Curcumin-enhanced NIR-II-responsive gold nanobipyramids for targeted HSP 90 inhibition

Zhenying Diao,Youcheng Liang,Yong Liu,Dou Zhang,Long Qiu,Jianbo Sun,Qiaoyou Lu,Yanlei Liu,Daxiang Cui,Ting Yin

Blockade of heat shock protein 90 (HSP90) expression in multimodal synergistic therapy has a great prospect for cancer treatment. Nanomaterials combined with bioinformatic analysis provides accurate guidance for the design of anti-HSP90 nanomedicines. Herein, a NIR-II-responsive nanoplatform was developed under bioinformatics guided to effectly inhibit HSP90 for enhanced synergistic mild-photothermal chemotherapy without any notable tissue damage. The nanoplatforms were assembled from NIR-II-responsive gold nanobipyramids (GNBs) combined with curcumin (Cur) via hydrophobic-hydrophobic interactions and hydrogen bonds. On the basis of drug discovery and network pharmacology, we found that Cur has impressive anti-HSP90 capability and analyzed its therapeutic mechanism against NSCLC. Under the irradiation of NIR-II light, the obtained GNBs-Cur blocked the expression of HPS90 and inhibited related antiapoptotic pathways, thus enhancing the mild PTT of GNBs under 1064 nm laser irradiation. Meanwhile, Cur served as chemotherapeutic agents to induce apoptosis in tumor cells. In vivo photoacoustic imaging-guided, GNBs-Cur achieved effective tumor elimination through mild-photothermal chemotherapy without systemic toxicity. Overall, this work provides a new therapeutic modality paradigm for potential NSCLC treatment on the basis of synergistic therapies.

Curcumin encapsulated zeolitic imidazolate frameworks modified polyetheretherketone promotes osteogenic capacity through immunomodulation

Sihan Ma,Heyi Gong,Duo Sun,Xin Fang,Xiao Han,Yue Liu,Yadi Su,Junqi Wang,Yu Wang,Jinghui Zhao

Controlling the immune response and enhancing osteogenesis are essential for the long-term success of biomaterial implantation. Although polyetheretherketone (PEEK) is widely used in orthopedic implants, its inherent bioinertness and poor osteogenic capacity limit its use in clinical applications. In this study, curcumin (CCM) and zeolitic imidazolate frameworks-8 nanoparticles (ZIF-8 NPs) were introduced onto PEEK substrate to obtain the SPEEK-CCM@ZIF-8 (SPZC) system, which enhances osseointegration through early immunomodulation. SPEEK-CCM@ZIF-8 induces macrophage polarization towards M2 phenotype to reduce inflammation, promotes osteogenic differentiation capacity of bone marrow mesenchymal stem cells (BMSCs), and enhances the osseointegration and osteogenic capacity of PEEK implants. In vivo evaluation further confirmed the enhanced osteointegration effect of SPZC. Our results suggest that the CCM@ZIF-8 coating confers good osteogenic capacity to PEEK and that SPZC will have great potential as an implant.

Curcumin Alleviates Arecoline-induced Oral Submucous Fibrosis via the FOSL1/MAPK8 Axis

Yin Lifen,Wang Xiao

Oral submucous fibrosis (OSF) is a precancerous lesion of the oral cavity. Areca nut consumption can cause OSF through sustained activation of buccal mucosal fibroblasts (BMFs). This study explored the effect of curcumin on arecoline-induced BMF activation and its mechanism of action. BMFs were isolated and identified by immunofluorescence detection of fibroblast surface markers vimentin and S100A4. After transfection with FOSL1- or MAPK8-related vectors, BMFs were activated by arecoline and treated with curcumin. Scratch and transwell assays were performed to detect cell migration. ChIP and luciferase reporter assays were conducted to detect the binding of FOSL1 to the MAPK8 promoter. RT-qPCR was used to detect FOSL1 and MAPK8 mRNA expression. Western blotting was used to detect FOSL1, MAPK8, COL1A1, α-SMA, Smad2, and p-Smad2 proteins. Curcumin treatment inhibited arecoline-induced fibroblast migration, reduced the expression of myofibroblast markers COL1A1, α-SMA, and p-Smad2, and downregulated the expression of FOSL1 and MAPK8. FOSL1 or MAPK8 overexpression enhanced migration and increased COL1A1, α-SMA, and p-Smad2 expression in curcumin-treated cells. FOSL1 bound to the MAPK8 promoter and promoted MAPK8 expression. Simultaneous FOSL1 overexpression and MAPK8 knockdown, compared to FOSL1 overexpression, reduced cell migration and inhibited COL1A1, α-SMA, and p-Smad2 expression. In conclusion, curcumin targets FOSL1 to reduce MAPK8 expression, thereby suppressing arecoline-induced fibroblast activation.

Schiff Base Mediated Food-Derived Peptide Supramolecular Self-Assembly as Curcumin Carriers

Longjiang Tian,Xinran You,Jingbo Liu,Yajuan Li,Shanglin Li,Xuemin Jin,Shugang Li,Fei Pan,Zhipeng Yu,Ting Zhang,Zhiyang Du

The fusion assembly strategy of supramolecular chemistry combined with dynamic covalent chemistry has provided novel insights into the design of precision nutrition and intelligent drug delivery carriers. This work involved the development of a supramolecular self-assembly originating from entropy- and enthalpy-driven dynamic covalent bonding on Schiff bases between egg white-derived peptide Gln-Ile-Gly-Leu-Phe (QIGLF) and glutaraldehyde (GA), denoted QIGLF–GA. The assembly exhibited outstanding assembly characteristics and multiwavelength autofluorescence properties. Benefiting from the potent facilitation of the dynamic covalent interaction of Schiff base on the noncovalent assembly force network, QIGLF–GA was afforded an encapsulation capacity of curcumin (Cur) of more than 22% (≫ 10%) and rationally inhibited P-glycoprotein-mediated cellular efflux and markedly elevated the efficacy of Cur in overcoming the intestinal epithelial absorption barrier to the circulation with the help of endocytosis. Furthermore, QIGLF–GA-Cur features responsive release under weakly acidic conditions, which dramatically contributes to the intracellular bioavailability of Cur.

The alginate dialdehyde crosslinking on curcumin-loaded zein nanofibers for controllable release

Jiawen Li,Yuanhao Zheng,Peng Wang,Hui Zhang

In this study, electrospun zein/alginate dialdehyde (AD) nanofibers were prepared by green crosslinking. The degree of crosslinking could reach 50.72 %, and the diameter of electrospun fibers ranged from 446.2 to 541.8 nm. The generation of AD and the bonding of crosslinking were further confirmed by the changes on characteristic peaks and conformational ratios in the infrared spectroscopy and secondary structure analysis. High concentrations of AD led to improved thermal stabilities, mechanical properties, and hydrophobicity. And the highly crosslinked nanofibers (Z-8) owned the highest elastic modulus (24.92 MPa), tensile strength (0.28 MPa), and elongation at break (8.14 %) among five samples. Moreover, Z-8 possessed a high swelling ratio of 5.45 g/g, and a low weight loss of 6.09 %. The samples could encapsulate curcumin efficiently and show controllable release behaviors based on different AD addition. And the oxidation resistance of nanofibers gradually improved, consistent with the release performances. This study indicated AD crosslinking favored the preparation and application of zein nanofibers, and the oxidized polysaccharide acted as the green crosslinking agent, which provided reference value for the application of polysaccharides in food-related electrospun materials.

Synergistic effects of oleanolic acid and curcumin nanoparticles in gastric ulcer prevention

Dan Zhang,Wei Tian,Li-Hang Chen,Tao Chen,Di Wu,Yinan Du,Jiangning Hu

Gastric ulcers (GC) are prevalent gastrointestinal disorders with complex etiologies, including Helicobacter pylori infection and prolonged use of non-steroidal anti-inflammatory drugs (NSAIDs). Curcumin (CUR), with its established anti-inflammatory and antioxidant properties, is limited in clinical application due to poor bioavailability. This study developed oleanolic acid-coated CUR nanoparticles (OC NPs) to enhance the efficacy of CUR in GC prevention. OC NPs were characterized by a spherical shape, demonstrating remarkable improvements in solubility and stability, maintaining structural integrity within biological systems and exhibiting excellent biocompatibility. In vitro studies showed OC NPs reduced inflammation and oxidative stress in GES-1 cells. In vivo , OC NPs effectively prevented ethanol-induced GC in mice by lowering inflammatory cytokines, while increasing antioxidant levels. Histological analysis confirmed enhanced gastric mucosal protection and reduced fibrosis and apoptosis. OC NPs demonstrated prolonged retention in the stomach, offering a targeted drug delivery system. These findings suggested OC NPs as a promising low-toxicity alternative for preventing GC.

Photocrosslinked carboxymethylcellulose-based hydrogels: Synthesis, characterization for curcumin delivery and wound healing

Jingfei Wang,Ying Ma,Qingye Meng,Yongyan Yang,Ruiting Zhang,Shuangling Zhong,Yan Gao,Wenqi He,Xuejun Cui

Skin could protect our body and regenerate itself to against dysfunctional and disfiguring scars when faced with external injury. As wound dressings, hydrogels are biocompatible, hydrophilic and have a 3D structure similar to the extracellular matrix (ECM). In particular, hydrogels with drug-releasing capabilities are in acute wound healing. In this paper, photocrosslinked hydrogels served as wound dressing based on sodium carboxymethylcellulose (CMC) were prepared to promote wound healing. Photocrosslinked hydrogels were prepared by grafting lysine and allyl glycidyl ether (AGE) onto CMC and encapsulating curcumin (Cur). The synthesized hydrogels had the unique 3D porous structure with a swelling ratio up to 1300 % in aqueous solution. The drug release ratios of the hydrogels were 20.8 % in acid environment, and 14.4 % in alkaline environment. Notably, the hydrogels showed good biocompatibility and antibacterial properties and also exhibited the ability to accelerate the process of skin wound healing while prevent inflammation and scar formation when applied to a mouse skin wound model. As a result, the prepared hydrogels Gel-CLA@Cur showed great potential in wound healing.

Multifunctional Ionic Composite Hydrogel with Curcumin Capsule for Promoting Infected Wound Healing

Hao-Jie Tan,Ming Yan,Shi-Yu Hu,Yuan-Yuan Mou,Xu Peng,Zhi-Guo Wang,Lingli Li,Maja Kuzmanović,Jia-Zhuang Xu,Zhong-Ming Li

Wound infection complicated by excessive inflammation remains a persistent challenge in clinical healthcare. Curcumin (Cur), a natural diketone compound, holds therapeutic potential in treating infected wounds. However, its application is limited by poor aqueous dispersibility and low stability. To overcome this dilemma, a versatile capsule as an effective Cur carrier is developed by encapsulating Cur in succinic acid-modified cyclodextrin via hydrophobic interactions, and then loading it into a Zn 2+ -mediated crosslinked injectable hydrogel for infected wound healing. The composite hydrogel exhibits adjustable rheological properties, modulus, swelling ratios, and degradation rate by control over Zn 2+ concentration. This unique tactic enables the sustained Cur release, ensuring long-term antioxidant activity and reactive oxygen species (ROS) scavenging. An in vivo experiment in a rat S. aureus -infected wound model demonstrates that the composite hydrogel significantly accelerates wound healing by massacring bacteria, inhibiting inflammation, as well as promoting collagen deposition and angiogenesis. Overall, this effort paves the way for arming Cur, offering a potent antibiotic-free candidate for bacteria-invaded wound management.

Synthesis, characterization and anti-cancer activity of Pluronic F68–curcumin conjugate micelles

Yuee Cai,Zhongqing Sun,Xiaobin Fang,Xiefan Fang,Fei Xiao,Yitao Wang,Meiwan Chen

Curcumin (CUR), a nontoxic polyphenol derived from the rhizome of turmeric (Curcuma longa), has been recognized as an anti-cancer and chemo-preventative agent. However, its clinical application for cancer treatment has been greatly limited due to its poor water-solubility and low bioavailability. To tackle this problem, Pluronic F68–CUR (F68–CUR) conjugate micelles, which are amphiphilic copolymers, were designed and synthesized in this study. These highly stable micelles with CUR concentrated in the core were formulated using the solvent evaporation method and were confirmed by Fourier transform infrared spectroscopy and nuclear magnetic resonance. Physicochemical characterization of F68–CUR conjugate micelles revealed that high drug loading content (DL%; 0.248 mg CUR/1 mg F68) was achieved, and the average particle size of micelles was 115.2 ± 3.0 nm. Compared with free CUR, a significantly higher cytotoxicity against human breast cancer cell line MDA-MB-231 was observed in F68–CUR conjugate micelles. The IC50 value of F68–CUR conjugate micelles was 1.95-fold lower than that of free CUR, indicating that the anti-cancer activity of CUR was significantly improved in the micelles. Furthermore, apoptotic studies demonstrated that F68–CUR conjugate micelles induced more cell apoptosis than that of free CUR. Taken together, these results demonstrate that F68–CUR conjugate micelles are promising to improve the clinical effectiveness of CUR in cancer treatment.

Enhanced wound healing properties of guar gum/curcumin-stabilized silver nanoparticle hydrogels

Bhubhanil Sakkarin,Talodthaisong Chanon,Khongkow Mattaka,Namdee Katawut,Wongchitrat Prapimpun,Yingmema Werayut,Hutchison James A.,Lapmanee Sarawut,Kulchat Sirinan

Biocompatible materials that act as scaffolds for regenerative medicine are of enormous interest. Hydrogel-nanoparticle composites have great potential in this regard, however evaluations of their wound healing and safety in vivo in animal studies are scarce. Here we demonstrate that a guar gum/curcumin-stabilized silver nanoparticle hydrogel composite is an injectable material with exceptional wound healing and antibacterial properties. We show that the curcumin-bound silver nanoparticles themselves exhibit low cytotoxicity and enhance proliferation, migration, and collagen production in in vitro studies of human dermal fibroblasts. We then show that the hydrogel-nanoparticle composite promotes wound healing in in vivo studies on rats, accelerating wound closure by > 40% and reducing bacterial counts by 60% compared to commercial antibacterial gels. Histopathology indicates that the hydrogel composite enhances transition from the inflammation to proliferation stage of healing, promoting the formation of fibroblasts and new blood vessels, while target gene expression studies confirm that the accelerated tissue remodeling occurs along the normal pathways. As such these hydrogel composites show great promise as wound dressing materials with high antibacterial capacity.

Folic Acid and PEI Modified Mesoporous Silica for Targeted Delivery of Curcumin

Xiaoxiao Sun,Nan Wang,Li-Ye Yang,Xiao-Kun Ouyang,Fangfang Huang

Nano anti-cancer drug carriers loaded with antineoplastic drugs can achieve targeted drug delivery, which enriches drugs at tumor sites and reduces the toxic side effects in normal tissues. Mesoporous silica nanoparticles (MSN) are good nano drug carriers, as they have large specific surface areas, adjustable pore sizes, easily modifiable surfaces, and good biocompatibility. In this work, polyethyleneimine (PEI) grafted MSN were modified with folic acid (FA) as an active target molecule using chemical methods. The product was characterized by SEM, TEM, Zetasizer nano, FTIR, and an N2adsorption and desorption test. MSN-PEI-FA are porous nano particles with an average particle size of approximately 100 nm. In addition, the loading rate and release behavior of MSN-PEI-FA were studied with curcumin as a model drug. The results show that when loading curcumin to MSN-PEI-FA at 7 mg and 0.1 g, respectively, the encapsulation efficiency was 90% and the cumulative release rate reached more than 50% within 120 h at pH = 5. This drug delivery system is suitable for loading fat-soluble antineoplastic drugs for sustained release and pH sensitive delivery.

Effect of curcumin on p38MAPK expression in DSS-induced murine ulcerative colitis

C-P Li,J-H Li,S-Y He,O Chen,L Shi

The aim of this study was to determine the therapeutic effect of curcumin on dextran sulfate sodium-induced ulcerative colitis (UC) and to explore the related mechanism. Sixty mice were randomly divided into 6 groups. A group was the normal control group; B group was the model group; C group was the 1.5 mg/kg dexamethasone group based on the B group; and D, E and F groups were 15, 30, and 60 mg/kg curcumin groups, respectively, based on the B group. The mice were killed 7 days after treatment; the expression of TNF-α and MPO in colon tissue was determined with ELISA, and colon p-p38MAPK and p38MAPK mRNA expression was evaluated by immunohistochemistry and RT-PCR, respectively. In the C, D, E, and F groups, TNF-α and MPO levels significantly decreased (P < 0.05), and the expression of p-p38MAPK also significantly decreased (P < 0.01). The expression of p38MAPK mRNA in the C, D, E, and F groups decreased (P < 0.01), and there was a statistically significant difference between the E and F groups (P < 0.01). Curcumin had a therapeutic effect, which probably played a role in UC treatment by inhibiting the p38MAPK signaling pathway, thereby reducing the release of TNF-α.
