A sensitive and selective fluorescence sensor for rutin was successfully developed. The sensor was fabricated from highly fluorescent nitrogen-boron co-doped carbon quantum dots (N,B-CDs) coated with mesoporous silica molecularly imprinted polymer (MIP). Rutin was detected by this sensor (N,B-CDs @ MIP) with a linear range of 0.08–30 μM and a detection limit as low as 0.05 μM, under optimal conditions. Meanwhile, the usefulness of the developed sensor was evaluated in samples of lake water, orange, and honey with satisfactory recoveries. Graphical

Oxidative stress is a major cause of ovarian aging and follicular atresia. There is growing evidence that showed potential roles of rutin in antidiabetic, anti-inflammatory, antitumor, antibacterial and antioxidant, although it is yet unclear what the underlying mechanism is. Here, we looked into the potential effects of rutin on oxidative stress in the prehierarchical small white follicles (SWFs) from 580-day-old (D580) laying chickens. According to the findings, aging D580 layer ferroptosis was much higher than it was for laying hens during the peak period (280-day-old, D280). In both naturally aged and d -gal-induced chicken SWFs, rutin treatment concurrently boosted cell proliferation and prevented apoptosis. In addition, rutin inhibited the increased ferroptosis in aging hens. Meanwhile, rutin markedly suppressed the elevated ferroptosis and descending antioxidant capacity of D280-culture-SWFs from chicken elicited by d -gal. Rutin's activation of the Nrf2/HO-1 pathway hastened the SWFs' verbal battle with oxidative damage and reduced ferroptosis. Furthermore, activation of the ferroptosis signal increased the oxidative damage in SWFs. In conclusion, rutin alleviated oxidative stress that was induced by ferroptosis in aging chicken SWFs through Nrf2/HO-1 pathway. These findings point to a novel mechanism by which rutin protects SWFs from oxidative stress by suppressing ferroptosis, which is presumably a fresh approach to slowing ovarian aging in laying hens.

The aim of this study was to establish a rapid detection method of rutin in food based on nitrogen-doped carbon quantum dots (N-CDs) as the fluorescent probe. N-CDs were prepared via a single-step hydrothermal process using citric acid as the carbon source and thiourea as the nitrogen source. The optical properties of N-CDs were characterized using an electron transmission microscope, X-ray diffractometer, Fourier-transform infrared spectrometer, and nanoparticle size potential analyzer. The UV/Vis absorption property and fluorescence intensity of N-CDs were also characterized using the respective spectroscopy techniques. On this basis, the optimal conditions for the detection of rutin by N-CDs fluorescent probes were also explored. The synthesized N-CDs were amorphous carbon structures with good water solubility and optical properties, and the quantum yield was 24.1%. In phosphate-buffered solution at pH = 7.0, Rutin had a strong fluorescence-quenching effect on N-CDs, and the method showed good linearity (R2= 0.9996) when the concentration of Rutin was in the range of 0.1–400 μg/mL, with a detection limit of 0.033 μg/mL. The spiked recoveries in black buckwheat tea and wolfberry were in the range of 93.98–104.92%, the relative standard deviations (RSD) were in the range of 0.35–4.11%. The proposed method is simple, rapid, and sensitive, and it can be used for the rapid determination of rutin in food.Keywords:nitrogen-doped carbon quantum dots;hydrothermal synthesis;fluorescence probe;rutin

This study aims to evaluate the heterocyclic aromatic amine (HAA) formation inhibited by cyanidin and rutin in chemical modeling systems and smoked chicken drumsticks. In the PhIP and β-carboline chemical modeling systems, cyanidin inhibited the PhIP, Harman, and Norharman formation while rutin inhibited the PhIP formation but promoted the Harman and Norharman formation compared with control sample ( P < 0.05). A mechanistic investigation confirmed that inhibiting the PhIP formation by cyanidin was mainly through trapping phenylalanine, creatine, creatinine, glucose, phenylacetaldehyde, and an aldol condensation product. In the smoked chicken drumsticks, cyanidin inhibited the PhIP, Harman, and Norharman formation dose-dependently compared to the control sample ( P < 0.05), with 2.0 % (w/v) cyanidin having the highest inhibitory effect. Moreover, cyanidin inhibited the formation of PhIP precursors and intermediates in the smoked chicken drumsticks. These results provide evidence for using phenolic compounds to reduce HAA formation in smoked meat products.

A [BMIM]PF 6 ion liquid (IL)-assisted synthesis of a rutin imprinted monolith (RIM) was carried out in an in-situ polymerization method. Bi-functional monomers and a ternary porogen containing IL was used for the RIM preparation and a L 9 (3 3 ) orthogonal factor design performed. Scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FT-IR) and N 2 adsorption method was for structural characterization of the RIMs. The monolith was directly used as stationary phase in liquid chromatography to test the retention selectivity, adsorption capacity and extraction application. The optimized porogen consists of 900 μL DMF, 144 μL ACN and 216 μL IL. The monolith RIM-13 obtained under the optimized conditions possessed improved adsorption performance, with a dynamic adsorption capacity of 6.695 mg/g, an imprinting efficiency of 4.841 and a selectivity α value of 4.821. Additionally, this monolith had also higher specific surface area , pore volume and permeability than that obtained without IL and the homogeneity of the imprint sites could be improved by using IL. When the RIM-13 was applied to the separation and purification of rutin from tartary buckwheat, a rutin product with a purity higher than 92 % can be obtained by one cycle. This molecular imprint solid-phase extraction (MI-SPE) is of potency to be applied to preparative-scale separation of other natural products.

Oxidative damage of neurons is one of the key pathological markers of Alzheimer’s disease (AD), which eventually leads to neuronal apoptosis and loss. Nuclear factor E2-related factor 2 (Nrf2) is a key regulator of antioxidant response and is considered to be an important therapeutic target for neurodegenerative diseases. In this study, the selenated derivative of antioxidant rutin (Se-Rutin) was synthesized with sodium selenate (Na2SeO3) as raw material by a simple electrostatic-compound in situ selenium reduction method. The effects of Se-Rutin on H2O2 induced oxidative damage in Pheochromocytoma PC12 cells were evaluated by cell viability, apoptosis, reactive oxygen species level and the expression of antioxidant response element (Nrf2). The results showed that H2O2 treatment significantly increased the level of apoptosis and reactive oxygen species, while the level of Nrf2 and HO-1 decreased. However, Se-Rutin significantly reduced H2O2 induced apoptosis and cytotoxicity, and increased the expression of Nrf2 and HO-1, both of which were better than that of pure rutin. Therefore, the activation of Nrf2/HO-1 signaling pathway may be the basis of Se-Rutin’s anti-oxidative damage to AD.

Rutin, as a biological flavonoid glycoside, has very important medicinal value. The accurate and rapid detection of rutin is of great significance. Herein, an ultrasensitive electrochemical rutin sensor based on β-cyclodextrin metal–organic framework/reduced graphene oxide (β-CD-Ni-MOF-74/rGO) was constructed. The obtained β-CD-Ni-MOF-74 was characterized by X-ray diffraction spectroscopy (XRD), X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), and nitrogen adsorption and desorption. The β-CD-Ni-MOF-74/rGO presented good electrochemical properties benefiting from the large specific surface area and good adsorption enrichment effect of β-CD-Ni-MOF-74 and the good conductivity of rGO. Under optimal conditions for the detection of rutin, the β-CD-Ni-MOF-74/rGO/GCE showed a wider linear range (0.06–1.0 μM) and lower detection limit (LOD, 0.68 nM, (S/N = 3)). Furthermore, the sensor shows good accuracy and stability for the detection of rutin in actual samples. Keywords:electroanalysis;carbon materials;electrochemical reduction;modified electrode

Heat stress (HS) leads to abnormal lipid metabolism in broilers, while rutin as a natural flavonoid with multiple biological functions shows potential to regulate antioxidant capacity and lipid metabolism. Therefore, this study aimed to investigate the effects of rutin supplementation on antioxidant and lipid metabolism functions in heat-stress broilers and to further explore its potential mechanisms. A total of 192 male broilers were randomly divided into three groups and treated with normal control (CON), heat stress (34 °C, HS), and HS with 500 mg kg -1 rutin supplementation (HS + Rutin), respectively. The results showed that dietary supplementation of 500 mg kg -1 rutin could significantly reverse the HS-induced liver damage. Rutin also mitigated HS-induced increases in liver triglyceride content and decreases in high-density lipoprotein and fatty acid binding protein contents. This might be achieved by increasing the expression of PPARα and decreasing the expression of SREBP1, which regulated the expression of downstream factors (such as CPT1, ACOX1, and ACC). Compared with the HS group, rutin increased mitochondrial DNA copy number and maintained mitochondrial homeostasis by regulating the expression of related genes (such as AMPK, PGC1-α, and SIRT3). In addition, rutin increased the activity of antioxidant enzymes, decreased the content of malondialdehyde, and activated the nuclear respiratory factor 2 (Nrf2) signaling pathway in the liver, which alleviated HS-induced oxidative stress. The effect of rutin in alleviating abnormal lipid deposition in HS broiler livers were correlated with indicators that reflected the antioxidant capacity and mitochondrial homeostasis. In conclusion, dietary 500 mg kg -1 rutin alleviates heat stress-induced hepatic abnormal lipid metabolism of broilers via improving antioxidant capacity to maintain mitochondrial homeostasis.

Intracerebral hemorrhage (ICH) presents a formidable challenge due to its high mortality and disability rates, primarily attributed to cerebral hematoma formation and ensuing neuroinflammation. Swift hematoma removal is paramount for prognosis, yet existing interventions carry risks and limitations. Notably, elevated CD47 expression on hematoma-associated RBC triggers a “don't eat me” signal, impeding hematoma clearance, while microglial/macrophage erythrophagocytosis exacerbates oxidative stress and the RBC lysate evokes neuroinflammation. To address this conundrum, a multifunctional nanomedicine (TD-CFR), employing DNA tetrahedra (TD) as a carrier for ICH treatment is introduced. The investigations reveal that CpG enhances the phagocytosis of CD47-expressing RBC by microglia/macrophages via lipid metabolism modulation. Integration of CpG into TD preserves its pro-phagocytic efficacy, while TD's double-stranded region enables efficient encapsulation of Rutin, a potent anti-inflammatory and antioxidant flavonoid. Capitalizing on disrupted blood-brain barrier integrity at the hemorrhage site, TD-CFR achieves robust enrichment within cerebral hematoma post-intravenous administration, augmented by folate receptor-mediated targeting of microglia/macrophages. Efficacy assessments in mouse and rabbit ICH models confirm TD-CFR's therapeutic benefits, including hematoma clearance, neuroinflammation suppression, and brain function restoration. Leveraging TD's high biosafety profile and dual active ingredient loading capacity, the study unveils a promising drug treatment paradigm for ICH.

A simple and rapid electrochemical method based on a composite of cadmium telluride quantum dots (CdTe QDs) and MXene is developed for the simultaneous determination of rutin and quercetin in food samples. The CdTe QD-MXene composite is synthesized via the in situ growth of CdTe QDs on MXene, which serves as a carrier and enhances electrical conductivity. Incorporating CdTe QDs into MXene interlayers effectively prevents agglomeration in MXene and provides more active sites for electrochemical determination. The developed electrochemical method can successfully determine rutin and quercetin, both individually and simultaneously, in aqueous solutions while achieving high stability and selectivity. Notably, the prepared sensor exhibits limits of detection of 3.300 × 10 −8 and 3.268 × 10 −7 M for the simultaneous determination of rutin and quercetin, respectively. Moreover, the sensing platform is used for the determination of rutin in buckwheat, locust rice, and apples, with results well comparable to those obtained using ultraviolet spectroscopy. Finally, the proposed sensor is employed to monitor the hydrolysis of rutin into quercetin in buckwheat using an electrochemical method for the first time. This study provides new ideas for the application of electrochemical sensors in food and drug science. Graphical

α-Monoglucosyl rutin (α-GR) is a structurally optimized rutin derivative, and its higher water solubility and bioavailability may help it exert better pharmacological effects in vivo . The present study examined the protective effects and potential mechanisms of α-GR on cyclophosphamide (CTX)-induced colonic injury in mice. The findings showed that α-GR significantly reduced serum levels of lipopolysaccharide (LPS) and diamine oxidase (DAO), effectively alleviating the histopathological damage in colon tissues caused by CTX exposure. Additionally, α-GR maintained the integrity of intestinal epithelial cells by promoting the secretion of secretory immunoglobulin A (sIgA) and upregulating the expression of tight junction proteins (Claudin-1 and ZO-1), while also reducing the levels of colonic pro-inflammatory factors and oxidative stress. In terms of gut microbiota, 16S rRNA gene sequencing and metabolomic analyses revealed that α-GR could effectively reverse the gut metabolic disorders and microbiota structural changes induced by CTX, promoted the growth of beneficial bacteria like norank_f_Muribaculaceae , Bacteroides, and Alloprevotella , and reduced the relative abundance of pathogenic bacteria ( Desulfovibrio ), while also revealing the complex correlation between them. In terms of mechanism research, α-GR further attenuated intestinal damage by regulating the TLR4/NF-κB and Nrf2/HO-1 pathways. This provides new ideas for the development of functional dietary supplements to treat CTX-associated intestinal toxicity.

A high‐level fava beans diet has been proven to enhance the texture characteristics of tilapia muscle, yet it can also induce hepatointestinal injury. Rutin, as a nutritional additive, has antioxidant and immune‐boosting effects. This study explored the regulatory effect of 150 and 300 mg/kg rutin on the muscle characteristics and hepatointestinal health of tilapia after being fed with a 60% fava beans diet. Two hundred and forty tilapias (average weight: 371.50 ± 9.29 g) were evenly divided into four groups and fed with the diet of groups C0 (0% fava bean), R0 (60% fava bean), R1 (60% fava bean + 150 mg/kg rutin), and R2 (60% fava bean + 300 mg/kg rutin) for 10 weeks, respectively. The results indicated that the growth performance in R1 and R2 showed no significant changes compared to R0, while muscle hardness, gumminess, and resilience increased, along with an elevation in muscle crude protein deposition. The addition of 150 and 300 mg/kg rutin enhanced the antioxidant capacity (AOC) of the muscles, liver, and intestines, alleviated liver damage by regulating liver lipid metabolism compared with R0. Dietary supplementation with 150 mg/kg rutin improved the composition of intestinal microbiota, significantly upregulating the abundance of beneficial bacteria Muribaculaceae unclassified and reduced the abundance of harmful bacteria Rhodobacteraceae , Chloroplast_unclassified , and Rothia . In summary, rutin can effectively function as a nutritional supplement to alleviate liver and intestinal damage caused by 60% fava bean feed, while also improving muscle texture simultaneously. This study provides a crucial theoretical basis for optimizing crispy tilapia aquaculture through rutin supplementation, promoting innovation in aquatic feed formulations.
