Enhanced dissolution and oral bioavailbility of cinacalcet hydrochlorde nanocrystals with no food effect

Xiaoqing Xu,Guoguang Chen,Yaning Li,Jingjing Wang,Jun Yin,Lili Ren

This study was aimed at improving oral bioavailbility and reducing the food effect of cinacalcet hydrochlorde (CINA), a poorly soluble drug for the treatment of chronic kidney disease, by preparing its nanocrystals (NCs) utilizing the precipitation–ultrasonication method. Based on the single factor method and Box–Behnken design, with the particle size and polydispersity index (PDI) as indexes, the optimal formulation was achieved. It was investigated that the particle size and PDI of the NCs prepared on the basis of optimal formulation were 244 ± 2 nm and 0.168 ± 0.001, respectively. The NCs were solidificated by lyophilization. Scanning electron microscopy, differential scanning calorimetry and x-ray powder diffraction were used to characterize the CINA-NCs, and there was no crystalline change during preparation and lyophilization. The CINA-NCs capsules prepared with 30% (w/v) MCC, 8% (w/v) CCNa and 2% (w/v) talcum powder by orthogonal experimental design presented an enhanced in vitro dissolution rate in four media compared with commercial tablets Sensipar® and raw material. The raw material, blank NCs and CINA-NCs were confirmed to be non-toxic to Caco-2 cells when the drug concentration was below 250 μg ml−1. In the in vivo pharmacokinetic study, the Cmax (the peak concentration of CINA in plasma) and AUC0–t (area under curve by trapezoidal area method) of the CINA-NCs capsules were approximately 1.90-fold and 1.64-fold greater than that of Sensipar® in the fasted state. Overall, this nanotechnology is a promising way to optimize the dosage form of CINA oral administration.

Influence of the TiO2 crystalline phase of MnOx/TiO2 catalysts for NO oxidation

Zhongyi An,Yuqun Zhuo,Chao Xu,Changhe Chen

Mn-based catalysts impregnated on TiO 2 with different crystalline phases (rutile (R), anatase (A), and P25 (P)) were studied for the oxidation of NO to NO 2 . 10%MnO x /TiO 2 (P) was the most active catalyst, giving 83% of NO conversion at 300 °C and a GHSV of 20000 h −1 . The catalyst activities were in the order MnO x /TiO 2 (P) > MnO x /TiO 2 (A) > MnO x /TiO 2 (R). The catalysts were characterized by X-ray diffraction, field emission scanning electron microscopy, X-ray photoelectron spectroscopy, H 2 temperature-programmed reduction, and O 2 temperature-programmed desorption. Compared to anatase and rutile, P25 gave better dispersion of MnO x on the support surface, suppressed the agglomeration of catalyst particles, and also produced more Mn 2 O 3 , which was more active for the oxidation of NO. In addition, P25 enhanced the reduction of MnO x , especially for Mn 2 O 3 , and the formation of easily desorbed O 2 − generated from the Mn 3+ –O bond.

Cascade-Targeted Nanoplatforms for Synergetic Antibiotic/ROS/NO/Immunotherapy against Intracellular Bacterial Infection

Xiaomei Dai,Xiaojun Liu,Xingxing Wang,Yongjie Zhang,Yu Li,Feng Gao

Intracellular bacteria in dormant states can escape the immune response and tolerate high-dose antibiotic treatment, leading to severe infections. To overcome this challenge, cascade-targeted nanoplatforms that can target macrophages and intracellular bacteria, exhibiting synergetic antibiotic/reactive oxygen species (ROS)/nitric oxide (NO)/immunotherapy, were developed. These nanoplatforms were fabricated by encapsulating trehalose (Tr) and vancomycin (Van) into phosphatidylserine (PS)-coated poly[(4-allylcarbamoylphenylboric acid)-ran-(arginine-methacrylamide)-ran-(N,N′-bisacryloylcystamine)] nanoparticles (PABS), denoted as PTVP. PS on PTVP simulates a signal of “eat me” to macrophages to promote cell uptake (the first-step targeting). After the uptake, the nanoplatform in the acidic phagolysosomes could release Tr, and the exposed phenylboronic acid on the nanoplatform could target bacteria (the second-step targeting). Nanoplatforms can release Van in response to infected intracellular overexpressed glutathione (GSH) and weak acid microenvironment. l-arginine (Arg) on the nanoplatforms could be catalyzed by upregulated inducible nitric oxide synthase (iNOS) in the infected macrophages to generate nitric oxide (NO). N,N′-Bisacryloylcystamine (BAC) on nanoplatforms could deplete GSH, allow the generation of ROS in macrophages, and then upregulate proinflammatory activity, leading to the reinforced antibacterial capacity. This nanoplatform possesses macrophage and bacteria-targeting antibiotic delivery, intracellular ROS, and NO generation, and pro-inflammatory activities (immunotherapy) provides a new strategy for eradicating intracellular bacterial infections.

Lignin-based nanoenzyme doped hydrogel for NO-enhanced chemodynamic therapy of bacterial infections

Ye Zhang,Yutong Wen,Yingying Li,Zhiqi Li,Zhen Wang,Bing Fan,Qiaoyu Li,Wanchen Cai,Ying Li

The infections triggered by bacteria often cause wound deterioration, and the development of safe and effective antimicrobial treatment is always highly desired. In this paper, naturally derived lignin was aminated to generate surface group functionalized lignin nanoparticles (NPs), efficiently loading Ag NPs and adsorbing L-arginine, to construct lignin-based nanoenzyme (SALL), which achieves synergistic antimicrobial treatment by chemodynamic therapy and NO gas therapy, while the dose of silver was decreased. The SALL is dispersed in eco-friendly hydrogel constructed using keratin and chitosan through realigned disulfide bond and diverse intermolecular interactions, the prepared SALL@K/C hydrogel has ideal rheological property, and strong adhesion capacity facilitating active bacteria capture, ensuring that the bacteria were immobilized within the effective range of reactive oxygen species and NO. The inhibition rates of S. aureus and E. coli were 98.5 % and 97.8 %, respectively. Meanwhile, the SALL@K/C hydrogel could achieve the delivery of hydrophobic drug curcumin for inhibiting inflammation. The study highlights a well-designed nanoenzyme-loaded hydrogel with excellent antibacterial, hemostatic, and antiinflammatory properties, offering new ideas for nanoenzyme design and antimicrobial hydrogel development.

Synergistic integration of metallic Bi and defective TiO2 for enhanced photocatalytic NO removal

Guangfu Yi,Ke Zhang,Sha Kang

The construction of hybrid catalysts is an effective method to enhance catalytic performance, but research on the positioning of the load is limited. In this study, we synthesized two photocatalytic materials by controlling the bismuth-titanium ratio and the site value of Bi modification in the catalyst. The surface-modified photocatalyst, namely 1:0.9 Bi/TiO 2 , exhibits a photocatalytic efficiency of 68.9 % for the abatement of ppb-level NO in the presence of visible irradiation. This efficiency was significantly superior to that of the corresponding one-step synthesized Bi/TiO 2 (48.7 %) and the TiO 2 (18.6 %). Furthermore, it promotes nitrate formation and suppresses the generation of toxic NO 2 . Capture experiments indicate that the same active species are involved in both surface-loaded and uniformly loaded catalysts. The differences in their performance can be attributed to the higher vacancy concentration in the surface-loaded catalysts and the different local coordination environments of vacancies in the catalysts, altering the adsorption capability of oxygen molecules. This work offers fresh perspectives for the generation of efficient photocatalysts.

Design and Evaluation of a Nonaqueous FeCl2/PEG200 System for Industrial NO Capture

Lantong Gou,Xiandong Hao,Zhiyong Xu,Wenbo Zhao

Nitric oxide (NO), a major air pollutant from coal-fired power plants, demands an efficient capture for environmental and industrial applications. Ferrous-based absorbents exhibit promising potential for NO capture due to rapid Fe2+-NO complexation. Herein, a novel nonaqueous FeCl2/PEG200 mixture absorbent was designed by employing FeCl2 as the Fe2+ source and PEG200 as the solvent. The FTIR/UV–vis spectrum confirmed Fe2+-NO complex formation and suggested possible reaction mechanisms. Dynamic sorption experiments were systematically conducted to evaluate sorption performance. Kinetic and thermodynamic analyses obtained activation energy, enthalpy change, and entropy change information. Most importantly, a new sorption kinetic model has been established, which can be used to predict the NO sorption capacity at any temperature and partial pressure. Cyclic sorption─desorption testing demonstrated a high desorption efficiency (>91%) and robust cyclic stability over 4 consecutive cycles. This work establishes the FeCl2/PEG200 system as an efficient, reusable, and scalable candidate for industrial NO capture.

Construction of a sustainably NO-releasing RGDC grafted chitosan oriented peripheral nerve scaffold

Wenxuan zhao,Yuzhe Chen,Xiaopei Wu,Xianzheng Dong,Honglian Dai

Peripheral nerve injury repair is hindered by an imbalanced regenerative microenvironment involving inflammation, oxidative stress, and inadequate blood supply. Traditional drugs are limited by poor blood-nerve barrier penetration and lack of multifunctionality. Nitric oxide (NO) is crucial for axonal growth, synaptic plasticity, and neuroprotection, offering anti-inflammatory, antioxidant and pro-angiogenic potential. However, uncontrolled NO release causes toxicity. To leverage NO therapeutically, sustained delivery is essential. This study developed a multifunctional nerve graft using a modified chitosan hydrogel grafted with RGDC peptide, featuring an internal orientation microstructure. Glutathione was incorporated as a precursor for the NO donor S-nitrosoglutathione (GSNO). This system promotes cell adhesion/migration and reduces oxidative stress. Results show the hydrogel is non-cytotoxic and synergistically enhances cellular adhesion. GSNO loading significantly reduces cellular reactive oxygen species (ROS) levels in human umbilical vein endothelial cells (HUVECs), facilitating their migration and angiogenesis. The GSNO-incorporated neural scaffold achieved sustained NO release and demonstrated significant antioxidant capacity. This innovative approach presents a promising strategy for clinical application in peripheral nerve injury repair.

Shape-persistent COF-derived functional carbon microspheres for No-carrier added 177Lu separation

Guoquan Ma,Haiyue Peng,Xisheng Fan,Yang Li,Jing Gao,Yingjiang Hu,Bo Li,Yuanyou Yang,Jingsong Zhang,Lijian Ma,Ning Liu,Jiali Liao,Feize Li

Porous carbon materials stemming from covalent organic frameworks (COFs) have emerged as pivotal candidates in various fields. However, no COF or relevant derivative has stepped into the recovery of no-carrier added (NCA) 177 Lu, one of the most important radionuclides in modern medicine, from irradiated target. In this work, shape-persistent COF-derived carbon microsphere (COF-CMS) was successfully synthesized via self-templated carbonization and subsequently functionalized with active component 2-ethylhexyl phosphoric acid mono-2-ethylhexyl ester (P507). The prepared functional COF-CMS@P507 has shown high selectivity and fast adsorption kinetics for Yb 3+ /Lu 3+ in static batch adsorption and great potential in dynamic column separation of Lu 3+ from Yb 3+ . More importantly, based on plenty of tracer experiments and practical investigations, a separation process flow was established to realize the isolation of 177 Lu from an irradiated 176 Yb target. Consequently, 177 Lu product can be effectively obtained in preferable recovery (86%) with high radinuclidic and chemical purity (≥99.99%). The present study provides a new idea for NCA 177 Lu production, which is also valuable for the development of lanthanide separation and functional COFs.

Puffy hierarchical nanonet structured multifunctional membranes for NO degradation and ultrafine dust filtration

Jiesong Tan,Shibo Yu,Jiahao Chen,Jinxin Gan,Feng Zhang,Shengui Ju,Yiqing Zeng,Zhaoxiang Zhong,Weihong Xing

Multifunctional nanofibrous catalytic membranes are used for comprehensive treatment of fine particulate matter (PM) and toxic gaseous contaminants. However, the need for high purification efficiency with low resistance presents a unique challenge. Herein, a method of optimizing the morphology of manganese oxide (MnO x ) on Zr-TiO 2 (TZ) nanofiber membranes was developed to remove ultrafine PM and nitric oxide (NO) efficiently with relatively low pressure drops. After tailoring the hydrothermal solution properties, the MnO 2 ultrafine nanowires (≈22 nm) and Mn 3 O 4 nano-octahedra assembled with scaffold nanofibers formed dual-network structures with high gas permeabilities, superior redox properties, and abundant surface acidities. The resultant M 2 TZ (2-MnO x /Zr-TiO 2 ) catalytic membranes exhibited exceptional NH 3 selective catalytic reduction (NH 3 -SCR) activity (T 90  = 215 °C) with a gas hourly space velocity of 75,000 h −1 , as well as outstanding SO 2 and H 2 O tolerance and long-term stability. Meanwhile, owing to the puffy hierarchical nanonet structure, the M 2 TZ catalytic membranes also show a high filtration efficiency of 99.99% for PM 0.3 removal with a low air resistance of 125 Pa, high air permeability of 1259 m 3  m −2 h −1 kPa −1 , and satisfactory PM capture at 300 °C. This work may shed light on the design of advanced multifunctional membranes for air pollutant purification.

Photocatalytic semi-flexible pavement for NO removal to alleviate urban automobile emissions pollution

Hang Diao,Bo Peng,Wenjing Kuang,Tianqing Ling,Ying Gao,Kaiyuan Luo

The escalating urban air pollution caused by vehicular nitrogen oxides (NO x ) necessitates sustainable solutions. Solar-driven photocatalytic NO removal represents a green approach, yet challenges remain in developing cost-effective photocatalysts with high removal efficiency. This study through the Fe-TiO 2 combined with thermal polycondensation method with g-C 3 N 4 build heterojunction generated Fe-TiO 2 /g-C 3 N 4 photocatalyst, in order to improve its NO catalytic efficiency. The optimized photocatalyst achieves a single-cycle NO removal rate of 52.3 %, demonstrating the 10.7 % improvement compared to pure TiO 2 . Following five cycles, the decay rate is only 5.52 %, which is significantly lower than the 12.45 % of TiO 2 , thus indicating superior long-term stability. Microscopic characterization shows that Fe can induce the generation of oxygen defects inside TiO 2 to promote the separation of photogenerated electron-hole pairs; and induce the formation of a built-in electric field to enhance the separation of photogenerated charges with the heterojunction formed by the construction of g-C 3 N 4 . Moreover, the designed photocatalytic semi-flexible pavement incorporating this photocatalyst exhibits 65.33 % NO removal efficiency per cycle, 10.95 % higher than traditional TiO 2 based systems, with a total decay rate of 1.71 % after five cycles. Additionally, the pavement demonstrates excellent mechanical properties, including fivefold higher rutting resistance than conventional asphalt pavements. This study provides a new sustainable pavement removal scheme for mitigating urban exhaust agglomeration pollution.

“Continuously Touching ROS into NO” by Hemodialysis Membranes for Preventing Hemodialysis-Associated Thrombocytopenia

Jian Ren,Lin Xu,Xianda Liu,Dongmei Tong,Shujing Wang,Shuxiang Zhang,Ran Wei,Weifeng Zhao,Changsheng Zhao

Hemodialysis membranes would easily accelerate excessive oxidative stress, leading to platelet adhesion and aggregation, which cause exacerbation of hemodialysis-associated thrombocytopenia. Notably, nitric oxide (NO) gas exerts an inhibitory effect on platelet activation by targeting key signaling molecules. Hence, we propose a strategy to inhibit platelet activation and adhesion by turning “harmful reactive oxygen species (ROS)” into “beneficial NO”, thereby preventing hemodialysis-associated thrombocytopenia. In this study, polydopamine-l-arginine (PDA-LA) molecules are coated onto the membrane surface to mainly enhance the mitigation of oxidative stress, while oxidized hyaluronic acid-l-arginine (OHA-LA) molecules are stably cross-linked into the membrane to ensure the sustained NO release for inhibition of platelet adhesion. As a result, the reactive oxygen and nitrogen species (RONS) scavenging capacities are improved to follow: H2O2 (50%), 2,2-diphenyl-1-picrylhydrazyl (DPPH•, 85.60%), 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonate) radical (ABTS•+, 82.81%), and O2•– (0.125 U/mg). Surprisingly, the concentration of continuously released NO is still approximately 0.590 μM after six cycles (0.557 μM in the first cycle). Importantly, the platelet adhesion experiment demonstrates a sustained inhibition of platelet adhesion, diminished by a factor of 100 per square centimeter (100/cm2). In a simulated sieving experiment, the BSA rejection ratio (97.52%) and urea sieving ratio (98.38%) both meet the basic requirements for hemodialysis membranes. Thus, the obtained PES/PVA/OA@PA membrane is a promising candidate for preventing hemodialysis-associated thrombocytopenia.

Iron single-atom catalysts confined in covalent organic frameworks for efficient oxygen evolution reaction

Xiao Wang,Lei Sun,Wei Zhou,Li Yang,Guoqing Ren,Hao Wu,Wei-Qiao Deng

The exploration of environmentally friendly and highly efficient oxygen evolution reaction (OER) catalysts is vital to large-scale, electrochemical renewable-fuels generation. Here, we report an iron single-atom catalyst (SAC) confined in a covalent organic framework (Fe-SAC@COF), which constitutes an unusual Fe–NO coordination in the skeleton. The as-prepared Fe-SAC@COF exhibits a high mass activity of 9.20 A mg −1 , which is 1.95 times higher than Ni species of the same coordination and 5.05 times higher than nanoparticulate Fe counterpart. Moreover, it shows, to the best of our knowledge, a record-low overpotential (290 mV) and Tafel slope (40 mV dec −1 ) among the reported atomically dispersed Fe-based catalysts and surpasses the benchmark Ir/C catalyst. The density functional theory calculation shows that the Fe–NO coordination exhibits low binding energy of oxygenated intermediates, which leads to an outstanding electrocatalytic OER performance. This work provides design strategies toward unusually coordinated SACs by prudent COF confinement for advanced electrocatalysis.
