pH-Responsive doxorubicin-loaded magnetosomes for magnetic resonance-guided focused ultrasound real-time monitoring and ablation of breast cancer

Na Tang,Yi Zhu,Ziwei Lu,Jiali Deng,Jiajing Guo,Xinyi Ding,Jingyi Wang,Rong Cao,An Chen,Zhongyi Huang,Hongwei Lu,Zhongling Wang

MR-guided focused ultrasound surgery (MRgFUS) is driving a new direction in non-invasive thermal ablation therapy with spatial specificity and real-time temperature monitoring. Although widely used in clinical practice, it remains challenging to completely ablate the tumor margin due to fear of damaging the surrounding tissues, thus leading to low efficacy and a series of complications. Herein, we have developed novel pH-responsive drug-loading magnetosomes (STPSD nanoplatform) for increasing the T2-contrast and improved the ablation efficiency with a clinical MRgFUS system. Specifically, this STPSD nanoplatform is functionalized by pH-responsive peptides (STP-TPE), encapsulating superparamagnetic iron oxide (SPIO) and doxorubicin (DOX), which can cause drug release and SPIO deposition at the tumor site triggered by acidity and MRgFUS. Under MRgFUS treatment, the increased vascular permeability caused by hyperthermia can improve the uptake of SPIO and DOX by tumor cells, so as to enhance ultrasound energy absorption and further enhance the efficacy of chemotherapy to completely ablate tumor margins. Moreover, we demonstrated that a series of MR sequences including T2-weighted imaging (T2WI), contrast-enhanced T1WI imaging (T1WI C+), maximum intensity projection (MIP), volume rendering (VR) and ADC mapping can be further utilized to monitor the MRgFUS ablation effect in rat models. Overall, this smart nanoplatform has the capacity to be a powerful tool to promote the therapeutic MRgFUS effect and minimize the side effects to surrounding tissues.

Inhibiting tumor oxygen metabolism and simultaneously generating oxygen by intelligent upconversion nanotherapeutics for enhanced photodynamic therapy

Dan Wang,Bin Xue,Tymish Y. Ohulchanskyy,Yubin Liu,Artem Yakovliev,Roman Ziniuk,Mengze Xu,Jun Song,Junle Qu,Zhen Yuan

Hypoxia is one of the hallmarks of solid tumor, which heavily restricts the clinical cancer therapy treatments, especially for the oxygen (O 2 ) -dependent photodynamic therapy (PDT). Herein, an intelligent multi-layer nanostructure was developed for decreasing the O 2 -consumption and elevating the O 2 -supply simultaneously. The cell respiration inhibitor –atovaquone (ATO) molecules were reserved in the middle mesoporous silicon layer, and thus were intelligently released at the tumor site after the degradation of gatekeeper of MnO 2 layer, which effectively inhibit tumor respiration metabolism to elevate oxygen content. Meanwhile, the degradation of MnO 2 layer can generate O 2 , further boosting oxygen content. Moreover, the inner upconversion nanostructures as the near infrared (NIR) light-transducers enable to activate photosensitizers for deep-tissue PDT. Systematic experiments demonstrate that this suppressing O 2 -consumption and O 2 -generation strategy improved oxygen supply to boost the singlet oxygen generation to eradicate cancer cells under NIR light excitation. Better still, superior trimodality imaging capabilities (computed tomography (CT), NIR-II window fluorescence , and tumor microenvironment-responsive T1-weighted magnetic resonance (MR) imaging) of the nanoplatform were evaluated. Our findings offer a promising aproach to conquer the serious hypoxia problem in cancer therapy by turning down the O 2 metabolism aveneue and simultaneously generating O 2 .

Ginsenoside Rk1-Loaded Manganese-Doped Hollow Titania for Enhancing Tumor Sonodynamic Therapy via Upregulation of Intracellular Reactive Oxygen Species

Ming Yang,Wenzhi Ren,Haijing Cui,Qiongyu Qin,Qiuye Wang,Weihao Zhu,Xiaoxia Wu,Chunshu Pan,Xiaopeng Qi,Aiguo Wu

Amplifying the intracellular reactive oxygen species (ROS) level remains an urgent challenge for efficient sonodynamic therapy (SDT) of tumors. Herein, by loading ginsenoside Rk1 with manganese-doped hollow titania (MHT), a Rk1@MHT sonosensitizer was conceived to strengthen the outcome of tumor SDT. The results verify that manganese-doping remarkably elevates the UV–visible absorption and decreases the bandgap energy of titania from 3.2 to 3.0 eV, which improves ROS production under ultrasonic irradiation. Immunofluorescence and Western blot analysis demonstrate that ginsenoside Rk1 can block the critical protein of the glutathione synthesis pathway, glutaminase, thus enhancing intracellular ROS by eliminating the endogenous glutathione-depleted pathway of ROS. Manganese-doping confers the nanoprobe T1-weighted MRI function (r2/r1 = 1.41). Moreover, the in vivo tests confirm that Rk1@MHT-based SDT eradicates liver cancer in tumor-bearing mice via dual upregulation of intracellular ROS production. In summary, our study provides a new strategy for designing high-performance sonosensitizer to achieve noninvasive cancer treatment.

Comparative Study on Pharmacokinetics of Four Long-Acting Injectable Formulations of Enrofloxacin in Pigs

Ahmad Salah Uddin,Sun Jichao,Cheng Fusheng,Li Bing,Arbab Safia,Zhou Xuzheng,Zhang Jiyu

A comparative study on pharmacokinetics of four long-acting enrofloxacin injectable formulations was investigated in 36 healthy pigs after intramuscular injection according to the recommended single dose @ 2.5 mg/kg body weight. The drug concentrations in the plasma were computed using high-performance liquid chromatography (HPLC) with fluorescence detection. WinNonLin5.2.1 software was used to analyze the experimental data and compared it under one-way ANOVA using SPSS software with a 95% confidence interval (CI). The main pharmacokinetic parameters, that is, the maximum plasma concentrations (Cmax), the time to maximum concentration (Tmax), area under the time curve concentration (AUCall) and Terminal half-life (T1/2) were 733.84 ± 129.87, 917.00 ± 240.13, 694.84 ± 163.49, 621.98 ± 227.25 ng/ml, 2.19 ± 0.0.66, 1.50 ± 0.37, 2.89 ± 0.24, 0.34 ± 0.13 h, 7754.43 ± 2887.16, 8084.11 ± 1543.98, 7369.42 ± 2334.99, 4194.10 ± 1186.62 ng h/ml, 10.48 ± 2.72, 10.37 ± 2.38, 10.20 ± 2.81, and 10.61 ± 0.86 h for 10% enrofloxacin (Alkali), 20% enrofloxacin (Acidic), Yangkang and control drug Nuokang® respectively. There were significant differences among Cmax, Tmax, and AUCall of three formulations compare with that of the reference formulation. No significant differences were observed among the T1/2 for tested formulations compare with the reference formulation. The pharmacokinetic parameters showed that the tested formulations were somewhat better compared to the reference one. The calculated PK/PD indices were effective for bacteria such as Actinobacillus pleuropneumoniae and Pasteurella multocida with values higher than the cut-off points (Cmax/MIC90≥10–12 and AUC/MIC90 ≥ 125). However, they were not effective against bacteria like Haemophilus parasuis, Streptococcus suis, E. coli, and Bordetella bronchiseptica where lower values were obtained.

A specific “switch-on” type magnetic resonance nanoprobe with distance-dominate property for high-resolution imaging of tumors

Junjie Liu,Wenyan Yu,Mingxu Han,Wei Liu,Zhenzhong Zhang,Kaixiang Zhang,Jinjin Shi

Magnetic resonance imaging (MRI) contrast agents (CAs) are a powerful tool for disease diagnosis. However, the “always on” state of clinically used CAs affects the specificity of tumor MR imaging in vivo . Herein, we constructed a tumor microenvironment “switch-on” type nanoprobe (TSN) with a distance-dominated property for specific tumor MR imaging, by employed a superparamagnetic hollow mesoporous iron oxide nanoparticle (HMIN) as the carrier to encapsulate T 1 -field CA (Gd), followed by blocking of the mesoporous channel with biocompatible hyaluronic acid (HA). In normal tissues with low-level hyaluronidase (HAase), Gd was restricted inside the HMIN, the distance between the two substances became quite close, and the magnetic field of Gd was interfered by the superparamagnetic HMIN, turning “off” the T 1 -field signal is “turned off”; in contrast, in tumor tissues with high-level HAase, Gd was rapidly released from the HMIN by specifically responding to overexpressed HAase, the distance between the two substances became significantly enlarged, and the magnetic field of Gd was not interfered, thus turning “on” the T 1 -field signal to achieve tumor-specific “switch on” MR imaging in a distance-controlled manner. The tumor/normal T 1 -field signal ratio after TSN treatment increased by 1.9-fold compared with free Gd-DTPA, because of its high Gd loading content, tumor-targeting ability of HA and the tumor-specific property. This distance-controlled switchable nanoprobe provides an alternative strategy for high-resolution tumor imaging with high specificity.

Optimization of Bi3+ in Upconversion Nanoparticles Induced Simultaneous Enhancement of Near-Infrared Optical and X-ray Computed Tomography Imaging Capability

Pengpeng Lei,Peng Zhang,Shuang Yao,Shuyan Song,Lile Dong,Xia Xu,Xiuling Liu,Kaimin Du,Jing Feng,Hongjie Zhang

Bioimaging probes have been extensive studied for many years, while it is still a challenge to further improve the image quality for precise diagnosis in clinical medicine. Here, monodisperse NaGdF4:Yb3+,Tm3+,x% Bi3+ (abbreviated as GYT-x% Bi3+, x = 0, 5, 10, 15, 20, 25, 30) upconversion nanoparticles (UCNPs) have been prepared through the solvothermal method. The near-infrared upconversion emission intensity of GYT-25% Bi3+ has been enhanced remarkably than that of NaGdF4:Yb3+,Tm3+ (GYT) with a factor of ∼60. Especially, the near-infrared upconversion emission band centered at 802 nm is 150 times stronger than the blue emission band of GYT-25% Bi3+ UCNPs. Such high ratio of NIR/blue UCL intensity could reduce the damage to tissues in the bioimaging process. The possibility of using GYT-25% Bi3+ UCNPs with strong near-infrared upconversion emission for optical imaging in vitro and in vivo was performed. Encouragingly, the UCL imaging penetration depth can be achieved as deep as 20 mm. Importantly, GYT-25% Bi3+ UCNPs exhibit a much higher X-ray computed tomography (CT) contrast efficiency than GYT and iodine-based contrast agent under the same clinical conditions, due to the high X-ray attenuation coefficient of bismuth. Hence, simultaneous remarkable enhancement of NIR emission and X-ray CT signal in upconversion nanoparticles could be achieved by optimizing the doping concentration of Bi3+ ions. Additionally, Gd3+ ions in the UCNPs endow GYT-25% Bi3+ UCNPs with T1-weighted magnetic resonance (MR) imaging capability.

Metal Ion/Tannic Acid Assembly as a Versatile Photothermal Platform in Engineering Multimodal Nanotheranostics for Advanced Applications

Tao Liu,Mingkang Zhang,Wenlong Liu,Xuan Zeng,Xianlin Song,Xiaoquan Yang,Xianzheng Zhang,Jun Feng

This study reports a family of photothermal materials, metal ion/tannic acid assemblies (MITAs). MITAs from FeIII, VIII, and RuIII afford excellent photothermal efficiency (η ≈ 40%). Sharply differing from the currently existing photothermal agents, MITAs are highlighted by merits including green synthesis, facile incorporation of diagnostic metal ions, and particularly topology-independent adhesion. Owing to the adhesion nature of MITAs, various kinds of MITA-based nanoengineerings are readily available via the self-adhesion of MITAs onto diverse templates, enabling MITAs well suited as a photothermal platform for versatile combination with other therapy approaches and imaging techniques. As a proof of concept, polymeric/inorganic nanoparticle/nanovesicle-supported FeIII-tannic acid (FeIIITA) is fabricated. The photothermal effect is shown to be unaffected by the template origin and type and FeIIITA thickness on the templates. We validate the potency of nanovesicle-supported FeIIITA (PNV@FeIIITA) for tumor-specific photoactivated utilizations, including NIR photothermal therapy with complete tumor elimination, photothermal imaging (PTI), and photoacoustic imaging (PAI) in addition to T1-MRI imaging. PNV@FeIIITA can be simultaneously equipped with functionalities, including T2-MRI imaging by additionally doping MnII and NIR fluorescence imaging by encapsulating a hydrophilic NIR fluoroprobe. MITA demonstrates unparalleled superiority as a photothermal platform in engineering multimodal theranostics for advanced applications.

Self-‘cycling, boosting and propelling’ multi-functional nanoplatforms for improving tumor starvation therapy

Qihang Ding,Gongcheng Ma,Yuxin Xie,Hongrong Shi,Qian Bai,Xuechuan Hong,Hui Zhou,Ling Mei,Jong Seung Kim,Shuai Zhang

Starvation therapy, utilizing glucose oxidase (GOx) to deplete glucose and disrupt tumor energy, has shown promise in cancer treatment. However, this approach alone is insufficient to block ATP generation in tumors. Here, we introduce a self-‘cycling, boosting and propelling’ strategy, overcoming the ‘Achilles' heel’ of traditional starvation therapy by regulating the energy supply with CO. The high concentration of H 2 O 2 in the tumor microenvironment reacts with MnO 2 to generate Mn 2+ and O 2 . Mn 2+ catalyzes a Fenton-like reaction to produce hydroxyl radicals ( OH) for chemodynamic therapy and T1-weighted magnetic resonance imaging (MRI), while O 2 further enhances GOx-mediated glucose consumption, producing more H 2 O 2 . This process not only recycles H 2 O 2 but accelerates CO release, disrupting ATP synthesis and inhibiting tumor energy supply. Additionally, the gas released enhances nanoplatform motility, improving tumor penetration and overcoming the limited permeability of conventional nanoparticles. In vitro and in vivo studies demonstrated superior antitumor and antimetastatic efficacy, coupled with excellent biocompatibility. This strategy offers a promising solution to traditional starvation therapy's challenges, with strong potential for clinical translation and transformative cancer treatment.

Dual-responsive nano-prodrug micelles for MRI-guided tumor PDT and immune synergistic therapy

Hui Guo,Fangzhe Liu,Enqi Liu,Shanshan Wei,Wenbo Sun,Baoqiang Liu,Guoying Sun,Lehui Lu

Micelles as nanocarriers not only offer new opportunities for early diagnosis and treatment of malignant cancers but also encounter numerous barriers in the path of efficient delivery of drugs to diseased areas in the body. To address these issues, we developed a pH/GSH responsive nano-prodrug micelle (NLG919/PGA–Cys–PPA@Gd) with a high drug-loading ratio and controlled drug release performance for MRI-guided tumor photodynamic therapy (PDT) and immune synergistic therapy. Under normal conditions, theranostic nanomicelles remained stable and in a photo-quenched state. Upon accumulation in the tumor site, however, the micelles demonstrated tumor microenvironment (TME) triggered photoactive formed-PPA (a photosensitizer) and NLG919 (an indoleamine 2,3-dioxygenase (IDO) inhibitor) release because the amide bonds of PGA–Cys–PPA and the disulfide linkage of Cys were sensitive to pH and GSH, respectively. More importantly, these micelles could avoid the undesired PPA leakage in blood circulation due to the conjugation between PPA and polymers. Furthermore, the obtained micelles could also enhance the contrast of T1-weighted MRI of tumors by virtue of their high relaxivity (r1 = 29.85 mM−1 s−1). In vitro and in vivo results illustrated that the micelles had good biocompatibility and biosafety. On the basis of the efficient drug delivery strategies in PDT and IDO pathway inhibition, this intelligent dual-drug delivery system could serve as an effective approach for MRI guided combination therapy of cancer.

Copper-thioguanine metallodrug with self-reinforcing circular catalysis for activatable MRI imaging and amplifying specificity of cancer therapy

Yuan Haifeng,Zhao Yan,Yang Chan,Zhang Cheng,Yang Yue,Meng Hongmin,Huan Shuangyan,Song Guosheng,Zhang Xiaobing

For chemotherapy, drug delivery systems often suffer from the inefficient drug loading capability, which usually cause systems toxicity and extra burden to excrete carrier itself. Moreover, the cancer therapeutic efficacy is also greatly limited by the specificity of tumor microenvironment for reactive oxygen species (ROS) based cancer therapeutic strategy ( e.g. , chemodynamic therapy). Herein, we have developed metal-drug coordination nanoplatform that can not only be responsive to tumor microenvironment but also modulate it, so as to achieve efficient treatment of cancer. Excitingly, by employing small molecule drug (6-thioguanine) as ligand copper ions, we achieve a high drug loading rate (60.1%) and 100% of utilization of metal-drug coordination nanoplatform (Cu-TG). Interestingly, Cu-TG possessed high-efficiently horseradish peroxidase-like, glutathione peroxidase-like and catalase-like activity. Under the tumor microenvironment, Cu-TG exhibited the self-reinforcing circular catalysis that is able to amplify the cellular oxidative stress, inducing notable cancer cellular apoptosis. Moreover, Cu-TG could be activated with glutathione (GSH) and facilitated for GSH triggered 6-TG release, higher selective therapeutic effect toward cancer cells, and GSH activated T 1 weight-magnetic resonance imaging. Based on the above properties, Cu-TG exhibited magnetic resonance imaging (MRI) guiding, efficient and synergistic combination of chemodynamic and chemotherapy with self-reinforcing therapeutic outcomes in vivo .
