Accurate and rapid detection of the causative agent of a disease is of great importance in controlling the spread of the disease. This work developed a biosensor with the Bi2Te3 family of topological insulators for detection of the SARS-CoV-2 virulence factor. The Bi2Te3 family is a three-dimensional topological insulator material with topologically protected surface states; the presence of these surface states facilitates charge transfer between the electrode and electrolyte interface. Compared with the detection performance of Bi2Se3, BiSbTeSe2, and a trivial insulator like Sb2Se3, Bi2Te3 exhibits superior characteristics. A Bi2Te3 electrochemical detection platform is utilized to fabricate a sensor that can detect SARS-CoV-2 DNA, RNA, and antigen for label-free target detection. The concentration range of DNA detection by the biosensor using Bi2Te3 is between 1.0 × 10–15 and 1.0 × 10–10 M, and the detection limit can reach 1.41 × 10–16 M. Furthermore, it exhibits excellent selectivity and maintains good stability even after being stored for 14 days. This study provides a new way to apply topological insulator materials in the field of biosensors and use their unique electronic structure to improve the accuracy and speed of disease detection and diagnosis.

Given the susceptibility of Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV-2) and Norovirus (NoV) to survive in cold chain food, thereby posing significant public health risks, we present a novel approach for biosensor development utilizing a graphene field-effect transistor (GFET) modified with nucleic acid aptamers. The biosensor’s innovative design incorporates 1-pyrenebutyric acid N-hydroxysuccinimide ester (PBASE) as a coupling agent to facilitate the attachment of nucleic acid aptamers onto channel graphene. This modification induces a redistribution of charge on the graphene surface, resulting in a shift of the Dirac point upon target capture by the nucleic acid aptamer. Through this pioneering methodology, we successfully engineered SARS-CoV-2 GFET and NoV GFET biosensors capable of detecting trace amounts of SARS-CoV-2 and norovirus within a rapid 5-minute timeframe, showcasing detection limits of 33 fg/mL and 6.17 pg/mL, respectively. Subsequently, we applied these sensors to detect SARS-CoV-2 in frozen meat and norovirus in shellfish, yielding promising results with excellent specificity and stability. This groundbreaking sensing mechanism holds significant promise for the detection of foodborne viruses across a diverse range of food samples.

The coronavirus disease-2019 pandemic reflects the underdevelopment of point-of-care diagnostic technology. Nuclei acid (NA) detection is the “gold standard” method for the early diagnosis of the B.1.1.529 (Omicron) variant of severe acute respiratory syndrome-coronavirus disease-2. Polymerase chain reaction is the main method for NA detection but requires considerable manpower and sample processing taking ≥ 3 h. To simplify the operation processes and reduce the detection time, exonuclease III (Exo III)-aided MoS 2 /AIE nanoprobes were developed for rapid and sensitive detection of the oligonucleotides of Omicron. Molybdenum disulfide (MoS 2 ) nanosheets with excellent optical absorbance and distinguishable affinity to single-strand and duplex DNAs were applied as quenchers, and aggregation-induced emission (AIE) molecules with high luminous efficiency were designed as donor in fluorescence resonance energy transfer-based nanoprobes. Exo III with catalytic capability was used for signal amplification to increase the sensitivity of detection. The composite nanoprobes detected the mutated nucleocapsid (N)-gene and spike (S)-gene oligonucleotides of Omicron within 40 min with a limit of detection of 4.7 pM, and showed great potential for application in community medicine.

The capping of mRNA and the proofreading play essential roles in SARS-CoV-2 replication and transcription. Here, we present the cryo-EM structure of the SARS-CoV-2 replication-transcription complex (RTC) in a form identified as Cap(0)-RTC, which couples a co-transcriptional capping complex (CCC) composed of nsp12 NiRAN, nsp9, the bifunctional nsp14 possessing an N-terminal exoribonuclease (ExoN) and a C-terminal N7-methyltransferase (N7-MTase), and nsp10 as a cofactor of nsp14. Nsp9 and nsp12 NiRAN recruit nsp10/nsp14 into the Cap(0)-RTC, forming the N7-CCC to yield cap(0) ( 7Me GpppA) at 5′ end of pre-mRNA. A dimeric form of Cap(0)-RTC observed by cryo-EM suggests an in trans backtracking mechanism for nsp14 ExoN to facilitate proofreading of the RNA in concert with polymerase nsp12. These results not only provide a structural basis for understanding co-transcriptional modification of SARS-CoV-2 mRNA but also shed light on how replication fidelity in SARS-CoV-2 is maintained.

Timely and accurate detection of the virus is of great significance to prevent the virus’s harm and control the epidemic. Here, an aptasensor based on the principle of promoting hybridization through aptamer conformational change was designed to quantitatively detect the spike (S) protein of SARS-CoV-2. When the S protein binds to the 3' end of the aptamer, the 5' end of the aptamer tansforms into a straight hybridization region, which will greatly facilitate the hybridization with complementary DNA (cDNA). In the absence of S protein, hybridization is less likely to occur due to the complex G-quadruplex structure of aptamer. According to this principle, cDNA is modified onto magnetic beads (MBs) or onto the optical fiber probe of an evanescent wave fluorescence aptasensor (EWFA) detection platform to capture the fluorescently labeled aptamer-S protein conjugate, two kinds of quantitative detection methods for SARS-CoV-2 S protein were established. In particular, simple, rapid and sensitive detection could be obtained based on the EWFA detection platform, in which the whole detection procedure including the measurement and regeneration takes only 14 min, the LOD is 5.34 ng/mL, the linear response range is 141.49 to 9507.36 ng/mL, and the optical fiber probe could be reused for 19 times. The EWFA detection platform is also potentially applicable to detect other protein biomarkers only by replacing the specifically modified optical fiber probes. Graphical

The outbreak of various epidemics has caused a significant impact on public health. Developing highly efficient techniques for the early detection of pathogenic viruses, e.g., Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), is the key to fast screening and diagnosis of patients. Here, a novel detection method with high sensitivity is developed to detect viral nucleic acids by employing nanoscale porphyrinic metal-organic frameworks (NPMOFs) as a photodynamic probe and magnetite nanoparticles as a trapping agent. Benefiting from the high content of porphyrin molecules as the organic linker with improved performance, NPMOFs exhibit the excellent photodynamic effect, which effectively transport energy to O 2 to produce highly active singlet oxygen ( 1 O 2 ) upon light activation. The presence of target nucleic acids allows the capture of NPMOFs probes with magnetite nanoparticles via gene complementary hybridization, resulting in the fast production of remarkable 1 O 2 based on photodynamic effect, which is utilized to decompose the indicator for signal readout. Results indicate that such method enables the detection of SARS-CoV-2 related RNA in the simulated swabs with the limit of detection (LOD) of 4.94 fM within 30 min, presenting giant potential for simple, sensitive and rapid sensing of viral nucleic acids without RNA amplification treatment. This strategy inaugurates a new direction of nanomaterials for signal transduction in biosensing application.

Strategies for the rapid detection of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) are critically needed due to conventional methods’ limitations: narrow range, virus mutation-induced failure, time-consuming, and complex operations. Herein, we propose a method utilizing CuInP2S6 nanosheet probes and Bio-Layer Interferometry (BLI) technology for the rapid (5–10 min), noninvasive, and broad-spectrum detection of the SARS-CoV-2 spike receptor binding domain (RBD) in human saliva. The nanoprobe exhibits a higher binding affinity to the RBD compared to most saliva proteins, allowing it to be immobilized on BLI sensors for easier detection of protein binding and elution. An eluent buffer containing Tween-20 and salts was employed to separate salivary proteins while retaining the RBD on the probes. This system can detect the RBD across a broad spectrum and a low limit of 25 ng/mL (S/N = 3) in less than 10 min. To validate this system, experiments with pseudoviruses showed accurate identification and binding of the RBD. Molecular dynamics simulations elucidated the mechanism for selective binding of probes to RBD. In conclusion, we propose a conceptual study based on an in situ strategy with inorganic nanoprobes alongside BLI techniques for convenient, noninvasive, and rapid detection of SARS-CoV-2 and its variants. This strategy is anticipated to inspire the design and implementation of nanoprobes for the rapid and selective detection of pathogens in the future.

The coronavirus disease 2019 (COVID-19) caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has resulted in widespread disease transmission, challenging the stability of global healthcare systems. Surface-enhanced Raman scattering (SERS) as an easy operation, fast, and low-cost technology illustrates a good potential in detecting SARS-CoV-2. In the study, one-step fabrication of gold-silver alloy nanoparticles (AuAgNPs) with adjustable metal proportions and diameters is employed as SERS substrates. The angiotensin-converting enzyme 2 (ACE2) functionalized AuAgNPs are applied as sensor surfaces to detect SARS-CoV-2 S protein. By optimizing the SERS substrates, ACE2/Au 35 Ag 65 NPs illustrate higher performance in detecting the SARS-CoV-2 S protein with a limit of detection (LOD) of 10 fg/mL in both phosphate-buffered saline (PBS) and pharyngeal swabs solution (PSS). It also provides excellent reproducibility with a relative standard deviation (RSD) of 7.7 % and 7.9 %, respectively. This easily preparable and highly reproducible SERS substrate has good potential in the practical application of detecting SARS-CoV-2.

Home testing technology strategy is critical for early screening of disease. However, current home testing technologies often require complex processes, which limits their application. In this study, a time-resolved cascade logic gate microfluidic chip (TCLMC) was revealed to enable capillary force-based one-step operation without manual intervention or professional equipment. By analogy with logic gates in the circuit, TCLMC could automatically control the fluid flow and regulate the incubation time to optimize the immunoassay . The limit of detection of TCLMC for SARS-CoV-2 and influenza B virus (Flu B) was 134.94 and 79.17 pg mL −1 within 10 min. Additionally, this study tested saliva samples from 12 Flu B patients and 24 healthy controls to verify its clinical application. The results showed that TCLMC had high sensitivity (100%), specificity (100%), and accuracy (100%). This study provides a new one-step strategy for home testing and demonstrates its great potential in the diagnosis field.

The outbreak of the pandemic caused by the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) calls for an urgent unmet need for developing a facial and cost-effective detection method. The requirement of well-trained personnel and sophisticated instrument of current primary mean (reverse transcription polymerase chain reaction, RT-PCR) may hinder the practical application worldwide. In this regard, a reverse transcription recombinase polymerase amplification (RT-RPA) coupled with CRISPR-Cas12a colorimetric assay is proposed for the SARS-CoV-2 detection. The methodology we have described herein utilizes DNA-modified gold nanoparticles (AuNPs) as a universal colorimetric readout and can specifically target ORF1ab and N regions of the SARS-CoV-2 genome. After the virus genome is amplified through RT-RPA, the resulting abundant dsDNA will bind and activate Cas12a. Under trans-cleavage degradation, the capped DNA substrate will be hydrolyzed gradually from AuNPs, demonstrating a change in the surface plasmon resonance (SPR), which can be facially monitored by UV–vis absorbance spectroscopy and naked eye observation. The high amplification efficiency from RT-RPA and Cas12a trans-cleavage process bring the sensitivity of our method to 1 copy of viral genome sequence per test. Notably, under the dual variations inspecting from the isothermal amplification and Cas12a activation process, the false positive events from other beta coronavirus members can be effectively avoided and thus significantly improve the specificity. Furthermore, the reliability of this colorimetric assay is validated by standard clinical samples from the hospital laboratory department. Through integration of the inherently high sensitivity and specificity from an RPA-coupled Cas12a system with the intrinsic simplicity of AuNP-based colorimetric assay, our method increases the practical testing availability of SARS-CoV-2.

The frequent emergence of variants of concern (VOC) of SARS-CoV-2 necessitates a sensitive and all-inclusive detection platform that remains viable despite the virus mutations. In this context, we targeted the receptor-binding domain (RBD) of glycoprotein (S-protein) of all VOC and constructed a consensus RBD (cRBD) based on the conserved amino acids. Then, we selected a high-affinity ssDNA novel aptamer specific for the cRBD by an in silico approach. The selected aptamer is utilized to fabricate a photonic crystal (PC)-decorated aptasensor (APC-sensor), which consists of polystyrene nanoparticles polymerized within a polyacrylamide hydrogel. cRBD-responsive ssDNA aptamers are crosslinked in the hydrogel network, which selectively bind to the cRBD and SARS-CoV-2 in saliva samples. The binding response can be visually monitored by swelling of the hydrogel and color generation by diffraction of light from PCs and can be quantified by the diffraction ring diameter or a spectrometer. The sensor delivers a LOD of 12.7 ± 0.55 ng mL–1 for the cRBD and 3 ± 18.8 cells mL–1 for SARS-CoV-2 in saliva samples, with a rapid response of 5 min. The sensor can be stored and regenerated without loss of activity. It can be utilized as a point-of-care testing (POCT) for SARS-CoV-2 diagnosis.

The COVID-19 pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has significantly impacted public health and social stability, highlighting the urgent need for rapid diagnostic methods to ensure timely clinical intervention. Herein, we develop a sensitive surface-enhanced Raman scattering (SERS) immunosensor based on a silver nanotripod (AgNT) array sandwich structure for simulating early SARS-CoV-2 detection. AgNT arrays modified with SARS-CoV-2 antibody serve as a capture substrate; antibody-modified gold nanoparticles (AuNPs) functionalized with dye molecules act as detection probes. Upon binding to analyte, these components form an AuNPs@AgNT sandwich immunosensor generating enhanced SERS signals for detection. The detection limit of AuNPs@AgNT for SARS-CoV-2 spike protein in phosphate-buffered saline (PBS) samples and saliva samples could reach 0.1 and 0.77 pg mL −1 , respectively. This sandwich immunosensor demonstrates excellent reproducibility with a relative standard deviation lower than 8% and maintains satisfactory detection specificity even in various saliva samples and other proteins. Furthermore, by integrating machine learning technology, the sandwich immunosensor enables accurate discrimination of multi-target samples. These results illustrate the promise of the SERS sandwich immunosensor for the early SARS-CoV-2 spike protein detection, which has a remarkable significance for the virus detection in the future. Graphical The front cover shows a surface-enhanced Raman scattering (SERS) immunosensor based on Ag nanotripod (AgNT) arrays. Au nanoparticles (AuNPs) are loaded with reporters (4-MBA, DTNB, TFMBA) and antibodies (Anti). The AgNT substrate is modified with 3-MPA and Anti. The immunosensor combined with machine learning enables detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein and distinction of tumor necrosis factor-α (TNF-α) and interleukin-1 (IL-1) in mixed samples.
