A method based on a fiber-optic interferometer to study the pH-dependent interaction between graphene oxide and single-stranded DNA chains is carried out with the assistance of a scanning electron microscope and a confocal laser scanning microscope. The various wavelength shifts of the interferometric fringes of the transmission spectrum reveal the different strengths of interaction between graphene oxide and single-stranded DNA in various pH environments. The present work demonstrates the feasibility of optical fibers for studying the interaction between graphene oxide and biomolecules. It provides a potential means to understand the intermolecular interactions. This technique might effectively supplement the existing tools.

Vascularization-ossification coupling is pivotal for bone defect repair. Neovascularization guides the migration of osteogenic precursor cells, and osteoblasts in the perivascular area support vascular regeneration through the secretion of cytokines. Delivery systems are being engineered to orchestrate the coupling of vascularization and ossification precisely. However, most delivery systems lack sensitivity to the inherent repair environment, hampering timely bone formation. This study developed a vascular environment-responsive biomaterial: a true bone ceramic/small intestine submucosa (TBC/SIS) scaffold loaded with nanoswitch and Apt19s-functionalized nanoparticles. This scaffold is designed to release the functionalized nanoparticles via an "unfolding–refolding" nanoswitch based on base pairing and protein-nucleic acid recognition. The TBC/SIS scaffold initiates neovascularization, prompting endothelial cells to secrete nucleolin. The nanoswitches detect nucleolin and trigger the release of nanoparticles, thereby creating a responsive vascular environment. Moreover, when modified with aptamer-19s, the nanoparticles can attract mesenchymal stem cells to perivascular areas, facilitating spatial coupling with blood vessels. Encapsulating miRNA-26a in nanoparticles enhances osteogenic activity and stimulates the secretion of vascular endothelial growth factor. As vascular endothelial cells infiltrate, nanoparticles are continuously released from the scaffold, establishing a positive feedback loop that promotes vascularization-ossification coupling and ultimately enhances bone regeneration. Our findings provide valuable insights for designing intelligent materials that utilize endogenous stimuli to enhance spatiotemporal interactions between vascularization and ossification. Further in-depth studies on the expression of nucleolin in vivo will continue to refine this promising strategy.

In this study, an electrochemical aptasensor dedicated to the sensitive detection of kanamycin (KAN) was constructed based on an Nb. Bbv CI-driven DNA walker-enabled signal amplification strategy. The sensor employed a gold electrode modified with polyethyleneimine-wrapped, nitrogen-doped multi-wall carbon nanotubes decorated with gold nanoparticles (N-MWCNTs-PEI-AuNPs). This composite was used to modify the gold electrode, providing an enhanced conductive interface and increasing the specific surface area for aptasensor construction. Meanwhile, a zirconium-based metal-organic framework decorated with gold nanoparticles (AuNPs@NH₂-UiO-66) was used both as an electrocatalyst and as a carrier for hairpin DNA (HP2), exhibiting strong catalytic activity toward the redox mediator thionine. In differential pulse voltammetry measurements, at the surface of the electrode, thionine undergoes a reversible, pH-dependent two-electron/two-proton transfer process at approximately −0.25 V on the electrode surface, generating a well-defined reduction current that serves as the analytical signal. Upon addition of KAN, the aptamer selectively binds to the analyte and dissociates from its complementary DNA (cDNA). The freed cDNA subsequently hybridizes with HP2 anchored on AuNPs@NH₂-UiO-66, thereby activating the Nb. Bbv CI-driven DNA walker, catalyzing the site-specific cleavage of HP2 and releasing multiple AuNPs@NH₂-UiO-66 labels. These released probes then hybridize with the captured hairpin DNA (HP1) fixed on the N-MWCNTs-PEI-AuNPs-modified gold electrode. The synergistic electrocatalytic effects of NH₂-UiO-66 and AuNPs significantly accelerate the electron transfer kinetics of thionine, leading to amplified voltammetric current responses that correlate proportionally with KAN concentration. The aptasensor demonstrated highly sensitive performance, including a linear detection range of 10 pM to 5 μM and a detection limit of 1.65 pM and a quantitation limit of 5.46 pM. The aptasensor also showed satisfactory selectivity and reproducibility and was successfully applied for KAN determination in real milk samples. This approach provides a robust signal amplification strategy and offers a promising tool for the rapid and sensitive detection of KAN in real-world samples.

Bark lice are common in terrestrial ecosystems, yet the poor taxonomic study of them renders species identification difficult. In this study, we investigated the species identification of the bark louse genus Neostenopsocus using DNA barcodes (675 bp fragment of the mitochondrial COI gene). We examined newly collected specimens and the type materials of some valid species of Neostenopsocus , redescribing 13 valid species and proposing 39 new synonyms. A total of 110 sequences of Neostenopsocus species were obtained from newly collected specimens and public data. The results show that the COI barcode is sufficient to distinguish most Neostenopsocus species, whereas more molecular marks and distribution data are necessary to identify the closely similar species. The family Stenopsocidae exhibits a rich biodiversity in China, yet species identification based on morphological characters remains problematic due to insufficient study and absent molecular data. DNA barcoding has been widely employed for rapid species identification in insects, particularly through standardized COI gene sequencing. In this study, we conducted species delimitation for Neostenopsocus in China based on the morphological characters and DNA barcodes, comprising 20 morphological species with 110 barcodes. We redescribed 13 species from China and proposed 39 new synonyms. This study supports the applicability of DNA barcoding for species identification in Neostenopsocus . The K2P distance between the morphological species of Neostenopsocus ranges from 0.0051 to 0.2040, with most exceeding 0.10. The maximum intraspecific divergence reaches 0.0778 in N. nepalensis . The external morphology of Neostenopsocus shows considerable structural uniformity, and the genitalia provide limited diagnostic value for species identification. Instead, the marking patterns of the head and forewing are considered to be useful for species identification. Therefore, delimiting closely related species demands an integrated taxonomy approach that combines extensive geographical sampling, detailed morphological analysis, and nuclear DNA data.

Smoking, a pivotal environmental risk factor, drives diseases including lung cancer through genetic and epigenetic alterations. While 2-Naphthylamine (2-NA), a tobacco-derived carcinogen, is established as a bladder carcinogen via DNA damage, its role in lung carcinogenesis remains mechanistically uncharacterized despite epidemiological associations. This study identifies bronchial epithelium as a direct target of 2-NA, demonstrating dose-dependent DNA damage in 16HBE cells (peak at 250 μM, 12 h; P < 0.05), accompanied by S-phase arrest, apoptosis, reduced proliferation, and reactive oxygen species (ROS) generation. 2-NA exposure upregulated the expression of circSHOC1, which is a circular RNA derived from the SHOC1 gene, in a dose-dependent manner. Functional assays revealed that circSHOC1 overexpression exacerbated 2-NA-induced DNA damage by enhancing ROS production and 8-hydroxy-2′-deoxyguanosine (8-OHdG) accumulation, whereas knockdown attenuated these effects. Mechanistically, circSHOC1 interacted with mitochondrial protein SLC25A3 (Solute Carrier Family 25 Member 3), a key oxidative stress regulator; SLC25A3 knockdown mitigated DNA damage ( P < 0.01), and co-transfection experiments confirmed SLC25A3 as a critical mediator of circSHOC1-driven genotoxicity. Collectively, this work provides the first experimental evidence that 2-NA induces bronchial epithelial DNA damage via a circSHOC1-SLC25A3-ROS axis, supporting a novel mechanism for tobacco-associated lung carcinogenesis and highlighting 2-NA as a potential pulmonary carcinogen.

Cutaneous squamous cell carcinoma (cSCC) is a common type of skin tumor with high incidence and significant disease burden. Current clinical treatments face challenges such as high recurrence rates and drug resistance, making the discovery of novel drugs and therapeutic targets critically important. Previous studies have confirmed that Mangiferin(MF) can inhibit the proliferation of various tumors, promote apoptosis, and regulate cellular oxidative stress. This study investigated the therapeutic effects and mechanisms of MF in cutaneous squamous cell carcinoma (cSCC). CCK-8 and EDU assays demonstrated that MF significantly inhibited A431 cell proliferation, induced apoptosis, and caused G0/G1 phase arrest. Network pharmacology, molecular docking, and molecular dynamics simulations identified HSP90α as a direct target of MF, which was highly expressed in clinical cSCC tissues and validated through CETSA and DARTS experiments. Genetic manipulation experiments revealed that HSP90α knockdown promoted apoptosis, while its overexpression accelerated proliferation and attenuated MF's effects. Mechanistically, MF induced ROS generation, leading to mitochondrial and DNA damage, which could be reversed by the antioxidant NAC. Furthermore, ROS-mediated activation of the MAPK pathway contributed to apoptosis and DNA damage. In vivo, MF effectively suppressed cSCC growth without causing organ toxicity. In conclusion, MF inhibits cSCC progression both in vitro and in vivo with minimal toxicity, primarily through targeting HSP90α and activating the ROS/MAPK pathway.

An aptamer-labelled fluorescent nanoprobe-integrated shell-isolated nanoparticle-enhanced fluorescence spectroscopy (SHINEFs) had been proposed for selective and sensitive detection of Tc. In the study, CdTe quantum dots were used as a fluorescent source; silica-coated silver nanoparticles (Ag@SiO2) were used as the SHINEFs material for signal amplification. The maximum SHINEFs was obtained with a 9 nm silica spacer. The linear range of the nanoprobe was from 0.2 to 400 μM, and the detection limit was 16.2 nM. Furthermore, our synthesized nanoprobe was applied to detect Tc in milk with a recovery rate of 96.50–101.80%. These results showed that the as-prepared nanoprobe was a rapid detection tool for Tc with high sensitivity and selectivity. The aptamer-modified nanoprobe can also be applied to the analysis of other substances by altering the aptamer.

Herba Ecliptae (HE) is a typical Chinese herbal medicine used in China for 1500 years. In the study, HE was extracted by various solvents to prepare five HE extracts. They were observed to possess a protective effect against ×OH-induced DNA damage, and scavenging effects on ×OH radical, ×O 2 − radical, DPPH×(1,1-diphenyl-2-picrylhydrazyl) radical, and ABTS× + (2,2′-azino-bis(3-ethyl-benzothiazoline-6-sulfonic acid) radical, and reduce Cu 2+ ion. The contents of total phenolics and wedelolactone in five extracts were determined respectively using Folin-Ciocalteu method and HPLC method. To identify which chemical component can be responsible for its effects, the correlation graphs between chemical contents and antioxidant abilities (1/IC 50 values) were plotted to calculate correlation coefficients (R values). Finally, MTT assay revealed that two HE extracts could effectively protect mesenchymal stem cells (MSCs) against ×OH-induced damage at 3-30 μg/mL. On the basis of mechanistic analysis, we concluded that: ( i ) HE can effectively protect against ×OH-induced damages to DNA and MSCs, thereby HE may have a therapeutic potential in MSCs transplantation or prevention of many diseases; ( ii ) the effects can be mainly attributed to total phenolics (R = 0.678) especially wedelolactone (R = 0.618); ( iii ) they exert antioxidant action via hydrogen atom transfer (HAT) and sequential electron proton transfer (SEPT) mechanisms.

Samarium doped cerium dioxide (CSO) nanostructures possessing a shell-like morphology were synthesised by a simple hydrothermal method. A novel nanocomposite membrane, comprising of CSO nanostructures, chitosan (CH) and a room temperature ionic liquid (RTIL) was deposited onto an indium-tin oxide (ITO) electrode for developing a DNA biosensor related to the breast cancer gene. The properties of the CSO and nano-composite membrane were studied by thermogravimetry (TG), X-ray diffraction (XRD), energy dispersive spectroscopy (EDS), scanning electron microscopy (SEM), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The hybridization capacity of the DNA biosensor was studied by differential pulse voltammetry (DPV) using [Fe(CN)6]3−/4− as an indicator. Under optimal conditions, the fabricated DNA biosensor could quantify a wide range of target DNA concentrations over the range of 1 × 10−13 to 1 × 10−6 M with good linearity (R = 0.9981) and a low detection limit of 1.56 × 10−14 M (3σ). Results showed that the fabricated shell-like CSO nanostructures and RTIL could enhance the electrical conductivity synergistically, which has great potential application in sensitive electrochemical biosensors.

The association of a plasmonic nano-antenna with single-molecule FRET technique presents new prospects to investigate the dynamics of biological molecules. However, the presence of a plasmonic nano-antenna significantly modifies the FRET rate and efficiency; this makes its applicability to the prevalent single-molecule FRET experiments unclear. Herein, using gold nanoparticle antennas of different sizes and DNA hairpins labelled with FRET pairs (Cy3 and Cy5) as the model system, we performed experiments to study the folding dynamics of single DNA hairpins at various salt concentrations. Our results indicate that gold nanoparticle antennas can enhance single-molecule fluorescence of Cy3 and Cy5 up to 3–5 folds, substantially reduce the FRET efficiency, and alter the obtained FRET efficiency histograms. However, the folding dynamics of DNA hairpins remains unaffected, and the correct kinetic and dynamic information can still be extracted from the seriously modified FRET efficiencies. Therefore, our experiments demonstrate the feasibility and compatibility for applying plasmonic nano-antennas to the mostly used single-molecule FRET assays, which provide a broad range of possibilities for the future applications of these nano-antennas in studying various essential biological processes.

DNA has proven of high utility to modulate the surface functionality of metal–organic frameworks (MOFs) for various biomedical applications. Nevertheless, current methods for preparing DNA–MOF nanoparticles rely on either inefficient covalent conjugation or specific modification of oligonucleotides. In this work, we report that unmodified oligonucleotides can be loaded on MOFs with high density (∼2500 strands/particle) via intrinsic, multivalent coordination between DNA backbone phosphate and unsaturated zirconium sites on MOFs. More significantly, surface-bound DNA can be efficiently released in either bulk solution or specific organelles in live cells when free phosphate ions are present. As a proof-of-concept for using this novel type of DNA–MOFs in immunotherapy, we prepared a construct of immunostimulatory DNA–MOFs (isMOFs) by intrinsically coordinating cytosine–phosphate–guanosine (CpG) oligonucleotides on biocompatible zirconium MOF nanoparticles, which was further armed by a protection shell of calcium phosphate (CaP) exoskeleton. We demonstrated that isMOFs exhibited high cellular uptake, organelle specificity, and spatiotemporal control of Toll-like receptors (TLR)-triggered immune responses. When isMOF reached endolysosomes via microtubule-mediated trafficking, the CaP exoskeleton dissolved in the acidic environment and in situ generated free phosphate ions. As a result, CpG was released from isMOFs and stimulated potent immunostimulation in living macrophage cells. Compared with naked CpG–MOF, isMOFs exhibited 83-fold up-regulation in stimulated secretion of cytokines. We thus expect this isMOF design with soluble CaP exoskeleton and an embedded sequential “protect–release” program provides a highly generic approach for intracellular delivery of therapeutic nucleic acids.

Background To investigate the potential mechanisms of chemoradiotherapy resistance in patients with bladder cancer. Methods We assessed three bladder cancer cell lines (5637, J82, and TCCSUP) for their sensitivity to chemoradiotherapy. Reverse transcription quantitative PCR (RT-qPCR) was used to detect the expression of specific genes after chemoradiotherapy or combined with olaparib. The Genome Cancer Atlas (TGCA) database was used to analyze possible radioresistance-related genes and the relationship between their expression and bladder cancer survival and prognostic indicators. Results The 5637 cell line showed the most significant sensitivity to chemoradiotherapy. The expression levels of DNA damage repair genes in 5637 cells did not significantly change after chemoradiotherapy. In contrast, the expression levels of BRCA1 and RBBP8 genes significantly increased in J82 and TCCSUP cells after chemoradiotherapy. After combination with olaparib, which is a poly (ADP-ribose) polymerase inhibitor that initiates DNA repair, 5637 cells were significantly inhibited by chemoradiotherapy. However, chemoradiotherapy inhibition on J82 cells was weakened when combined with olaparib. A high reactive expression of BRCA1 and RBBP8 after combination with olaparib suggested that olaparib was ineffective because it did not induce synthetic lethality in which inhibiting PARP by olaparib coincided with suppression of BRCA1/2 expression result in cancer cell death. Conclusions The expression levels of RBBP8 and BRCA1 genes were associated with sensitivity to radiotherapy and chemotherapy in bladder cancer, and an increase in reactive expression after treatment led to worse sensitivity. Therefore, the reactive expression levels of BRCA1 and RBBP8 after chemoradiotherapy may be useful in evaluating the efficacy of olaparib combination therapy.
