Multifunctional Nanomedicine with Microenvironment-Regulated Controlled Release for Promoting Healing of Infected Diabetic Wounds

Ketao Yan,Yidan Chen,Tiantian Feng,Ziwei Xiang,Yuzhu Long,Yuzhu Yuan,Wei Wen,Xiuhua Zhang,Shengfu Wang,Suxiao Wang,Jichao Liang,Huayu Xiong

Diabetic wounds pose an ongoing challenge due to their difficult healing and complex microenvironmental nature, and the presence of bacterial infections significantly exacerbates this issue. Here, leveraging the characteristics of the bacterial infection microenvironment, we developed a nanomedicine based on a hollow covalent organic framework (HCOF) for the synergistic application of chemodynamic therapy (CDT) and photodynamic therapy (PDT) in antimicrobial treatment. With HCOF as the carrier, photosensitizer Chlorin e6 (Ce6) is loaded by electrostatic adsorption, and then a manganese dioxide (MnO2) nanoshell was grown on the Ce6@HCOF surface to obtain Ce6@HCOF@MnO2. The nanomedicine exhibits a glutathione (GSH) stimulus response, thereby releasing Mn2+ and Ce6. Mn2+ engages in a Fenton-like reaction with the naturally occurring hydrogen peroxide (H2O2) at the wound site, resulting in the production of highly reactive hydroxyl radicals, while Ce6 generates singlet oxygen under laser irradiation, thus achieving CDT/PDT dual-mode synergistic therapy. Notably, Ce6@HCOF@MnO2 has not only demonstrated the ability to promote collagen deposition but also accelerated skin regeneration and aided in the repair of diabetic wounds. This pioneering approach possesses significant potential for the multifaceted management of bacterial infections and the diseases they precipitate.

Fabrication, Characterization, Interactions, and Controlled Release of Quercetin by Surface-Functionalized Nanoparticle-Protein Corona

Zhiheng Zhang,Baicun Chen,David Julian McClements,Wenqi Yin,Hangyan Ji,Zhengyu Jin,Chao Qiu

This study investigated the interactions between surface-modified starch nanoparticles and digestive enzymes (pepsin and trypsin), with emphasis on enzyme activity modulation, protein corona formation, and quercetin release kinetics. Three quercetin-loaded nanoparticle formulations were prepared: unmodified starch nanoparticles (QSNP), amine-functionalized (QNH 2 ), and carboxyl-functionalized (QCOOH). The protein corona formation on the starch nanoparticles was found to be strongly influenced by their surface properties, as shown by quartz crystal microbalance, atomic force microscopy, circular dichroism, and thermal analyses. QNH 2 formed a thick and soft cationic protein corona, which greatly reduced enzyme activities (especially trypsin), whereas the corona effect was minimal for QSNP. The quercetin release of QNH 2 (36.2%) was lower than that of QSNP (53.6%), which was attributed to corona thickness/charge differences. Thermal analysis confirmed that the nanoparticles remained stable after adsorption of proteins, with QNH 2 being the most thermally stable, followed by QSNP and QCOOH. These findings suggest that modulating the surface charge and functional groups of nanoparticles can alter their interactions with digestive enzymes, which may be used to control the release of encapsulated bioactive compounds. These insights are critical for the design of effective oral delivery systems for nutraceutical and pharmaceuticals.

Potential applications of PLGA/PVA coaxial nanofibers with controlled release for guiding tissue regeneration

Huinan Zhang,Dongchao Ji,Kunlong Zhao,Zhuoheng Li,Shuo Yang,Peng Wang,Wenxin Cao,Yu Sun

Biomedical scaffolds are increasingly used in bone repair due to their exceptional ability to support cell growth and proliferation. This study developed a multifunctional poly(lactic-co-glycolic acid) (PLGA)/polyvinyl alcohol (PVA)/metronidazole coaxial electrospun nanofiber membrane to overcome the limitations of current bone tissue self-repair mechanisms. Optimization of the coaxial electrospinning parameters significantly improved the membrane's overall performance. Mechanical property testing revealed that the tensile strength increased from 4.304 ± 0.079 MPa to 6.915 ± 0.032 MPa as the shell layer feeding rate was increased. Drug release studies demonstrated a marked reduction in the initial burst release of metronidazole as the shell layer thickness increased. The release amount decreased from 86% to 34% by the third hour, and the release continued over the course of one week. Furthermore, the in vitro release model transitioned from first-order kinetics to Peppas-Sahlin kinetics. In vitro studies confirmed that the metronidazole-loaded coaxial fiber membrane exhibited excellent biocompatibility, antibacterial properties, and osteogenic potential. In conclusion, PLGA/PVA controlled-release nanofiber membranes loaded with antibacterial drugs offer great promise for bone tissue regeneration therapies.

Design of Multifunctional SC-PLA Pesticide Carrier System and Study of Controlled-Release Performance

Xuanxuan Wang,Ruizhe Wang,Dongxia Han,Yaling Zhou,Qinwei Gao

To construct a high-performance avermectin (Avm) carrier system, this study utilized the advantages of stereocomplex (SC) crystal formation between poly (L-lactic acid) (PLLA) and poly (D-lactic acid) (PDLA) to prepare Avm-loaded stereocomplex polylactic acid (SC-PLA) nanoformulations via the emulsion solvent evaporation method. The results showed the successful formation of SC-PLA after introducing PDLA into the PLLA matrix, and the influence of SC-PLA crystallinity enabled the fabrication of tunable Avm@SC-PLA nanospheres with a regular spherical morphology. Avm@SC-PLA exhibited controlled release characteristics and possessed pH-responsive properties with specific release behaviors under pH 5.5, 7.4, and 8.0 conditions. The Avm@SC-PLA sustained-release nano system had a series of advantages, including controllable particle size, efficient drug loading, excellent sustained-release performance, good UV-shielding ability, high stability, favorable spreadability, and strong affinity for different leaves. In conclusion, the Avm@SC-PLA nanoformulation not only achieves effective loading and stable encapsulation of Avm but also possesses good structural stability and environmental responsiveness. It provides a novel PLA-based carrier strategy for the efficient delivery of Avm and holds potential application value in the pesticide and pharmaceutical fields.

Lotus leaf flavonoid-loaded zein/chitosan composite films for controlled-release active food packaging

Xinrui Gao,Zhili Ma,Huiqing Wu,Wen Huang,Dexin Zhang

Lotus leaf flavonoids (LLF), derived from an abundant agricultural byproduct, were incorporated into zein/chitosan (ZC) matrices to fabricate multifunctional edible films with controlled-release properties for active food packaging applications. LLF-ZC composite films containing different flavonoid loadings (0.1–0.3%, w / v ) were prepared and systematically evaluated in terms of physicochemical, mechanical, thermal, structural, and bioactive properties. The incorporation of LLF significantly increased film thickness, density, tensile strength, and thermal stability, while maintaining low water vapor and gas permeability. Structural characterization by SEM, ATR-FTIR, and XRD confirmed good compatibility between LLF and the ZC matrix, mainly attributed to hydrogen bonding and hydrophobic interactions. LLF addition markedly enhanced the antioxidant and antibacterial performance of the films, with DPPH and ABTS radical scavenging activities exceeding 90% at higher LLF contents and concentration-dependent inhibition against Escherichia coli and Staphylococcus aureus . Among the tested formulations, the 0.2% LLF-ZC film exhibited the most balanced overall performance in terms of mechanical strength, barrier properties, and bioactivity. The release behavior of LLF was strongly influenced by the polarity of food simulants, with the highest cumulative release observed in 50% ethanol, followed by water and 95% ethanol. Kinetic analysis indicated that the release profiles were best described by the Peppas-Sahlin and Ritger-Peppas models, demonstrating that Fickian diffusion was the dominant release mechanism. Overall, these results highlight the potential of LLF-ZC films as sustainable active packaging materials, particularly for semi-fatty food systems.

Catechyl lignin enhances self-assembly for Pickering emulsion stabilization and the controlled-release application

Lingjuan Luo,Ya Ma,Dingfu Wang,Fangfang Wang,Zhicheng Jiang,Bi Shi

Lignin structure significantly affects the self-assembly ability into lignin nanoparticles (LNPs) and their properties for the specific applications. Herein, several lignin oligomers with distinctive chemical structures were extracted from the typical biomasses and self-assembled into LNPs, which were further used to stabilize Pickering emulsion. The linearly arranged catechyl unit and the abundant phenolic hydroxyl groups in the castor seed coats-derived lignin oligomers would promote their π-π stacking and hydrogen bonding interactions for the self-assembly. Associating with further fractionation of the catechyl lignin, the hydrophilic part with large molecular weight were separated, and self-assembled into small and uniform LNPs. The catechyl unit in the catechyl lignin self-assembled LNPs endowed high hydrophilicity of the LNPs surface with rich negative charge. The uniform LNPs could stabilize Pickering emulsion with smaller vacuoles, which showed high stability maintaining at least 15 days. Subsequently, the LNPs-stabilized Pickering emulsion was applied for curcumin encapsulation, improving its storage and controllable release. This study may contribute to the application of curcumin in food and drug formulations, increase the utilization efficiency of pharmaceutical molecules and reduce their usages, creating economic and alleviating environmental issue.

A hydrophobic and high-toughness elastomer-modified bio-based film-coated controlled-release fertilizer

Denglun Chen,Lulu Cao,Meiqi Han,Junyin Li,Zhaohui Tong,Chunyan Tang,Yuechao Yang,Shugang Zhang

The development of bio-based controlled-release coating materials for controlled-release fertilizers has become a key research focus in sustainable agriculture, driven by their renewability, cost-effectiveness, and environmental benefits in contrast to conventional petrochemical counterparts, which are often non-renewable, expensive, and slow to degrade. However, bio-based materials utilized for controlled-release coatings, such as vegetable oil and lignin, usually contain hydrophilic groups, which lead to poor hydrophobicity. Furthermore, the coating shell has poor toughness and is prone to rupture. These deficiencies ultimately result in suboptimal nutrient release synchronization with crop growth cycles. In this study, a hydrophobic and high-toughness bio-based polymer coating was prepared by combining an elastomer as a modifier with the bio-based film materials. The bio-based polymer coating modified with 20% elastomer (derived from pentaethylene glycol, castor oil, and isophorone diisocyanate) exhibits high cross-linking density (high gel content), which significantly enhances the coating's mechanical properties and barrier function, thereby successfully mitigating the limitations on its release performance. Specifically, the ultimate tensile strength increases from 1.87 MPa to 14.09 MPa, the elongation at break increases from 6.06% to 15.22%, the water contact angle rises from 111.86° to 126.09°, and the controlled-release period extends from 98 days to 140 days. This study presents a new approach for incorporating the elastomer to enhance bio-based controlled-release fertilizer coating materials. In the long run, this approach is expected to offer technical insights for facilitating resource utilization and the development of sustainable agriculture.

Synthesis and Insecticidal Activity of Enzyme-Triggered Functionalized Hollow Mesoporous Silica for Controlled Release

Amir E. Kaziem,Yunhao Gao,Shun He,Jianhong Li

In the present study, enzymatic responsive controlled release formulations (CRFs) were fabricated. The CRFs were achieved by anchoring mechanically interlocked molecules using α-cyclodextrin onto the surface pore rims of hollow mesoporous silica (HMS). The CRFs were characterized using Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, and thermogravimetric analysis. The results showed that the CRFs had extraordinary loading ability for chlorantraniliprole (42% w/w) and could effectively preserve chlorantraniliprole against degradation under thermal conditions and UV radiation. The CRFs have been proven to be enzyme-sensitive. The release ratio of chlorantraniliprole from CRFs can be accelerated observably when external α-amylase was introduced. The persistence of CRFs was evaluated by regular sampling feeding experiment using Plutella xylostella as the target insect. The results showed that the larval mortality of P. xylostella was much higher than that of Coragen under all concentrations after 14 days, which proved that CRFs had remarkable persistence.

Smart pH-Responsive Controlled-Release Fertilizers Using Castor Oil Alkyd Resin-Based Polyurethane Coatings

Boning Su,Xiangjie Zhao,Ziyao Li,Bin Gao,Denglun Cheng,Junyin Li,Zekun Wang,Jiahui Liu,Chenghao Liu,Tianlin Shen,Yuanyuan Yao,Shugang Zhang,Yuechao Yang

Biobased controlled-release fertilizers (CRFs) contribute to sustainable agricultural development. However, their nutrient release often lacks sufficient responsiveness to environmental fluctuations, limiting overall fertilizer use efficiency. This study aimed to develop smart and green CRFs capable of modulating nutrient release in response to soil pH variations. A low-cost dehydrated castor oil (DCO)-based or castor oil (CO)-based alkyd resin (DCOCA/COCA) was used as the foundation for polyurethane (PU) reactions to prepare a DCOCA/COCA-coated CRF (DCOCAPF/COCAPF). Next, a one-step polymerization reaction was performed between HDI and differing amounts of 3-dimethylamino-1,2-propanediol (DMAPD), which contains a tertiary amine group with pH-responsive properties. The resulting products (DMAPDIs) were then subjected to a further polyurethane reaction with COCA to produce pH-responsive CRFs (DMAPDIPFs). The DMAPDIPFs exhibited excellent pH responsiveness with approximately 29% increase in the nitrogen release rate and 7-day reduction in the nitrogen release longevity under acidic conditions compared to that under neutral conditions, requiring only 1% of DMAPDI by total mass of the coating materials. This work provides valuable insights for developing smart CRFs that can autonomously adjust their nutrient release characteristics in response to environmental stimuli to increase the fertilizer utilization rate and thereby promote sustainable agricultural development.

Protonated pentaethylenehexamine-functionalized hydrochar for adsorption of anionic pharmaceuticals and controlled-release of pesticides

Lei Cui,Kun Ye,Shu-Han Chai,Qian Zhang,Bing Li

The inappropriate use and discharge of pesticides and pharmaceuticals pose a serious threat to ecosystems, which can be effectively mitigated with environmentally friendly hydrochar. Here, a protonated pentaethylenehexamine-functionalized hydrochar (PEHA-HC) was successfully synthesized through preparation of glycidyl ester-functionalized hydrochar, amination grafting with PEHA, and protonation. The performance of PEHA-HC in removing two anionic pharmaceuticals, 2,4-dichlorophenoxyacetate (2,4-D Na) and diclofenac sodium (DS), was systematically investigated. The rich protonated amino groups on the surface endow PEHA-HC with a positive charge and a high point of zero charge, enabling efficient adsorption of these chemicals across a broad pH range, with the largest adsorption capacities exceeding 417.78 and 679.43 mg·g −1 , respectively. The adsorption kinetics followed the pseudo-second-order model, while the adsorption isotherms conformed to the Langmuir equation. The primary mechanisms for adsorption of these anions by PEHA-HC were electrostatic attraction and anion exchange. Furthermore, PEHA-HC was explored as a stimuli-responsive carrier for controlled pesticide delivery. The release behavior of the 2,4-D@PEHA-HC formulation was highly sensitive to environmental pH and ionic strength. Release profiles were well expressed with the Ritger-Peppas model (release exponent n  = 0.37–0.65), implying a combined diffusion-erosion mechanism or Fickian diffusion. Notably, 2,4-D@PEHA-HC demonstrated high biological efficacy for rapeseed. Thus, PEHA-HC functions not only as a high-capacity adsorbent for removing anionic pharmaceuticals from aqueous but also as an intelligent carrier for constructing environmentally responsive controlled-release pesticide formulations. These dual functionalities highlight its potential in reducing pharmaceutical pollution in aquatic systems while improving agrochemical utilization efficiency.

pH/glutathione dual-responsive magnetic carboxymethyl chitosan nanoparticles for the controlled release of astaxanthin

Chenchen Li,Xintong Zhang,Weiye Xiu,Mingfu Yuan,Jingyang Wang,Diaomei Ma,Chunran Han

Astaxanthin (AST) bioavailability is inherently limited by poor stability. In this study, pH/glutathione (GSH) dual-responsive carboxymethyl chitosan (CMCS) superparamagnetic iron oxide (SPION) nanoparticles were prepared, loaded with AST (CMCS-SPION-AST), and characterized. Its stability, responsive release, and kinetics were studied. The results showed that CMCS-SPION successfully encapsulated AST. The composite showed excellent stability (88.66 % AST retention at 4 °C, light-protected) and the highest cumulative release (82.11 % at pH 5.5/10 mM GSH), which was higher than the release rates of previously reported single pH-responsive carriers. The release curves of CMCS-SPION-AST at pH 5.5, pH 7.4/10 mM GSH, and pH 5.5/10 mM GSH showed good model fit (R 2  > 0.9), indicating non-Fickian diffusion. The release process represents a synergistic effect of diffusion and dissolution. This study provides novel insight for the development of responsive drug delivery systems and expands the application prospects of AST as a functional natural pigment in the food industry.

Liposome Crosslinked Polyacrylamide/DNA Hydrogel: a Smart Controlled-Release System for Small Molecular Payloads

Danya Lyu,Shanshan Chen,Weiwei Guo

A novel stimuli-responsive hydrogel system with liposomes serving as both noncovalent crosslinkers and functional small molecules carriers for controlled-release is developed. Liposomes can crosslink polyacrylamide copolymers functionalized with cholesterol-modified DNA motifs to yield a DNA hydrogel system, due to the hydrophobic interaction between cholesteryl groups and the lipid bilayer of liposomes. Functional information encoded DNA motifs on the polymer backbones endow the hydrogel with programmable smart responsive properties. In a model system, the hydrogel exhibits stimuli-responsive gel-to-sol transformation triggered by the opening of DNA motifs upon the presence of a restriction endonuclease enzyme, EcoR I, or temperature change, realizing the controlled-release of liposomes which are highly efficient carriers of active small molecules payloads. Two active molecules, 1,1-dioctadecyl-3,3,3,3-tetramethylindodicarbocyanine perchlorate (DiIC18(5)) and calcein, are chosen as the hydrophobic and hydrophilic model payloads, respectively, to address the feasibility of the releasing strategy. Moreover, the hydrogel exhibits injectable property as well as self-recovery behaviors.
