Prediabetes, a prevalent metabolic condition characterised by elevated blood glucose levels not reaching type 2 diabetes mellitus criteria, represents a critical intervention window for diabetes prevention. Konjac glucomannan (KGM) exhibits efficacy in diabetes management, but the structure–function relationships underlying its bioactivity are poorly defined. This study investigated the impact of acetylation degree (DS) on KGM's anti-prediabetic potential. Structural characterisation revealed that acetylation and deacetylation altered DS while preserving the KGM's backbone, accompanied by changes in molecular weight and viscosity. Given the pivotal role of gut microbiota in the development of diabetes, in vitro fermentation experiments using faeces from prediabetic individuals revealed that KGM with varying DS (AcKGM) enriched Bacteroides uniformis and Bacteroides coprocola , with enrichment positively correlating with DS. Consistently, mono-culture assays demonstrated preferential utilization of highly acetylated AcKGM by both species. Prediabetic animal experiments further demonstrated that AcKGM improved glucose and lipid metabolism, suppressed inflammation, and ameliorated hepatosteatosis, in a DS-dependent manner. Notably, these effects were associated with DS-dependent enrichment of B. uniformis and upregulation of beneficial faecal metabolites, including D-pantothenic acid. Collectively, these findings establish that higher DS enhances KGM's efficacy in modulating gut microbiota and alleviating prediabetes, providing mechanistic insights for structure-guided optimisation of KGM-based therapeutics.

The objective of this study was to fabricate and characterize antimicrobial films based on Agar/Konjac glucomannan (KA) incorporated with carvacrol (CV) (1%, 1.5% and 2%). The physical, structural and antimicrobial properties were studied to evaluate the effects of carvacrol on the composite films. Results indicated that the addition of 2% CV markedly enhanced the tensile strength (36.72 MPa), elongation at break (100.19%), hydrophobic properties and UV barrier properties compared with KA film. X-ray diffraction, Fourier transform infrared spectroscopy , and scanning electron microscopy indicated good compatibility and the formation of intermolecular hydrogen bonds. Additionally, the addition of 2% CV showed strong antimicrobial activities against the most common food spoiling bacteria, Staphylococcus aureus and Escherichia coli . Finally, the potential application of the films was evaluated by the packing test of chicken, where KA-2% CV was able to prolong the shelf life of refrigerated chicken breast from 5 days to 9 days. Overall, the composite films incorporating 2% carvacrol was a promising antimicrobial packing material to displace petrochemical-based plastics to extend the shelf life of foods .

In this work, konjac glucomannan (KGM)-based film reinforced with pullulan (PL) and acai berry extract (ABE) was developed by solvent casting method. The as-prepared films performed pH-sensitive properties, which can be potentially applied for fish freshness detection. Rheology , Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM) were used to characterize chemical structure and morphology of ABE-loaded KGM/PL (KP) films (KP-ABE). FT-IR spectrum indicated that hydrogen bond dominated the formation of KP-ABE films. Adding PL contributed to enhanced mechanical properties of KGM film with increased tensile strength (TS) from 21.25 to 50.27 MPa and elongation at break (EAB) from 10.64 to 19.19 %. Incorporating ABE upgraded flexibility, UV-shielding, thermostability , water barrier (decreased Water vapor permeability (WVP) from 2.07 to 1.67 g·mm/m 2 ·day kPa), antioxidant, and antibacterial ability of KP films, but weakened TS. In addition, KP-ABE films can reflect fish freshness in real time through color variability. Therefore, KP-ABE films exhibited potential applications in intelligent food packaging materials.

A composite coating containing konjac glucomannan (KGM) and CaCO 3 was fabricated on AA5052 alloy through a self-curing process, and its surface morphology and component were characterized by scanning electron microscope (SEM), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD) and thermogravimetric analysis (TGA). The effect of Ca(OH) 2 content on the protective property of the composite coating for aluminum alloy in 3.5 wt% NaCl solution was evaluated by electrochemical impedance spectroscopy measurements. Moreover, the inhibitory performance of the coating on the pitting corrosion of metal was examined by potentiodynamic polarization curves and atomic force microscopy (AFM). The images of scanning electron microscope show the composite coating with a thickness of 50 μm has well-defined branch-shaped structure. The FT-IR spectra confirm the successful fabrication of the polymer coating through the formation of CaCO 3 . The results of electrochemical impedance spectroscopy suggest that the resistance of the aluminum alloy sample coated with such protective layer is improved by six orders of magnitude, compared with the bare metal. After 60 d of immersion, the polarization curve data and AFM analysis show the pitting corrosion occurring on AA5052 surface is prevented due to the barrier property of coating.

Orally administered colon-targeted delivery vehicles are of major importance in the treatment of inflammatory bowel disease (IBD). However, it remains a challenge to maintain the integrity of such delivery vehicles during treatment, particularly in the gastric environment, which may cause untimely drug release before reaching the targeted colon. Herein, an oral colon-targeted drug delivery system (OCDDS) based on acetylated konjac glucomannan (AceKGM) has been developed in this work, which accomplishes colonic localization release and targets local inflammatory macrophages. The AceKGM nanoparticle-loading curcumin (Cur) was successfully fabricated by emulsion solvent evaporation techniques. DLS, AFM, and SEM were used in order to evaluate the nanoparticles’ diameter as well as their in vitro drug release profile, and reactive oxygen species (ROS) scavenging results showed that the OCDDS considerably retained the activity of Cur treated with simulated gastric fluid (SGF) and controllably released in simulated intestinal fluid (SIF). In addition, the adhesion experiment results indicated that the nanoparticle could accumulate on the colonic macrophages. Evaluations in colitis mice showed that the treatment significantly alleviated the symptoms of colitis by decreasing the local level of myeloperoxidase (MPO) and the disease activity index (DAI) score in mice. In summary, the results of our research demonstrate that Cur–AceKGM nanoparticles exhibit significantly improved therapeutic efficacy compared to orally administered free Cur and can be developed as an effective drug delivery vehicle for IBD treatment.

To enrich the application of nanocomposite hydrogels, we introduced two types of nanocellulose (CNC, cellulose nanocrystals; CNF , cellulose nanofibers) into the soy protein isolate(SPI)- konjac glucomannan (KGM) composite hydrogel system, respectively. The similarities and differences between the two types of nanocellulose as textural improvers of composite gels were successfully explored, and a model was developed to elaborate their interaction mechanisms. Appropriate levels of CNC (1.0 %) and CNF (0.75 %) prolonged SPI denaturation within the system, exposed more buried functional groups, improved molecular interactions , and strengthened the honeycomb structural skeleton formed by KGM. The addition of CNC resulted in greater gel strength (SKC1 2708.53 g vs. Control 810.35 g), while the addition of CNF improved the elasticity (SKF0.75 1940.24 g vs. Control 405.34 g). This was mainly attributed to the reinforcement of the honeycomb-structured, water binding and trapping, and the synergistic effect of covalent (disulfide bonds) and non-covalent interactions (hydrogen bonds, ionic bonds) within the gel network. However, the balance and interactions between proteins and polysaccharides were disrupted in the composite system with excessive CNF addition (≥0.75 %), which broken the stability of the honeycomb-like structure. We expect this study will draw attention on potential applications of CNC and CNF in protein-polysaccharide binary systems and facilitate the creation of novel, superior, mechanically strength-regulated nanofiber composite gels.

In this work, we prepared polyvinyl pyrrolidone (PVP) microfibers incorporated water-in-oil (W/O) emulsions. The W/O emulsions were fabricated by hexadecyl konjac glucomannan (HKGM, emulsifier), corn oil (oil phase) and purple corn anthocyanins (PCAs, water phase). The structures and functions of emulsions and microfibers were characterized by confocal laser scanning (CLSM) and scanning electron microscopy (SEM), Fourier transform infrared (FT-IR), Raman and nuclear magnetic resonance (NMR) spectroscopy. The results showed that W/O emulsions exhibited good storage stability for 30 d. Microfibers presented ordered and uniform arrays. Compared with pure PVP microfiber films, the addition of W/O emulsions with PCAs improved the water resistance (WVP from 1.28 to 0.76 g mm/m 2 day kPa), mechanical strength (Elongation at break from 18.35 % to 49.83 %), antioxidation (free radical scavenging rate from 2.58 % to 16.37 %), and antibacterial activity (inhibition zone against E. coli : 27.33 mm and inhibition zone against S. aureus : 28.33 mm) of microfiber films. Results showed that microfiber film exhibited controlled release of PCAs in W/O emulsions, and about 32 % of the PCAs were released from the microfiber film after 340 min. The as-prepared microfiber films exhibited potential applications for food packaging.

The inferior anti-washout property of injectable calcium phosphate cement (CPC) limits its wider application in clinic. In this study, the improvement of anti-washout performance of CPC by addition of konjac glucomannan or guar gum, which was dissolved in the CPC liquid, was first studied. The influence of KGM/GG blend with different mass ratios on the anti-washout property, compressive strength and in vitro cytocompatibility of CPC was estimated. The results revealed that small amount of KGM or GG could obviously enhance the anti-washout property of CPC. Moreover, the washout resistance efficiency of KGM/GG blend was better than KGM or GG alone. The addition of KGM/GG blend slightly shortened the final setting time of CPC. Although the introduction of KGM/GG blend reduced the compressive strength of CPC, the compressive strength still reached or surpassed that of human cancellous bone. The best KGM/GG mass ratio was 5:5, which was most efficient at not only reducing CPC disintegration, but also increasing compressive strength. The addition of KGM/GG blend obviously promoted the cells proliferation on the CPC. In short, the CPC modified by KGM/GG blend exhibited excellent anti-washout property, appropriate setting time, adequate compressive strength, and good cytocompatibility, and has the potential to be used in bone defect repair.
The addition of KGM/GG blend significantly improved the anti-washout property of CPC. The best KGM/GG mass ratio was 5:5, which was most efficient in reducing the CPC disintegration.

Neutral polysaccharides such as konjac glucomannan, starch and pullulan are abundant in nature and have unique property. Their nanofibers hold great potential for biomedicine, which however, are seldom applied in the field due to the lack of crosslinking method. In this work, we report a periodate oxidation - adipic acid dihydrazide (ADH) crosslinking strategy to prepare robust and biocompatible neutral polysaccharide nanofibers. Neutral polysaccharides with adjacent dihydroxyl groups are firstly partially oxidized with periodate to give dialdehyde polysaccharides, and their electrospun nanofibers are then crosslinked with ADH to form dihydrazone crosslinkers. The resulting crosslinked neutral polysaccharide nanofibers exhibit high water resistance and excellent mechanical properties because of the high reactivity of Schiff base crosslinking reaction. Moreover, the crosslinked neutral polysaccharide nanofibers show good biocompatibility due to the low toxicity of ADH. These robust and biocompatible neutral polysaccharide nanofibers are expected to seek extensive applications in a variety of biomedical fields.

Here, inspired by the hydrophilic/hydrophobic theory, a novel konjac glucomannan/poly (methyl methacrylate)/chlorogenic acid (KGM/PMMA/CGA) food packaging film was successfully fabricated via microfluidic spinning technology (MST). The results of fourier transform infrared spectroscopy and x-ray diffraction confirmed the formation of hydrogen bonds in the films, which lead to the enhanced mechanical properties. Thermogravimetric analysis and differential scanning calorimetry showed excellent thermal stability of the films. Water vapor permeability (1.47 × 10 −5 ± 0.11 g/(m⋅h⋅kPa)) and water contact angle (89.2°) measurement proved that the films were hydrophobic. The good swelling degree (85.18 ± 15.65%) indicated film’s potentials in releasing CGA. More importantly, KGM played a key role in the antibacterial activities against Staphylococcus aureus (8.5 ± 3.5 mm) and Escherichia coli (6.5 ± 2.1 mm) by utilizing its hydrophilicity. Thus, our present work may provide a new idea for constructing active food packaging films with significant performances based on hydrophilic/hydrophobic strategy.

Konjac glucomannan-gelatin (KGM-GEL) based electrospun nanofibrous films are promising materials for food packaging but suffer from high hydrophilicity and weak mechanical properties. To overcome these limitations, KGM-GEL-based nanofibrous films were reinforced using polycaprolactone (PCL) nanofibers as a crosslinking layer via two-step electrospinning technique. The results show that the bilayer nanofibrous films composed of PCL nanofibers and KGM-GEL nanofibers display a homogenous morphology. Additionally, FTIR and X-ray diffraction analysis shows that physicochemical properties of each biopolymer are preserved in bilayer nanofibrous films. The hydrophobicity of KGM-GEL-based nanofibers is improved dramatically with PCL nanofibers as indicated by water contact angle (WCA) and WVP measurement. All bilayer nanofibrous films have achieved improvements in mechanical properties and thermal stability. It can be concluded that crosslinking with biocompatible synthetic polymers PCL nanofibrous films is an effective pathway for the preparation of natural biopolymers-based nanofibrous films with improved properties, which may be a promising material for food packaging.

ABSTRACT Naringin (NR) is a prominent citrus bioflavonoid, but the application is greatly limited by its low bioavailability. To solve this problem, NR was encapsulated in carboxymethyl konjac glucomannan (CKGM) and chitosan (CS) to form core-shell nanoparticles (CS/CKGM-NR NPs). The physicochemical properties of CS/CKGM-NR NPs with ideal encapsulation efficiency of 82.70 ± 0.30% were characterized by fourier transform infrared spectroscopy and differential scanning calorimetry. The morphology and core-shell structure were investigated by scanning electron microscopy and confocal laser scanning microscopy . Moreover, the releasing behavior of NR was evaluated in In Vitro digestion mode, suggesting that core-shell CS/CKGM-NR NPs were successfully prepared (diameter 700 nm, +35 mV). The underlying mechanism indicated that carboxyl groups of CKGM were cross-linked with ammonium ion of CS by electrostatic interaction. Moreover, the releasing efficiency of NR in simulated gastric fluid (SGF, 20.62 ± 0.59%), simulated intestinal fluid (SIF, 17.08 ± 0.01%) and simulated colon fluid (SCF, 18.82 ± 0.61%) were greatly lowered compared with that of naked NR (52.70 ± 0.01% in SGF, 46.42 ± 3.30% in SIF, 69.35 ± 12.63% in SCF). The findings implied that CS/CKGM-NR NPs were created in core-shell form with controlled-releasing effect. This study provided new insight into the fabrication of CS/CKGM-NR NPs that could be developed as a potential nano-delivery with controlled-releasing effect.
