Rapid discovery of drug-introduced multiple organ dysfunction via NIR-II fluorescent imaging

Pu Jiang,Ruihu Song,Yue Hu,Xin He,Zewei Zhang,Xuemei Wei,Zhiming Wang,De-an Guo,Hao Chen

The precise and rapid monitoring of multiple organ dysfunction is crucial in drug discovery. Traditional methods, such as pathological analysis, are often time-consuming and inefficient. Here, we developed a multiplexed near-infrared window two (NIR-II) fluorescent bioimaging method that allows for real-time, rapid, and quantitative assessment of multiple organ dysfunctions. Given that existing probes did not fully meet requirements, we synthesized a range of NIR-II hemicyanine dyes (HDs) with varying absorption and emission wavelengths. By modifying these dyes, we achieved high spatial and temporal resolution imaging of the liver, kidneys, stomach, and intestines. This method was further applied to investigate disorders induced by cisplatin, a drug known to cause gastric emptying issues along with liver and kidney injuries. By monitoring the metabolic rate of the dyes in these organs, we accurately quantified multi-organ dysfunction, which was also confirmed by gold-standard pathological analysis. Additionally, we evaluated the effects of five aristolochic acids (AAs) on multiple organ dysfunction. For the first time, we identified that AA-I and AA-II could cause gastric emptying disorders, which was further validated through transcriptomics analysis. Our study introduces a novel approach for the simultaneous monitoring of multi-organ dysfunction, which may significantly enhance the evaluation of drug side effects.

Ni-doped carbon dots unlock dual NIR-II photothermal and photodynamic cancer therapy

Jun Song,Minghao Kang,Yongping Li,Jiaqing Guo,Huibo Wang,Liwei Liu,Junle Qu,Shuai Ye

Due to their excellent biocompatibility, outstanding water dispersibility, and multifunctional integration capability, carbon dots have emerged as highly promising materials for cancer phototherapy. In this study, nickel-doped carbon dots (Ni-CDs) were successfully synthesized via a one-step hydrothermal method, which enables efficient and uniform Ni incorporation within the carbon framework. Ni-CDs exhibit strong absorbance in the range of 840–1100 nm. They have a reactive oxygen species (ROS) production rate of 3.27% and a photothermal conversion efficiency of 61.33% under 1064 nm laser irradiation. The enhanced dual-mode performance can be attributed to Ni-induced nonradiative relaxation and improved electron transfer. They are the first reported nickel-doped carbon dots with synergistic therapeutic capabilities of PDT/PTT in the NIR-II region. In vitro and in vivo experiments demonstrated that Ni-CDs can effectively induce tumor cell death, with no significant toxic damage observed in normal tissues/organs. This study highlights the potential of Ni-CDs as a multifunctional nanoplatform for deep-tissue cancer treatment, providing a reference for the design of materials for the synergistic combination of photothermal and photodynamic therapy of deep tumors.

Sentinel lymph node identification using NIR-II ultrabright Raman nanotags on preclinical models

Binge Deng,Yan Wang,Xiangdong Bu,Jin Li,Jingsong Lu,Linley Li Lin,Yaohui Wang,Yao Chen,Jian Ye

Surface-enhanced Raman spectroscopy (SERS) nanotags have garnered much attention as promising bioimaging contrast agent with ultrahigh sensitivity, but their clinical translation faces challenges including biological and laser safety. As breast sentinel lymph node (SLN) imaging agents, SERS nanotags used by local injection and only accumulation in SLNs, which were removed during surgery, greatly reduce biological safety concerns. But their clinical translation lacks pilot demonstration on large animals close to humans. The laser safety requires irradiance below the maximum permissible exposure threshold, which is currently not achievable in most SERS applications. Here we report the invention of the core-shell SERS nanotags with ultrahigh brightness (1 pM limit of detection) at the second near-infrared (NIR-II) window for SLN identification on pre-clinical animal models including rabbits and non-human primate. We for the first time realize the intraoperative SERS-guided SLN navigation under a clinically safe laser (1.73 J/cm 2 ) and identify multiple axillary SLNs on a non-human primate. No evidence of biosafety issues was observed in systematic examinations of these nanotags. Our study unveils the potential of NIR-II SERS nanotags as appropriate SLN tracers, making significant advances toward the accurate positioning of lesions using the SERS-based tracer technique.

Microenvironment-Activatable Long-Wavelength NIR-II Visualization and Synergistic Treatment of Pulmonary Embolism

Xue Meng,Jianwen Song,Zekun Du,Yongyou Tao,Ji Qi

Pulmonary embolism (PE) is a life-threatening thrombotic condition, yet its precise diagnosis and effective treatment remain a major clinical challenge. In this study, we report the development of a microenvironment-activatable nanoplatform that enables hypoxia-triggered long-wavelength second near-infrared (NIR-II) fluorescence imaging for accurate thrombus visualization, combined with photothermal therapy (PTT) and controlled hydrogen sulfide (H2S) release for synergistic therapeutic intervention. We first synthesize an N-oxide-based molecular probe that undergoes hypoxia-induced structural transition, thereby activating both NIR-II fluorescence and photothermal properties. This probe, together with a thermosensitive H2S donor, is encapsulated into nanoparticles, which are further functionalized with bis-serotonin groups to achieve thrombus-specific targeting via the myeloperoxidase and hydrogen peroxide-mediated aggregation. In a PE mouse model, the nanoagent enables highly selective and sensitive visualization of thrombi via activatable NIR-II fluorescence imaging after intravenous injection, achieving a remarkable signal-to-noise ratio exceeding 120. The combined PTT and H2S therapy significantly enhance thrombolytic efficacy compared to conventional treatment (e.g., urokinase). Importantly, the nanoagent exhibits excellent biocompatibility in vivo, with minimal risk of hemorrhagic complications. This high-performance hypoxia-responsive platform offers a promising strategy for the precise diagnosis and effective treatment of PE and other cardiovascular diseases.

Non-Small Cell Lung Cancer Treatment via Cu9S8-Mediated NIR-II Photothermal Therapy

Huifang Hao,Zhe Yu,Cheng Cao,Yingao Jiao,Yu Cao,Xinyuan Cui,Yan Gao,Dengyao Gao,Shengsheng Cui,Chenyu Wang,Yanlei Liu

Non-small cell lung cancer (NSCLC) treatment is often compromised by acquired resistance and, in the context of photothermal therapy (PTT), by tumor thermotolerance─a protective stress response mediated by the molecular chaperone heat shock protein 90 (HSP90). To address this dual challenge, we developed a multifunctional nanoplatform (Cu9S8@Lum NPs) that synergistically integrates NIR-II PTT with HSP90 inhibition. This nanosystem is based on a Cu9S8 nanocore with high photothermal conversion efficiency, loaded with the HSP90 inhibitor Luminespib and designed for pH-responsive drug release. This spatiotemporally coordinated strategy enables concurrent tumor hyperthermia and molecular interference with the heat-shock defense mechanism. In vitro, the nanoparticles effectively suppressed HSP90 expression, exacerbated cellular oxidative stress, and induced mitochondrial apoptosis, resulting in markedly enhanced cytotoxicity compared to PTT alone. In vivo, the nanoplatform demonstrated effective tumor accumulation and, under mild NIR-II irradiation, achieved significant tumor growth inhibition in a syngeneic model. This work presents a mechanism-informed nanomaterial strategy to overcome thermotolerance in NSCLC, advancing the development of synergistic cancer nanotherapeutics.

Targeted Inflammatory Microenvironment Remodeling and NIR-II Photothermal Therapy for Abdominal Aortic Aneurysm

Yijun Liu,Peng Qiu,Huifang Hao,Ang’ang Ding,Zhaoyu Wu,Jiahao Lei,Zhaoxi Peng,Hongji Pu,Chenlin Zeng,Hongbin Guo,Xiaodong Wu,Huaxiang Lu,Jinbao Qin,Ruihua Wang,Haoqi Liu,Kaichuang Ye,Xinwu Lu

Abdominal aortic aneurysm (AAA) is a degenerative aortic disease with high rupture-associated mortality. There is no effective medical therapy for early intervention, inflammatory microenvironment remodeling is a crucial therapeutic strategy. Herein, we designed a multifunctional nanoplatform, Cu 9 S 8 @MCC950-Belnacasan-cRGDfK (CS@MBR) for AAA treatment. This platform employed a Cu 9 S 8 core for near-infrared-II (NIR-II) photothermal therapy (PTT), encapsulated NLRP3 inflammasome inhibitors (MCC950 and Belnacasan), and coated with a cRGDfK peptide for active targeting of the aneurysm. In vitro, CS@MBR with mild PTT inhibited NLRP3 inflammasome activation, decreased pro-inflammatory macrophage polarization, and prevented the phenotype switching of vascular smooth muscle cells. In vivo, CS@MBR demonstrated excellent accumulation at the aneurysm site in an angiotensin-II-induced murine AAA model. Upon NIR-II irradiation, the treatment effectively inhibited AAA development, damage of elastic fibers, and imbalance of the inflammatory microenvironment. This study presents a promising, targeted nanotherapeutic strategy for the treatment of AAA and other inflammatory vascular diseases.

Pb-Doped Ag2Se Quantum Dots with Enhanced Photoluminescence in the NIR-II Window

Mengxuan Yu,Xiaohu Yang,Yejun Zhang,Hongchao Yang,Haoying Huang,Zan Wang,Jinyi Dong,Rong Zhang,Ziqiang Sun,Chunyan Li,Qiangbin Wang

Ag 2 Se quantum dots (QDs) as an effective biological probe in the second near-infrared window (NIR-II, 1000–1700 nm) have been widely applied in bioimaging with high tissue penetration depth and high spatiotemporal resolution. However, the ions deficiency and crystal defects caused by the high Ag + mobility in Ag 2 Se crystals are mainly responsible for the inefficient photoluminescence (PL) of Ag 2 Se QDs. Herein, a tailored route is reported to achieve controllable doping of Ag 2 Se QDs in which Ag is exchanged by Pb via cation exchange (CE), which is unattainable by direct synthetic methods. The Pb-doped Ag 2 Se QDs (denoted as Pb:Ag 2 Se QDs) present fire-new optical features with significantly enhanced PL intensity of 4.2 folds. Photoelectron spectroscopy confirms that Pb acts as an n-type dopant for Ag 2 Se QDs and therefore the electronic impurities provide additional carriers to fill the traps. Moreover, the general validity of this method is demonstrated to convert different sized Ag 2 Se into Pb:Ag 2 Se QDs, so that a wide range of NIR-II PL with high intensity is obtained. The bright NIR-II emission of Pb:Ag 2 Se QDs is further successfully performed in lymphatic system mapping.

NIR-II-Activated Yolk–Shell Nanostructures as an Intelligent Platform for Parkinsonian Therapy

Yao Liu,Daoming Zhu,Jingshan Luo,Xiaojia Chen,Liqian Gao,Wei Liu,Tongkai Chen

Despite the relative severity of Parkinson’s disease (PD), to date, there has been only limited success in preventing or treating this condition owing to the low permeability of the blood–brain barrier (BBB), which makes the cerebral delivery of pharmaceutical agents very challenging. In the present study, we explored an approach to increasing BBB permeability via the use of mesoporous silica-encapsulated gold nanorods (MSNs-AuNRs) that could reliably achieve a robust photothermal effect in response to second near-infrared (NIR-II) irradiation. To test the potential anti-Parkinsonian activity, we loaded MSNs-AuNRs with the frequently utilized anti-PD agent quercetin (QCT) to yield MSNs-AuNRs@QCT. Following NIR-II irradiation, we observed a dramatical increase in QCT transfer through the BBB, indicating that MSNs-AuNRs may enhance BBB permeability via the photothermal effect. These findings were further supported by rat pharmacokinetic studies, which clearly revealed that the combination of MSNs-AuNRs and NIR-II irradiation was associated with significantly improved brain accumulation. In a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced murine model of PD, we also found that intravenous MSNs-AuNRs@QCT delivery and subsequent NIR-II irradiation was sufficient to markedly reduce the neurological abnormalities in these mice. Together, this combination offers promising potential as a nanoplatform for neuroprotective drug delivery and treatment of PD.

Lanthanide/Cu2–xSe Nanoparticles for Bacteria-Activated NIR-II Fluorescence Imaging of Infection

Leilei Sun,Sisi Shi,Zeming Wu,Yao Huang,Chenhui Ji,Craig A. Grimes,Xinxin Feng,Qingyun Cai

A material system enabling specific NIR-II fluorescence imaging of Gram-positive bacteria is described. The material system is based on the electrostatic binding of Cu2–xSe and vancomycin-modified NaGdF4:Nd,Yb@NaGdF4 downconversion nanoparticles (DCNPs), the fluorescence of which is weak owing to the spectral overlap of Cu2–xSe absorption with the DCNP NIR emission. The presence of Gram-positive bacteria precisely disconnects the bond between vancomycin-modified DCNPs and Cu2–xSe, thus enabling a strong fluorescent signal. In vivo studies show that the material system can be specifically activated at the site of Gram-positive bacterial infection but is essentially nonfluorescent in the area of Gram-negative bacterial infection.

Extracellular matrix-degrading STING nanoagonists for mild NIR-II photothermal-augmented chemodynamic-immunotherapy

Zhan Meixiao,Yu Xiangrong,Zhao Wei,Peng Yongjun,Peng Shaojun,Li Jingchao,Lu Ligong

Regulation of stimulator of interferon genes (STING) pathway using agonists can boost antitumor immunity for cancer treatment, while the rapid plasma clearance, limited membrane permeability, and inefficient cytosolic transport of STING agonists greatly compromise their therapeutic efficacy. In this study, we describe an extracellular matrix (ECM)-degrading nanoagonist (dNAc) with second near-infrared (NIR-II) light controlled activation of intracellular STING pathway for mild photothermal-augmented chemodynamic-immunotherapy of breast cancer. The dNAc consists of a thermal-responsive liposome inside loading with ferrous sulfide (FeS2) nanoparticles as both NIR-II photothermal converters and Fenton catalysts, 2′3′-cyclic guanosine monophosphate-adenosine monophosphate (cGAMP) as the STING agonist, and an ECM-degrading enzyme (bromelain) on the liposome surface. Mild heat generated by dNAc upon NIR-II photoirradiation improves Fenton reaction efficacy to kill tumor cells and cause immunogenic cell death (ICD). Meanwhile, the generated heat triggers a controlled release of cGAMP from thermal-responsive liposomes to active STING pathway. The mild photothermal activation of STING pathway combined with ICD promotes anti-tumor immune responses, which leads to improved infiltration of effector T cells into tumor tissues after bromelain-mediated ECM degradation. As a result, after treatment with dNAc upon NIR-II photoactivation, both primary and distant tumors in a murine mouse model are inhibited and the liver and lung metastasis are effectively suppressed. This work presents a photoactivatable system for STING pathway and combinational immunotherapy with improved therapeutic outcome.Graphical Abstract

Cellulose Nanocrystal Films with NIR-II Circularly Polarized Light for Cancer Detection Applications

Di Lu,Mengfei Li,Xiaoqing Gao,Xiao Yu,Lihong Wei,Shoujun Zhu,Yan Xu

Near-infrared circularly polarized light is attractive for wide-ranging applications. However, high-performance near-infrared circularly polarized light is challenging to realize. Here, we show that left-handed chiral photonic cellulose nanocrystal (CNC) films produced from ultrasonicated suspensions enable right-handed circularly polarized luminescence with a dissymmetry factor of −0.330 in the second near-infrared window (NIR-II). We present a theoretical analysis of the adverse effect of structural defects and luminescence intensity heterogeneity on the right-handed circularly polarized luminescence glum inside the bandgap and the occurrence of left-handed circularly polarized luminescence at the band edges. We demonstrate the potential of the chiral photonic CNC films with NIR-II circularly polarized light for cancer cell discrimination. The present work identifies key scientific questions in CNC-based circularly polarized luminescence materials research.

Bioactive NIR-II gold clusters for three-dimensional imaging and acute inflammation inhibition

Huizhen Ma,Xiaoning Zhang,Ling Liu,You Huang,Si Sun,Ke Chen,Qi Xin,Pengfei Liu,Yuxing Yan,Yili Wang,Yuan Li,Haile Liu,Ruoli Zhao,Kexin Tan,Xinzhu Chen,Xun Yuan,Yonghui Li,Ying Liu,Haitao Dai,Changlong Liu,Hao Wang,Xiao-Dong Zhang

Strong fluorescence and high catalytic activities cannot be achieved simultaneously due to conflicts in free electron utilization, resulting in a lack of bioactivity of most near-infrared-II (NIR-II) fluorophores. To circumvent this challenge, we developed atomically precise Au22 clusters with strong NIR-II fluorescence ranging from 950 to 1300 nm exhibiting potent enzyme-mimetic activities through atomic engineering to create active Cu single-atom sites. The developed Au21Cu1 clusters show 18-fold higher antioxidant, 90-fold higher catalase-like, and 3-fold higher superoxide dismutase–like activities than Au22 clusters, with negligible fluorescence loss. Doping with single Cu atoms decreases the bandgap from 1.33 to 1.28 eV by predominant contributions from Cu d states, and Cu with lost electron states effectuates high catalytic activities. The renal clearable clusters can monitor cisplatin-induced renal injury in the 20- to 120-minute window and visualize it in three dimensions using NIR-II light-sheet microscopy. Furthermore, the clusters inhibit oxidative stress and inflammation in the cisplatin-treated mouse model, particularly in the kidneys and brain.
