Sensing systems based on cholinesterase and carboxylesterase coupled with different transduction technologies have emerged for pesticide screening owing to their simple operation, fast response, and suitability for on-site analysis. However, the broad spectrum and specificity screening of pyrethroids over organophosphates and carbamates remains an unmet challenge for current enzymatic sensors. Human serum albumin (HSA), a multifunctional protein, can promote various chemical transformations and show a high affinity for pyrethroids, which offer a route for specific and broad-spectrum pyrethroid screening. Herein, for the first time, we evaluated the catalytic hydrolysis function of human serum albumin (HSA) on the coumarin lactone bond and revealed that HSA can act as an enzyme to catalyze the hydrolysis of the coumarin lactone bond. Molecular docking and chemical modifications indicate that lysine 199 and tyrosine 411 serve as the catalytic general base and contribute to most of the catalytic activity. Utilizing this enzymatic activity, a broad specific ratiometric fluorescence pyrethroids sensing system was developed. The binding energetics and binding constants of pesticides and HSA show that pyrethroids bind to HSA more easily than organophosphates and carbamates, which is responsible for the specificity of the sensing system. This study provides a general sensor platform and strategy for screening pesticides and reveals the catalytic activity of HSA on the hydrolysis of the coumarin lactone bond, which may open innovative horizons for the chemical sensing and biomedical applications of HSA.

A novel photosensitizer hybrid film (Ir(cumr)2(dabpy)+@NWF-g-MAH) has been designed and synthesized by anchoring a coumarin–Ir(III) complex on a polymer substrate. Photocatalytic tests show that Ir(cumr)2(dabpy)+@NWF-g-MAH displays a long lifetime of over 650 h under visible-light irradiation. The hydrogen evolution efficiency of Ir(cumr)2(dabpy)+@NWF-g-MAH is nearly 25 times higher than that of [Ir(ppy)2(dabpy)]+@NWF-g-MAH in 100 h, and optimizing the average concentration of Ir(cumr)2(dabpy)+@NWF-g-MAH in the hydrogen evolution system improves the hydrogen evolution amount to 12 790 μmol m–2. This photocatalytic system achieves the best synergy of hydrogen evolution efficiency and lifetime so far. The high performance is derived from the sterically bulky substrate effectively inhibiting the photodegradation of the photosensitizer and the coumarin group with strong visible-light absorption in the visible region. This work provides a novel direction for developing a durable and efficient Ir(III) complex for photocatalytic application.

Chitosan grafting coumarin-3-carboxylic acid (CS-g-CCA) fiber with fluorescent function and enhanced tensile strength was successfully prepared by in-situ wet-spinning. FTIR and NMR results demonstrate that CCA is successfully grafted onto the CS molecule chains. As the grafting rate increases from 4.2 to 15.8 %, the spinning solution viscosity increases from 22 to 54 Pa·s. SEM observations show that the CS and CS-g-CCA fiber surfaces and cross-sections exhibit homogeneity and smoothness. Likewise, as the drawing ratio increases from 1.0 to 1.4, 2D WAXS patterns illustrate the molecular chain oriented significantly along the drawing direction. The CS-g-CCA fiber (the grafting rate of 15.8 %) exhibits a maximum breaking strength of 1.06 cn/dtex, increasing by 20 % more than the CS fiber. Meanwhile, it has a peak fluorescence intensity of around 480 nm. In addition, the continuous preparation process simplifies the technological route and improves the preparation efficiency of CS-g-CCA fiber. As-prepared CS-g-CCA fiber with enhanced tensile strength and elevated fluorescence efficiency lays the foundation for potential application in fluorescent probes , anti-counterfeiting, and biomedicine fields.

Lanthanide-doped biocompatible nanoparticles have promising applications in near-infrared second region imaging due to their high chemical stability, enhanced photostability and sharp emission bandwidth. However, the weak light absorption capacity limits the application of rare-earth nanoparticles (RENPs) for bioimaging. We prepared a coumarin-derived dye sensitized NaYGdF 4 :Yb,Er nanoparticle probe, in which the organic dye enhances photon absorption through the sensitization process, improving the luminescence efficiency of the rare earth particles near 1000 and 1500 nm. In addition, good water solubility and stability of the probe are imparted by coating the particles with amphiphilic polymers distearoyl phosphatidylethanolamine-polyethylene glycol (DSPE-PEG) and polyacrylic acid. This composite probe with good biocompatibility and NIR II luminescence properties can be used for vascular imaging, providing a tool for the detection of hematologic-related diseases.

Background and Purpose The short biological half-life limits the therapeutic use of glucagon-like peptide-1 (GLP-1) and chemical modification to improve the interaction of peptides with serum albumin represents an effective strategy to develop long-acting peptide analogues. Coumarin, a natural product, is known to bind tightly to plasma proteins and possesses many biological activities. Therefore, we designed and synthesized a series of coumarin-modified GLP-1 derivatives, hypothesizing that conjugation with coumarin would retain the therapeutic effects and prolong the biological half-life of the conjugates. Experimental Approach Four cysteine-modified GLP-1 analogues (1–4) were prepared using Gly 8 -GLP-1(7–36)-NH 2 peptide as a starting point. These analogues were conjugated with two coumarin maleimides to yield eight compounds (conjugates 6–13) for testing. Activation of human GLP-1 receptors, stability to enzymic inactivation in plasma and binding to human albumin were assessed in vitro . In vivo , effects on oral glucose tolerance tests (OGTT) in rats and on blood glucose levels in db/db mice were studied. Key Results Most conjugates showed well preserved receptor activation efficacy, enhanced albumin-binding properties and improved in vitro plasma stability and conjugate 7 was selected to undergo further assessment. In rats, conjugate 7 had a longer circulating t 1/2 than exendin-4 or liraglutide. A prolonged antidiabetic effect of conjugate 7 was observed after OGTT in rats and a prolonged hypoglycaemic effect in db/db mice. Conclusions and Implications Cysteine-specific coumarin conjugation with GLP-1 offers a useful approach to the development of long-acting incretin-based antidiabetic agents. Conjugate 7 is a promising long-lasting GLP-1 derivative deserving further investigation.

The optical behavior of a simple intramolecular charge transfer (ICT) fluorescent probe, a coumarin-nitrobenzene conjugate (CNB), has been described to detect cyanide (CN−) selectively in buffered aqueous media. The cyanide addition on the electron-deficient alkene bridge interrupts the π-conjugation in CNB, which obstructs the ICT process and results in a color change from yellow to colorless and a significant fluorescence enhancement at 410 nm. Job's plot analysis revealed a 1 : 1 stoichiometry for the interaction between the probe and cyanide anion with a detection limit of 0.14 μM. The probe was also used for fabrication of test strips that could be used in practical and efficient CN− test kits.

The chirality of amino acids plays an important role in biological and medical sciences. The development of achiral small-molecule probes that can simultaneously determine the absolute configuration, enantiomeric excess, and total concentration of amino acids is significant. We reported the currently available achiral coumarin aldehyde probe that could form Schiff bases with free amino acids at room temperature to induce CD signals and change UV-vis signals. The red-shifted UV-vis signals were independent of the substrate's chirality and could be used to determine the total concentration. Conversely, the enantioselective CD signals could be used to determine the absolute configuration and enantiomeric excess. The usefulness and practicability of this sensing method were demonstrated by analyzing 6 non-racemic phenylalanine samples with different enantiomeric compositions and concentrations.

Due to the low bioavailability and severe toxic side effects caused by the lack of selectivity of traditional chemotherapy drugs, the targeted delivery of chemotherapy drugs has become the key to tumor treatment. The activity and transmembrane potential of mitochondria in cancer cells were significantly higher than that of normal cells, making them a potential target for chemotherapeutic drug delivery. In this study, triphenylphosphine (TPP) based mitochondria targeting polylactic acid (PLLA) nanoparticles (TPP-PLLA NPs) were synthesized to improve the delivery efficiency of anticancer drugs. The carrier material was characterized by 1H NMR and FT-IR and 7-hydroxyl coumarin (7-HC) was successfully loaded into TPP-PLLA to form 7-HC/TPP-PLLA NPs. Further studies showed that TPP-PLLA NPs were primarily accumulated in the mitochondrial and 7-HC/TPP-PLLA NPs had higher antitumor activity. Taken together, our results indicated that TPP-PLLA NPs could be a promising mitochondria-targeted drug delivery system for cancer therapy.

According to the mechanism of hydrazine to ester bond elimination, a novel ratiometric fluorescent probe (FCP) based on the fluorescein-coumarin structure is designed and synthesized for detecting hydrazine. The obvious red shift in the absorption and fluorescence spectrum is caused by the hydrolysis of the ester bond of FCP by upon mixing with hydrazine. The proposed FCP probe is shown to have linear detection ranges from 0 to 250 nM for hydrazine, and LOD is 0.364 nM. In addition, this promising probe possesses high sensitivity and selectivity for monitoring the intracellular hydrazine, thus, has great potential to be applied in early diagnosis of disease. Meanwhile, it has been successfully applied to the detection and cell imaging of hydrazine in actual water samples.

Herein, an effective integrated electrochemiluminescence (ECL) bioprobe was constructed based on F-couSiPc as co-reaction accelerator of tris (2,2′-bipyridine-n,n’)-, dichloride, hexahydrate (II) (Ru(bpy) 3 2+ ) and TiO 2 MOFs as carrier for developing a competitive-type ECL strategy for the sensing of deoxynivalenol (DON). Initially, TiO 2 MOFs with high specific surface area and regular pore structure was employed to immobilize Ru(bpy) 3 2+ and Mercapto-beta-cyclodextrin (β-CD), then sensitizer of F-couSiPc was introduced into the system by encapsulating into the cup-shaped cavity of β-CD, which mainly ascribed to the supermolecular self-assembly behavior, perfectly avoiding the aggregation of F-couSiPc. Due to the shortened electronic transmission distance, reduced energy loss and less reagent consumption, the integrated ECL reaction pattern between the luminescent Ru(bpy) 3 2+ and sensitizer of F-couSiPc has shown improved luminous efficiency compared with the common intermolecular ECL reactions. Meanwhile, the (3-Aminopropyl) triethoxysilane (APTES), perylene-3,4,9,10-tetracarboxylic acid (PTCA) and carbon nanohorn (CNHs) nanohybrid (APTES-PTCA@CNHs), combing the remarkable advantages of CNHs with the desirable electronic and optical properties of PTCA, served as matrix to immobilize antigen (DON) with an increasing amount, and on account of the high affinity between the APTES and DON, a stable ECL performance was obtained. Based on the competitive immunoreactions, a facile and sensitive ECL immunosensor was constructed for the detection of DON, with excellent linear range from 0.1 pg/mL to 20 ng/mL and a detection limit of 0.03 pg/mL (S/N = 3).

Effective discrimination of cysteine (Cys) from biothiols and other amino acids has gained increasing attention due to the essential roles of this molecule in biological systems. In this work, we report a new strategy for the detection of Cys in aqueous media and live cells using a simple Cu2+ ensemble (L–Cu2+). A coumarin–salicylaldehyde hydrazone fluorescent ligand (L) was initially synthesized and well characterized. L features high affinity towards Cu2+ (Ka = 1.995 × 106 M−1), accompanied by effective quenching of fluorescence in HEPES buffer. The in situ generated chemosensing ensemble, L–Cu2+, is able to specifically respond to Cys over other biothiols and amino acids through a displacement approach. As a result, a remarkable recovery of fluorescence and UV-vis absorption spectra can be observed. L–Cu2+ showed high selectivity and sensitivity towards Cys. The detection limit is estimated to be 15 nM. Using L–Cu2+ as the fluorescent probe, imaging of Cys in live A549 cells was successfully demonstrated.

In this study, a coumarin-based probe (probe 1) bearing a maleimide group was used to rapidly and selectively detect thiols. After inducing thiols, this probe presented a significant fluorescence enhancement (≥22-fold), whereas other amino acids that had no sulfhydryl group just generated slight changes. The detection limits were determined to be as low as 7.41 nM for Cys, 6.06 nM for GSH, and 7.73 nM for Hcy based on S/N = 3. Furthermore, this probe possessed excellent water solubility and successfully detected thiols in PBS (pH 7.4, 99.5%). More importantly, the practical application of probe 1 for the selective detection of thiols was successfully demonstrated in living T24 cells.
