PEGylated GdF3:Fe Nanoparticles as Multimodal T1/T2-Weighted MRI and X-ray CT Imaging Contrast Agents

Lile Dong,Peng Zhang,Pengpeng Lei,Shuyan Song,Xia Xu,Kaimin Du,Jing Feng,Hongjie Zhang

Contrast agents for multimodal imaging are in high demand for cancer diagnosis. To date, integration of T1/T2-weighted magnetic resonance imaging (MRI) and X-ray computed tomography (CT) imaging capabilities in one system to obtain an accurate diagnosis still remains challenging. In this work, biocompatible PEGylated GdF3:Fe nanoparticles (PEG-GdF3:Fe NPs) were reasonable designed and synthesized as multifunctional contrast agents for efficient T1/T2-weighted MRI and X-ray CT multimodal imaging. Owing to the enhanced permeability and retention effect in vivo, strong T1 contrast, evident T2 contrast, and X-ray CT signals in a tumor lesion can be observed after intravenous injection of PEG-GdF3:Fe NPs. Therefore, PEG-GdF3:Fe NPs could be used as potential multimodal contrast agents for cancer diagnosis.

Longitudinal Multi-tracer PET/CT for Monitoring the Progression of Diabetic Cardiac Autonomic Neuropathy: A Preclinical Study

Jing Gu,Hui Wang,Yulin He

Rationale and Objective This study utilized micro-positron emission tomography/computed tomography (micro-PET/CT) to track the longitudinal and dynamic progression of cardiac autonomic nerve injury in diabetic rats. Our objective was to map the precise timeline of neural damage relative to disease stages, defining a sensitive imaging window for the early diagnosis of diabetic cardiac autonomic neuropathy (DCAN). Materials and Methods Following streptozotocin induction of type 2 diabetes, rats underwent biweekly micro-PET/CT imaging for up to 17 weeks. Sympathetic and parasympathetic innervations were thoroughly evaluated using [ 11 C]C-MDA and [ 11 C]C-donepezil ([ 11 C]C-DNP), respectively. [ 11 C]C-triphenylmethylphosphonium ([ 11 C]C-TPMP) provided a perfusion baseline to characterize neuro-perfusion mismatches. Quantitative tracer uptake was correlated with histopathological remodeling and the myocardial expression of neurotrophic factors (NGF, GAP-43, CNTF) quantified via quantitative real-time polymerase chain reaction (RT-qPCR). Results Longitudinal PET/CT imaging unveiled a distinct chronological cascade of cardiac deterioration. Parasympathetic denervation ([ 11 C]C-DNP depletion) manifested first at week 6, followed by sympathetic denervation ([ 11 C]C-MDA deficit) at week 8. Conversely, myocardial perfusion deficits ([ 11 C]C-TPMP) remained absent until week 16, escalating to severe metabolic dysfunction by week 17. Histopathology confirmed progressive myocardial inflammation, while RT-qPCR indicated a late-stage, yet insufficient, compensatory upregulation of neurotrophic factors. Conclusion This multi-tracer longitudinal investigation establishes that parasympathetic denervation precedes sympathetic impairment during DCAN development, with both deficits predating microvascular and metabolic crises. These insights offer a critical, noninvasive diagnostic window for early DCAN detection and clinical monitoring.

Bo Wu,Shu-Ting Lu,Hui Yu,Ru-Fang Liao,Huan Li,B.V. Lucie Zafitatsimo,Yu-Shuang Li,Ying Zhang,Xiao-Lei Zhu,Hong-Guang Liu,Hai-Bo Xu,Shi-Wen Huang,Zhen Cheng

Multifunctional nanomaterials with simple structure and good biosafety, integrating multimodal imaging and therapeutic functions, can facilitate the development of clinical cancer treatments. Here, a simple but powerful pure bismuth based nanoparticle (Gd-PEG-Bi NPs) was developed from pure Bi NPs and gadolinium-diethylenetriaminepentaacetic acid-bis-tetradecylamide, which not only shows high quality MRI/CT/PAI triple-modal imaging, but can also be a potent photothermal therapy agent under the guidance of the triple-modal imaging. The Gd-PEG-Bi NPs showed good stability and excellent biocompatibility. In vitro and in vivo study demonstrated that Gd-PEG-Bi NPs have ultrahigh X-ray attenuation coefficient, short T1 relaxation time in MRI, and strong PAI signal. Following the imaging diagnosis, the excellent light-to-heat conversion efficiency of Gd-PEG-Bi NPs was capable of suppressing the tumor growth effectively under near-infrared laser radiation in vivo . Such multifunctional nanoparticles were ideal candidates for cancer diagnosis and treatment.

Au/Polypyrrole@Fe3O4 Nanocomposites for MR/CT Dual-Modal Imaging Guided-Photothermal Therapy: An in Vitro Study

Wei Feng,Xiaojun Zhou,Wei Nie,Liang Chen,Kexin Qiu,Yanzhong Zhang,Chuanglong He

Construction of multifunctional nanocomposites as theranostic platforms has received considerable biomedical attention. In this study, a triple-functional theranostic agent based on the cointegration of gold nanorods (Au NRs) and superparamagnetic iron oxide (Fe3O4) into polypyrrole was developed. Such a theranostic agent (referred to as Au/PPY@Fe3O4) not only exhibits strong magnetic property and high near-infrared (NIR) optical absorbance but also produces high contrast for magnetic resonance (MR) and X-ray computed tomography (CT) imaging. Importantly, under the irradiation of the NIR 808 nm laser at the power density of 2 W/cm2 for 10 min, the temperature of the solution containing Au/PPY@Fe3O4 (1.4 mg/mL) increased by about 35 °C. Cell viability assay showed that these nanocomposites had low cytotoxicity. Furthermore, an in vitro photothermal treatment test demonstrates that the cancer cells can be efficiently killed by the photothermal effects of the Au/PPY@Fe3O4 nanocomposites. In summary, this study demonstrates that the highly versatile multifunctional Au/PPY@Fe3O4 nanocomposites have great potential in simultaneous multimodal imaging-guided cancer theranostic applications.

Synthesis of Silver-Based Plasmas by Plant Extracts for In Vitro PTT/PDT/CT Synergistic Antitumor Therapy

Yiliang Xie,Baowen Dong,Xuan Zhang,Jimin Fan,Jianying Lin,Bing Zhang,Zhihuan Zhao

In this study, biosynthetic Ag@AgCl plasma is used as a photosensitizer for the first time to construct a photothermal/photodynamic/chemotherapy and pH-responsive synergistic antitumor nanomedicine delivery system CS-Ag@AgCl/C. The Ag@AgCl and AgNPs synthesized by the extracts of alpine banana, C itrus reticulata , and Citrus sinensis are characterized. The Ag@AgCl NPs synthesized with banana extract have a regular crystal shape and a granular size of 12 nm, and they are more stable than AgNPs synthesized with the other two extracts. In addition, Ag@AgCl plasma used as a photosensitizer can reduce the dependence of CS-Ag@AgCl/C on oxygen in the photodynamic process. After coating with chitosan, though the photothermal conversion efficiency of CS-Ag@AgCl/C decreases from 44.06% to 29.51%, the pH response character of it increases, and the pH response plays an important role in drug release experiments. Under an acidic environment of pH = 5.0 phosphate-buffered saline buffer, the drug release rate of the samples increases significantly. In the synergistic antitumor experiment, the cell survival rate increases from 36.83% to 78.69% after chitosan-coated Ag@AgCl/C NPs. When CS-Ag@AgCl/C-DOX+808 nm is co-cultured with HeLa cells, the cell survival rate is only 14.89%, which indicates that the constructed plasma drug delivery system has excellent photothermal/photodynamic/chemotherapy combined treatment capability.

AuNCs–LHRHa nano-system for FL/CT dual-mode imaging and photothermal therapy of targeted prostate cancer

Ze Wang,Liang He,Songtian Che,Huiyuan Xing,Lin Guan,Zhe Yang,Xingchen Li,Andrei V. Zvyagin,Quan Lin,Wenrui Qu

As the most common cancer in men worldwide, prostate cancer has a serious impact on people's health. Until now, the development of a platform for integrating tumor targeting, imaging and an effective treatment for prostate cancer has remained challenging. Herein, a nano-system is designed to improve both diagnosis and treatment for prostate cancer. We successfully synthesized an AuNCs–LHRHa nano-system by combining PEI-modified gold nanoclusters (AuNCs) with LHRH analogues (LHRHa). Due to the good tunable optical properties and photothermal properties of AuNCs, the nano-system can not only achieve efficient fluorescence/computed tomography dual-mode imaging, but can also be used for photothermal therapy (PTT). After modifying the LHRHa antibody of a prostate tumor, AuNCs–LHRHa can be more effectively recognized by the gonadotropin-releasing hormone receptors (GnRH-R) on the membrane of RM-1 cells, enhancing the tumor cell uptake of the nano-system, improving the targeting accuracy and PTT therapy efficacy for prostate cancer. It is hoped that the nano-system, which combines dual-mode imaging and targeted therapy, will provide a promising strategy for the integration of FL/CT diagnosis and PTT therapy for GnRH-R positive prostate cancer.

Automatic-degradable Mo-doped W18O49 based nanotheranostics for CT/FL imaging guided synergistic chemo/photothermal/chemodynamic therapy

Yilong Tian,Wenhui Yi,Qiuya Shao,Minghai Ma,Lei Bai,Rundong Song,Ping Zhang,Jinhai Si,Xun Hou,Jinhai Fan

The development of nanotherapeutics that can efficiently inhibit tumors and rapidly degrade in both cured and healthy tissues remains a research imperative. Herein, we developed tumor microenvironment (TME)-responsive Mo-doped W 18 O 49 nanobundles (MoWO NBs), which are co-assembled hyaluronic acid (HA) and doxorubicin (DOX) to achieve CT/FL imaging-guided multimodal cancer therapy. MoWO NBs exhibit multiple merits: (1) MoWO NBs degrade rapidly in the normal physiological environment but slowly in the weakly acidic TME, which leads to the selective accumulation of MoWO NBs at tumor sites and can prolong the therapeutic effect. (2) The existing Mo 4+ and W 5+ ions in MoWO NBs bestow the NBs with Fenton-like reaction activity to generate cytotoxic hydroxyl radicals (OH) from endogenous H 2 O 2 . Meanwhile, overexpressed glutathione (GSH) can reduce Mo 6+ and W 6+ ions in MoWO NBs to lower valence states, further enhancing the chemodynamic activity of MoWO NBs. (3) Under NIR-II laser irradiation, MoWO with a photothermal conversion efficiency of 45.5% could trigger powerful photothermal therapeutic effects in vitro and in vivo . (4) Owing to the fluorescence emission of DOX and the high Z-elements of molybdenum and tungsten, fluorescence (FL) imaging and computed tomography (CT) imaging have the potential to be used to monitor the biodistribution of Mo-W 18 O 49 @DOX@HA (MoWODH) in vivo . More importantly, the synergistic Chemo/PTT/CDT of MoWODH NCs greatly inhibited tumor growth. This work may broaden the path for smart nanomaterials and TME-responsive therapies.

Photothermal and Acid-Responsive Fucoidan-CuS Bubble Pump Microneedles for Combined CDT/PTT/CT Treatment of Melanoma

Jiaojiao Tao,Bingjie Wang,Yu Dong,XiangYan Chen,Shuang Li,Tianze Jiang,Xia Zhao

Transdermal cancer therapy faces great challenges in clinical practice due to the low drug transdermal efficiency and the unsatisfactory effect of monotherapy. Herein, we develop a novel bubble pump microneedle system (BPMN-CuS/DOX) by embedding sodium bicarbonate (NaHCO3) into hyaluronic acid microneedles (MNs) loaded with fucoidan-based copper sulfide nanoparticles (Fuc-CuS NPs) and doxorubicin (DOX). BPMN-CuS/DOX can generate CO2 bubbles triggered by an acidic tumor microenvironment for deep and rapid intradermal drug delivery. Fuc-CuS NPs exhibit excellent photothermal effect and Fenton-like catalytic activity, producing more reactive oxygen species (ROS) by photothermal therapy (PTT) and chemodynamic therapy (CDT), which enhances the antitumor efficacy of DOX and reduces the dosage of its chemotherapy (CT). Simultaneously, DOX increases intracellular hydrogen peroxide (H2O2) supplementation and promotes the sustained production of ROS. BPMN-CuS/DOX significantly inhibits melanoma both in vitro and in vivo by the combination of CDT, PTT, and CT. In short, our study significantly enhances the effectiveness of transdermal drug delivery by constructing BPMNs and provides a promising novel strategy for transdermal cancer treatment with multiple therapies.

Cell membrane-functionalized bismuth oxyiodide nanodots as potential contrast agent for gastrointestinal tract imaging

Meng Jin,Yunyan Ling,Limin Jin,Xuecen Wang,Xin Jiang,Quanxiao Li

Various noninvasive imaging modalities, including X-ray and computed tomography (CT) imaging, have provided significant advantages in accurately assessing the conditions of the gastrointestinal (GI) tract. The corresponding contrast agent with high resolution and biosafety is one of the most crucial focuses in the diagnosis of GI diseases. In this study, considering the requirements of good biocompatibility and biosafety, a novel imaging platform composed of the high-performance contrast agent based on macrophage membrane-coated BiOI nanodots (BiOI@M) was designed via a facile approach. The BiOI@M exhibited outstanding long-term stability and low toxicity. Significantly, BiOI@M demonstrated superior imaging ability compared to clinical barium sulfate. Higher CT values of BiOI@M were observed at equivalent concentrations to barium sulfate, thereby clearly outlining the stomach, small intestine, caecum, and colon after the oral administration of BiOI@M solutions in X-ray and CT imaging. Taken together, BiOI@M could serve as a novel efficient contrast agent in X-ray/CT imaging of the GI tract and diagnosis of GI diseases.

Highly efficient dual-modal phosphorescence/computed tomography bioprobes based on an iridium complex and AuNP polyiohexol composite nanoparticles

Yao Yu,Youshen Wu,Jiajun Liu,Yiming Liu,Daocheng Wu

On the basis of finite-difference time-domain simulations, a novel strategy is developed to prepare highly efficient bis(2-(2′-benzothienyl)pyridinato-N, C3′)iridium (BTP) and AuNP polyiohexol composite nanoparticles (BAPI NPs) as dual-modal phosphorescence/computed tomography (CT) bioprobes. In these bioprobes, BTP and AuNPs are both encapsulated with polyiohexol NPs. All bioprobe components perform two functions here: AuNPs could be used as both a CT contrast agent and a phosphorescence enhancement reagent of BTP with a metal-enhancement fluorescence effect. The results showed that BAPI NPs were almost spherical in shape, with an average size of 50.36 ± 3.8 nm and a higher total contrast agent loading ratio of 69.4%. Fourier transform infrared spectra confirmed that AuNPs and BTP are encapsulated in BAPI NPs. It is shown that they have lower toxicity for tissues and cells, their phosphorescence intensities are 8.27-fold that of BTP polyiohexol NPs (BPI NPs), the average phosphorescence intensity of the BAPI NPs was 1.46 times higher than that of BPI NPs and 5.85 times that of BTP alone in vivo. Improved CT imaging is obtained at a low dose of polyiohexol in vivo. These bioprobes not only have highly efficient and excellent dual-modal imaging, but they also save the use of various materials, indicating that these bioprobes are the potential dual-modal probes of the future.

Non-invasive fast assessment of hepatic injury through computed tomography imaging with renal-clearable Bi-DTPA dimeglumine

Ma Li,Pan Jinbin,Shu Gang,Pan Haiyan,Li Jingang,Li Dong,Sun Shaokai

Enhanced computed tomography (CT) imaging with iodinated imaging probes is widely utilized for the diagnosis and evaluation of various liver diseases. However, these iodine-based imaging probes face intractable limitations including allergic reactions and contraindications. Herein, we propose the utilization of renal-clearable iodine-free bismuth chelate (Bi-DTPA dimeglumine) for the non-invasive fast assessment of hepatic ischemia–reperfusion injury (HIRI) via CT imaging for the first time. Bi-DTPA dimeglumine offers several advantages such as simple synthesis, no purification requirement, a yield approaching 100%, large-scale production capability (laboratory synthesis > 100 g), excellent biocompatibility and superior CT imaging performance. In a normal rat model, the administration of Bi-DTPA dimeglumine resulted in a significant 63.79% increase in liver CT value within a very short time period (30 s). Furthermore, in a HIRI rat model, Bi-DTPA dimeglumine enabled the rapid differentiation between healthy and injured areas based on the notable disparity in liver CT values as early as 15 min post-reperfusion, which showed a strong correlation with the histopathological analysis results. Additionally, Bi-DTPA dimeglumine can be almost eliminated from the body via the kidneys within 24 h. As an inherently advantageous alternative to iodinated imaging probes, Bi-DTPA dimeglumine exhibits promising prospects for application in liver disease diagnosis.

Bismuth Chelate-Mediated Computed Tomography Perfusion Imaging of Acute Ischemic Stroke In Vivo

Lu Zhao,Li Ma,Xuetao Jia,Xiaoxue Wang,Wenjing Hou,Gang Shu,Bing Han,Dianxun Fu,Dong Li,Shao-Kai Sun,Jinbin Pan

Contrast-enhanced computed tomography perfusion (CTP) imaging is a critical clinical tool for evaluating cerebral blood flow after acute ischemic stroke (AIS), enabling accurate diagnosis and treatment planning. However, conventional iodine-based contrast agents may lead to allergic reactions and are contraindicated in patients with thyroid disorders. This study introduces Bi-DTPA dimeglumine, a renal-clearable and iodine-free bismuth chelate, for AIS perfusion assessment via CTP imaging. Bi-DTPA dimeglumine is synthesized through a simple and scalable process with nearly 100% yield (laboratory synthesis >100 g) and demonstrates excellent biocompatibility and efficient renal clearance. In a middle cerebral artery occlusion (MCAO) rat model, Bi-DTPA dimeglumine-enhanced CTP imaging accurately visualized vascular perfusion deficits, showing reduced cerebral blood volume (CBV) and cerebral blood flow (CBF), alongside prolonged time-to-maximum (Tmax) and mean transit time (MTT). Compared to iohexol, Bi-DTPA dimeglumine provided superior vascular contrast in computed tomography angiography (CTA), with contrast-to-noise ratio (CNR) improvements of 129.8 and 93.5% in normal and MCAO rats, respectively. Notably, CTP imaging revealed distinct reperfusion dynamics in ischemic regions postrecanalization. This iodine-free imaging agent marks a significant advancement in precise AIS diagnosis and post-treatment evaluation, addressing key limitations of existing contrast agents while enhancing diagnostic accuracy and safety.
