Au/NaYF4 : Yb,Er Binary Superparticles: Synthesis and Optical Properties

Junwei Zhao,Jian Wu,Junfei Xue,Qiannan Zhu,Weihai Ni

Graphical Colloidal superparticles (SPs) are nanoparticle (NP) assemblies in the form of colloidal particles. Assembling nanoscopic objects into mesoscopic or macroscopic composite architectures allows for the bottom-up fabrication of functional nanomaterials. In this study, a method for single-step self-assembly synthesis of Au/NaYF 4 : Yb,Er SPs was developed using oil-in-water (O/W) microemulsions to simultaneously encapsulate gold nanoparticles (AuNPs) and NaYF 4 : Yb,Er upconversion nanoparticles (UCNPs) via evaporation at room temperature. The synthesized Au/NaYF 4 : Yb,Er SPs possess good dispersibility and stability. When the number of AuNPs added is increased, the SPs exhibit decreased upconversion luminescence, which can be ascribed to the Förster resonance energy transfer (FRET) from the NaYF 4 : Yb,Er UCNPs to the AuNPs. Time-resolved measurements of the green emission further confirm the existence of a new decay route corresponding to the FRET process. Our research provides a facile and versatile strategy for the synthesis of novel multifunctional nanocomposites with tunable upconversion luminescence properties, which can be of great significance in biological applications.

An electronic tongue based on conjugated polymers for the discrimination and quantitative detection of tetracyclines

Shiyong Chen,Jie Wang,Zhikun Shang,Yun Ding,Aiguo Hu

A fluorescent sensor array has been developed based on conjugated polymers (CPs) having six different skeletons for the detection of tetracyclines (TCs), which are known as environmental pollutants. CPs were synthesized from confined nanoreactors in a controlled manner. The fluorescent response occurs through the fluorescence resonance energy transfer (FRET) effect. By utilizing linear discriminant analysis (LDA), effective differentiation of TCs was accomplished with a very low detection concentration (66 nM). Moreover, the sensor array exhibited a highly sensitive ability to quantitatively distinguish different concentrations of TCs. Finally, the sensor array's potential for detecting TCs in aqueous solutions has been successfully demonstrated, widening its applications in practical environments. With simple preparation process, a low cost of detection and high sensitivity, the experimental results indicate that the CP-based sensor array is a promising platform for the sensitive and quantitative detection of TCs, and provides a good reference for future scientific research.

Aptamer based ratiometric determination of DON by exploiting the FRET between carbon dots and graphene oxide

Meijun Lu,Tianlin Wang,Tiange Li,Yan Ma,Xianqing Huang,Libin Wan,Fayun Wang,Lianjun Song

A ratiometric aptasensor for deoxynivalenol (DON) detection was developed based on FRET between graphene oxide (GO) and carbon dots. Blue carbon dots, functionalized with aptamers, act as energy donors, while GO functions as the energy acceptor. In the absence of DON, the addition of GO quenches the fluorescence of the blue carbon dots, while the fluorescence of red carbon dots, used as internal reference signals, remains unchanged. When DON is introduced, the aptamer selectively binds to the target, increasing the distance between the donor and acceptor, disrupting FRET, and gradually restoring the blue fluorescence. The sensor demonstrated a strong linear relationship within the concentration range of 0.05–200 ng/mL of DON, with a detection limit of 14.7 pg/mL. The superior analytical performance and high sensitivity indicate the potential of the sensor for DON detection in real sample matrices.

An autocatalytic hybridization circuit-based FRET aptasensor for detection of low-abundant sulfameter in human serum

Yanping He,Ruifen Tian,Weili Shen,Jingrui Zhang,Chen Tao

Exposure to antibiotics is considered a potential risk factor for human health. Yet, the extensive and cost-effective detection of low-abundant antibiotics in complex matrices remains a significant challenge. Herein, an aptamer and an autocatalytic hybridization circuit (AHC) were used to fabricate a fluorescence resonance energy transfer (FRET) platform to detect sulfameter (SME) in human serum. The AHC system comprised two mutually motivated hybridization chain reactions (HCR) modules, ultimately producing long-branched DNA copolymeric nanowires. This mutually reciprocal activation of two HCR modules enables continuous signal amplification, providing the AHC system with wide linear range and high sensitivity for the SME detection. Compared to the HCR-based aptasensor, the AHC-based aptasensor exhibited a wider linear range and improved sensitivity (3.3 times greater). Under optimal conditions, the fluorescent AHC-based aptasensor demonstrated a linear range (R 2 was 0.996) from 0.5 to 2000 nM, with a low detection limit of 0.301 nM (S/N = 3). The fluorescent aptasensor was also validated by SME-spiked human serum samples, showing average recoveries ranging from 96.40 % to 109.30 %, with a relative standard deviation below 10.45 %. Furthermore, when tested on six human serum samples, the aptasensor results were consistent with those obtained from the commercial ELISA method. These findings demonstrate that the proposed aptasensor provides a promising approach for the practical monitoring of low-abundant SME in human serum.

Investigation of the in vivo integrity of polymeric micelles via large Stokes shift fluorophore-based FRET

Houbing Zhang,Hongjun Li,Zhiting Cao,Jinzhi Du,Lifeng Yan,Jun Wang

Polymeric micelles hold great potential for anticancer drug delivery. Sufficient integrity of polymeric micelles after intravenous injection is critical for successful drug delivery to solid tumors , but accurate measurement of the in vivo micellar integrity remains challenging. Methods based on Förster resonance energy transfer (FRET) to monitor the in vivo micellar integrity are frequently used. However, the self-quenching effect of these FRET fluorophores used has been improperly ignored and has caused inaccurate measurements. Herein, we report a FRET-based approach using the large Stokes shift (LSS) fluorophores NBD-X and MS735 as the donor and acceptor, respectively, to investigate the integrity of polyethylene glycol- block -poly(ε-caprolactone) (PEG-PCL) micelles. We established a mathematical formula for the integrity calculation, and an in vitro verification experiment showed that the formula results exactly matched the simulated results. Our results demonstrated that PEG-PCL micelles gradually dissociated in blood circulation, but approximately 60% of the micelles in plasma remained intact 72 h after intravenous ( i.v. ) injection. This LSS fluorophore-based FRET approach can be used to accurately monitor the integrity of nanoparticles , and this study demonstrates that most of PEG-PCL micelles maintain their aggregation state during blood circulation for anticancer drug delivery.

On-Site Detection of Thiamethoxam Using a Portable FRET Aptasensor with Upconversion Nanoparticles and Graphene Oxide

Enyu Zhou,Dou Che,Yeshun Zhang,Hui Yan

Thiamethoxam (THI), a widely used neonicotinoid pesticide, is of crucial significance for the accurate detection of THI in terms of environmental and food safety. In this study, a portable fluorescent aptasensor was developed for the rapid and sensitive detection of thiamethoxam (THI). Based on the principle of fluorescence resonance energy transfer (FRET), the sensor system consists of THI-specific aptamers (UCNPs-apt) and upconversion nanoparticles (UCNPs) functionalized with graphene oxide (GO). Compared to conventional fluorophores, UCNPs exhibit minimal background fluorescence, thereby minimizing interference in complex matrices such as food samples. Key parameters, including GO concentration (0.15 mg/mL), UCNPs/GO incubation time (40 min), pH (7.4), and THI reaction time (30 min), were optimized to maximize fluorescence quenching and target-induced recovery. The sensor showed a linear detection range of 0.1–100 ng/mL and a limit of detection (LOD) of 0.088 ng/mL, with excellent selectivity and reproducibility. The method was validated using peach and mulberry leaf samples, yielding results consistent with ELISA (recoveries: 98.79–113.04%; relative standard deviations: 0.46–1.92%). The integrated portable system, leveraging aptamer specificity and compact instrumentation, enables on-site monitoring and addresses the urgent need for rapid field-deployable detection of THI residues.

Integrity of polymeric micelles regulates bioavailability of cyclosporine A: A FRET-based analysis during oral delivery

Haihan Sun,Jiansong Zhao,Boyuan Liu,Jingxin Gou,Haibing He,Yu Zhang,Tian Yin,Xiangqun Jin,Xing Tang

This study comprehensively investigated the effect of structural integrity on the oral delivery efficacy of mPEG- b -PCL polymeric micelles (PMs) using fluorescence resonance energy transfer (FRET) technology. During mucus penetration, the integrity loss of PMs resulted from the “hydration effect” of mucins and the “solubilization effect” of bile salts. Increasing surface PEG density or decreasing particle size <50 nm resisted the “hydration effect”, while shortening PCL chain or increasing aggregation number alleviated the “solubilization effect”. Fluorescence colocalization studies demonstrated that low surface PEG density and small particle size enhanced the cellular uptake of PMs, but high surface PEG density preserved the intracellular integrity of PMs. Approximately 30 %–60 % of endocytosed PMs were expelled within 4 h, correlating with their cellular uptake efficiency. Transcellular transport results indicated that 5 %–25 % of PMs underwent transcytosis within 4 h, 0.5 %–10 % of initial PMs remained intact. In situ intestine perfusion data confirmed the consistency between the in vitro findings and the actual performance of PMs in vivo . Two types of PMs were selected to encapsulate cyclosporine A (CyA) based on their enhanced integrity and transcytosis efficiency. The PM with a short amphiphilic block and loose structure exhibited comparable in vitro release and in vivo performance to Neoral®, with a C max of 1356.28 ± 170.01 ng/mL, a T max of 2 h, and an AUC 0-t of 16,408.92 ± 1166.78 ng/mL*h. However, the PM with a long amphiphilic block and ordered core PCL arrangement exhibited sustained release of CyA both in vitro and in vivo , decreasing CyA accumulation in major organs and improving the oral bioavailability of CyA, with a C max of 757.07 ± 66.19 ng/mL, a T max of 9.33 ± 2.31 h, and an AUC 0-t of 21,938.44 ± 2183.59 ng/mL*h.

Rapid detection of procalcitonin using dual-color carbon quantum dots as fluorescent probes in FRET assay

Yuxin Tian,Yuwei Du,Zhang Chen,Li Li,Ruhong Yan,Xiaoqiang Li,Juan Yue

FRET, known as Förster Resonance Energy Transfer, serves as a versatile technique enabling precise quantitative assessment of protein concentrations with minimal disruption to biological processes. In this study, we have developed two types of fluorescent carbon quantum dots, B-CDs and G-CDs, serving as donor and acceptor probes, respectively, for FRET-based procalcitonin (PCT) detection. The B-CDs exhibit strong blue fluorescence (excitation/emission peaks at 360/442 nm) as donors, while the G-CDs display faint green fluorescence (excitation/emission peaks at 449/520 nm) as acceptors. These CDs are conjugated with PCT antibodies to form immune probes. Upon antigen binding, the antibody quickly binds and reacts with the antigen. Concurrently, the fluorescence energy from the donor is transferred to the acceptor, leading to a decrease in the blue fluorescence intensity emitted by B-CDs. The ratio of the fluorescence intensity between the green probe (G-CDs-Ab2) and the blue probe (B-CDs-Ab1) exhibited a positive correlation with the concentration of PCT antigen in the low concentration range of 0.5 to 5 ng/mL. Consistent results were observed in both phosphate-buffered saline (PBS) and diluted human serum. In the concentration range of 0.5–5 ng/mL in PBS buffer, the linear equation is expressed as F A /F D  = −0.0045 ×  c [PCT] ^2 + 0.0021 ×  c [PCT]  + 0.3232, with a limit of detection (LOD) of 0.1 ng/mL based on 3σ K −1 . For concentrations spanning from 5-80 ng/mL, the equation is F A /F D  = 0.0021 ×  c [PCT]  + 0.4212. These findings suggest that carbon dot-labeled immune probes could offer a novel approach for sensitive detection of disease biomarkers.

FRET-triggered fluorescence switching: a g-CNQDs/Cu-TCPP “turn-on” aptasensor for ultrasensitive GP73 detection

Zhou Yu,Wu Runqiang,Fan Lingling,Qui Pengtao,Deng Xiaohua,Tan Xiaohong,Dong Shuaikang,Li Canru,Chen Jiejing,Liang Jintao,Li Guiyin

Hepatocellular carcinoma (HCC) is one of the most lethal malignancies worldwide, and early diagnosis is crucial. Golgi protein 73 (GP73) has emerged as a promising serum biomarker for HCC. However, current detection methods often fail to meet routine screening requirements due to limitations in sensitivity and operational simplicity. To address these challenges, we have developed a novel fluorescent aptamer-based sensor for highly sensitive GP73 detection based on the fluorescence resonance energy transfer (FRET) mechanism between graphitic carbon nitride quantum dots (g-CNQDs) and a copper-based metal–organic framework (Cu-TCPP). g-CNQDs were covalently conjugated with a GP73-specific aptamer to serve as the fluorescent donor, while two-dimensional Cu-TCPP nanosheets acted as the efficient acceptors. Fluorescence was quenched upon donor–acceptor interaction via FRET. In the presence of GP73, aptamer–target binding disrupted FRET interaction, separating the donor from the acceptor and restoring fluorescence in a concentration-dependent manner. Under optimal conditions, the sensor exhibited excellent linearity over a concentration range 1.0–225.0 ng mL⁻¹, with a detection limit as low as 0.907 ng mL⁻¹. Recoveries for spiked human serum samples ranged from 95.96% to 103.85%, with relative standard deviations (RSDs) of 0.38%–5.48%. The developed aptamer sensor demonstrated excellent sensitivity, selectivity, and stability, providing a powerful tool for early HCC diagnosis and offering strong potential for real-time analytical applications. Graphical A novel fluorescent aptamer sensor for GP73 detection by leveraging the fluorescence resonance energy transfer (FRET) mechanism between graphite-phase carbon nitride quantum dots (g-CNQDs) and a copper-based organic framework (Cu-TCPP). This platform achieved a good linear relationship with GP73 concentration in the range of 1.0–225.0 ng mL -1 for GP73 concentration, and a detection limit of 0.907 ng mL -1 .

Design and Synthesis of FRET-Mediated Multicolor and Photoswitchable Fluorescent Polymer Nanoparticles with Tunable Emission Properties

Jian Chen,Peisheng Zhang,Gang Fang,Pinggui Yi,Fang Zeng,Shuizhu Wu

Novel multicolor and photoswitchcable fluorescent polymer nanoparticles were prepared by one-step miniemulsion via methyl methacrylate (MMA) copolymerization with 4-ethoxy-9-allyl-1,8-naphthalimide (EANI), allyl-(7-nitro-benzo[1,2,5]oxadiazol-4-yl)-amine (NBDAA) and spiropyran-linked methacrylate (SPMA). Under visible-light conditions, SPMA moieties in polymer nanoparticles are colorless and nonfluorescent, by varying the incorporating ratio of two dyes (EANI and NBDAA), fluorescence resonance energy transfer (FRET)-mediated emission signatures can be tuned so that the nanoparticles exhibit multiple colors under a single wavelength excitation. Moreover, the fluorescence emission of EANI and NBDAA dyes in nanoparticles can be reversibly switched “on” and “off” through the FRET process by the alternating irradiation of UV and visible light. This class of novel photoswitchable multicolor fluorescent polymer nanoparticles may find potential applications in multiplexed bioanalysis.

Aptamer based fluorometric determination of ATP by exploiting the FRET between carbon dots and graphene oxide

Cheng Xia,Cen Yao,Xu Guanhong,Wei Fangdi,Shi Menglan,Xu Xiaoman,Sohail Muhammad,Hu Qin

The authors describe a fluorometric aptamer based assay for adenosine triphosphate (ATP). It is based on the use of carbon dots (CDs) and graphene oxide (GO). The resultant CD-aptamer is adsorbed on the surface of GO via π-stacking and hydrophobic interaction, and the fluorescence of CD-aptamer is quenched via fluorescence resonance energy transfer (FRET) between CDs and GO. If ATP is present, it will bind to the aptamer and the CD-aptamer will be desorbed from GO. This will suppress FRET and the fluorescence of the CDs is restored. Under the optimal conditions and at typical excitation/emission wavelengths of 358/455 nm, the assay has a 80 pM detection limit and a linear range that extends from 0.10 to 5.0 nM concentrations of ATP. The method was successfully applied to the determination of ATP in yogurt samples. This method can also be conceivably applied to the detection of other analytes for which appropriate aptamers are available. Schematic of a novel fluorometric ATP assay based on the fluorescence resonance energy transfer (FRET) between aptamer modified carbon dots (CD-aptamer) and graphene oxide (GO). CD-aptamer was used as the energy donor and molecular recognition probe, and GO acted as energy acceptor. This assay exhibits high sensitivity and selectivity with a detection limit as low as 80 pM.

A novel FRET-based fluorescent chemosensor of β-cyclodextrin derivative for TNT detection in aqueous solution

Lijuan Feng,Cuiyan Tong,Yao He,Bingxin Liu,Chunyu Wang,Jie Sha,Changli Lü

A new fluorescent sensor based on single water-soluble β-cyclodextrin (β-CD) molecule as the support carrier of donor and acceptor complex was designed, and used for TNT detection by fluorescence resonance energy transfer (FRET). In this sensing platform, per-6-amino-β-CD (per-6-NH 2 -β-CD) was used as the detection vehicle. The probe dye of fluorescein 5(6)-isothiocyanate (FITC) was covalently linked onto the per-6-NH 2 -β-CD rim with the grafting molar ratio of 1:1, the residual amino groups of per-6-NH 2 -β-CD can adsorb TNT molecules by forming Meisenheimer complex (TNT–amine complex) with TNT. The absorbtion spectrum of this complex has a spectral overlapping with the emission of FITC in aqueous solution, so it can strongly suppress the fluorescence emission of the adjacent FITC through FRET on β-CD vehicle. This FRET-based fluorescent sensing technique provides a facile, ultrasensitive and selective detection method for TNT molecule. The observed linear fluorescence intensity change could allow the quantitative detection TNT with the detection limit of 20   nM in water.
