In the treatment of foodborne antibiotic-resistant bacterial infections, the use of bactericidal nanozymes is one of the most promising non-antibiotic therapeutic strategies. However, this approach faces problems such as the inability to monitor the bactericidal effect in real time. In this work, novel ZnGa 2 O 4 :Cr/Mn persistent luminescent nanoparticles (PLNPs) with dual functionality of afterglow and catalysis were directly synthesized. The ZnGa 2 O 4 :Cr/Mn PLNPs not only possess inherent near-infrared persistent luminescence but also exhibit peroxidase- and oxidase-like nanozyme activities, making them applicable to autofluorescence-free afterglow imaging and drug-resistant bacteria eradication. The nanozyme activity of PLNPs was optimized without compromising their afterglow properties, and the bactericidal efficacy against Staphylococcus aureus and MRSA under acidic conditions was validated. These results provide a foundation for potential infection therapies. This dual-functional platform offers a new strategy for afterglow imaging-guided treatment of drug-resistant foodborne infections, with promising applications in food safety control, particularly for real-time tracking and elimination of Staphylococcus aureus contamination.

Conventional wound dressings exhibit limited functionality and bacterial-infected wounds may contribute to the development of drug resistance. Consequently, there is a critical demand for multifunctional nanocomposite hydrogels with enhanced antibacterial efficacy to facilitate rapid healing of infected wounds. Here, we developed a cascade reactor hydrogel (APT/OC) for releasing nitric oxide (NO) to achieve near-infrared (1064 nm) light-activated photothermal/photodynamic/gas synergistic therapy for drug-resistant bacteria-infected wounds. APT/OC hydrogel is prepared by inducing with polyphenolic amino acids and reversible crosslinking, using hyaluronic acid (HA), carboxymethyl chitosan (CMCS), L-arginine (L-Arg), polydopamine (PDA) and mesoporous titanium dioxide (m-TiO 2 ). In vitro and in vivo analyses revealed that the synergistic therapeutic effects triggered by near-infrared irradiation significantly inhibited the proliferation and biofilm formation of E. coli and MRSA ,and markedly accelerated the healing of drug-resistant wounds. Overall, the proposed hydrogel shows great potential in meeting the complex repair requirements arising from antibiotic-resistant bacterial infections of wounds.

Bacterial infection reduces quality of wound healing and even cause severe complications, but most current dressings adopt single antibacterial mode showed limited effectiveness. Herein, a plant extract quercetin (Que) composite chitosan/hyaluronic acid hydrogel (named CBCQ) with near-infrared (NIR) triggered controllable NO releasing was developed for efficient photothermal therapy/nitric oxide (PTT/NO) synergistic antibacterial and wound healing. Specifically, CBCQ hydrogel with chemical/physical crosslinked network was fabricated via host-guest interaction, metal coordination and Schiff base reaction. It exhibited porous structure, excellent injectability, self-healing, adhesion, antioxidant, and biocompatible properties suitable for wound dressings. Notably, superior PTT effect generated from Que-Fe 3+ has been demonstrated, which served as a photothermal switch for the controllable NO release, thereby enable to utilize the physiological functions of NO at different concentrations, improving antibacterial efficiency by integrating PTT and NO benefits, and further allowing sustained antibacterial through Que when NIR was off. Importantly, CBCQ hydrogel achieved better healing of skin incision than commercial 3M medical glue and Film, and significantly improved the healing rate and quality of Methicillin-resistant Staphylococcus aureus (MRSA) infected wound by efficient and sustained antibacterial activity, inhibiting inflammation, and promoting collagen deposition and angiogenesis, highlighting its promising application in skin incisions and infected full-thickness skin wounds.

Background : Biofilm infection caused by multidrug-resistant bacteria is difficult to eradicate by conventional therapies. Photodynamic therapy (PDT) is an effective antibacterial method for fighting against biofilm infection. However, the blocked photosensitizers outside of biofilm greatly limit the efficacy of PDT. Methods : Herein, a novel acid-responsive superporogen and photosensitizer (SiO 2 -P Ce6-IL ) was developed. Because of the protonation of the photosensitizer and the high binding energy of the polyionic liquid, SiO 2 -P Ce6-IL changed to positive SiO 2 -P IL + in an acidic microenvironment of biofilm infection. SiO 2 -P IL + could combine with negatively charged extracellular polymeric substances (EPS) and create holes to remove the biofilm barrier. To strengthen the interaction between SiO 2 -P IL + and EPS, SiO 2 -P IL + of high charge density was prepared by grafting the high-density initiation site of ATRP onto the surface of the SiO 2 base. Results : Due to the rapid protonation rate of COO - and the strong binding energy of SiO 2 -P IL + with EPS, SiO 2 -P Ce6-IL could release 90% of Ce6 in 10 s. With the stronger electrostatic and hydrophobic interaction of SiO 2 -P IL + with EPS, the surface potential, hydrophobicity, adhesion and mechanical strength of biofilm were changed, and holes in the biofilm were created in 10 min. Combining with the release of photosensitizers and the porous structure of the biofilm, Ce6 was efficiently concentrated in the biofilm. The in vitro and in vivo antibacterial experiments proved that SiO 2 -P Ce6-IL dramatically improved the PDT efficacy against MRSA biofilm infection. Conclusion : These findings suggest that SiO 2 -P Ce6-IL could rapidly increase the concentration of photosensitizer in biofilm and it is an effective therapy for combating biofilm infection.

Bacterial infection poses a significant impediment in wound healing, necessitating the development of dressings with intrinsic antimicrobial properties. In this study, a multilayered wound dressing (STPU@MTAI 2 /AM 1 ) was reported, comprising a surface-superhydrophobic treated polyurethane (STPU) sponge scaffold coupled with an antimicrobial hydrogel. A superhydrophobic protective outer layer was established on the hydrophilic PU sponge through the application of fluorinated zinc oxide nanoparticles (F-ZnO NPs), thereby resistance to environmental contamination and bacterial invasion. The adhesive and antimicrobial inner layer was an attached hydrogel (MTAI 2 /AM 1 ) synthesized through the copolymerization of N -[2-(methacryloyloxy)ethyl]- N, N, N -trimethylammonium iodide and acrylamide, exhibits potent adherence to dermal surfaces and broad-spectrum antimicrobial actions against resilient bacterial strains and biofilm formation. STPU@MTAI 2 /AM 1 maintained breathability and flexibility, ensuring comfort and conformity to the wound site. Biocompatibility of the multilayered dressing was demonstrated through hemocompatibility and cytocompatibility studies. The multilayered wound dressing has demonstrated the ability to promote wound healing when addressing MRSA-infected wounds. The hydrogel layer demonstrates no secondary damage when peeled off compared to commercial polyurethane sponge dressing. The STPU@MTAI 2 /AM 1 -treated wounds were nearly completely healed by day 14, with an average wound area of 12.2% ± 4.3%, significantly lower than other groups. Furthermore, the expression of CD31 was significantly higher in the STPU@MTAI 2 /AM 1 group compared to other groups, promoting angiogenesis in the wound and thereby contributing to wound healing. Therefore, the prepared multilayered wound dressing presents a promising therapeutic candidate for the management of infected wounds. Statement of significance Healing of chronic wounds requires avoidance of biofouling and bacterial infection. However developing a wound dressing which is both anti-biofouling and antimicrobial is a challenge. A multilayered wound dressing with multifunction was developed. Its outer layer was designed to be superhydrophobic and thus anti-biofouling, and its inner layer was broad-spectrum antimicrobial and could inhibit biofilm formation. The multilayered wound dressing with adhesive property could easily be removed from the wound surface preventing the cause of secondary damage. The multilayered wound dressing has demonstrated good abilities to promote MRSA-infected wound healing and presents a viable treatment for MRSA-infected wound.

Background Scutellariae Radix - Coptidis Rhizoma herb pair is a classic traditional Chinese medicine (TCM) combination with heat-clearing, dampness-drying and detoxifying effects, extensively applied clinically against bacterial infections. Traditional water decoction and ethanol pretreatment are mainstream extraction approaches, whereas existing comparisons merely emphasize quantitative variations of bioactive compounds, neglecting the underlying supramolecular assembly differences of effective substances. Comparative analysis of solvent-induced supramolecular variations provides a novel perspective to interpret differential pharmacodynamic material bases of TCM formulas. Methods Macroscopic and microscopic morphologies of HH-0 and HH-50 were firstly characterized by visual inspection, SEM and DLS. Their phytochemical profiles were subsequently analyzed by UHPLC-Q-Orbitrap HRMS and HPLC. Antibacterial activities against MRSA were further quantified via turbidimetry and plate assays. UV, FT-IR and MD simulations revealed divergent supramolecular assembly behaviors. Finally, untargeted metabolomics and assembly-peptidoglycan interaction simulations elucidated the underlying anti-MRSA mechanisms. Results The solvent pretreatment endowed HH-0 and HH-50 supramolecules with distinct characteristics. HH-50 featured rougher supramolecular surfaces and higher Zeta potential with favorable dispersibility. The two supramolecules shared consistent active constituents, and HH-50 showed elevated berberine abundance and enhanced anti-MRSA activity. Notably, HH-50 maintained superior antibacterial efficacy and interference against peptidoglycan synthesis and energy metabolism even at identical berberine doses. This confirmed that divergent supramolecular conformations determined antibacterial differences, as diminished hydrogen bonds and weakened van der Waals and electrostatic interactions enhanced the binding affinity between assemblies and MRSA. Conclusion Beyond chemical composition, solvent pretreatment (water versus 50% ethanol) markedly modulates supramolecular assembly behaviors and subsequent assembly-bacteria interactions, thereby regulating the anti-MRSA efficacy of the Scutellariae Radix - Coptidis Rhizoma herb pair. This work emphasizes that supramolecular configurations, rather than simple differences in chemical contents, dominate the pharmacological activities of traditional Chinese medicine preparations. Persistent bacterial infections and the imbalance in immune regulation induced by oxidative stress present a significant challenge in diabetic wound healing. In this study, we developed a novel dual-network hydrogel system composed of chitosan/polyacrylic acid (PEC) and polyoxometalate PMo12 (PMo12-PEC) loaded with baicalein (BA), designated as PMo12-BA-PEC, for the treatment of methicillin-resistant Staphylococcus aureus (MRSA)-infected diabetic wounds. The hydrogel demonstrated enhanced mechanical strength and elasticity, facilitating effective wound adherence and accommodating tissue movement. Upon 808 nm near-infrared (NIR) light irradiation, the photothermal properties of PMo12 enabled controlled low-temperature hyperthermia and accelerated BA release. Remarkably, the hydrogel achieved an antibacterial efficacy of 99.45% ± 0.12% against MRSA following 5 minutes of NIR exposure, while exhibiting potent antioxidant and anti-inflammatory capabilities to scavenge reactive oxygen species and mitigate inflammatory responses. Comprehensive evaluation revealed that the PMo12-BA-PEC hydrogel significantly promoted angiogenesis, enhanced collagen deposition, inhibited bacterial growth, and modulated immune regulation, thereby accelerating the wound healing process in diabetic rat models. These findings suggest that the PMo12-BA-PEC hydrogel represents a promising biomaterial platform for clinical management of diabetic wounds and potential treatment of oxidative stress-related disorders.

Bacterial infection and insufficient angiogenesis caused by immune dysregulation pose significant challenges to diabetic wound healing. Effectively combating bacterial infection and modulating endothelial function through the polarization of M2 macrophages to promote angiogenesis are considered as effective strategies for treating diabetic wounds. However, local treatment that achieves dual effects of antibacterial and immunomodulation through simple pathways remains limited. There is a critical need for the development of a wound dressing that possesses both antimicrobial activity and the ability to modulate the immune response within the wound microenvironment. Herein, the positively charged chitosan-derived carbon dots (CCDs) with excellent antibacterial activity were synthesized by a one-step hydrothermal method, demonstrating significant efficacy against methicillin-resistant Staphylococcus aureus (MRSA). The carbomer hydrogel (CH) was designed for loading CCDs to be the hybrid hydrogel (CCDs@CH). CCDs@CH not only possessed pH responsiveness and enhanced mechanical properties that enabled adaptation to the wound microenvironment, but also exhibited pronounced immunomodulatory and pro-angiogenic activities. In a rat model of MRSA-infected skin wound, CCDs@CH significantly accelerated wound healing, resulting in a healing rate of 72.3 % on day 7 and 98.7 % on day 14. Furthermore, a reduction in bacterial load at the wound site by over 90 % was observed in CCDs@CH group, whose cure was also confirmed by histological examination. This topical hydrogel provides a straightforward and effective approach for treating infectious wounds and offers new insights into treating diabetic wounds.

Diabetic wounds represent a major clinical challenge due to their susceptibility to bacterial biofilm infections, persistent oxidative stress, and impaired tissue regeneration. Current therapeutic approaches typically address these pathological factors individually, limiting their overall efficacy in achieving complete wound closure. There is thus an urgent need for integrated platforms capable of simultaneously targeting multiple barriers to healing. Here we show that a self-healing hydrogel composed of quaternized chitosan-phenylboronic acid (QCSP), ceria nanoparticles (CeNPs), and embryonic stem cell-derived exosomes (ESC-Exo) effectively eradicates methicillin-resistant Staphylococcus aureus (MRSA) biofilms, scavenges reactive oxygen species, and promotes tissue regeneration in diabetic mice. The QCSP matrix provides inherent antibacterial activity and injectable, self-healing properties, while CeNPs function as catalytic antioxidants through reversible Ce 3+ /Ce 4+ redox cycling. Incorporation of ESC-Exo further enhances angiogenesis and re-epithelialization. In an MRSA-infected diabetic wound model, this multifunctional hydrogel achieves near-complete wound closure within 12 days alongside significant biofilm clearance and collagen deposition. This work demonstrates that integrating natural polymer-based hydrogels with nanozymes and bioactive exosomes offers a promising strategy for managing complex chronic wounds.

Methicillin-resistant Staphylococcus aureus (MRSA) infection and compromised immunity are the severe complications associated with implantation surgery in diabetes mellitus. Enhancing the antibacterial and immunomodulatory properties of implants represents an effective approach to improve the osseointegration of implant in diabetes mellitus. Herein, guanidination carbon dots (GCDs) with antibacterial and immunoregulatory functions are synthesized. The GCDs demonstrate killing effect on MRSA without detectable induced resistance. Additionally, they promote the polarization of macrophages from the M1 to M2 subtype, with the inhibiting pro-inflammatory cytokines and promoting anti-inflammatory factors. Correspondingly, GCDs are immobilized onto sulfonated polyether ether ketone (SP@GCDs) using a polyvinyl butyraldehyde (PVB) coating layer through soaking-drying technique. SP@GCDs maintain stable antibacterial efficacy against MRSA for six consecutive days and retain the immunomodulatory function, while also possessing the long-term storage stability and biocompatibility of more than 6 months. Moreover, SP@GCDs significantly promote the proliferation and mineralization of osteoblasts. SP@GCDs facilitate osteogenesis through immunoregulatory. Additionally, SP@GCDs exert stable antibacterial and immune regulatory functions in implantation site of a diabetes rat, effectively promoting implant osseointegration regardless of the MRSA infection. These findings provide valuable insights into implant modification through designing nanomaterials with multifunction for enhancing osseointegration of diabetes mellitus, suggesting the promising clinical application prospects.

Nanozymes, featuring intrinsic biocatalytic effects and broad-spectrum antimicrobial properties, are emerging as a novel antibiotic class. However, prevailing bactericidal nanozymes face a challenging dilemma between biofilm penetration and bacterial capture capacity, significantly impeding their antibacterial efficacy. Here, this work introduces a photomodulable bactericidal nanozyme (ICG@hMnO x ), composed of a hollow virus-spiky MnO x nanozyme integrated with indocyanine green, for dually enhanced biofilm penetration and bacterial capture for photothermal-boosted catalytic therapy of bacterial infections. ICG@hMnO x demonstrates an exceptional capability to deeply penetrate biofilms, owing to its pronounced photothermal effect that disrupts the compact structure of biofilms. Simultaneously, the virus-spiky surface significantly enhances the bacterial capture capacity of ICG@hMnO x . This surface acts as a membrane-anchored generator of reactive oxygen species and a glutathione scavenger, facilitating localized photothermal-boosted catalytic bacterial disinfection. Effective treatment of methicillin-resistant Staphylococcus aureus -associated biofilm infections is achieved using ICG@hMnO x , offering an appealing strategy to overcome the longstanding trade-off between biofilm penetration and bacterial capture capacity in antibacterial nanozymes. This work presents a significant advancement in the development of nanozyme-based therapies for combating biofilm-related bacterial infections.

Bacterial infection and local growth factor deficiency are two of the major causes of the nonunion of diabetic wounds. Antimicrobial peptides (AMPs) are believed to be alternatives to antibiotics against drug-resistant bacterial infections. 8-Bromoadenosine-3’, 5’-cyclic monophosphate (8Br-cAMP) can promote cells to secrete growth factors and accelerate cell proliferation . In the present study, we constructed a hydrogel with antimicrobial peptide Jelleine-1 (J-1) and 8Br-cAMP without any other gelators or chemical crosslinking agents. The hydrogel was proved to promote the secretion of transforming growth factor-β (TGF-β) and vascular endothelial growth factor-A (VEGFA) in vitro and in vivo . Notably, it exhibited potent potential for wound healing in methicillin-resistant Staphylococcus aureus ( MRSA ) infected diabetic wounds. This would be attributed to the retention of AMPs and 8Br-cAMP on the wound site by the hydrogel system. In addition, the hydrogel also showed good biodegradability , proper stability, and good biocompatibility. This study would shed light on the development of carrier-free and multifunctional hydrogel for wound healing. Statement of significance Bacterial infection and local growth factor deficiency are two of the major causes for the nonunion of refractory wounds. In the present study, an injectable carrier-free hydrogel was constructed of a natural antimicrobial peptide J-1 and 8Br-cAMP by eco-friendly physical crosslinking without any other gelators or chemical crosslinking agents. The hydrogel exhibited excellent antimicrobial activity and was proved to promote the secretion of TGF-β and VEGFA in vitro and in vivo . Correspondingly, the hydrogel showed exceptionally wound healing effects in the wound model of MRSA infected diabetic rats. This study would provide an alternative strategy or a potential hydrogel dressing for the treatment of chronic or refractory wounds.
