Biocompatible cationic pullulan-g-desoxycholic acid-g-PEI micelles used to co-deliver drug and gene for cancer therapy

Lili Chen,Fangling Ji,Yongming Bao,Jing Xia,Lianying Guo,Jingyun Wang,Yachen Li

The greatest crux in the combination of chemotherapy and gene therapy is the construction of a feasible and biocompatible carrier for loading the therapeutic drug and gene simultaneously. Here, a new amphiphilic bifunctional pullulan derivative (named as PDP) synthesized by grafting lipophilic desoxycholic acid and low-molecular weight (1   kDa) branched polyethylenimine onto the backbone of pullulan was evaluated as a nano-carrier for the co-delivery of drug and gene for potential cancer therapy. PDP exhibited good blood compatibility and low cytotoxicity in the hemolysis and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays, respectively. By self-assembly process, the amphiphilic PDP polymer formed cationic core-shell nanomicelles in aqueous solution with an average diameter of 160.8   nm and a zeta potential of approximate 28   mV. The PDP micelles had relative higher drug encapsulation efficiency (84.05%) and loading capacity (7.64%) for doxorubicin (DOX), an effective anti-tumor drug, demonstrating sustained drug release profile and good DNA-binding ability. The flow cytometry and confocal laser scanning microscopy showed that PDP/DOX micelles could be successfully internalized by MCF-7 cells, and presenting higher cytotoxicity against MCF-7 cells than that of free DOX. Furthermore, PDP micelles could efficiently transport tumor suppressor gene p53 into MCF-7 cells, and the expressed exogenous p53 protein induced MCF-7 cells to die. More excitedly, in comparison with single DOX or p53 delivery, the co-delivery of DOX and gene p53 using PDP micelles displayed higher cytotoxicity, induced higher apoptosis rate of tumor cells and blocked more effectively the migration of cancer cells in vitro . In tumor-bearing mice, co-delivery of DOX and p53 also exhibited enhanced antitumor efficacy as compared with single delivery of DOX or p53 alone. In summary, these results demonstrated that it is highly promising to use cationic PDP micelles for effectively co-delivering functional gene and chemotherapeutic agent, and thus improving antitumor efficacy and systemic toxicity.

Developing fisetin-AgNPs incorporated in reinforced chitosan/pullulan composite-film and its application of postharvest storage in litchi fruit

Zhiwen Qi,Pujun Xie,Chen Yang,Xingying Xue,Hongxia Chen,Hao Zhou,Hua Yuan,Guliang Yang,Chengzhang Wang

In this study, Toxicodendron vernicifluum fisetin chelated silver nanoparticles (FT-AgNPs) with outstanding antioxidant and antimicrobial activities were constructed via self-assembly. To surprise, 0.6 wt% FT-AgNPs was compatibly dispersed into the 1:1 chitosan/pullulan (CS/PUL, CP) matrix. The hydrogen bonding and electrostatic interaction between FT-AgNPs and CP, slightly increased the CP thermal stability, and greatly enhanced the tensile strength to 61.2 MPa, water vapor permeability below 20 kg/m 2 •d. Furthermore, after treated with the composite hydrocolloid film (FT-AgNPs/CP), the reactive oxygen species level of the treated Aspergillus niger cells was significantly increased, and the membrane permeability was enhanced. It effectively slowed down the decay of litchi fruit induced by microbial infection under the storage at 25 °C (15 d of the 0.6 % FT-AgNPs/CP treatment vs 9 d of the control). In addition, 0.024 μg/kg Ag + residual in lichi pulp verified the qualified safety of the application of the 0.6 % FT-AgNPs/CP.

Improving the tribocorrosion performance of plasma electrolytic oxidized coatings on AZ31B magnesium alloy using pullulan as an electrolyte additive

Luyao Xu,Dejian Zhang,Huijuan Su,Peng Yu,Yong Wan,Huilai Sun

This study aimed to investigate the enhancement of the corrosion and tribocorrosion performance of AZ31B magnesium (Mg) alloys after plasma electrolytic oxidization (PEO) treatment using pullulan as an electrolyte additive . The effects of different concentrations of pullulan in the electrolyte solutions on the mechanical and corrosion-resistance properties of the PEO coatings were investigated. The microstructures and chemical compositions of the coatings were investigated via scanning electron microscopy, energy dispersive spectrum, and X-ray diffraction. The corrosion resistance of the samples in simulated body fluid was evaluated via potentiodynamic polarization and electrochemical impedance spectroscopy . The tribocorrosion performance was determined by simultaneously recording the change in the friction coefficient and the open circuit potential over time during sliding experiments. The results indicated that the addition of pullulan to the silicate-based electrolyte solutions resulted in an increase in the unit-area adsorptive capacity of the negative ions at the anode/electrolyte interface, which enhanced the compactness of the PEO coating. The PEO coating produced in electrolyte solution containing 1.0 g/L pullulan exhibited the greatest thickness (50 μm), the lowest surface roughness (1.25 μm), the highest corrosion resistance, and the best anti-tribocorrosion performance, all of which corroborated the potential application of PEO-coated Mg alloys as biological implants .

An ecofriendly coaxial antibacterial and anticorrosion nanofiber pullulan-ethyl cellulose embedded with carvacrol coating for protection against marine corrosion

Siwei Wu,Quantong Jiang,Dongxiao Han,Shuai Yuan,Xia Zhao,Jizhou Duan,Baorong Hou

Coaxial electrospun coatings with antibacterial and anticorrosion properties have a marked potential to protect against corrosion in marine environments. Ethyl cellulose is a promising biopolymer for corrosion caused by microorganisms owing to its high mechanical strength, nontoxicity, and biodegradability. In this study, a coaxial electrospun coating loaded with antibacterial carvacrol (CV) in the core and anticorrosion pullulan (Pu) and ethyl cellulose (EC) as a shell layer was successfully fabricated. The formation of core–shell structure was confirmed using transmission electron microscopy. [email protected] coaxial nanofiber had small diameters, uniform distribution, smooth surface, strong hydrophobicity, and no fractures. Electrochemical impedance spectroscopy was used to analyze corrosion of the electrospun coating surface in a medium containing bacterial solution. The results indicated significant corrosion resistance of the coating surface. In addition, the antibacterial activity and mechanism of coaxial electrospun were studied. The [email protected] nanofiber coating exhibited excellent antibacterial properties by effectively increasing the permeability of cell membranes and killing bacteria, as determined by plate counts, scanning electron microscopy, cell membrane permeability, and the activity of alkaline phosphatase. In summary, the coaxial electrospun pullulan-ethyl cellulose embedded with CV coating can be used as antibacterial and anticorrosion materials and may have potential applications in the field of marine corrosion.

A biodegradable pH and glutathione dual-triggered drug delivery system based on mesoporous silica, carboxymethyl chitosan and oxidized pullulan

Qian Rui,Jun Gao,Zheng-Zhi Yin,Junyao Li,Wenrong Cai,Datong Wu,Yong Kong

A simple and smart drug controlled delivery system is developed in this work. Biodegradable mesoporous silica nanoparticles (BMSN) were first synthesized by introducing disulfide during the synthesis of mesoporous silica nanoparticles (MSN), which were used for the loading of methotrexate (MTX), an anti-cancer drug. The MTX loaded BMSN (BMSN-MTX) was then encapsulated in the hydrogels of carboxymethyl chitosan (CMCS)/oxidized pullulan (OPL) generated through Schiff base reaction. The acylhydrazone bonds (–N=CH–) between CMCS and OPL are prone to be hydrolyzed in acidic medium while the disulfide linkage (–S–S–) in the BMSN can be cleaved in the presence of glutathione (GSH), and thus the delivery of MTX from the BMSN-MTX-gel can be triggered by both pH and GSH. The results of release kinetics reveal that the delivery of MTX from the biodegradable hydrogels is controlled by Higuchi model. Finally, good biocompatibility and pronounced cytotoxicity of the developed BMSN-MTX-gel are confirmed by cytotoxicity test .

Preparation and characterization of ultrasound-assisted essential oil-loaded nanoemulsions stimulated pullulan-based bioactive film for strawberry fruit preservation

Arif Rashid,Abdul Qayum,Qiufang Liang,Lixin Kang,Husnain Raza,Zhuzhong Chi,Runhao Chi,Xiaofeng Ren,Haile Ma

Ultrasound is a high-energy approach that can help with homogenization and dispersion in cavitation. In this study, nanoemulsions of curcumin and orange essential oil were prepared with ultrasound treatment at different times. The ultrasound-treated nanoemulsions for 10 min exhibited the smallest droplet size, the best storage, and higher thermal stability. The pullulan-based film with ultrasound-assisted nanoemulsions exhibited improved water vapor permeability and moisture content and the highest tensile strength and elongation at break. The structural analysis showed that ultrasonic treatment enhanced the H-bond, resulting in a more orderly molecular arrangement and intermolecular compatibility. Furthermore, the bioactive film had the maximum oil retention time. It possessed excellent bacteriostatic properties against Escherichia coli and Staphylococcus aureus due to the smallest oil droplets and uniform distribution in the film matrix. Besides, the weight loss and deterioration of the strawberry fruit were effectively reduced, thus prolonging the shelf life.

Cytocompatible, hemocompatible, and bioactive hydrogel fabricated using decellularized tendon, carboxymethyl chitosan and oxidized pullulan with antibacterial and ROS-scavenging properties

Lei Nie,Xiaoyue Ding,Letian Yan,Man Li,Peng Ding,Oseweuba Valentine Okoro,Armin Shavandi

In recent years, hydrogels based on decellularized extracellular matrix (dECM) from tendons have been used in tissue scaffold preparation and guided tissue regeneration. However, pure dECM hydrogel may be limited in biomedical applications due to slow gelation kinetics, poor mechanical properties, and other suboptimal characteristics. To enhance functionality, dECM hydrogel can be remodelled using natural polysaccharide polymers. In this study, carboxymethyl chitosan (CMCS) and oxidized pullulan (OPL) were employed to remodel dECM synthesized from porcine Achilles tendon, resulting in the fabrication of dECM-based composite hydrogels (CLD). Scanning electron microscopy (SEM) images and X-ray computed microtomography (Micro-CT) confirmed the interconnected microstructure of CLD hydrogels. Furthermore, the CLD hydrogels exhibited favorable self-healing ability and adhesion properties. In vitro hemolysis test using the red blood cells demonstrated good hemocompatibility of CLD hydrogels. CLD hydrogels exhibited favorable antibacterial capabilities against Escherichia coli and Staphylococcus aureus, as well as effective reactive oxygen species (ROS) scavenging capabilities and good cytocompatibility. Collectively, these findings suggested that the fabricated dECM-based remodelling hydrogels hold potential in future biomedical applications.

Fabrication and postharvest application of alginate/pullulan composite films integrated with citral-loaded α-CD-MOFs for sustainable cherry storage

Mou Linyun,Li Jianlong,Lu Ya

Cherries are highly susceptible to spoilage after harvest due to physiological and biochemical changes. This poses considerable challenges to food safety. Common fungal pathogens responsible for decay include Aspergillus niger and Cladosporium cladosporioides . Citral, an efficient antifungal agent, is thought to combat these pathogens through interactions with phytase (phyA) and N-myristoyltransferase (NMT), a hypothesis explored through molecular docking simulations. Using the Independent Gradient Model based on Hirshfeld partition (IGMH) analysis, it was found that citral encapsulated within α -cyclodextrin metal-organic frameworks ( α -CD-MOFs) forms strong van der Waals forces and exhibits hydrogen bonding. These interactions enhance the stability of the composite material. Incorporating citral-loaded α -CD-MOFs into a matrix of sodium alginate and pullulan resulted in a novel composite film. This film exhibits significantly improved flexibility and tensile properties, which is beneficial for fruit packaging. Meanwhile, the Higuchi and Ritger–Peppas models both indicate that citral is released by diffusion from a uniform polymeric matrix. When applied to cherry preservation, the film exhibited significantly better cherry preservation at 4 °C than at 30 °C ( P  < 0.05). Simultaneously, the combination of low temperature with the film’s barrier properties and sustained antimicrobial release synergistically inhibited microbial growth and fruit metabolism, effectively delaying spoilage and supporting its application in both cold-chain and ambient-temperature transport.

Antioxidant-antibacterial caffeic acid-derived carbon dots film based on pullulan/chitosan: Preparation, characterization, and application in chicken breast preservation

Jiacheng Chen,Yanzhao Zheng,Yanlan Ma,Jinxue Hou,Guanghui Li,Jingxin Sun,Ziyang Huang,Ming Huang

Synthesized multifunctional carbon dots (CA-CDs) from caffeic acid, exhibiting antioxidant and antibacterial properties. • Developed a novel, fully bio-based food packaging film by incorporating CA-CDs into a pullulan/chitosan matrix. • Extended the shelf life of chilled chicken breast by 4 days by effectively inhibiting microbial growth and oxidation. Caffeic acid-derived carbon dots (CA-CDs) were synthesized and characterized by their potent antioxidant capacity, broad-spectrum antibacterial activity, and favorable cytocompatibility. SEM analysis further elucidated the antibacterial mechanism, revealing that CA-CDs induce irreversible bacterial membrane disruption. Subsequently, these bioactive CA-CDs were integrated into a pullulan/chitosan (Pul/CS) matrix to fabricate multifunctional composite films (Pul/CS/CA-CDs). Incorporation of CA-CDs at different loadings (0.025%, 0.05%, and 0.1% w/v) led to progressively enhanced functional properties of the films, with the highest loading (Pul/CS/CA-CDs 3 ) producing the most pronounced improvements. In particular, compared with the pristine Pul/CS film, the addition of CA-CDs significantly enhanced UV-blocking ability, mechanical strength, and water vapor barrier performance ( p  < 0.05). The Pul/CS/CA-CDs3 film achieved ABTS and DPPH radical scavenging efficiencies of 92.65% and 90.53%, respectively, indicating strong antioxidant performance. It also showed superior antibacterial activity against Escherichia coli and Staphylococcus aureus . During chicken breast storage at 4 °C for 12 days, Pul/CS/CA-CDs 3 packaging significantly inhibited spoilage compared with the control, reducing total viable counts by 1.41 log CFU/g and markedly decreasing TVB-N (14.31 mg/100 g), TBARS (0.55 mg MDA/kg), and carbonyl content (2.37 nmol/mg protein). These changes indicate effective suppression of microbial proliferation and oxidative deterioration, resulting in improved meat preservation. The results confirm that CA-CDs-reinforced Pul/CS films act as effective active packaging materials for meat preservation, where their antioxidant, antimicrobial, and barrier properties synergistically suppress microbial proliferation and oxidative deterioration during storage. Enhanced hydrophobicity, antioxidant activity, and antimicrobial efficacy of edible starch-based films incorporating pullulan and geraniol for fresh salmon preservation

Youning Zou,Jiahao Ren,Yiting Fan,Yifang Cao,Yijia Xie,Xinglong Xiao,Fengsong Liu

The development of sustainable food packaging materials remains a critical challenge due to inherent limitations in hydrophobicity, antioxidant activity, and antimicrobial efficacy. Herein, a novel starch-based film incorporating pullulan and geraniol was designed to overcome these shortcomings through hydrogen bonding and intermolecular entanglement. Structural analyses using Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) confirmed strong molecular interactions, while scanning electron microscopy (SEM) and light transmittance studies demonstrated enhanced compatibility and improved optical properties. Notably, the Starch-Pullulan-Geraniol film exhibited superior hydrophobicity, with a contact angle of 68.11°, reduced water solubility (37.56 %), and minimal water vapor permeability (1.35 ×10⁻ 10 g·m⁻ 1 ·s⁻ 1 ·Pa⁻ 1 ). Mechanical performance was also enhanced, achieving a reduced Young’s modulus of 130.34 MPa, a tensile strength of 22.66 MPa, and a maximum elongation at break of 52.81 %. Furthermore, the incorporation of pullulan and geraniol conferred remarkable antimicrobial and antioxidant properties, significantly improving the film’s effectiveness in food preservation. When applied to salmon storage, the film successfully maintained appearance, lipid stability, and microbial safety, highlighting its potential as an advanced bio-based packaging material. These findings underscore the Starch-Pullulan-Geraniol film as a promising alternative to conventional packaging, offering enhanced functionality for extending food shelf life.

Advancing aqueous zinc-ion battery performance with pullulan through preferential adsorption on zinc anode surfaces and enhanced zinc ion desolvation

Jin Bao,Wenping Zeng,Yun Huang,Zhongwei Zhao,Jiajun He,Zhanpeng Du,Fengliang Wang,Huihui Li,Heguo Zeng,Xing Li,Mingshan Wang,Yuanhua Lin,Bingshu Guo,Bo Yu,Haijun Cao

Aqueous zinc-ion batteries (AZIBs) have emerged as a prominent contender in energy storage devices, largely due to their substantial theoretical capacity, and competitive cost. Nevertheless, the uncontrollable proliferation of zinc dendrites and the emergence of parasitic side effects present a significant challenge to the practical implementation of AZIBs. Herein, a novel low-cost and environmentally friendly electrolyte additive, pullulan polysaccharide (PLL), was incorporated into a common ZnSO 4 electrolyte to stabilize the Zn anode of high-performance AZIBs. Experimental and theoretical calculations demonstrated that PLL can modulate the interaction of Zn 2+ with H 2 O molecules, reducing the number of H 2 O molecules around Zn 2+ and thereby facilitating the desolvation of Zn 2+ . Furthermore, the PLL molecules adsorbed on the surface of the zinc anode formed a protective layer that permitted the uniform deposition of Zn 2+ . As anticipated, the cycle life of the Zn//Cu asymmetric cells using 2M ZnSO 4 + 1 wt% PLL as the electrolyte exceeded 3700 cycles, with a high average Coulombic Efficiency of 99 %. The Zn//Zn symmetric cell with PLL as the electrolyte exhibited excellent cycling stability, demonstrating prolonged operational longevity that exceeded 1200 h. Furthermore, full cells with PLL additive displayed enhanced long-term durability when coupled with sodium vanadate (NVO) cathode.

A Study of Hydrophobically Modified Pullulan Nanoparticles with Different Hydrophobic Densities on the Effect of Anti-Colon Cancer Cell Efficiency

Zhang Yi,Jiang Qing,Liu Xinyi,Peng Liping,Tang Xinyi,Li Ling,Ling Xiao,Yang Xiaoping,He Chunlian,Tao Xiaojun,Hou Defu

To discuss the effect of hydrophobic groups of a polymer on the structural properties and function of polymer nanoparticles (NPs), we grafted chenodeoxycholic acid (CDCA) with pullulan (PU) to form hydrophobically modified PU (PUC). Three PUC polymers, namely, PUC-1, PUC-2, and PUC-3, with different degrees of substitution were designed by changing the feed ratio of CDCA and PU. 1H-NMR spectra showed that the PUC polymer was successfully synthesized, and the degrees of hydrophobic substitution for PUC-1, PUC-2, and PUC-3 were calculated to be 10.66%, 13.92%, and 16.94%, respectively. The PUC NPs were prepared by the dialysis method and were shown to be uniformly spherical by transmission electron microscopy (TEM). The average sizes were about (220±10) nm, (203±7) nm, and (163±6) nm under dynamic light scattering (DLS) for PUC-1 NPs, PUC-2 NPs, and PUC-3 NPs, respectively. Drug release experiments showed that the three PUC/DOX NPs exhibited good sustained release. At 48 h, the IC50 of doxorubicin in inhibiting colon cancer HCT116 cells was 0.0904 μg/mL. A cell study showed that PUC-3/DOX NPs had the highest uptake efficiency by HCT116 cells with the most cytotoxicity and inhibited the migration of HCT116 cells with the highest efficiency. The structural properties and function of polymer NPs were closely related to the hydrophobic groups in the polymer, and NPs with higher hydrophobicity showed a smaller size, higher drug capacity, and greater cell efficiency.
