Background Alzheimer's Disease (AD) is a serious neurodegenerative disease and there is currently no effective treatment for AD progression. The use of TCM as a potential treatment strategy for AD is an evolving field of investigation. Asafoetida (ASF), an oleo-gum-resin isolated from Ferula assa-foetida root, has been proven to possess antioxidative potential and neuroprotective effects, which is closely associated with the neurological disorders. However, the efficacy and further mechanisms of ASF in AD experimental models are still unclear. Methods A cognitive impairment of mouse model induced by scopolamine was established to determine the neuroprotective effects of ASF in vivo , as shown by behavioral tests, biochemical assays, Nissl staining, TUNEL staining, Immunohistochemistry, western blot and qPCR. Furthermore, the PC12 cells stimulated by H 2 O 2 were applied to explore the underlying mechanisms of ASF-mediated efficacy. Then, the UPLCM analysis and integrated network pharmacology approach was utilized to identified the main constitutes of ASF and the potential target of ASF against AD, respectively. And the main identified targets were validated in vitro by western blot, qPCR and immunofluorescence staining. Results In vivo , ASF treatment significantly ameliorated cognitive impairment induced by scopolamine, as evidenced by improving learning and memory abilities, and reducing neuronal injury, cholinergic system impairment, oxidative stress and apoptosis in the hippocampus of mice. In vitro , our results validated that ASF can dose-dependently attenuated H 2 O 2 -induced pathological oxidative stress in PC12 cells by inhibiting ROS and MDA production, as well as promoting the activities of SOD, CAT, GSH. We also found that ASF can significantly suppressed the apoptosis rate of PC12 cells increased by H 2 O 2 exposure, which was confirmed by flow cytometry analysis. Moreover, treatment with ASF obviously attenuated H 2 O 2 -induced increase in caspase-3 and Bax expression levels, as well as decrease in Bcl-2 protein expression. KEGG enrichment analysis indicated that the PI3K/Akt/GSK3β/Nrf2 /HO-1pathway may be involved in the regulation of cognitive impairment by ASF. The results of western blot, qPCR and immunofluorescence staining of vitro assay proved it. Conclusions Collectively, our work first uncovered the significant neuroprotective effect of ASF in treating AD in vivo . Then, we processed a series of vitro experiments to clarify the biological mechanism action. These data demonstrate that ASF can inhibit oxidative stress induced neuronal apoptosis to foster the prevention of AD both in vivo and in vitro , and it may exert the function of inhibiting AD through PI3K/Akt/GSK3β/Nrf2/HO-1pathway.

In this work, Yb3+/RE3+ (RE = Er/Ho) co-doped Zn3Mo2O9 phosphors were synthesized by high-temperature solid-state reactions. Under 980 nm excitation, the upconversion (UC) luminescence thermal enhancement was obtained for Zn3Mo2O9:Yb3+/RE3+ phosphors. The green emission intensity of the Zn3Mo2O9:Yb3+/Er3+ sample was increased 5 times from 373 to 573 K. The red emission intensity of the Zn3Mo2O9:Yb3+/Ho3+ sample was enhanced 7.92 times. The anomalous thermal enhancement of UC emission was induced by the negative thermal expansion (NTE) of the Zn3Mo2O9 host. The energy transfer rate from the sensitizer (Yb3+) to the activator (RE3+) was enhanced because of the lattice contraction and distortion for NTE materials. Compared with the UC emission of Er3+single doped Zn3Mo2O9 sample, the luminescence thermal enhancement was absent, which contributed to proving the physical mechanism. The temperature sensing properties of the Zn3Mo2O9:Yb3+/Er3+ and Zn3Mo2O9:Yb3+/Ho3+ samples were also investigated based on the fluorescence intensity ratio (FIR) technology. The absolute sensitivity (SA) and relative sensitivity (SR) of Zn3Mo2O9:Yb3+/Er3+ phosphor reached 0.0060 K−1 and 0.72% K−1, which is based on the thermal coupling levels (2H11/2, 4S3/2) FIR of Er3+ ions. In addition, the SA and SR of Zn3Mo2O9:Yb3+/Ho3+ phosphor reached 0.0119 K−1 and 0.86% K−1, that is based on the non-thermal coupling levels (5S2/5F4, 5F5) FIR of Ho3+ ions. The research results indicate that the Zn3Mo2O9 host shows NET. The Yb3+/RE3+ co-doped Zn3Mo2O9 phosphors are good materials for highly sensitive optical temperature measurement, which can be used to develop thermally enhanced ratiometric optical thermometers.

SrWO4 phosphors doped with Ho3+(Er3+)/Yb3+ are successfully prepared by a high temperature solid-state reaction method. The upconversion (UC) luminescence properties of all the samples have been investigated under 980 nm excitation. Strong green emissions are obtained in the SrWO4:Yb3+/Ho3+ and SrWO4:Yb3+/Er3+ samples with the naked eyes. In a temperature range going from 303 K to 573 K, the UC emission spectra of the phosphors have been measured. Then the temperature sensing properties also have been discussed via fluorescence intensity ratio (FIR) technology. For the SrWO4:Yb3+/Ho3+ phosphor, the FIR technologies based on thermal coupling levels (TCLs)(5F4,5F5) and non-thermal coupling levels (non-TCLs)(5S2, 5F4/5F5) are used for investigating the sensitivity. The results show that the maximum absolute sensitivity reaches 0.0158 K−1 with non-TCLs. As for Yb3+/Er3+ codoped SrWO4 phosphor, the maximum absolute sensitivity reaches 0.013 K−1 with TCLs (2H11/2,4S5/2) at a temperature of 513 K. These significant results demonstrate that the SrWO4:Ho3+(Er3+)/Yb3+ phosphors are robust for optical temperature sensors.

Pulmonary hypertension is divided into five groups, and chronic exposure to hypoxia causes pulmonary hypertension in the third group -- high altitude pulmonary hypertension, which is commonly seen in patients with chronic obstructive pulmonary disease, sleep-disordered breathing, and interstitial lung disease. Its clinicopathological features include right heart overload and right ventricular hypertrophy, with severe patients potentially experiencing right heart failure or even death. The pathogenesis of high altitude pulmonary hypertension is complex; it includes oxidative stress, mitochondrial dysfunction, apoptosis, and pyroptosis. Pharmacological therapy may alleviate the condition if immediate descent to lower altitudes or supplemental oxygen is not possible. Herein, we preliminarily investigated the effect of methyleugenol on high altitude pulmonary hypertension through in vitro and in vivo experiments. Methyleugenol alleviated pulmonary arteriole constriction by acting on potassium ion channels, and inhibited the proliferation of pulmonary artery smooth muscle cells(PASMCs) by decreasing and reactive oxygen species levels, as well as inhibiting the expression of HIF-1α. The cardiac systolic function improved and mitochondrial structure of pulmonary artery smooth muscle cells in rats with high altitude pulmonary hypertension was significantly improved after methyleugenol gavage intervention, and malondialdehyde levels in serum of rats were decreased while those of superoxide dismutase and glutathione increased, suggesting that the reduction of oxidative stress may be one of the mechanisms to improve mitochondrial function. Western blot experiments showed that methyleugenol regulated the oxidative stress level in rats by activating Nrf2/HO-1 signaling pathway, activate p27Kip1, inhibit CDK4/CCND1/ p-ERK1/2 and inhibited HIF-1α/RASSF1/Ras axis to inhibit cell proliferation, thereby improving the occurrence and development of high altitude pulmonary hypertension in rats. The results of this study reveal that methyl eugenol has the potential to alleviate pulmonary vasoconstriction and remodeling in rat models of high-altitude pulmonary hypertension, providing a scientific basis for the application of this compound in improving pulmonary vascular remodeling.

Continuous remodeling of the heart can result in adverse events such as reduced myocardial function and heart failure. Available evidence indicates that ferroptosis is a key process in the emergence of cardiac disease. P2 family purinergic receptor P2X7 receptor (P2X7R) activation plays a crucial role in numerous aspects of cardiovascular disease. The aim of this study was to elucidate any potential interactions between P2X7R and ferroptosis in cardiac remodeling stimulated by angiotensin II (Ang II), and P2X7R knockout mice were utilized to explore the role of P2X7R and elucidate its underlying mechanism through molecular biological methods. Ferroptosis is involved in cardiac remodeling, and P2X7R deficiency significantly alleviates cardiac dysfunction, remodeling, and ferroptosis induced by Ang II. Mechanistically, Ang II interacts with P2X7R directly, and LYS-66 and MET-212 in the C-type domain form a binding complex with Ang II. P2X7R blockade influences HuR-targeted GPX4 and HO-1 mRNA stability by affecting the shuttling of HuR from the nucleus to the cytoplasm and its expression. These results suggest that focusing on P2X7R could be a possible therapeutic approach for the management of hypertensive heart failure.

Fucoxanthin (Fx) has garnered significant interest due to its exceptional biological properties. However, its efficacy in enhancing food quality and human health is contingent upon the solubility of the compound in water and its physicochemical stability. Therefore, nanocarriers must be developed to enhance the stability and biocompatibility of Fx. In this study, oxidized paramylon and Fx self-assembled nanoparticles (Fx-OEP) were prepared via the anti-solvent method, with a loading rate of 82.47 % for Fx. The Fx-OEP exhibited robust storage and photostability. In vitro simulated digestion assays demonstrated that Fx-OEP effectively protected Fx from premature gastric release, while achieving a release efficiency of 72.17 % in the intestinal phase. Fx-OEP has the capacity to scavenge a range of reactive oxygen species (ROS) induced by cellular oxidative stress. Treatment with Fx-OEP resulted in a significant reduction in ROS accumulation in insulin-resistant HepG2 cells, which was attributed to the activation of the nuclear factor E2-related factor 2/heme oxygenase-1 (Nrf2/HO-1) pathway. This, in turn, activated insulin receptor substrate 1/glucose transporter type 4 (IRS1/GLUT4), promoting cellular glucose absorption and utilization. These findings indicate the potential of self-assembled nanoparticles based on oxidized paramylon as a new type of nanocarrier for delivering hydrophobic substances.

High-fat diets (HFDs) impair the intestinal barrier by increasing permeability and oxidative stress, which contributes to obesity-related disorders. Ulva lactuca, a marine green alga, produces extracellular vesicles (UEVs) that remain underexplored as functional food components. In this study, UEVs were isolated and characterized, showing a cup-like shape morphology with average diameter of 188.8 nm. Functional assays demonstrated that UEV treatment attenuated oxidative stress, reduced apoptosis, and restored tight junction proteins. In a gut-on-a-chip model, UEVs improved transepithelial electrical resistance (TEER) and enhanced mucin secretion under HFD-mimicking stress. Mechanistic studies revealed that UEVs activated the Nrf2/Keap1/HO-1 signaling pathway, promoting nuclear translocation of Nrf2 and upregulation of antioxidant genes. Collectively, these findings highlight UEVs as novel food-derived nanostructures with dual functions in oxidative stress reduction and intestinal barrier protection, supporting their potential in functional food applications targeting metabolic gut disorders.

Ba3Sc2F12:Yb3+, Ln3+ (Ln = Er, Ho, Tm) crystals with various morphologies have been synthesized via a one step hydrothermal route. X-ray diffraction (XRD), scanning electron microscopy (SEM), and up conversion photoluminescence (UCPL) spectra were used to characterize the samples. The influences of surfactants, pH values, and molar ratio of F−/Sc3+ on the crystal phase, size and shape of final products have been studied in detail. The aspect ratio of products increased gradually with the increase of F−/Sc3+ molar ratio. Additionally, the luminescence properties of Ba3Sc2F12:Yb3+, Ln3+ (Ln = Er, Ho, Tm) crystals were systematically studied. The blue emission is attributed to the 1G4 → 3H6 transition of Tm3+; the green emission can be obtained due to the 2H11/2/4S3/2 → 4I15/2 transitions of Er3+ and the 5S2/5F4 → 5I8 transition of Ho3+; the red emission comes from the 4F9/2 → 4I15/2 transition of Er3+ and the 1G4 → 3F4 transition of Tm3+. Based on the generation of red, green, and blue emissions in the different Ln ion-doped Ba3Sc2F12, the white light emission can be obtained by appropriately doping Yb3+, Er3+, and Tm3+ in the present Ba3Sc2F12 crystals due to the color superposition principle. Here, the sample Ba3Sc1.5856F12:0.4Yb3+, 0.01Er3+, 0.0044Tm3+ crystals showed suitable intensity ratio of blue, green and red (RGB) emissions resulting in bright white light with CIE-x = 0.274 and CIE-y = 0.287, which was illustrated by a photograph under excitation of 980 nm. The prepared Ba3Sc2F12:Ln3+ phosphor has potential applications in the fields of three dimensional displays, back lighting and white light sources.

Osteoarthritis (OA) is a progressive degenerative joint disorder characterized by articular cartilage degradation, persistent oxidative stress, and chronic inflammation. Targeting oxidative damage within the joint microenvironment represents a promising avenue for disease modification, offering potential to preserve cartilage integrity, alleviate inflammatory cascades, and ultimately delay structural progression. Lipid nanoparticles were prepared by thin-film hydration and extrusion, then embedded into GelMA microspheres via a microfluidic droplet platform with UV crosslinking to form a sustained-release system. Biocompatibility, ROS-scavenging, anti-inflammatory, and chondroprotective effects were evaluated in vitro and in vivo, while transcriptome sequencing was applied to elucidate system-induced molecular alterations. In vitro , Api-LNP@GMS markedly attenuated ROS accumulation and inflammatory responses in IL-1β-stimulated chondrocytes, as demonstrated by ROS fluorescence imaging, immunofluorescence, Western blotting, and qRT-PCR analyses. In vivo , intra-articular administration in anterior cruciate ligament transection (ACLT) rats effectively alleviated joint inflammation while preserving cartilage integrity and functional capacity. Moreover, transcriptome-wide RNA sequencing, integrated with pathway enrichment analysis and validated by molecular assays, revealed robust activation of the Nrf2–HO-1 antioxidant signaling axis. Api-LNP@GMS is a multifunctional antioxidant biomaterial platform that integrates a natural therapeutic with a sustained-release delivery system, offering a safe and effective strategy to reprogram the osteoarthritic microenvironment and mitigate disease progression. Mg 3 (Sb, Bi) 2 is a promising mid- to low- temperature thermoelectric material owing to its earth-abundant composition. However, its performance is limited by low intrinsic carrier concentration and the inherent trade-off among thermoelectric parameters. Herein, large-grain sized Mg 3.01 Sb 1.3 Bi 0.69 Te 0.01 Ho 0.07 are fabricated via melting method at 1423 K using a tantalum-sealing technique. A high peak zT of 1.6 at 700 K is realized, along with a peak power factor of 24 μW cm −1 K −2 and a low κ L of 0.41 W m −1 K −1 . The superior performance originates from synergistic manipulation of electron and phonon transport. Ho doping optimizes carrier concentration up to 7.2 × 10 19 cm −3 due to its small electronegativity difference with Mg, while the large grain ensures high carrier mobility. Simultaneously, atomic-scale alloy disorder suppresses phonon propagation, resulting in a substantial reduction in lattice thermal conductivity. This work highlights the great potential of synergistic transport engineering for high-performance Mg 3 (Sb, Bi) 2 -based thermoelectric materials.

Acute renal injury (AKI) has a high incidence rate and mortality, but current treatment methods are limited. As a kind of nanomaterial with enzyme-like activity, nanozyme has shown outstanding advantages in treating AKI according to recent reports. Herein, we assess the potential of manganese-based nanozymes (MnO2–BSA NPs) with excellent biosafety in effectively alleviating AKI. Our findings in vitro and in vivo reveal that MnO2–BSA NPs exert regulatory effects on oxidative stress, inflammation, and apoptosis. These effects are mediated through activation of the Nrf2/HO-1 and PI3K/Akt/NF-κB pathways. Notably, it was observed that the cytoprotective effect of MnO2–BSA NPs is abrogated upon inhibition of Nrf2 expression, highlighting the important role of this transcription factor in cellular protection. In summary, the study demonstrates the protective effect of MnO2–BSA NPs in AKI and provides the molecular mechanisms involved, which can contribute to the advancement of potential therapeutic interventions for nanozyme-based treatments.
