Improved mechanical and antimicrobial properties of zein/chitosan films by adding highly dispersed nano-TiO2

Liangfan Qu,Guiyun Chen,Shuang Dong,Yaoxing Huo,Zhonglin Yin,Shuhong Li,Ye Chen

Highly dispersible TiO 2 nanoparticles were prepared through the hydrolysis of titanium alcohol, and a series of TiO 2 -modified zein/chitosan (CS) films were prepared through a process involving blending, solution casting and evaporation. The mechanical properties and the antibacterial capacity of the zein/CS/TiO 2 films were characterized to explore the effects of different TiO 2 nanoparticle contents. Results showed that the addition of nanoscale TiO 2 remarkably improved the mechanical properties and thermal stability of the zein/CS/TiO 2 film. The tensile strength of the zein/CS/TiO 2 films significantly increased (p<0.05) with the increasing TiO 2 loadings in films, reaching a maximum of 28.33 ± 1.53 MPa at 0.15 wt % TiO 2 nanoparticles. The antibacterial effect of the composite films on Gram-positive bacteria ( S. aureus ) was more significant than that of Gram-negative bacteria ( E. coli and S. enteritidis ), and the antibacterial effect of UV light was better than that of dark conditions. In particular, the inhibition zones of zein/CS/TiO 2 (0.15%) film against E. coli , S. enteritidis and S. aureus under UV light condition increased by 21.78%, 21.45%, and 26.44%, respectively, compared with the control. This study suggested that adding highly dispersible TiO 2 nanoparticles is an effective method for improving the mechanical and antimicrobial properties of zein/CS films for various applications, such as medical and food packaging.

Development of a resveratrol–zein–dopamine–lecithin delivery system with enhanced stability and mucus permeation

Huang Wenquan,Li Shaomin,Li Zhixian,Zhu Wei,Lu Shan,Jiang Yanbin

The development of multi-functional nanoparticles is important in overcoming the poor stability and low mucus permeation of traditional drug delivery systems. Hence, the encapsulation of a model drug, resveratrol (RES), into zein–polydopamine–lecithin (ZPLs) nanoparticles was investigated through a delicately controlled phase separation process. The surface of the nanoparticles obtained was successfully modified with lecithin, which endowed them with neutral and hydrophilic properties. Due to the strong charge repulsion of the particles, the fabricated ZPLs showed high stability under the conditions of a saline solution, various pH, long-term storage and high-speed centrifugation. In vitro study suggested that the RES exhibited remarkably sustained release performance after loading into ZPLs. The quantificational experiment of the Transwell system suggested that most of the ZPLs could pass through mucus and exhibited high mucus permeation. This study provides a new opportunity to prepare RES-ZPLs nanospheres as a smart drug nanocarrier, with enhanced stability and mucus permeation.

Effects of frequency ultrasound on the properties of zein-chitosan complex coacervation for resveratrol encapsulation

Xiaofeng Ren,Ting Hou,Qiufang Liang,Xi Zhang,Di Hu,Baoguo Xu,Xinxiang Chen,Meram Chalamaiah,Haile Ma

In this study, resveratrol was successfully encapsulated using zein-chitosan complex coacervation. The encapsulation efficiency was markedly improved (51.4%) after chitosan coating at 1:2.5 zein/chitosan ratio, compared with 38.6% using native zein . Analysis of multi-model frequency ultrasound treatment effects on resveratrol encapsulation using zein-chitosan complex coacervation showed that 28/40 kHz dual-frequency ultrasound led to the highest encapsulation efficiency (65.2%; 31.9% increase) and loading capacity (5.9%; 31.1% increase) of resveratrol, followed by multi-frequency ultrasound at 20/28/40 kHz (17.8% encapsulation efficiency increase; 17.8% loading capacity increase). Dual-frequency ultrasound treatment significantly reduced the zein-chitosan complex coacervation particle size and reduced their distribution, however, did not change the zeta potential. Fourier transform infrared spectroscopy and fluorescence spectroscopy analysis demonstrated that ultrasound treatment had no effect on secondary structure of zein-chitosan complex but markedly decreased the fluorescence emission intensity. Differential scanning calorimetry and X-ray diffraction results indicated that Dual-frequency ultrasound treatment improved the thermal stability of zein-chitosan complex coacervation but had no effect on the crystal structure. Atomic force microscopy and scanning electron microscopy images revealed uniform distribution of zein-chitosan complex coacervation followed by ultrasonic treatment.

Effect of drying temperature on structural and thermomechanical properties of konjac glucomannan-zein blend films

Chong Li,Kao Wu,Yuehong Su,Saffa B. Riffat,Xuewen Ni,Fatang Jiang

Konjac glucomannan (KGM)/zein blend films were successfully prepared by solution casting at different drying temperatures (40, 50, 60, 70 and 80   °C). The effects of drying temperature on the films' structural, thermomechanical, mechanical and water barrier properties were investigated. Microstructural observations indicated that zein particles were homogeneously dispersed in KGM continuous matrix, and the blend film dried at 60   °C showed the most compact and smooth surface. Dynamic mechanical thermal analysis curves showed that with increasing drying temperature from 40 to 60   °C, glass transition temperature (Tg) of films increased; however, with further increase in temperature, the Tg decreased, indicating the compatibility of film components was the highest when dried at 60   °C. The hydrophobicity of blend film dried at 60   °C was significantly stronger than that of other blend films, supported by the highest water contact angle, and the lowest swelling ratio and solubility. Moreover, the film dried at 60   °C showed the highest tensile strength , elongation at break, and the lowest water vapor permeability. Therefore 60   °C was preferred for KGM/zein blend film preparation. This study indicated that intermolecular interactions among film components were greatly influenced by the drying temperature, and should be carefully noticed for film preparation.

Folate-conjugated zein/Fe3O4 nanocomplexes for the enhancement of cellular uptake and cytotoxicity of gefitinib

Pang Jiafeng,Li Zhixian,Li Shaomin,Lin Shuying,Wang Hongdi,Xie Qiuling,Jiang Yanbin

Integrating various functional components into a single nano-platform is an ideal but challenging strategy for cancer therapy. Herein, a facile approach to fabricating multi-targeted nano-drug delivery systems which can be monitored is reported. Superparamagnetic iron oxide nanoparticles (SPIONs) and gefitinib (GEF) were encapsulated into folic acid-conjugated zein (Fa–zein) nanocomplexes (GEF-FSZs) with good dispersity, high GEF loading efficiency and pH-dependent release profile. The uptake of water-insoluble GEF was facilitated by encapsulating GEF into a zein-based nanocomplex, the magnetic responsive property endowed by SPIONs and the conjugation of Fa, which resulted in enhanced toxicity to A549 cells. The endocytosis study indicated that macropinocytosis and clathrin/caveolae-independent endocytosis exerted great influence on the internalization of GEF-FSZs. These results implied that GEF-FSZs could be a promising candidate for controlled and targeted drug delivery.

Co-encapsulation of quercetin and resveratrol: Comparison in different layers of zein-carboxymethyl cellulose nanoparticles

Long Chen,Wen Xu,Zhongyu Yang,David Julian McClements,Xinwen Peng,Zhenlin Xu,Man Meng,Yidong Zou,Guanxiong Chen,Zhengyu Jin

Three nanoparticles were fabricated for the co-delivery of quercetin and resveratrol. Nanoparticles consisted of a zein and carboxymethyl cellulose assembled using antisolvent precipitation/layer-by-layer deposition method. Nanoparticles contained quercetin in the core and resveratrol in the shell, resveratrol in the core and quercetin in the shell or both quercetin and resveratrol in the core. The particle sizes of nanoparticles were 280.4, 214.8, and 181.8 nm, respectively. Zeta-potential was about −50 mV and PDI was about 0.3. The different positions of polyphenol distribution nanoparticles could reduce the competition between the two polyphenols, the encapsulation rate, loading rate and storage stability reached up to 91.7 %, 5.37 % and 97.1 %, respectively. FT-IR showed that hydrophobic and electrostatic interactions were the main driving forces of nanoparticle assembly. XRD showed that two polyphenols were successfully encapsulated in nanoparticles. TGA showed that distributing the nanoparticles in different layers would enhance thermal stability. TEM and SEM showed that polysaccharides attached to the surface of nanoparticles formed a core-shell structure with uniform particle size. All three nanoparticles could release two polyphenols slowly in simulated gastrointestinal digestion, Korsmeyer-Peppas was the most suitable kinetic release model. Therefore, biopolymer-based nanocarriers can be created to enhance the loading, stability, and bioaccessibility of co-encapsulated nutraceuticals.

Supramolecular design of coordination bonding architecture on zein nanoparticles for pH-responsive anticancer drug delivery

Hongshan Liang,Bin Zhou,Jing Li,Wei Xu,Shilin Liu,Yan Li,Yijie Chen,Bin Li

A pH-responsive system by constructing a designable coordination bonding-based metal-tannic acid (TA) architecture on zein nanoparticles (NPs) has been investigated. Film formation was initiated by the adsorption of the polyphenol and directed by pH-dependent, multivalent coordination bonding. The prepared metal-TA coated zein NPs (zein-TA/metal NPs) demonstrated good stability to maintain particle size in cell culture medium at 37   °C. The microstructure of the NPs was revealed by transmission electron microscopy (TEM). To confirm the surface chemical information of the NPs, XPS analysis was performed. Furthermore, in vitro viability studies revealed that the zein-TA/metal NPs showed no significant cytotoxicity against HepG2 cells for 24   h. Because of the pH-responsive coordination bonding between TA and metal ions, the functional property of the metal-TA films was tailored for drug delivery. Biocompatible AuNPs were produced using zein-TA/metal NPs as reducing and stabilizing agents which were promising in the photothermal therapy of cancers and other diseases.

Environmentally friendly zein/ethylcellulose nanofiber air filtration materials with tunable hydrophobicity and high filtration efficiency

Naiteng Wei,Feng Yang,Yaxin Zhao,Huafeng Tian,Yujuan Jin,Rakesh Kumar

Due to people's environmental awareness and the continuous improvement of the living environment requirements, the pollution problem of fine particles has attracted widespread attention and great importance. Therefore, the development of new green and environmentally friendly air filtration materials with high efficiency and low resistance is ongoing. In this work, eco-friendly zein/ethylcellulose blende nanofiber membranes with different fiber morphologies, diameter sizes, and hydrophobicity are prepared by electrospinning technology, and their performance in the field of air filtration and purification is investigated, to make them highly efficient for the adsorption of small pollutants of various polarities. The experiments showed that the hydrophobicity of the nanofiber membrane was adjusted by changing the ratio of zein and ethylcellulose, and the addition of ethylcellulose improved the thermal stability and use temperature of the composite nanofiber membrane. The filtration efficiency of the nanofiber membrane can reach more than 85 % for small particle pollutants of different polarities, and both sides of the tested membrane have high filtration capacity, which can still be maintained after three times of reused. This gives it great potential and broad application prospects in the field of air filtration.

Enhanced bioaccessibility of cyclolinopeptides via zein-cyclodextrin nanoparticles: Simulated gastrointestinal digestion and cellular uptake study

Peifang Chen,Yong Wang,Zizhe Cai,Xuanxuan Lu

Cyclolinopeptides (CLS) are hydrophobic cyclic peptides in flaxseed with multiple bioactive activities. This study developed zein ( Z )-cyclodextrin (CD) binary nanoparticles (NPs) as an oral delivery system for CLS. Z -CD NP had a smaller diameter (D h ) and better encapsulation effect on CLS. Formation of CLS-loaded NPs was driven by hydrogen bonds and electrostatic interactions. Presence of CD improved the thermal, pH and storage stabilities of NPs. Besides, CD prevented premature release of CLS in the stomach and enhanced the bioaccessibility of CLS to a maximum of 86.71 % ± 2.20 %. Lipid-raft-mediated endocytosis was involved in the cell uptake of NPs, where the addition of CD significantly facilitated the uptake of NPs. Z -CD NPs also enhanced absorption and reduced secretion of CLS after digestion. Overall, this study provides a simple approach to enhance the oral delivery efficiency of CLS by modulating Z-based NPs with CD.

Efficient stabilizing effect of low-dose zein/xanthan gum nanoparticles at the oil-water interface

Lida Wu,Yitong Jiang,Chong Chen,Weibo Zhang,Jiayue Guo,Siyuan Liu,Pengjie Wang,Fazheng Ren,Baocai Xu,Yao Hu

The inherent propensity for aggregation necessitates the use of high concentrations of protein-polysaccharide nanoparticles to achieve stable Pickering emulsions. This study employed xanthan gum (XG) to mitigate the pronounced aggregation of zein nanoparticles by structure construction, thereby enhancing the emulsifying efficiency of zein/XG (Z/XG) nanoparticles. The Z/XG nanoparticles displayed significantly enhanced dispersity, with the absolute ζ-potential increasing from 6.25 mV to approximately 55 mV as the zein/XG ratio was optimized from 1:0 to 1:0.2. The Z/XG nanoparticles exhibited core–shell structure with zein as the core and XG as the shell, facilitated by electrostatic interactions. This structural transformation enables Z/XG nanoparticles to more readily adsorb at the oil-water interface. The addition of excess XG can further augment the stability of Z/XG emulsions due to the formation of XG networks in the continuous phase preventing the coalescence of oil droplets. It was demonstrated that a low concentration of 0.4 % Z/XG nanoparticles is sufficient to maintain emulsion stability under typical conditions of heating, pH, and ionic treatments. Overall, this study has developed a highly effective strategy to overcome the application challenges associated with protein-polysaccharide Pickering emulsions, broadening their potential applications in the food, cosmetic, and pharmaceutical industries.

Fabrication and characterization of electrospun zein nanofibers incorporated with citronellol-rich Origanum vulgare essential oil

Mayire Abulitifu,Wei Qin,Wang Yingbo,Bai Xi

The citronellol-rich Origanum vulgare essential oil (OEO), which exhibits excellent antioxidant and antimicrobial activities, has limited use in food packaging owing to its high volatility and instability. This problem can be solved using electrospinning. In this study, OEO was incorporated into the zein matrix via emulsion electrospinning. OEO was obtained by hydro-distillation; gas chromatography–mass spectrometry analysis revealed that citronellol (44.34%) was a predominant compound in OEO. Scanning electron microscopy images demonstrated that the fibers had a bead-free and uniform structure with a diameter ranging from 215 ± 36 to 260 ± 72 nm. The X-ray diffractograms indicate the amorphous structure of the zein fibers. Fourier transform infrared absorption spectroscopy revealed an interaction between OEO and zein. The addition of OEO to the zein nanofibers affected its secondary structure. Thermogravimetric analysis showed that the OEO-loaded (OEO/zein) nanofibers exhibited high thermostability and that the water contact angle was enhanced. The encapsulation efficiency and loading capacity were in the range of 71.56–85.8% and 8.88–40.93%, respectively. The antioxidant activity of the OEO/zein nanofibers ranged from 45.40% to 71.50%. The OEO/zein fibers displayed antimicrobial activity against Staphylococcus aureus and Escherichia coli . In conclusion, OEO/zein nanofibers are promising alternatives for active food packaging.

Hyaluronic acid-coated zein nanoparticle-mediated resveratrol therapy for the reduction of cisplatin-associated nephrotoxicity

Yuan Ning,Ping Chen,Zhengnan Shen,Xing Liu,Huan Gu

Cisplatin (CDDP) is commonly used as an anticancer drug in clinical practice, but severe nephrotoxicity restricts it from exerting anticancer effects. Natural drugs, such as resveratrol, can alleviate the side effects of cisplatin, but their low solubility and gastrointestinal effects prevent them from working. Herein, we developed nanoparticles for kidney injury consisting of a biocompatible material, zein, as a carrier. HA-Zein/Res NPs were fabricated using low-molecular-weight hyaluronic acid coatings. This preparation is non-cytotoxic to renal tubular epithelial cells and can be used with confidence. Low-molecular-weight hyaluronic acid has inflammation-targeting properties and CDDP damage causes renal inflammation. Owing to this property of the low-molecular-weight hyaluronic acid coating, in vivo imaging experiments in mice demonstrated that the HA-Zein/Res NPs enabled more nanoparticles to accumulate in the renal sites affected by inflammation. Efficient resveratrol delivery alleviated kidney injury, and experiments demonstrated that HA-Zein/Res NPs could treat kidney injury while reducing the serum creatinine and urea nitrogen levels in mice. Collectively, these results indicated that this nanomaterial is a promising agent for reducing the clinical nephrotoxicity of cisplatin.
