Effect of Fatty Acid Chain Length on the Crystallization Behavior of Trans-free Margarine Basestocks during Storage

Peng Hu,Xuebing Xu,Liangli (Lucy) Yu

In order to obtain margarine free of trans-fatty acids, four interesterified basestocks were prepared by chemical interesterification (CIE) of oil blends. Different ratios of palm stearin, palm olein and soybean oil were mixed without and with 1) fully hydrogenated Acer truncatum oil (FHATO), 2) fully hydrogenated rapeseed oil or 3) palm kernel oil containing a similar amount of saturated, monounsaturated and polyunsaturated fatty acids, but different saturated fatty acid length for CIE. Compared to the physical blends, the CIE samples demonstrated lower slip melting points and decreased solid fat contents, especially at high temperatures, indicating that the CIE samples might have improved mouthfeel. In all CIE samples, the β crystal form disappeared and only the β’ crystal form was observed, except for sample 2, which contained a mixed β and β’ forms. Furthermore, in all CIE samples, except sample 1, the β' crystal forms began transforming to β form after only two cycles of higher temperature treatments indicating that the CIE sample with FHATO had the most resistance to temperature fluctuation during storage which may be attributed to its longer saturated chains. In conclusion, the CIE basestocks containing longer saturated fatty acids could be more suitable for margarine use.

Cis-trans isomerism-driven thermal and phase transitions in oleic and elaidic acids and their derived polymers

Minghao Wang,Haomin Yu,Jiawei Jiang,Wencong Xu,Guofeng Xu,Si Wu

Cis-trans isomerism plays a crucial role in regulating the thermal properties and phase transitions of materials, which is essential for the development of advanced functional materials. By influencing melting points and glass transition temperatures, cis-trans isomerism enables precise control over material performance in applications such as thermal sensors, self-healing coatings, thermal sensors, and temperature-responsive films. Here we report the impact of cis-trans isomerism in double bonds on the thermal and phase transitions of oleic acid (OA) and elaidic acid (EA) and their derived polymers, with a focus on glass transition temperature ( T g ) and melting point ( T m ). A series of molecules and copolymers with varying cis and trans double bond ratios are synthesized to examine how these isomeric configurations affect thermal behavior. Thermal and structural properties are characterized using differential scanning calorimetry (DSC) and nuclear magnetic resonance (NMR) spectroscopy. The results show a clear correlation between isomeric composition and the polymers' T g and T m , illustrating that the incorporation of cis-trans isomers can be effectively used to fine-tune thermal properties. This research offers valuable insights into the design of advanced polymeric materials with tailored thermal characteristics, paving the way for applications in various high-performance sectors.

A Triple Shape Memory Material of Trans-Polyisoprene/Polycaprolactone with Customizable Response Temperature Controlled by Crosslinking Density

Wenzhe Zhang,Chenglong Zou,Qizhu Pan,Guixin Hu,Huihong Shi,Yi Zhang,Xiyang He,Yiheng He,Xiucheng Zhang

Body-temperature responsive shape memory polymers (SMPs) attract a lot of attention with the application in biomedicine. Multiple SMPs regulate the response temperature through the proportion of their stationary and reversible phases by changing the molecular weight of the polymer or the components of the composite. Herein, a crosslinking reaction of trans-polyisoprene (TPI) and polycaprolactone (PCL) initiated by benzoyl peroxide gave a SMP of TPI/PCL, whose shape memory property is realized by stage-division crystallization zone. Under the fixed ratio of TPI:PCL, the crystallization performance of TPI/PCL is controlled by the crosslinking density. The response temperatures of the components in TPI/PCL are selected for the stage-division crystallization zone, making excellent shape memory performance at corresponding response temperatures. For the further achievement in controllable response temperature within a certain range, the relationship model between crosslinking density and response temperature is established, obtaining the customizable response temperature ranging 37–55 °C. The presented facile regulation method for response temperature will provide a new idea for the SMPs development.

Stereoselective Synthesis and Chain Microstructure Control of Trans-1,4-polyisoprene Enabled Via TiCl4/MgCl2 Ziegler-Natta Catalyst

Zhou Jing,Zhang Gongguo,Si Chongdian,Jin Manman,He Aihua,Niu Qingtao

Series of isoprene polymerizations with different polymerization time were performed by TiCl 4 /MgCl 2 type Ziegler-Natta catalysts. Polymerization kinetics showed catalytic activity had two-stage polymerization feature and the polymer chain microstructure gradually changed from the trans -1,4 and cis -1,4 mixed structure with low molecular weight ( M w ) as well as broad molecular weight distribution ( M w / M n ) at the early stage, to the final high trans -1,4 structure with high M w and narrow M w / M n . The high trans -1,4 units can enhance the tensile strength and hardness of the synthetic polyisoprenes, which are widely used for golf ball cover, medicinal material and shape memory material. Polymers were mainly isolated into Fraction C, D and G by the stepwise isothermal crystallization fractionation method. Fraction C and D had high trans -1,4 content, high trans -1,4-[II] dyad sequence concentration, high trans - trans unit linkage content and high crystallinity, which were generated by high trans -1,4 stereoselective active centers. While G fraction had low trans -1,4 content, low trans -1,4-[II] dyad sequence concentration, low trans - trans unit linkage content and low crystallinity, which were formed by the active center with low trans -1,4 stereoselectivity. The stereoselective transformation of active centers as well as trans -1,4 stereospecific polymerization mechanism of isoprene with TiCl 4 /MgCl 2 -AlEt 3 catalyst system was discussed. Graphical

trans-4-[4-(Dimethylamino)styryl]-1-methylpyridinium iodide@cyclopentanocucurbit[6]uril as a fluorescent probe for anion recognition

Cheng Si Yuan,Yang Xi Nan,Tao Zhu,Wei Lian Tong,Zhao Wei Wei,Jiang Dao Fa,Jin Yan Mei,Gao Jie,Ma Pei Hua

Abstract The host–guest binding characteristics of cyclopentanocucurbit[6]uril (CyP 6 Q[6]) and cyanine dye trans -4-[4-(dimethylamino)styryl]-1-methylpyridinium iodide ( t -DSMI) have been studied. The results show that CyP 6 Q[6] and t -DSMI form a 1:1 inclusion system, generating strong fluorescence. On this basis, the fluorescent probe t -DSMI@CyP 6 Q[6] has been constructed, and its anion recognition ability has been investigated. In a neutral environment, BF 4 − , H 2 PO 4 − , and I − elicit obvious specific responses from the probe, since they have stronger binding forces. This probe shows greater sensitivity than the previously reported probe t -DSMI@HMeQ[6]. Graphic abstract Since the complex formed by CyP 6 Q[6] and t -DSMI can produce strong fluorescence at pH > 4.6, t -DSMI@CyP 6 Q[6] was constructed to recognize the anions at pH  ≅  5. The results show that in a near neutral environment, BF 4 − , H 2 PO 4 − , and I − elicit obvious specific responses from the probe.

Real-time measurement of the trans-epithelial electrical resistance in an organ-on-a-chip during cell proliferation

Jinlong Liu,Wenjie Zhao,Meiyan Qin,Xiaofeng Luan,Yuang Li,Yang Zhao,Chengjun Huang,Lingqian Zhang,Mingxiao Li

The trans-epithelial electrical resistance (TEER) is widely used to quantitatively evaluate cellular barrier function at the organ level in vitro. The measurement of the TEER in organ-on-chips (organ chips) plays a significant role in medical and pharmacological research. However, due to the limitation of the electrical equivalent model for organ chips, the existing TEER measurements usually neglect the changes of the TEER during cell proliferation, resulting in the low accuracy of the measurements. Here, we proposed a new whole-region model of the TEER and developed a real-time TEER measurement system that contains an organ chip with a plate electrode. A whole region circuit model considering the impedance of the non-cell covered region was also established, which enables TEER measurements to be independent of the changes in the cell covered region. The impedance of the non-cell covered region is here attributed to the resistance of the porous membrane. By combining the real-time measurement system and the whole region model, subtle changes in cellular activity during the proliferation stage were measured continuously every 6 minutes and a more sensitive TEER response was obtained. Furthermore, the TEER measurement accuracy was also verified by the real-time measurement of the TEER with stimulation using the permeability enhancer ethylene glycol-bis(2-aminoethylether)-N,N,N′,N′-tetraacetic acid (EGTA). The obtained results indicated that the new proposed whole region model and the real-time measurement system have higher accuracy and greater sensitivity than the traditional model.

Dual-Responsive Micelles Co-delivering All-Trans Retinoic Acid and Paclitaxel for Enhanced Eradication of Osteosarcoma Stem Cells

Zhiyu Chen,Junhong Chen,Shuang Liu,Xu Zhou,Yichuan Zhang,Yunsheng Jiang,Xiao Liu,Yuan Zhang,Xiangling Pu,Hui Zhang

Osteosarcoma is the most prevalent primary bone cancer affecting children and adolescents, characterized by its high invasiveness and the challenges in treatment posed by multidrug resistance. Cancer stem cells significantly contribute to poor clinical outcomes; however, current targeted therapies remain limited in their effectiveness. In this study, we developed a novel dual-responsive nanomedicine designed to target the microenvironment of osteosarcoma cancer stem cells (OCSCs) and osteosarcoma itself, which selectively releases all-trans retinoic acid and paclitaxel, termed mPCDAP. Our findings indicate that mPCDAP rapidly releases all-trans retinoic acid to suppress stemness under conditions of elevated glutathione, subsequently increasing intracellular reactive oxygen species levels in tumor cells. This mechanism further facilitates the release of paclitaxel under conditions of highly reactive oxygen species, thereby inducing apoptosis in osteosarcoma cells. Additionally, mPCDAP was effectively internalized by both osteosarcoma cells and OCSCs, with in vitro and in vivo results demonstrating significant synergistic antitumor effects, promoting apoptosis and markedly reducing tumor stemness. This study presents a novel approach and promising prospects for targeted therapy of osteosarcoma and OCSCs.

Echinacoside alleviates type 2 diabetes mellitus through inhibiting hepatic gluconeogenesis via gut bacterial-fungal trans-kingdom network reconstruction

Li Fan,Jian Liu,Lin Li,Xueping Yang,Qingwei Zhao,Lijuan Zhao

Background Echinacoside (ECH), a natural phenylethanoid glycoside, has demonstrated protective effects against type 2 diabetes mellitus (T2DM). However, the mechanism underlying the low bioavailability yet advantageous anti-diabetic of ECH remains unresolved. Purpose To elucidate the mechanism of ECH against T2DM through gut microbiota-mediated host metabolism for the first time. Study design and methods A T2DM mouse model was established using a high-fat diet in combination with streptozotocin injection. The therapeutic effects of ECH against T2DM were evaluated by measuring fasting blood glucose (FBG), insulin resistance, glucose intolerance, blood lipids and organ damage in mice. Fecal 16S rRNA and ITS sequencing techniques were employed to characterize the composition of gut microbiota, followed by analysis of bacterial-fungal trans-kingdom network. Metabolomics was conducted to assess the ECH-induced metabolite profile alterations. Additionally, the predicted mechanism of ECH on T2DM was investigated through measuring the hepatic gluconeogenesis markers and inflammation by western blotting, immunohistochemistry, enzymatic assays and antimicrobial mixture (ABX) experiments. Results ECH exhibited significant protective effects against T2DM, as evidenced by reductions in FBG and fasting insulin levels, improvements in glucose and insulin tolerance, attenuations of hyperlipidemia, and alleviation of liver, kidney, and colon damage in T2DM mice. Furthermore, ECH modulated gut microbiota by decreasing the abundances of conditional pathogenic intestinal bacteria ( Klebsiella and Escherichia-Shigella ) and fungi ( Debarymoyces ), while increasing beneficial bacteria ( Lactobacillus ) and fungi ( Wallemia and Penicillium ). Moreover, ECH could restore the disrupted trans-kingdom network between gut fungi and bacteria, thereby suppressing the inflammation-mediated hepatic gluconeogenesis via downregulation of FBP1, PCK1 and G6PC expression. Correspondingly, ABX experiments indicated that once the regulatory function of gut microbiota imbalance was blocked, the anti-T2DM effects of ECH were weakened, accompanied by a failure to improve the levels of inflammation and key gluconeogenic markers in T2DM mice. Conclusion This study presents novel evidence indicating that ECH alleviates T2DM through inhibiting hepatic gluconeogenesis via gut bacterial-fungal trans-kingdom network reconstruction. These findings suggest that ECH may serve as a promising therapeutic strategy for T2DM management, providing new insights for the prevention and treatment of clinical diabetes and its complications.

Isodimorphic-Like Crystallization and Mechanical Properties of Unsaturated Poly(butene succinate): Crucial Role of Cis/Trans Isomeric Contents

Wangkai Xiang,Lingling Ni,Xing Zhang,Jing Li,Jian Zhou,Ying Zheng,Chengtao Yu,Bao Wang,Junfeng Liu,Pengju Pan

Geometric configuration of C═C bonds along the polymer backbone plays a crucial role in determining the physical properties of the resulting materials. However, the key role of cis / trans -C═C units in the crystallization and physical property of stereoisomeric polymers has not been well understood. Herein, we report a unique series of linear unsaturated polyesters, poly(butene succinate) (PBuS), with the precisely controlled cis / trans -C═C unit contents synthesized via melt polycondensation. High-molecular-weight cis / trans -polyesters without detectable isomerization or saturation of C═C bonds were prepared in the used polycondensation method. We find that the synthesized PBuS was crystallizable across the entire range of cis / trans unit ratios and showed the unique isodimorphic-like crystallization behavior, with an asymmetric pseudo-eutectic point at ∼31% cis -unit content. Notably, cis -PBuS crystallized much faster than its trans -counterpart, highlighting the profound effect of C═C isomerism on crystallization behavior. Mechanical properties of PBuS correlated strongly with the crystallinity and crystalline structure, showing low strength and modulus but high flexibility around the pseudo-eutectic composition. This work elucidates the crucial role of cis / trans isomerism in polymer crystallization and provides a rational way for designing the aliphatic polyesters with tailored physical properties.

Unveiling the Trans-Cis Isomerization Mechanism in Alicyclic PA10C: the Vital Role of Water Cluster and Hydrogen Bonds

Chen Chen,Xiaoyue Yuan,Chunlei Zhu,Zonglin Qiu,Yang Wang,Xin Li,Yuancheng Zhang,Xiaomeng Zhang,Wei Zhao,Peng Fu,Zhe Cui,Xinchang Pang,Minying Liu

Stereoisomerism exerts critical influence on the physicochemical properties and biofunctionality. Precise regulation of stereoisomeric composition enables programmable manipulation of polymeric aggregation states and macroscopic properties. Herein, the trans – cis isomerization mechanism inherent to alicyclic PA10C was comprehensively deciphered through an integrated experimental and computational methodology. Primarily, regulated polymerization was conducted under varied thermal regimes within a reactor environment to strategically manipulate the stereoisomeric ratio. Experimental results demonstrated that isomerization transpires at temperatures as low as 170°C, with trans -PA10C as the predominant configuration. Subsequently, the energy barrier of trans- to -cis isomerization of PA10C was calculated using Density Functional Theory (DFT) to be 72.9 kcal/mol. This suggests that configurational inversion necessitates thermal activation exceeding 300°C, exhibiting a substantial discrepancy with experimental observations at 170°C. Theoretical simulations further corroborated that H 2 O markedly reduced the activation energy to 29.2 kcal/mol, as water cluster facilitate rapid proton transfer through dynamic hydrogen bond during polymerization. Furthermore, trans -PA10C macromolecules adopt a centrosymmetric structure, which facilitates dense packing into crystalline regions with an energy barrier of 7.6 kcal/mol. Therefore, trans -to- cis isomerization requires overcoming the hydrogen bond energy, thereby resulting in the thermodynamic predominance of the trans -configuration. Ultimately, stereoisomerism dynamics of cis/trans -PA10C was carried out, and their characteristic critical isomerization temperature were determined to be 204°C and 300°C, respectively.

Thymol and trans-cinnamaldehyde induce apoptosis through a metacaspase-dependent mitochondrial pathway in food-spoilage yeast Zygosaccharomyces rouxii

Huxuan Wang,Zhonghua Peng,Cong Wang,Yanan Zhu,Fei Xia,Hongmin Sun

This work aimed for the first time to provide detailed insights into thymol and trans -cinnamaldehyde's mechanisms of action on the food-spoilage yeast Zygosaccharomyces rouxii and offers evidence in favor of the activation of an apoptosis-like phenotype. The action mechanisms of thymol and trans -cinnamaldehyde were investigated by the measurement of a series of typical apoptotic features using flow cytometer or microplate reader. Moreover, quantitative reverse transcription PCR (QRT-PCR) was performed to investigate the effects of thymol and trans -cinnamaldehyde on the transcription of key regulators of apoptosis in Z. rouxii . The results indicated that the treatment of Z. rouxii with thymol or trans -cinnamaldehyde (minimum inhibitory and subinhibitory concentrations) triggered reactive oxygen species (ROS) accumulation, elevated intracellular Ca 2+ level, and depolarized mitochondrial membrane potential (MMP) coupled with hallmarks of apoptosis including mitochondrial cytochrome c (cyt c) release, metacaspase activation, phosphatidylserine (PS) exposure, and DNA fragmentation. Moreover, thymol or trans -cinnamaldehyde treatment upregulated the transcription of proapoptotic regulators including Yca 1, Dnm 1, Nuc 1, Ndi 1, and Mmi 1 and downregulated the transcription of antiapoptotic regulators of Fis 1 and Cdc 48, further confirming the apoptosis induced by thymol and trans -cinnamaldehyde in Z. rouxii . In summary, thymol and trans -cinnamaldehyde probably induced apoptosis through a metacaspase-dependent mitochondrial pathway in Z. roxuii . These findings provide theoretical support for the development of safe and efficient agents used in food preservation. Practical Application The results will provide a new idea for the systematic analysis of the antifungal mechanisms of thymol and trans -cinnamaldehyde and also provide a theoretical support for the development and application of natural food preservatives, which is of positive significance for the effective control of the spoilage caused by Z. rouxii in food processing and storage and the protection of consumers’ health.

C/EBPα promotes porcine pre-adipocyte proliferation and differentiation via mediating MSTRG.12568.2/FOXO3 trans-activation for STYX

Weimin Lin,Lei Chen,Wenjing Meng,Kai Yang,Shengjuan Wei,Wei Wei,Jie Chen,Lifan Zhang

As a key adipogenic marker, C/EBPα (CCAAT/enhancer binding protein α) is also an important factor in regulating targets containing CCAAT element for transcription, whose products include coding and non-coding RNAs (ncRNAs). However, knowledge of the mechanism of C/EBPα affecting pre-adipocyte proliferation and adipogenesis through regulating ncRNA is still limited. In this study, we firstly conducted an investigation concerning the impact of C/EBPα knockdown on porcine pre-adipocytes by using RNA sequencing (RNA-Seq) to identify the role of key ncRNAs, especially lncRNAs and their correlated mRNAs in regulating proliferation and differentiation of porcine pre-adipocytes. 97 differentially expressed (DE) mRNAs and 4 DE lncRNAs were identified in si-C/EBPα groups compared with the si-NC groups. Meanwhile, we found C/EBPα directly target the promoter of a novel lncRNA , namely MSTRG.12568.2 , which was trans -correlated with STYX (serine/threonine/tyrosine interacting protein), an important candidate gene for regulating cell proliferation . Moreover, FOXO3 (forkhead box O3) was identified as a co-regulator with MSTR.12568.2 for STYX . Overexpression and knockdown of any of the MSTRG.12568.2 , STYX , and FOXO3 increased and decreased the levels of pre-adipocyte proliferation and differentiation, respectively, which demonstrated that they played a positive role in adipogenesis of pre-adipocytes. Furthermore, our results revealed that FOXO3 was necessary for MSTRG.12568.2 to trans -activate STYX . We revealed that C/EBPα regulated pre-adipocyte proliferation and differentiation through mediating trans -activation of MSTRG.12568.2 -FOXO3 to STYX . These results provide a novel regulation signal for C/EBPα to influence porcine pre-adipocyte proliferation and differentiation and greatly benefit to our understanding of molecular mechanism regulating subcutaneous adipogenesis.
