In this study, the interface-modified ternary blends based on polylactic acid (PLLA)-polycaprolactone (PCL)-polycarbonate diols (PCDL) were prepared via suspension flame spray. Morphological analysis showed that the interface modification of PCDL significantly improves the compatibility of PLLA and PCL. Mechanical analysis revealed that the addition of PCDL resulted in great enhancement of the toughness of the PLLA/PCL composite coatings with a dramatic 326 % increase of the fracture strain for 10PLLA/90PCL/10PCDL compared with 10PLLA/90PCL. Further physicochemical and antibacterial test revealed no significant difference among PLLA/PCL and PLLA/PCL/CAM composite coatings with and without PCDL. Time-course release analysis showed that the release mechanism of the composite coating generally remains the same when the PCDL added is lower than the main components in PLLA/PCL, and even changed from the original abnormal diffusion to Fick diffusion. Overall, the ternary blending approach of PLLA-PCL-PCDL using suspension flame spray is a promising technique with great potential drug-delivery applications with different mechanical requirements.

Introduction: Hierarchical nanofibrous scaffolds are emerging as a promising bone repair material due to their high cell adhesion activity and nutrient permeability. However, the existing method for hierarchical nanofibrous scaffolds fabrication is complicated and not perfectly suitable for further biomedical application in view of both structure and function. In this study, we constructed a hierarchical nanofibrous poly (l-lactic acid)/poly(ε-caprolactone) (PLLA/PCL) scaffold and further evaluated its bone healing ability. Methods: The hierarchical PLLA/PCL nanofibrous scaffold (PLLA/PCL) was prepared by one-pot TIPS and then rapidly mineralized at room temperature by an electrochemical deposition technique. After electrode-positioning at 2 V for 2 hrs, a scaffold coated with hydroxyapatite (M-PLLA/PCL) could be obtained. Results: The pore size of the M-PLLA/PCL scaffold was hierarchically distributed so as to match the biophysical structure for osteoblast growth. The M-PLLA/PCL scaffold showed better cell proliferation and osteogenesis activity compared to the PLLA/PCL scaffold. Further in vivo bone repair studies indicated that the M-PLLA/PCL scaffold could accelerate defect healing in 12 weeks. Conclusion: The results of this study implied that the as-prepared hydroxyapatite coated hierarchical PLLA/PCL nanofibrous scaffolds could be developed as a promising material for efficient bone tissue repair after carefully tuning the TIPS and electrodeposition parameters.

Solid polymer electrolytes (SPE) are of great importance for developing the next-generation all-solid-state lithium metal batteries, which are featured by their high safety and extraordinary energy density. Herein, by using the highly efficient azide-alkyne click chemistry, a series of long-subchain hyperbranched copolymers from various compositions of AB2 macromonomers of PCL-2N3 (polycaprolactone) and PS-2N3 (polystyrene) were synthesized and employed as SPE polymer host. Taking advantage of the hyperbranched structure, the crystallization of PCL was successfully suppressed, and endows the SPE with high ionic conductivity and high mechanical properties simultaneously. By doping with LiFSI, the SPEs thereof exhibit high ionic conductivity (1.59 × 10−4 S cm−1 at 80 °C), broad electrochemical stability window (>4.6 V) and high lithium-ion transference number (>0.40). The electrolyte film based on the HB-II (the hyperbranched PCL/PS copolymer with 70 wt% PCL and 30 wt% PS) shows an ionic conductivity of 1.36 × 10−5 S·cm−1 at 80 °C and a high lithium transference number of 0.49. The all-solid-state LiFePO4//Li cell with HB-II-based SPE delivers a high and stable discharge capacity (133 mAh·g−1 at 1 C, with 90% capacity retention after 300 cycles) and exhibits long lifetime up to over 1200 cycles, owing to its relatively high ionic conductivity and good interfacial stability.

Background An ideal wound dressing should exhibit good biocompatibility, minimize pain and infection, absorb excess exudates, and maintain a moist environment. However, few clinical products meet all these needs. Therefore, the aim of this study was to fabricate a multifunctional double layer nanofibrous scaffolds (DLS) as a potential material for wound dressing. Materials and methods The scaffold was formed from mupirocin and lidocaine hydrochloride homogeneously incorporated into polycaprolactone as the first layer of scaffolds and chitosan as the second layer of scaffolds nanofibers through electrospinning. The fabricated nanofibrous scaffolds were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, thermogravimetric analysis, differential scanning calorimetry, and measurement of swelling ratio, contact angle, drug release, and mechanical properties. Furthermore, antibacterial assessment, live/dead cell assays, and MTT assays were used to investigate the antibacterial activity and cytotoxicity of the nanofibrous scaffolds. Results The morphology of the nanofibrous scaffolds was studied by scanning electron microscopy, showing successful nanofibrous scaffolds. Fourier transform infrared spectroscopy demonstrated the successful incorporation of the material used to produce the produced nanofibrous scaffolds. Thermal studies with thermogravimetric analysis and differential scanning calorimetry indicated that the DLS had high thermal stability. The DLS also showed good in vitro characteristics in terms of improved swelling ratio and contact angle. The mechanical results revealed that the DLS had an improved tensile strength of 3.88 MPa compared with the second layer of scaffold (2.81 MPa). The release of drugs from the scaffold showed different profiles for the two drugs. Lidocaine hydrochlo ride exhibited an initial burst release (66% release within an hour); however, mupirocin exhibited only a 5% release. Furthermore, the DLS nanofibers displayed highly effective antibacterial activities against Staphylococcus aureus, Escherichia coli , and Pseudomonas aeruginosa and were nontoxic to fibroblasts. Conclusion The fabricated DLS exhibited excellent hydrophilicity, cytocompatibility, sustained drug release, and antibacterial activity, which are favorable qualities for its use as a multifunctional material for wound dressing applications.

Although blending brittle polylactide (PLA) with flexible poly(ε-caprolactone) (PCL) holds promise for developing biodegradable materials with tunable mechanical properties, their poor compatibility poses a significant challenge to effective toughening. In this study, PLA/PCL blends with varying Dibutyl itaconate (DBI) content were prepared via melt blending, and the effects of DBI loading on the properties of the blends were systematically investigated. The results demonstrated that the incorporation of DBI enhanced the intermolecular interactions between PLA and PCL chains, thereby improving their compatibility. The phase interface between the PLA matrix and the PCL dispersed phase became so progressively blurred with increasing DBI content that it was nearly indistinguishable at a loading of 7 phr, demonstrating a significant improvement in compatibility. With the increase in DBI content, both the elongation at break and impact strength of the blends exhibited a trend of initial increase followed by a decrease. At the optimal loading, the maximum values reached 314.3% and 29.5 kJ/m 2 , representing enhancements of 3.4-fold and 4.9-fold, respectively, compared to those of the neat PLA/PCL blend. Simultaneously, the crystallization rate and enzymatic degradation rate of the blends increased continuously with higher DBI content. This study opens a new avenue for developing high-toughness PLA-based blend materials without compromising their biodegradability. Summary DBI enhanced both elongation at break and impact strength of PLA/PCL blends. The incorporation of DBI improved the compatibility between PLA and PCL. The crystallization capability of the modified blends was enhanced. The modified blends showed an increased degradation rate.

For enhancing the storability and quality of postharvest fresh products, an electrospun nanofiber membrane composed of polylactic acid (PLA), polycaprolactone (PCL), perilla essential oil (PEO) and zeolitic imidazolate framework (ZIF-8) (ZIF-8@PEO/PLA/PCL) was developed for active food packaging. The ZIF-8@PEO/PLA/PCL electrospun fibers integration were continuous nanofibers with ZIF-8 nanoparticles affixed to their surface and 759.1 nm average diameter. The ZIF-8@PEO/PLA/PCL nanofiber membranes had improved tensile strength of 14.35 MPa and anti-deformation capability of 9.33 MPa and reduced thermal stability compared with PEO/PLA/PCL. The adhesion of ZIF-8 nanoparticles maintained the nanofiber surface hydrophobicity and improved the water vapor barrier quality of 0.21 g mm/m 2 h. The ZIF-8@PEO/PLA/PCL nanofiber membrane showed excellent antibacterial ability within 24 h test and biosafety confirmed by the zebrafish co-culture experiments. The storage test revealed that the ZIF-8@PEO/PLA/PCL membrane delayed the postharvest quality degradation of blueberry fruits to extend their shelf-life to 28 days by maintaining firmness, color, and nutritional quality. These results indicated that the nanofiber membrane has significant potential for fruit preservation as functional packaging material.

Bypass vascular grafting is an effective treatment for cardiovascular disease. Currently, small-diameter vascular grafts (SDVGs, inner diameter <6 mm) for peripheral revascularization, especially for lower limb revascularization, are in high demand. Previously, we prepared and investigated PLGA@PCL SDVGs with core/shell fibers. In this study, PLGA@PCL/elastin core/shell fibers (PCEs) SDVGs were further prepared by electrospinning, so as to enhance the vascular regeneration performance through the introduction of exogenous soluble elastin. The cross-linking effects of elastin with different cross-linking reagents on PCEs SDVGs were subsequently explored. Among them, PCEs SDVGs cross-linked with 1-ethyl-3-(3-(dimethylamino)propyl) carbodiimide (EDCI) and N-hydroxysuccinimide (NHS) retained the most soluble elastin content and achieved a compliance of 1.74 ± 0.62%/mmHg × 10–2, a bursting pressure of 1750 mmHg, and a tensile strength of 2.09 ± 0.51 MPa, which are mostly close to those of human saphenous vein. Subsequently, in vivo transplantation in the abdominal aorta showed that all implanted SDVGs cross-linked by EDC/NHS (PCEsEN) remained patent without aneurysm formation. Histological staining results further showed that the introduction of elastin not only promoted the degradation of SDVGs but also accelerated the formation of neointimal tissue and suppressed the calcification. Thus, PCEsEN shows promising potential to be translated into SDVGs in clinic.

Heparin-like polymers have a good anticoagulant effect due to some groups similar to heparin. Herein, sulfonated keratin(SK) was prepared by chain transfer radical polymerization of 3-sulfopropyl methacrylate(SPMA) to improve blood coagulation nature of keratin. Poly(ε-caprolactone)(PCL)/SK nanofibrous mats with the ratio of 7 and 3 were then fabricated by electrospinning. In vitro cytotoxicity and blood compatibility tests were performed to assess the biocompatibility. Viability of NIH 3T3 cells on PCL/SK mats was higher than that on the pristine PCL mats, indicating their good cytocompatibility. These sulfonated keratin-containing mats enhanced endothelial cell growth, while inhibited smooth muscle cell proliferation and reduced platelet adhesion in the presence of GSH and GSNO, as a result of NO generation. Furthermore, the biocomposite mats prolonged the activated partial thromboplastin time(APTT) effectively without hemolysis. Taken together, PCL/SK mats are potential for applications in vascular tissue engineering.

Hydrogel is widely used as a scaffolding material for tissue engineering due to its excellent cytocompatibility and potential for biofunctionalization. However, its poor mechanical property limits its further application. Fabrication of fiber-reinforced hydrogel composite scaffolds has emerged as a solution to overcome this problem. However, existing strategies usually produce nonporous composite scaffolds, where the interfiber pores are completely filled with hydrogel. This design can hinder oxygen and nutrient exchange between seeded cells and the culture medium, thereby limiting cell invasion and colonization within the scaffold. In this study, sodium alginate (SA) hydrogel was exclusively grafted onto the surface of the constituent fibers of the melt electrowritten scaffold while preserving the porous structure. The grafted hydrogel amount and pore size were precisely controlled by adjusting the SA concentration and the crosslinking ratio (SA: CaCl2). Experimental results demonstrated that the porous composite scaffolds exhibited superior swelling capacity, degradation ratio, mechanical properties, and biocompatibility. Notably, at an SA concentration of 0.5% and a crosslinking ratio of 2:1, the porous composite scaffold achieved optimal cell adhesion and viability. This study highlights the critical importance of preserving porous structures in composite scaffolds for tissue-engineering applications.

Micro-nano bioactive glass (MNBG) has bone regenerative potential. However, the difficulty of MNBG molding restricts its clinical applications in bone regeneration. Exosomes carry proteins, lipids, and nucleic acids for communication between cells. In this study, we adhered human exfoliated deciduous teeth (SHED)-derived exosomes (SHED-Exos) to electrospun micro-nano bioactive glass/polycaprolactone (MNBG/PCL) membrane to control inflammation and promote bone regeneration. MNBG/PCL membrane showed biocompatibility and osteogenic ability. Next, we demonstrated that SHED-Exos internalized into macrophages attenuated LPS-induced expression of M1-macrophage markers iNOS, IL-6, and IL-1β and upregulated M2-macrophage marker IL-10 expression, indicating a switch from M1 to M2 macrophage phenotype. MNBG/PCL-loaded SHED-Exos membrane supported the survival and growth of mouse bone marrow stromal cells (mBMSCs). When M1 macrophages were co-cultured with mBMSCs on the membrane, the SHED-Exos-loaded membrane showed higher Runx2, Alp, and Ocn gene expression. Our findings indicate that SHED-Exos-functionalized MNBG/PCL membrane has anti-inflammatory and osteoinductive potential, suggesting its potential as a candidate material for bone regeneration.

Commercially available guided bone regeneration (GBR) membranes often exhibit limited mechanical properties or bioactivity, leading to poor performance in repairing bone defects. To surmount this limitation, we developed a Janus structural composite membrane (Mg-MgO/PCL) reinforced by dual Mg (Mg sheets and MgO NPs) by using a combined processing technique involving casting and electrospinning. Results showed that the addition of Mg sheets and MgO NPs enhanced the mechanical properties of the composite membrane for osteogenic space maintenance, specifically tensile strength (from 10.2 ± 1.2 to 50.3 ± 4.5 MPa) and compression force (from 0 to 0.94 ± 0.09 N mm–1), through Mg sheet reinforcement and improved crystallization. The dense cast side of the Janus structure membrane displayed better fibroblast barrier capacity than a single fiber structure; meanwhile, the PCL matrix protected the Mg sheet from severe corrosion due to predeformation. The porous microfibers side supported preosteoblast cell adhesion, enhanced osteogenesis, and angiogenesis in vitro, through the biomimetic extracellular matrix and sustainable Mg2+ release. Furthermore, the Mg-MgO/PCL membrane incorporating 2 wt % MgO NPs exhibited remarkable antimicrobial properties, inducing over 88.75% apoptosis in Staphylococcus aureus. An in vivo experiment using the rat skull defect model (Φ = 5 mm) confirmed that the Mg-MgO/PCL membrane significantly improved new bone formation postsurgery. Collectively, our investigation provides valuable insights into the design of multifunctional membranes for clinical oral GBR application.

Introduction: Drugs and biocompatible nanoparticles have raised significant potential in advancing the bone regeneration. Electrospinning technology enables the full realization of the value of drugs and nanoparticles. Methods: In this study, we have successfully fabricated core–sheath nanofibers solely composed of polycaprolactone (PCL) polymer. Simvastatin (SIM) was confined to the core of the nanofibers while nanohydroxyapatite (nHA) was loaded on the nanofiber surface. Results: All the prepared nanofibers exhibited a cylindrical micromorphology, and the core–sheath structure was exploited using a Transmission Electron Microscope. X-ray pattern results indicated that SIM was in an amorphous state within nanofibers, while Fourier Transform InfraRed spectroscopy showed excellent chemical compatibility among SIM, nHA, and PCL. The actual loading of nHA within the nanofiber was determined by a thermogravimetric test due to the high melting point of nHA. Core–sheath nanofibers could release SIM for 672 h, which was attributed to the core–sheath structure. Furthermore, nanofibers loaded with SIM or nHA had a positive impact on cell proliferation, with the core–sheath nanofibers displaying the most favorable cell proliferation behavior. Discussion: Such a synergistic facilitation strategy based on materials and nanostructure may encourage researchers to exploit new biomedical materials in future.
