Cytochrome P450 monooxygenases (P450s) play pivotal roles in diverse insect physiology, including growth, development, xenobiotic detoxification, and olfaction. In the present study, we identified and characterized a novel P450, LmCYP3136A1, from Locusta migratoria , which lacks the conserved cysteine residue critical for canonical P450 activity in its heme-binding motif. Phylogenetic analyses indicated that LmCYP3136A1 clusters within the CYP3 clan. LmCYP3136A1 was predominantly expressed in the gastric caecum and midgut, with moderate expression in the antenna, and its transcript levels peaked in the 5th-instar nymphal stage. Its expression was significantly induced 6.22-fold by carbaryl and 9.44-time by chlorpyrifos and was also upregulated 1.51-fold by the aggregation pheromone 4-vinylanisole (4-VA) and 1.41-fold by the plant volatile 2-ethylpyrazine. RNAi-mediated silencing enhanced locust susceptibility to malathion and carbaryl, increasing mortality by 35% and 42%, respectively, but not to deltamethrin and chlorpyrifos. Furthermore, LmCYP3136A1 silencing significantly reduced electroantennogram (EAG) responses to eight volatiles, including 4-VA and decanal, with reductions of up to 83%. Molecular docking revealed ligand binding via hydrogen bonds and π-π stacking. Our findings demonstrate putative dual roles for LmCYP3136A1 in mediating specific insecticide detoxification and regulating olfactory sensitivity, expanding understanding of P450 functional diversity and structural plasticity in insects.

Chlorinated pharmaceuticals like diclofenac (DCF) pose persistent environmental risks due to their bioaccumulation and toxicity, necessitating advanced remediation strategies. Herein, an integrated electrochemical system was developed, combining a self-nitrogen-doped carbon nitride (CN)-supported PdNi (CN@PdNi) cathode for electrocatalytic hydrodechlorination (ECH) with a CN anode for oxidative mineralization in a single, sequential process. The CN@PdNi cathode achieved 98.6% ECH efficiency, selectively cleaving C–Cl bonds to mitigate toxicity, which is attributed to its structural merits, including a well-dispersed, albeit partially clustered, nanoparticle distribution, metallic Pd/Ni phases, predominant Pd 0 /Ni 0 states, and enhanced porosity. Subsequent CN anodic oxidation enabled 96.8% total organic carbon (TOC) removal, ensuring near-complete mineralization. Mechanistic studies via Electron paramagnetic resonance (EPR), quenching, LSV, and Tafel slopes revealed atomic hydrogen (*H) generated at Pd 0 sites as the primary reactive species, augmented by electron transfer. The system exhibited robust performance across varied conditions and exceptional stability (< 1% efficiency loss over 11 consecutive cycles). Toxicity assessments showed 3–5 Log 10 unit reductions (ECOSAR) and mung bean germination rates improving from 68.7 to 99.8%. This dual-functional detoxification-mineralization strategy overcomes the limitations of single-step processes and offers a scalable, sustainable solution for efficient remediation of halogenated pollutants.

Profenofos residues in the environment pose a high risk to mammals and non-target organisms. In this study, the biodegradation and detoxification of profenofos in an efficient degrading strain, Cupriavidus nantongensis X1 T , was investigated. Strain X1 T could degrade 88.82 % of 20 mg/L profenofos in 48 h. The optimum temperature and inoculation amount of strain X1 T for the degradation of profenofos were 30–37 °C and 20 % ( V /V), respectively. Metabolic pathway analysis showed that strain X1 T could degrade both profenofos and its main metabolite 4-bromo-2-chlorophenol. Metabolite toxicity analysis results showed that dehalogenation was the main detoxification step in profenofos biodegradation. The key gene and enzyme for profenofos degradation in strain X1 T were also explored. RT-qPCR shows that organophosphorus hydrolase (OpdB) was the key enzyme to control the hydrolysis process in strain X1 T . The purified enzyme OpdB in vitro had the same degradation characteristics as strain X1 T . Divalent metal cations could significantly enhance the hydrolysis activity of strain X1 T and enzyme OpdB. Meanwhile, strain X1 T could degrade 60.89 % of 20 mg/L profenofos in actual field soil within 72 h. This study provides an efficient biological resource for the remediation of profenofos residual pollution in the environment.

The huge application of synthetic dyes caused a severe impact in the environment. In the present study, a physico-chemical strategy of heterogeneous-Fenton catalyzed by the natural ferrous ore has been established for toxic chemical degradation , of which the complex and high-expense repetitive pH adjustment procedures were escaping. And this natural heterogeneous catalyst also could be recycled and sustainable for toxic substances treatment involved in synergetic adsorption and oxidation. The siderite , served as an adsorbent and catalyst for the degradation of methylene blue (MB). Siderite exhibited a better adsorption capacity with a saturated adsorption capacity of ∼11.08 mg/g. Batch adsorption experiments have verified that adsorption rate and adsorption equilibrium followed pseudo-second-order rate model and Langmuir isotherm equation, respectively. The combination with H 2 O 2 , showed significant enhancement of MB degradation without any pH adjustment. The effect of siderite dosage, H 2 O 2 dosage, MB concentration, initial pH, and reaction temperature on MB degradation was investigated, which also has indicated the excellent catalytic performance of siderite. About 99.71% of MB was degraded in 480 min with initial pH of 7.0, reaction temperature of 25 °C, siderite, and H 2 O 2 dosage of 2.5 g/L and 122.38 mM, respectively. It was found that siderite could be reused and remained high degradation efficiency on MB after 5 times reutilization, which also could demonstrate the sustainable and effective process to degrade organic pollution. The generation of reactive species including ·OH and O2·- have been confirmed based on scavenger test and electron spin resonance (ESR) analysis, which was dominated by heterogeneous reaction. The possible degradation mechanisms of MB have been predicted based on spectrum scanning and GC-MS analysis. Moreover, acute toxicity assessment with marine photobacterium Vibrio fisheri was conducted to investigate the toxicity change in the adsorption/oxidation coupled process. This sustainable heterogeneous-Fenton technology has been verified as a promising and applicable process for toxic organic chemicals removal due to effective mineralization and detoxification assisted with the natural ore mineral through the simple operation and mild condtions.

Propamocarb is a pesticide widely used to control cucumber downy mildew. The overuse of propamocarb has resulted in residues and phytotoxicity. However, the detoxification and metabolic process of propamocarb have not been documented well. Our previous work showed differences in the propamocarb residues among the different genotypes of cucumber and their regulation by multiple genes. Based on the already reported data on gene expression profiles under propamocarb treatment, we identified the glutathione pathway, including six different genes (Csa4M303130,Csa3M133380,Csa5M409710,Csa7M395820,Csa3M597320, andCsa1M571280), involved in propamocarb detoxification. The qPCR analysis showed thatCsa1M571280(CsGSH2) was most significantly and differentially expressed at 48 h after propamocarb spray in the cucumber varieties Y3F604 (low propamocarb residues) and M729 (high propamocarb residues). In Y3F604,CsGSH2expression increased from 6 to 48 h after spraying propamocarb, and the expression was positively correlated with propamocarb residues, whereas M729 showed no significant difference inCsGSH2expression. Therefore, we presumedCsGSH2as a key gene in managing propamocarb residues. Gene functional analysis showed that propamocarb residues decreased inCsGSH2-overexpressing plants and increased inCsGSH2-antisense plants. Overexpression ofCsGSH2enhanced glutathione (GSH) accumulation and glutathione S-transferase (GST), glutathione reductase (GR), and glutathione peroxidase (GPX) activities, probably for propamocarb detoxification. The activity of antioxidant enzymes (SOD, POD, CAT, and APX) increased to maintain a high antioxidant capacity inCsGSH2-overexpressing plants. The superoxide (O2−), hydrogen peroxide (H2O2), and malondialdehyde (MDA) levels decreased inCsGSH2-overexpressing plants, promoting the antioxidant system composed of ascorbic acid and glutathione (AsA-GSH). Thus, we conclude thatCsGSH2alleviates propamocarb residues and phytotoxicity by enhancing cucumber’s antioxidant and glutathione detoxification potential. Keywords:cucumber;propamocarb;antioxidant;glutathione;detoxification

A novel material of self-shaped ZnO-embedded biomass carbon foam pellets (ZnO@BCFPs) was successfully synthesized and used as fluidized particle electrodes in three-dimensional (3D) electro-peroxone systems for metronidazole degradation. Compared with 3D and 2D + O3systems, the energy consumption was greatly reduced and the removal efficiencies of metronidazole were improved in the 3D + O3system. The degradation rate constants increased from 0.0369 min−1and 0.0337 min−1to 0.0553 min−1, respectively. The removal efficiencies of metronidazole and total organic carbon reached 100% and 50.5% within 60 min under optimal conditions. It indicated that adding ZnO@BCFPs particle electrodes was beneficial to simultaneous adsorption and degradation of metronidazole due to improving mass transfer of metronidazole and forming numerous tiny electrolytic cells. In addition, the process of metronidazole degradation in 3D electro-peroxone systems involved hydroxyethyl cleavage, hydroxylation, nitro-reduction, N-denitrification and ring-opening. The active species of ·OH and ·O2−played an important role. Furthermore, the acute toxicity LD50and the bioconcentration factor of intermediate products decreased with the increasing reaction time.Keywords:electro-peroxone process;particle electrode;metronidazole;degradation mechanism;detoxification

Downsizing supported metal of Fe 0 -based materials in dealing with ever-growing pharmaceutical contamination issues are highly desired but still challenging. Herein, we develop a modular design protocol to synthesize a family of ultrafine alloy nanoclusters embedded in a nitrogen-enriched carbon framework (NEC@FeM (M=Pd, Pt, Au)). Engineering the fine structure of nitrogen-enriched carbon and interfacial interaction with bimetallic nanoclusters largely suppresses bimetallic overgrowth, eventually producing supported ultrafine (< 4 nm), highly dispersed, well-alloyed nanoclusters. The alloying Fe 0 with M for ultrafine nanoclusters optimize the electron flow, chemical stability, and catalytic property, resulting in the obtained NEC@FeM exhibiting excellent doxorubicin (DOX) degradation performance. Density functional theory calculation combined with experimental results attribute the outstanding DOX degradation ability of NEC@FePd originally to the synergistic effect of enhanced DOX affinity and efficient electrons transfer from Fe to DOX. This work provides a paradigm for the design of ultrafine Fe 0 -based nanocluster with application in pollution control.

A surface oxygen vacancy-engineered WO 3-x film with enhanced photoelectrocatalytic (PEC) activity for the detoxification of deoxynivalenol (DON) was successfully synthesized via an improved sol-gel method. The presence of oxygen vacancy was verified by high-resolution transmission electron microscopy, X-ray photoelectron spectroscopy, X-ray diffraction, Raman spectra and electron spin resonance spectroscopy. The introduction of oxygen vacancy into WO 3-x film substantially enhanced its absorption in visible region, and facilitated its transfer and separation of photogenerated charges. This made the WO 3-x film to have excellent PEC activity for the detoxification of DON under visible light illumination at a low bias of +0.3 V. The percentage of DON removed was up to 86.4 % after being treated with PEC for 2 h. When the bias potential applied was further increased, there were no obvious changes in the degradation efficiency of the film. The cell viabilities were dramatically enhanced with prolonged time, which were over 95 % after 2 h PEC treatment compared to the control. Through the analysis of eight intermediates that were separated and identified by high performance liquid chromatography-mass spectrometry, it can be concluded that the reduction of DON and the cleavage of double bond at C9 C10, epoxy group at C12 13 and hydroxyl group at C3 in DON could greatly reduce the toxicity, and the detoxification mechanism of this PEC system was proposed. This work can provide further insights into DON detoxification using PEC system with low bias.

The potential of bile acids (BAs) to reduce aflatoxin B 1 (AFB 1 ) residues and toxicity in Litopenaeus vannamei was evaluated. Both juveniles and subadults were treated with 0, 0.05, 0.15 and 0.25 g/kg BAs for 60 days followed by 10-d AFB 1 exposure (2000 μg/kg), and fifteen shrimp (five shrimp were pooled into one sample, n = 3) from each treatment were collected at five time points (30, 60, 63, 66 and 70 d). All parameters were determined using accepted and standard methods with acceptable accuracy (recovery) of 90–110%. Results demonstrated that BAs reduced the AFB 1 residues in shrimp (limit of detection: 0.01 μg/L, relative standard deviation < 10% and recovery: 92.1–96.8%). BAs increased the detoxification of AFB 1 and decreased the levels of oxidative stress products by increasing Phase II and antioxidant systems, avoiding AFB 1 -induced deterioration of shrimp meat and health risks to human. The confidence level was 95%.

Cadmium (Cd) contamination in rice threatens rice food safety. Organosilicon shows substantial potential to reduce the uptake and accumulation of Cd in crops. However, the current understanding of the mechanisms by which organosilicon reduces Cd toxicity in rice remains fragmented. In a one-season pot experiment using soil with a total Cd concentration of 1.04 mg·kg−1, we examined organosilicon-modulated physiological responses and Cd-transport gene expression in two rice varieties. The three organosilicon compounds tested were KH590 (3-mercaptopropyltrimethoxysilane), DMDCS (dimethyldichlorosilane), and D6 (dodecamethylcyclohexasiloxane). Two-way ANOVA confirmed significant cultivar × treatment interactions for Cd in all organs (p< 0.05). DMDCS treatment reduced grain Cd in Shaoxiang 100 by about 43.4%. Subcellular fractionation revealed that DMDCS enhanced Cd sequestration in root and culm cell walls (60.8% increase in culm), while D6 and KH590 showed greater efficacy in leaves. Gene expression analysis indicated that KH590 most potently suppressed the root uptake gene OsNramp5 (76% reduction), whereas DMDCS and D6 exerted stronger inhibition on the long-distance transport gene OsLCT1 (up to 91.8%). Principal component analysis further confirmed that KH590 primarily inhibited root uptake, DMDCS mainly enhanced root Fe oxide deposition and culm Cd retention, while D6 best maintained biomass and reduced late-stage Cd transport to grains, increasing yield by 28%. These findings provide evidence for differential pathways through which organosilicon compounds mitigate Cd stress in rice under pot conditions, offering a theoretical reference for reducing Cd pollution in farmland.

Aflatoxin B1 (AFB1) is the most common and toxic among the aflatoxin group. Improper storage of both raw and processed peanut oil makes it susceptible to AFB1 production. Here, a closed-loop photocatalytic reactor consisting of a glass tube coated with TiO 2 as a catalyst has been developed to study the photocatalytic detoxification of AFB1 in peanut oil. The removal rates of AFB1 in the photocatalytic process were apparently higher than that of the photolysis process. It is inappropriate to approximate Langmuir–Hinshelwood kinetics by a first-order kinetic expression based on the experimental kinetic data for the photocatalytic process. The newly proposed Weibull distribution model provides a good description of the kinetics of the photocatalytic process over the ranges of initial AFB1 concentrations and light intensities studied. The key parameters affecting the detoxification efficiency in terms of the reliable life of this model are described.

Helicoverpa armigera Hübner (Lepidoptera: Noctuidae) is a globally devastating pest and has evolved with varying levels of resistance to a broader spectrum of insecticides. CarEs are major enzymes involved in insecticide detoxification and metabolic resistance. Four CarE genes were identified and cloned from H. armigera, and the expression pattern analyses indicated that they were predominantly expressed in the detoxifying tissues and larval feeding periods. The insecticide inductive assays further suggested that these CarE genes with expressions were significantly upregulated after beta-cypermethrin, lambda-cyhalothrin, malathion, and chlorpyrifos exposures. The purified recombinant proteins of both CarE016B and CarE016C by bacterial expression exhibited significantly higher catalytic efficiency toward α-naphthyl acetate and also showed relatively stronger binding with both β-cypermethrin and λ-cyhalothrin compared to the CarE006C and CarE015A. In vitro metabolism assay with HPLC suggests that CarE016B and CarE016C have the ability to metabolize β-cypermethrin, λ-cyhalothrin, and malathion with varying hydrolase activities. GC-MS/MS analysis identified 3-phenoxybenzaldehyde (3-PBAld) as the metabolite of both β-cypermethrin and λ-cyhalothrin. Homology modeling and molecular docking analyses further indicate that these three types of insecticides could be anchored into the active pocket of both CarEs. Collectively, these results demonstrate that both CarE016B and CarE016C play a critical role in the detoxification of pyrethroid and organophosphate insecticides in H. armigera. This study provides the foundations for a comprehensive understanding of the role of the CarEs family in insecticide detoxification and resistance in H. armigera.
