Electrohydrodynamic jet 3D printing of PCL/PVP composite scaffold for cell culture

Kai Li,Dazhi Wang,Kuipeng Zhao,Kedong Song,Junsheng Liang

Controlled printing of biodegradable and bioresorbable polymers at desired 3D scaffold is of great importance for cell growth and tissue regeneration. In this work, a novel electrohydrodynamic jet 3D printing technology with the resultant effect of electrohydrodynamic force and thermal convection was developed, and its feasibility to fabricate controllable filament composite scaffolds was verified. This method introduces an effective thermal field under the needle to simultaneously enhance the ink viscosity, jetting morphology controllability and printing structure solidify. The fabrication mechanisms of thermal convection on jetting morphology and printed structures feature were investigated through theoretical analysis and experimental characterization. Under optimized conditions, a stable and finer jet was formed; then with the use of this jet, various 3D structures were directly printed at a high aspect ratio ~30. Furthermore, the PCL/PVP composite scaffolds with the controllable filament diameter (~10 μm) which is closed to living cells were printed. Cell culture experiments showed that the printed scaffolds had excellent cell biocompatibility and facilitated cellular proliferation in vitro. It is a great potential that the developed electrohydrodynamic jet 3D printing technology might provide a novel approach to directly print composite synthetic biopolymers into flexibly scale structures for tissue engineering applications.

Functionalization of PCL-based nanofibers loaded with hirudin as blood contact materials

Zhiwen Zheng,Xin Dai,Xueyang Li,Chang Du

Blood-contacting materials with good mechanical property, excellent anticoagulant function and promoting effect on endothelialization are in great demand for clinical application such as vascular grafts in treating cardiovascular diseases. In this study, electrospinning nanofiber scaffolds of polycaprolactone (PCL) were functionalized by oxidative self-polymerization of dopamine (PDA) on the surface followed by the modification of anticoagulant recombinant hirudin (rH) molecules. The morphology, structure, mechanical property, degradation behavior, cellular compatibility and blood compatibility of the multifunctional PCL/PDA/rH nanofiber scaffolds were evaluated. The diameter of the nanofibers was between 270–1030 nm. The ultimate tensile strength of the scaffolds was around 4 MPa and the elastic modulus increased with the amount of rH. The degradation tests in vitro indicated that the nanofiber scaffolds began to crack on the 7th day, but still maintained the nanoscale architecture within a month. The cumulative release of rH from the nanofiber scaffold was up to 95.9 % at 30th day. The functionalized scaffolds promoted the adhesion and proliferation of endothelial cells, while resisting platelet adhesion and enhancing anticoagulation effects. The hemolysis ratios of all scaffolds were <2 %. The nanofiber scaffolds are promising candidates for vascular tissue engineering.

Multimaterial Printing of Serpentine Microarchitectures with Synergistic Mechanical/Piezoelectric Stimulation for Enhanced Cardiac-Specific Functional Regeneration

Kang Han,Mao Mao,Liyan Fu,Yabo Zhang,Yuming Kang,Dichen Li,Jiankang He

Recreating the natural heart's mechanical and electrical environment is crucial for engineering functional cardiac tissue and repairing infarcted myocardium in vivo. In this study, multimaterial-printed serpentine microarchitectures are presented with synergistic mechanical/piezoelectric stimulation, incorporating polycaprolactone (PCL) microfibers for mechanical support, polyvinylidene fluoride (PVDF) microfibers for piezoelectric stimulation, and magnetic PCL/Fe 3 O 4 for controlled deformation via an external magnet. Rat cardiomyocytes in piezoelectric constructs, subjected to dynamic mechanical stimulation, exhibit advanced maturation, featuring superior sarcomeric structures, improved calcium transients, and upregulated maturation genes compared to non-piezoelectric constructs. Furthermore, these engineered piezoelectric cardiac constructs demonstrate significant structural and functional repair of infarcted myocardium, as evidenced by enhanced ejection and shortening fraction, reduced fibrosis and inflammation, and increased angiogenesis. The findings underscore the therapeutic potential of piezoelectric cardiac constructs for myocardial infarction therapy.

Amphiphilic poly(methacryloxyethyl sulfobetaine)-block-poly(ε-caprolactone) blended polyvinyl chloride plastics: Study on the Relationship between molecular structure and antibiofouling properties

Rong-Chao Xue,Nan Hu,Rong-Xu Zhao,Ze-Zhong Lin,Li-Ping Yu,Bin Wang

The blending of amphiphilic copolymers with plastics is an important method for preparing antibiofouling medical plastics. However, enhancing the enrichment of amphiphilic copolymers on the plastic surface, thereby improving the antibiofouling performance of the plastics remain challenging tasks. Herein, we designed and synthesized a series of amphphilic copolymers, denoted as PSB- b -nPCL. These copolymers possess similar chemical compositions but exhibit distinct topological structures. Antibiofouling flexible polyvinyl chloride (PVC) films were prepared by blending PSB- b -nPCL with PVC. These films showed excellent mechanical and thermal properties. X-ray photoelectron spectroscopy, water contact angle and hydration capacity tests demonstrated that PSB- b -nPCL which bearing more PCL blocks were more prone to migrating to the film surface, thereby enhancing the film’s hydrophilicity. Furthermore, anti-protein adsorption, anti-bacterial and anti-platelet adhesion properties of the films were investigated. It was found that topological structure can obviously affect the antibiofouling performance of films. Specifically, films blended with PSB- b -nPCL containing more PCL blocks exhibited superior performance. The optimal blended film achieved a reduction of 96.4% in bovine albumin adsorption, 82% in platelet adhesion, and nearly complete inhibition of bacterial adhesion. We anticipate that the strategy presented in this work will pave a new path for the development of high performance antibiofouling plastics.

Metabolomic and Proteomic Analyses Unveil That Polyethylene Glycol–Polycaprolactone-Loaded Curcumin Nanoparticles Induce Mitochondrial Dysfunction and Metabolic Reprogramming to Suppress Melanoma Growth

Yixun Zhang,Xin Zhou,Wenlian Zheng,Jianwei Chen,Qianfeng Xu,Zhenguo Liang,Hongli Zhang,Jundong Lin,Wenjie Xie,Yangjia Zhuo,Muqi Chen,Fen Zou,Biyan Wen,Zeheng Tan,Weide Zhong,Huikang Yang,Ronghua Yang

Melanoma is highly dependent on cellular metabolism, but current treatments show limited effectiveness. Despite the promising antitumor properties of curcumin, its clinical utility is limited by its poor bioavailability and potential systemic toxicity. To address these challenges, we formulated curcumin-loaded polyethylene glycol–polycaprolactone (PEG–PCL) nanoparticles, denoted as PEG–PCL@Cur. We employed omics technologies to investigate their impact on melanoma at the molecular level. In vitro studies revealed that PEG–PCL@Cur notably suppressed the growth, movement, and invasiveness of A375 melanoma cells in a dose-dependent manner. Untargeted metabolomics identified 347 metabolites that were significantly altered. A decrease in fumaric acid levels indicated the suppression of oxidative phosphorylation and a shift in metabolism. Proteomics further confirmed the downregulation of key mitochondrial proteins, such as SDHA, pointing to mitochondrial dysfunction. Mechanistically, PEG–PCL@Cur increased reactive oxygen species (ROS) production by raising NADH levels and inhibiting SDHA expression. This led to a reduction in the level of fumaric acid production and a disruption of the tricarboxylic acid (TCA) cycle. Additionally, PEG–PCL@Cur hindered the generation of inorganic phosphate (Pi), resulting in a decreased level of ATP production. These metabolic changes promoted apoptosis and enhanced melanoma cell death. In vivo, PEG–PCL@Cur significantly curtailed tumor growth in a melanoma xenograft model and displayed an enhanced biocompatibility. Histological analysis showed extensive tumor necrosis, diminished proliferation markers, and elevated levels of apoptosis indicators. Overall, our integrated metabolomic and proteomic approach highlights how PEG–PCL@Cur targets SDHA-mediated metabolic reprogramming and mitochondrial dysfunction. This discovery positions PEG–PCL@Cur as a promising nanoplatform for melanoma therapy and offers valuable insights into the development of targeted nanomedicines.

Adsorption isotherm, kinetics simulation and breakthrough analysis of 5-hydroxymethylfurfural adsorption/desorption behavior of a novel polar-modified post-cross-linked poly (divinylbenzene-co-ethyleneglycoldimethacrylate) resin

Jiayi Zheng,Xianda He,Chiliu Cai,Jiangxiong Xiao,Yao Liu,Zhe Chen,Baoying Pan,Xiaoqing Lin

A polar modified post-cross-linked poly (divinylbenzene- co -ethyleneglycol-dimethacrylate) (PCL-PDE) resin was synthesized by suspension polymerization of ethylene glycol dimethacrylate (EGDMA) and divinylbenzene (DVB), and a post-cross-linked reaction. After characterization, the adsorption behaviors of 5-hydroxymethylfurfural (5-HMF) on PCL-PDE resin were determined in comparison with the starting copolymers PDE resin. The equilibrium adsorption capacity of 5-HMF on PCL-PDE resin was much larger than PDE resin and the increase rate was greater than 52.6%. The equilibrium data of 5-HMF onto PCL-PDE resin were found to be better fitted by the Langmuir isotherm model. The kinetic data shows that the adsorption reached equilibrium in a short time (less than 20 min) can be fitted by the pore diffusion model (PDM) at various operating conditions. The effective pore diffusion coefficient was dependent upon adsorption temperature, and were 6.706 × 10 −10 , 8.958 × 10 −10 , 1.136 × 10 −9 and 1.429 × 10 −9  m 2  s −1 at 288, 298, 308 and 318 K, respectively. Furthermore, the effects of feed flow rate ( Q f  = 0.6, 1.5, 3.0 and 6.0 mL min −1 ) and initial 5-HMF concentration ( c f  = 0.52, 1.02, 2.00 and 4.96 g L −1 ) on the adsorption were investigated systematically. Besides, a general rate model (GRM) was used to predict adsorption breakthrough curves of 5-HMF. The simulation results are highly consistent with the experimental data, indicating that the GRM can successfully simulate this process. In the desorption process, the desorption capacity reaches 99.6% of adsorbed capacity, suggesting that the PCL-PDE resin exhibited good reusability. Therefore, it could be suggested that the PCL-PDE resin has a potential application in the separation and purification of 5-HMF.

Controlled-release of indomethacin trigged by inflammation-response for wound care

Yanping Huang,Ye Dan,Nianhua Dan,Yining Chen

An inflammatory-responsive intelligent controlled-release drug-loaded membrane (polycaprolactone/poly (hydroxyethyl methacrylate hydroxyethyl-methacrylate -2-(2-(1-(4-chlorobenzoyl)-5-methoxy-2-methyl-1H-indol -3-yl) acetoxy) ethyl meth acylate) PCL/P(HEMA-HEIn) was prepared. Drug precursors HEIn was synthesized by the esterification reaction between indomethacin (Indo) and hydroxyethyl methacrylate (HEMA) and the structure of HEIn was confirmed by 13 C NMR and ESI-MS. Through in-situ crosslinking polymerization and solvent evaporation, the drug-loaded membranes were fabricated. Furthermore, the morphology, chemical and physical properties demonstrated that HEIn and HEMA were copolymerized and well dispersed in the film. The drug released from PCL/P(HEME-HEIn) could be triggered by inflammation environment and the release amount increased along with the degree of inflammation. Results of in vivo evaluation proved that the PCL/P(HEME-HEIn) film had the capacity of inhibiting inflammation, which was beneficial to wound healing. The proposed strategy towards PCL/P(HEME-HEIn) opened new window for drug controlled-releasing via inflammatory response.

Fabrication and characterization of electrospinning/3D printing bone tissue engineering scaffold

Yinxian Yu,Sha Hua,Mengkai Yang,Zeze Fu,Songsong Teng,Kerun Niu,Qinghua Zhao,Chengqing Yi

Electrospinning and three-dimensional (3D) printing is widely used to fabricate bone tissue engineering scaffolds. However, cells are difficult to infiltrate into the nanoporous structure of traditional electrospun scaffolds by electrospinning, and also 3D printing techniques have the disadvantage of low print resolution. In order to solve these problems, we fabricated a 3D composite scaffold by infusing PCL/gelatin dispersed nanofibers into the meshes of PCL printing scaffold. The morphology of the composite scaffold was evaluated by a scanning electron microscope (SEM), which shows the micro-scale (100–300 μm) porous structure. The porosity of the composite scaffold is as high as 79.32 ± 8.32%. Mechanical testing results indicated that the compressive modulus of the 3D composite scaffold (30.50 ± 0.82 MPa) is significantly higher than that of the lyophilized electrospun scaffold (18.55 ± 0.56 MPa), which is attributed to the 3D printing scaffold. An in vitro study indicated that the 3D composite scaffolds have good biocompatibility. Through CCK-8 assay and fluoresce staining characterization, MC3T3-E1 cells exhibit a better proliferation and infiltration on the composite scaffold than on the PCL printing scaffold, which is due to the microporous structure of the electrospun scaffold. Altogether, these results confirm the good potential of an electrospun/3D printing composite scaffold applied to bone tissue repair.

Novel Airflow-Field-Driven Melt Spinning 3D Printing of Tubular Scaffolds Based on Polycaprolactone Blends

Junyuan Zhang,Zilong Peng,Mengjie Wang,Yinan Li,Jinyin Wu,Yifan Jiang,Chaolong Liu,Guqiang Li,Lin Xu,Hongbo Lan

The fabrication of various 3D tissue engineering tubular scaffolds with fibrous structures, to assist the human body in rapidly repairing a variety of ailments, is receiving more and more attention. Due to the inefficiency of the majority of fibrous preparation techniques, the question of how to rapidly produce the requisite three-dimensional tubular microfiber scaffold structures has become an urgent problem. In this study, an efficient polymer fiber preparation method was developed, using a high-speed airflow drive. Melt blending of polycaprolactone (PCL), polylactic acid (PLA), and tributyl citrate (TBC), was used for the printing material, to achieve the efficient preparation of tubular microfiber scaffolds with different structures. The scaffold diameter was as small as 2 mm, the wall thickness was up to 100 μm, and the fiber injection efficiency reached 15.48 g/h. By utilizing simulations to optimize the printing parameters and by adjusting the printing settings, it was possible to achieve a controlled fiber diameter in the range of 3 μm to 15 μm. In addition, plasma treatment was applied to the microfibers’ surface, to increase their wettability, and the efficiency of the hydrophilic modification was demonstrated. Furthermore, the mechanical property test demonstrated that the fibers have a tensile strength of 1.36 ± 0.16 MPa and a tensile strain of 30.8 ± 3.5%. The radial compressive strain of the tubular scaffold could reach 60% of the original scaffold’s diameter. Finally, the in vitro degradation of the fibers at various pH values was tested. The results showed that, under alkaline conditions, the surface of the fibers would be severely crushed and the rate of deterioration would increase.Keywords:airflow-field-driven;3D printing;microfiber scaffold;polycaprolactone (PCL);polylactic acid (PLA)

A facile strategy for fabricating PCL/PEG block copolymer with excellent enzymatic degradation

Haipeng Wang,Dejin Tong,Lei Wang,Lei Chen,Na Yu,Zhanxiong Li

Poly(ω-caprolactone) (PCL) has been the focus in many research initiatives, especially regarding various ways though which to introduce reactive sites to the polyester chains. In this work, hydroxyl groups were covalently introduced into the chain of PCL by reacting ester groups with 6-amino-1-hexanol. Poly(ethylene glycol) (PEG) was crosslinked to the PCL chain through esterification , by using N,N’-carbonyldiimidazole (CDI) as catalyst. The prepared PCL-PEG copolymers were characterized by various analytical techniques such as Fourier transform infrared spectroscopy (FT-IR), 1 H NMR and X-ray photoelectron spectroscopy (XPS). Static water contact angles measurement indicated that hydrophilicity of the copolymer films modified by PEG has improved considerably. The copolymer films were prepared by casting the polymer solution onto a stainless plate. The degradation of the copolymer samples, in a phosphate buffer solution containing a type of Lipase (Aspergillus Oryzae), was observed at different time intervals. The results of weight loss ratio, during the degradation process, indicated that blocking PEG chains onto the PCL chains can effectively accelerate the process of enzymatic degradation of the resulting copolymer. The changes in molecular weight of the copolymer films were investigated by Gel Permeation Chromatography (GPC) and the copolymers were characterized by 1 H NMR during degradation. Surface morphology of PCL and PCL-PEG polymers, before and after degradation, was observed by Scanning electron microscopy (SEM), and emphasized that the enzymatic degradation of PCL-PEG copolymers occurred faster by comparison with PCL, starting from the surface layer of the films.

Preparation of pCL-20/PTFE-Al composites for sensibility reduction and combustion improvement

Lanting Yang,Zhaokang Chen,Lihao Wang,Zhenwei Zhang,Ruihao Wang,Changping Guo

The high sensitivity of high explosive hexanitrohexaazaisowurtzitane (CL-20) limits its applicability, and the performance optimization brought by the synergistic effect between nanothermite and CL-20 is expected to expand its application prospects. In this study, polydopamine (PDA) was introduced to modify CL-20 (denoted as pCL-20), and a quasi-core/shell pCL-20/polytetrafluoroethylene-aluminum (pCL-20/PTFE-Al) composite was successfully prepared by ultrasonic-assisted composite method. The performance characterization results demonstrate that the composite exhibits significantly better decomposition heat release than the mixed sample, a reduction in combustion time from 408 ms to 268 ms, a more stable combustion flame, and more focused energy release. At the same time, the comparison of composite combustion with different condition variables reflects the combustion advantages of the selected proportional conditions in this study. The characteristic drop height (H 50 ) increased from 24.05 cm to 42.15 cm, the explosive percentage of friction sensitivity reduced from 100 % to 70 %, and the sensitivity of the composite sample decreased significantly when compared to the raw CL-20. It improves the combustion performance while reducing the sensitivity, thereby enhancing the application potential and providing a reference for the high-energy desensitization of energetic materials.

Preparation and characterization of PCL@β-TCP composite antibacterial scaffolds by additive manufacturing

Chen Zhang,Yi Qi,Binying Dai,Runyi Lin,Jinchao You,Yaomei Wang,Shunxue Xing,Yuansong Ye,De-Jing Li

Poly(ε-caprolactone) (PCL) and β-tricalcium phosphate (β-TCP) are promising materials for bone tissue engineering due to their complementary biodegradability and osteoconductivity. However, their individual limitations hinder clinical application. In this study, PCL@β-TCP composite scaffolds containing 5%, 10%, and 20% β-TCP were fabricated via fused deposition modeling (FDM). The effects of β-TCP content on the physicochemical, mechanical, and antibacterial properties of the scaffolds were systematically investigated. Results showed that increasing β-TCP content enhanced hydrophilicity (contact angle reduced from 89.1° to 66.9°), accelerated biodegradation (higher weight loss after 103 days in PBS), and improved antibacterial activity, as evidenced by larger inhibition zones against E. coli and S. aureus. These findings demonstrate that β-TCP incorporation effectively enhances the bioactivity and antibacterial performance of PCL scaffolds, highlighting their potential for bone tissue engineering applications, particularly in non-load-bearing contexts.
