Hydroxyapatite (HA) has good biocompatibility and biological activity which can be widely used in bone defect repair. Zinc oxide nanoparticles (ZnO NPs) can promote osteogenesis by enhancing angiogenesis and exhibit notable antibacterial properties. Sintering temperatures critically influence the physicochemical and biological properties of bioceramic scaffolds. In this study, porous ZnO/HA bioceramic scaffolds were fabricated at various sintering temperatures, and their structural, mechanical, and biological properties were systematically investigated. The results demonstrated that increasing sintering temperature led to reduced porosity and degradation rate, accompanied by a significant improvement in compressive strength. Biocompatibility remained consistently high across all temperatures, with all scaffolds exhibiting excellent cell compatibility and osteogenic ability. However, antibacterial efficacy declined with increasing temperature. Notably, bioceramic scaffolds sintered at 950 °C achieved an optimal balance of porosity, mechanical strength, and osteogenic potential, making them promising candidates for repairing critical-sized bone defects.

Surgical intervention is a primary treatment for osteosarcoma, often resulting in a tumorous bone defect with an irregular shape. Postoperative management is essential to minimize tumor recurrence risks and promote bone regeneration. To address these issues, we developed a multifunctional injectable, rapidly photo-curable hydrogel composed of gelatin methacryloyl/carboxymethyl chitosan/hyaluronic acid (GelMA/CMCS/HA), modified with vanadium-doped mesoporous bioactive glass (VMBG). The exceptional injectability enables seamless adaptation to irregular bone defects, offering a significant advantage over preformed implants, while the rapid photocurability of the hydrogel ensures stable fixation within minutes, thereby reducing potential risks during surgery. Furthermore, this platform exhibits dual therapeutic efficacy, characterized by antitumor activity and osteogenic induction. In vitro assessments demonstrated that V(V)/V(IV) valence cycling-driven ROS generation mediated its potent antitumor efficacy. Additionally, concurrent enhancement of alkaline phosphatase activity and osteogenic marker expression validated its osteogenic potential. The CMCS incorporation promoted healing at the defect site, while the HA addition created binding sites for cell adhesion and growth, thereby improving scaffold bioactivity. Collectively, this study presents the development and validation of a multifunctional GelMA/CMCS/HA hydrogel, highlighting its dual capability for bone regeneration and tumor suppression within tumor-associated bone microenvironments.

The treatment of osteoporotic bone defects remains challenging due to the pathological microenvironment with impaired regenerative capacity and imbalanced bone formation and resorption. In this study, a methacrylated gelatin (GelMA) and methacrylated chitosan (CSMA) composite hydrogel coating is developed on poly(phthalazinone ether nitrile ketone) (PPENK), showing pro‐osteogenic and anti‐osteoclastic effects in vitro. Nano‐zinc oxide (nano‐ZnO), nano‐hydroxyapatite (nano‐HA), and phosphate‐ester‐functionalized alendronate (ALNP) are incorporated to confer antibacterial activity, promote osteogenesis, and enable enzyme‐responsive release of anti‐osteoporosis alendronate (ALN). SEM/EDS results confirm the homogeneous dispersion of the nanoparticles within the porous hydrogel. The stability of the hydrogel coating is verified by smooth scratch edges and no visible detachment in the cross‐hatch test. ALN release experiments confirm that the release amount of ALN is positively correlated with alkaline phosphatase (ALP) concentration, indicating significant enzyme‐responsive characteristics. In vitro studies show that the hydrogel coating on PPENK exhibits antibacterial activity. Nano‐HA and nano‐ZnO enhance the expression of osteogenic‐related genes and proteins in MC3T3‐E1 pre‐osteoblasts. The released ALN inhibits the receptor activator of nuclear factor‐κB ligand (RANKL)‐induced differentiation of RAW264.7 macrophages into osteoclasts. Overall, this surface modification strategy integrates enzyme‐responsive ALN release with composite hydrogel coating on PPENK, realizing antibacterial, osteogenic, and anti‐osteoclastic effects in vitro. This study reports a methacrylated gelatin/methacrylated chitosan/nano‐HA/nano‐ZnO/phosphate‐ester‐functionalized alendronate hydrogel coating on vinyl‐modified PPENK (G/C/H/Z/A@PPENK‐DB). The composite hydrogel coating enables enzyme‐responsive release of the anti‐osteoporotic drug alendronate via hydrolysis of phosphate ester bonds by alkaline phosphatase. In vitro tests confirm its favorable antibacterial, osteogenic, and anti‐osteoclastic effects, showing great potential for the repair of osteoporotic bone defects.

Tumor-associated macrophage (TAM) polarization is related to mitochondrial function, stability, and ROS production. The development of a mitochondria-targeted nanosystem with immunomodulatory impact on TAM repolarization is crucial for macrophage-targeted immunotherapy. First, zirconium(IV) coordinated with tetra-kis(4-caboxyphenyl) porphyrin to synthesize PCN-224, which encapsulated CO-releasing molecule 401 (CORM-401) via the hollow structures (PCN-CORM), and then, an amphiphilic copolymer containing triphenylphosphine (TPP) was synthesized to load PCN-CORM via self-assembly and surface-coated with hyaluronic acid (HA) to obtain the mitochondria-targeted nanosystem (HA@MR@PCN-CORM). Mediated by HA and TPP motifs, HA@MR@PCN-CORM efficiently targeted mitochondria of TAMs via HA-CD44 interaction and re-exposed TPP groups by digesting HA and caused reactive oxygen species (ROS) and carbon dioxide (CO) storm under a laser irradiation of 660 nm (20 mW cm–2), and the killing rate of M2-like TAMs reached 66.8% at a PCN-224 concentration of 15 μg/mL. Moreover, HA@MR@PCN-CORM, through depletion of GSH, inhibition of glutamine metabolism, and metabolic reprogramming, resulted in an increased ratio of M1/M2-type TAMs from 0.29 to 1.20 and an in vivo tumor volume inhibition rate of 93.4% with synergistically enhanced infiltration of CD3+/CD4+, CD3+/CD8+ T lymphocytes, and DCs in tumor tissues. The nanosystem was exhibited as a promising strategy for depletion and modulation of TAMs in an immunosuppressive tumor microenvironment for antitumor therapy.

A reduced graphene oxide (RGO)/gold nanorod (AuNR)/hydroxyapatite (HA) nanocomposite was designed and successfully synthesized for the first time. An anticancer drug, 5-fluorouracil (5FU), was chosen as a model drug to be loaded in RGO/AuNR/HA. The fabricated RGO/AuNR/HA-5FU showed robust, selective targeting and penetrating efficiency against HeLa cells due to the good compatibility and nontoxicity of HA, and showed excellent synergetic antitumor effects through combined chemotherapy (CT) by 5FU and photothermal therapy (PTT) by both RGO and AuNRs under near-infrared (NIR) laser irradiation. More importantly, this synergistic dual therapy based on RGO/AuNR/HA can also minimize side effects in normal cells and exhibits greater antitumor activity because of a multi-stage drug release ability triggered by the pH sensitivity of HA in the first stage and the combined photothermal conversion capabilities of RGO and AuNRs by means of the NIR laser irradiation in the second stage. This study suggests that the novel RGO/AuNR/HA multi-stage drug delivery system may represent a promising potential application of multifunctional composite materials in the biomedical field.

Biofilm-associated infections and the lack of successful tissue integration of biomaterial surfaces are the two main barriers to the long-term service of implanted biomaterials. Development of novel biocompatible antimicrobial materials has provided insights into their potential biomedical applications. Many clinical studies have successfully proved that hydroxyapatite coating has excellent osteogenic activity but lacks antibacterial infection in the early stages after implantation. Rare earth (Re) elements have become promising antibacterial biocides and bone-forming effects. Antibacterial capacity of 14 rare earth elements (Eu, Gd, Ce, Nd, Y, La, Pr, Er, Sm, Ho, Tb, Yb, Lu, Dy) was assayed. The gadolinium (Gd) showed outstanding broad-spectrum antibacterial activity against both Gram-positive and Gram-positive bacteria. Here, we report Gd-HA coatings deposited on titanium (Ti) substrate by liquid thermal spraying. The grain size of Gd-HA decreased slightly after Gd 3+ incorporation. The antibacterial properties of Gd-HA composite coatings were determined against Gram-negative pathogens Escherichia coli and Gram-positive pathogens Staphylococcus epidermidis . The anti-infection performances were assessed by examining bacteria adhesion and biofilm formation on the coatings. The in vitro cytotoxicity of the Gd-doped HA coatings was further measured on human osteoblast cell line by CCK-8 method. The thermal sprayed HA-Re composite coatings show improved antimicrobial and biocompatible properties and great applicable potential in orthopedics.

The increasing demand for carbon fiber/polyether-ether-ketone (CF/PEEK) composites in marine and aerospace engineering is driven by their potential for weight reduction and enhanced structural flexibility. However, the inherent weak interface poses a challenge, leading to stress concentration and structural failures. Herein, aqueous PEKC-COOH sizing agent on CF as a soft unit (SCF) and hydroxyapatite (HA) nanomaterial as a rigid unit establish a regulable active interface structure, synergistically enhancing the interface adhesion through mechanical interlocking and chemical bonding of the CF/PEEK composites. Under external loading, the PEKC-COOH sizing agent allows for uncurling of molecular chains at the interface, promoting slip of HA nanomaterials and dissipating a significant amount of energy during fracture progression. The SCF-HA/PEEK composites exhibited significant enhancements in interlaminar shear strength and flexural strength, with an increase of 65.1% and 49.8%, respectively, compared with the pristine composites. Meanwhile, the salt-spray resistance of the composites has been significantly improved. Following a 72-h salt spray test, the ILSS retention ratios of modified SCF-GB/PEEK composites were determined to be 91.7%. Remarkably, the use of cationic, non-ionic, and zwitterionic surfactants reduced the growth time of HA on the fiber surface in simulated body fluids to 15 min compared to several days using conventional methods. This study not only improves production efficiency and reduces manufacturing costs but also provides a research foundation for industrial manufacturing. Furthermore, the novel interface structure design broadens the applicability of CF/PEEK composites in hygrothermal and salt-spray environments. Highlights Regulable interface was formed by in situ growth of HA on fiber sizing agent. HA nanomaterial growth time in SBF was reduced from several days to 15 min. Modified composites increase ILSS by 65.1% and flexural strength by 49.8%. Modified composites maintain 91.7% ILSS after a 72-h salt-spray resistance test.

Breast cancer is a malignant tumor that poses a significant threat to women's health and single therapy fails to play a good oncological therapeutic effect. Synergistic treatment with multiple strategies may make up for the deficiencies and has gained widespread attention. In this study, sulfhydryl-modified hyaluronic acid (HA-SH) was covalently crosslinked with polydopamine (PDA) via a Michael addition reaction to develop an injectable hydrogel, in which PDA can be used not only as a matrix but also as a photothermal agent. After HSA and paclitaxel were spontaneously organized into nanoparticles via hydrophobic interaction, hyaluronic acid with low molecular weight was covalently linked to HSA, thus conferring effectively delivery. This photothermal injectable hydrogel incorporates PTX@HSA-HA nanoparticles, thereby initiating a thermochemotherapeutic response to target malignancy. Our results demonstrated that this injectable hydrogel possesses consistent drug delivery capability in a murine breast cancer model, collaborating with photothermal therapy to effectively suppress tumor growth, represented by low expression of Ki-67 and increasing apoptosis. Photothermal therapy (PTT) can effectively stimulate immune response by increasing IL-6 and TNF-α. Notably, the treatment did not elicit any indications of toxicity. This injectable hydrogel holds significant promise as a multifaceted therapeutic agent that integrates photothermal and chemotherapeutic modalities.

The burgeoning implantable biodevices have unlocked new frontiers in healthcare, promising personalized monitoring strategies tailored to specific needs. Herein, hyaluronic acid (HA) is harnessed to create fully biocompatible, acidity-sensitivity and cleft-adjustable neuromorphic devices. These HA-biodevices exhibit remarkable sensitivity to pH variations, effectively mimicking biological acid-sensing ion channels (ASICs) through protonation reactions between electronegative atoms and hydrogen ions, even at ultralow driving voltage (5 mV). They can monitor joint cartilage acidity by tracking changes in proton concentration and successfully diagnose the onset of arthritis. Furthermore, by adjusting the synaptic device's cleft distance, which determines responsiveness, power efficiency and plasticity, HA-based neuromorphic devices can be tailored to meet the unique demands of various implantation sites, providing both high-sensitivity and low-heat dissipation, thus broadening their application scopes. Moreover, the HA-biodevices maintain stable performance across various bending degrees, up to a curvature radius of 7.5 mm, with flexibility and deformation resilience enabling installation on joints of varying curvatures. The combination of all-biocompatibility, high sensitivity, low heat dissipation, ultralow low power (2 pW), and extraordinary deformation tolerance paves the way for the development of versatile, multipurpose medical monitoring devices with immense potential in the field of healthcare.

Developing bone scaffolds with biomimetic architecture and bioactive signaling is essential for effective orthopedic repair. In this study, 16.6 wt% MgO incorporated hydroxyapatite (MgO/HA) scaffolds with a Gyroid Triply Periodic Minimal Surface (TPMS) architecture were fabricated using a multi-stage route combining Digital Light Processing (DLP) 3D printing and Spark Plasma Sintering (SPS). The resulting scaffolds maintained ∼60% porosity and exhibited an increase in compressive strength (7.27 MPa) compared to HA (4.24 MPa). The incorporation of MgO led to an increase in the degradation rate, facilitating the sustained release of bioactive Mg²⁺ ions. In vitro biological evaluations demonstrated a potent multi-lineage cellular response: the scaffolds enhanced rat mesenchymal stem cell (rMSC) proliferation and upregulated key osteogenic markers (RUNX2, ALP, and OCN), while simultaneously stimulating human umbilical vein endothelial cells (HUVECs) by significantly upregulating angiogenic gene expressions (VEGF and Ang-1). These results indicate that the combination of MgO doping and TPMS architecture successfully establishes a robust osteogenesis-angiogenesis coupling effect alongside optimized mechanical performance, providing a highly promising candidate platform for patient-specific bone tissue engineering.

Poly(TA-Arg) coating with self-supplying NO fabricated via ethanol-water system. • Poly(TA-Arg) sample possesses a low degradation rate. • HA@CS nanoparticles synthesized with optimal HA:CS ratio. • HA@CS sample promotes endothelialization via PI3K-Akt-eNOS and Nrf2 pathways. Rapid degradation and inadequate re-endothelialization hinder the clinical application of biodegradable magnesium (Mg) alloy stents, while thrombosis and bacterial colonization further compromise early implant performance. To address these challenges, a hierarchical bilayer coating, consisting of an inner poly(α-thioctic acid)-L-arginine (Poly(TA-Arg)) layer and an outer hyaluronic acid/chitosan (HA@CS) nanoparticle layer (particle size, ∼240 nm), was fabricated on a fluorinated ZE21B Mg alloy. The Poly(TA-Arg) layer supported endothelial cell (EC)-mediated endogenous nitric oxide (NO) production. It also provided strong interfacial adhesion and corrosion protection, reducing the corrosion current density to 5.60 × 10 −8 A cm −2 . The HA@CS layer exhibited antioxidant activity, prolonged clotting time, low hemolysis, and > 99% antibacterial efficacy, as determined by plate counting. The bilayer coating promoted EC proliferation while inhibiting smooth muscle cell growth. RNA sequencing suggested that the pro-endothelialization effect was associated with PI3K-Akt- endothelial nitric oxide synthase-related signaling and an Nrf2-related antioxidant response. In a rat abdominal aorta model, the coated wire was surrounded by a thin, well-organized neointima with endothelial coverage, minimal inflammation, and no evident thrombosis. No obvious histopathological abnormalities were observed in major organs, and hematological parameters remained within normal ranges. This multifunctional coating provides a potential strategy for improving the vascular compatibility of biodegradable Mg stents. Repairing orbital bone defects remains a complex challenge in craniofacial surgery. Existing bone repair materials are plagued by issues such as intricate preparation processes and inadequate consideration of the spatiotemporal characteristics of bone healing. This study aimed to enhance the osteoinductive activity of poly (glycolide-co-caprolactone) (PGCL)-based materials through bicomponent modification, thereby providing a basis for the design of clinical bone repair materials. Hydroxyapatite/calcium sulfate/PGCL (HA/CS/PGCL) composite microspheres were fabricated via electrostatically assisted spray evaporation (ESASE). Their physicochemical properties and in vitro/in vivo osteogenic efficacy were evaluated using degradation-mineralization assays, MC3T3-E1 cell differentiation assessments and a rabbit orbital bone defect model. Innovatively, CS mesocrystal pore structure regulation was introduced to synergize with HA, achieving multidimensional optimization of surface hydrophilicity, sequential calcium ion release and mechanical properties. Results showed that in vitro mineralization formed native-like hydroxyapatite structures within 14 days. On HA/CS/PGCL surfaces, the proliferation activity of MC3T3-E1 cells reached 1.8 times that of the PGCL group after 7 days, with a 1.6-fold increase in alkaline phosphatase activity. In vivo, the composite microspheres achieved a new bone volume ratio (BV/TV) of 78.72% at 10 weeks, resulting in complete closure of the bone defect. Their trabecular structural parameters (trabecular thickness, Tb.Th; trabecular number, Tb.N) outperformed those of the single-component groups. Histological staining confirmed their ability to induce ordered collagen arrangement and the formation of dense new bone. In conclusion, the HA/CS synergistic modification strategy achieved the sequential release of calcium ions during bone repair and spatial regulation of the microsphere surface pore structure. The superior performance of HA/CS/PGCL microspheres in repairing complex bone defects lays an experimental foundation for clinical translation. Further research into the interactions between ion release and cell signaling is warranted.
