Preparation of Cholesteryl-Modified Aminated Pullulan Nanoparticles to Evaluate Nanoparticle of Hydrophobic Degree on Drug Release and Cytotoxicity

Huabing Yuan,Wu Zhong,Rong Wang,Ping Zhou,Yu Nie,Wei Hu,Xiaojun Tao,Ping Yang

The formation of nanoparticles (NPs) and surface properties such as size and charge are affected by the amphiphilic property of polymer, which is vital for evaluating their function. Here, we synthesized cholesteryl-modified aminated pullulan polymers (CHPNs) with different amounts of cholesterol succinate (CHS). We characterized the three hydrophobically modified polymers (CHPN1, CHPN2, and CHPN3) (CHS: ) by Fourier transform infrared spectrometry. Dynamic light scattering (DLS) was used to measure particle size and zeta potential of CHPN NPs. The particle sizes of the three NPs CHPN1, CHPN2, and CHPN3 were 178.0, 144.4, and 97.8 nm, respectively. The particle size was related to the cholesteryl substitution of polymers to a certain extent: the stronger the hydrophobicity, the smaller the particle size. In 48 h, the drug release for CHPN3 and CHPN1 NPs was 57.8% and 72.7%. Thus, the NPs showed good sustained drug release: the greater the degree of hydrophobic substitution, the better the sustained release. The cytotoxicity findings were reversed: CHPN1 NPs, with low hydrophobic substitution, showed the best inhibition of Lewis lung cancer cells.

Fabrication and characterization of crosslinked pea protein isolated/pullulan/allicin electrospun nanofiber films as potential active packaging material

Xiwen Jia,Xin Li,Jinhai Zhao,Baohua Kong,Hao Wang,Qian Liu,Hui Wang

In this study, pea protein isolated/pullulan electrospun nanofiber films (PPI/PUL NFs) were successfully cross-linked by heating through Maillard reaction . PPI/PUL NFs were characterized by scanning electron microscopy (SEM), fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and water contact angles (WCAs). The morphological analysis illustrated that the cross-linking degree of these NFs varied with the glucose content, and the maximum glucose content was 3.0% (w/v). The fiber structure was broken down when the glucose content exceeded 3.0% (w/v), which was attributed to partial swelling and fusion of nanofibers after cross-linked by glucose. FTIR results indicated that the Maillard reaction occurred between the PPI and glucose during the thermal crosslinking process. Cross-linking dramatically improved the thermal stability, WCAs, insolubility and barrier properties of PPI/PUL NFs, which were needed to be solved urgently in biobased food packaging material. The cross-linked PPI/PUL NFs with a 2.0% glucose showed the most hydrophobic surface (WCAs of 104.5° and water vapor transmission rate of 29.3%). The antimicrobial activity of allicin-loaded PPI/PUL NFs (PPI/PUL-AC NFs) was evaluated, and the maximum size of the inhibition zone of cross-linked PPI/PUL-AC NFs against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) was 16.5 mm and 12.8 mm, respectively. This study not only offered an effective pathway to protect the biological activity of allicin under heating conditions by electrospinning but also provided a reference on novel crosslinking approach of PPI/PUL NFs, which allowed PPI/PUL-AC NFs to be a highly promising platform for antimicrobial food packaging.

A novel pullulan oxidation approach to preparing a shape memory sponge with rapid reaction capability for massive hemorrhage

Weishi Zheng,Zheng Zhang,Yuyu Li,Lujie Wang,Feiya Fu,Hongyan Diao,Xiangdong Liu

Traditional hemostatic materials on the market have considerable limitations for coping with massive bleeding. Herein, we developed an injectable hemostatic sponge via a pullulan oxidation approach. After an oxidation using NaIO 4 at room temperature, the resulting oxidized pullulan provides aldehyde groups to cross-link with the residual hydroxyl groups via the acetal reaction, leading to a hydrogel for lyophilization, thereby producing a porous sponge having highly interconnected macro-pores 89.4–195.4 μm in size. Experimental results show that the oxidized pullulan (OP) sponge possesses good biocompatibility and hemocompatibility. The optimized sponge, [email protected] (prepared using 0.5:1 mass ratio of NaIO 4 and pullulan), exhibited a high fluid absorption ratio, excellent shape memory property, and robust mechanical elasticity. In addition, the [email protected] sponge realized a remarkable hemostatic property, which can be reflected by a lower whole blood clotting index and better aggregation capacities of both blood cell and platelet when compared to commercial gelatin sponge and medical gauze. Furthermore, in vivo hemostatic results demonstrate that [email protected] sponge can accomplish rapid hemostasis times for a rat femoral artery injury model (mean value of 40 s) and liver volume defect injury model (mean value of 33.8 s). In contrast, the commercial gelatin sponges presented 230 s and 100 s under the same test condition, respectively. Our data also show that the [email protected] sponge provides a good antibacterial effect on both Escherichia coli and Staphylococcus aureus . This study demonstrated a great application potential using pullulan-based material for massive hemorrhage cases.

Insights into ultrasonic treatment on the properties of pullulan/oat protein/nisin composite film:mechanical, structural and physicochemical properties

Lixin Kang,Qiufang Liang,Huanxin Chen,Qiusuo Zhou,Zhuzhong Chi,Arif Rashid,Haile Ma,Xiaofeng Ren

This study developed two novel food packaging films, oat protein/pullulan (Op/Pul) and Nisin-loaded oat protein/pullulan (Nis@Op/Pul) films. Ultrasound was introduced to improve its mechanical, structural and physicochemical properties. The Op/Pul film has lower light transmittance, water vapour and oxygen permeability (OP) and improved film uniformity than pure oat protein and pullulan film. The addition of Nisin led to a significant decrease in the composite films' transparency, moisture content, and total soluble matter (TSM). The ultrasound treatment significantly increased the elongation at break and transparency of Nis@Op/Pul film by 18.37% and 8.03% and decreased its TSM and OP by 8.33% and 2.78%, respectively, compared to the conventional method. The structure analysis shows ultrasound enhances intermolecular hydrogen bonding, reduces the crystallinity and formed a more regular, uniform surface. Moreover, the Nis@Op/Pul film prepared by ultrasound treatment could effectively delay the decay and deterioration of fresh strawberries and prolong their shelf life.

Multi-scale ultrasound induced composite coacervates of whey protein and pullulan polysaccharide on emulsion forming and stabilizing mechanisms

Abdul Qayum,Arif Rashid,Qiufang Liang,Lixin Kang,Zahoor Ahmed,Muhammad Hussain,Muhammad Safiullah Virk,John-Nelson Ekumah,Xiaofeng Ren,Haile Ma,Song Miao

A non-destructive technique known as multi-scale ultrasound (MSU) was employed to modify the emulsion consisting of glycosylated bovine whey protein (WP) and pullulan (Pu). To assess the effect on the structural and emulsifying properties of the WP-Pu, the formulated emulsion, was treated with divergent MSU at (single: 20 kHz, 40 kHz, and 60 kHz; dual: 20–40 kHz, 40–60 kHz, and 20–60 kHz; and tri: 20–40-60 kHz) frequency for a duration of 30 min. The tri-frequency, treated emulsion showed improved emulsifying stability compared to the control and MSU-treated single, and dual-frequency samples, as indicated by the particle size, structural morphology, and adsorbed protein. The molecular docking and numerous spectral analysis provided evidence that WP can undergo successful phenolation. This modified form of WP then interacts with Pu through various forces, including H-bonding and other mechanisms, resulting in the formation of a composite emulsion. The rheological properties revealed that both the control emulsion and the MSU-treated emulsion exhibited non-Newtonian pseudoplastic flow behavior. This behavior is characterized by shear thinning, where the viscosity decreases with increasing shear rate. The shear rates tested ranged from 1 to 300 1/s, additionally, the degree of crystallinity increased from 18.2° to 19.4°. Overall, the tri-frequency effect was most pronounced compared to single and dual-frequency. Ultrasonication, an emerging non-thermal technology, proves to be an efficient approach for the formulation of WP-Pu composites. These composites have significant potential for use in drug delivery systems and functional foods.

Development of functional, sustainable pullulan-sodium alginate-based films by incorporating essential oil microemulsion for chilled pork preservation

Ze Wei,Lingli Huang,Xinyu Feng,Feng Cui,Ruijie Wu,Qingjun Kong,Keyu Sun,Jianhua Gao,Jun Guo

Developing safe, eco-friendly, and functionally edible packaging materials has attracted global attention. Essential oils, can be incorporated into packaging materials as antioxidant and antibacterial agents. However, their high volatility and discontinuous film matrix issues may cause a rough film surface, limiting the application in food packaging. In this study, thyme essential oil microemulsion (TEO-M) was prepared and incorporated into a pullulan-sodium alginate (PS) film. The TEO-M incorporation endowed the PS film with antioxidant and UV protection properties. The antioxidant activities of the TEO-M-incorporated PS film were significantly better than those of the TEO-C (thyme essential oil coarse emulsion)-incorporated PS film. In comparison to TEO C, the distribution of TEO-M in the film is more uniform. Lipid oxidation and the growth of microorganisms in chilled pork were inhibited by incorporating TEO-M at a concentration of 50 mg/mL in the PS film (PS-50M). After 10 days of storage at 4 °C, the total viable count (TVC) of chilled pork preserved in the PS-50M material was significantly reduced compared to the control group ( P  < 0.05). This study shows that incorporating TEO-M in the PS film provides a method for applying essential oils in food packaging, which may have great potential in the food industry.

Effects of grape seed extract on the properties of pullulan polysaccharide/xanthan gum active films for apple preservation

Meixia Zheng,Yujing Zhu,Yuanhong Zhuang,Kok Bing Tan,Jianfu Chen

Grape seed extract (GSE) was added to pullulan polysaccharide (PP)/xanthan gum (XG) as composite film (PP/XG/GSE or PXG). The observed composite morphology indicated their biocompatibility. Sample PXG100 (contain 100 mg/L GSE) demonstrated the best mechanical properties, with tensile strength of 16.62 ± 1.27 MPa, and the elongation at break of (22.60 ± 0.48)%. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activity of PXG150 were the highest at (81.52 ± 1.57)% and (90.85 ± 1.54)%, respectively. PXG films also demonstrated inhibitory effects on Staphylococcus aureus , Escherichia coli , and Bacillus subtilis . The PXG films could also prolong the shelf life of fresh-cut apples because it could decrease the rate of weight loss and retain more vitamin C and total polyphenol even on the 5 th day. The weight loss rate of PXG150 was decreased from (8.58 ± 0.6)% (control) to (4.15 ± 0.19)%. It was able to achieve vitamin C and total polyphenol retention rate of 91 % and 72 %, respectively, which was significantly higher that the control sample. Therefore, GSE had contributed in enhancing the antibacterial, antioxidant properties, mechanical strength, UV protection and water resistance in PXG composite films. This effectively extend the shelf life of fresh-cut apples, which it will be an excellent food packaging material.

Lipid phase behavior governs microstructural retention and sustained α-tocopherol release in pullulan composite films for walnut kernel preservation

Xueli Liu,Xiaoxue Wang,Junxia Xia,Bing Qi,Kuizhang Yao,Dayong Zhou,Liang Song

Lipid phase behavior governs microstructural evolution of nanoscale emulsions. • Ordered viscoelastic fluid states enable nanodroplet stabilization. • Nanoscale emulsion architecture is preserved within solid pullulan matrices. • Embedded lipid nanodomains function as steric obstacles to retard solute diffusion. • Phase-engineered composite films intercept oxidative cascades in walnut kernels. The entrapment of lipophilic bioactives within hydrophilic polymer matrices presents physical challenges regarding phase separation and uncontrolled release. This study evaluates a strategy based on tuning lipid phase behavior to modulate the microstructural evolution of α-tocopherol-loaded nanoemulsions and derived pullulan films. A 40% coconut oil proportion induced a transition from a disordered supercooled liquid to an ordered viscoelastic fluid. This state facilitated the formation of stable nanodroplets with a mean diameter of 18.59 nm and a polydispersity index of 0.24 by reducing the interfacial tension to 5.07 mN·m −1 and the critical micelle concentration to 0.01 g·L −1 . Conversely, a 60% coconut oil proportion caused microstructural crowding and interfacial bridging. This highly homogeneous architecture was preserved within the solid film, yielding a tensile strength of 26.51 MPa, an elongation at break of 15.14%, and reduced permeabilities for oxygen (0.14 × 10 −4  g·m −1 ·s −1 ) and water vapor (1.04 × 10 −13  kg·m −1 ·s −1 ·Pa −1 ). Kinetic modeling confirmed a quasi-Fickian diffusion profile in which the embedded nanodomains functioned as steric obstacles. This tortuous diffusion path restricted solute mobility, suppressing the initial release and achieving an 88.88% retention of tocopherol activity. Application on walnut kernels demonstrated preservation efficacy, evidenced by a 39.94% reduction in peroxide value and a 38.56% decrease in thiobarbituric acid reactive substances compared to the control group. Regulating internal lipid phase behavior provides a predictable microstructural framework for developing active packaging systems to extend the shelf life of lipid-rich foods. Fabrication of complex hydrogels based on pullulan, nanocellulose and carbon dots using photo-Fenton process to eliminate doxycycline contamination

Meng Liu,Wenzhuo Lv,Fan Zeng,Canhui Wang,Xin Guo,Bo Zhou,Yiqiang Wu

Overcoming the inherent activity–stability trade-off in photo-Fenton systems requires advanced material architectures that integrate multifunctional synergies. In this study, we developed a multifunctional hydrogel, termed PCCF, fabricated by confining Fe 3 O 4 @polydopamine (PDA) nanoparticles within a three-dimensional (3D) pullulan–nanocellulose matrix specifically tailored for efficient doxycycline (DOX) elimination. PCCF exhibited a hierarchically engineered structure based on three synergistic strategies: (1) a pullulan (Pu)–cellulose nanofibrils (CNF)@carbon dots (CD) co-network with a hierarchically porous architecture (surface area: 230.6 m 2 /g), enabling ultrahigh adsorption capacity (Q m  = 272.9 mg/g); (2) PDA-mediated nanoparticle immobilization to achieve spatial uniformity and reduce catalyst leaching (< 5 ppb Fe after five cycles); and (3) an optically transparent and mechanically resilient hydrogel framework (94.2 % transmittance at 500 nm, tensile strength: 41.7 kPa, elongation at break: 84.5 %). These integrated features synergistically improved the photo-Fenton degradation rate constant of PCCF by 3.03-fold compared to Fe 3 O 4 nanoparticles, while maintaining 90.4 % catalytic efficiency after five cycles. This work establishes a Pu hydrogel-based structural paradigm that harmonizes catalytic activity with long-term stability, offering a versatile blueprint for high-performance environmental remediation systems.

Conductive polyacrylamide/pullulan/ammonium sulfate hydrogels with high toughness, low-hysteresis and tissue-like modulus as flexible strain sensors

Hongyao Ding,Bingyao Wang,Xu Yang,Jie Liu,Wei Sang,Xuewen Li,Yuefang Wen,Hui Li,Xiaodong Shen

Conductive hydrogels have great potential for applications in flexible wearable sensors due to the combination of biocompatibility, mechanical flexibility and electrical conductivity. However, constructing conductive hydrogels with high toughness, low hysteresis and skin-like modulus simultaneously remains challenging. In the present study, we prepared a tough and conductive polyacrylamide/pullulan/ammonium sulfate hydrogel with a semi-interpenetrating network. Ammonium sulfate promoted the formation of low-energy-dissipating motifs between polymer chains, reinforcing the gel matrix and resulting in excellent mechanical properties, including a high stretchability of 2063 %, a high strength of 890 kPa, and a high toughness of 4268 kJ/m 3 . The hydrogen bonds formed within the network endowed the gels with low-hysteresis under deformation. The unique semi-interpenetrating network structure provided the gels with a tissue-like low modulus. Additionally, the resulting hydrogels exhibited a high conductivity of 2.39 S/m and excellent anti-freezing properties, making them suitable for flexible strain sensors. These sensors demonstrated high sensitivity over a broad strain window of 0.1–1500 %, enabling the detection of various human motions and the recognition of different languages. These findings emphasize the potential of the composite hydrogels as wearable strain sensors for flexible devices.

Chitosan-pullulan edible coating loaded with dihydromyricetin: Enhanced antioxidant activity and barrier properties to prolong Cantonese sausages' shelf-life

Mingyue Yang,Xiaoyan Xu,Xingguo Tian

Cantonese sausages are susceptible to oxidative deterioration during storage. Compared with synthetic antioxidants, dihydromyricetin (DMY) is a natural active substance with various functions such as antioxidant and antimicrobial. In this study, edible coating solutions loaded with DMY were prepared based on chitosan (CS) and pullulan (PUL) to prolong the shelf-life of Cantonese sausages. FT-IR and XRD results showed good compatibility between DMY and CS-PUL. It was found that 8 % DMY was physically cross-linked with CS and PUL mainly through hydrogen bonding, which enhanced the barrier and mechanical properties of the films. Meanwhile, the antioxidant property, antimicrobial efficiency and thermal stability of the films were also significantly improved ( p  < 0.05). The results of storage experiments of Cantonese sausages showed that the 8 % DMY-CP coating solution could effectively retard the lipid oxidation of Cantonese sausages at 25 °C, and maintain the appearance and flavour of products at storage on the 28th d. This study proved DMY's potential as a natural alternative to synthetic additives in polysaccharide-based edible coatings, providing a green and sustainable solution to combat lipid oxidation in meat products.

Intra-articular injection of platinum nanozyme-loaded silk fibroin/pullulan hydrogels relieves osteoarthritis through ROS scavenging and ferroptosis suppression

JunWei Feng,Xia Deng,Peng Hao,ZongDong Zhu,Tao Li,XinWei Yuan,Jiang Hu,Yue Wang

Reactive oxygen species (ROS)-mediated ferroptosis plays a critical role in the development of osteoarthritis (OA). Consequently, it is speculated that anti-ferroptosis agents could represent a novel therapeutic strategy for managing OA. In this study, a hydrogel incorporating platinum (Pt) nanozyme was synthesized by dispersing Pt nanoparticles (NPs) within a matrix of silk fibroin (SF) and oxidized pullulan (oxPL). This hydrogel allows for a substantial and sustained release of up to 30 days. The gelation time (from 140.3 ± 42.3 s to 460.0 ± 40.0 s), swelling capacity (from 57.7 ± 3.8 % to 24.0 ± 7.0 %), and degradation rate (from 60.3 ± 4.7 % to 32.0 ± 4.6 %) of the hydrogels can be modulated by adjusting the Pt NP content. The Pt@SF/oxPL hydrogel effectively eliminates ROS due to its catalase-like and superoxide dismutase-like enzymatic properties. In vitro studies demonstrated that Pt@SF/oxPL efficiently mitigated the process of ferroptotic cell death in chondrocytes. More critically, intra-articular administration of Pt@SF/oxPL showcased therapeutic advantages by both protecting and stimulating the regeneration of cartilage throughout the progression of OA. Collectively, this study suggests that Pt@SF/oxPL hydrogels could potentially serve as an effective treatment for OA, presenting a novel nanozyme-based therapeutic approach for this condition.
