As a common macromolecular carbohydrate, pectin has a strong affinity for Pb 2+ . An ethylenediamine modified pectin (EP 48 ) with 48 % of amidation was prepared and exhibited great removal efficiency towards Pb 2+ in our previous study. However, the EP 48 has drawbacks in adsorption including low mechanical strength and difficulty in separation. In this study, EP 48 was compounded with sodium alginate (Alg) and Fe 3 O 4 to synthesize EP 48 /Alg/Fe 3 O 4 microsphere. The physicochemical properties and Pb 2+ adsorption characteristics of microsphere were analyzed. It was found that the microsphere exhibited good thermal stability, mechanical strength, porous structure, as well as acid tolerance. The pseudo-second-order model well described the kinetics of adsorption process, indicating the chemical adsorption is dominant. The Langmuir model fitted the experimental data well, and the maximum adsorption capacity reached 175.19 mg/g. Adsorption-desorption experiments showed that the removal rate of the microsphere maintained over 98.9 % after 10 cycles. The X-ray photoelectron spectroscopy (XPS) analyses revealed that the potential adsorption mechanism included ion-exchange and chelation. The above results suggested its potential use for the removal of Pb 2+ from wastewater.

Tea is a popular beverage with a long history of safe and healthy use. Tea polysaccharide is a bioactive component extracted from tea, which has attracted more and more attention in recent decades. In this article, an acidic polysaccharide Gougunao tea polysaccharide (GPS) was isolated from Gougunao green tea by hot water extraction and ethanol precipitation. After purification by a diethylaminoethyl (DEAE) Sepharose Fast Flow column and a Sephacryl S-400 column, several homogalacturonan (HG) and rhamnogalacturonan-I (RG-I) fractions were obtained. Fraction GPS2b with the highest yield was selected for structural characterization by methylation and nuclear magnetic resonance (NMR) analysis. GPS2b was found to be an HG-type pectic polysaccharide (degree of methyl esterification [DE], 51.6%) with low molecular weight (Mw, 36.8 kDa). It was mainly composed of →4)-α-GalpA- (1→ and →4)-α-GalpA-6-OMe-(1→. In addition, a minor highly branched RG-I domain was identified in this fraction. The investigation of structural features of tea polysaccharides can provide insights to understand their structure-bioactivity relationship.

In the present study, a novel simple and sensitive method was developed for the determination of pectin based on the decarboxylation derivatization reaction and surface-enhanced Raman scattering (SERS) without complicated separation steps. The derivatization reaction can be controlled by the experimental parameters such as reaction time, temperature and the amount of hydrochloric acid. Additionally, the method was used to accurately and reliably detect pectin added in honey or apple, which can be detected at levels as low as 0.5 or 0.01 ppm, respectively. Based on the conventional decarboxylation reaction, a simple and sensitive SERS method was proposed for the detection of pectin, which shows potential for practical application.

Deacetylated konjac glucomannan (DKGM)/calcium pectin (Ca-PEC) composite films were prepared by calcium hydroxide to realize deacetylation of konjac glucomannan (KGM) and the crosslinking of pectin at the same time. Fourier transform infrared spectroscopy (FTIR) indicates that there are intermolecular hydrogen bond and crosslinking structure between DKGM and Ca-PEC. The physical properties of the composite films are characterized by mechanical properties, water vapor permeability (WVP), swelling degree (SD), water solubility (WS), thermogravimetry (TG) and Opacity. The results exhibit that deacetylation of KGM and crosslinking structure of pectin reduce the swelling degree and water solubility of the films, as well as improve its thermal stability and tensile strength. These are attributed to enhanced intermolecular interactions, more compact entanglement of molecular chains and more solid spatial network structures. When pectin content is 40%, the combination property of DKGM / Ca-PEC composite film is the best.

Traditional aerogels lack specific functional groups for the adsorption of Pb2+, which results in a low adsorption capacity and limits the application scope. Novel porous pectin-based aerogels (PPEAs) were prepared by incorporating polyethylenimine (PEI) using ethylene glycol diglycidyl ether (EGDE) as a cross-linker for the removal of Pb2+from water. The cross-linking mechanism, morphology, mechanical strength, thermal stability, adsorption properties, and mechanism of the aerogels were investigated. The aerogels possessed several desirable features, such as a large maximum Pb2+adsorption capacity (373.7 mg/g, tested at pH 5.0), ultralight (as low as 63.4 mg/cm3), high mechanical strength (stress above 0.24 MPa at 50% strain), and easy recyclability. Meanwhile, the equilibrium adsorption data was well described by the Langmuir–Freundlich (Sips) model and the kinetic adsorption process was well fitted using the pseudo-second-order model. The donor groups, such as -NH2, and oxygen-containing functional groups were responsible for the Pb2+adsorption, which was confirmed by the FTIR and XPS analysis. The excellent characteristics mean that PPEAs are highly effective adsorbents in the remediation of lead-containing wastewater.Keywords:pectin;PEI;EGDE;aerogel;Pb2+;adsorption

Furfural is an important platform chemical that can be produced by dehydration of pentose, which is an important building block of lignocellulosic biomass. In this study, xylose was chosen as the reaction substrate, and a carbon-based solid acid prepared from pectin was used as the catalyst in a mixed solvent consisting of water and γ-valerolactone (GVL). FTIR results showed that the sulfonated catalyst had typical characteristic peaks of sulfonic groups, while the results based on NH3-TPD and elemental analysis showed that the sulfonation temperature changed the surface acid concentration and acid site intensity of the catalyst. A furfural yield of 80.4% and a xylose conversion of 100% were obtained at 170 °C for 60 min. The conversions and product yields were influenced by the reaction temperature, reaction time, solvent composition, and catalyst acidity. Different sulfonation temperatures can change the acidity of the catalyst, which determines the catalytic performance of the catalyst.

This work aimed to extract gelatinous chicory root pectin (CRP) and evaluated the rheological behavior of the dispersions and gels. CRP was extracted by citric acid (CEP), alkaline (AEP), ammonium oxalate (OEP) and sodium citrate (SEP). The yield, molecular weight (Mw) and the degree of esterification (DE) of pectin samples varied from 8.8 to 14.8% (w/w), 204 to 336 k Da and 4.0 to 47.4%, respectively. AFM studies showed self-organize on mica of CEP, revealing a random coil conformation due to the interaction of multiple branching, whereas, AEP exhibited long linear filamentous structures. The flow behavior study verified the pseudoplastic character of CEP and SEP at 25 °C, while OEP and AEP belonged to dilatant fluid, besides, a closed hysteresis loop was observed when the CEP concentration increased to 1.5%. OEP gel was thermo insensitive and stiff, AEP gel presented most sensitive to calcium ion but more brittle, and SEP was observed a weak syneresis in spite of the poor gelation property. The texture analysis indicated OEP gel had a superior firmness and chewiness. These findings demonstrated that CRP may be attractive as a thickener or gelling agent to modulate textures of sugar-free and calcium content food.

Background Pectin methylesterase (PME) is a hydrolytic enzyme that catalyzes the demethylesterification of homogalacturonans and controls pectin reconstruction, being essential in regulation of cell wall modification. During fruit ripening stage, PME-mediated cell wall remodeling is an important process to determine fruit firmness and softening. Strawberry fruit is a soft fruit with a short postharvest life, due to a rapid loss of firm texture. Hence, preharvest improvement of strawberry fruit rigidity is a prerequisite for extension of fruit refreshing time. Although PME has been well characterized in model plants, knowledge regarding the functionality and evolutionary property of PME gene family in strawberry remain limited. Results A total of 54 PME genes (FvPMEs) were identified in woodland strawberry (Fragaria vesca ‘Hawaii 4’). Phylogeny and gene structure analysis divided these FvPME genes into four groups (Group 1–4). Duplicate events analysis suggested that tandem and dispersed duplications effectively contributed to the expansion of the PME family in strawberry. Through transcriptome analysis, we identified FvPME38 and FvPME39 as the most abundant-expressed PMEs at fruit ripening stages, and they were positively regulated by abscisic acid. Genetic manipulation of FvPME38 and FvPME39 by overexpression and RNAi-silencing significantly influences the fruit firmness, pectin content and cell wall structure, indicating a requirement of PME for strawberry fruit softening. Conclusion Our study globally analyzed strawberry pectin methylesterases by the approaches of phylogenetics, evolutionary prediction and genetic analysis. We verified the essential role of FvPME38 and FvPME39 in regulation of strawberry fruit softening process, which provided a guide for improving strawberry fruit firmness by modifying PME level.

Pectin has a complex structure, and different structures lead to different biological functions. Citrus pectin (PC) has been widely proven to possess immunomodulatory activity. This study compared and analyzed the anti-allergic activities of PC and polygalacturonic acid (PGA) based on intestinal microbiota, metabolomics, and molecular docking. The results showed that dietary interventions with PC and PGA significantly relieved allergic symptoms, intestinal damage, and the contents of specific antibodies IgE and IgG1. Among them, PGA exhibited a significant suppressive effect on histamine and alleviated the Th1/Th2 imbalance caused by food allergies. Furthermore, after fermentation by intestinal flora, the metabolites of both PC and PGA activated the NOTCH/NF-κB pathway through the gut-spleen axis to mitigate food allergies. Interestingly, the linear main chain of PGA was more easily degraded by the intestinal flora, thereby promoting histamine metabolism. In contrast, the complex side chains of PC facilitated the production of co-metabolites by the host microbiota, allowing them to participate in immune regulation. This study investigated the structure-activity relationship of pectin substances, providing a theoretical basis for the high-value utilization of citrus processing byproducts and the development of targeted nutritional intervention strategies.

Self-healing hydrogels have important application in hemostasis and wound repairing. In this research, pectin based self-healing hydrogel was fabricated with conjugated polyphosphate for hemostatic and wound healing applications. The hydrogel formed without any stimulus and hydrogel kept its biocompatibility; at the same time, the hydrogel degraded completely by enzyme and in vivo . The polyphosphate conjugated hydrogel also showed self-healing property and sustained release performance with strong coagulation characteristic. More importantly, the in vivo experiment revealed that the polyphosphate conjugated hydrogel reduced the blood loss and hemostasis time in hemorrhage model; meanwhile, the hydrogel accelerated the wound repairing rate of the open wound by preventing bacterial invasion. Altogether, the PolyP conjugated hemostatic pectin-based hydrogel is a good candidate as wound dressing material applied in clinic or open wound repairing.

To prolong the storage period of blueberry and improve its shelf-life quality, an edible coating based on chitosan was developed, and different contents of pectin were added to the coating. In this study, Jerusalem artichoke residue was used as a source of pectin, which is a byproduct of the processing of inulin. The extracted pectin has a low cost and high quality, which is very suitable for the preservation of coating. The coating was prepared and the chemical properties of the coating were characterized by SEM, XRD, TG, and FTIR. The barrier properties of the coating were analyzed by thickness, water content, solubility, and water vapor permeability. The results showed that the pectin coating exhibited excellent performance in blueberry preservation. Following 16 days of storage, the decay and weight loss rates of blueberry treated with 0.2% pectin coating decreased by 33 and 22%, respectively. Moreover, the organic acid consumption of the coated blueberry slowed and the anthocyanins were better preserved. As a low-cost, safe, and efficient technology, the pectin chitosan composite coating has significant potential in the berries preservation industry. Keywords:Jerusalem artichoke;pectin;coating;blueberry preservation

Pectin with various degrees of esterification (DE) leads to different food processing directions and has the distinct potential for modulating human health. Investigations of pectin–gut microbiota interactions may contribute towards better understanding of the structure–function mechanism. In this study, in vitro batch fermentation (artificial colon model) was used to illustrate the differential impacts of pectin with different DE on the gut microbiota and metabolome of healthy adults. The results indicated that low-esterified pectin L13 showed better-sustained abilities in terms of the diversity of microbiota and promoted the abundance of Clostridiaceae and Lachnospiraceae at family levels, and Bacteroides and Lachnospira at genus levels. High-esterified pectin H121 induced less Enterococcus and Clostridium . Data from untargeted metabolomics revealed the alterations of intracellular metabolites including fatty acids, amino acids, and organic molecules by various types of pectins. Inositol was the unique intracellular metabolite that was significantly upregulated by low-esterified pectin L13. All types of pectins could increase the level of acetic acid, but butyric acid was only enriched by pectin L13.
