This study aims to assess the toxicity of the commonly-spread titanium dioxide nanoparticles (TiO 2 NPs) by evaluating the exposure impact of the particles on both freshwater algae Chlorella pyrenoidosa and zebrafish liver cell line (ZFL), the two common in vitro models in toxicological studies. To compare the toxic effects of TiO 2 NPs with different physiochemical properties, three types of manufactured TiO 2 were used: bulk TiO 2 , Degussa P25 TiO 2 , and ultrafine TiO 2 NPs. Both short and long-term biological responses of green algae, such as the effect on the cell growth rate, pigment autofluorescence, and esterase activity were investigated. The dosage, physical property of TiO 2 particles, and their interactions with algal cells affect cellular growth, especially after short-term exposure. The hydrodynamic size plays a critical role in determining the acute toxicity to C. pyrenoidosa in terms of autofluorescence and esterase activity, while all types of TiO 2 NPs show toxic effects after exposure for 14 days. However, this observation is not seen when studying the effect of introduced particles in ZFL, for the precipitated Degussa P25 TiO 2 showed the highest cellular inhibition. Interestingly, despite the obvious overall toxicity toward C. pyrenoidosa , the photocatalytical properties of TiO 2 NPs may contribute to the enhanced photosynthesis in the low concentration range (< 40 µg mL −1 ). Overall, we found that the physical interactions between TiO 2 particles and the cells, particles’ size and dispersibility play critical role in the cytotoxic effect for both algal and ZFL cells, while the photocatalytical properties of TiO 2 particles may produce mixed effects on the cytotoxicity of green algae.

Despite that the phycosphere was an important niche for the proliferation of various bacteria and antibiotic resistance genes (ARGs), the factors that affect the colonization of bacteria and ARGs in the phycosphere are still poorly understood. In this study, sterile C. pyrenoidosa co-cultured with bacteria from different sources and provided with polylactic acid microplastic (PLA MPs) and florfenicol (FF) was examined. Results showed that bacteria promoted the growth of C. pyrenoidosa and increased its chlorophyll contents. PLA MPs and FF also showed positive effects on C. pyrenoidosa due to the “Hormesis effect”. The occurrence of bacteria in the phycosphere was significantly affected by their sources and the addition of PLA MPs and FF. However, the core microbiota of the phycosphere in each group was similar. Additionally, PLA MPs and FF proliferated the abundance of phenicol-related ARGs (especially flo R) and mobile genetic elements in the phycosphere. Notably, PLA MPs and FF enhanced the abundance of Flavobacterium , a potential host of ARGs. Our results highlighted the important roles of bacteria in microalgae and demonstrated exogenous pollutants could promote the spread of ARGs between surrounding environments and the phycosphere, which provide new insights into the occurrence and spread of ARGs in the phycosphere.

Infection and oxidative stress seriously hinder the healing of diabetic wounds, resulting in various serious health and clinical problems. Herein, a sustainable biological hydrogen (H2)-producing hyaluronic acid–based hydrogel patch (HAP-Chl) was constructed by loading an imidazolium-based poly(ionic liquid) (PIL) flocculated live Chlorella as a diabetic wound dressing. The PIL can flocculate Chlorella through electrostatic interactions between PIL and Chlorella to form Chlorella agglomerates, endowing the Chlorella in the central agglomerates with the ability to continuously produce H2 for 24 h under mild conditions. Combining the membrane disruption-related bactericidal mechanism of PIL and the antioxidant properties of the produced H2, HAP-Chl was determined to be antibacterial and antioxidant. In addition to exhibiting biocompatible and nontoxic activities, subsequent Staphylococcus aureus-infected chronic wound studies revealed that HAP-Chl is capable of promoting the healing of chronic wounds by effectively killing bacteria, reducing extensive ROS, relieving inflammation, and promoting the deposition of mature collagen and angiogenesis. This study provides a new strategy for constructing an in situ sustainable H2-producing hydrogel, enabling the formation of novel antibacterial and antioxidant material platforms with potential for wound dressing applications.

Algal proteins are an emerging source of functional foods. Herein, Chlorella pyrenoidosa protein (CPP)/xanthan gum-based hydrogels (HG) and beeswax-gelled oleogels (OG) are adopted to fabricate bigels. The phase inversion of bigels can be regulated by the ratio of OG and HG: As the OG increased, bigels turn from OG-in-HG (OG/HG) to a semicontinuous state and then HG-in-OG (HG/OG). In OG/HG bigels (OG ≤ 50 %), hydrophilic CPP acts as the emulsifier at the interface of OG and HG, while beeswax emulsifies the system in HG/OG bigels (OG = 80 %). A semicontinuous bigel appears during the transition between HG/OG and OG/HG. The increase of OG can enhance the viscoelasticity, hardness, adhesiveness, chewiness, and thermal stability. OG/HG bigels exhibit stronger thixotropic recovery and oil-holding capacity than HG/OG bigels. In the in-vitro digestion and food 3D printing, the high specific surface area and the highest thixotropic recovery caused by the emulsion structure of the OG/HG bigel (OG = 50 %) are conducive to the release of free fatty acids and molding of 3D-printed objects, respectively. This study provides a new approach to structure the gelled water-oil system with CPP and helps to develop edible algal proteins-based multiphase systems in food engineering or pharmacy.

In this study, ultrasound-assisted natural deep-eutectic solvents (NaDESs) were used to extract Chlorella. A safe and stable Ect-Pan DES (DES formed by ectoin and panthenol) was prepared by mixing different molar ratios of ectoin and panthenol. Compared with traditional extraction methods, Ect-Pan DES combined with ultrasonic treatment not only saves time but also improves the extraction efficiency of Chlorella (5.32 mg g−1). Chlorella extract has good skin permeability and can penetrate the skin cuticle to the epidermis and dermis. Chlorella extract has good anti-wrinkle and firming effects on the human body along with its biosafety, biocompatibility, and antioxidant activity. Therefore, Chlorella extract has potential for application as an anti-wrinkle agent for anti-wrinkle cosmetics.

A highly tolerant Chlorella sp. achieved efficient lambda-cyhalothrin transformation. • Multi-omics revealed coordinated detoxification and metabolic remodeling. • Intracellular–extracellular remodeling drove adaptive LC responses. • XDA-R55 treatment reduced lambda-cyhalothrin-induced oxidative stress and toxicity. • Mechanisms underlying microalgal pesticide adaptation were elucidated. Lambda-cyhalothrin (LC), a widely used pyrethroid insecticide, poses persistent ecological risks due to its toxicity and environmental persistence. In this study, a microalga strain, Chlorella sp. XDA-R55, was investigated as a potential biological platform for LC-contaminated water remediation. XDA-R55 maintained active growth under 20 mg/L LC exposure and promoted 96.6% LC removal within 6 days without external carbon supplementation. Physiological analyses revealed that XDA-R55 alleviated LC-induced oxidative stress by enhancing antioxidant defense systems and maintaining cellular redox homeostasis. Integrated transcriptomic and metabolomic analyses indicated that XDA-R55 facilitates the detoxification and transformation of LC by upregulating genes associated with glutathione metabolism, biosynthesis of secondary metabolites, and ABC transporters. Several putative transformation products, such as 4-hydroxybenzaldehyde, were identified and exhibited transient accumulation followed by decline. Ecological safety assessments demonstrated that water treated with XDA-R55 significantly alleviated oxidative stress, metabolic disturbances, and immune suppression in zebrafish, with most biomarkers recovering to levels comparable to those in the control group. Moreover, as a photoautotrophic organism, XDA-R55 facilitated LC removal and transformation without external carbon supplementation, providing a sustainable and environmentally compatible strategy for pesticide-contaminated water remediation. These findings provide mechanistic insights into microalgal adaptation and LC transformation under pesticide stress, and highlight the potential of Chlorella sp. XDA-R55 for mitigating the ecological risks associated with LC-contaminated water. Download: Download high-res image (189KB) Download: Download full-size image

Microalgal-bacterial granular sludge (MBGS) offers an efficient solution for low-carbon sewage treatment by eliminating challenging sludge-water separation processes. However, the impact of extracellular polymeric substances (EPS)-enveloped micro-environments on microalgae and the role of microalgae-derived phytohormones in bacterial signaling in MBGS remain poorly understood. This study investigated the characteristics of Chlorella vulgaris within an EPS-enveloped micro-environment using bio-entrapment in natural polysaccharide-protein (PS-PN) hydrogel granules. Results showed that PS-PN hydrogel bio-entrapment facilitates microalgal cell aggregation and proliferation, and upregulated pathways for photosynthesis, carbon metabolism, and antioxidant defense, suggesting that aggregated growth in PS-PN granules enhanced carbon fixation, organic metabolism, and environmental adaptation. In addition, tryptophan in the hydrogel activated the tryptamine pathway of C. vulgaris , increasing indoleacetic acid (IAA) production to 212.88 ± 7.86 μg/L (vs. 1.95 ± 0.10 μg/L in suspended cells). This IAA alleviated Pseudomonas stutzeri growth inhibition under sulfamethoxazole (SMX) stress. The microalgae-bacteria symbiotic system seeded with aggregated-growth microalgae exhibited superior growth and antioxidant enzyme activity under SMX stress. This study highlights the advantages of aggregated-growth microalgae in supporting both their survival and that of symbiotic bacteria under antibiotic stress, and provides new insights into the role of microalgae-derived phytohormones in microalgae-bacteria signaling and stress resistance within MBGS.

Microalgae biomass is regarded as a potential feedstock for valuable compounds such as pigments, lipids and proteins. However, development of single molecule extraction processes is the most common practice. A green multiproduct extraction approach is needed for economically sustainable process development of the microalgal industry. Therefore, this study aims to investigate the sequential extraction of lutein and lipid from dry and wet Chlorella pyrenoidosa biomass pretreated with a choline chloride-based deep eutectic solvent (DES) under a sustainable biorefinery scheme. In this context, we have assessed the kinetic modeling of the solid–liquid extraction process for the aforementioned compounds, focusing on the effects of temperature and time. The maximum lutein (3.80 mg g−1) and lipid (95.0 mg g−1) contents from dry biomass were obtained at 45 °C in 40 min and at 70 °C in 90 min, respectively. From wet biomass, the maximum lutein (2.57 mg g−1) and lipid contents (87.47 mg g−1) were obtained at 35 °C in 40 min and at 70 °C in 90 min, respectively. The kinetics of the solvent-based extraction process for lutein and lipids were assessed via first-order and second-order kinetic models with an associated investigation of kinetic parameters, such as rate constants, saturation concentration and activation energies. We found that temperature is an important parameter that influences the extraction of all compounds and also has a significant impact on the kinetic parameters. Toxicity evaluation of the DES and economic assessment of DES vs. ionic liquids (ILs) were performed. The synthesis cost of the DES is lower than that of ILs, and Escherichia coli JM109 survivability assessment confirms the DES as a non-toxic solvent. The present study provides valuable insights into the sequential extraction for a high-value multiproduct biorefinery.

The frequent occurrence of algal blooms in surface water has attracted more and more attention, which caused many water quality problems, including disinfection byproducts (DBPs). Algal organic matter (AOM) including intracellular organic matter (IOM) and extracellular organic matter (EOM), was a well-known precursor to DBPs formation in drinking water. This study evaluated the effect of ultraviolet (UV) irradiation on the cell integrity, IOM release and DBPs formation during subsequent chlorination of Chlorella sp. Results showed the damage rates of algal cells increased to 40.1% after the high UV irradiation of 528 mJ/cm 2 , which contributed to the release of IOM. In addition, UV irradiation was effective in reducing the formation of haloacetic acids (HAAs) both in AOM and IOM, but promoted the formation of nitrogenous DBPs (N-DBPs) from AOM in subsequent chlorination. Furthermore, neutral pH exerted a positive effect on the formation of DBPs. UV irradiation decreased the bromine substitution factor (BSF) value of AOM at a high bromide level. The BSF values increased with increasing of the concentration of bromide. Moreover, more amino acids and low molecular weight precursors were produced after UV irradiation in filtered supernatant, which contributed to the formation of N-DBPs with algal chlorination. Overall, this information demonstrated pre-oxidation of UV irradiation could be used to treat the algal-rich drinking water.

The priority pollutant chromium (Cr) was ubiquitous and great efforts have been made to reduce Cr (VI) into less-toxic Cr (III) by alga for the convenient availability and low expense. However, the functional role of organelle inside the algal cell in Cr (VI) reduction was poorly understood. In this study, organelles in green algae Chlorella vulgaris were extracted and further decorated for Cr (VI) reduction tests. Results showed that the chloroplast exhibited not only adsorption ability of total Cr (21.18% comparing to control) but also reduction potential of Cr (VI) (almost 70% comparing to control), whose most suitable working concentration was at 17 μg/mL. Furtherly, the isolated thylakoid membrane (ITM) showed better Cr (VI) reduction potential with the presence of sodium alginate (SA), even though the Hill reaction activity (HRA) was inhibited. As for photosystem II (PSII), the addition of mesoporous silica SBA-15 enhanced the reduction ability through improving the light-harvesting complex (LHC) II efficiency and electron transport rate. On the whole, the reduction ability order of the three kinds of materials based on chloroplast in C. vulgaris was PSII@SBA-15 > Chloroplast > ITM@SA. The attempt made in this study to reduce the Cr (VI) with C. vulgaris organelles might not only offer basement to detect the potential action mechanism of Cr (VI) reduction by C. vulgaris but also provide a new sight for the scavenge of heavy metal with biological materials.

The response mechanisms of microalgal mutant Chlorella PY-ZU1 cells were investigated in their removal of antibiotic tilmicosin from wastewater under 15% CO 2 . Low concentrations (0.01–2 mg L −1 ) of tilmicosin in wastewater stimulated the growth of microalgal cells, whereas high concentrations (5–50 mg L −1 ) of tilmicosin significantly inhibited cell growth. When initial tilmicosin concentration increased from 0 to 50 mg L −1 , fractal dimension of microalgal cells monotonically increased from 1.36 to 1.62 and cell size monotonically decreased from 4.86 to 3.75 μm. In parallel, malondialdehyde content, which represented the degree of cellular oxidative damage, monotonically increased from 1.92 × 10 −7 to 7.07 × 10 −7 nmol cell −1 . Superoxide dismutase activity that represented cellular antioxidant capacity first increased from 2.59 × 10 −4 to the peak of 6.60 × 10 −4 U cell −1 , then gradually decreased to 2.39 × 10 −4 U cell −1 . The maximum tilmicosin removal efficiency of 99.8% by Chlorella PY-ZU1 was obtained at the initial tilmicosin concentration of 50 mg L −1 .

This study investigates the biomass and carbon fixation rate of Chlorella sp. by the main valuable elements in converter steel slag in F/2 seawater medium. The results indicate that Ca, Mg, P, Si, Fe, and Mn can increase the biomass and carbon fixation rate of Chlorella sp. , while Cu, Zn, Cr, and Al can decrease the biomass and carbon fixation rate of Chlorella sp. . Three machine learning methods known as Back Propagation neural network (BPNN), decision tree (DT), and random forest (RF) were applied to construct the prediction model for the carbon fixation rate of Chlorella sp. based on real-life experimental data obtained from single factor experiments. The overall results exhibited that the BPNN model is better than the DT model and RF model to predict the carbon fixation rate of Chlorella sp. . Finally, the maximum carbon fixation rate for Chlorella sp. predicted by BPNN model is 50.86 mg/(L·d), which was 2.46 times that of the control group, under the optimum conditions of Ca 5.77 g/L, Mg 4.74 g/L, P 1.27 g/L, Si 6.31×10 −1 g/L, Fe 6.50×10 −4 g/L, Mn 5.00×10 −5 g/L, Cu 2.51×10 −6 g/L, Zn 4.98×10 −6 g/L, Cr 0 g/L, and Al 0 g/L in F/2 seawater medium.
