Biomimetic macrophage membrane-camouflaged curcumin nanocrystals attenuate myocardial fibrosis after myocardial infarction

Na Deng,Chan Liu,Aihua Xie,Man Jiang,Binyi Xia,Yadi Zhou,Zhipeng Wen,Ming Li,Xiaoyun Si,Tongkai Chen,Xiangping Lin

Myocardial infarction is one of the leading causes of heart failure. Following myocardial infarction, activated intrinsic immunity and an inflammatory response secrete various growth factors that contribute to the activation and proliferation of myocardial fibroblasts, inducing uncontrollable neo-dysfunction and heart failure. The transforming growth factor β1 (TGF-β1)/Smad signaling pathway is closely associated with ventricular remodeling and fibrosis post-myocardial infarction. Curcumin (CUR) inhibits the activation of the TGF-β1/Smad signaling pathway and collagen synthesis to alleviate heart fibrosis. However, its clinical applicability is hampered by its low bioavailability and poor water solubility. As a hydrophilic polymer, Polyvinylpyrrolidone K90 (PVP K90) can interact with CUR molecules to increase the solubility of CUR in water and improve the problem of its poor water solubility. Hence, in this work, we prepared curcumin nanocrystals (CN) used CUR and K90 by the method of solvent-exchange precipitation, which greatly increased the specific surface area and thus improved the dissolution properties. Subsequently, we prepared biomimetic CN by utilizing the chemotaxis of macrophages to regions of inflammation and extracting CN encapsulated within macrophage membranes (MCN). The outstanding characteristics of MCN were observed by transmission electron microscopy, FTIR and ultraviolet spectrophotometer. According to the cell uptake results, the macrophage membrane-coated nanoparticles were more effective in reaching RAW264.7 cells and the inflammatory damage sites. Further, the results of cardiac ultrasound and pathological staining demonstrated that MCN can alleviate the inflammatory area after infarction, facilitating the transport and release of CUR to the myocardial infarction site. The mechanism is attributed to MCN inhibiting the TGF-β1/Smad signaling pathway and improving myocardial fibrosis. Our findings offer a fresh approach to cardiac fibrosis medication treatment. This "inflammation-oriented" active targeting strategy provides a completely biomimetic concept for the precise intervention of myocardial fibrosis.

FRET-based fluorometry assay for curcumin detecting using PVP-templated Cu NCs

Zhixia Yao,Hanmeng Liu,Yaosheng Liu,Yongxing Diao,Guangxing Hu,Qifang Zhang,Zhuang Li

A novel fluorometric strategy is proposed for detecting curcumin by polyvinyl pyrrolidone-templated Cu NCs (PVP–Cu NCs) as a fluorescent probe which exhibits excitation/emission peaks at 380/510 nm. The fluorescent excitation and emission spectra of PVP-Cu NCs have a striking overlap with the UV–vis spectrum of curcumin, and the fluorescence lifetime of PVP-Cu NCs decreases after the addition of curcumin. Curcumin leads to fluorescence quenching based on fluorescence resonance energy transfer . This method allows for the determination of curcumin in the range of 0.1–10 μg mL −1 and the detection limit is 21 ng mL −1 . Furthermore, this method displays good selectivity and is successfully applied for real sample analysis.

Fabrication of soy protein isolate/cellulose nanocrystal composite nanoparticles for curcumin delivery

Songyan Wang,Yuqing Lu,Xiao-kun Ouyang,Junhong Ling

In this study, we developed novel complex nanoparticles as carriers for curcumin (Cur) delivery by using soy protein isolate (SPI) and cellulose nanocrystals (CNC) as polymer matrices. We found that the SPI-to-CNC mass ratio influenced the stability and physical properties of the SPI-CNC complex nanoparticles. Moreover, SPI-CNC complex nanoparticles had a relatively small size (197.7 ± 0.2 nm) and low polydispersity index (0.14) at a 6:1 mass ratio. The nanosystem was relatively stable at different pH values (3–9), temperatures (30–90 °C), and salt concentrations (0–40 mmol/L). Furthermore, the complex nanoparticles exhibited a high encapsulation efficiency (88.3%) and sustained release during simulated gastrointestinal digestion. Therefore, SPI-CNC complex nanoparticles are a promising delivery system for hydrophobic bioactive compounds.

Effect of Curcumin Addition on the Properties of Biodegradable Pectin/Chitosan Films

Qingtong Xie,Xudong Zheng,Liuting Li,Liqun Ma,Qihui Zhao,Shiyuan Chang,Lijun You

A pectin/chitosan matrix-loaded curcumin film (PCCF) with a deep eutectic solvent (DES) as the solvent and plasticizer was prepared in this study. Different quantities of curcumin (identified as PCCF-0, PCCF-1, PCCF-2. PCCF-3) were loaded on the pectin/chitosan film in order to evaluate their effects on the film properties. Results showed that curcumin could interact with the pectin/chitosan matrix and form a complex three-dimensional network structure. PCCF could promote the thickness, tensile strength, thermal properties, antioxidant and antiseptic capacities, but deteriorate the light transmission and elongation at the same time. The addition of curcumin would change the color of the film, without significantly affecting the moisture content. The tensile strength of PCCF-3 reached the maximum value of 3.75 MPa, while the elongation decreased to 10%. Meanwhile, the water-resistance properties of PCCF-3 were significantly promoted by 8.6% compared with that of PCCF-0. Furthermore, PCCF showed remarkable sustained antioxidant activities in a dose-dependent manner. PCCF-3 could inhibit DPPH and ABTS free radicals by 58.66% and 29.07%, respectively. It also showed antiseptic capacity on fresh pork during storage. Therefore, curcumin addition could improve the barrier, mechanical, antioxidant and antiseptic properties of the polysaccharide-based film and PCCF has the potential to be used as a new kind of food packaging material in the food industry. Keywords:chitosan;pectin;curcumin;film;DES

Curcumin prevents obesity by targeting TRAF4-induced ubiquitylation in m6A-dependent manner

Yushi Chen,Ruifan Wu,Wei Chen,Youhua Liu,Xing Liao,Botao Zeng,Guanqun Guo,Fangfang Lou,Yun Xiang,Yizhen Wang,Xinxia Wang

Obesity has become a major health problem that has rapidly prevailed over the past several decades worldwide. Curcumin, a natural polyphenolic compound present in turmeric, has been shown to have a protective effect on against obesity and metabolic diseases. However, its underlying mechanism remains largely unknown. Here, we show that the administration of curcumin significantly prevents HFD-induced obesity and decreases the fat mass of the subcutaneous inguinal WAT (iWAT) and visceral epididymal WAT (eWAT) in mice. Mechanistically, curcumin inhibits adipogenesis by reducing the expression of AlkB homolog 5 (ALKHB5), an m6A demethylase, which leads to higher m6A-modified TNF receptor-associated factor 4 (TRAF4) mRNA. TRAF4 mRNA with higher m6A level is recognized and bound by YTHDF1, leading to enhanced translation of TRAF4. TRAF4, acting as an E3 RING ubiquitin ligase, promotes degradation of adipocyte differentiation regulator PPARγ by a ubiquitin–proteasome pathway thereby inhibiting adipogenesis. Thus, m6A-dependent TRAF4 expression upregulation by ALKBH5 and YTHDF1 contributes to curcumin-induced obesity prevention. Our findings provide mechanistic insights into how m6A is involved in the anti-obesity effect of curcumin.

Dual functional matrix metalloproteinase-responsive curcumin-loaded nanoparticles for tumor-targeted treatment

Fangyuan Guo,Qiafan Fu,Chenhao Jin,Xugang Ji,Qinying Yan,Qingliang Yang,Danjun Wu,Ying Gao,Weiyong Hong,Aiqin Li,Gensheng Yang

The limitations of anticancer drugs, including poor tumor targeting and weak uptake efficiency, are important factors affecting tumor therapy. According to characteristics of the tumor microenvironment, in this study, we aimed to synthesize matrix metalloproteinase (MMP)-responsive curcumin (Cur)-loaded nanoparticles (Cur-P-NPs) based on amphiphilic block copolymer (MePEG-peptide-PET-PCL) with MMP-cleavable peptide (GPLGIAGQ) and penetrating peptide (r9), modified to improve tumor targeting and cellular uptake. The average size of Cur-P-NPs was 176.9 nm, with a zeta potential of 8.1 mV, and they showed drug entrapment efficiency and a loading capacity of 87.07% ± 0.63% and 7.44% ± 0.16%, respectively. Furthermore, Cur release from Cur-P-NPs was sustained for 144 h at pH 7.4, and the release rate was accelerated under enzyme reaction condition. The MTT assay demonstrated that free P-NPs had favorable biosafety, and the anti-proliferative activity of Cur-P-NPs was positively correlated with Cur concentration in MCF-7 cells. Additionally, the results of cellular uptake, in vivo pharmacokinetics, and biodistribution showed that Cur-P-NPs had a good effect on cellular uptake and tumor targeting, resulting in the best bioavailability in tumor therapy. Therefore, Cur-P-NPs, as a promising drug delivery system, might lead to a new and efficient route for targeted therapy in clinical practice.

Odorranalectin modified PEG–PLGA/PEG–PBLG curcumin-loaded nanoparticle for intranasal administration

Xinrui Li,Jing Su,Zul Kamal,Pengcheng Guo,Xinyi Wu,Lina Lu,Hongbing Wu,Mingfeng Qiu

Curcumin (Cur) is a promising drug for neurological diseases. Nevertheless, the application of Cur has been limited due to its difficulty in penetrating blood-brain barrier (BBB). Intranasal drug delivery, a noninvasive alternative delivery of Cur, can effectively help Cur cross BBB and inert into central nervous system directly. Odorranalectin (OL) which is the smallest lectin can prolong the residence time of Cur in the nasal mucosa and promote cellular uptake. In this work, a nasal delivery system incorporating OL modified Cur-loaded nanoparticles (Cur–OL–NPs) was developed and expected to bypass BBB and promote the absorption of Cur. We conjugated OL to polyethylene glycol-poly (lactic-co-glycolic acid) (PEG–PLGA), and combined polyethylene glycol-poly (γ-benzyl-L-glutamate) (PEG–PBLG) and OL–PEG–PLGA to prepare nanoparticles to improve the stability, bioavailability and targeting of Cur. Compared with unmodified NPs, increased efficiency of Cur–OL–NPs cellular uptake by Calu-3 cells had been obtained with no severe toxicity. Furthermore, in vivo pharmacokinetic studies also showed that Cur–OL–NPs had higher relative bioavailability. Thus, it is concluded that the results indicated that OL–NPs as carriers of Cur had a promising future in nasal drug delivery system.

Synergistic antibacterial effects of curcumin modified silver nanoparticles through ROS-mediated pathways

Zhiyong Song,Yang Wu,Huajuan Wang,Heyou Han

Due to their remarkable antibacterial properties, silver nanoparticles (Ag NPs) and curcumin (CCM) have been widely used in the antimicrobial field. In our study, we have fabricated the uniform and stable silver/curcumin composite nanoparticles by a facile ultrasound treatment process and the synergistic antibacterial activity were evaluated. The curcumin not only played a role of reducing agent but also acted as a capping agent. The antibacterial effects of silver/curcumin (cAgNPs) were studied by measuring the growth curve and surface plate assay based on the E. coli and B. subtilis , which showed concentration dependent bacteriostatic and bactericidal effects of cAgNPs. The presence of CCM enhance the binding of Ag to bacterial membrane and Ag + release in comparison to that without CCM, so that creating a temporary and local high Ag + concentration near the surface of the bacterium, meanwhile, generation of more reactive oxygen species, lead to membrane damage, bacterial lipases and induce leakage of intracellular contents followed by bacterial death that lead to growth inhibition of the bacteria. The antibacterial effects were characterized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM), the effect which were further found to decrease by introducing antioxidant N -acetyl- l -cysteine (NAC) act as a reactive oxygen species (ROS) scavenging agent. These initial data suggest that cAgNPs have a highly antibacterial efficient and might have potential to be developed as an effective antimicrobial nanomaterial.

Improved bioavailability of curcumin in ovalbumin-dextran nanogels prepared by Maillard reaction

Jin Feng,Shanshan Wu,Hua Wang,Songbai Liu

In this work, ovalbumin-dextran nanogels were fabricated via the Maillard reaction followed by a heat-gelation process and their potential to improve curcumin bioavailability was investigated using an in vitro gastrointestinal tract. The result of electrophoresis, conjugation efficiency and yield, and far-UV circular dichroism verified the covalent attachment of dextran to ovalbumin by the Maillard reaction. The conjugates were then heated at the isoelectric point of ovalbumin to give stable nanogels. The transmission electron microscopy observation suggested that nanogels displayed a spherical structure. Curcumin was loaded into nanogels by a pH-driven method and the incorporation of curcumin affected the morphology of nanogels marginally. The transformation of curcumin in nanogels was significantly higher than that in ovalbumin nanoparticle. On the other hand, the bioaccessibility was similar for curcumin in nanogels and nanoparticle. The ovalbumin-dextran nanogels hold the potential as delivery systems for curcumin to fortify functional foods and beverages.

Curcumin-loaded redox-responsive mesoporous silica nanoparticles for targeted breast cancer therapy

Nana Li,Zhi Wang,Yongtai Zhang,Kai Zhang,Jianxu Xie,Ying Liu,Wansi Li,Nianping Feng

Hypothesis: The antitumor applications of curcumin (CUR) are limited because of its low water solubility, poor stability, and low bioavailability. We developed novel nanocarrier systems for tumour targeting and controlled CUR release and evaluated their therapeutic efficacy.Experiments: The surface of mesoporous silica nanoparticles (MSN) was modified with hyaluronan (HA) or polyethyleneimine-folic acid (PEI-FA) via disulfide bonds. The capacity of the resultant nanocarriers (MSN-HA and MSN-PEI-FA, respectively) for CUR delivery was evaluated in a breast cancer cell line and a mouse xenograft model.Findings: MSN/CUR-PEI-FA and MSN/CUR-HA were cytotoxic to MDA-MB-231 breast cancer cells. Both formulations showed an enhanced cellular uptake compared with that of a non-targeted nanocarrier, with a greater cellular uptake of FA-modified nanoparticles than that of HA-modified nanoparticles. Accordingly, MSN-PEI-FA showed more precise targeting and higher accumulation in tumours than did MSN-HA, as visualized by live imaging. Both types of nanoparticles had good biocompatibility and low toxicity, and MSN/CUR-PEI-FA inhibited the tumour growth to a greater degree than did free CUR. Thus, MSN/CUR-PEI-FA are a promising drug delivery system for the treatment of breast cancer.

Preparation, characterization and in vitro release properties of pectin-based curcumin film

Pan Jie,Cao Duanlin,Ma Xuemei,Yang Jing

Curcumin is a kind of natural polyphenol whose functions are anti-tumor, anti-oxidant, anti-inflammatory and anti-microbial with low side effects. Solubility of Cur in water is poor, which results in low bioavailability, rapid metabolism, instability and poor absorption in vivo. Based on the crosslinking mechanism of pectin-calcium, the curcumin-pectin film (PT-Curf) was prepared by casting method, where chitosan and sodium alginate were added to the film, forming a system for blending modification to improve the performance of PT-Curf. Furthermore, the release of the curcumin films in vitro was studied and the effects of natural polysaccharides on the drug delivery system were discussed. According to our results, the natural polysaccharides involving pectin, chitosan and sodium alginate were excellent carriers of the fat-soluble drug Cur, and Cur was uniformly dispersed in the composite films in different crystal forms. Out of different drug delivery media, the three films exhibited different release rate and sustained release effect in vitro. However, the pectin-sodium alginate curcumin film (PT-SA-Curf) exhibited the most sustained release effect, the pectin-chitosan curcumin film (PT-CS-Curf) was second.

The stability and in vitro digestion of curcumin emulsions containing Konjac glucomannan

Shenghua He,Cheng Gu,Deguo Wang,Weili Xu,Rongchun Wang,Ying Ma

Konjac glucomannan (KGM) was used to characterize the structure of the curcumin emulsions stabilized with whey protein isolate (WPI). With an increasing concentration of KGM from 0.1% to 0.3%, the fat droplet size decreased significantly (p < 0.05) with increasing KGM content. The retention of curcumin in emulsions with KGM was significantly higher (p < 0.05) than the emulsions without KGM, after heating at 85 °C for 90 min. These two emulsions showed good stability in the pH range of 2–8 in addition to pH 5. Both emulsions with/without KGM were stable at 0–80 mM salt concentration, however the curcumin emulsion with KGM was unstable at 100 mM NaCl concentration. The droplet size analysis and microscopic observations indicated that the aggregation of fat droplets in these two emulsions occurred under simulated gastric fluid (SGF) conditions with a digestion time of 10 min. There were more undigested fat droplets in the curcumin emulsion with KGM than the emulsion without KGM under simulated intestinal fluid conditions after 120 min of digestion. The addition of KGM in WPI-stabilized curcumin emulsions slowed down the release rate of free fatty acids and curcumin from the emulsion droplets. These findings indicated that KGM could be used to achieve a controlled and sustainable release of bioactive compounds from emulsions.
