An ultrasensitive chemiluminescent immunosensor for the detection of human leptin using hemin/G-quadruplex DNAzymes-assembled signal amplifier

Yuezhen He,Xiaoxun Wang,Yuzhong Zhang,Feng Gao,Yongxin Li,Hongqi Chen,Lun Wang

In this work, we reported a sensitive chemiluminescent immunosensor for the detection of human leptin by using hemin/G-quadruplex DNAzymes to amplify detection signal. In this sensing system, the primary antibody (anti-human leptin) was firstly bound to the 96-well plates, and human leptin and biotinylated secondary antibody were successively combined to form sandwich-type immune complex through specific interactions. Then streptavidin labeled with hemin/G-quadruplex DNAzymes was assembled to the sandwich-type immunocomplex by streptavidin–biotin interaction. The DNAzymes exhibited an excellent catalytic activity to the chemiluminescent reaction of luminol with hydrogen peroxide in strong alkaline solution, leading to significant enhancement in response signal. Under the optimum conditions, the proposed immunosensor showed high sensitivity and selectivity with a low detection limit of 1.9   pg   mL −1 and a wide linear response range of human leptin from 10 to 1000   pg   mL −1 . The immunosensor was used to detect human leptin in serum, and the results were in good agreement with the data obtained by conventional ELISA method.

Proximity hybridization triggered hemin/G-quadruplex formation for construction a label-free and signal-on electrochemical DNA sensor

Fenglei Gao,Taotao Fan,Jing Wu,Sha Liu,Yan Du,Yao Yao,Fuyi Zhou,Yu Zhang,Xianjiu Liao,Deqin Geng

We describe a novel label-free and signal-on electrochemical DNA sensing platform via proximity hybridization triggered hemin/G-quadruplex formation based on the direct electron transfer of hemin . The thiolated modified G-DNA1 was first immobilized onto the Au electrode surface . In the presence of target DNA, Y-junction-structure ternary complex can be formed to trigger the proximity assembly of G-DNA1, hemin, and G-DNA2, which leads to the formation of hemin/G-quadruplex for generation an amplified electrochemical signal by differential pulse voltammetry . The observed signal gain was sufficient to achieve a demonstrated detection limit of 54 fM, with a wide linear dynamic range from 10 −13 to 10 −9 M and discriminated mismatched DNA from perfect matched target DNA with a high selectivity. Benefiting from the one step proximity dependent hemin/G-quadruplex formation, the assay method is extremely simple and can be carried out within 40   min. The advantages of free of any label conjugation step, and versatility make it a promising candidate for point-of-care testing and commercial application.

Determination of phenol by flow-injection with chemiluminescence detection based on the hemin-catalysed luminol–hydrogen peroxide reaction

Wenwen Liu,Wei Cao,Weihua Liu,Kang Du,Pixue Gong

This study established a novel flow injection (FI) methodology for the determination of phenol in aqueous samples based on luminol chemiluminescence (CL) detection. The method was based on the inhibition that phenol caused on the hemin-catalysed chemiluminescence reaction between luminol and hydrogen peroxide in alkaline solution. Optimum conditions and possible mechanisms have been investigated. The linear range was 2.0   ×   10 −9 to 4.0   ×   10 −7   g   mL −1 for phenol. The proposed method is sensitive with a detection limit of 4.0   ×   10 −10   g   mL −1 . The relative standard deviation for 11 measurements was 2.3% for 1.0   ×   10 −7 g   mL −1 phenol. The method was applied for the determination of phenol in waste water samples. The results obtained compared well with those by an official method.

Colorimetric and photothermal dual-modal analysis of a cancer-related gene based on G-quadruplex-hemin cascade signal amplification

Hongling Tao,Yanyue Chen,Jiawen Liu,Jiali Liu,Yunhui Yang,Rong Hu

p53 gene is one of the most important anticancer genes and has been recognized as a typical biomarker for the early diagnosis of cancers. In this paper, a portable point-of-care testing (POCT) platform was developed for the colorimetric and photothermal dual-modal analysis of the p53 gene. Long concatemer G4 structures with high catalytic activity were formed by a target-driven hybridization chain reaction, and their catalytic properties can further be enhanced with the help of iodide ions. This dual-modal method realized the sensitive multicolor visual point-of-care detection of the p53 gene, where color variations from colourless to blue were easily distinguished by the naked eye. The temperature output was achieved by effective photothermal conversion and regulation guided by the accumulated concatemer G4 complexes in the TMB–H2O2 system, avoiding the need for complex instruments. The proposed dual-modal assay for p53 gene detection is cost-effective, simple, highly sensitive (3.34 pM), stable, and remarkably selective. It was also applied to determine the concentration of the p53 gene in spiked serum samples.

Streptomycin-Triggered Suppression of G-Quadruplex/Hemin DNAzyme Activity: A Rapid Label-Free Colorimetric Strategy for Food Safety Monitoring

Siying Ma,Bingqian Weng,Yingying Huang,Shaoyun Chen

A rapid, simple, and stable detection method for streptomycin has been established based on the coloration reaction of 3,3′,5,5′-tetramethylbenzidine (TMB) catalyzed by peroxidase-mimicking DNAzymes (PMDs). It is discovered that the catalytic activity of PMDs constructed with G-quadruplex and hemin is weakened in the presence of streptomycin. In the presence of streptomycin, the OD 450nm value of the TMB colorimetric sample catalyzed by PMDs is significantly lower than that of the control sample (without streptomycin). Hence, the detection of streptomycin can be accomplished by detecting the change of OD 450nm (△OD 450nm ). Furthermore, when the concentration of the substrate TMB is lower, the catalytic activity of PMDs is inhibited more significantly by streptomycin. Under optimized conditions, this method has good detection specificity, could effectively distinguish streptomycin from other antibiotics (ampicillin, chloramphenicol, geneticin, and kanamycin), has a limit of quantitation (LOQ) of 5 mg/L, excellent linear performance, good repeatability, and a total detection time of less than 2 h, and can be employed for the detection of actual samples (laver and egg drop soup). This method circumvents the complex preparation process of experimental materials, is simple to operate, does not require fluorescent labeling, and is of low cost, providing a novel method for food safety supervision.

Facile synthesis of hemin@PMo12 nanocomposite with peroxidase-mimicking activity for colorimetric detection of ascorbic acid in fresh fruits

Zhongmei Chi,Shiqi Chu,Jiahui Ma,Guocheng Liu,Huanxiang Wang,Xiuli Wang

Facile synthesis of Hemin@PMo 12 via manual grinding, no complex equipment and harsh conditions required. • Solid-phase Hemin@PMo 12 enables easy recovery, good reusability and high stability. • Hemin@PMo 12 shows excellent peroxidase-mimicking activity, outperforming natural HRP in kinetic parameters. • The sensor achieves accurate AA detection in apple samples via UV–vis and smartphone analysis. Polyoxometalate-based nanocomposites, as promising peroxidase mimetics for biosensing, have drawn extensive attention. In this study, a facile manual grinding method was developed for the synthesis of Hemin@PMo 12 nanocomposites from hemin and phosphomolybdic acid (H 3 PMo 12 ), without complicated equipment or harsh reaction conditions. Hemin@PMo 12 exhibited excellent peroxidase-mimicking activity, capable of catalyzing the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H 2 O 2 ) to form a blue oxidized product. Ascorbic acid (AA) could specifically reduce the oxidized TMB, leading to a noticeable color fading and a significant decrease in absorbance, which enabled the colorimetric detection of AA. The proposed method based on Hemin@PMo 12 demonstrated a wide linear range of 3–200 μM and a low limit of detection (LOD) of 1.1 μM for AA detection. Furthermore, the developed sensor was successfully applied to the quantitative determination of AA in fresh fruit samples (e.g., apple), yielding satisfactory recoveries ranging from 98.4% to 102.7%. Notably, the solid-phase nature of Hemin@PMo 12 endowed it with prominent advantages of easy recovery from the reaction system and good reusability. These advantages well made up for the inherent drawbacks of traditional nanozymes, namely the difficulty in separation and the restriction to single-use applications. With the merits of simple synthesis, easy recovery, reusability, high sensitivity, and good selectivity, Hemin@PMo 12 provided a promising candidate for the development of practical, low-cost colorimetric sensors for AA detection in food analysis and related fields. A portable and quantitative detection of xanthine and xanthine oxidase based on cascade enzymatic reactions of Fe3O4@MWCNTs/Hemin

Wang Yijie,Li Qiulan,Xia Qinghai,Yang Yaling,Yang Dezhi

Sensitive and effective detection of xanthine oxidase (XOD) is essential for assessing human health. Herein, a dual-enzyme-based cascaded reaction system was developed for xanthine and xanthine oxidase assay based on the peroxidase (POD)-like catalytic activity of Fe 3 O 4 @MWCNTs/Hemin nanocomposite. The XOD could catalyze xanthine to produce H 2 O 2 , which is subsequently converted by Fe 3 O 4 @MWCNTs/Hemin into hydroxyl radicals (·OH) to oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) to produce a light blue. Notably, it is demonstrated that the modification of MWCNTs and Hemin could significantly enhance the POD-like activity from 9.78 × 10 −8  M s −1 ( V max value of Fe 3 O 4 ) to 23.34 10 −8  M s −1 ( V max value of Fe 3 O 4 @MWCNTs/Hemin), which makes the detection of xanthine oxidase more sensitive. By using the smartphone to capture the colorimetric signal of oxidized TMB (oxTMB), the assay can detect xanthine and XOD as low as 0.250 µM and 0.216 mU mL −1 , respectively. The reliability of the method in human serum suggested satisfactory spiked recoveries in the range of 92.350–105%. This work provided a new insight for Fe 3 O 4 -based nanozyme design and monitoring XOD level in blood. Graphical Abstract

Generation of high-valent iron-oxo porphyrin cation radicals on hemin loaded carbon nanotubes for efficient degradation of sulfathiazole

Sijing Chen,Zhen Li,Longsheng Wu,Lu Liu,Jingping Hu,Huijie Hou,Sha Liang,Jiakuan Yang

Hemin has attracted considerable interest as an efficient catalyst recently, however, its direct application is inefficient due to severe molecular aggregation. Immobilizing hemin on various supports is a feasible approach to address this issue. In this work, a CNTs-hemin catalyst was prepared by loading hemin onto multiwalled carbon nanotubes (CNTs) through ball milling. Compared with hemin, CNTs-hemin demonstrates remarkably enhanced performance in the peroxymonosulfate system, with a 650-fold improvement of apparent rate constant, reaching 97.8% degradation of sulfathiazole in 5 min. High-valent iron-oxo porphyrin cation ((Porp) +• Fe IV =O) radicals are proposed as the dominant reactive species in the CNTs-hemin/peroxymonosulfate system instead of sulfate radicals (SO 4 •- ), hydroxyl radicals (•OH), superoxide radicals (O 2 •- ) and singlet oxygen ( 1 O 2 ). More in-depth mechanisms reveal that the strong electron transfer between CNTs and hemin promotes the generation of (Porp) +• Fe IV =O radicals through a heterolysis pathway. This research enriches the understanding of the catalytic mechanism of supported biomimetic catalysts for PMS activation and provides a perspective on the role of support materials for catalytic activity .

Label-free and dual-mode biosensor for HPV DNA based on DNA/silver nanoclusters and G-quadruplex/hemin DNAzyme

Junyang Chen,Mengke Wang,Chenyu Zhou,Jiabao Zhang,Xingguang Su

Specific and cost-effective methodologies for human papillomavirus (HPV) gene detection are significant for clinical diagnosis and cancer control. Herein, a label-free and fluorimetric/colorimetric dual-mode sensing strategy was developed for the quantitative determination of HPV DNA based on the integration of fluorescent DNA-silver nanoclusters (DNA/AgNCs) and G-quadruplex/hemin DNAzyme. The fluorimetric sensing strategy was based on the phenomena that the fluorescence enhancement of DNA/AgNCs obtained in proximity of guanine-rich DNA sequences and the photoinduced electron transfer (PET) effect between the electron donor (DNA/AgNCs) and electron receptor (G-quadruplex/hemin). The colorimetric sensing strategy was relied on the peroxidase-like activity of G-quadruplex/hemin DNAzyme. By integrating DNA/AgNCs and DNAzyme, this dual-mode strategy could produce two independent signals to improve the analytical diversity and accuracy. Under optimized conditions, the fluorimetry and colorimetry of the strategy displayed a linear range of 0.01–4 and 0.02–4 nM, with the low detection limit of 2.3 and 5.2 pM, respectively. Additionally, this dual-mode strategy has been successfully applied to HPV DNA analysis in different real samples with excellent results. Moreover, the sensing platform could be developed for different HPVs DNA assay by only adjusting the recognition sequence, which provided a universal strategy for various kinds of virus analysis.

A colorimetric and fluorescent dual-mode sensor based on bifunctional G-quadruplex-hemin complex for the determination of Pb2+

Tai Ye,Haohao Chen,Long Bai,Min Yuan,Hui Cao,Liling Hao,Xiuxiu Wu,Fengqin Yin,Fei Xu

Due to the threat of lead pollution to health, environmental and food safety, developing simple and fast detection methods is highly required. Whereas, traditional single-mode probe suffers from limited application scenario. In this study, a colorimetric and fluorometric dual-mode probe for Pb 2+ determination was constructed by using bifunctional G-quadruplex-hemin complex. In this dual-mode probe, enzyme strand and substrate strand of 8–17 DNAzyme are labeled with G-quadruplex-hemin complex and fluorophore, respectively. In the absence of Pb 2+ , the self-assembly of enzyme strand and substrate strand inhibits intrinsic mimic peroxidase of G-quadruplex-hemin complex by base-pairing, which also quench the fluorescence via in proximity effect. When the DNAzyme is activated by Pb 2+ , the quenched fluorescence is restored as well as the inherent peroxidase mimetic activity, leading to dual signal output. Under optimal conditions, this dual-mode probe exhibit a good linear relationship between logarithm of Pb 2+ concentration and signal difference within the range from 1.5 nM to 20 nM and 0.5 nM to 10 nM for colorimetric and fluorescence mode, respectively. The detection limits for the corresponding mode were estimated to be 1.29 nM and 0.16 nM, respectively. This dual-mode probe also successfully applied for the spiked river water assay with satisfactory recovery in the range of 93.2 %–115.3 %. This work paves a new way for DNAzyme based dual-mode probe construction.

Cu–MOF/hemin: a bionic enzyme with excellent dispersity for the determination of hydrogen peroxide released from living cells

Hong Cui,Shuaishuai Cui,Siyuan Zhang,Qiuju Tian,Yunfeng Liu,Ping Zhang,Mingxiu Wang,Jialing Zhang,Xiangjun Li

The stability, repeatability and sensitivity of an electrochemical biosensor material are closely connected with the dispersibility of metal organic frameworks (MOFs) in aqueous media. Herein, a nanocomposite based on Cu–MOF/hemin, which is not only highly water-soluble but also simple and efficient in synthesis, was used for the construction of a non-enzymatic sensor to detect hydrogen peroxide (H2O2). The Cu–MOF/hemin was characterized via scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS)-mapping, X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS) and thermal gravimetric analysis (TGA), which indicate that hemin and the Cu–MOF were successfully combined. As a H2O2 electrochemical biomimetic enzyme, the Cu–MOF/hemin exhibited excellent electrocatalytic performance, which was confirmed by the electrochemical experiments and chromogenic reactions, and the possible mechanism of the reactions has been deduced. The electrochemical sensor based on the biomimetic enzyme exhibited an extended linear detection range from 0.01–5.0 mM (R = 0.998), low detection limit of 4.14 μM, and high selectivity and stability under the optimized conditions. More importantly, the practical application ability of the sensor was verified by the test of H2O2 in human serum samples and it could be used for the real-time detection of H2O2 released from living cells with satisfactory results. Therefore, this novel nanocomposite has certain potential in preparing electrochemical sensing platforms for nonenzymatic biosensing and provides a new method for clinical diagnosis and real-time monitoring.

Sensitive and label-free chemiluminescence detection of malathion using exonuclease-assisted dual signal amplification and G-quadruplex/hemin DNAzyme

Hao Wu,Jun Wu,Hongyong Wang,Yaling Liu,Guoqing Han,Pei Zou

Malathion is one of the most commonly used organophosphorus pesticides that can cause serious harm to the ecological environment and human health. Herein, we demonstrated a label-free chemiluminescent aptasensor for the sensitive detection of malathion based on exonuclease-assisted dual signal amplification and G-quadruplex/hemin DNAzyme. Upon the addition of malathion, the aptamer probe specifically bound to the target to form a complex malathion-S3, leaving a duplex S1-S2. The complex malathion-S3 was digested by exonuclease I and the target was released. The released target was recycled to perform exonuclease I-assisted signal amplification. Furthermore, after treatment with exonuclease III, the duplex S1-S2 was converted into the secondary target ST. The secondary target ST interacted with the hairpin H1 to form a complex H1-ST, which was further digested by exonuclease III and released the secondary target. The released secondary target was recycled to perform exonuclease III-assisted signal amplification. After complete amplification, large numbers of G-quadruplex/hemin DNAzymes were generated. Under the optimal experimental conditions, the prepared aptasensor showed an excellent linear response to malathion with a detection limit of 0.47 pM. The relative standard deviations were in the range of 4.2–6.9%. Moreover, the aptasensor was successfully applied to detect malathion in spiked food and traditional Chinese medicine samples.
