Biodegradable PLLA-based cast films were prepared via melt blending poly(L-lactic acid) (PLLA) with poly(L-lactic acid-co-butylene itaconate) (PLBI), poly(L-lactic acid-co-glycolic acid) (PLGA), and poly(L-lactic acid-co-butylene itaconate-co-glycolic acid) (PLBIGA) copolymers to enhance mechanical, barrier, and degradation properties for dairy packaging applications. Copolymers were synthesized using a one-step melt polycondensation method, and the film properties were effectively controlled by adjusting the blending ratios. All blended films exhibited significantly improved toughness while maintaining high transparency. Specifically, PLLA/PLBIGA20 % film demonstrated a 90.1 % reduction in oxygen transmission rate (OTR) and a 54.3 % reduction in water vapor transmission rate (WVTR) compared to pure PLLA, and PLLA/PLBI20 % film reduced ultraviolet (UV) transmittance to 53 % in the 280–320 nm range. The soil degradation rates of blended films were notably higher than pure PLLA, with distinct degradation mechanisms observed for each copolymer; notably, PLLA/PLBIGA20 % film degraded simultaneously in both amorphous and crystalline regions. Blended films met food-contact safety standards and, when applied to commercial yogurt packaging, effectively preserved lactic acid bacteria viability, outperforming pure PLLA film. These findings demonstrate the strong potential of PLLA-based blended films as sustainable packaging solutions for dairy products.

Effects of montmorillonite (MMT) on the crystallization behavior of poly(L-lactic acid) (PLLA) were investigated by variable-temperature FTIR spectroscopy. The variations of carbonyl band (1800–1720 cm −1 ) of different PLLA/MMT nanocomposites were focused due to its strong intensity and the involved abundant structure information. Difference spectrometry was used to evaluate the structural variations of PLLA after introducing MMT, and perturbation correlation moving window two-dimensional analysis (PCMW2D) split the temperature range into two sub-regions, i.e., 32–116 ℃ and 116–152 ℃, on the basis of the spectral variation. Two-dimensional correlation spectroscopy (2DCOS) was further applied to such sub-regions in order to find the change order between varied PLLA polymorphs. The results showed that less addition of MMT (≤3%) would lead to a well-exfoliated structure, which not only had no nucleation effect for PLLA, but also delayed the cold crystallization to a higher temperature compared with the one of pure PLLA. However, a higher addition of MMT (≥5%) would lead to an intercalated structure, which acted as a nucleating agent and thus advanced the cold crystallization to a lower temperature. Nevertheless, the introduction of MMT cannot affect the phase transition order between the amorphous, the intermediate, the α′- and the α′-PLLAs based on 2DCOS results.

Poly(l-lactic acid) (PLLA) featuring desirable biodegradability and biocompatibility has been recognized as one of the promising eco-friendly biomaterials. However, low crystallization and poor mechanical and chemical performances dramatically hamper its practical application. In this work, we report that functionalized cellulose/PLLA composite superhydrophobic stereocomplex films with controllable water adhesion and protein adsorption can be fabricated by a facile approach for the first time. First, cellulose is surface-modified by means of two silanization modification methods. Then, superhydrophobic cellulose/PLLA composite films are prepared through a solvent-evaporation-induced phase separation method. The two cellulose/PLLA composite films exhibit extreme water repellency but tunable water adhesion from sticky to slippery. The protein adsorption capacity of the cellulose/PLLA composite films can also be regulated. In addition, the stereocomplexation of the composite film provides excellent mechanical properties with an elongation at break of 22.36%, which is 237.8% higher than that of a pure PLLA film, which is more suitable for biomaterials. Furthermore, good biodegradability of the PLLA composite films in nature enables the bio-based composites as alternative materials to replace conventional petroleum-based polymers. The superhydrophobic films have also been demonstrated for many applications, including slippery surfaces, liquid transportation without loss, and antifouling.

Orderly hierarchical structure with balanced mechanical, chemical, and electrical properties is the basis of the natural bone microenvironment. Inspired by nature, we developed a piezocatalytically-induced controlled mineralization strategy using piezoelectric polymer poly-L-lactic acid (PLLA) fibers with ordered micro-nano structures to prepare biomimetic tissue engineering scaffolds with a bone-like microenvironment (pcm-PLLA), in which PLLA-mediated piezoelectric catalysis promoted the in-situ polymerization of dopamine and subsequently regulated the controllable growth of hydroxyapatite crystals on the fiber surface. PLLA fibers, as analogs of mineralized collagen fibers, were arranged in an oriented manner, and ultimately formed a bone-like interconnected pore structure; in addition, they also provided bone-like piezoelectric properties. The uniformly sized HA nanocrystals formed by controlled mineralization provided a bone-like mechanical strength and chemical environment. The pcm-PLLA scaffold could rapidly recruit endogenous stem cells, and promote their osteogenic differentiation by activating cell membrane calcium channels and PI3K signaling pathways through ultrasound-responsive piezoelectric signals. In addition, the scaffold also provided a suitable microenvironment to promote macrophage M2 polarization and angiogenesis, thereby enhancing bone regeneration in skull defects of rats. The proposed piezocatalytically-induced controllable mineralization strategy provides a new idea for the development of tissue engineering scaffolds that can be implemented for multimodal physical stimulation therapy.

Artesunate, as a semi-synthetic artemisinin derivative, exhibits a broad-spectrum bactericidal effect via the release of singlet oxygen. However, its release is slow due to the difficulty breaking the endoperoxide bridge bond. Ammoniated graphene oxide could be electrostatic adsorbed with artesunate and efficiently transfer thermal energy to artesunate by interfacial heat transfer, thereby facilitating the breaking of the endoperoxide bridge bond. In detail, artesunate was anchored on graphene oxide by a one-pot hydrothermal synthesis method and introduced to the PLLA scaffold fabricated by selective laser sintering. Under near-infrared light irradiation, graphene oxide could convert light to heat through photothermal conversion. Then the heat energy was transferred to artesunate by interfacial heat transfer, accelerating the release of singlet oxygen. Meanwhile, graphene oxide could achieve the purpose of thermal ablation for bacterial infection by increasing the local temperature. Based on the combination of singlet oxygen and thermal ablation therapy, the scaffold possesses a high bactericidal ability triggered by near-infrared light. Under the near-infrared light irradiation, the singlet oxygen production from artesunate increased by 39 %, and the antibacterial ratio against S. aureus and E. coli reached 91.6 % and 93.3 %, respectively. Additionally, the scaffold was favorable cytocompatibility with mBMSCs. All the results highlight the great potential of artesunate-mediated graphene oxide scaffold in eliminating bacterial infections.

Poor tendon repair is often a clinical challenge due to the lack of ideal biomaterials. Electrospun aligned fibers, resembling the ultrastructure of tendon, have been previously reported to promote tenogenesis. However, the underlying mechanism is unclear and the aligned fibers alone are not capable enough to commit teno-differentiation of stem cells. Here, based on our observation of reduced expression of histone deacetylases (HDACs) in tendon stem/progenitor cells (TSPCs) cultured on aligned fibers, we proposed a strategy to enhance the tenogenesis effect of aligned fibers by using a small molecule Trichostatin A (TSA), an HDAC inhibitor. Such a TSA-laden poly ( l -lactic acid) (PLLA) aligned fiber (A-TSA) scaffold was successfully fabricated by a stable jet electrospinning method, and demonstrated its sustained capability in releasing TSA. We found that TSA incorporated aligned fibers of PLLA had an additive effect in directing tenogenic differentiation. Moreover, the in situ implantation study in rat model further confirmed that A-TSA scaffold promoted the structural and mechanical properties of the regenerated Achilles tendon. This study demonstrated that HDAC was involved in the teno-differentiation with aligned fiber topography, and the combination of HDAC with aligned topography might be a more efficient strategy to promote tenogenesis of stem cells. Statement of Significance Electrospun aligned fibers, resembling the ultrastructure of tendon, have been previously reported to promote tenogenesis. However, the underlying mechanism is unclear and the aligned fibers alone are not capable enough to commit teno-differentiation of stem cells. The uniqueness of our studies are as follows, based on our observation of reduced expression of histone deacetylases (HDACs) in tendon stem/progenitor cells (TSPCs) cultured on aligned fibers, we proposed a strategy to enhance the tenogenesis effect of aligned fibers by using a small molecule Trichostatin A (TSA), a HDAC inhibitor. Such a TSA-laden poly ( l -lactic acid) (PLLA) aligned fiber (A-TSA) scaffold was successfully fabricated by a stable jet electrospinning method, and demonstrated its sustained capability in releasing TSA. The incorporation and subsequent release of bioactive small molecule TSA into electrospun aligned fibers allows a controllable manner for both biochemical and physical regulation of tenogenesis of stem cells both in vitro and in vivo . Collectively, the present study provides a model of “translating the biological knowledge learned from cell-material interaction into optimizing biomaterials (from Biomat-to-Biomat)”.

The inability of articular cartilage to self-repair following injuries frequently precipitates osteoarthritis, profoundly affecting patients' quality of life. Given the limitations inherent in current clinical interventions, an urgent need exists for more effective cartilage regeneration methodologies. Previous studies have underscored the potential of electrical stimulation in cartilage repair, thus motivating the investigation of innovative strategies. The present study introduces a three-dimensional scaffold fabricated through a composite technique that leverages the synergy between piezoelectricity and biofactors to enhance cartilage repair. This scaffold is composed of polylactic acid (PLLA) and barium titanate (BT) for piezoelectric stimulation and at the bottom with a collagen-coated layer infused with fibroblast growth factor-18 (FGF-18) for biofactor delivery. Designed to emulate the properties of natural cartilage, the scaffold enables controlled generation of piezoelectric charges and the sustained release of biofactors. In vitro tests confirm that the scaffold promotes chondrocyte proliferation, matrix hyperplasia, cellular migration, and the expression of genes associated with cartilage formation. Moreover, in vivo studies on rabbits have illustrated its efficacy in catalyzing the in situ regeneration of articular cartilage defects and remodeling the extracellular matrix. This innovative approach offers significant potential for enhancing cartilage repair and holds profound implications for regenerative medicine.

Owing to the unpredictable size of wounds and irregular edges formed by trauma, nanofibers' highly customizable and adherent in situ deposition can contribute to intervention in the healing process. However, electrospinning is limited by the constraints of conventional polymeric materials despite its potential for anti-inflammatory and antimicrobial properties. Here, inspired by the Janus structure and biochemistry of nanometal ions, we developed an in situ sprayed electrospinning method to overcome bacterial infections and immune imbalances during wound healing. The bilayer fiber scaffold has a hydrophobic outer layer composed of polycaprolactone (PCL) and a hydrophilic inner layer composed of gelatin, poly (L-lactic acid) (PLLA), and magnesium oxide nanoparticles (MgO NPs), constituting the PCL/PLLA-Gelatin-MgO (PPGM) electrospun scaffold. This electrospun scaffold blocked the colonization and growth of bacteria and remained stable on the wound for continuous anti-inflammatory properties to promote wound healing. Furthermore, PPGM electrospinning modulated collagen deposition and the inflammatory microenvironment in the full-thickness skin model, significantly accelerating vascularization and epithelialization progression. This personalized Janus electrospun scaffold has excellent potential as a new type of wound dressing for first aid and wound healthcare.

Biodegradable shape-memory polymers (SMPs) which are functional materials with applicability for medicine devices are designed to acquire their therapeutically relevant shape and drug release after implantation. In the work, an amphiphilic polymer (PVAD) is synthesized by using polytetrahydrofuran (PTMG), vinyl acetate (VAc), acrylic acid (AA), tetramethyltetravinylcyclotetrasiloxane (D 4 vi ) as raw materials. PVAD encapsulating hydrophilic drug as switching phase and poly( l -lactide) (PLLA) as fixing matrix construct an SM system with the characteristic of “reservoir-matrix” drug release. The shape recovery ratio ( R r ) of medicated PVAD/PLLA reaches 99 % by heat-water stimulation. The effects of release temperature and SM on drug release are investigated. With the release temperature increasing, the medicated PVAD/PLLA accelerates drug release and shows burst release initially, while the drug release for the medicated PLLA changes slightly. The drug release rate goes up after 3 rounds of SM. The mechanism of SM system controlling drug release is put forward based on structural changes. The yield strength and elongation at break of medicated PVAD/PLLA are 29.8 MPa and 44.6 %, respectively. It opens up new perspectives for drug carrier matrices in Pharmaceutical Sciences.

Tendon injuries are common debilitating musculoskeletal diseases with high treatment expenditure in sports medicine. The development of tendon-biomimetic scaffolds may be promising for improving the unsatisfactory clinical outcomes of traditional therapies. In this study, we combined an advanced electrospun nanofiber yarn-generating technique with a traditional textile manufacturing strategy to fabricate innovative nano-micro fibrous woven scaffolds with tendon-like anisotropic structure and high-strength mechanical properties for the treatment of large-size tendon injury. Electrospun nanofiber yarns made from pure poly L-lactic acid (PLLA) or silk fibroin (SF)/PLLA blend were fabricated, and their mechanical properties matched and even exceeded those of commercial PLLA microfiber yarns. The PLLA or SF/PLLA nanofiber yarns were then employed as weft yarns interlaced with commercial PLLA microfiber yarns as warp yarns to generate two new types of nanofibrous scaffolds (nmPLLA and nmSF/PLLA) with a plain-weaving structure. Woven scaffolds made from pure PLLA microfiber yarns (both weft and warp directions) (mmPLLA) were used as controls. In vitro experiments showed that the nmSF/PLLA woven scaffold with aligned fibrous topography significantly promoted cell adhesion, elongation, proliferation, and phenotypic maintenance of tenocytes compared with mmPLLA and nmPLLA woven scaffolds. Moreover, the nmSF/PLLA woven scaffold exhibited the strongest immunoregulatory functions and effectively modulated macrophages towards the M2 phenotype. In vivo experiments revealed that the nmSF/PLLA woven scaffold notably facilitated Achilles tendon regeneration with improved structure by macroscopic, histological, and ultrastructural observations six months after surgery, compared with the other two groups. More importantly, the regenerated tissue in the nmSF/PLLA group had excellent biomechanical properties comparable to those of the native tendon. Overall, our study provides an innovative biological-free strategy with ready-to-use features, which presents great potential for clinical translation for damaged tendon repair.

The treatment of tenacious diabetic wounds still remains an enormous challenge in clinics, originated from the complicated pathological microenvironment of wound sites. Therefore, it's urgently required to develop one type of innovative dressing patch with appropriate microstructure and multifunctions to regulate the pathological microenvironment and promote the regeneration of diabetic wounds. In this study, novel gelatin (Gel)/poly (L-lactic acid) (PLLA) nanofibrous yarns loading with or without Salvia miltiorrhiza Bunge-Radix Puerariae herbal compound (SRHC) are fabricated by using our modified electrospinning strategy, which are further interlaced into nanofibrous woven fabrics respectively, serving as biofunctional dressing patches for potential diabetic wound treatment application. The actual photographs and SEM images confirm that all the different nanofibrous textiles with or without SRHC exhibit a uniform interwoven structure of warp and weft, and the internal nanofibers present bead-free morphology and uniaxially oriented structure along the longitudinal axis of nanofibrous yarns. Moreover, all the different nanofibrous woven fabrics are demonstrated to possess strong mechanical properties and great surface wettability. The in vitro cell characterization shows that the addition of SRHC can significantly promote the attachment and proliferation of human dermal fibroblasts (HDFs), and also dramatically inhibit the secretion levels of proinflammatory factors of M1 macrophages. The in vivo diabetic mouse full-thickness skin model experiments reveal that the as-developed SRHC-loaded Gel/PLLA nanofibrous textile shows the best performances referring to shorten wound healing time (100 % wound closure after 18 days of treatment) and high-quality regeneration (i.e., enhance collagen deposition, improve re-epithelialization and neovascularization, and increase hair follicles), which assuredly finds great interests serving as an innovative dressing patch for the treatment of hard-to-heal diabetic wounds.

Bone defect regeneration depends on the population and lifespan of M2 macrophages, which are regulated by dual signals generated by the “physical” spatial configuration of biological tissues and “molecular” chemokines. Herein, inspired by the reprogramming of macrophages, immunoengineered porous microspheres are constructed to accelerate bone repair through the regulation of both “physical” and “molecular” signals. The porous structure of injectable poly ( l -lactic acid) (PLLA) microspheres prepared by the microfluidic technique provides a “physical signal” for osteogenic differentiation. Additionally, interleukin (IL)-4-loaded liposomes (Ls) are modified on PLLA microspheres through amide bonds to produce IL-4/Ls/PLLA microspheres, providing a “molecular signal” in stimulating the differentiation of macrophages to M2 type. It is confirmed that IL-4/Ls/PLLA microspheres could induce M2-macrophages polarization and potentiate osteoblast proliferation and differentiation while coculturing with macrophages and osteoblasts in vitro. Besides, IL-4/Ls/PLLA microspheres are proved to promote bone defect regeneration by inducing the conversion of M1 macrophages to M2 through dual biosignal-functional regulation in both the calvaria defect and maxillary sinus defect models. Overall, the immuno-reprogrammed IL-4/Ls/PLLA microspheres achieve the precise immuno-reprogramming of macrophages by dual biosignal-functional regulation. This immune reengineering strategy paves a way for clinical bone defect treatment.
