Biomimetic Nano-Immunoactivator via Ionic Metabolic Modulation for Strengthened NIR-II Photothermal Immunotherapy

Xiaodan Wei,Honglin Huang,Junhan Guo,Ningxi Li,Qingzhi Li,Tian Zhao,Geng Yang,Lulu Cai,Hong Yang,Chunhui Wu,Yiyao Liu

Reprogramming the immunologically “cold” environment of solid tumors is currently becoming the mainstream strategy to elicit powerful and systemic anticancer immunity. Here, a facile and biomimetic nano-immunnoactivator ( CuS/Z@M 4T1 ) is detailed by engineering a Zn 2+ -bonded zeolitic imidazolate framework-8 (ZIF-8) with CuS nanodots (NDs) and cancer cell membrane for amplified near-infrared-II (NIR-II) photothermal immunotherapy via Zn 2+ metabolic modulation. Taking advantage of the NIR-II photothermal effect of CuS NDs and the acidic responsiveness of ZIF-8, CuS/Z@M 4T1 rapidly causes intracellular Zn 2+ pool overload and disturbs the metabolic flux of 4T1 cells, which effectively hamper the production of heat shock proteins and relieve the resistance of photothermal therapy (PTT). Thus, amplified immunogenic cell death is evoked and initiates the immune cascade both in vivo and in vitro as demonstrated by dendritic cells maturation and T-cell infiltration. Further combination with antiprogrammed death 1 (aPD-1) achieves escalated antitumor efficacy which eliminates the primary, distant tumor and avidly inhibits lung metastasis due to cooperation of enhanced photothermal stimulation and empowerment of cytotoxic T lymphocytes by aPD-1. Collectively, this work provides the first report of using the intrinsic modulation property of meta-organometallic ZIF-8 for enhanced cancer photoimmunotherapy together with aPD-1, thereby inspiring a novel combined paradigm of ion-rich nanomaterials for cancer treatment.

Water-Soluble High-Quality Ag2Te Quantum Dots Prepared by Mutual Adaptation of Synthesis and Surface Modification for In Vivo Imaging

Xue-Hui Shi,Ying-Ying Dai,Lei Wang,Zhi-Gang Wang,Shu-Lin Liu

Near-infrared (NIR) in vivo fluorescence imaging has exhibited the distinct advantage of high optical resolution at deeper penetration into biological tissues. Ag2Te quantum dots (QDs), with a relatively narrow band gap, show great promise for fluorescence emission at long wavelengths in the second near-infrared (NIR-II) window for bioimaging. However, existing Ag2Te QDs have severely hindered the application of in vivo bioimaging due to their poor fluorescence brightness and stability, so it is important to prepare Ag2Te QDs with high quantum yield and stability as well as high biocompatibility in the NIR-II window. Herein, we designed an integrated method for the preparation of water-soluble Ag2Te QDs by mutual adaptation of QD synthesis and surface modification. We first synthesized high-quality Ag2Te QDs with different NIR-II emission wavelengths and the photoluminescence quantum yields (PLQYs) up to 6.51% by rapidly injecting the TBP-Te precursor into a hot solvent to form a highly fluorescent Ag2Te core. Then water-dispersible Ag2Te QDs were obtained by direct exchange of the hydrophobic Ag2Te QD surface ligands with thiol ligands. The PLQY of the water-soluble Ag2Te QDs obtained by this method can still be maintained at 4.94%. With these highly bright and stable Ag2Te QDs, the abdominal vessels, hindlimb arterial vessels, venous vessels, sacral lymph nodes, and tumor vessels were visualized non-invasively in vivo in the NIR-II window in mice. The results demonstrate that the integrated strategy of QD synthesis and modification provides valuable technical support for further in-depth applications of Ag2Te QDs.

Fluorinated Chitosan Mediated Synthesis of Copper Selenide Nanoparticles with Enhanced Penetration for Second Near-Infrared Photothermal Therapy of Bladder Cancer

Shaohua Zhang,Guangzhi Li,Dashi Deng,Yizhi Dai,Zhuang Liu,Song Wu

The emergence of near-infrared-II (NIR-II) light activated photomedicines has extended the penetration depth for noninvasive theranostics, especially for photothermal nanomedicines. Intravesical instillation therapy of bladder cancer (BCa) is restricted greatly due to the short residence time and the low penetration ability of therapeutic agents in clinic. Therefore, improving the mucoadhesiveness and penetration of therapeutic agents are crucial factors in treatment of BCa. The development of NIR-II light triggered photothermal transduction agents for enhanced penetration and photothermal ablation of BCa post-intravesical instillation has not yet been investigated. Herein, Cu 2- x Se biocompatible particles are prepared through a fluorinated chitosan (FCS)-mediated one-pot aqueous approach, which possesses enhanced penetrating ability and high photothermal conversion capability for effective NIR-II photothermal therapy (PTT). Owing to the outstanding transmucosal ability of FCS, FCS capped Cu 2- x Se nanoparticles exhibit much better transmucosal and penetration performance than their non-fluorinated counterparts to significantly improve the therapeutic efficacy of NIR-II PTT to ablate orthotopic bladder tumors. This study presents a promising phototherapy agent based on Cu 2- x Se modified with FCS for efficient NIR-II PTT for BCa treatment post-intravesical instillation.

Targeted nanobody complex enhanced photodynamic therapy for lung cancer by overcoming tumor microenvironment

Zhang Qing,Wu Lian,Liu Shaozheng,Chen Qingjie,Zeng Lingpeng,Chen Xuezhong,Zhang Qing

Background To investigate the efficacy of a PLGA-based nanobody complex in photodynamic therapy (PDT) and NIR-II imaging in A549 tumor hypoxic model.MethodIR1048-MZ was firstly synthesized by conjugating a nitro imidazole group to IR1048. IR1048-MZ and Cat were then encapsulated in PLGA-SH solution. Anti-EGFR-Nanobody was also expressed and purified, and finally Anti-EGFR-Nanobody@PLGA-IR1048MZ-Cat (Nb@IC-NPs) nanobody complex was obtained based on the formation of desulfide bond between PLGA-SH and Anti-EGFR-Nanobody. Size distribution and morphology were characterized by TEM and DLS. Spectrum of Nb@IC-NPs towards NTR was measured by UV and fluorescence, while the particle’s selective response was studied using fluorescence. The uptake of Nb@IC-NPs in A549 cells was observed by flow cytometry and CLSM. In the meantime, its’ catalytic ability that decomposes H2O2 both extra-and intra-cellular was observed by fluorescence and CLSM. In vitro photodynamic toxicity of Nb@IC-NPs was examined by MTT, Live/Dead Cell Staining, Flow Cytometry and Apoptosis Assay. Tumor-bearing model was constructed to observe a semi-quantitative fluorescent distribution and the possibility of NIR-II fluorescence/photoacoustic (PA) imaging. Effect of Nb@IC-NPs on enhancing A549 tumor hypoxia and expression profile of HIF-1α was investigated in the presence of NIR. An A549 tumor metastasis model was also constructed to confirm the complex’ potential to destroy primary tumor, inhibit lung metastasis, and prolong mice’ survival. Lastly, impact of Nb@IC-NPs on mice’ main organs and blood indices was observed. Results Nb@IC-NPs was successfully fabricated with good homogeneity. The fluorescent absorbance of Nb@IC-NPs showed a linear relationship with the concentration of NTR, and a higher concentration of NTR corresponded to a stronger photoacoustic signal. In addition, Nb@IC-NPs showed a stable selectivity toward NTR. Our results also suggested a high efficient uptake of Nb@IC-NPs in A549 cells, which was more efficient than IC-NPs and IR1048-MZ alone. In vitro assays confirmed the effects of Nb@IC-NPs on catalytic O2 generation even in hypoxic cells. The cell viability was upregulated with the nanocomplex at the absence of the laser, whereas it was dramatically declined with laser treatment that excited at 980 nm. Nb@IC-NPs achieved tumor hypoxia NIR-II/PA imaging through assisting A549 gathering. When NIR was applied, Nb@IC-NPs can significantly relieve A549 cellular/tumor hypoxia by generating more reactive oxygen species (ROS), which in turn helps lower the expression level of HIF-1α. In summary, Nb@IC-NPs based PDT can efficiently decimate A549 primary tumor, inhibit metastatic lung cancer, and prolong the lifespan of the mice under tolerable dosage. At last, in vivo toxicity tests of the nanocomplex showed its biosafety to the main organs and normal blood indices values. Conclusion Nb@IC-NPs improves tumor hypoxia through catalytic reaction and lowers the expression level of HIF-1α. It achieves tumor PA imaging through intensified NIR-II fluorescence signal that caused by response of the complex to the lesion’s nitroreductase (NTR). Nb@IC-NPs based PDT can efficiently kill A549 primary tumor, inhibit a lung metastasis, as well as prolong mice’ survival cycle.

Bioorthogonal Regulated Metabolic Balance for Promotion of Ferroptosis and Mild Photothermal Therapy

Ying Huang,Huisi Zhao,Yu Zhang,Chuanqi Zhao,Jinsong Ren,Xiaogang Qu

The nanozyme with NADPH oxidase (NOX)-like activity can promote the consumption of NADPH and the generation of free radicals. In consideration of that the upregulation of glucose-6-phosphate dehydrogenase (G6PD) would accelerate the compensation production of NADPH, for inhibition of G6PD activity, our designed bioorthogonal nanozyme can in situ catalyze pro-DHEA to produce G6PD inhibitor and dehydroepiandrosterone (DHEA) drugs to inhibit G6PD activity. Therefore, the well-defined platform can disrupt NADPH homeostasis, leading to the collapse of the antioxidant defense system in the tumor cells. The enzyme-like activity of PdCuFe is further enhanced when irradiated by NIR-II light. The destruction of NADPH homeostasis can promote ferroptosis and, in turn, facilitate mild photothermal therapy. Our design can realize NADPH depletion and greatly improve the therapeutic effect through metabolic regulation, which may provide inspiration for the design of bioorthogonal catalysis

APE1-Activated and NIR-II Photothermal-Enhanced Chemodynamic Therapy Guided by Amplified Fluorescence Imaging

Xiaofeng Bi,Jinyue Feng,Xiyuan Feng,Dianpeng Li,Yumin Wang,Shulin Zhao,Liangliang Zhang

The development of intelligent nanotheranostic technology that integrates diagnostic and therapeutic functions holds great promise for personalized nanomedicine. However, most of the nanotheranostic agents exhibit “always-on” properties and do not involve an amplification step, which may largely limit imaging contrast and restrict therapeutic efficacy. Herein, we construct a novel nanotheranostic platform (Hemin/DHPs/PDA@CuS nanocomposite) by assembling DNA hairpin probes (DHPs) and hemin on the surface of PDA@CuS nanosheets that enables amplified fluorescence imaging and activatable chemodynamic therapy (CDT) of tumors. The cancer-relevant APE1 triggers nucleic acid amplification with DHPs to generate activatable and amplified fluorescence signals for discriminating cancer cells from normal cells. Meanwhile, excessive G-quadruplex/hemin-based DNAzyme are also activated, and they function as Fenton-like catalysts to catalyze the production of highly toxic hydroxyl radicals (•OH) for CDT. Moreover, owing to the excellent photothermal conversion efficiency in the near-infrared-II (NIR-II) window, the PDA@CuS not only improves the catalytic performance of CDT but also furnishes PTT. A remarkable antitumor therapeutic effect is demonstrated both in vitro and in vivo. Therefore, the Hemin/DHPs/PDA@CuS nanocomposite is expected to provide a promising avenue for precise imaging-guided antitumor therapy.

Design of donor–acceptor conjugated polymers based on diketopyrrolopyrrole for NIR-II multifunctional imaging

Mengyu Cao,Xuezhen Ma,Chang Wang,Wentao Zou,Fang Wang,Bing Yu,Hailin Cong,Youqing Shen

Diketopyrrolopyrrole (DPP) is an excellent photosensitizer and photothermal agent with the advantages of good planarity, strong electron affinity, high electron mobility, easy purification, easy structural modification and high molar absorption coefficient. It is regarded as one of the ideal choices for the design and synthesis of efficient organic photovoltaic materials. Therefore, two kinds of donor–acceptor (D–A) conjugated polymers were designed and synthesized with DPP as the acceptor, and their optical properties and applications in the near-infrared region were studied. The quantum yield (QY) of PBDT-DPP is 0.46%, and the highest temperature reached within 10 minutes after irradiation with a 660 nm laser is 60 °C. Another polymer, EDOT-DPP, has a QY of 0.48%, and its semiconductor polymer nanoparticle aqueous solution can reach 60 °C within 12 minutes under laser irradiation, achieving photothermal treatment of nude mice tumors. Both polymer NPs have good biocompatibility and promising applications in bioimaging and photothermal therapy.

De Novo siRNA nanoplatform with NIR-II imaging for treatment of carotid atherosclerotic plaques

Jialu Gao,Hua Zhu,Huanhuan Cai,Qihang Ding,Xiaoxing Xiong,Tong Zhang,Bo Chen,Wumei Wang,Xinpeng Ju,Jianying Huang,Xiaodong Zeng,Jong Seung Kim,Xuechuan Hong

Reducing foam cell formation and accumulation within plaques is a critical strategy for treating atherosclerosis. Although current lipid-lowering therapies have significantly improved the management of atherosclerosis, the risk of cardiovascular events remains. Thus, it is necessary to explore next-generation therapies to reduce foam cell formation, inflammation, and oxidative stress. To address these challenges, we present an innovative near-infrared II (NIR-II) fluorescent dye coupled with a lesion-targeting peptide, forming a molecular self-assembly nanoplatform for effective in vitro small interfering RNA (siRNA) delivery. This platform downregulates Epsin1/2, a family of ubiquitin-binding endocytic adaptors, upregulates the expressions of ABCG1 and LRP-1 proteins, and markedly reduces foam cell formation. Additionally, this system enables real-time tracking of plaque accumulation, therapeutic response, and efficacy through in vivo noninvasive NIR-II imaging. Systemic siRNA delivery significantly suppresses lipid accumulation, plaque size, and necrotic core area in the carotid plaque model of ApoE −/− mice. These results suggest that this multi-functional nanoplatform could become an effective tool for NIR-II image-guided siRNA delivery for personalized treatment of atherosclerosis.

Cr3+-Nd3+-Yb3+ Co-Activated NIR I to NIR II Mechanoluminescence Toward Biomechanics Detection

Ziyi Niu,Pengfei Zhang,Wenbo Chen,Junyang Xia,Zhili Zhang,Jinwei Liu,Yanyan Li,Bitao Liu,Lei Zhao

Near-infrared (NIR) mechanoluminescence (ML) holds significant promise for in vivo biomechanical detection and imaging due to its self-powered excitation and self-recovery capabilities. However, its broader application remains constrained by the limited availability of suitable ML materials and the narrow spectral range of NIR emission. In this study, we report the Y 3 Ga 5 O 12 (YGO): Cr 3+ , Nd 3+ , Yb 3+ /polydimethylsiloxane (PDMS) NIR-ML composite with broadband NIR emission tunable from 600 to 1100 nm, covering both the NIR-I and NIR-II windows. The ML of composite device is driven by an interfacial triboelectric effect arising from the mechanical interaction between the inorganic particles and the polymer matrix. Comparative studies on photon penetration through biological tissues reveal that co-doping with Nd 3+ and Yb 3+ markedly enhances tissue penetration depth. Specifically, the luminescence decay rate in biological tissues is reduced by 20.8% relative to Cr 3+ single doping, indicating improved optical persistence and signal transmission. These findings demonstrate the potential of full-spectrum NIR-ML devices for high-resolution, deep-tissue biomechanical sensing and imaging. This work provides a generalizable design strategy for the development of broadband NIR-ML materials tailored for in vivo mechanical diagnostics.

Oligopeptide modified ICG/Cisplatin@CaCO3 for targeted osteosarcoma NIR-II imaging guided multimodal therapy

Li-chen Ji,Jia-qing Huang,Xu-gang Zhong,Qi-hong Sun,Yong Fan,Li Yin,Qiong Zhang,Yu Cai,Zeju He

Osteosarcoma, as the most prevalent primary malignant bone tumor, has consistently shown suboptimal clinical treatment outcomes. In this manuscript, we proposed an efficacious and novel phototherapy strategy for osteosarcoma based on a photoresponsive near-infrared (NIR) material, CaCO 3 -ICG-DDP-PEG-PT (CIDP-PT), which contained calcium carbonate (CaCO 3 ) and Cisplatin (DDP) via the gas dispersion method, and indocyanine green (ICG) was encapsulated through a nanoprecipitation reaction by employing DSPE-PEG 2000 -COOH to enhance the stability and hydrophilicity. The targeting oligopeptide (PT) for osteosarcoma cells was inserted into the outer membrane of nanoparticles, thereby further enhancing the tumor-targeting capability through receptor-mediated binding. The prepared CIDP-PT nanoparticles exhibited high targeting efficiency and multifunctionality. In vitro and in vivo experiments demonstrated that CIDP-PT significantly enhanced the therapeutic efficacy for osteosarcoma via the NIR-II photoresponsive and tumor-targeting approach. It achieves efficient tumor cell eradication through multiple modalities while maintaining an exceptional level of biocompatibility. Hence, the CIDP-PT nanocarriers possess considerable practical value for future clinical applications in adjunct osteosarcoma phototherapy.

NIR-II photo- and sono-responsive hyaluronic acid-capped nanozymes for glioblastoma-targeting theranostics

Huanran Qu,Yuanyuan Zhang,Minghao Chen,Shiyang Shao,Jianqiang Chen,Yundi Wu,Xilong Wu

Glioblastoma is the most aggressive and heterogeneous astrocytic tumor, with its intracranial location limiting the efficacy of conventional therapies. This study presents the synthesis of a multifunctional nanocomplex utilizing borophene nanosheets (B NSs) as a substrate. Gold nanoparticles (Au NPs) are modified onto the surface of B NSs to create Schottky heterojunctions (B Au). This is followed by co-precipitation solvothermal synthesis incorporating silver sulfide (Ag 2 S) and hyaluronic acid (HA) as a capping agent, yielding B-Au-Ag 2 S-HA (BAA-HA). The Schottky heterojunction reduces the bandgap and accelerates charge carrier separation, significantly enhancing the sonodynamic therapy (SDT) efficiency of B NSs. In comparison to B NSs, BAA-HA exhibits significantly improved photothermal conversion efficiency under 1064 nm laser irradiation, facilitating the cascade catalysis of glucose oxidase-like (GOx-like) and catalase-like (CAT-like) enzymes. This accelerates glucose and H 2 O 2 decomposition, increasing O 2 supply to amplify SDT efficacy and induce immunogenic cell death (ICD), inducing a robust anti-tumor immune response. Ultrasound-targeted microbubble destruction technology is employed to transiently open the blood-brain barrier, allowing for targeted delivery of BAA-HA to glioblastoma cells via HA-mediated recognition of the CD44 receptor. Additionally, the NIR-II fluorescence properties of Ag 2 S enable precise tumor imaging, guiding multimodal synergistic therapy. This platform provides a promising strategy for treating deep-seated tumors, integrating therapeutic and diagnostic functions to enhance efficacy and precision.

NIR-II Photoacoustic-Cavitation Boosted Metabolic Reprogramming Regulation Toward the Localized TNBC Growth Inhibition

Leilei Zhang,Haoheng Bai,Weiqi Sun,Luqi Wang,Minyan Duan,Qianying Liu,Chao Zhou,Xueyang Fang,Tao Zhang

Triple-negative breast cancer (TNBC) treatment with traditional phototherapeutics generally suffers from short light penetration depth and limitations of the hypoxic tumor microenvironment. Developing effective therapeutic modalities tailored to TNBC’s features remains highly desirable yet challenging. Herein, we propose a near-infrared dual windows (NIR-II) photoacoustic (PA) cavitation-based strategy to amplify oxidative stress and induce metabolic disruption in TNBC cells, achieving effective tumor suppression. With a fabricated nanoagent (DTG), we integrated the oxygen-independent therapeutic effects of photomechanical damage, cavitation-promoted oxidative damage and glucose oxidase (GOx)-mediated glucose depletion. Specifically, under NIR-II pulsed laser (PL) irradiation, PA cavitation generates collapsing bubbles and shock waves, causing mechanical injury to tumor cells. Meanwhile, GOx depletes intratumoral glucose, blocking glycolysis, reducing adenosine triphosphate (ATP) production, and generating hydrogen peroxide (H2O2) that would be further converted into more toxic hydroxyl radicals (•OH) by the photocavitation to suppress the tumors effectively. In addition, the strong NIR-II PA signal produced from the DTG facilitates high-resolution deep-tissue imaging to provide precise therapeutic guidance. Thus, these findings establish the multimodal strategy as a potent candidate for TNBC management.
