L-arginine modified mesoporous bioactive glass with ROS scavenging and NO release for periodontitis treatment

Haiyan Yao,Emine Sumeyra Turali Emre,Yuan Fan,Jiaolong Wang,Feng Liu,Junchao Wei

Periodontitis is a chronic inflammatory disease characterized by progressive alveolar bone resorption, and excessive reactive oxygen species (ROS) is a key factor to disease progression. Therefore, scavenging ROS to alleviate inflammation and promote bone regeneration are promising strategies to treat periodontitis. In this study, L-arginine (L-Arg) was used to modify mesoporous bioactive glass (MBG), forming L-Arg modified MBG (MBG@L-Arg), which showed effective ROS-scavenging and NO release properties in cells, and realize the protection and restoration of cell's activity in ROS-rich microenvironment. Furthermore, MBG@L-Arg can induce macrophage polarization from M1 to M2 phenotype, and promote the osteogenic differentiation of MC3T3-E1 cells and human periodontal ligament stem cells (hPDLSCs). MBG@L-Arg also regulated anti-inflammatory and antioxidant systems by inhibiting the NF- κ B signaling pathway and activating the Nrf2 signaling pathway. Besides, NO-PKG signaling pathway was also activated, further promoting bone regeneration. The in vivo results demonstrated that MBG@L-Arg can efficiently inhibit inflammation-induced tissue destruction and promote osteogenesis regeneration. The quantitative bone loss in MBG@L-Arg group was 1.03 ± 0.05 mm, significantly lower than that of the periodontitis group (1.47 ± 0.13 mm), implying that MBG@L-Arg can work as multifunctional materials for periodontal tissue regeneration.

Targeted delivery of NO donor and ROS scavenger for synergistic treatment of rheumatoid arthritis

Rongrong Luo,Jingjing Su,Wenying Zhang,Wenguang Huang,Qianqian Zhou,Pengchao Sun,Yongxing Zhao

Rheumatoid arthritis (RA) is characterized by high level of reactive oxygen species (ROS) and proinflammatory cytokines, which facilitate the activation of the inflammatory signaling such as NF-κB pathway and exacerbate the development of inflammation. Herein, we designed a nanodrug by encapsulating the NO donor S-nitrosoglutathione (GSNO) into an emulsion and coating the surface with a polydopamine (PDA) layer to yield GSNO@PDA, which simultaneously scavenged the extra ROS and suppressed NF-κB signaling for potent RA treatment. To enhance the cellular uptake and NO generation efficiency, dextran sulfate (DS) and Cu 2+ were anchored on the surface of GSNO@PDA to obtain the final formulation GSNO@PDA@DS. Our results demonstrated that GSNO@PDA@DS were successfully prepared and the modification of DS effectively boosted the cellular uptake of GSNO@PDA@DS. Moreover, GSNO@PDA@DS lowered cellular ROS and elevated intracellular NO, resulting in a decrease of M1 phenotype, inhibition of NF-κB pathway and down-regulation of proinflammatory cytokine tumor necrosis factor-α (TNF-α). Further in vivo studies confirmed that GSNO@PDA@DS significantly relieved symptoms and bone erosion by regulating the microenvironment of RA, highlighting the potential of GSNO@PDA@DS for RA therapy through ROS scavenging and NO-mediated suppression of inflammatory signaling.

Construction of nano-biomimetic coatings capable of catalyzing NO-generation based on the lysozyme phase transition reaction to improve the biocompatibility of polyurethane

Changjiang Pan,Naiquan Yang,Jie Chen,Yan Ji,Linhong Deng,Qingping Xiong

As a versatile biomaterial, polyurethane (PU) finds extensive use in blood-contacting medical devices, including vascular grafts, central venous catheters, hemodialysis catheters, artificial heart valves, and blood oxygenators, etc. However, surface-induced thrombosis and difficulty in surface endothelialization after implantation are potential factors limiting their long-term use. In the present study, a lysozyme nanoparticle biomimetic coating (PTL) was first built on the PU surface based on the phase transition reaction of lysozyme. Subsequently, through sequential amidation reactions, heparin with anticoagulant performances and selenocystamine capable of catalyzing nitric oxide (NO) generation were immobilized onto the PTL surface, yielding a multifunctional nano-biomimetic coating with NO-generation and anticoagulant activities. The coating exhibited outstanding wettability, the ability to inhibit fibrinogen adsorption, and the capacity to catalyze NO generation from NO donors continuously. The as-prepared coating could reduce platelet adhesion, activation, and hemolysis, displaying superior anticoagulant performance. Meanwhile, the coating could promote endothelial cell (EC) proliferation and functional expression (vascular endothelial growth factor (VEGF) and NO) while significantly impeding smooth muscle cell (SMC) adhesion and proliferation, showing a good ability to facilitate endothelialization. Additionally, in the presence of NO donors, the coating could catalyze the NO liberation from donors continuously, further enhancing the blood compatibility and facilitating EC growth selectively. As a consequence, the strategy of this study can simultaneously impart PU with excellent hemocompatibility and rapid endothelialization, providing a new way for constructing anticoagulant and endothelium-promoting nano-biomimetic coatings on PU surfaces.

An angiotensin-converting enzyme inhibitory peptide LVLPGE exerts an antihypertensive effect and improves endothelial function via eNOS/NO/cGMP pathway

Zishan Dou,Yang Xu,Jingli Liu,Wei Chen

LVLPGE is an angiotensin-converting enzyme (ACE) inhibitory peptide extracted from broccoli protein hydrolysates. The antihypertensive mechanism of LVLPGE was explored in the present study. The results indicated that the IC 50 value of LVLPGE was 19.49 μM, and its binding to ACE occurred via a static fluorescence quenching involving the secondary structure changes of ACE. In addition, LVLPGE competitively bound to the active site of ACE through hydrogen bonding. Furthermore, LVLPGE markedly enhanced the levels of the vasodilator nitric oxide (NO) in the Ang I-induced HUVECs model, suppressed the expression of vasoconstrictor endothelin-1 (ET-1), and increased the phosphorylation of eNOS. Moreover, LVLPGE was found to enhance the production of cyclic guanosine monophosphate (cGMP), which leading to a subsequent decrease in intracellular calcium concentration, ultimately facilitating the relaxation of vascular smooth muscles and lowering the blood pressure. These results demonstrated that LVLPGE achieves the antihypertensive effects by inhibiting ACE activity and activating the eNOS/NO/cGMP signaling pathway. This study provides new insights into the antihypertensive mechanism of LVLPGE and contributes to the high-value utilization of broccoli resources.

The superior performance of Nb-modified Cu-Ce-Ti mixed oxides for the selective catalytic reduction of NO with NH3 at low temperature

Xiaoqiang Wang,Yue Liu,Qingji Ying,Weiyuan Yao,Zhongbiao Wu

In this paper, a series of Nb modified Cu-Ce-Ti mixed oxides (Nb y CCT) were synthesized using wetness impregnation method for the selective catalytic reduction of NO with ammonia (NH 3 -SCR). The activity tests indicated that Nb y CCT catalysts had shown superior SCR performances. Among which, Nb 0.05 CCT catalyst exhibited a NO conversion of more than 90% within a broad operation temperature window (180–360 °C) under the GHSV of 40,000 h −1 . After that, corresponding characterizations were performed to correlate the physical structures and surface properties to catalytic performances. These results revealed that the strong interactions among the active phases induced by Nb addition could not only enhance the redox property of the catalysts but also inhibit the over-oxidation of NH 3 , which would be beneficial to SCR performance within a wide temperature range and good reaction selectivity. Furthermore, DRIFTS results suggested that the doping of Nb could promote the formation of NO 2 , thereby enhancing the “fast” SCR reaction process (Langmuir-Hinshelwood reaction pathway). Moreover, the Nb 0.05 CCT catalyst showed a better sulfur tolerance compared to Nb free catalyst that over 85% NO conversion could be retained after 12 h SCR reaction in the presence of SO 2 and H 2 O.

Effect of preparation methods on the performance of CeO2/Al2O3 catalysts for selective catalytic reduction of NO with NH3

Rui-tang Guo,Yue Zhou,Wei-guo Pan,Jie-nan Hong,Wen-long Zhen,Qiang Jin,Cheng-gang Ding,Shi-yi Guo

CeO 2 /Al 2 O 3 catalysts prepared by three methods were investigated for selective reduction of NO with NH 3 . It was found that the catalyst prepared by the single step sol–gel method had the best SCR activity and SO 2 resistance performance. From the results of BET, XRD, TPD and TPR, it can be concluded that large surface area, strong interaction, highly dispersed nano-crystalline ceria , high NH 3 adsorption capacity and good redox ability might be the main reasons for the excellent performance of CeO 2 /Al 2 O 3 catalyst prepared by the single step sol–gel method.

A multifunctional platform with single-NIR-laser-triggered photothermal and NO release for synergistic therapy against multidrug-resistant Gram-negative bacteria and their biofilms

Zhao Baohua,Wang He,Dong Wenjing,Cheng Shaowen,Li Haisheng,Tan Jianglin,Zhou Junyi,He Weifeng,Li Lanlan,Zhang Jianxiang,Luo Gaoxing,Qian Wei

Background Infectious diseases caused by multidrug-resistant (MDR) bacteria, especially MDR Gram-negative strains, have become a global public health challenge. Multifunctional nanomaterials for controlling MDR bacterial infections via eradication of planktonic bacteria and their biofilms are of great interest. Results In this study, we developed a multifunctional platform (TG-NO-B) with single NIR laser-triggered PTT and NO release for synergistic therapy against MDR Gram-negative bacteria and their biofilms. When located at the infected sites, TG-NO-B was able to selectively bind to the surfaces of Gram-negative bacterial cells and their biofilm matrix through covalent coupling between the BA groups of TG-NO-B and the bacterial LPS units, which could greatly improve the antibacterial efficiency, and reduce side damages to ambient normal tissues. Upon single NIR laser irradiation, TG-NO-B could generate hyperthermia and simultaneously release NO, which would synergistically disrupt bacterial cell membrane, further cause leakage and damage of intracellular components, and finally induce bacteria death. On one hand, the combination of NO and PTT could largely improve the antibacterial efficiency. On the other hand, the bacterial cell membrane damage could improve the permeability and sensitivity to heat, decrease the photothermal temperature and avoid damages caused by high temperature. Moreover, TG-NO-B could be effectively utilized for synergistic therapy against the in vivo infections of MDR Gram-negative bacteria and their biofilms and accelerate wound healing as well as exhibit excellent biocompatibility both in vitro and in vivo. Conclusion sOur study demonstrates that TG-NO-B can be considered as a promising alternative for treating infections caused by MDR Gram-negative bacteria and their biofilms.

Development of novel quinoline-NO donor hybrids inducing human breast cancer cells apoptosis via inhibition of topoisomerase I

Guiying Wu,Hui Zhong,Ying Wang,Li Chen,Jianbo Sun

A number of NO-releasing quinoline derivatives have been designed and synthesized by introducing NO donor to quinoline carboxylic acid fragment. The anti-proliferation of all target compounds was evaluated against human cancer cell lines (HCT-116, MCF-7, and A549), MCF-7/ADR and normal cell (MCF-10A). Most compounds showed cytotoxic activity on cancer cells and drug-resistant cells with IC 50 values in the range of 0.62–5.51 μM. Importantly, these compounds showed low toxicity to normal cells (4.21–34.08 μM). Further mechanism studies showed that the most potent compound 9 could release high concentration of NO and inhibit the activity of topoisomerase I. In addition, 9 regulated apoptosis-related proteins, generated ROS and blocked MCF-7 cells in G2/M phase to induce cell apoptosis. Furthermore, the P-gp-mediated transport was also influenced by 9 . And 9 could significantly inhibit the growth of tumor in vivo without observable organ-related toxicities. Overall, as a novel NO-releasing quinoline derivative, 9 was worthy for further in-depth study.

Effect of morphology on the performance of MnOx catalysts for selective catalytic reduction of NO with NH3

Tian Gu,Chengzhi Wang,Denghui Chen,Mengmeng Han

Modulating the physicochemical properties of catalysts through the construction of special crystal structures or microscopic morphology provides new insights for developing high-performance low-temperature denitrification catalysts. This paper explores the effect of morphology structure on catalyst performance and identifies the dominant driving factors. The catalyst activity sequence was found to be MnO x spiny nanospheres (Mn-SNS) > MnO x nanowires (Mn-N) > MnO x nanospheres (Mn-NS) > MnO x nanoparticles (Mn-P). Among these, the Mn-SNS catalyst prepared by the hydrothermal method exhibited the best catalytic activity and the good resistance to water and sulfur. It achieved over 70% NO x conversion at 150 °C, and maintained 100% at 200–350 °C. In the presence of SO 2 and H 2 O, the NO x conversion was maintained at 87%. The spherical structure consisting of nanospikes provides the Mn-SNS catalysts with a large specific surface area, more (Mn 4+ +Mn 3+ ), abundant acid sites, and stronger reducibility, thus exhibiting excellent performance through the "fast-SCR" pathway. It also inhibits oxide crystallization and provides more sites for the adsorption activation of NO x and NH 3 . The nanowire structures can provide a large specific surface area and a high concentration of Mn 3+ and Mn 4+ for MnO x catalyst. Characterization analyses revealed that the specific surface area, the ratio of (Mn 4+ +Mn 3+ )/Mn, and the number of Lewis acid sites (L-NH 3 ) are the most important factors affecting the catalytic activity. This present empirical analysis provides new ideas for designing high-performance catalysts with a large specific surface area.

X-ray-triggered NO-released Bi–SNO nanoparticles: all-in-one nano-radiosensitizer with photothermal/gas therapy for enhanced radiotherapy

Fangmei Zhang,Shikai Liu,Na Zhang,Ye Kuang,Wenting Li,Shili Gai,Fei He,Arif Gulzar,Piaoping Yang

Hypoxia in tumor cells is regarded as the most crucial cause of clinical drug resistance and radio-resistance; thus, relieving hypoxia of tumor cells is the key to enhancing the efficacy of anticancer therapy. As a gas signal molecule of vasodilatation factors, nitric oxide (NO) can relieve the hypoxia status of tumor cells, thereby, enhancing the sensitivity of tumor cells to radiotherapy. However, considering complications of vascular activity, the level of NO required for radiotherapy sensitization cannot be obtained in vivo. In view of this, we design and fabricate a multifunctional bismuth-based nanotheranostic agent, which is functionalized with S-nitrosothiol and termed Bi–SNO NPs. X-rays break down the S–N bond and simultaneously trigger large amount of NO-releasing (over 60 μM). Moreover, the as-prepared Bi–SNO NPs not only possess the capability of absorbing and converting 808 nm NIR photons into heat for photothermal therapy, but also have the ability to increase X-ray absorption and CT imaging sensitivity. In addition, the collaborative radio-, photothermal-, and gas-therapy of Bi–SNO in vivo was further investigated and remarkable synergistic tumor inhibition was realized. Finally, no obvious toxicity of Bi–SNO NPs was observed in the treated mice within 14 days. Therefore, the Bi–SNO developed in this work is an effective nano-agent for cancer theranostics with well-controlled morphology and uniform size (36 nm), which could serve as a versatile CT imaging-guided combined radio-, photothermal- and gas-therapy nanocomposite with negligible side effects.

Butterfly-shaped dendrimers with photosensitizer recruitment function for enhanced light-controlled precise cascaded ROS and NO release in multimodal diabetic foot ulcers infections treatment

Xiangyu Huang,Zucheng Shen,Qi Deng,Chengfei Zhao,Xiaofeng Cai,Shaoteng Huang,Mingbo Zhang,Yanzhuo Lv,Dongliang Yang,Junyang Zhuang,Ning Li

Diabetic foot ulcers (DFU) infections have been a major setback in wound healing, yet there is no effective therapeutic intervention. Here we report a nano-multidrug co-delivery system based on arginine-terminal butterfly-shaped peptide dendrimers (C&D@Z) that effectively kill bacteria and promote wound healing. Firstly, upon the near-infrared light irradiation, reactive oxygen species (ROS) was generated from the loaded photosensitizers, and then mediated bacteria killing. Next, the excessive ROS reacted with the designed butterfly-shaped dendrimers to generate sustained and abundant nitric oxide (NO), which exerted bactericidal action, as well as produced anti-inflammatory and angiogenesis effects. The co-delivery system, facilitated by the generation of NO, penetrated deeply into the biofilm, thereby effectively eliminating bacteria. The enhanced bactericidal and healing efficiency was investigated through biofilm penetration and wound healing experiments. Collectively, our system leveraged the highly-branched and cavity structures of the butterfly peptide dendrimer to realize the tight contact between the arginine groups and photosensitizer to achieve efficient conversion from ROS to NO, and the C&D@Z exhibited superior NO production, optimal photodynamic therapy utilization and effective penetration performance through cascade reaction between photodynamic therapy and NO therapy, may serve as a promising therapeutic approach for DFU infections.

Piezocatalytic Pt@BMO-Gel Dressing for In Situ Generation of H2 and NO to Treat Infectious Inflammation Related to Incontinence

Jiawen Wang,Jinfu Wang,Zi Wang,Jiayue Wu,Jiaxuan Liao,Shenyuan Zhan,Xiaoye Chen,Xinhao Wang,Yongbao Wei,Qidong Zhou,Liefu Ye,Jinfeng Wu

Incontinence-associated dermatitis (IAD) leads to persistent tissue damage, antibiotic-resistant infections, and neurogenic inflammation, greatly impairing patients' quality of life. Current therapies fail to simultaneously eliminate resistant bacterial biofilms, neutralize alkaline conditions, and break the cycle of ammonia (NH 3 ) regeneration. Here, a piezoelectric gas therapy strategy based on platinum-decorated bismuth molybdate load in gelatin methacrylate (GelMA) hydrogel (Pt@BMO-Gel) is introduced. Under ultrasound excitation, Pt@BMO generates high piezoelectric potential to improve charge separation, selectively adsorbs NH 3 on BMO (001) crystal facets, efficiently degrades NH 3 to normalize pH, and enables sustained release of H 2 and NO for dual antibacterial and anti-inflammatory effects. Mechanistic insights from theoretical calculations show that the Pt (111) and BMO (001) surfaces lower energy barriers for H 2 formation and N─H bond cleavage, conducive to the generation of NO and H 2 . Transcriptomic profiling reveals downregulation of key nitrogen metabolism genes (arcC/hutH) in methicillin-resistant Staphylococcus aureus (MRSA) and disruption of the (p)ppGpp-mediated stringent response, effectively eradicating bacterial persistence via metabolic reprogramming. This study establishes Pt@BMO as an efficient piezoelectric catalyst for wound therapy, presenting a novel approach in medicine that transforms harmful NH 3 into therapeutic H 2 and NO, offering a promising treatment paradigm for infected wounds, such as incontinence-associated dermatitis.
