Targeting fermentation via quality indicators mapping during the overall processing of Xanthoceras sorbifolium Bunge black tea: Investigation of stable metabolic pathways and key substances

Hongyuan Zhao,Siyuan Wang,Meixi Zhong,Jiayi Xiao,Wenjie Guan,Haiwei Ren,Yuan Liu,Xiaoxiao Feng

This study demonstrates flavor compound variation and the stable pattern of Xanthoceras sorbifolium Bunge Black Tea (XSBT) at different processes through multi-omics techniques. The dominant aroma compounds were identified as alcohols, aldehydes, esters, and ketones during processing and correlation analysis preliminarily shows that fermentation is the decisive stage for quality formation, as evidenced by the contents of tea polyphenols (84.98 ± 0.39 mg/g), total flavonoids (7.30 ± 0.06%) and water extracts (32.57 ± 0.45%). The flavor evolution of XSBT was mainly formed through lipid oxidation and the Maillard reaction during fermentation stage by the content alternations of aldehydes and ketones. Metabolomic interrogation of the fermentation stage further disclosed that α-linolenic acid metabolism constitutes the pivotal pathway, yielding 1-penten-3-ol and 1-penten-3-one (ROAV = 57.11) as key volatile metabolites, which reveals a stable and active lipid metabolism during fermentation. These findings could provide material, pathway and key component support for improving XSBT fermentation quality.

Molecular insights into the mechanisms of a leaf color mutant in Anoectochilus roxburghii by gene mapping and transcriptome profiling based on PacBio Sequel II

Huang Huiming,Zou Hui,Lin Hongting,Dai Yimin,Lin Jiangbo

Plants with partial or complete loss of chlorophylls and other pigments are frequently occurring in nature but not commonly found. In the present study, we characterize a leaf color mutant ‘ arly01 ’ with an albino stripe in the middle of the leaf, which is an uncommon ornamental trait in Anoectochilus roxburghii . The albino “mutant” middle portion and green “normal” leaf parts were observed by transmission electron microscopy (TEM), and their pigment contents were determined. The mutant portion exhibited underdevelopment of plastids and had reduced chlorophyll and other pigment (carotenoid, anthocyanin, and flavonoid) content compared to the normal portion. Meanwhile, comparative transcript analysis and metabolic pathways mapping showed that a total of 599 differentially expressed genes were mapped to 78 KEGG pathways, most of which were down-regulated in the mutant portion. The five most affected metabolic pathways were determined to be oxidative phosphorylation, photosynthesis system, carbon fixation & starch and sucrose metabolism, porphyrin and chlorophyll metabolism, and flavonoid biosynthesis. Our findings suggested that the mutant ‘ arly01 ’ was a partial albinism of A. roxburghii , characterized by the underdevelopment of chloroplasts, low contents of photosynthetic and other color pigments, and a number of down-regulated genes and metabolites. With the emergence of ornamental A. roxburghii in southern China, ‘ arly01 ’ could become a popular cultivar due to its unique aesthetics.

In Situ Mapping of Activity Distribution of V(II)/V(III) and Onset Potential Distribution of Hydrogen Evolution Side Reaction in Vanadium Flow Batteries

Hao Lin,Yang Xu,Xiao Chen,Zengxian Fang,Tian Yan,Kaijie Ma,Le Liu,Jingyu Xi

Vanadium flow batteries (VFBs) face a challenge with the low reaction rates of the V(II)/V(III) redox couple, which limits the performance of VFBs. Additionally, the negative electrode in VFBs is often accompanied by the persistent hydrogen evolution reaction (HER), which is difficult to eliminate. Therefore, understanding the spatial distribution of activity on the negative electrode and the HER side reaction on the electrode surface is of critical importance. This study proposes a weak measurement imaging method to characterize the spatial distribution of surface activity and HER onset potential on the negative electrode in VFBs). This method enables the visualization and in situ detection of key parameters such as the absolute values of |i pa |, |i pc |, |∆E|, |i pc /i pa |, and the HER onset potential. By comparing three different types of graphite felts with varying activity levels, it validates the feasibility of this method. Furthermore, electrochemical stability tests are conducted to study the electrodes repeatability, uniformity, and durability. This method holds promise in guiding the design of electrodes with enhanced activity, good reversibility, minimized HER side reactions, and uniform distribution.

Mapping the kidney distribution of Huangkui capsule-derived components in diabetic nephropathy treatment: a mass spectrometry imaging and metabolomics approach nominates quercetin and its conjugates as bioactive candidates

Liu Chang,Li Ping,Han Jinwei,Zheng Ying,Yan Guangli,Wang Mengmeng,Wang Yuhang,Chen Xiangmei,Yan Xiaotong,Sun Hui,Guan Shihan,Wang Xijun

The Huangkui capsule (HKC) demonstrates significant efficacy against diabetic nephropathy (DN), yet its bioactive components that exert effects within the target organ, the kidney, have remained poorly characterized. This study aimed to elucidate the renal in situ effective constituents of HKC by integrating multimodal mass spectrometry analyses. A spontaneous DN model was established using db/db mice. Physiological parameters, biochemical indices, and histopathological assessments confirmed the model’s success and the therapeutic effects of HKC intervention. Serum pharmacochemistry based on UPLC-Q-TOF-MS/MS characterized 24 absorbed components. Among them, quercetin, isoquercetin, and their metabolites exhibited the strongest correlation with pharmacodynamic indicators. Crucially, matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-TOF-MSI) directly visualized and identified these quercetin-derived conjugates, particularly quercetin glucuronide, within kidney tissues, suggesting their potential as primary in situ bioactive candidates. This study is the first to spatially resolve the kidney distribution of HKC’s active constituents, providing a scientific basis for understanding its mechanism of action and advancing quality control protocols. In vitrometabolic mapping of neobavaisoflavone in human cytochromes P450 and UDP-glucuronosyltransferase enzymes by ultra high-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry

Jinjin Xu,Mengsen Li,Zhihong Yao,Yezi Zhang,Shishi Li,Liufang Hu,Zifei Qin,Frank J. Gonzalez,Xinsheng Yao

Neobavaisoflavone (NBIF), a phenolic compound isolated from Psoralea corylifolia L., possesses several significant biological properties. However, the pharmacokinetic behaviors of NBIF have been characterized as rapid oral absorption, high clearance, and poor oral bioavailability. We found that NBIF underwent massive glucuronidation and oxidation by human liver microsomes (HLM) in this study with the intrinsic clearance ( CL int ) values of 12.43, 10.04, 2.01, and 6.99 μL/min/mg for M2, M3, M4, and M5, respectively. Additionally, the CL int values of G1 and G2 by HLM were 271.90 and 651.38 μL/min/mg, respectively, whereas their respective parameters were 59.96 and 949.01 μL/min/mg by human intestine microsomes (HIM). Reaction phenotyping results indicated that CYP1A1, 1A2, 2C8, and 2C19 were the main contributors to M4 (34.96 μL/min/mg), M3 (29.45 μL/min/mg), M3 (13.16 μL/min/mg), and M2 (63.42 μL/min/mg), respectively. UGT1A1, 1A7, 1A8, and 1A9 mainly catalyzed the formation of G1 (250.87 μL/min/mg), G2 (438.15 μL/min/mg), G1 (92.68 μL/min/mg), and G2 (1073.25 μL/min/mg), respectively. Activity correlation analysis assays showed that phenacetin- N -deacetylation was strongly correlated to M3 ( r  = 0.860, p  = 0.003) and M4 ( r  = 0.775, p  = 0.014) in nine individual HLMs, while significant activity correlations were detected between paclitaxel-6-hydroxylation and M2 ( r  = 0.675, p  = 0.046) and M3 ( r  = 0.829, p  = 0.006). There was a strong correlation between β-estradiol-3- O -glucuronide and G1 ( r  = 0.822, p  = 0.007) and G2 ( r  = 0.689, p  = 0.040), as well as between propofol- O -glucuronidation and G1 ( r  = 0.768, p  = 0.016) and G2 ( r  = 0.860, p  = 0.003). Moreover, the phase I metabolism and glucuronidation of NBIF revealed marked species differences, and mice are the best animal model for investigating the metabolism of NBIF in humans. Taken together, characterization of NBIF-related metabolic pathways involving in CYP1A1, 1A2, 2C8, 2C19, and UGT1A1, 1A7, 1A8, 1A9 are helpful for understanding the pharmacokinetic behaviors and conducting in-depth pharmacological studies.

Metabolism and disposition of corylifol A from Psoralea corylifolia: metabolite mapping, isozyme contribution, species differences and identification of efflux transporters for corylifol A-O-glucuronide in HeLa1A1 cells

Yang Li,Jinjin Xu,Chunxia Xu,Zifei Qin,Shishi Li,Liufang Hu,Zhihong Yao,Frank J. Gonzalez,Xinsheng Yao

Corylifol A (CA), a phenolic compound from Psoralea corylifolia, possessed several biological properties but poor bioavailability. Here we aimed to investigate the roles of cytochromes P450s (CYPs), UDP-glucuronosyltransferases (UGTs) and efflux transporters in metabolism and disposition of CA.Metabolism of CA was evaluated in HLM, expressed CYPs and UGTs. Chemical inhibitors and shRNA-mediated gene silencing of multidrug resistance-associated proteins (MRPs) and breast cancer resistance protein (BCRP) were performed to assess the roles of transporters in CA disposition.Three oxidated metabolites (M1–M3) and two glucuronides (M4–M5) were detected. The intrinsic clearances (CLint) values of M1 and M4 in HLM were 48.10 and 184.03 μL/min/mg, respectively. Additionally, CYP1A1, 2C8 and 2C19 were identified as main contributors with CLint values of 13.01–49.36 μL/min/mg, while UGT1A1, 1A7, 1A8 and 1A9 were with CLint values ranging from 85.01 to 284.07 μL/min/mg. Furthermore, activity correlation analysis proved CYP2C8, UGT1A1 and 1A9 were the main active hepatic isozymes. Besides, rats and monkeys were appropriate model animals. Moreover, dipyridamole and MK571 both could significantly inhibit M4 efflux. Gene silencing results also indicated MRP4 and BCRP were major contributors in HeLa1A1 cells.Taken together, CYPs, UGTs, MRP4 and BCRP were important determinants of CA pharmacokinetics.
