Trivalent copper chelate-luminol chemiluminescence system for highly sensitive CE detection of dopamine in biological sample after clean-up using SPE

Lin Wang,Ying Liu,Haoyue Xie,Zhifeng Fu

A transition metal chelate unstable at a high oxidation state, diperiodatocuprate (III) (K 5 [Cu(HIO 6 ) 2 ], DPC), was synthesized and applied in the luminol-based chemiluminescence (CL) system for highly sensitive CE end-column detection of dopamine (DA). This method was based on the fact that DA enhanced the CL emission resulting from the reaction between luminol and DPC in alkaline medium. The DPC-luminol-DA CL system showed very intensive emission and very fast kinetic characteristics, thus resulting in a high sensitivity in flow-through detection mode for CE. Under optimal conditions, the linear range was 1.0 × 10 −8 –5.0 × 10 −5 g/mL ( R 2 = 0.9984) with a limit of detection of 6.0 × 10 −9 g/mL (S/N = 3). The RSDs of the peak height and the migration time were about 4.2 and 2.4% for a standard sample at 3.0 × 10 −6 g/mL ( n = 5), respectively. The presented method has been successfully used for the determination of DA in commercial preparation and human urine samples after clean-up using SPE.

Au@BSA microspheres-luminol and a novel luminescent Zeolitic Imidazolate Framework were used for potential-resolved electrochemiluminescence to detect dual targets

Qihao Zhu,Weiran Mao,Chengli Zhang,Yuan Zhou,Zhiyong Tang,Chao Yu

Here, a novel electrochemiluminescence biosensor based on potential-resolved strategy was firstly prepared for the detection of dual targets α2,3-sialylated glycans and α2,6-sialylated glycans. This is the first time that Au@BSA microsphere was used to connect with luminol to enhance its ECL intensity, and it can generate ECL signals at positive potential. Zeolitic Imidazolate Framework-8 (ZIF-8) and Meso-tetra (4-carboxyphenyl) porphyrin (TCPP) were linked using a one-pot method to synthesize a novel luminescent ZIF (L-ZIF) named TZZ, which can emit ECL signals at negative potential. Moreover, magnetite microspheres were used to construct a sandwich-type biosensor to obtain higher sensitivity and reduce background signals. In addition, the biosensor manufactured directly in solution have a wider linear range than constructed on electrode because it has more available space than the electrode surface. Due to the above advantages, the prepared ECL biosensor exhibited high sensitivity, stability and broader linear range, even for practical analysis. Therefore, the prepared ECL biosensor will become a promising method for determination of α2,3-sialylated glycans and α2,6-sialylated glycans in clinical applications in the future. What is more, it provides a potential method for detection of other multi-targets.

Ultrasensitive electrochemiluminescence biosensor for the detection of tumor exosomes based on peptide recognition and luminol-AuNPs@g-C3N4 nanoprobe signal amplification

Xuejiao Liu,Qiaoe Wang,Jun Chen,Xu Chen,Wensheng Yang

Highly sensitive determination of tumor exosomes is significant for early diagnosis of cancers and precision therapy. Herein, a sandwich peptide-based electrochemiluminescence (ECL) biosensor was developed for determination of phosphatidylserine (PS)-positive exosomes, a promising biomarker for early diagnosis of ovarian malignancy. A PS-specific binding peptide with high affinity was immobilized on Au nanoflowers (AuNFs) modified biosensing interface for recognition and capture of exosomes. Meanwhile, g-C 3 N 4 nanosheet loaded with luminol capped AuNPs (Lum-AuNPs@g-C 3 N 4 ) nanocomposite was used as the ECL signal nanoprobe. The g-C 3 N 4 nanosheets with large surface area were not only utilized as the carrier to immobilize more peptides for recognition of exosomes but also used to catalyze co-reactant H 2 O 2 decomposition to achieve the ECL signal amplification of luminol-H 2 O 2 system. Under optimal conditions, the biosensor showed superior performances compared with most currently available methods, including wider linear range across 5 orders of magnitude and a lower detection limit (LOD) down to 39 particles μL −1 . Moreover, the biosensor could be applicable for determination of exosomes in complex biological samples. This study indicates the combination of peptide recognition with nanoprobe as a label for signal amplification in sandwich ECL biosensing is a great promising strategy for sensitive and cost-effective determination of exosomes.

Flow-injection chemiluminescence of the luminol–potassium periodate system enhanced by TGA–capped CdTe quantum dots for the determination of theophylline

Min Zhou,Qiqi Chen,Ailian Wang,Juanhua Li,Yongjun Ma

The chemiluminescence (CL) behaviour of the luminol–potassium periodate system enhanced by CdTe quantum dots capped with thioglycolic acid (TGA–CdTe QDs) was studied using kinetic experiments, CL spectra, UV–vis absorption spectra and fluorescence spectra. The production of oxygen-containing reactant intermediates (O 2 •− and OH • ) in the present CL system was verified by CL. The possible CL mechanism was discussed in detail. Furthermore, theophylline (THP) was determined based on its enhancement of the CL intensity of the CdTe QDs–luminol–potassium periodate system coupled with a flow-injection technique. Under these optimized conditions, the linear range was found to be from 1.0 × 10 −8 to 1.0 × 10 −5 g/mL with a detection limit of 2.8 × 10 −9 g/mL (3σ). The recoveries for the determination of THP in tablets were from 98.2 to 99.6%.

Immunosensors for dual tumor biomarker detection based on ternary electrochemiluminescence using confined CeO2 nanozyme as Co-reactant enhancers for luminol-O2 system

Chao Lu,Xue Fan,Fengna Xi,Yucheng Zhou

Sensitive and parallel individual detection of multiple tumor biomarkers is critical for early diagnosis and therapeutic monitoring of pancreatic cancer. Herein, a ternary electrochemiluminescence (ECL) system was developed by employing nanochannel-confined cerium dioxide (CeO 2 ) nanozyme as co-reactant enhancers for the luminol-dissolved oxygen (DO) system, enabling parallel individual immunoassays of carcinoembryonic antigen (CEA) and carbohydrate antigen 199 (CA199). Vertically-ordered mesoporous silica film (VMSF) was rapidly grown on an indium tin oxide (ITO) electrode, forming nanochannel array with thickness of 97 nm and diameters of 2.7 nm. CeO 2 nanozyme was synthesized in situ within the VMSF nanochannels via electrochemical deposition. Transmission electron microscopy (TEM), high-angle annular dark-field scanning TEM (HAADF-STEM), and energy-dispersive X-ray spectroscopy (EDX) elemental mapping confirmed CeO 2 nanozyme was confined in nanochannels with high dispersion. CeO 2 nanozyme exhibited peroxidase-like catalytic activity and enhanced the ECL intensity of the luminol-DO system by 13.9-fold under near-neutral conditions through catalysis of both oxygen reduction and luminol oxidation. After derivatization of the VMSF outer surface with epoxy groups, antibodies specific to CEA or CA199 were covalently immobilized to construct two immunorecognition interfaces. Target binding formed antigen-antibody complexes, increasing interfacial resistance and hindering diffusion of the ECL emitters, enabling a signal-off immunoassay mode. The resulting immunosensors displayed wide detection linear ranges from 0.1 pg mL −1 to 100 ng mL −1 for CEA and 0.1 × 10 −3 mU mL −1 to 1.0 × 10 4 mU mL −1 for CA199, with limit of detection (LOD) of 1.3 fg mL −1 and 0.5 × 10 −4 mU mL −1 , respectively. Both immunosensors exhibited good selectivity, reproducibility, and storage stability.

Electrochemiluminescence quenching of luminol by CuS in situ grown on reduced graphene oxide for detection of N-terminal pro-brain natriuretic peptide

Xiaojian Li,Peng Lu,Bin Wu,Yaoguang Wang,Huan Wang,Bin Du,Xuehui Pang,Qin Wei

A novel electrochemiluminescence (ECL) signal-off strategy based on CuS in situ grown on reduced graphene oxide (CuS-rGO) quenching luminol/H 2 O 2 system was firstly proposed. Luminol was grafted on the surface of Au@Fe 3 O 4 -Cu 3 (PO 4 ) 2 nanoflowers (Luminol-Au@Fe 3 O 4 -Cu 3 (PO 4 ) 2 ) which exhibited excellent catalytic effect towards the reduction of H 2 O 2 to enhance the ECL intensity of luminol. Cu 3 (PO 4 ) 2 nanoflowers showed large surface area which can immobilize more Fe 3 O 4 and Au nanoparticles . The quenching mechanism of CuS-rGO was due to ECL resonance energy transfer (RET). The spectral overlap between fluorescence spectrum of Luminol-Au@Fe 3 O 4 -Cu 3 (PO 4 ) 2 and UV–vis absorption spectrum of CuS-rGO revealed that resonance energy transfer was possible. Au nanoparticles were immobilized on the surface of CuS-rGO to capture secondary antibodies. After a sandwich-type immunoreaction, a remarkable decrease of ECL signal was observed. Under the optimal conditions, the immunosensor showed excellent performance for N-terminal pro-brain natriuretic peptide (NT-proBNP) detection with a wide detection range from 0.5 pg mL −1 to 20 ng mL −1 and a low detection limit of 0.12 pg mL −1 (S/N = 3). The prepared NT-proBNP immunosensor displayed high sensitivity, excellent stability and good specificity.

Copper ion modified graphitic C3N4 nanosheets enhanced luminol-H2O2 chemiluminescence system: Toward highly selective and sensitive bioassay of H2S in human plasma

Jun-Tao Cao,Wen-Sheng Zhang,Xiao-Long Fu,Hui Wang,Shu-Hui Ma,Yan-Ming Liu

A high-efficient chemiluminescence (CL) platform for highly selective and sensitive H 2 S detection was constructed on the basis of the quenching effect of S 2 − on the copper ion modified graphitic carbon nitride nanosheets (Cu 2+ -g-C 3 N 4 NSs) enhanced luminol-H 2 O 2 system. Cu 2+ -g-C 3 N 4 NSs with horseradish peroxidase-like catalytic activity were prepared and provide a great improvement for luminol-H 2 O 2 system. The presence of S 2 − induced the formation of CuS precipitate on g-C 3 N 4 NSs surface. The precipitate can block the catalytic Cu 2+ sites on the g-C 3 N 4 NSs surface, resulting in a great CL decrease of CL system. Based on such a mechanism, a simple, highly selective and sensitive CL biosensor for H 2 S detection was designed. Under the optimized conditions, luminol-H 2 O 2 -Cu 2+ -g-C 3 N 4 NSs system gave a decrease of CL intensity with the Na 2 S concentration increasing. The CL biosensor is in a linear range of 10.0 pM–50.0 nM and the detection limit for detecting Na 2 S is as low as 2.0 pM. Moreover, the method here has enjoyed a successful application for determining H 2 S in human plasma samples and the recovery is between 95.7% and 110.0%.

Development of luminol-based chemiluminescence approach for ultrasensitive sensing of Hg(II) using povidone-I2 protected gold nanoparticles as an efficient coreactant

Halawa Mohamed Ibrahim,Wu GuoXing,Li Bing Shi

In this work, we fabricated gold nanoparticles (AuNPs) capped with both polyvinyl pyrrolidone (PVP) and iodine (I 2 ) to act as efficient chemiluminescent coreactants for luminol. AuNPs synthesis was based on the direct chemical reduction of Au 3+ with NaBH 4 in the presence of PVP-I 2 complex. The successful synthesis of PVP-I 2 @AuNPs was confirmed with scanning electron microscopy (SEM) and UV-vis spectrophotometry. Chemiluminescence (CL) intensity of luminol was greatly enhanced, upon its chemical reaction with chemisorbed I 2 on AuNPs surfaces owing to the excellent catalytic activity of AuNPs. The PVP-I 2 @AuNPs/luminol CL sensing system was successfully applied for determination of Hg 2+ ions and the results displayed linearity in a wide range from 0.5 to 2000 nM and an ultrasensitive response to 1.0 nM Hg 2+ . The detection limit of Hg 2+ ions was 0.1 nM, which was 100 times lower than the limit value (10 nM) defined by the U.S. Environmental Protection Agency in drinkable water. This ultrasensitive luminogenic system for Hg 2+ detection also exhibited excellent selectivity among 13 types of metals, suggesting that the luminol/PVP-I 2 @AuNPs system is a promising sensor for real-time detection of Hg 2+ . Co-metal-organic-frameworks with pure uniform crystal morphology prepared via Co2 + exchange-mediated transformation from Zn-metallogels for luminol catalysed chemiluminescence

Xue Qian Tang,Bo Wen Xiao,Chun Mei Li,Dong Mei Wang,Cheng Zhi Huang,Yuan Fang Li

Cation exchange-mediated transformation from Zn-metallogels (MOGs), which was a mild facile strategy relative to the demanding hydrothermal method , was employed to develop Co 2   + metal-organic frameworks (Co-MOFs) at room temperature . The obtained Co-MOFs was of uniform octahedral morphology and possessed high activity to catalyze luminol chemiluminescence without extra oxidants. By adding cysteine, the CL emission of luminol-Co-MOFs system was further enhanced. Based on this phenomenon, Co-MOFs was utilized to build a practical sensing platform for cysteine determination. Under the optimized conditions, the relative CL intensity (ΔI) was proportional to the concentration of cysteine in the range of 2–10   μM, and the detection limit was 0.49   μM (3S/N). Moreover, the established method was applied to the determination of cysteine in commercially available pharmaceutical injections.

Double-enhanced sandwich electrochemiluminescence aptasensor based on g-C3N4–Au-luminol nanocomposites and ZnCuS nanosheets for highly sensitive detection of mucin 1

Kaida Kuang,Yao Lu,Yang Chen,Pei Zhang,Nengqin Jia

The traditional luminol electrochemiluminescence (ECL) sensing suffers from low signal response and instability issues. Here, an Au/ZnCuS double-enhanced g -C 3 N 4 -supported luminol ECL aptasensor is constructed for the sensitive detection of human mucin 1 (MUC1). In this platform, g -C 3 N 4 of a large specific surface area is beneficial to load more luminol illuminants. Au nanoparticles promote the decomposition of H 2 O 2 coreactants to generate more reactive oxygen (•OH and O 2 • − ) intermediates, while ZnCuS can immobilize the aptamer and simultaneously catalyze H 2 O 2 decomposition, realizing the double-wing signal amplification. Under optimal conditions, this sensor shows a good detection capability within 1.0 × 10 −4 –1.0 × 10 3  ng mL −1 and a low detection limit of 5.0 × 10 −5  ng mL −1 , as well as ideal stability, selectivity, and reproducibility. This double-enhanced aptasensor highlights a new signal-enhancement approach for early biomarker detection.

Sensitive and visual determination of sulfamethazine in milk and drinking water using aggregation-induced emission fluorescent sensor based on luminol-europium nanocomposites

Jie Mingsha,Lan Shengkai,Lu Congran,Guo Ruipeng,Qu Xuanxuan,Zheng Qiuchen,Xiang Qisen

Considering the excessive residue and serious harm of sulfamethazine (SM 2 ), the sensitive determination of SM 2 has attracted wide attention in ensuring food security and human health. Herein, using the unique fluorescence property of aggregation-induced emission (AIE), we reported a simple and sensitive detection approach of SM 2 residues in milk and drinking water. Firstly, luminol-europium nanocomposites (luminol-Eu NCs) were self-assembly synthesized via AIE strategy. Then, the fluorescence emission of luminol-Eu NCs could be quenched by SM 2 due to the inner filter effect (IFE). As a result, the fluorescent sensor based on luminol-Eu NCs displayed a wide linear response for SM 2 in a concentration range of 0.1–500 µmol/L and a detection limit of 90 nmol/L was obtained. Furthermore, semi-quantitative detection of SM 2 on test papers was realized via a smartphone-assisted visual sensing device. Therefore, this fluorescent sensor shows a potential value in the field of food safety assessment. Graphical abstract An aggregation-induced emission fluorescent sensor based on luminol-europium nanocomposites was developed and applied for sensitive and visual detection of sulfamethazine in milk and drinking water.

Remarkably Enhanced Luminol/H2O2 Chemiluminescence with Excellent Peroxidase-like Activity of FeCoNi-based Metal-Organic Xerogels for the Sensitive Detection of Dopamine

Fangshuo Liu,Shiyu Xia,Ala’a Mhmoued Abdllh Alboull,Zhiyong Dong,Hongzhan Liu,Chengda Meng,Fengxia Wu,Guobao Xu

Metal-organic gels (MOGs) are a category of metal-organic smart soft materials with large specific surface areas, loose porous structures, and open metal active sites. In this work, trimetallic Fe(III)Co(II)Ni(II)-based MOGs (FeCoNi-MOGs) were synthesized at room temperature via a simple and mild one-step procedure. Fe3+, Co2+, and Ni2+ were the three central metal ions in it, while 1,3,5-benzenetricarboxylic acid (H3BTC) served as the ligand. The solvent enclosed in it was then removed by freeze-drying to get the corresponding metal-organic xerogels (MOXs). The as-prepared FeCoNi-MOXs have excellent peroxidase-like activity and can significantly enhance luminol/H2O2 chemiluminescence (CL) by more than 3000 times, which is very effective compared with other reported MOXs. Based on the inhibitory effect of dopamine on the CL of the FeCoNi-MOXs/luminol/H2O2 system, a simple, rapid, sensitive, and selective CL method for dopamine detection was established with a linear range of 5–1000 nM and a limit of detection of 2.9 nM (LOD, S/N = 3). Furthermore, it has been effectively used for the quantitative measurement of dopamine in dopamine injections and human serum samples, with a recovery rate of 99.5–109.1%. This research brings up prospects for the application of MOXs with peroxidase-like activity in CL.
