Probiotics were found to be effective in ameliorating the microbial dysbiosis and inflammation caused by intestinal pathogens. However, biological challenges encountered during oral delivery have greatly limited their potential health benefits. Here, a model probiotic (Lactobacillus rhamnosus) was encapsulated in an intestinal-targeted hydrogel to alleviate bacterial enteritis in a novel mode. The hydrogel was prepared simply by the self-cross-linking of thiolated hyaluronic acid. Upon exposure to H2S which were excreted by surrounding intestinal pathogens, the hydrogel can locally degrade and rapidly release cargos to compete with source pathogens in turn for binding to the host. The mechanical properties of hydrogel were studied by rheological analysis, and the ideal stability was achieved at a polymer concentration of 4% (w/v). The morphology of the optimal encapsulation system was further measured by a scanning electron microscope, exhibiting uniform payload of probiotics. Endurance experiments indicated that the encapsulation of L. rhamnosus significantly enhanced their viability under gastrointestinal tract insults. Compared with free cells, encapsulated L. rhamnosus exerted better therapeutic effect against Salmonella-induced enteritis with negligible toxicity in vivo. These results demonstrate that this redox-responsive hydrogel may be a promising encapsulation and delivery system for improving the efficacy of orally administered probiotics.

To study the effects of mixed starter consisted of different fermentative type of lactic acid bacteria (LAB) on the fermentation of suansun , two lactic acid bacteria ( Leuconostoc citreum NM-12 and Lactobacillus plantarum L 01 ) isolated from Chinese traditional fermented vegetable were used in the preparation of suansun . The fermentation was carried out at ambient temperature (around 25 °C)for 96 h by inoculating different mixing ratios of LAB (inoculated fermentation) or using natural microbes (natural fermentation). The changes of pH, titratable acid (TA), microbe communities, free sugars, organic acids, nitrite and volatile compounds during fermentation were evaluated. Suansun treated with high Leuconostoc citreum ratio inoculation exhibited a quickly change in pH and TA, resulting from the rapid increase in the number of viable cells, at the early stage of fermentation and produced more mannitol (0.12–0.46 mg/mL) and acetic acid (0.93–3.56 mg/mL). However, Suansun treated with high Lactobacillus plantarum ratio inoculation had lower pH and higher TA at the later stage of fermentation and produced more lactic acid (5.32–7.68 mg/mL). No mannitol was detected in suansun when only Lactobacillus plantarum was inoculated in fermentation. No p-cresol was produced in the inoculated fermentation with mixed starter culture , in addition to the production of ethyl acetate and 2.3-butanedione, which had a positive effect on the flavor of suansun . In summary, this study demonstrated the application value of mixed starter consisted of different fermentative type of LAB. LAB types and mixing ratios greatly affected the types and concentration of metabolites in suansun fermentation.

The current study was conducted to investigate the effects of zinc enrichment on the characteristics, metabolites and antioxidant activity of L. plantarum DNZ-4. Electron microscopy analysis showed that the ultrastructure of the zinc-enriched strain changed greatly both for surface and inside as compared to the control strain. The zinc-enriched strain showed higher acid production ability resulting in lower pH value. Resistance assays demonstrated that the zinc-enriched strain has stronger tolerance ability to acid, bile salt and H 2 O 2 than that of the control strain. Metabolites analysis indicated that the various metabolites in the zinc-enriched strain were altered significantly as a result of amino acids, energy and fatty acid metabolism affected by zinc enrichment. Furthermore, the antioxidant properties of the zinc-enriched strain, including radical scavenging capacity and the antioxidative enzyme activity increased significantly. These results suggest that zinc addition had a positive effect on the physiologic function of L. plantarum DNZ-4.

To improve the storage stability and antioxidant activity of yogurt, an exopolysaccharide (EPS)-producing strain, Lactobacillus fermentum SN4-1, was isolated from traditional Qinghai yak yogurt, and the EPS biosynthetic pathway was analyzed via whole-genome sequencing. A novel EPS (EPS-2) from SN4-1 was purified by anion-exchange chromatography and characterized by HPLC, HPGPC, and FT-IR. SN4-1 was combined with a commercial starter, and its effects on yogurt quality during cold storage were evaluated. Results showed that EPS-2 mainly consisted of mannose, glucose, and galactose (molar ratio 5.93:1.19:1) with an average molecular weight of 40.3 kDa. SN4-1 contained an EPS gene cluster ( eps-2 ) and multiple probiotic-related genes. During cold storage, the EPS content in yogurt with SN4-1 peaked at 144.21 mg kg -1 on day 5. Accordingly, the yogurt exhibited higher hardness, adhesiveness, cohesiveness, and viscosity, with adhesiveness increasing by 198.81%. Its water-holding capacity was 10.35% higher than that of the control on day 15. Moreover, the yogurt demonstrated strong radical scavenging capacities (DPPH 89.49%, ·OH 76.32%, O 2 - 63.23%, ABTS ∙+ 90.14%). These findings highlight the potential of SN4-1 in developing stable, clean-label probiotic yogurt.

Lact. gasseri alleviates dyslipidaemia and pancreatic injury, reducing the incidence of T2DM in a diabetes-prone mouse model. • The probiotic suppresses IL-1β–mediated trained immunity-like priming by reducing H3K4me3 enrichment at the Il-1β promoter. • Lact. gasseri modulates host methionine–one‑carbon metabolism by limiting intestinal methionine availability. • These findings link intestinal nutrient availability to systemic inflammatory programming and metabolic disease. Trained immunity-like epigenetic priming contributes to persistent metabolic inflammation in type 2 diabetes mellitus (T2DM), but the upstream metabolic and microbiota-dependent regulatory mechanisms remain unclear. Here, we performed an exploratory study to examine whether Lactobacillus gasseri ( Lact. gasseri ) attenuates metabolic dysfunction by suppressing methionine-dependent epigenetic priming of innate immune cells, and to characterize how metformin regulates Lact. gasseri colonisation in the intestine. A diabetes-prone mouse model was established using high-fat diet (HFD) and low-dose streptozotocin (STZ); a standard-chow plus STZ group was included to control for the direct β-cell toxicity of STZ. Metabolic phenotypes, histopathology, inflammatory responses, and intestinal barrier function were assessed. Bone marrow-derived macrophages (BMDMs) were rested ex vivo for 4 days in the absence of any in vivo stimulus and then subjected to an unrelated secondary LPS/IFN-γ challenge in order to evaluate primed responsiveness, and methionine–one‑carbon metabolism, H3K4me3, and cytokine expression were analysed by RT-qPCR, western blotting, ELISA and ChIP-qPCR. Lact. gasseri significantly reduced diabetes incidence and improved glucose tolerance and insulin sensitivity, ameliorated dyslipidaemia and pancreatic injury in HFD/STZ-treated mice. Mechanistically, Lact. gasseri was associated with reduced methionine availability and with lower absolute abundance of systemic and bone marrow methyl donors, accompanied by decreased H3K4me3 at the Il-1β promoter and attenuated IL-1β-mediated trained immunity-like priming. Methionine restriction in macrophages recapitulated these effects, whereas methionine replenishment restored histone methylation and IL-1β maturation, supporting a metabolic–epigenetic axis while not excluding contributions from other nutrient-sensitive pathways. Notably, metformin enhanced intestinal colonisation of Lact. gasseri without directly promoting bacterial growth in vitro, an effect accompanied by improved mucus barrier integrity and concordant upregulation of P53 and MUC2. Because no combined Lact. gasseri plus metformin arm was included in vivo, metabolic synergy was not tested in this study. Together, these exploratory findings describe a candidate microbiota-linked mechanism in which Lact. gasseri constrains methionine-dependent epigenetic priming of innate immune cells, connecting intestinal nutrient availability to systemic inflammatory memory and providing a rationale—rather than proof—for probiotic–drug combination strategies in T2DM. Microorganisms in Chinese rice wine (CRW), a complex ecosystem, in which microbial interactions play a key role in flavor development. However, the correlation between core microbiota and volatile flavor substances and physicochemical indicators during the fermentation of CRW remains unclear. In this study, we investigated the dynamic changes in microbial communities, physicochemical indicators, and volatile flavor substances throughout the fermentation of CRW. A total of 31 volatile compounds were identified, among which 10—including 8 esters, 1 alcohol, and 1 aldehyde—were determined to be the main contributors to CRW flavor. Correlation analysis revealed significant associations among the microbial taxa, flavor compounds, and physicochemical indices. Saccharomyces , Lactobacillus , Rhizomucor , Saccharopolyspora , Lactococcus , Thermomyces , and Wickerhamomyces were identified as the core microbial genera involved in CRW fermentation. Moreover, Lactiplantibacillus plantarum 15 ( L. plantarum 15) was selected for inoculation experiments. Among different inoculation levels, 10 7 colony-forming units (CFU)/mL exhibited the most pronounced effect in enhancing CRW flavor. In particular, inoculation with 10 6 CFU/mL significantly improved both physicochemical indicators and volatile compound profiles compared to the sterile control, indicating optimal microbial activity at this concentration. These findings provide valuable insights into microbial contributions to flavor formation and offer guidance for optimizing the CRW fermentation through microbial modulation.

Soy protein is a multimeric globular protein that is less likely to be metabolized and utilized by lactic acid bacteria (LAB) during fermentation compared to milk-derived proteins. Little genetic information is available regarding LAB-mediated hydrolysis of soy protein. In this study, we sequenced the genomes of 46 Lactobacillus helveticus ( L. helveticus ) strains to analyze genetic-level factors affecting soy protein hydrolytic activity. The analysis revealed that the soy protein hydrolysis capability of these L. helveticus strains showed no correlation with their isolation sources, geographic origins, extracellular protease genes, peptide transporter genes, or peptidase gene distribution. Through genome-wide association analysis (GWAS), we identified two significantly associated missense SNPs (SNP-482062 and SNP-1780770) linked to soy protein hydrolysis activity. These two SNPs changed the local tertiary structure of amino acid permease and ornithine decarboxylase, respectively. Enzymatic assays confirmed that the mutations significantly affected enzyme activity, thereby validating their critical role in soy protein hydrolysis. This study elucidates the genetic basis for differential soy protein hydrolysis capacity among L. helveticus strains, offering practical guidance for selecting highly proteolytic strains to develop soy-based fermented dairy alternatives.

Background: The purpose of this study is to observe LP45 ( Lactobacillus plantarum 45) to investigate the mechanism by which LP45 attenuates oxidative stress-induced damage and regulates the osteoblast-osteoclast balance. Materials and Methods: The oxidative stress level and osteoblast- and osteoclast-related proteins were detected by immunofluorescence staining, Western blotting, ROS fluorescent probe and ELISA. Osteoblast cell proliferation capacity was determined by the CCK-8 assay. X-ray observation and HE staining were used to detect the effect of LP45 on osteoporosis. Results: The expression level of SHP2 and Src was significantly increased, and the expression levels of NOX4, P22, P47, IL-1β, NLRP3, IRF3, RANK, β-catenin and INF-β were inhibited in LP45 group and LPS + LP45 group as compared to those in LPS group. Compared with that in LPS group, the concentration of SOD was increased and the concentration of MDA was decreased in LPS + LP45 group. The protein expressions of OPG, RANKL, RUNX3, RANK and β-catenin in LP45 group and LPS + LP45 group increased. The protein expressions of NF-κB, CREB and AP-1 in LP45 group and LPS + LP45 group decreased significantly. The results were also confirmed by immunofluorescence staining and ROS fluorescent probe. X-ray observation and HE staining showed that LP45 could inhibit the progression of osteoporosis. Conclusion: LP45 can exert its antioxidant effect by inhibiting the production of oxidative stress to activate the SHP2 signaling pathway, thus promoting osteoblast differentiation and repressing osteoclast formation to maintain bone homeostasis and improve bone metabolism.

Objective Zhi-zi-chi decoction (ZZCD) is a classical antidepressant traditional Chinese medicine (TCM) formula. However, the microbiological basis of its antidepressant effects remains unclear. The objective of this study was to investigate the microbial basis of ZZCD. Methods The bacteria that exhibit enhanced growth in ZZCD extracts were identified through in vitro culture. A depressed rat model was subsequently established to assess the antidepressant effects of the bacteria. A GC-MS-based metabolomics approach and molecular docking simulation were employed to investigate the potential mechanism of the antidepressant effects of the bacteria from the perspective of changes in the metabolic profile of the gut microbiota. Finally, the antioxidant effects of gut microbial metabolites following Lactobacillus rhamnosus treatment were validated through cellular experimentation. Results ZZCD was able to promote the growth of L. rhamnosus . Furthermore, L. rhamnosus was demonstrated to increase the percentage of sucrose preference, reduce the immobility time in tail suspension and forced swimming tests, and alleviate depressive symptoms in rats. Additionally, L. rhamnosus was shown to alter the metabolic profile of gut microbiota. A total of six metabolites (ethanamin, butyric acid, 2-methylbutanoic acid, behenic acid, 3-methylvaleric acid and 1-hexadecanol) associated with depressive behavior in rats were identified. All six metabolites demonstrated the capacity to bind to Keap1 in molecular docking simulations. Gut microbial metabolites after L. rhamnosus treatment increased Nrf2 levels in HT22 cells. Conclusion L. rhamnosus may be one of the microbial bases for the antidepressant effects of ZZCD. Its effects may be related to the regulation of gut microbiota metabolism and anti-oxidative stress.

To improve the postharvest preservation of cherry tomatoes and combat pathogenic, both bacterial and fungal contamination (particularlyAlternaria alternata), a novel biodegradable coating was developed based on a water-in-water (W/W) Pickering emulsion system. The emulsion was stabilized byL. plantarum(Lactobacillus plantarum), with maltodextrin (MD) as the dispersed phase and hydroxypropyl methylcellulose (HPMC) as the continuous phase. Characterization of emulsions at varying concentrations revealed that the optimized W/W-PL^8 film exhibited superior stability, smooth morphology, and low water vapor permeability (WVP = 220.437 g/(m2·24 h)), making it a promising candidate for fruit and vegetable preservation. Furthermore, the coating demonstrated strong antioxidant activity, with scavenging rates of 58.99% (ABTS) and 94.23% (DPPH), along with potent antimicrobial effects, showing inhibition rates of 12.8% againstEscherichia coliand 23.7% againstStaphylococcus aureus. Applied to cherry tomatoes, the W/W-PL^8 coating significantly reduced respiration rates, minimized decay incidence, and maintained nutritional quality during storage. Remarkably, the coating successfully controlledAlternaria alternatacontamination, enhancing the storage duration of cherry tomatoes. These findings highlight the potential of W/W-PL^8 as an eco-friendly and functional packaging material for fresh produce preservation.

In this study, litchi polysaccharides were obtained from unfermented or fermented pulp by Lactobacillus fermentum (denoted as LP and LPF, respectively). The differences between LP and LPF in the colonic fermentation characteristics and modulatory of gut microbiota growth and metabolism were investigated with an in vitro fecal fermentation model. Results revealed that the strategies of gut bacteria metabolizing LP and LPF were different and LPF with lower molecular weight (Mw) was readily utilized by bacteria. The monosaccharide utilization sequence of each polysaccharide was Ara > Gla > GalA > GlcA ≈ Glu ≈ Man. Moreover, LPF promoted stronger proliferation of Bifidobacterium , Megamonas , Prevotella , and Bacteroides and higher SCFAs production (especially acetic and butyric acids) than LP. Correlation analysis further revealed that Mw could represent an essential structural feature of polysaccharides associated with its microbiota-regulating effect. Overall, Lactobacillus fermentation pre-treatment of litchi pulp promoted the fermentation characteristics and prebiotic activities of its polysaccharide.

Deoxynivalenol (DON) is a common environmental pollutant that poses a serious health risk to humans worldwide. This study was aim to explore whether gut microbiota is involved in DON-induced intestinal toxicity as well as to reveal effect of probiotics derived from gut microbiota in protecting intestinal barrier and to elucidate mechanism. We found that DON caused disturbed gut microbiota, particularly Lactobacillus murinus ( L. murinus ) deficiency. DON enhanced M1 macrophage polarization and decreased tight junction protein expression. Microbiota transplantation experiments showed that transfer of DON-disrupted microbiota to healthy mice resulted in delivery of DON-induced intestinal toxicity. Besides, DON lost its damaging effect on macrophage and intestinal barrier in antibiotic-treated mice. Further intervention experiments revealed that L. murinus induce macrophage conversion from M1 to M2 phenotype through secreted extracellular vesicles (EVs) to alleviate DON-induced intestinal barrier disruption. Mechanistically, EVs activate TLR2 to promote M2 macrophage polarization and release IL-10, which in turn enhances intestinal barrier function. Upon successful translation of its efficacy into clinical practice, EVs created from L. murinus could be a novel possible treatment strategy for DON-induced gut disease.
