potential targets and molecular mechanism of Lut in alleviating the GPS-induced increase in vascular endothelial permeability. The CD44 pathway and Lut may be an effective target and antibiotic alternative, respectively, to prevent the increased vascular endothelial permeability caused by GPS.

Dual function AgNPs@UiO-66 based ratiometric electrochemical sensor for rapid and sensitive determination of luteolin in plants

Zixuan Wang,Juan Peng,Houlin Li,Ganghui Chu

Luteolin is a variety of bioactive flavonoids, it is of great significance to quickly and accurately determine the content of luteolin. This paper established a simple and rapid ratiometric electrochemical sensor for the sensitive determination of luteolin. Nano-flowering zirconium-based metal-organic frameworks (UiO-66) were synthesized and silver nanoparticles (AgNPs) were directly grown on the surface of UiO-66 to prepare AgNPs@UiO-66 composites. Then, AgNPs@UiO-66 was drop-coated on the electrode surface to prepare the ratiometric electrochemical sensor. AgNPs@UiO-66 has the dual function of providing a stable internal reference signal and enhancing the electrochemical signal. Under optimal conditions, the peak current of luteolin (I Luteolin ) increased with increasing luteolin concentration, while the peak current of AgNPs@UiO-66 (I AgNPs@UiO-66 ), which is used as an internal reference signal, remained relatively stable. I Luteolin /I AgNPs@UiO-66 showed a good linear relationship in the range of 0.07 - 0.9 μM and 0.9 - 40 μM, and the limit of detection (LOD) was 0.017 μM (S/ N = 3). The sensor detected luteolin in peanut shells with recoveries ranging from 96.7 % to 107 %, which agreed with ultraviolet-visible spectroscopy. These results demonstrate the possibility of this ratiometric electrochemical sensor for detecting luteolin in real life.

Sensitive Detection of Luteolin Using Thioglycolic Acid–Capped Cadmium Telluride/Cadmium Sulphide Quantum Dots as the Fluorescent Probe

Yizhong Shen,Shaopu Liu,Zhengqing Liu,Youqiu He

A sensitive and simple method for the determination of luteolin (LTL) was developed based on the fluorescence quenching effect of LTL for thioglycolic acid–capped (TGA-capped) CdTe/CdS quantum dots (QDs). Under optimum conditions, a good linear relationship was obtained from 0.3 to 20.0 µg · mL−1 with a correlation coefficient of 0.9972, and the detection limit was 7.2 ng · mL−1. The fluorescence quenching mechanism has been proposed on the basis of electron transfer supported by ultraviolet-visible (UV-Vis) absorption, fluorescence (FL) spectroscopy. The proposed method was successfully applied to the determination of LTL in commercial capsules and human urine samples. It manifested several advantages such as high sensitivity, short analysis time, low cost, and ease of operation.

A Double Network Composite Hydrogel with Self-Regulating Cu2+/Luteolin Release and Mechanical Modulation for Enhanced Wound Healing

Yue Li,Yunpeng Wang,Yuanyuan Ding,Xi Fan,Liansong Ye,Qingqing Pan,Bowen Zhang,Peng Li,Kui Luo,Bing Hu,Bin He,Yuji Pu

Designing adaptive and smart hydrogel wound dressings to meet specific needs across different stages of wound healing is crucial. Here, we present a composite hydrogel, GSC/PBE@Lut, that offers self-regulating release of cupric ions and luteolin and modulates mechanical properties to promote chronic wound healing. The double network hydrogel, GSC, is fabricated through photo-cross-linking of gelatin methacrylate, followed by Cu2+-alginate coordination cross-linking. On one hand, GSC allows for rapid Cu2+ release to eliminate bacteria in the acidic pH environment during inflammation and reduces the hydrogel’s mechanical strength to minimize tissue trauma during early dressing changes. On the other hand, GSC enables slow Cu2+ release during the proliferation stage, promoting angiogenesis and biocompatibility. Furthermore, the inclusion of pH- and reactive oxygen species (ROS)-responsive luteolin nanoparticles (PBE@Lut) in the hydrogel matrix allows for controlled release of luteolin, offering antioxidant and anti-inflammatory effects and promoting anti-inflammatory macrophage polarization. In a murine model of Staphylococcus aureus infected wounds, GSC/PBE@Lut demonstrates exceptional therapeutic benefits in antibacterial, anti-inflammatory, angiogenic, and tissue regeneration. Overall, our results suggest that smart hydrogels with controlled bioactive agent release and mechanical modulation present a promising solution for treating chronic wounds.
