Astaxanthin-containing hollow salt derived from Haematococcus pluvialis protein-chitosan complex with strong antioxidant capacity as a promising salt substitute

Ming He,Ronggang Liu,Chenglong Sun,Miyao Sun,Haitao Wang,Mingqian Tan

Excessive dietary sodium chloride (NaCl) intake is strongly associated with hypertension and an elevated risk of stroke and cardiovascular disease. In this study, astaxanthin-containing hollow salt was prepared using a spray-drying method from an emulsion containing a high concentration of NaCl, stabilized by a complex of Haematococcus pluvialis protein (HPP) and chitosan (CS). The interaction between HPP and chitosan, as well as the effects of varying concentrations of HPP-CS complexes on salt emulsion, were investigated. The HPP-CS complex at a ratio of 1:2 and pH 5 exhibited strong binding and emulsifying abilities. Cryo-scanning electron microscopy revealed that emulsions stabilized by the HPP-CS complex exhibited smaller particle sizes and a more uniform distribution. As the HPP-CS complex concentration increased, the emulsion exhibited higher structural stability, excellent fluidity, and improved thermal stability. The stability of emulsion was significantly improved by HPP-CS complex. These results confirmed the potential application of salt emulsion stabilized by HPP-CS complex in hollow salt preparation. Hollow salt prepared by spray drying exhibited a smooth surface, uniform particle size distribution, and microspheres with a diameter of less than 10 μm. The hollow salt had a sodium content of 0.22 g per gram. Sensory evaluation and electronic tongue results showed that there was no significant difference in saltiness between hollow salt and traditional NaCl under the same mass. High astaxanthin retention and oxidation stability during storage at 50 °C for 15 days were observed in the hollow salt. In addition, the protective effect on fried steaks stored overnight revealed that the astaxanthin-containing hollow salt significantly reduced the lipid hydroperoxide and malondialdehyde content. Therefore, the hollow salt, based on an emulsion stabilized with the HPP-CS complex, may serve as a potential low-salt substitute and dietary supplement.

Extraction of astaxanthin from Haematococcus pluvialis utilizing acidic deep eutectic solvents: A comparison with ionic liquids and organic solvents

Wanting Cheng,Qi’an Yu,Huiling Huang,Kun Hu,Jing Gao

Ionic liquids (ILs) and organic solvents have been extensively utilized for the extraction of astaxanthin (Ast). Nonetheless, the large-scale extraction of Ast still necessitates enhancements in extraction efficiency, environmental sustainability, and a decrease in economic costs. Acidic deep eutectic solvents (DESs) formulated with DL-menthol and fatty acids was designed to extract Ast from Haematococcus pluvialis ( H. pluvialis ). The properties of the DESs, along with its extraction efficiency, antioxidant activity, stability, and recycling were assessed and contrasted with tributyloctylphosphine chloride ([P 4448 ]Cl) and ethanol. Quantum chemical calculation was applied to understand the extraction mechanism of DES. The extraction efficiency peaked at 91.25 % when using a DL-menthol:acetic acid (MAA) mixture at an ultrasonic power of 300 W for 30 min, with a solid-to-liquid ratio of 1:20 g/ml, surpassing that of [P 4448 ]Cl extraction (69.91 %) and ethanol extraction (59.34 %). Furthermore, the extracts demonstrated the highest storage stability after MAA extraction, and MAA also showed the best recycling capabilities as compared with [P 4448 ]Cl and ethanol. However, the DPPH· and intracellular ROS scavenging capacities were slightly lower than those after [P 4448 ]Cl extraction. In general, acidic DESs exhibit the highest extraction efficiency and stability of Ast as compared with ILs and organic solvents, while their antioxidant activity requires further improvement. In addition, 25 % of sugar could be released by enzymatic hydrolysis the H. pluvialis residue to further produce bioethanol. This study will provide a theoretical foundation for the extraction of biomolecules and the design of efficient method to comprehensive utilization of microalgae resources.

Optimally Aligned Nerve Scaffolds with Sustained Astaxanthin Release Improve the Inflammatory Microenvironment through Mitophagy Activation

Xuehan Jin,Shengfu Liu,Jiaqi Fang,Feng Chen,Bo Xu,Liping Nan,Shihong Zhao,Zhong Wu,Zhiyuan Guan,Kun Tao,Junjian Liu

Effective repair of peripheral nerve injury (PNI) depends on the scaffold orientation and immunomodulatory capabilities of functionalized scaffolds, both of which substantially influence nerve regeneration. In this study, composite nerve scaffolds incorporating astaxanthin (AXT) and polycaprolactone (PCL) are developed to investigate the influence of scaffold orientation and blend concentration on cellular behavior, including adhesion, migration, and proliferation. In vitro analysis identifies 0.2% AXT/PCL fabricated at a rotational speed of 400 rpm as the optimal configuration for facilitating directed cell growth and guiding nerve repair. Moreover, the controlled release of AXT improves the microenvironment by preserving mitochondrial homeostasis, promoting mitophagy, and reducing oxidative stress and inflammation. In vivo assessments reveal that the AXT/PCL group (0.2% AXT/PCL-400) achieves better morphological, histological, electrophysiological, and functional recovery than the PCL, AXT/PCL+M0, and AXT/PCL+M4 groups, approaching the outcomes observed in the autograft (Auto) group. Moreover, the AXT/PCL+M4 group demonstrates better regenerative outcomes than the PCL and AXT/PCL+M0 groups, underscoring the critical role of mitophagy in regulating the regenerative microenvironment.

Effects of hydroxyproline supplementation in low fish meal diet on growth, immunity and intestinal health of Litopenaeus vannamei

Menglin Shi,Liangjuan Sun,Liutong Chen,Kangyuan Qu,Beiping Tan,Shiwei Xie

This experiment investigated the impacts of hydroxyproline (Hyp) supplementation in low fish meal diets on growth, antioxidant capacity, immunity and intestinal health of Litopenaeus vannamei . Six diets were formulated for the experiment, with the high fish meal diet containing 25 % fish meal (HF) and the remaining diets incorporating a mixture of soybean protein concentrate (SPC) and Clostridium autoethanogenum protein (CAP) replacing 15 % fish meal. These diets were supplemented with varying levels of Hyp (0 %, 0.2 %, 0.4 %, 0.6 %, and 0.8 %, designated as LF, H1, H2, H3, and H4, respectively). A total of 960 healthy shrimp (0.32±0.00 g) were divided into 6 groups at random; each group had 4 duplicates, each containing 40 shrimp. The shrimp were fed four times a day for a period of 51 days. The results indicated that dietary Hyp supplementation had no significant effects on growth performance and whole body composition of L. vannamei ( P > 0.05). The antioxidant enzyme activities of hemolymph and hepatopancreas were significantly decreased after fish meal was replaced, and were significantly increased after dietary Hyp supplementation ( P < 0.05). The expression of antioxidant-related genes in intestinal and hepatopancreas was significantly increased after dietary Hyp supplementation ( P < 0.05). In the intestine, the gene expression of penaeidins 3 ( pen3 ), dorsal , anti-lipopolysaccharide factor ( alf ) and crustins ( cru ) in the H2 group is the highest, and the gene expression of B-cell lymphoma-2 ( bcl2 ) in the H3 group is the highest ( P < 0.05). The genes expression of tumor necrosis factor-alpha ( tnf-a ), Bcl-2-assoxicated x protein ( bax ) and cytochrome c ( cyc-t ) in LF group reached the maximum ( P < 0.05). Intestinal H&E stained sections showed that low-fish meal diet reduced the mucosal folds height of shrimp, and H2 and H3 groups significantly increased the mucosal fold height of shrimp ( P < 0.05). Therefore, adding Hyp to the diet can improve the antioxidant capacity, immunity and intestinal health of L. vannamei .

New Horizons of Astaxanthin-loaded Akkermansia muciniphila as an Integrated Dietary Supplement: Physicochemical Structures and Gastrointestinal Fate

Ma Xue-Ke,Shao Xin,Xu Tian,Yan Hong,Teng Hui,Chen Zhe-Xin,Hu Meng-Xin

Astaxanthin (ASX) gains wide interest in food and nutritional sciences due to its great antioxidant and anti-inflammatory properties. However, ASX faces challenges such as poor water solubility, low bioavailability, and sensitivity to environmental factors. In this study, we employ Akkermansia muciniphila ( AKK ) with mucoadhesive property as a carrier for ASX to enhance ASX stability and bioaccessibility. The complex multilayered cell envelope of AKK cell provided space for ASX loading. Immersion loading yielded the highest ASX encapsulation efficiency (10.80 ± 2.29 μg/mg in ASX@ AKK group and 11.47 ± 3.52 μg/mg in ASX@ AKK -Heat group), whereas osmoporation loading and vacuum loading yielded ASX encapsulation efficiencies of 4–6 μg/mg. The encapsulated ASX demonstrated improved thermostability, tolerating pasteurization, and was predominantly amorphous in state, interacting with AKK primarily through hydrophobic interactions and partial hydrogen bonding interactions. The ASX@ AKK system facilitated precise ASX release in the intestine and markedly enhanced bioaccessibility, increasing approximately 25.7-fold in simulated intestinal fluid and 77.6-fold in simulated colonic fluid. Importantly, ASX encapsulation did not impair AKK ’s adhesive affinity to mucus. Therefore, the newly developed integrated dietary supplements have both the functional activity of inactivated AKK as a postbiotic and the antioxidant property of the intestine-targeted released ASX, showing dual-functional activities. These findings highlight a paradigm shift in the functional application of probiotics, showcasing their potential as innovative vehicles for targeted nutrient delivery with improved bioaccessibility and synergistic health effects.

Determination of 21 amino acids in grass carp, clam and shrimp by enzymatic hydrolysis coupled with high-performance liquid chromatography and tandem mass spectrometry

Chengwen Liu,Xulong Chen,Yan Ouyang,Liping Sun,Shaoyou Lu

A simple and sensitive method was developed and validated for the simultaneous determination of 21 amino acids in three aquatic products including grass carp ( Ctenopharyngodon idella ), clam ( paphia undulata ) and shrimp ( Metapenaeus ensis ) with high-performance liquid chromatography coupled with ionization tandem mass spectrometry (HPLC-MS/MS). Firstly, aquatic products samples were pretreated by enzymatic hydrolysis and the efficiencies of different hydrolysis conditions were investigated. Then both chromatographic and mass spectrometric conditions were optimized to improve sensitivity and accuracy for the analytes. Under the optimized conditions, the limits of detection (LODs) for 21 amino acids were in the range from 0.02 μg/g to 1.78 μg/g. The correlation coefficients and recoveries were ranged from 0.997 to 0.999 and 80–115 %, respectively, and the relative standard deviation was within the range of 0.6–8.7 %. The results showed that 21 amino acids were all detected in samples. Except for asparagine, glutamine, tryptophan, cystine, cysteine, aspartic acid and glutamic acid, there is no significant difference in the measurement of amino acids by HPLC-MS/MS and amino acid analyzer.

Design of anti-saponification Ca-based catalyst with porous-shell structure from crustacean waste for biodiesel production

Mengjie Zhang,Xiangpu Jiao,Dezhang Ren,Chenxin Huang,Yang Li,Duofu Chen,Zhibao Huo

The rapid development of the biodiesel industry is of significant importance for achieving carbon emission reduction in the transportation sector. However, the presence of trace amounts of water in the feedstock during biodiesel synthesis may lead to severe saponification reactions, thereby reducing the yield of fatty acid methyl esters. In this study, based on our exploration of the characteristic of porous carbon to absorb moisture from the environment, we synthesized a CaO catalyst with porous carbon coating via discarded shrimp shell biomass. Because the carbonization temperature of organic matter in shrimp shells is lower than the decomposition temperature of CaCO 3 , the CO 2 generated during pyrolysis can erode the pyrolytic carbon and form porous structure. The mesoporous carbon shell facilitates the diffusion of oil and methanol molecules while adsorbing moisture in the alcohol phase. This structure effectively minimizes the interaction between CaO and water, thereby suppressing saponification reactions and ensuring the smooth progression of the desired transesterification reaction. Through a series of physicochemical characterizations, the structure and water absorption performance of the catalyst were confirmed. The as-prepared CaO@PC-900 catalyst demonstrated the ability to maintain a biodiesel yield of over 85% in a reaction system with a water content of 3%, significantly surpassing the performance of commercial CaO and previously reported Ca-based solid bases. This work provides a new approach to the design of anti-saponification transesterification catalysts, while also offering a novel pathway for the synthesis of porous carbon from discarded crustacean biomass.

Enhancement of Colorimetric pH-Sensitive Film Incorporating Amomum tsao-ko Essential Oil as Antibacterial for Mantis Shrimp Spoilage Tracking and Fresh-Keeping

Yunxia He,Yuay Yuan,Yuanyuan Gao,Mianhong Chen,Yingying Li,Ying Zou,Liangkun Liao,Xiaotong Li,Zhuo Wang,Jihua Li,Wei Zhou

Anthocyanin-based smart packaging has been widely used for food freshness monitoring, but it cannot meet the requirements of smart films with antibacterial properties. This study aimed to enhance the antibacterial properties of intelligent films by incorporatingAmomum tsao-koessential oil (AEO) for mantis shrimp spoilage tracking and keeping the product fresh. A smart film was designed by introducing AEO and purple potato anthocyanin (PPA) to a polyvinyl alcohol/cellulose nanocrystal (PVA/CNC) polymer matrix. Our findings revealed that APP and AEO imparted the smart film with a favorable oxygen barrier, UV protection, mechanical properties, and antioxidant and pH/NH3-sensitive functions. Interestingly, the PVA/CNC-AEO-PPA film achieved 45.41% and 48.25% bactericidal efficacy againstS. putrefaciensandV. parahaemolyticus, respectively. Furthermore, a visual observation confirmed that the target film (PVA/CNC-AEO-PPA) changed color significantly during mantis shrimp spoilage: rose red—light red—pink—light gray—dark gray. Meanwhile, the PVA/CNC-AEO-PPA film retarded the quality deterioration of the mantis shrimp effectively. The PVA/CNC-AEO-PPA film shows great application potential in mantis shrimp preservation and freshness monitoring; it is expected to become a rapid sensor for detecting seafood quality non-destructively and a multifunctional film for better preservation of product quality.

Cadmium-induced oxidative stress, histopathology, and transcriptomic changes in the hepatopancreas of Fenneropenaeus merguiensis

Zhimin Ju,Charles Brighton Ndandala,Yiguo Lei,Vicent Michael Shija,Junliang Luo,Panmei Wang,Chongqing Wen,Huafang Liang

Cadmium (Cd) is a threatening heavy metal pollutant to aquatic organisms. The hepatopancreas plays a crucial role in combating environmental stress and enhancing the immune defense of aquatic species. The exact mechanism behind the damage induced by Cadmium in the hepatopancreas of Fenneropenaeus merguiensis is unclear. Thus, this study aimed to elucidate the potential mechanisms of Cadmium's impact on the hepatopancreas of F. merguiensis through physiological, histological, and transcriptomic analyses. After 72 h of F. merguiensis exposure to the 0.5 mg/L concentration of cadmium results in the reduction of superoxide dismutase and catalase activities, coupled with an increase in hydrogen peroxide and malondialdehyde levels, which leads to damage in both the gills and hepatopancreas. This causes severe histological changes in the gills and hepatopancreas of F. merguiensis , characterized by irregularities in the gill filaments and ruptures in the basal membrane of the hepatopancreas. Comparative transcriptomic analysis of F. merguiensis revealed the upregulation of several genes associated with pathways related to metabolism (ko01100), antigen processing and presentation (ko04612), and cell apoptosis (ko04215), indicating adaptive response mechanisms to Cadmium exposure. Transcriptomic analysis also showed that Cadmium exposure induced the expression of genes related to detoxification and apoptosis, such as heat shock proteins, metallothionein, and Caspase-3 , suggesting adverse effects on the normal physiological functions of F. merguiensis . Our study provides comprehensive insights into the toxicological mechanisms and impacts of Cadmium on F. merguiensis.

Biocompatible zein-gum Arabic nanoparticles for astaxanthin encapsulation and topical delivery: Preparation and characterization

Wenpeng Song,Wei Wei,Biao Zhou,Ning Lv,Heng Qin,Fei Huan,Xiaoling Zhang

Astaxanthin (AST), a potent antioxidant, holds great promise for skin health and cosmetic applications but is limited by poor water solubility and stability. This study presents the development of zein nanoparticles modified with gum arabic (GA) for the encapsulation of AST, serving as a topical nanocarrier for AST to enhance its dermal bioavailability and topical delivery. The optimized zein-GA-AST nanoparticles (Z-GA-AST) exhibited a spherical nanostructure with a mean particle size of 199.03 ± 4.63 nm (PDI <0.2) and achieved a high encapsulation efficiency of 90.34 ± 1.74 %. FT-IR spectroscopy analysis confirmed the successful encapsulation of AST within the nanoparticles. The AST-loaded nanoparticles exhibited significantly superior antioxidant activity compared to free AST. At an equivalent astaxanthin concentration of 10 μg/mL, the ABTS and DPPH radical scavenging efficiencies of the nanoparticles were at least 153 % and 128 % higher than those of free AST, respectively. The incorporation of GA enhanced the physicochemical stability and astaxanthin retention of the nanoparticles under various conditions, including different salt ion concentrations, temperatures, and storage durations, by facilitating the formation of a more compact nanostructure. The skin permeation behavior of the nanoparticles was evaluated using Franz diffusion cells and porcine skin. The results demonstrated that Z-GA2-AST1 exhibited significantly higher skin retention (48.37 %) and lower astaxanthin permeation (10.20 %) compared to free astaxanthin. This study provides valuable insights into the preparation and characterization of biocompatible nanoparticles for AST encapsulation, could become a potential formulation for topical delivery applications in cosmetics and topical drug delivery systems.

Sphingopyxis sp. YF1 and metabolite astaxanthin alleviate MC-LR-induced kidney injury

Yafang Li,Jia Wei,Xiaofang Liang,Jiangping Wang,Sisi Yan,Lemei Zhu,Zhen Ding,Chunhua Zhan,Xiaoya Ren,Fei Yang

Microcystin-LR (MC-LR) is a widely distributed cyanotoxin that causes kidney injury. Our previous work identified Sphingopyxis sp. YF1 as a novel bacterial strain capable of efficiently degrading MC-LR, but its protective role and mechanisms against MC-LR-induced kidney injury remain unexplored. In this study, the bioactive metabolite of strain YF1 was identified by mass spectrometry. The beneficial effect of strain YF1 and its bioactive metabolite against MC-LR-induced kidney injury was validated in vivo and in vitro . We found strain YF1 effectively ameliorated MC-LR induced kidney disruption and functional impairment. Mechanistically, strain YF1 significantly improved oxidative stress (SOD, CAT, GSH, and MDA) and inhibited pro-inflammatory factors expression (including IL-6 , IL-18 , IL-1β , and TNF-α ) induced by MC-LR. Notably, strain YF1 not only reduce MC-LR level in mice but also produce a bioactive metabolite astaxanthin. Consistently, both in vivo and in vitro experiments demonstrated that astaxanthin alleviated MC-LR-induced kidney injury by modulating antioxidant pathways and inflammatory cascades. This study suggested that strain YF1 could alleviate MC-LR-induced kidney injury by removing MC-LR and generating astaxanthin that mediated antioxidant and anti-inflammatory effects. These findings highlighted microbial-based antidotes for simultaneously eliminating MC-LR and mitigating its harmful effects.

Fabrication and characterization of chitosan/anthocyanin intelligent packaging film fortified by cellulose nanocrystal for shrimp preservation and visual freshness monitoring

Dan Zheng,Shumin Cao,Dengming Li,Yihan Wu,Peijun Duan,Shanshan Liu,Xing Li,Xiaoyu Zhang,Yisheng Chen

In this study, a multi-functional packaging film was fabricated, utilizing the natural polysaccharide chitosan (CS) as the base material, integrating natural blueberry anthocyanin (AN) as pH-responsive indicator, and reinforced with cellulose nanocrystals (CNCs). The implications of addition levels of CNCs on the characteristics of the films were systematically investigated, resulting in that CS-AN-CNCs 9 % film exhibited optimal performance. Specifically, the film showed a substantial enhancement in maximum tensile strength from 15 MPa to 35 MPa; On the other hand, the swelling degree properties, the oxygen permeability and water vapor permeability decreased from 159.2 % to 92.0 %, from 51.7 g/(m 2 d) to 12.2 g/(m 2 d), from 31.6 × 10 −12  g/(m·s·Pa) to 1.6 × 10 −12  g/(m·s·Pa), respectively. Moreover, the CS-AN-CNCs 9 % film exhibited antioxidant, antibacterial, coupled with a color metrically responsive to pH variations, displaying great potential in indicating the shrimp freshness and delaying spoilage. Another notable advantage of the-prepared packaging material lies in its completely biodegradability, therefore meeting the requirement of environmental protection. Therefore, the prepared CS-AN-CNCs film as an intelligent packaging solution with potential applications in food preservation and freshness monitoring applications.
