Acid-Triggered H2O2 Self-Supplying Nanoplatform for 19F-MRI with Enhanced Chemo–Chemodynamic Therapy

Ruijuan Zhang,Qian Ma,Gaofei Hu,Leyu Wang

The real-time tracking and efficacy evaluation of therapeutic nanoplatforms especially in deep-tissues is of great importance but faces challenges. Meanwhile, chemodynamic therapy (CDT), relying on Fenton reaction by converting H2O2 into toxic hydroxyl radicals (•OH), has drawn wide interests in the fabrication of nanozymes for tumor therapy, while endogenous H2O2 is usually insufficient for effective CDT. Here, we report the pH-responsive multifunctional nanoplatforms consisting of copper peroxide (CP) nanoparticles, paclitaxel (PTX) and perfluoro-15-crown-5-ether (PFCE), for 19F magnetic resonance imaging guided and enhanced chemo–chemodynamic synergetic therapy with self-supplied H2O2 stemmed from the decomposition of CP nanoparticles under acid conditions in tumor. The decomposition of CP nanoparticles further promotes the release of PTX for enhanced chemotherapy. Both in vitro and in vivo results indicate that the efficient generation of •OH and drug release effectively inhibits tumor growth. Furthermore, 19F MRI signal can clearly track the fate of nanoplatforms in tumor and guide tumor treatment. This work provides a promising strategy for the rational design and construction of multifunctional nanoplatforms for imaging-guided synergistic therapy of deep seated tumor.

Synthesis and characterization of superparamagnetic iron oxide nanoparticles as calcium-responsive MRI contrast agents

Pengfei Xu,Zhiwei Shen,Baolin Zhang,Jun Wang,Renhua Wu

Superparamagnetic iron oxide nanoparticles (SPIONs) as T2 contrast agents have great potential to sense calcium ion (Ca 2+ ) using magnetic resonance imaging (MRI). Here we prepared calcium-responsive SPIONs for MRI, formed by combining poly(ethylene glycol) (PEG) and polyethylenimine (PEI) coated iron oxide nanoparticle (PEI/PEG-SPIONs) contrast agents with the straightforward calcium-sensing compound EGTA (ethylene glycol tetraacetic acid). EGTA was conjugated onto PEI/PEG-SPIONs using EDC/sulfo-NHS method. EGTA-SPIONs were characterized using TEM, XPS, DSL, TGA and SQUIID. DSL results show that the SPIONs aggregate in the presence of Ca 2+ . MRI analyses indicate that the water proton T2 relaxation rates in HEPES suspensions of the EGTA-SPIONs significantly increase with the calcium concentration because the SPIONs aggregate in the presence of Ca 2+ . The T2 values decreased 25% when Ca 2+ concentration decreased from 1.2 to 0.8   mM. The aggregation of EGTA-SPIONs could be reversed by EDTA. EGTA-SPIONs have potential as smart contrast agents for Ca 2+ -sensitive MRI.

Biomineralization-Derived Gadolinium Nanoparticles for Dual-Modal 1H/19F MRI-Guided Photodynamic Tumor Therapy

Si Tingzhao,Li Tuohuan,Xiao Wenqing,Wang Kai,Pan Yue,Zhu Yuyu,Cao Zhong

The integration of conventional 1 H MRI with heteronuclear 19 F MRI enables the concurrent acquisition of complementary anatomical and molecular imaging information using a single instrument. Here, we report a T 1 -T 2 dual-modal MRI platform based on gadolinium-albumin (Gd-BSA) nanostructures synthesized through a biomimetic mineralization strategy. To endow multifunctionality, perfluoropentaphenyl-15-crown-5 ether (PFCE) and the photosensitizer IR780 were coencapsulated, yielding the theranostic probe IR780@PFCE@Gd-BSA (IPGB). IPGB exhibited strong 19 F MRI signals and T 1 -T 2 contrast enhancement on clinical and preclinical MRI systems (1.5, 3.0, and 9.4 T), confirming its potential for 1 H/ 19 F multimodal imaging. Beyond imaging, IPGB exhibited superior photodynamic therapy (PDT) efficacy compared with free IR780. In vivo , IPGB achieved passive tumor accumulation with prolonged retention, enabled high-resolution multimodal MRI, and significantly suppressed breast tumor growth after PDT. Collectively, these results highlight IPGB as a viable multifunctional nanoplatform for cancer diagnosis, real-time multimodal imaging, and image-guided therapy.

In vivo membrane engineering traps Gd-based MRI contrast agents for detecting microhepatocellular carcinoma

Chunping Mao,Jie Yi,Fuan Deng,Wanning Zhu,Jiaru Zhao,Hualing Li,Luoxing Xia,Jianing Ji,Yuxiang Xiong,Jiayao Cai,Lingdong Jiang,Jun Shen,Guobin Hong,Lu Zhang

Accurate detection of microhepatocellular carcinoma (HCC) remains a major clinical challenge owing to the limited specificity and sensitivity of current imaging modalities. Herein, we present a dual-injection magnetic resonance imaging (MRI) peptidic probe based on in vivo membrane engineering, achieving in situ signal amplification and molecularly precise imaging. The first injection of programmable nanoparticles coassembled from two peptide monomers, incorporating a GPC3-targeting ligand, a β sheet–forming motif, a dibenzocyclooctyne (DBCO) handle, and porphyrin IX (PpIX) for fluorescence tracking. Following systemic administration, the nanoparticles high-specifically bind to GPC3-overexpressing tumor membranes and transform into surface-anchored nanofibrils, exposing confined DBCO groups. A second injection of azide-modified Gd-DOTA enables rapid copper-free click conjugation on the nanofibrillar scaffold, yielding a nearly fourfold increase in longitudinal relaxivity. MRI demonstrated strong T 1 -weighted signal enhancement and high tumor-to-liver contrast in Hepa1-6 tumor-bearing mice. In vivo membrane engineering strategy establishes a generalizable platform for receptor-guided molecular imaging and early cancer detection.

Programmable macrophage-polarizing nanoparticles for MRI-guided early detection and treatment of pulmonary metastases

Sui Meiju,Xie Kairu,Luo Yingying,Wang Haiqiang,Sheng Hao,Yue Sen,Liu Xiaoxun,Li Sha,Suk Jung Soo,Chen Shizhen,Chen Daiqin,Zhou Xin

Metastatic lung cancer remains a leading cause of cancer-related mortality worldwide, underscoring the urgent need for early detection and intervention strategies. Current diagnostic and therapeutic methods often fall short in sensitivity and efficacy, particularly for early-stage disease. In this study, we develop CaCO 3 @quercetin-Mn nanoparticles (CQM NPs) integrating dual-stage macrophage reprogramming with T 1 -weighted magnetic resonance imaging (MRI) enhancement for theranostic management of pulmonary metastasis. CQM NPs exploit dynamic macrophage polarization: initial M2 polarization promotes nanoparticles accumulation in metastatic lesions, enabling detection of lesions as small as 0.11 mm, while subsequent acid/glutathione (GSH)-triggered degradation reprograms tumor-associated macrophages to antitumor M1 phenotype, suppressing metastatic growth by 81.64% and preventing circulating tumor cells (CTCs) from colonizing the lungs by 78.08%. These findings demonstrate a significant improvement in MRI sensitivity and an integrated approach to therapy, contrasting with traditional methods that separate these challenges. This “detect-to-treat” paradigm bridges the critical gap between imaging sensitivity and immunomodulatory therapy, offering a blueprint for precision nanomedicine in metastatic cancers. Manganese-Doped Zeolite Imidazolium-Based Nanoplatform for MRI-Guided Sonodynamic and Chemodynamic Cancer Therapy

Qi Xu,Qiqi Qi,Zhimin Mo,Han Hu,Yanru Shao,Qianyuan He,Zushun Xu

Multimodal sonodynamic therapy (SDT) is one of the main approaches for future tumor therapy; however, the application of SDT is still hindered by hypoxia and glutathione (GSH) overexpression in the tumor microenvironment (TME). Based on this, we designed manganese-doped zeolitic imidazolate framework-67 (ZIF-67) nanocrystals and encapsulated them with cell membranes to obtain Mn-ZIF-67@CMs (MF@CMs). The ZIF-67 nanocrystals have natural M-N active sites and show potential for application in SDT. Since the TME contains a large amount of H2O2, which reacts with Mn2+ to release O2. Meanwhile, Co3+ and Mn3+ can consume GSH in the TME, which in turn boosts the effectiveness of SDT. Additionally, the reaction that produces Co2+ and Mn2+ exhibits a Fenton-like mechanism, which further contributes to the therapeutic effects of chemodynamic therapy (CDT). Furthermore, due to the magnetic resonance imaging (MRI) T1 imaging properties of Mn2+, these nanoparticles (MF@CMs) also hold potential for integrated diagnosis and treatment. The MF@CMs designed in this study have good biocompatibility, oxygen-producing capacity, and GSH-consuming capacity, indicating the potential for clinical application.

Metal-Ion-Activity Magnetic Nanoprobes for MRI Sensing of Cu2+ Levels in Tumor Malignancy

Yuehao Gan,Qiyue Wang,Hui Du,Yuqi Wang,Dao Shi,Longfei Pan,Daishun Ling,Fangyuan Li

Metal ions are essential for enzymatic regulation, redox balance, and signal transduction, with dysregulation increasingly implicated in cancer, neurodegeneration, and inflammation. Copper ions (Cu 2+ ), in particular, are closely associated with tumor progression and malignancy. However, conventional binding-based magnetic resonance imaging (MRI) probes lack the sensitivity to detect trace metal ion fluctuations in vivo. Here, a metal-ion-activity magnetic (MIAM) nanoprobe is reported that integrates activity-based chemical sensing with magnetic resonance energy transfer for signal amplification. MIAM nanoprobe consists of superparamagnetic iron oxide nanoparticles (SPIONs) linked to gadoteric acid (Gd-DOTA) via a Cu 2+ -cleavable adipic acid dihydrazide moiety. The T 1 -MRI signal is initially quenched as SPIONs suppress Gd-DOTA's relaxivity. Upon exposure to elevated Cu 2+ , Cu 2+ -catalyzed reaction breaks the covalent bond in the MIAM nanoprobe and then releases free Gd-DOTA, resulting in an activated T 1 -MRI signal. The catalytic nature of this reaction enables multiple turnover events, thereby facilitating signal amplification and enabling sensitive detection of Cu 2 ⁺ levels down to ≈10 µ m in vitro, facilitating the identification of tumor malignancy and the evaluation of therapeutic response to Cu 2+ chelators. By converting metal ion activity into quantifiable MRI contrast, the MIAM nanoprobe offers a noninvasive platform for diagnosing and tracking various metal-ion-associated diseases.

Dual-Functional Nanoliposome with High BPA Loading for Targeted MRI-Guided BNCT of Glioblastoma

Yaxin Qin,Xiaoyan Sun,Zhicheng Zhang,Yufan Yang,Qi Dai,Xin Tan,Ruolin Jiang,Xiaoyan Bao,Linjie Wu,Changran Geng,Yuanhao Liu,Xiaoping Sun,Xingyan Liu,Minoru Suzuki,Qinchun Wei,Min Han

Glioblastoma is a highly malignant brain tumor with few available therapeutic options, for which boron neutron capture therapy (BNCT) has emerged as a promising precision radiotherapy approach. However, its efficacy remains suboptimal due to inadequate tumor targeting of boron agents and lack of in vivo visualization. Herein, a gadolinium–boron integrated lipid nanocarrier (BPA-F&DOTA-Gd@LIPO-ANG) was developed for targeted boron delivery and MRI-guided BNCT. BPA-F and DOTA-Gd were coloaded into lipid nanocarriers using a microfluidic system, with Angiopep-2 modification to enhance blood–brain barrier penetration and glioma targeting. Following cellular uptake, BPA-F&DOTA-Gd@LIPO-ANG effectively increased boron accumulation in glioma cells, inducing significant apoptosis and DNA damage upon neutron irradiation. Intravenously injected into orthotopic glioma model mice, BPA-F&DOTA-Gd@LIPO-ANG selectively accumulated in the tumor site, increasing tumor-to-normal tissue (T/N) and tumor-to-blood (T/B) ratios. Enhanced boron accumulation facilitated greater cytotoxic effects mediated by high-energy rays from the boron capture reaction, leading to significantly prolonged mice survival. In addition, in vitro and in vivo MRI validations confirmed its MRI visualization capability, meeting the demand for boron drug imaging for BNCT diagnosis and treatment integration. Overall, these results suggest that BPA-F&DOTA-Gd@LIPO-ANG is a promising targeted boron delivery system for MRI-guided BNCT in glioblastoma treatment.

Biomimetic Catalase-Templated Nanoprobes for MRI-Guided Oxygen-Supplemented Photodynamic Therapy in Breast Cancer

Wen Gu,Dinghua Liu,Yanyan Wu,Weitao Yang,Yanjing Yun,Bingbo Zhang,Shiyuan Liu

Photodynamic therapy (PDT) faces critical challenges in practical application due to tumor hypoxia and the lack of precise imaging guidance. To address these limitations, we engineered Gd@CAT Ce6 (GCC) via catalase (CAT)-mediated biomimetic synthesis, where CAT serves as a structural template for the green synthesis of gadolinium-based nanoparticles, an enzymatic oxygenator through H 2 O 2 decomposition, and a hydrophobic host for photosensitizer chlorin e6 (Ce6) loading. GCC leverages CAT's enzymatic activity to decompose tumor-overexpressed H 2 O 2 into oxygen, effectively mitigating hypoxia while amplifying Ce6-mediated reactive oxygen species (ROS) generation under laser irradiation. In vitro studies confirmed a uniform nanostructure (approximately 10 nm), high longitudinal relaxivity (r 1 = 10.9 m m −1 s −1 ), and potent ROS production. In vivo magnetic resonance imaging (MRI) demonstrated significant tumor accumulation via the enhanced permeability and retention (EPR) effect, extending the imaging window to 1–2 h for precise therapy guidance. Notably, GCC combined with laser irradiation suppressed 4T1 tumor growth by 87.84% in mice, outperforming controls, while exhibiting good biocompatibility in blood and organ toxicity assays. This work presents an enzyme-based theranostic strategy that synergizes real-time imaging with self-oxygenating PDT, offering a promising solution to overcome hypoxia-driven therapeutic resistance.

Zeolitic imidazolate framework nanoparticles loaded with gadolinium chelate as efficient T1 MRI contrast agent

Zhou Weixiu,Shen Jiyun,Lin Jiaomin,An Miao,An Lu,Tian Qiwei,Yang Shiping

Gd–chelate magnetic resonance imaging (MRI) contrast agents used in conventional clinical medicine still have some shortcomings, such as their low relaxation rates and poor imaging sensitivity. In this work, a Gd–chelate (Gd–DTPA), with a comparison made to the commercial contrast agent Magnevist was loaded into a zeolitic imidazolate framework (ZIF-8) to construct the efficient T 1 MRI contrast agent Gd–DTPA@ZIF-8. At 0.5 T, the longitudinal relaxivity value ( r 1 ) of Gd–DTPA@ZIF-8 reaches 29.60 mM −1  s −1 , which is about 6 times higher than that of Gd–DTPA. In terms of the retention of Gd–DTPA in ZIF-8, no obvious Gd 3+ ions are released in water and complete culture solution over a long time period. Haemolysis and cytotoxicity experiments revealed that Gd–DTPA@ZIF-8 has good biocompatibility. In addition, the in vivo MRI results prove that Gd–DTPA@ZIF-8 can significantly enhance the T 1 -weighted MRI contrast effect of the tumour site. These advantages make Gd–DTPA@ZIF-8 a potential candidate for biomedical applications.

Fe/Mn Multilayer Nanowires as High-Performance T1-T2 Dual Modal MRI Contrast Agents

Xiaoming Cao,Liyuan Gu,Shike Hu,Aiman Mukhtar,Kaiming Wu

A lot of nanomaterials are using T1-T2dual mode magnetic resonance (MR) contrast agents (CAs), but multilayer nanowire (NW) with iron (Fe) and manganese (Mn) as T1-T2dual modal CAs has not been reported yet. Herein, we synthesized a Fe/Mn multilayer NW with an adjustable Fe layer, as T1-T2dual-mode CAs. The relaxation performance of Fe/Mn multilayer NW was studied at 1.5 T. Results show that, when the length of the Fe layer is about 10 nm and the Mn is about 5 nm, the r1value (21.8 mM−1s−1) and r2value (74.8 mM−1s−1) of the Fe/Mn multilayer NW are higher than that of Mn NW (3.7 mM−1s−1) and Fe NW (59.3 mM−1s−1), respectively. We predict that our Fe/Mn multilayer NW could be used as T1-T2dual mode MRI CAs in the near future. Keywords:dual modal;MRI;CAs;nanowire;relaxation

Methotrexate–Mn2+ based nanoscale coordination polymers as a theranostic nanoplatform for MRI guided chemotherapy

Yan Wu,Li Xu,Jiwen Qian,Leilei Shi,Yue Su,Youfu Wang,Dawei Li,Xinyuan Zhu

Theranostic nanoplatforms based on magnetic resonance imaging (MRI) technology have drawn much attention due to their inherent advantages such as non-invasiveness and high spatial resolution, but their construction is usually complicated and the compositions are uncontrollable. Herein, a simple and controllable theranostic nanoplatform by replacing a chelating agent with a chemotherapeutic drug directly was proposed. In detail, we elaborately synthesized Mn2+ and methotrexate (MTX) based nanoscale coordination polymers (NCPs) coated with poly(ethylene glycol) (PEG), MTX-Mn@PEG, in which Mn2+ ions served as MRI contrast agents (CAs), MTX was used as a chemotherapy drug and PEG provided a stable environment for nanoparticles, respectively. The obtained MTX-Mn@PEG NCPs showed a relatively uniform size, a pH-responsive feature and long blood circulation to enrich in the tumor location through the enhanced permeability and retention (EPR) effect. We determined the composition and coordination mechanism of MTX-Mn@PEG through thermogravimetric analysis (TGA), Fourier transform infrared (FI-TR) spectroscopy, powder X-ray diffraction (XRD) and so on. We determined the composition and coordination mechanism of MTX-Mn@PEG with the molar ratio of MTX to Mn2+ as 1 : 1. Then we confirmed that the antitumor ability was mediated by MTX through MTT and cell apoptosis assay in vitro. In vivo MR imaging and antitumor assays demonstrated the excellent effect of the NCPs in diagnosis and therapy of tumors. Overall, these novel MTX-Mn@PEG NCPs provide a theranostic nanoplatform for cancer treatment.
