A ratio luminescence probe was developed for detecting Staphylococcus aureus ( S. aureus ) based on luminescence energy transfer (LET) using double-wavelength emission (550 nm and 812 nm) upconversion nanoparticles (UCNPs) as donor, gold nanoparticles (AuNPs) as acceptor and the aptamer for S. aureus as the specific recognition and link unit. The LET process could cause luminescence quenching because of the spectral overlap between the acceptor and the donor at 550 nm. In the presence of S. aureus , S. aureus selectively combined with the aptamer, and the AuNPs left the surface of UCNPs, which weakened the quenching effect and restored the luminescence of UCNPs. Based on this, the ratio detection was realized by monitoring the change of the luminescence signal of the probe at 550 nm and taking the luminescence signal at 812 nm as the reference signal. Crucially, the probe has a fast reaction speed, with a reaction time of 25 min, and the detection of S. aureus is realized in the concentration range of 5.0 × 10 3 –3.0 × 10 5 CFU/ml, with the detection limit of 106 CFU/ml. Therefore, the ratio probe has great potential for detecting of S. aureus in food because of its high sensitivity, fast speed and good selectivity.

Thymol showed good antimicrobial activity, however, the poor aqueous solubility limits it to apply in food industry. α-Lactalbumin can be used to delivery hydrophobic molecules, then enhancing their biological activities. The study investigates the potential of α-lactalbumin to expand the application range of thymol, further to evaluate the antimicrobial activity of the α-lactalbumin-thymol complexation. Multi-spectroscopy techniques and computational simulations have confirmed the successful complexation, driving mainly by van der Waals force. The α-lactalbumin-thymol complexation exhibited the superior antimicrobial activity than thymol against Escherichia coli and Staphylococcus aureus , as indicated by lower or comparable minimum inhibitory concentration (336 μg/mL and 224 μg/mL, respectively), fewer colony forming units, and larger inhibition zone diameters. Furthermore, α-lactalbumin enhanced the degree of membrane damage by thymol. The complexation preserved no obvious cytotoxicity against HeLa cells. This study indicates that the α-lactalbumin-thymol complexation chelates hold promise as natural antimicrobial agents in the food processing industry.

Methicillin-resistant Staphylococcus aureus (MRSA) poses a challenge for clinical treatment and combining antibiotics with other agents might be a promising strategy to overcome this challenge. This study explored the synergistic antibacterial activity of baicalin (traditional Chinese medicine extract) and the narrow-spectrum beta-lactam antibiotic oxacillin sodium, both of which are poorly active against MRSA in vitro . The combination of baicalin and oxacillin sodium showed a synergistic effect with a fractional inhibitory concentration index of 0.5. Mechanistically, the supplementation of baicalin increased the permeability of bacterial cell walls and cell membranes, enhancing oxacillin sodium entry and bactericidal action. The combination of baicalin and oxacillin sodium also significantly inhibited MRSA USA300 biofilm formation by further reducing polysaccharide intercellular adhesion production. Therefore, the combination of baicalin and oxacillin sodium offers a new therapeutic option for addressing clinical MRSA resistance. Further studies, including clinical trials, will be required to validate the observed in vitro results.

Efforts to curtail the escalating health threat posed by methicillin-resistant Staphylococcus aureus (MRSA), a formidable superbug, necessitate the development of innovative treatment strategies. Leveraging potential compounds from natural sources in tandem with antibiotics has emerged as a promising approach against MRSA. These strategies should enhance the antibiotic efficacy, reduce dosage and toxicity, and bypass MRSA resistance. In this study, we used a checkerboard assay to illustrate the significant synergistic anti-MRSA effect of shikimic acid (SA), a naturally occurring compound, and ceftiofur (CF). Time-kill curves further revealed that a combination of 1/4 of the minimum inhibitory concentration (MIC) of SA and 1/8 MIC of the sodium CF eradicated MRSA within 2 h, with no noticeable toxicity observed with these concentrations. In vivo experiments confirmed that this combination therapy demonstrated robust antimicrobial activity against MRSA-induced bacteremia in mice, significantly reducing bacterial loads in the kidneys, liver, and spleen, attenuating inflammatory cell infiltration, and alleviating pathological damage. This study not only offers a compelling strategy, capitalizing on the synergistic potential of SA and CF, to rapidly address antibiotic resistance but also contributes significantly to the refinement of antimicrobial therapeutic strategies.

Methicillin-resistantStaphylococcus aureus(MRSA) poses a significant clinical challenge due to its multidrug resistance, particularly to β-lactam antibiotics. This study comprehensively evaluated the natural compound emodin for its anti-MRSAactivity, mechanisms of action, and potential for synergy with β-lactam antibiotics. Our findings demonstrate that emodin dose-dependently inhibitsMRSAgrowth and abrogates biofilm formation at 2× MIC. Mechanistic studies revealed that emodin compromises cell membrane and wall integrity, induces oxidative stress, and downregulates the virulence factors SPA and EsxA. Furthermore, emodin acted synergistically with β-lactam antibiotics: it enhanced the ability of cefalexin to block bacterial adhesion and invasion of HaCat cells, and potentiated the efficacy of amoxicillin in clearingMRSAfrom infected macrophages. In conclusion, emodin employs a multi-target mechanism againstMRSAand can resensitize the bacterium to conventional β-lactam antibiotics, presenting a promising strategy for combination therapy that may help curb antibiotic use and resistance development.

Accurate and rapid identification of methicillin-resistant Staphylococcus aureus (MRSA) is of important clinical significance. In this study, a novel aptamer-based fluorometric assay was developed for detection of MRSA in clinical samples by coupling with immunomagnetic separation . The S. aureus cells in clinical specimens were enriched by magnetic separation. Following lysis by staphylococcal lysin , the PBP2a proteins were released from S. aureus cells and detected by the aptamer-based fluorometric assay. Without lengthy period of bacteria cultivation in the traditional susceptibility testing, this test has an overall testing time of only 2 h with the detection limit of 2.63 × 10 3 and 1.38 × 10 3 CFU/mL in PBS and spiked nasal swab, respectively. Since it is simple, rapid and sensitive, this method could be used for the detection of MRSA in various clinical samples.

We report here a facile synthesis of ZnS/CdS heterojunction nanoparticles on an indium–tin oxide (ITO) electrode and their application in the ultrasensitive photoelectrochemical detection of Staphylococcus aureus (S. aureus). The ZnS/CdS/ITO electrode was prepared using one-pot electrodeposition in an acidic solution containing ZnCl2, CdCl2 and Na2S2O3. The optimal ZnS/CdS heterojunction nanoparticles with a Zn/Cd atomic ratio of 1 : 1 showed a high photoelectrochemical response to L-cysteine. L-Cysteine-encapsulated liposome (cysteine@liposome) immunonanocapsules were prepared and used as the labels for photoelectrochemical detection of S. aureus. By coupling cysteine@liposome immunonanocapsule labeling with immunomagnetic separation/enrichment and photoelectrochemical analysis using the ZnS/CdS/ITO electrode, sensitive photoelectrochemical detection of S. aureus was achieved. Under optimal conditions, the linear range for photoelectrochemical detection of S. aureus was from 1 to 4000 CFU mL−1. The proposed method was successfully used for photoelectrochemical detection of S. aureus in milk and juice samples.

In confronting the significant challenge posed by multidrug-resistant (MDR) pathogens, particularly methicillin-resistant Staphylococcus aureus (MRSA), the development of innovative anti-infective strategies is essential. Our research focuses on sortase A (SrtA), a vital enzyme for anchoring surface proteins in S. aureus. We discovered that plantamajoside (PMS), a phenylpropanoid glycoside extracted from Plantago asiatica L. (Plantaginaceae), acts as an effective and reversible inhibitor of SrtA, with a notable IC50 value of 22.93 µg/mL. This breakthrough provides a novel approach to combat both resistance and virulence in MRSA. PMS significantly inhibits S. aureus adhesion to fibrinogen, reducing biofilm formation and hindering the anchoring of staphylococcal protein A to the cell wall. Live-dead cell assays demonstrated increased survival rates in PMS-treated MRSA-infected A549 cells. Fluorescence quenching experiments revealed a robust interaction between PMS and SrtA, with mechanistic analyses pinpointing the critical R197 amino acid residue as the target site. In vivo, PMS was highly effective in a Galleria mellonella infection model, reducing mortality rates in MRSA-infected larvae. Additionally, PMS demonstrated therapeutic efficacy in a mouse pneumonia model, improved survival rates, reduced the bacterial load in pulmonary tissues, and mitigated lung damage. These results validate PMS as a promising compound to mitigate MRSA virulence and thwart resistance by targeting SrtA. This study highlights PMS as a leading candidate for controlling MRSA infections, showing the potential of targeting specific bacterial mechanisms in the fight against MDR infections.

Two-dimensional Ag nanoplates exhibit robust facet-mediated catalytic activity for ozone adsorption and decomposition, accompanied by asymmetric particle dissolution, ROS generation, and concomitant Ag + release. • The synergistic antibacterial effects of Ag nanoparticles and ozone confer superior efficacy, effectively overcoming the defense mechanisms of drug-resistant bacteria and eradicating multidrug-resistant S. aureus strains. The overuse of antibiotics drives the emergence and dissemination of bacterial resistance, thereby escalating global risks associated with exposure to multidrug-resistant (MDR) bacteria, a challenge demanding urgent worldwide attention. Recent studies show that certain bacteria can even develop resistance phenotypes against engineered antibacterial nanomaterials, including silver (Ag)-based nanomaterials. However, effective strategies to counteract such resistance remain elusive. Herein, we propose a simple, low-cost synergistic antibacterial strategy that enhances the antimicrobial efficacy of Ag nanoparticles via ozone, a green, environmentally benign disinfectant widely deployed in water and surface treatment. Mechanistically, Ag nanoparticles exert morphology- and crystal facet-dependent antibacterial effects through three interrelated pathways: catalytic decomposition of ozone, generation of reactive oxygen species (ROS), and release of Ag + ions. Compared with the most used spherical Ag nanoparticles, two-dimensional Ag nanoplates (Ag NPLs) demonstrate significantly superior eradication efficiency against MDR Staphylococcus aureus strains in both in vitro and in vivo settings. This enhanced activity arises from the preferential {100} and {110} facet-mediated adsorption and decomposition of ozone, as well as more favorable interactions at the nano–cell interface. Overall, this ozone-assisted synergistic antibacterial approach presents a promising alternative for combating MDR pathogens, highlighting its substantial potential to mitigate environmental contamination and reduce infection risks posed by resistant microbes.

Polyhexamethylene guanidine (PHMG) is widely used to eliminate biofilms in the dairy industry; however, sublethal chemical stress inadvertently forces pathogens into a viable but non-culturable (VBNC) state. This study examined PHMG-induced VBNC formation in biofilm-associated Staphylococcus aureus under laboratory and food-relevant conditions. PHMG at 4.0 mg/mL (0.4%) eliminated culturability within 3 h, whereas flow cytometry detected a viable fraction of 25.70 ± 3.83% in TSB-grown biofilms at 25 °C. This shift was primarily driven by acute oxidative stress, marked by a 1.56-fold accumulation of intracellular ROS levels and compensatory increases in catalase and superoxide dismutase activities. Concurrently, the cells underwent severe metabolic shutdown, characterized by intracellular ATP depletion, membrane depolarization, and cell shrinkage. Transcriptional profiling of energy- and virulence-related genes confirmed this systemic reprogramming, revealing altered expression in key energy metabolism and stress survival pathways. Crucially, this dormant state was highly unstable in food environments. When transferred back to milk, the VBNC cells rapidly resuscitated within 4 h and successfully resumed hemolysin production. Ultimately, these findings highlight the urgent need for the dairy industry to re-evaluate current PHMG disinfection strategies and closely monitor hidden VBNC risks to ensure food safety.

Drug-resistant Staphylococcus aureus (DRSA) infections present a formidable therapeutic challenge severely threatening human health. To address this, we engineered a metal−phenolic network (MPN) platform that co-immobilizes whole-cell DRSA with immunostimulatory metal ions (i.e., Fe3+, Mn2+, Zn2+) under mild conditions. Among these, the iron-coordinated MPN (SA@FeMPN) exhibited superior potency, leveraging a dynamic valence transition mechanism to robustly amplify the TLR2/NF-κB signaling. This mechanism promoted functional polarization of macrophages to the M1 state and drove dendritic cell maturation. In a murine systemic infection model, SA@FeMPN elicited robust cellular and humoral immunity, achieving a 2−18-fold greater reduction in bacterial burden across major organs compared to other MPNs, and established durable immunological memory to prevent infection relapse. Collectively, this work establishes iron-coordinated MPNs as a rational design platform for next-generation immunotherapy and identifies SA@FeMPN as a promising therapeutic vaccine candidate that not only eradicates established DRSA infections but also confers lasting protection against recurrence.

Staphylococcus aureus infections, particularly those caused by multidrug-resistant strains and associated with biofilm formation, pose a major therapeutic challenge in clinical practice. The objective of this study was to evaluate the antibacterial and antibiofilm activity of mocravimod (KRP-203), an FDA-approved S1P receptor modulator, against clinical S. aureus isolates and to explore its underlying mechanism of action. The antibacterial activity of KRP-203 was assessed against methicillin-susceptible S. aureus (MSSA) and methicillin-resistant S. aureus (MRSA) using MIC determination, time-kill assays, and biofilm inhibition models. KRP-203 exhibited strong bactericidal activity against planktonic MSSA and MRSA, with MIC values ranging 6.25–50μM. Time-kill assays demonstrated rapid bacterial eradication at 8× MIC within 2 h, showing superior killing kinetics compared with vancomycin. At sub-inhibitory concentrations, KRP-203 inhibited biofilm formation by up to 70% and reduced viable bacterial counts in mature biofilms by >2.5 logs. To elucidate the antibacterial mechanism, whole-genome sequencing and quantitative proteomic analyses were performed. These analyses revealed mutations in membrane-associated genes, including glnQ and BCAT, and significant alterations in proteins related to membrane integrity and redox regulation. Consistently, functional assays confirmed that KRP-203 disrupts bacterial cell membrane, as evidenced by dose-dependent membrane depolarization, increased permeability, and direct binding to cardiolipin and phosphatidylglycerol. Molecular docking further predicted a favorable interaction between KRP-203 and GlnQ. In conclusion, KRP-203 demonstrated notable antibacterial and antibiofilm activity against S. aureus, likely through membrane integrity disruption. While these findings highlight its potential as a repurposed antibacterial agent, further studies are required to fully elucidate its molecular targets, optimize antibacterial efficacy, and evaluate its in vivo safety profile.
