Synthesis of water dispersible and biocompatible nanodiamond composite via photocatalytic surface grafting of zwitterionic polymers for intracellular delivery of DOX

Lijing Dai,Meiying Liu,Wei Long,Xin Hu,Hui Ouyang,Yulin Feng,Fengjie Deng,Yuanqing Wen,Xiaoyong Zhang,Yen Wei

As a member of nano-carbon compound, nanodiamonds (ND) with diverse structure, small size and relative low toxicity has been extensively explored for different applications especially as delivery vehicles for drugs. However, surface functionalization of ND with polymers is still required to overcome their poor water dispersibility and improve drug delivery performance. In this work, we reported a simple photocatalytic atom transfer radical polymerization (ATRP) method for surface functionalization of ND by using FeBr 3 as the photoredox catalyst. Owing to the introduction of water dispersible 2-methacryloyloxyethyl phosphorylcholine (MPC), resultant NDs-MPC displayed improved water dispersibility and low toxicity. The anticancer agent DOX could be loaded on NDs-MPC efficiently and controlled release from ND-P-DOX complexes. The cell viability and cell imaging results demonstrated that DOX can be transported into cells and maintained its corresponding therapeutic effect. More importantly, as compared with traditional ATRP, photocatalytic ATRP could not only operate under rather mild conditions but also could avoid utilization of expensive and toxic agents. Considered the advantages of photocatalytic ATRP and ND, the method developed in this work could be a promising strategy for fabrication of multifunctional ND-based composites for different applications.

Functionalized N-hydroxypropyl trimethylammonium chloride chitosan as an ICG/DOX co-delivery nanocarriers for targeted and chemo-photothermal combination therapy

Chen Zhang,Zheng Jin,Yuhong Lin,Yiyu Wang,Kai Zhao

Doxorubicin (DOX) is considered one of the most effective chemotherapy drugs and is used as a first-line drug for many cancers. However, it has shown serious side effects such as cardiotoxicity and colonic ulcers , which limit its clinical use. Targeted delivery to tumor cells by nanocarriers or decreasing DOX dosage is considered to be an effective way to reduce the toxic and side effects of DOX on normal human tissues. In this paper, the mannose-functionalized N-2-HACC nanoparticles (MN NPs) loaded with DOX and Indocyanine Green (ICG) (DOX/ICG@MN NPs) were synthesized by the ion-crosslinking method, which were used for the in vitro targeted photothermal and chemical therapy. The results of FT-IR, particle size analysis, and SEM showed that ICG and DOX were successfully loaded, and DOX/ICG@MN NPs had great dispersibility and size. The results of the cell safety test showed that MN NPs had good safety. The photothermal stability tests revealed that the ICG in DOX/ICG@MN NPs had excellent thermal stability and photothermal conversion efficiency than the free ICG. The DOX release of DOX/ICG@MN NPs is pH-responsive, which promoted the effective release in slightly acidic environments at the tumor site. Owing to the specific targeting ability of mannose , DOX/ICG@MN NPs could be effectively uptake into HCT116 cells. In the presence of Near-Infrared irradiation (NIR), DOX/ICG@MN NPs exhibited synergistic treatment effects against tumor cells, which could significantly reduce the dosage of DOX and reduce the toxic side-effects on normal tissues. Therefore, this co-delivery system is considered to have significant potential for targeting tumors and chemo-photothermal therapy.

Beibei Zhu,Jun Shi,Chongchong Liu,Jingguo Li,Shaokui Cao

As a novel two-dimensional material, Ti 3 C 2 MXenes has attracted lots of attention in biomedical filed for its large surface area and excellent near-infrared (NIR) responsiveness. In this paper, an in-situ growth and self-assembly approach was employed to combine gold nanorods (GNRs) with Ti 3 C 2 nanosheets to prepare intelligent sandwich-like Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids . Compared with Ti 3 C 2 nanosheets, in-situ growth Ti 3 C 2 @GNRs possessed excellent photothermal conversion efficiency (45.89%), caused by the distinguished photothermal synergy between GNRs and Ti 3 C 2 nanosheets. Moreover, the high specific surface area of Ti 3 C 2 MXene and outstanding adhesion performance of PDA endowed Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids with superior drug loading ability for doxorubicin hydrochloride (DOX) (95.88%). Besides, Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids displayed distinct pH/NIR responsive drug release properties upon NIR irradiation owing to the strong π-π stacking interaction between Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 and DOX, along with the excellent NIR-responsiveness of Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 . This paper offers a practicable method to prepare Ti 3 C 2 MXene-based nanoplatform with synergistically enhanced NIR drug release behavior, brilliant biocompatibility and high drug loading efficiency, which is expected to be applied in remote cancer therapy.

Ruiyi Dai,Xiangrong Peng,Bi Lin,Danyang Xu,Ruichan Lv

In this research, a NIR II luminescence imaging and enhanced chemo-/photothermal therapy system of CuS–DOX–Nd/FA NPs for breast cancer and lymph node tracing under single 808 nm irradiation is proposed. Nd–DTPA molecular cluster with the NIR II imaging effect as the carrier was designed to load the ultrasmall CuS nanoparticles and chemotherapeutic drug doxorubicin hydrochloride (DOX). The composite probe is used for tumor lesion imaging and tracking the breast cancer sentinel lymph nodes with simultaneous chemo-/photothermal therapy (PTT) for breast cancer under the single 808 nm laser. This designed probe not only has high permeability and retention (EPR) targeting effect but also can respond to the tumor microenvironment (TME), realizing more precise and efficient release of DOX at the cancer focus. At the same time, CuS as a drug carrier has a good photothermal therapy effect (photothermal conversion efficiency: 27.9%). The serialized released chemotherapy DOX and synergistic PTT effect can be used to the treat the in situ breast cancer land and simultaneously kill the metastasis cancer. The system made the combined molecular clusters Nd–DTPA achieve NIR II imaging of tumor lesions of breast cancer and lymph node to obtain the integration of diagnosis of the transferred disease for better prognosis. The feasibility of the system had obvious tumor growth inhibition effect with NIR II imaging guided is verified by a series of in vitro and in vivo experiments.

Glucose-modified PEG-PLA self-assembled nano-micelles optimize DOX enrichment, tissue penetration, and tumor suppression effects in pancreatic ductal adenocarcinoma

Zhijie Liang,Jinzhuai Li,Hai Huang,Hongmian Jiang,Lifeng Luo,Shaorong Li,Huali Huang,Fangdi Hu

Pancreatic ductal adenocarcinoma (PDAC) poses significant therapeutic challenges due to its unique tumor microenvironment, which is characterized by collapsed blood vessels and elevated intratumoral pressure, severely hindering drug penetration and distribution. Consequently, we synthesized glucose-modified PEG-PLA (polyethylene-polylactice acid) nanoparticles as a drug delivery system for doxorubicin (DOX). We successfully enhanced the permeability and targeting capability of the nanoparticles in pancreatic cancer cells using glycosylation modification, significantly increasing drug accumulation within the tumor microenvironment. Our study demonstrated excellent cellular uptake of the nanoparticles in vitro and their improved penetration in 3D tumor spheroid models. Additionally, in vivo experiments revealed a notable increase in nanoparticle accumulation at the tumor site, along with favorable drug release characteristics. Hence, glucose-modified PEG-PLA nanoparticles hold substantial potential for clinical application in PDAC treatment, effectively enhancing the therapeutic efficacy of chemotherapeutic agents.

A CuS/Ag/Pt@ICG/DOX nanoplatform with cascaded nanozyme catalysis for tumor microenvironment remodeling and synergistic photodynamic/photothermal/chemodynamic therapy

Wen-Qi Zhu,Dong Cheng,Lin-Song Li,Peng-Wei Chen,Bang-Bang Liu,Mei-Xia Zhao

This work introduces the rational design of a hierarchically structured, multi-enzyme integrated therapeutic platform, referred to as HCuS/Ag/Pt/ICG/DOX (CAPID). The platform is constructed by depositing dual-enzyme-active Ag/Pt on copper sulfide nanoparticles (CuS NPs) to remodel the tumor microenvironment (TME), followed by loading indocyanine green (ICG) as a near-infrared (NIR) fluorophore and doxorubicin (DOX) as a chemotherapeutic payload. The CAPID platform utilizes catalase to decompose H 2 O 2 into oxygen (O 2 ), which synergizes with endogenous H 2 O 2 to initiate a Fenton-like reaction, generating cytotoxic hydroxyl radicals (·OH). The redox cycling between Cu 2 + ions and intracellular thiols depletes GSH, amplifying oxidative stress cascades. NIR irradiation achieved 88.9 % tumor inhibition via spatiotemporal tri-therapy (chemo/PDT/PTT) synergy, thereby addressing the limitations of monotherapy through nanoplatform engineering.

pH-sensitive magnetic drug delivery system via layer-by-layer self-assembly of CS/PEG and its controlled release of DOX

Juan Wu,Xi Wang,Binglong Zhu,Qinting He,Fang Ren,Fei Tong,Wei Jiang,Xianghong He

Cancer is one of the biggest killers threat to human life and health and is still difficult to treat mainly due to the lack of targeted drug delivery stages and limitations associated with delivering drugs to targeted cancer tissues. The goal of this work was to develop a magnetic, pH-sensitive formulation for targeted delivery of chemotherapeutic agents to cancer cells. Then the functional drug delivery system (Fe3O4@CS/PEG-DOX) was synthesized by the layer-by-layer (LbL) self-assembly technique. And the drug loading content is calculated to be 19.1%. In addition, the Fe3O4@CS/PEG-DOX exhibited excellent pH-sensitivity, 73.1% DOX was released within 48 h at pH 4.0. Furthermore, all the release behaviors fit the Higuchi model very well and the dissolution of CS/PEG layers played a key role on DOX release from Fe3O4@CS/PEG-DOX. The results of toxicity analysis in human liver hepatocellular carcinoma cells (HepG2) revealed that Fe3O4@CS/PEG-DOX exhibited high anti-tumor activity, while the Fe3O4@CS/PEG nanocomposites were practically non-toxic. Therefore, all the results demonstrated that the Fe3O4@CS/PEG-DOX could have an important impact on the development of targeted intracellular delivery nanodevices for cancer therapy.

Tumor Microenvironment-Responsive Nanoplatform of Cu-Doped ZIF-8 Dual-Loaded with ICG and DOX for Photothermal-Enhanced Chemodynamic Therapy/Chemotherapy

Tao Yang,Tao Wang,Tao Shen,Mingrong Dong,Jinkun Liu,Ming Ni,Yan Zhu

Targeting tumor-specific characteristics of acidic microenvironment, elevated H2O2 levels, and thermosensitivity, this study proposed and developed a copper-doped ZIF-8 nanoplatform coloaded with indocyanine green (ICG) and doxorubicin hydrochloride (DOX), designated as Cu-ZIF-8@ICG&DOX (CZID), to establish a multimodal therapeutic system integrating chemodynamic therapy, photothermal therapy, and chemotherapy. Results showed that the 25% copper doping level optimized the structure, achieving 7.46% actual doping content while limiting the average nanoparticle size to 99.6 nm. Dual loading of ICG and DOX induced morphological transition to spherical core–shell architectures, as confirmed by zeta potential reversal, with maximum drug loading capacities of 23.01 μg/mg for ICG and 122.43 μg/mg for DOX. The ZIF-8 framework exhibited pH-responsive degradation under acidic conditions. Released Cu2+ ions mediated continuous hydroxyl radical generation through glutathione-depletion redox cycling, confirming chemodynamic efficacy. Under 808 nm laser irradiation, ICG enabled enhanced photothermal conversion, showing concentration- and power-dependent temperature elevation. Cellular assays revealed efficient CZID internalization by MCF-7 cells, generating significantly higher intracellular reactive oxygen species levels under irradiation and more intense dead cell staining compared to CZI and CZD groups. Such an enhancement was attributed to photothermally accelerated ZIF-8 degradation, promoting Cu2+ ions and DOX release, thereby strengthening chemodynamic–chemotherapy synergy. At the working concentration of 50 μg/mL, CZID induced 95.36% apoptosis in MCF-7 cells while maintaining over 70% viability in MCF-10A normal cells, validating its precise antitumor potential.

A novel pH-responsive hollow mesoporous silica nanoparticle (HMSN) system encapsulating doxorubicin (DOX) and glucose oxidase (GOX) for potential cancer treatment

Kaiwu Cheng,Yu Zhang,Yaojia Li,Zhiguo Gao,Fanghui Chen,Kai Sun,Peijing An,Chen Sun,Yong Jiang,Baiwang Sun

The multi-therapy modality is based on the combination and synergy of multiple single treatment modalities and materials chemistry. Herein, a novel pH-responsive doxorubicin (DOX) delivery system based on hollow mesoporous silica nanoparticles (HMSNs) was constructed, which was designed to treat cancer via synergistic chemotherapy and starvation therapy. Logically, starvation therapy could improve the therapeutic efficiency by sensitizing the drug molecules and activating the chemotherapy; glucose oxidase (GOX) coupled to the surface of HMAN deprived the cancer cells of glucose for starvation therapy and increased the abnormality of the tumor microenvironment by locally reducing the pH, aggravating hypoxia and promoting the concentration of poisonous H2O2. Increased acidity could accelerate the depolymerization of pH-sensitive polyelectrolyte multilayers (PEM) wrapped on the most external surface, causing the spatial control of DOX release within tumor cells. Detailed physical characterization certified the successful synthesis of DOX/GOX@HMSN-PEM, which was approximately 180 nm in diameter. In vitro experiments demonstrated that GOX could effectively promote drug release, and DOX/GOX@HMSN-PEM possessed high drug loading rate and stability. Excellent cellular uptake performances were revealed by confocal laser scanning microscopy (CLSM). The positively charged nanoparticles could specifically target the cancer cells overexpressing negative charges. In addition, they were found to have an excellent effect on the cytotoxicity and apoptosis. The promoted drug release via combined starvation therapy and drug therapy suggests that this novel type of pH-responsive hollow mesoporous silica nanoparticles may serve as a new drug delivery system for cancer treatment.

Construction of a Drug Delivery System via pH-Responsive Polymeric Nanomicelles Containing Ferrocene for DOX Release and Enhancement of Therapeutic Effects

Chenggang Song,Fan Yang,Ruixuan Ji,Yin Lv,Zhong Wei

We report an amphiphilic block copolymer via poly(ethylene glycol) methyl ether-Dlabile-poly(caprolactone)-ferrocene (mPEG-Dlabile-PCL-Fc) to deliver anticancer drug doxorubicin (DOX). Lipase Novozyme-435 was used as a catalyst for ring-opening polymerization with ε-caprolactone, and an acid-sensitive Schiff base was used to connect the hydrophilic and hydrophobic parts; the ferrocene provided ferrous ions and was introduced at the end of the amphiphilic copolymer. The resulting copolymers were characterized by 1H NMR/13C NMR and could be self-assembled in an aqueous solution to form nanomicelles with PCL-Fc as a hydrophobic core and mPEG as a hydrophilic shell. Transmission electron microscopy showed that the micelles were spherical and nanosized before and after DOX loading. The blank micelles also showed good biocompatibility. The drug-loaded polymeric nanomicelles exhibited a positive anticancer effect relative to the copolymers without ferrocene; the therapeutic effect of drug-loaded micelles containing ferrocene was more obvious. In vitro drug release results also showed that the polymer had a good pH response. Confocal microscopy also showed that polymeric micelles can effectively deliver and release the drug; the polymer containing ferrocene also leads to significantly improved ROS levels in tumor cells. Ferrocene can effectively and synergistically inhibit tumor cells with DOX.

Cell Penetrating Peptide-Based Self-Assembly for PD-L1 Targeted Tumor Regression

Feng Guo,Junfeng Ke,Zhengdong Fu,Wenzhao Han,Liping Wang

Cell penetrating peptides (CPPs) are peptides that can directly adapt to cell membranes and then permeate into cells. CPPs are usually covalently linked to the surface of nanocarriers to endow their permeability to the whole system. However, hybrids with lipids or polymers make the metabolism much more sophisticated and even more difficult to determine. In this study, we present a continuous sequence of 18 amino acids (FFAARTMIWY(d-P)GAWYKRI). It forms nanospheres around 170 nm, which increase slightly after loading with siRNA and DOX. Notably, it can be internalized by cancer cells mainly through electronic interactions and PD-L1-mediated endocytosis. Compared with poly-l-lysine and polyethyleneimine, it has a much higher efficiency (about four times) of gene transduction while lowering toxicity. In the treatment of cancer, it causes apoptosis (21%) and inhibits the expression of SURVIVIN protein in vitro. In vivo, it shows good biocompatibility as there are no changes in mice’s body weight. When administering peptide-siRNA-DOX, tumor growth is inhibited the most (about three times). These results above prove the sequence to be a good candidate for gene therapy and drug delivery.Keywords:peptide;self-assembly;gene therapy;PD-L1
