Kanamycin (KAN) residues in food pose a serious risk to human health, necessitating the development of rapid and sensitive detection methods. Herein, a novel surface-enhanced Raman scattering (SERS) aptasensor for ultrasensitive detection of kanamycin, which integrates hollow Au/Ag alloy nanourchins (HAAA-NUs) tags with high-density “hot spots” and exonuclease III (Exo III)-assisted cyclic amplification. In the presence of KAN, its specific binding with the KAN aptamer (KAN-Apt) triggers the release of complementary DNA (cDNA), followed by hybridization with hairpin DNA (HAP) and Exo III-mediated digestion, resulting in cyclic amplification of target DNA. The amplified target DNA serves as a bridging mediator that specifically hybridizes with the DNA1 immobilized on Fe 3 O 4 @Au nanoparticles (NPs) and the DNA2 anchored on PATP-loaded HAAA-NUs, leading to the formation of a Fe 3 O 4 @Au NPs-DNA1/target DNA/DNA2-HAAA-NUsPATP complex, which subsequently produces a strong SERS signal. In addition, the background signal can be effectively reduced through magnetic separation. Under optimal conditions, the SERS aptasensor exhibited a wide detection range for KAN from 1 to 10 5 fg/mL with a good linear correlation (R 2 = 0.9913), and a limit of detection (LOD) as low as 0.12 fg/mL. It also performed satisfactorily in the analysis of real samples (milk and honey), achieving good recoveries ranging from 89.72% to 112.54%. Moreover, the method demonstrated excellent specificity and stability.

Based on the reversible covalent binding between borates and glycoproteins, we proposed a scheme that can specifically recognize and detect immunoglobulin G (IgG). The application of magnetic materials to enrich SERS probes can provide an enhanced signal and repeatability. The results showed that the prepared sandwich-like complex displayed high sensitivity and excellent selectivity for IgG. The Raman peak intensity showed a good linear relationship in the concentration range of 1 mg/mL – 1 ng/mL. The linear equation was y = 657.93x + 2963.9, R2 = 0.996. Parallel testing of this sandwich-like complex proved to have excellent repeatability. This method provided a new possibility for clinical non-immunity and the quantitative detection of IgG.

By in situ synthesis of gold nanoparticles (AuNPs) within the acid-etched (AE) MIL-101 (Cr) framework, AE-MIL-101 (Cr) nanocomposites embedded with AuNPs (AuNP/AE-MIL-101 (Cr)) were prepared as surface-enhanced Raman scattering (SERS) substrate. AuNPs are uniformly distributed and stabilized inside the metal-organic framework (MOF), thus forming more SERS hotspots. The SERS performance of AuNP/AE-MIL-101 (Cr) was evaluated using 4-mercaptophenylboronic acid (4-MPBA), 4-mercaptobenzoic acid (4-MBA), benzidine, and rhodamine 6G (R6G). The SERS substrate displays satisfying stability with very low background signal. When benzidine is used as the Raman reporter, the limit of detection (LOD) can reach 6.7 × 10 −13 mol·L −1 , and the relative standard deviation (RSD) of the intra- and inter-batch repetitive tests is less than 5.2%. On this basis, we developed a method for the detection of human carboxylesterase 1 (hCE 1) in human serum using AuNP/AE-MIL-101 (Cr) nanocomposite as SERS substrate and enzyme-linked immunosorbent assay (ELISA) colorimetric substrate as SERS marker. This method was used to determine hCE 1 in clinical serum samples without complicated sample pretreatment, and the detection results were consistent with the data determined by ELISA. In the concentration range 0.1–120 ng·mL −1 , the SERS signal intensity of benzidine at 1609 cm −1 gradually decreases with the increase of hCE 1 concentration (R 2 = 0.9948). The average recoveries of hCE 1 in human serum are in the range 84 to 108%, with RSDs lower than 7.7%. Graphical abstract
By using AuNP/acid etching-MIL-101(Cr) metal organic framework (MOF) as SERS substrate and enzyme-linked immunosorbent assay (ELISA) colorimetric substrate as the SERS marker, a rapid and sensitive method for the determination of human carboxylesterase 1 (hCE1) in human serum samples has been developed.

Plasmonic gold nanorods play important roles in nowadays state-of-the-art plasmonic sensing techniques. Most of the previous studies and applications focused on gold nanorods with relatively small aspect ratios, where the plasmon wavelengths are smaller than 900 nm. Gold nanorods with large aspect ratios are predicted to exhibit high refractive-index sensitivity (Langmir 2008, 24, 5233–5237), which therefore should be promising for the development of high-performance plasmonic chemical- and bio-sensors. In this study, we developed gold nanorods with aspect ratios over 7.9, which exhibit plasmon resonances around 1064 nm. The refractive index (RI) sensitivity of these nanorods have been evaluated by varying their dielectric environment, whereby a sensitivity as high as 473 nm/RIU (refractive index unit) can be obtained. Furthermore, we have demonstrated the large-aspect-ratio nanorods as efficient substrate for surface enhanced Raman spectroscopy (SERS), where an enhancement factor (EF) as high as 9.47 × 108was measured using 4-methylbenzenethiol (4-MBT) as probe molecule. Finally, a type of flexible SERS substrate is developed by conjugating the gold nanorods with the polystyrene (PS) polymer. The results obtained in our study can benefit the development of plasmonic sensing techniques utilized in the near-infrared spectral region. Keywords:large aspect ratios;gold nanorods;refractive index sensitivities;SERS;plasmonic sensing

The large decline in breast cancer mortality has been driven by rapid development and continuous progress in diagnostic techniques. However, it remains difficult to diagnose malignant lesions in distal branching ducts using mainstream techniques such as immunofluorescence ultrasound, molybdenum target, and ductoscopy. Combined with existing reports, we found that CEA level by ELISA in 13 patient 's nipple aspirate fluids (NAF) may be positively correlated with the malignancy of breast lesions. Here, considering the low content of CEA in NAF and the need for automated techniques for batch clinical testing, a magnetic induced mixing surface-enhanced Raman spectroscopy technique (SERS) was used to quantitatively analysis CEA concentration in NAF. In the appearance of CEA, an antibody-conjujated silver nano-probe (AgNPs-Ab1/4-ABP) can be linked onto the surface of another antibody-conjujated magnetic-probe. This immune sandwich-like structure enables more AgNPs-Ab1/4-ABP to remain in situ in the magnetic field. The magnetic spontaneous collection effect brings second SERS signal amplification at 1140 cm −1 approximately. After testing 10 clinical samples, the signal of Magnetic-Field Amplified SERS was not affected by the complex components in NAF, and a good linear range of 0.5–20 ng mL −1 could be achieved. This method might potentially be useful in the differential diagnoses of benign lesions with breast cancer and clinical follow-up of non-surgical intervention nipple discharge patients.

Novel hollow Au Ag alloy nano urchins were synthesized via Ag seeds growth method, and self-assembly coated on the wall and end-tip of silica fiber for fiber probe fabrication. The nano urchins homogeneously distributed on fiber surface because of fiber silanization. The sizes and tip sharpness of the nano-urchins could be controlled by Ag seeds. The elements distribution analysis indicated there was high Ag content in tip-top for better surface enhance Raman scattering performance. The detectable concentration could be as low as 10 −8 M using crystal violet molecules as analyte. Moreover, the fiber probes were stable in air, due to Au in the alloy. This fiber probe could be used for low content single molecular analysis.

Medicinal and edible homologs (MEHs) offer significant preventive and therapeutic benefits for various diseases and health functions. However, the widespread application of MEHs faces significant challenges, particularly in quality control, rapid identification, and authentication. In this study, we present a novel approach that combines surface-enhanced Raman spectroscopy (SERS) based on spectral set, referred to as “SERSome”, with deep learning to develop an identification model for analyzing MEHs. The platform uses silver nanoparticles prepared via reduction with NaBH 4 , activation with sodium borohydride, and aggregation with calcium ions. The method avoids the use of additional protective agents during the reaction process, thereby reducing interference from the protective agents and other materials. Additionally, the method also overcomes the fluorescence interference from MEHs. SERSomes provide comprehensive molecular fingerprint recognition, significantly enhancing the system's detection accuracy. The introduction of Ca 2+ as aggregator promotes the aggregation of silver nanoparticle, significantly enhancing the electromagnetic field of the nanoparticle system. The study achieves up to 98% accuracy in identifying specific MEHs. The integration of SERSomes with deep learning offers a promising methodology for the rapid and modern detection of MEHs, advancing the MEHs industry and contributing to the protection of human health.

The electromagnetic field redistribution in metal nanostructure–film systems exhibits great capability of confining light on plain film, which has great significance in surface-enhanced Raman scattering (SERS) of a molecule on plain film or 2D material. In this work, the SERS spectra of a monolayer molecule on metal or dielectric film were investigated with the help of a metal nanoparticle–nanowire structure. The results indicated that SERS intensity from an adjacent nanoparticle was much greater than that from an individual nanowire, which was confirmed by the simulated electric field distribution through finite method. The polarization dependence of both experimental and simulated data pointed out the importance of electromagnetic field redistribution in this phenomenon. Furthermore, the shape independence of a nanoparticle found in this work greatly improved the application prospect of this nanoparticle–nanowire assisted SERS on film.

Combining both localized surface plasmon polaritons (LSPPs) and propagating surface plasmon polaritons, remote surface-enhanced Raman scattering (SERS) emerges as a novel sensing technology in recent years, which could avoid the overlap of incident light and inelastic scattering light in SERS. Compared to traditional SERS, it has novel applications in sensors, plasmon-driven surface-catalyzed reactions, Raman optical activity, etc. However, the weak Raman intensity of remote SERS impedes its further application. In this work, we demonstrated that the remote SERS signals could be enhanced by more than 100% through the subwavelength interference in dual-path-excited Ag-branched nanowire dimer and nanowire–nanoparticle systems. Our experiment has revealed that remote SERS intensities could be modulated by polarization and phase differences of two incident lights illuminating at two separate nanowire terminals. The simulated electromagnetic field distributions through the finite-difference time-domain (FDTD) method indicate that subwavelength interference occurs in Ag nanowires, which causes the Raman intensities collected at a remote site is greatly influenced by the coherent superposition of propagating surface plasmon polaritons (PSPPs). Our work on this coherent enhancement could not only promote the application of remote SERS but also enlarge the research on light manipulating in the subwavelength.

Tumor-associated macrophages, the largest population of immune cells in the tumor microenvironment (TME), dominate the complexity of TME due to their highly phenotypic plasticity, leading to personalized tumor progression, differentiated drug response, and tolerance. However, current methods for the interpretation of cellular heterogeneity involve destructive, cumbersome, and high-cost sample pretreatment procedures. Herein, we present surface-enhanced Raman scattering cytomics (SERS-Cytomics) as a non-destructive and low-cost approach for macrophage phenotyping and metabolic profiling. SERS-Cytomics is capable of providing the whole molecular fingerprints of living cells at a single-cell level. A deep-learning model enables accurate classification of three phenotypes of macrophages exceeding 95%. Further, the Shapley additive explanations analysis is adopted to screen the differential metabolic signals, where two metabolic biomarkers at 1071 (glucose) and 1440 (cholesterol) cm−1 display the maximum weight in the classification of M1 and M2 macrophages, respectively. Correlation explanation of SERS-Cytomics and transcriptomics reveals the involvement of the glycolytic pathway and lipid metabolism in M1 and M2 macrophages, respectively. Therefore, SERS-Cytomics enables an efficient and non-destructive analytical tool for cellular heterogeneity explanation at a single living cell level.

Attracting target molecules in solution to electromagnetic hot spots is one of the most important problems in the application of surface-enhanced Raman spectroscopy (SERS). In this work, an ultrasensitive SERS detection system with electrokinetic preconcentration is proposed, in which triboelectric nanogenerator (TENG) is employed to provide the electrostatic driving force for molecular enrichment on SERS substrate. During TENG operation, an electric field is generated in solution and provides sufficient driving force for the movement of target molecules. Thanks to the unique output characteristics of TENG, effective enrichment of molecules in triboelectric preconcentration (TEPC) is achieved in 30 s, which exhibits much higher efficiency compared to the enrichment in 10 min with a DC source. Moreover, the TEPC assisted SERS detection system exhibits excellent sensitivity with the limit of detection (LOD) at 10 −15 M. Furthermore, a self-powered TEPC assisted SERS system is designed, which could use environmental mechanical energy and realize outdoor real-time sensitive detection .

An ionic liquid (IL)-based liposome was utilized as a substrate to construct a SERS platform. The isotropy of the IL outer surface together with its ion-exchange property led to the array-like growth of Au nanoparticles (NPs), generating hot-spots and resulting in anionic probes being present on the hot-spot regions. The simultaneous strategy of enrichment and localization endowed the platform with ability to detect trace amounts of anionic probes.
