Sensitive SERS assay for glyphosate based on the prevention of L-cysteine inhibition of a Au–Pt nanozyme

Jun Ma,Gege Feng,Ying Ying,Yong Shao,Yongxin She,Lufei Zheng,A. M. Abd EI-Aty,Jing Wang

Herein, an indirect SERS sensing assay was developed for the determination of glyphosate (Gly) in tap water. The mechanism of detection was based on relieving the inhibitory effect of L-cysteine (L-cys) on a Au–Pt nanozyme by combining Gly with L-cys through divalent copper ions (Cu2+). In this method, a novel nano-chain-like Au–Ag composite with good repeatability was successfully fabricated to detect SERS signals of oxTMB without disturbing TMB Raman signals. Under optimal conditions, the SERS signal intensity of oxTMB (at 1605 cm−1) was proportional to the concentration of Gly. The results showed a good linear response over the concentration ranges of 10 μg L−1 to 1000 mg L−1. The limit of detection and limit of quantitation of Gly were found to be 5 μg L−1 and 10 μg L−1, respectively. In addition, good anti-interference ability against interfering cations and structural analogues deserves to be mentioned. This SERS assay can be used for detection of Gly in tap water that can meet the needs of practical detection.

A turn-on fluorescent sensor based on carbon dots from Sophora japonica leaves for the detection of glyphosate

Jingzhou Hou,Xianfeng Wang,Shiyu Lan,Chao Zhang,Changjun Hou,Qiang He,Danqun Huo

Carbon dots (CDs) having low cost and low toxicity and synthesized via a green route were applied to establish a fluorescent nanoprobe for the measurement of glyphosate. The synthesis was realized via a one-pot hydrothermal procedure using Sophora japonica leaves as the carbon source. It was found that electron transfer occurred between Fe3+ and the as-prepared CDs. Therefore, Fe3+ exhibited a specific dynamic-quenching toward CDs. However, the electron transfer process was inhibited by glyphosate. The fluorescence of the quenched CDs/Fe3+ system was recovered by the addition of glyphosate. It resulted from the strong complexation between Fe3+ and the functional groups (like –PO3H2 and –COOH) in the glyphosate molecule. These functional groups captured Fe3+ from the CD/Fe3+ system to reduce the electron transfer. With such a design, the rapid detection of glyphosate could be realized by this turn-on fluorescent sensor based on the CD/Fe3+ system. Under optimal conditions, the CD/Fe3+ system showed a concentration-dependent fluorescent response toward glyphosate in the linear range from 0.1 to 16 ppm. The limit of detection was calculated to be as low as 8.75 ppb (3σ/S). In addition, the successful detection of glyphosate in real samples with satisfactory recoveries exhibited a practical application of the CD/Fe3+ nanoprobe in food safety and environmental monitoring.

Attapulgite-based herbicide particles with smart wetting surface for environmental-safe glyphosate delivery induced by consecutive base-acid stimuli

Nana Jing,Chaowen Chen,Chuang Chen,Xue Yuan,Xueqi Ma,Guopeng Teng,Jia Zhang,Xingjiang Li,Zhengyan Wu

To minimize the pesticide loss and enhance the efficiency of active ingredient utilization, the pH-responsive controlled-release pesticide with easy operation has been widely applied despite for its potential on bothering the non-target organisms while using acid or alkaline alone. To overcome that obstacle, we developed an environmentally safe herbicide particle with a smart wetting surface that can be regulated through consecutive base-acid stimuli to maintain stable pH levels. Herein, attapulgite (ATP) is mixed with glyphosate (Gly) initially and the resultants are coated by suspensions of phenylamine-modified ATP (ph-ATP) and amino silicon oil (ASO), ultimately the outmost polyvinyl pyrrolidone (PVP) are constructed to acquire the smart herbicide (AG@ph-AA@PVP). The base may destroy the outmost PVP membrane, and the later acid treatment turns ph-AA layer gradually from hydrophobic to hydrophilic due to protonation of aniline groups, both of which allow water to invade further and promote Gly release. Therefore, the AG@ph-AA@PVP appears with excellent base-acid sensitivity and controlled-release performance, being demonstrating by the burst Gly release in water and significant weeds control efficiency in pot experiments. The interaction of the materials and the surface wettability were analyzed for explaining the controlled-release mechanism. Besides, AG@ph-AA@PVP possesses high environmental stability and good biosafety. This work develops a simple and feasible technology for environmental-safe and on-demand Gly delivery, assisting for advancement of sustainable and green agriculture.

An on–off–on fluorescent probe for the detection of glyphosate based on a Cu2+-assisted squaraine dye sensor

Shuhua Zhao,Lei Shi,Xiufeng Zhang,Xiaoran Sun,Wenxuan Zhu,Lijia Yu

The herbicide glyphosate, N-(phosphonomethyl)glycine, has been widely used in the past 40 years, and has had many adverse effects on human health. Here, we constructed a convenient “on–off–on” fluorescent platform for detection of glyphosate via Cu2+ modulated squaraine dye fluorescence quenching. The squaraine dye F-0 exhibited strong fluorescence, which could be quenched by the addition of Cu2+. However, the addition of glyphosate restored the fluorescence intensity of F-0 due to the formation of a Cu2+-glyphosate complex. F-0 was utilized as a fluorescent probe for the quantitative detection of glyphosate, with the lowest detection limit of 13.16 nmol L−1. Furthermore, this method demonstrated high selectivity and anti-interference capabilities. The successful monitoring of glyphosate in real samples was achieved using this detection strategy.

Rational Design of a Molecularly Imprinted Sensor on a Biomass Carbon Platform for Glyphosate Monitoring in Traditional Chinese Medicines

Xin Wang,Delai Zhou,Xuxia Liu,Guodi Lu,Jia Hou,Jian Xu,Fude Yang

A molecularly imprinted electrochemical sensor was developed for the selective and sensitive detection of glyphosate in Traditional Chinese Medicine samples. An excellent conductive hierarchical porous carbon substrate made from sodium alginate and ammonium chloride co-carbonization was used to build the sensor. The molecularly imprinted polymer layer was systematically designed using Density Functional Theory calculations, which identified nicotinamide as the optimal functional monomer. A deep eutectic solvent was utilized as an effective green eluent for template removal. Under optimized conditions, the sensor demonstrated a wide linear detection range from 1.0 × 10−9to 1.0 × 10−6M with an exceptionally low detection limit of 8.8 × 10−10M. The sensor exhibited satisfactory reproducibility (RSD = 3.35%,n= 6), repeatability (RSD = 5.0% over 6 cycles), and robust stability (retaining >90% initial response after 10 days). The sensor displayed satisfactory recovery rates of 94.47–112.23% and RSD values ranging from 1.37–3.01% when applied to real traditional Chinese medicine samples, thereby confirming its accuracy and practical utility for glyphosate residue analysis in complex matrices. This study introduces an effective sensing platform that integrates rational design principles with environmentally friendly synthesis strategies for quality control in traditional medicine applications.

Insights into the enhanced adsorption of glyphosate by dissolved organic matter in farmland Mollisol: effects and mechanisms of action

Jiao Yaqi,Jia Junxin,Gu Jiaying,Wang Sa,Zhou Qin,Li Hui,Li Li

Dissolved organic matter (DOM) is easy to combine with residual pesticides and affect their morphology and environmental behavior. Given that the binding mechanism between DOM and the typical herbicide glyphosate in soil is not yet clear, this study used adsorption experiments, multispectral techniques, density functional theory, and pot experiments to reveal the interaction mechanism between DOM and glyphosate on Mollisol in farmland and their impact on the environment. The results show that the adsorption of glyphosate by Mollisol is a multilayer heterogeneous chemical adsorption process. After adding DOM, due to the early formation of DOM and glyphosate complex, the adsorption process gradually became dominated by single-layer chemical adsorption, and the adsorption capacity increased by 1.06 times. Glyphosate can quench the endogenous fluorescence of humic substances through a static quenching process dominated by hydrogen bonds and van der Waals forces, and instead enhance the fluorescence intensity of protein substances by affecting the molecular environment of protein molecules. The binding of glyphosate to protein is earlier, of which affinity stronger than that of humic acid. In this process, two main functional groups (C–O in aromatic groups and C–O in alcohols, ethers and esters) exist at the binding sites of glyphosate and DOM. Moreover, the complexation of DOM and glyphosate can effectively alleviate the negative impact of glyphosate on the soil. This study has certain theoretical guidance significance for understanding the environmental behavior of glyphosate and improving the sustainable utilization of Mollisol. Graphical abstract

Two swords combination: Smartphone-assisted ratiometric fluorescent and paper sensors for dual-mode detection of glyphosate in edible malt

Kai Deng,Haipeng Guo,Xueying Li,Te Li,Tingting Di,Runran Ma,Doudou Lei,Yining Zhang,Jiabo Wang,Weijun Kong

The long-term and excessive use of glyphosate (GLY) in diverse matrices has caused serious hazard to the human and environment. However, the ultrasensitive detection of GLY still remains challenging. In this study, the smartphone-assisted dual-signal mode ratiometric fluorescent and paper sensors based on the red-emissive gold nanoclusters (R-AuNCs) and blue-emissive carbon dots (B-CDs) were ingeniously designed accurate and sensitive detection of GLY. Upon the presence of GLY, it would quench the fluorescence of B-CDs through dynamic quenching effect, and strengthen the fluorescence response of R-AuNCs due to aggregation-induced enhancement effect. Through calculating the GLY-induced fluorescence intensity ratio of B-CDs to R-AuNCs by using a fluorescence spectrophotometer, low to 0.218 μg/mL of GLY could be detected in lab in a wide concentration range of 0.3–12 μg/mL with high recovery of 94.7–103.1% in the spiked malt samples. The smartphone-assisted ratiometric fluorescent sensor achieved in the 96-well plate could monitor 0–11 μg/mL of GLY with satisfactory recovery of 94.1–107.0% in real edible malt matrices for high-throughput analysis. In addition, a portable smartphone-assisted ratiometric paper sensor established through directly depositing the combined B-CDs/R-AuNCs probes on the test strip could realize on-site measurement of 2–8 μg/mL of GLY with good linear relationship. This study provides new insights into developing the dual-signal ratiometric sensing platforms for the in-lab sensitive detection, high-throughput analysis, and on-site portable measurement of more trace contaminants in foods, clinical and environmental samples.

Dual beneficial activities of Paenibacillus ottowii NPDY10: glyphosate degradation, modulation of soil microbial communities, and antagonism against phytopathogenic fungi

Deng Yun,Niu Yuqing,Xu Qing,Xie DongRong,Su Yan,Liu DuanHua,Chen Yong-He,Han YongHe,Tian Dagang

Background The widespread use and environmental persistence of the herbicide glyphosate raise significant concerns for human and ecological health. Developing efficient and sustainable remediation strategies is therefore crucial. Microbial degradation and biocontrol are two promising approaches. This study aimed to isolate and characterize a bacterial strain capable of both degrading glyphosate and antagonizing soil-borne fungal pathogens, and to evaluate its impact on soil microbial communities. Results This study isolated and characterized a novel bacterial strain, NPDY10, identified as Paenibacillus ottowii , which exhibits dual beneficial functions: efficient glyphosate degradation and broad-spectrum antagonism against soil-borne fungal pathogens. Under optimal conditions (pH 7–7.5, 30–35 °C), strain NPDY10 completely degraded 400 mg L⁻¹ of glyphosate within four days, utilizing the herbicide as a carbon source. Metabolic profiling via UPLC-MS/MS confirmed glyphosate degradation via the aminomethylphosphonic acid (AMPA) pathway In a rice paddy field trial, the application of NPDY10 reduced glyphosate levels in paddy water, and a visible foam layer formed, serving as a visual indicator of active metabolism. Furthermore, strain NPDY10 demonstrated broad-spectrum antifungal activity in vitro, inhibiting 17 plant-pathogenic fungi with inhibition rates ranging from 60.00% to 92.00%. Analysis of soil microbial communities in a subsequent maize cultivation season revealed that NPDY10 treatment increased bacterial diversity and suppressed the relative abundance of pathogenic fungal genera such as Fusarium and Gibberella . Conclusion Paenibacillus ottowii NPDY10 represents a promising multi-functional agent for sustainable agriculture, potentially combining glyphosate bioremediation with direct and indirect biocontrol effects through modulation of soil microbial communities, thereby contributing to improved soil health.

A novel hydrazide-based fluorescent chemosensor and its application to recognize Cu2+ and glyphosate in environmental and biological systems

Dong Zhang,Xiaohui Pang,Xinfu Zhu,Yi Wang,Meiqing Zhu

Background Although copper ions (Cu 2+ ) and glyphosate (GLY) tend to exist in the environment or living organisms at low doses, they gradually accumulate with exposure time and may eventually cause irreversible damage to living organisms. The existing detection methods are effective, but most of them lack the ability to detect multiple pollutants simultaneously or suffer from low sensitivity and poor selectivity. Therefore, it is important to develop a sensor that can efficiently and sensitively detect copper ions and glyphosate, but also has the potential for practical applications. Results In this study, a novel hydrazide-based DHBC capable of sequentially detecting Cu 2+ and glyphosate (GLY) was developed. The DHBC forms a complex with Cu 2+ , causing fluorescence quenching. Upon exposure to GLY, functional groups such as carboxyl, phosphate, and amino groups coordinate with Cu 2+ , displacing DHBC and restoring fluorescence intensity. The mechanism has been validated by various characterization methods such as HRMS and Job curves, DFT calculation, etc. The DHBC has excellent detection performance, including high selectivity, strong anti-interference, fast response (less than 1 min), and high sensitivity (Cu 2+ : 0.09 μM, GLY: 0.05 μM). The quantitative analysis of Cu 2+ /GLY in cycling experiments and actual samples demonstrated the great potential of DHBC for practical applications. The DHBC successfully imaged Cu 2+ /GLY in live models such as HeLa cells and zebrafish, demonstrating its broad applicability and stability in a wide range of biological models. Significance The high accuracy of DHBC in environmental pollution monitoring and its stable performance in biological modeling make it an important tool for assessing the impact of pollutants on ecosystems and human health. In addition, the wide applicability and real-time imaging capability of DHBC provide reliable technical support for environmental science, toxicology, and health risk assessment and have important prospects for scientific research and practical applications.

Co-exposure to polystyrene nanoplastics and glyphosate exacerbates NETs-mediated pyroptosis by activating the NLRP3 inflammasome in mouse liver

Huan Zeng,Jingyi Qi,Qiyi Chen,Ruowei Zhang,Chen Guo,Jinghua Zhao,Huimei Liang,Junlong Sun,Wei Wang,Houhui Song

Nanoplastics (NPs), highly prevalent due to large-scale plastic production, and glyphosate (Gly), the most utilized herbicides worldwide, are ubiquitous environmental contaminants. Growing concerns highlight that NPs can act as vectors for various pollutants like Gly, but their combined toxic effects in mammals and the underlying mechanisms remain poorly understood. In this study, the hepatotoxicity and potential mechanisms under the exposure of polystyrene nanoplastics (PSNPs) and/or Gly in vivo and in vitro were investigated. Mice were treated with PSNPs (25 mg/kg/day) and/or Gly (50 mg/kg/day) by oral gavage for 5 weeks. Results showed that exposure to PSNPs or Gly caused liver injury in mice, with co-exposure resulting exacerbated hepatotoxicity, evidenced by increased neutrophil infiltration and ultrastructural damages, elevated oxidative stress (LPO, H 2 O 2 , T-AOC and CAT), increased neutrophil chemokines (CCL2, CXCL12) and marker of neutrophil extracellular traps (NETs) formation (MPO), and upregulated pyroptosis-related factors (TLR4, NF-κB, NLRP3, ASC, Caspase-1, GSDMD, IL-1β, IL-18). In addition, a co-culture system of peripheral blood neutrophils (PBNs) and AML12 cells was established, revealing that co-exposure amplified ROS production, NETs formation (SYTOX Green staining) and pyroptosis. Notably, inhibition of the NLRP3 inflammasome significantly reduced NETs production, and degradation of NETs substantially decreased pyroptosis, demonstrating feedback between NETs and the NLRP3 inflammasome that drives inflammation induced by PSNPs and Gly. These results highlight that co-exposure exacerbated NETs-mediated pyroptosis through NLRP3 inflammasome activation. Our study equips new reference for understanding the mechanistic insights and health implications of the combined toxicity of PSNPs and Gly.

An artificial neural network model based on CDs@MOF/COF composites for the ultra-sensitive fluorescence detection of glyphosate

Yingfei Hui,Yuli Wei,Hao Guo,Mingyue Wang,Liping Peng,Borong Ren,Yanyan Ma,Wu Yang

Glyphosate (Gly) overuse threatens public health and ecosystems, necessitating highly sensitive detection. This work developed a novel ratiometric fluorescent probe, CDs@Cu-MOF/TMTA-COF, by integrating carbon dots (CDs), metal organic frameworks (MOFs), and covalent organic frameworks (COFs). We encapsulated CDs within Cu-MOF in-situ grown onto sp 2 ‑carbon conjugated TMTA-COF. Gly binding to Cu 2+ causes MOF structural collapse, releasing CDs and enhancing its fluorescence at 425 nm. Simultaneously, Gly inhibits competitive absorption and acceptor-photoinduced electron transfer process between TMTA-COF and Cu-MOF, increasing COF fluorescence at 550 nm. This dual-signal change enables ratiometric Gly detection from 0.2 to 40 μM, with a detection limit of 0.07 μM. This probe was successfully used to measure Gly in soil, Yellow River water and soybean (recoveries: 95.3–104.1 %). Additionally, the back propagation neural network further enables fast detection of Gly. This work demonstrates the potential of MOF/COF hybrids for fluorescence sensing for pesticide analysis and paves the way for intelligent sensing platforms.

Adverse Effects of Glyphosate and Microcystin-LR on Fish Health: Evidence from Structural and Functional Impairments in Zebrafish Gills

Yidan Zhang,Han Hu,Linmei Song,Zhihui Liu,Junguo Ma,Xiaoyu Li

Simple SummaryThe coexistence of two environmental pollutants, glyphosate (GLY, a widely used herbicide) and microcystin-LR (MC-LR, a toxic byproduct from harmful algal blooms) has become an issue that cannot be ignored. The adverse effects of both individual and combined exposures to GLY and MC-LR on aquatic organisms warrant increasing attention. This study demonstrates that exposure to GLY and MC-LR, either individually or in combination, can induce toxic effects on fish gill structure, potentially impairing their physiological function and ultimately threatening fish survival. Overall, the findings of this study not only enhance our understanding of the detrimental impacts of these pollutants on aquatic animals but also provide valuable insights for ecological and health risk assessments.Glyphosate (GLY) and microcystin-LR (MC-LR) frequently co-occur in natural water bodies. In this study, a subacute exposure test was conducted on zebrafish treated with 3.5 mg/L GLY and 35 μg/L MC-LR, individually and in combination, for 21 d to determine their effects on the gills of zebrafish and their potential mechanisms. The hematoxylin and eosin staining and scanning electron microscopy examination results showed that GLY and MC-LR exposure caused structural damage to gills. Biochemical analysis revealed Na+-K+-ATPase activity decreased, and the levels of reactive oxygen species, 8-hydroxy-2′-deoxyguanosine, and malondialdehyde increased, inducing oxidative damage to DNA and lipids of gills. Meanwhile, the inflammatory and immune function of the gill was significantly influenced, as evidenced by the alteration of the expression of tumor necrosis factor-α, interleukin-1β, complement 3, and immunoglobulin M. RNA-seq results revealed that GLY and/or MC-LR treatment induced transcriptional changes in the fish gills, which may affect various biological functions, and the lipid metabolism disruption potentially involved in the aforementioned process. Integrating histopathological, biochemical, and transcriptomic analyses, this study revealed that both individual and combined exposures to GLY and MC-LR had adverse effects on zebrafish gills, with combined exposure appears to result in more pronounced adverse effects, potentially compromising fish health.
