In Vitro Degradation and Photoactivated Antibacterial Activity of a Hemin-CaP Microsphere-Loaded Coating on Pure Magnesium

Lixin Long,Yang Song,Xiaoyi Tian,Lanyue Cui,Chengbao Liu,Shuoqi Li,Yu Wang,Rongchang Zeng

Photoactivated sterilization has received more attention in dealing with implant-associated infections due to its advantages of rapid and effective bacteriostasis and broad-spectrum antibacterial activity. Herein, a micro-arc oxidation (MAO)/polymethyltrimethoxysilane (PMTMS)@hemin-induced calcium-bearing phosphate microsphere (Hemin-CaP) coating was prepared on pure magnesium (Mg) via MAO processing and dipping treatments. The morphology and composition of the coating were characterized via scanning electron microscopy, Fourier transform infrared spectrometer, X-ray diffractometer and X-ray photoelectron spectrometer. Corrosion behavior was evaluated through electrochemical and hydrogen evolution tests. The release of Fe3+ions at different immersion times was measured with an atomic absorption spectrophotometer. Antibacterial performance and cytotoxicity were assessed using the spread plate method, MTT assay and live/dead staining experiment. The results showed that the corrosion current density of the MAO/PMTMS@(Hemin-CaP) coating (4.41 × 10−8A·cm−2) was decreased by two orders of magnitude compared to that of pure Mg (3.12 × 10−6A·cm−2). Photoactivated antibacterial efficiencies of the Hemin-CaP microspheres and MAO/PMTMS@(Hemin-CaP) coating reached about 99% and 92%, respectively, which we attributed to the photothermal and photodynamic properties of hemin with a porphyrin ring. Moreover, based on the release of Fe3+ions, the MC3T3-E1 pre-osteoblasts’ viability reached up to 125% after a 72 h culture, indicating a positive effect of the coating in promoting cell growth. Thus, this novel composite coating holds a promising application as bone implants. Keywords:magnesium;degradable biomaterial;antibacterial coating;biocompatibility;hemin

Hemin loaded Zn−N–C single-atom nanozymes for assay of propyl gallate and formaldehyde in food samples

Hong Li,Qiulan Li,Qiao Shi,Yijie Wang,Xiuwei Liu,Hao Tian,Xinrui Wang,Dezhi Yang,Yaling Yang

Single-atom nanozymes (SAzymes) show distinct advantages in catalytic activity and selectivity owing to their stability and special characteristic of maximum atomic utilization. Inspired by the structure of natural horseradish peroxidase (HRP), we developed a simple method for specific determination of both propyl gallate (PG) and formaldehyde (HCHO) by utilizing the intrinsic peroxidase mimics activity of hemin (hem) loaded Zn–nitrogen–carbon single-atom nanozymes (Zn–N–C@hem SAzymes). Zn–N–C@hem was prepared via a salt-template strategy and self-assembly, where hemin exhibits enhancing peroxidase-like activity can catalyze oxidation of colorless PG to yellow product. Upon introduction of HCHO into Zn–N–C@hem/PG system, complete suppression of PG oxidation was showed, resulting in distinguished decrease in absorbance. The colorimetric sensors of PG and HCHO based on Zn–N–C@hem/PG were developed at their respective linear range of concentration 1.25–200 mg/kg and 5–250 mg/kg. The practicability of the rapid analysis of PG and HCHO in food samples has been verified with reliable results.

Peptide nanotube/hemin composite with enhanced peroxidase activity for the detection of dopamine in food and drug samples

Xiaolin Mei,Ya Wang,Fang Li,Rui Yang,Yan Zhao,Xiupei Yang

Inspired by natural enzymes, artificial enzymes have been widely studied due to their ease of mass production, robustness to harsh environments and high stability. In this work, a peptide nanotube/hemin composite (KL@hemin) as an artificial enzyme was prepared by immobilizing hemin onto self-assembled peptide nanotubes (PNTs). The successful loading of hemin was determined by a series of characterizations. The multiple noncovalent interactions between the PNTs and hemin endow KL@hemin with strong stability. Subsequent enzyme activity tests showed that the prepared KL@hemin exhibited enhanced peroxidase activity. Further experiments indicate that PNTs as carriers can not only protect hemin from dimerization to maintain   enzyme activity but also increase the affinity of hemin to the substrate for faster binding and accelerate mass transfer, thus promoting the whole catalytic process. Coupled with a peroxidase-catalyzed chromogenic system, a colorimetric method for dopamine detection was constructed based on KL@hemin. The strategy shows high sensitivity and selectivity and has been applied to the determination of dopamine in dopamine injection and meat samples.

In situ formation of G-quadruplex/hemin nanowires for sensitive and label-free electrochemical sensing of acid phosphatase

Qian Bi,Feng Qiu,Ruo Yuan,Yun Xiang

The abnormal variation of the concentration of acid phosphatase (ACP) in human body can be related to several pathophysiological diseases. Herein, a highly sensitive and non-label electrochemical biosensor for detecting ACP on the basis of target-induced inhibition of metal ion/pyrophosphate (PPi) coordination and G-quadruplex/hemin nanowires signal amplification is described. The target ACP molecules can hydrolyze PPi into phosphate to prevent the coordination between Cu 2+ and PPi, and the un-coordinated Cu 2+ further triggers cyclic cleavage of a G-quadruplex sequence (GS)-containing hairpin probes by the Cu 2+ -dependent DNAzyems on the sensor electrode to release the GS, which leads to in situ formation of the GS/hemin nanowires via the stacking of the free GS on the surface-immobilized GS with the presence of hemin . The many hemin molecules in the nanowires can thus yield dramatically enhanced current response for detecting ACP sensitively with the detection limit of 0.45 U/L. Besides, low levels of ACP present in diluted serum samples can also be selectively detected with this method. In addition to its sensitivity and selectivity, the developed method can realize substantial signal amplifications without using enzymes or nanomaterials, which can facilitate the establishment of convenient and sensitive strategies for detecting low concentrations of phosphatase for disease diagnosis.

The oxidase-like activity of hemin encapsulated by single-ring GroEL mutant and its application for colorimetric detection

Wang Xiaoqiang,Xu Baomei,Liu Zhenzhen

Supramolecular anchoring of metalloporphyrins in a protein is an attractive approach to the generation of artificial enzymes. Here, we employ the hydrophobic nanocage of single-ring mutant of bacterial GroEL protein for this purpose. We found that multiple monomeric hemin cofactors can be efficiently loaded into the protein nanocage. The as-prepared biohybrid possessed an oxidase-like catalytic activity and followed the typical Michaelis–Menten kinetics and a ping-pong mechanism in the H 2 O 2 -mediated oxidation of model substrates. In comparison with natural peroxidase, the artificial enzyme exhibited higher affinity for the model substrate. A simple and sensitive colorimetric method for the quantitative detection of H 2 O 2 and glucose was also developed based on the artificial enzyme, with the detection limits determined to be 3.0 μM for H 2 O 2 and 5.0 μM for glucose, respectively. The protein nanocage-based artificial enzyme is very flexible and is envisioned to be adapted readily for binding other metal complexes and catalysis of other reactions.

Magnetic graphene oxide-supported hemin as peroxidase probe for sensitive detection of thiols in extracts of cancer cells

Sai Bi,Tingting Zhao,Xiaoqiang Jia,Peng He

Magnetic graphene oxide (GO)–hemin probes containing disulfide bonds are simply and effectively synthesized through amide reaction to covalently link magnetic particles to GO surface and π – π stacking interaction between hemin and GO to immobilize hemin on GO. Based on the strong nucleophilicity of sulfhydry, we have developed a colorimetric detection system for thiols by using glutathione (GSH) as a model analyte . Upon the introduction of GSH to the fabricated magnetic particle (MP)-GO–hemin probes, the disulfides can be readily reduced by thiols, resulting in the release of GO–hemin hybrids to solution. Due to the existence of hemin on GO surface, the released GO–hemin that has the intrinsic peroxidase-like activity can catalyze the oxidation of ABTS 2− by H 2 O 2 to form the colored radical product ABTS ·− . A broad linear dynamic range of 10 −10   M to 10 −6   M GSH is achieved with a detection limit of 8.2×10 −11   M (3 σ ). Moreover, the new probe is successfully applied to the detection of non-protein thiols and protein thiols in the extracts of Ramos cells, which shows favorable correlationship with the results obtained by electrochemical method . In addition, the MP-GO–hemin probe can detect non-protein thiols in Ramos extracts as low as 500 cells. In this assay, the prepared MP-GO–hemin conjugates are thoroughly characterized by SEM , AFM , UV–Vis, FT-IR, and Raman spectroscopy.

A Cu2O/hemin-GO nanozyme with aptamer-enhanced peroxidase-mimic activity for colorimetric detection of kanamycin in milk

Yaoting Mou,Lu Han,Yanhao Yin,Jianghua Liu,Yanqing Xu,Yuhang Tian,Yanfang Wu,Dongfei Chen,Yemin Guo,Xia Sun,Falan Li

Kanamycin (KAN) is an aminoglycoside antibiotic employed for the treatment of bacterial infections in livestock and poultry and is widely used as a veterinary drug in animal husbandry. Its overuse poses serious threats to public health and ecosystems; while traditional detection methods cannot meet the requirements of widespread application and rapid detection, developing rapid and reliable technology for KAN detection is essential. In this study, a colorimetric aptasensor was developed to rapidly detect KAN residues, leveraging the enhanced catalytic effect of aptamers on the peroxidase-like activity of the Cu2O/hemin-graphene oxide (Cu2O/hemin-GO) nanozyme. In the presence of hydrogen peroxide, the Cu2O/hemin-GO nanozyme demonstrated excellent peroxidase-like catalytic activity, oxidizing 3,3′,5,5′-tetramethylbenzidine (TMB). The adsorption of aptamers on the nanozyme surface increased the negative charge density and significantly enhanced the affinity of the nanozyme for the positively charged chromogenic substrate TMB, boosting the catalytic activity nearly threefold. Upon exposure to KAN, the specific binding of aptamers to KAN reduced their adsorption on the Cu2O/hemin-GO nanozyme, leading to a decrease in the peroxidase-like activity of the Cu2O/hemin-GO nanozyme and a corresponding reduction in color change. The aptasensor exhibited excellent sensitivity with a linear detection range from 0.1 pM to 1 μM and a detection limit of 16 fM. The recovery rates for KAN in milk samples ranged from 92.05% to 110.71%. Furthermore, the colorimetric aptasensor demonstrated high selectivity and reproducibility. Overall, the colorimetric approach achieved the economical, simple and sensitive detection of KAN residues, indicating its promising potential in real-world applications. In addition, this sensing method could be applied to other targets by replacing the aptamer, providing a strategy for developing sensors facilitating rapid detection.

VS4 Nanodendrites with Narrow Bandgaps in Activating Dissolved Oxygen for Boosted Chemiluminescence and Hemin Detection by Unexpected Quenching

Abubakar Abdussalam,Hongzhan Liu,Islam Mohamed Mostafa,Baohua Lou,Dmytro Viktorovych Snizhko,Yuriy Tymofiiovych Zholudov,Wei Zhang,Guobao Xu

Chemiluminescence (CL)-based analytical methods utilize luminophores that need to be activated with an oxidizing agent to trigger CL emission. Despite its susceptibility to decomposition when exposed to external light or trace metals, hydrogen peroxide (H2O2) has been widely used to develop chemiluminescent methods due to the limited number of suitable alternatives for activating chemiluminescent luminophores. Also, analytical methods based on the well-known luminol/H2O2 CL system have low sensitivity. Dissolved oxygen (DO) is a naturally abundant and environmentally benign alternative oxidant for luminol and other CL luminophores. However, DO alone is inactive and needs an efficient catalyst or a coreaction accelerator for its activation. Because of the narrow bandgap of VS4 (ca. 1.12 eV), it can facilitate fast electron-transfer kinetics with an acceptor molecule such as DO. Here, we introduce vanadium tetrasulfide (VS4) to boost CL for the first time. Under the optimized conditions, VS4 nanodendrite catalyzes the generation of reactive oxygen species by activating DO which subsequently reacts with luminol to generate intense CL. It enhances the CL intensity of luminol/DO by about 10,000 times. Surprisingly, hemin remarkably quenches the generated CL of luminol/DO/VS4 nanodendrites, which is completely opposite to its typical enhancement of luminol CL. Based on the remarkable concentration-dependent quenching of the luminol/DO/VS4 nanodendrite CL by hemin, we have developed a sensitive CL method that can selectively detect hemin in the linear concentration range of 1–250 nM and achieved a limit of detection of 0.11 nM. The practical utility of the developed method was demonstrated by the determination of hemin in a pharmaceutical drug for the treatment of acute intermittent porphyria and in human serum. This study demonstrates that VS4 holds great promise in analytical method development.

Imidazole-enhanced luminol/H2O2 chemiluminescent biosensing for SARS-CoV-2 nucleocapsid protein with enzymatic regulation of hemin switch

Hang Ao,Jinshan Xiong,Wencheng Xiao,Wenrui Hu,Jie Wu,Huangxian Ju

Owing to the continuous mutations of the coronavirus, the sensitive and accurate detection of the SARS-CoV-2 nucleocapsid protein is still necessary in clinical diagnosis. Aim to the challenge of sensitivity in chemiluminescence (CL) imaging detection and the need for high-throughput clinical screening, this work used imidazole (Im) as an enhancer for hemin-DNA/luminol/H 2 O 2 CL system to propose a CL imaging method for SARS-CoV-2 nucleocapsid protein detection. In this system Im could reduce the hemin-DNA catalyzed oxidation energy of luminol by H 2 O 2 to promote the CL emission through the accelerated breaking of O-O bond of dianionic cyclic peroxide. The enhancement mechanism was demonstrated with theoretical calculations and several characterization techniques. By combining the proximity-induced initiator release with immunological recognition, an initiator-triggered DNA nanomachine was designed to achieve the release of abundant primers for activating hemin-DNA switch. This signal amplification strategy led to strong CL emission for highly sensitive imaging detection of the target. The Im-enhanced hemin-DNA/luminol/H 2 O 2 CL imaging assay exhibited a linear detection range over 4 orders of magnitude with a detection limit down to 2.75 pg/mL for SARS-CoV-2 nucleocapsid protein, exhibiting promising potential of both Im sensitization and the proposed DNA nanomachine to improve the sensitivity of high-throughput CL biosensing for the early diagnosis and clinical screening of the coronavirus and other diseases.

Aptamer-functionalized biomimetic supramolecular nanozyme constructed by dipeptide, glutaraldehyde and hemin and its excellent sensing performances for tetrodotoxin

Hongchao Ma,Xifeng Ma,Tiantian Dong,Xinyu Bian,Xiaokang Zhang,Yanhui Wei

Bioinspired nanozymes hold promise for simulating natural processes and creating optimized functional systems, but their application is hindered by limited catalytic activity and selectivity. These challenges can be addressed by reconstructing enzymatic active sites to enhance catalytic efficiency and integrating biological recognition units for specificity. In this work, we developed a peroxidase-mimicking nanozyme by stabilizing hemin on a supramolecular scaffold of diphenylalanine (FF) and glutaraldehyde (GA). To enable specific recognition, we conjugated a tetrodotoxin (TTX) aptamer, yielding the He@FF/GA-Apt composite nanozyme. This nanozyme demonstrated robust catalytic activity in 3,3′,5,5′-tetramethylbenzidine (TMB) oxidation. The TTX aptamer conferred specific TTX recognition, with the aptamer-TTX complex blocking the nanozyme active site and reducing its activity. Based on this mechanism, we created a dual-mode TTX detection method using UV–vis spectroscopy and smartphone RGB analysis. The UV–vis mode achieved a linear range of 1.0–40.0 ng mL −1 and a limit of detection (LOD) of 0.61 ng mL −1 , while the smartphone mode had a LOD of 1.43 ng mL −1 in a linear range of 2.0–40.0 ng mL −1 . Both methods performed well in real samples, with recoveries of 96.29 %–102.57 % (UV–vis mode) and 92.07 %–109.46 % (RGB mode). In comparation, the UV–vis mode offers high sensitivity but requires lab equipment, whereas smartphone RGB mode enables rapid on-site detection despite a little lower sensitivity. This work provides a promising approach for developing target-specific nanozyme sensors.

Analysis of poly(ADP-ribose) polymerase-1 by enzyme-initiated auto-PARylation-controlled aggregation of hemin-graphene nanocomposites

Yong Liu,Xiaolin Xu,Haitang Yang,Ensheng Xu,Shuangshuang Wu,Wei Wei,Jin Chen

Poly(ADP-ribose) polymerase-1 (PARP-1) is a highly conserved nuclear enzyme, which binds tightly to damaged DNA and plays a key role in DNA repair, recombination, proliferation, and genomic stability. However, due to the poor electrochemical and optical activity of PARP-1 and its product PAR, only a few studies on its activity detection method have been reported. Herein, we report a simple and sensitive colorimetric strategy to monitor PARP-1 activity based on enzyme-initiated auto-PARylation-controlled aggregation of hemin-graphene nanocomposites (H-GNs). PARP, activated by dsDNA, catalyzed its substrate nicotinamide adenine dinucleotide (NAD+) to polymerize as a poly(ADP-ribose) polymer (PAR). PAR possesses several negative charges, and its charge density is twice that of a single-stranded DNA, which greatly impacts the dispersibility of H-GNs; due to their peroxidase-like catalytic activities, H-GNs can catalyze the chromogenic reaction of TMB and H2O2. As a result, in the presence of different PARP-1 activities, the supernatant of the corresponding solution contained different amounts of dispersed H-GNs and showed different colors after the chromogenic reaction that could be discerned easily by the absorbance or the color changes of the solution. The method was simple, sensitive, and reliable. The proposed method displays a linear range from 0.05 to 1 U with a detection limit of 0.03 U. In addition, this new method has been successfully applied to detect PARP-1 activity in human serum and different cancer cells and evaluate PARP-1 inhibitors.
