Enhanced catalytic degradation of tetracycline antibiotic by persulfate activated with modified sludge bio-hydrochar

Jia Wei,Yitao Liu,Yuhan Zhu,Jun Li

In this research, a one-pot prepared modified sludge bio-hydrochar (IBHC) was prepared to activate persulfate (PS) for the degradation of tetracycline (TC) antibiotic. The obtained IBHC bearing defect structure, dispersed iron and large amounts of surface organic functional groups, acts as an outstanding modified biomass carbonaceous material for catalyzing PS to improve the removal efficiency of TC as high as 99.72%. The IBHC + PS system can remove TC effectively with a relative low IBHC dosage (0.2 g·L −1 ), limited PS consumption (5 mmol·L −1 ) and wide pH values (2–10). In addition, the degradation of TC still keep in 94.70% after 5 rounds reuse proving that IBHC possesses excellent stability and practicability. During the activation, both and • OH were generated and the contribution of each component on the TC degradation in IBHC + PS system was explored. Furthermore, the degradation pathways of TC were proposed based on the results of LC-MS.

Development of a highly sensitive immunoassay for detecting aminopyrine abuse in herbal tea

Zian Wang,Zhiwei Liu,Tian Guan,Xi Zeng,Runlin Shen,Zhaodong Li,Yi Lei,Zhenlin Xu,Zhili Xiao,Hongtao Lei,Dingqiang Huang

With the popularity of herbal tea in China, many food fraudsters have added illegal drugs to herbal tea to enhance its functions, among which aminopyrine is widely abused as an antipyretic and analgesic. Presently, there is no immunoassays for aminopyrine, and it is difficult to achieve real-time detection in the field. Based on a polyclonal antibody of aminopyrine with high specificity and sensitivity, an optimal combination of coating antigen/antibody was obtained by screening different coating antigens. On this basis, a sensitive ic-ELISA method was established to detect aminopyrine in herbal tea. The detection limit of the ic-ELISA was 0.18 ng mL−1, which was much lower than the 100 ng mL−1 required as a standard. The method had good consistency with LC-MS in the detection of actual samples and could be used as a reliable method for the detection of aminopyrine in herbal tea.

A salting-out assisted liquid-liquid extraction method for 25 emerging pesticides in follicular fluid

Xin Wei,Liang Cao,Mingrui Zhou,Jiana Huang,Xiaoyan Liang,Nijie Li,Yanan Pan,Renli Yin,Huashou Li,Adela Jing Li

Emerging pesticides of neonicotinoids (NEOs) and “Universal Pesticides” (UPs) are a growing global concern due to their growing commercial importance and potential risks to human health. The currently available analytical methods for these pesticides in biomonitoring were usually tailored for limited number of analytes, or were time consuming and costly. In this study, an efficient and sensitive method for the analysis of 16 NEOs and nine UPs in human follicular fluid (FF) was developed by using a salting-out assisted liquid-liquid extraction (SALLE) method and liquid chromatography-tandem mass spectrometry (LC-MS/MS). Method performance was evaluated by calibration linearity ( r  > 0.99), sensitivity at limits of quantification (0.01–0.50 ng/mL), accuracy at relative recoveries (81–117%) and precision at relative standard deviations (≤16%). The developed method was further validated by analyzing 21 human FF samples that were collected from a hospital in Guangzhou, China. Among the 25 study analytes, two NEOs and six UPs had their detection rates over 85% and medians at 0.048–0.808 ng/mL in the FF samples. Considering the well-known toxicity of these pesticides and their metabolites, it is urgent to figure out exposure profiles of study pesticides and potential reproductive risk for women. To the best of our knowledge, this study is the first to develop and apply the SALLE method in the extraction of 16 NEOs and nine UPs simultaneously in human FF.

Expanding Metabolome Coverage in LC–MS/MS Analysis through Hydralazine-Based Multifunctional Derivatization

Zhiye Yan,Weiwei Tang,Bin Li

Metabolomics is a potent tool used for discovering disease biomarkers and investigating intricate mechanisms of diseases by analyzing metabolite changes in biological systems. However, challenges in metabolite detection have not been fully addressed due to their vast physicochemical diversity and broad concentration range, impacting comprehensive profiling of the metabolome, particularly for certain metabolites with limited detection sensitivity. Therefore, a novel chemical derivatization method based on hydralazine (HZN) was developed for highly sensitive detection of various poorly ionized small metabolites in biological samples using liquid chromatography–tandem mass spectrometry (LC–MS/MS). HZN-based multifunctional derivatization mainly targeted metabolites with carbonyl, carboxyl, and phosphoryl groups, such as free fatty acids, keto-acids, hydroxy acids, bile acids, fatty aldehydes, and phosphoryl compounds. Main parameters affecting the derivatization efficiency were investigated, such as the condensation reagent, and optimal conditions were obtained. The labeling of HZN enabled the generation of characteristic fragment ions through collision-induced dissociation, avoiding the laboriousness of deducing MS/MS fragmentation and facilitating large-scale quantitative analysis. The sensitivities of HZN-derivatized metabolites improved significantly, ranging from 10 to 5000-fold, and the limits of quantitation for derivatized metabolites varied from 5 pM to 100 nM, depending on their structures. Furthermore, alterations of various gut microbiota-derived metabolites in mice fed with a high-fat diet were unraveled using the HZN-based derivatization method, indicating its promising applications in the investigation of disease-related changes in the metabolome.

Rapid determination of trace EDTA in wines and beers by LC-MS/MS

Xiaoqun Wei,Linzhou Zhuang,Chaomin Wu,Wenrui Chen,Zhiyong Li,Baojun Xu

A novel method for quantitative analysis of ethylenediaminotetracetic acid (EDTA) in wines and beers by liquid chromatography coupled to tandem mass spectrometry has been developed. Positive-ion mode was applied as the scan mode, amine species were firstly used as the derivatization reagents of EDTA, and phenylboronic acid was used to react with maltose to eliminate its matrix interference. Primary amines could efficiently eliminate the interference from metal ions (Fe(III) and Cu(II)). The responsive intensity of EDTA with amine derivatization could be improved over 300 times than that without adding amine species. The recovery rate was significantly increased to 104.1% after adding phenylboronic acid at 20.0 mg/L as compared to the control spike-and-recovery experiment results (1.3–1.7%) of beer samples without adding phenylboronic acid. The validated method showed excellent detection sensitivity (0.001 mg/L for LOD (limit of detection) and 0.004 mg/L for LOQ (limit of quantification)) with satisfactory recovery rates (76.0%–97.2%), and good precision (1.40–5.40% RSD values). The optimized method has been satisfactorily applied to the analysis of actual wine and beer samples. It can be proposed as a powerful analytical method of EDTA in wines and beers, and can even be adapted to detect other chelates and polydentate compounds.

Rapid quantitative profiling of kynurenine metabolites in human plasma using LC-MS/MS

Zhu Lian,Liu Jin,Li Kailin,Wang Yang,Liu Fengjiao,Xie Liu,Zhang Yan,Cai Wei

Tryptophan is an essential amino acid for the human body, and its metabolic processes are closely related to a variety of physiological and pathological processes, including neurodegenerative diseases, immune modulation, and cardiovascular diseases. Simultaneous detection of tryptophan and its nine metabolites is challenging and previous methods may have shortcomings of poor sensitivity, selectivity, and specificity in complex matrices. The technology of LC-MS/MS, with its characteristics of high sensitivity and selective, provides a powerful tool for the detection of biomarkers. A rapid and effective method for the simultaneous determination of kynurenine metabolites (Xanthurenic acid (XA), DL-Tryptophan (TRP), Kynurenine (KYN), 3-Hydroxyanthranilic acid (3-HAA), Quinolinic acid (QUIA), Kynurenic acid (KYNA), 3-Hydroxy-DL-kynurenine (3HK), 2-Picolinic acid (2PA), 5-Hydroxytryptamine (5-HT), and Anthranilic acid (AA)) was established in human plasma using ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) technology. Methanol precipitation was used for sample pretreatment, with [2H5]-L-Tryptophan (TRP-D5) as the internal standard. Mass spectrometric detection was performed in positive electrospray ionization mode using multiple reaction monitoring on a triple-quadrupole mass spectrometer. The method’s selectivity, linearity, quantification limit, detection limit, precision, recovery rate, matrix effect, and stability are evaluated. The results show that the linear ranges of 2PA, QUIA, 3HK, 5HT, KYN, 3HAA, TRP, XA, KYNA and AA are 0.25-50, 0.9–225, 1.2–120, 1.4–700, 2-2000, 0.15–150, 180-180000, 0.1–100, 0.1–100, and 0.2–100 ng/mL, respectively; all analytes have good linear relationships with a determination coefficient R2 greater than 0.99. The precision relative standard deviation (RSD) is less than 15%; the spiked recovery rate is 87.54%-107.43%, with an RSD of 2.76%-12.19%. Additionally, the matrix effects for 10 analytes are summarized. At all three QC levels, the matrix effect in plasma is 88.44% to 107.85%, and the RSD is 2.97%-14.62%. These results indicate that there is no significant matrix effect. Stability was evaluated by comparing the measured concentrations after storage with the initial concentrations and expressed as percentages of the initial values. All analytes remained within 85–115% of the initial concentrations under the tested conditions. This study describes the development and validation of a rapid, sensitive, and robust UHPLC–MS/MS method for the simultaneous quantification of ten kynurenine pathway metabolites—including the precursor tryptophan, key intermediates, and neurotoxic/neuroprotective end products-in human plasma. The method employs a simple protein precipitation procedure, a single deuterated internal standard (TRP-D5), and a 13-minute chromatographic run on a C8 column, providing adequate separation of metabolites with diverse physicochemical properties.

HeuSMA: A Multigradient LC-MS Strategy for Improving Peak Identification in Untargeted Metabolomics

Yao-Yu Chen,Na An,Yan-Zhen Wang,Peng-Cheng Mei,Jun-Di Hao,Song-Mei Liu,Quan-Fei Zhu,Yu-Qi Feng

Metabolomics, which involves the comprehensive analysis of small molecules within biological systems, plays a crucial role in elucidating the biochemical underpinnings of physiological processes and disease conditions. However, current coverage of the metabolome remains limited. In this study, we present a heuristic strategy for untargeted metabolomics analysis (HeuSMA) based on multiple chromatographic gradients to enhance the metabolome coverage in untargeted metabolomics. This strategy involves performing LC-MS analysis under multiple gradient conditions on a given sample (e.g., a pooled sample or a quality control sample) to obtain a comprehensive metabolomics data set, followed by constructing a heuristic peak list using a retention index system. Guided by this list, heuristic peak picking in quantitative metabolomics data is achieved. The benchmarking and validation results demonstrate that HeuSMA outperforms existing tools (such as MS-DIAL and MZmine) in terms of metabolite coverage and peak identification accuracy. Additionally, HeuSMA improves the accessibility of MS/MS data, thereby facilitating the metabolite annotation. The effectiveness of the HeuSMA strategy was further demonstrated through its application in serum metabolomics analysis of human hepatocellular carcinoma (HCC). To facilitate the adoption of the HeuSMA strategy, we also developed two user-friendly graphical interface software solutions (HPLG and HP), which automate the analysis process, enabling researchers to efficiently manage data and derive meaningful conclusions (https://github.com/Lacterd/HeuSMA).

Temperature-associated metabolic responses of Aspergillus cristatus revealed by untargeted LC-MS metabolomics

Lu Lingqing,Zhang Rongrong,Jie Wenli,Zhou Hang,Yang Yi,He Yunxue,Ren Mingzheng,Zhou Lihong

Temperature is a key environmental factor affecting fungal growth, reproduction, and metabolism. In this study, the industrially important fungus Aspergillus cristatus was investigated by integrating physiological phenotyping with liquid chromatography-mass spectrometry (LC-MS)-based untargeted metabolomics to systematically characterize its morphological traits, antioxidant activity, and untargeted metabolic profiles at 20 °C, 25 °C, 30 °C, and 35 °C. The results showed that approximately 30 °C was the most favorable temperature for the growth of Aspergillus cristatus . Metabolomic analysis indicated that Aspergillus cristatus exhibited a pattern of metabolic redirection under different temperature conditions, reflecting a multilayered and coordinately changing metabolic mode associated with temperature adaptation. At low temperatures (20 °C and 25 °C), unsaturated lysophospholipids, such as PC O-18:2, accumulated substantially, suggesting their involvement in membrane adaptation, whereas high temperature was associated with changes in specific phospholipid molecules. In terms of oxidative stress defense, flavonoid- and phenolic-related metabolites showed more prominent changes under low and optimal temperatures, whereas glutathione-related metabolites increased significantly at high temperature, indicating a greater reliance on glutathione-associated antioxidant defense. Regarding energy and osmotic balance, reserve carbohydrates accumulated at the optimal temperature, consistent with coordination between growth and stress resistance, whereas high-temperature stress was associated with a shift toward sugar alcohol accumulation and a metabolic pattern consistent with metabolic deceleration. This study presents the metabolic changes of Aspergillus cristatus under different temperature conditions from a systems-level metabolic perspective and suggests that Aspergillus cristatus may adapt to environmental change by coordinating growth-related metabolism with stress defense-related metabolism. In addition, these findings may provide a reference for future studies on fermentation regulation and stress resistance in Aspergillus cristatus .

Xiangwu Liu,Bangyan Song,Xiaoxia Jiang,Ya Yang,Ping Lu,Deyu Hu

A quick, easy, cheap, effective, rugged and safe liquid chromatography-tandem mass spectrometry technology was used to determine benziothiazolinone in citrus. Orthogonal test and single-factor analysis were conducted to identify the optimal extraction method of benziothiazolinone. The good linearity (R2 > 0.9949), high accuracy and precision, matrix effect and satisfactory recoveries (79.31%–98.68%) at three spiked levels (0.02, 0.2 and 2 mg/kg) with low relative standard deviation values (0.63%–8.68%) were validated. The limits of detection and quantification were within 0.2–0.8 and 20 μg/kg, respectively. This study could provide guidance for the reasonable use of this fungicide and for the establishment of maximum residue level.

LC-MS-based metabolomics reveals metabolite dynamic changes during irradiation of goat meat

Wei Jia,Zibian Fan,Qingyun Shi,Rong Zhang,Xin Wang,Lin Shi

The current study applied an untargeted metabolomics approach by ultra high performance liquid chromatography quadrupole-orbitaltrap high resolution mass spectrometry (UHPLC-Q-Oritrap-MS) to identify the chemical composition of irradiated goat meat and investigate the effect of irradiation on its metabolic profile and meat quality. A total of 103 metabolites were identified as differential metabolites responsible for metabolic changes in irradiated goat meat, which were involved in phenylalanine , tyrosine and tryptophan biosynthesis, phenylalanine metabolism and purine metabolism. Differential metabolites comprising amino acids , nucleotides and their derivatives were determined as the discriminating factors responsible for the meat quality during irradiation. Specifically, the levels of L-phenylalanine, L-isoleucine, L-histidine, guanosine, guanine, creatinine, glutathione and nicotinic acid were increased while inosine 5′-monophosphate (IMP) and guanosine 5′-monophosphate (GMP) were decreased. Overall, except for L-phenylalanine and guanine, other related metabolites significantly decreased with storage. This study contributes to a comprehensive understanding of the effect of irradiation doses and storage time on goat meat metabolism at the molecular level, so as to assess the quality of irradiated goat meat. Satisfactory results with linearity (R 2  > 0.995), precision (RSD less than 8.9%) and recovery (83%-106%) were obtained, demonstrating that the untargeted mebabolomics approach was appropriate for monitoring the changes of small molecular metabolites in irradiated goat meat and irradiation is a feasible method for goat meat preservation .

Anti-Inflammatory Activity of Panax notoginseng Flower Saponins Quantified Using LC/MS/MS

Junchen Liu,Yuehang Wu,Wenrui Ma,Hongyan Zhang,Xianyao Meng,Huirong Zhang,Miaomiao Guo,Xiao Ling,Li Li

Panax notoginseng(Burk) F. H. Chen is a traditional Chinese medicinal and edible plant. However,Panax notoginsengflower (PNF) is rarely used. Therefore, the purpose of this study was to explore the main saponins and the anti-inflammatory bioactivity of PNF saponins (PNFS). We explored the regulation of cyclooxygenase 2 (COX–2), a key mediator of inflammatory pathways, in human keratinocyte cells treated with PNFS. A cell model of UVB-irradiation-induced inflammation was established to determine the influence of PNFS on inflammatory factors and their relationship with LL–37 expression. An enzyme-linked immunosorbent assay and Western blotting analysis were used to detect the production of inflammatory factors and LL37. Finally, liquid chromatography–tandem mass spectrometry was employed to quantify the main active components (ginsenosides Rb1, Rb2, Rb3, Rc, Rd, Re, Rg1, and notoginsenoside R1) in PNF. The results show that PNFS substantially inhibited COX–2 activity and downregulated the production of inflammatory factors, indicating that they can be used to reduce skin inflammation. PNFS also increased the expression of LL-37. The contents of ginsenosides Rb1, Rb2, Rb3, Rc, and Rd in PNF were much higher than those of Rg1, and notoginsenoside R1. This paper provides data in support of the application of PNF in cosmetics. Keywords:Panax notoginsengflower;UVB;inflammatory factor;antibacterial peptide LL-37;total saponin;LC/MS/MS
