Mesenchymal stem cells in response to exposed rod-heights of TiO2 nanorod films

Fei Ge,Mengfei Yu,Jun Lin,Cuixia Yu,Wenjian Weng,Kui Cheng,Huiming Wang

Cellular responses are strongly sensitive to surface structure, so the optimization of the structures is essential in biomaterial research. In this work, the exposed nanorod-heights in TiO2 nanorod films were adjusted for enhancing cellular responses. The adjustment was realized by incorporating mesoporous bioactive glass (MBG) into the nanorod films via a sol–gel method. The exposed nanorod-heights in the films could be changed from the original ∼300 nm to ∼200 nm and ∼100 nm. The cellular responses on the nanostructured surfaces were evaluated through culturing mesenchymal stem cells (MSCs). The results showed that the films with shortened nanorod-heights had better cellular responses and could accelerate osteogenic differentiation and mineralization, and the films with 100 nm nanorod-height provided the best surface for cell growth. This is attributed to the nanostructure with the shortened nanorod-heights being well recognized by the cells, consequently the cells grew with a faster osteogenic differentiation through a strengthened BMP-smads signal pathway.

Lyophilizable Stem Cell-Hybrid Liposome with Long-Term Stability and High Targeting Capacity

Tianyuan Zhang,Xixi Zheng,Ruyi Lin,Hao Sun,Honghui Wu,Jinsong Zhang,Shihan Chen,Yaosheng Li,Donghang Xu,Jianqing Gao

The hybridization of liposome with stem cell membranes is an emerging technology to prepare the nanovehicle with the capacity of disease-responsive targeting. However, the long-term storage of this hybrid liposome has received limited attention in the literature, which is essential for its potential applicability in the clinic. Therefore, the preservation of long-term activity of stem cell-hybrid liposome using freeze-drying is investigated in the present study. Mesenchymal stem cell-hybrid liposome is synthesized and its feasibility for freeze-drying under different conditions is examined. Results reveal that pre-freezing the hybrid liposome at −20 °C in Tris buffer solution (pH 7.4) containing 10% trehalose can well preserve the liposomal structure for at least three months. Notably, major membrane proteins on the hybrid liposome are protected in this formulation and CXCR4-associated targeting capacity is maintained both in vitro and in vivo. Consequently, the hybrid liposome stored for three months demonstrates a comparable tumor inhibition as the fresh-prepared one. The present study provides the first insights into the long-term storage of stem cell hybrid liposome using lyophilization, which may make an important step forward in enhancing the long-term stability of these promising biomimetic nanovehicle and ease the logistics and the freeze-storage in the potential clinical applications.

Nanofibril-Structured Granular Hydrogels Harness Stem Cell Retention and Immunoregulation in Diabetic Microenvironment

Qian Yang,Yali Miao,Jinshui Luo,Chunhui Li,Xining Li,Yunhua Chen,Yingjun Wang

Granular hydrogel matrices have shown significant advantages in mesenchymal stem cell (MSC) delivery and tissue ingrowth due to their minimally invasive injection capabilities and porous structures. However, creating granular hydrogels that simultaneously mimic the nanofilamentous architecture of the natural extracellular matrix (ECM) and enhance stem cell retention and in vivo immunoregulation in a diabetic microenvironment remains challenging. In this study, we present a nanoengineered supramolecular granular hydrogel with a nanofibrillar structure designed to improve stem cell retention and regulate immune responses under diabetic conditions. The granular hydrogel matrix is assembled based on multiple hydrogen bonding and hydrophobic interactions, exhibiting a range of tunable features, including shear-thinning, injectability, self-healing, and 3D printability. Furthermore, enriched with manganese dioxide (MnO2)-amyloid fibril (AF) nanohybrids, the granular hydrogel supports MSC adhesion and stemness maintenance and can modulate the reactive oxygen species microenvironment by converting H2O2 into oxygen, thereby promoting cell viability and osteogenic differentiation of MSCs with the sustained release of Mn2+. In a two-week diabetic rat model study, the granular hydrogel demonstrates enhanced in vivo cell retention and anti-inflammatory immunomodulation properties, underscoring its potential as a promising matrix for stem cell therapy and immune regulation in diabetic conditions.

Pretreatment greatly facilitates ethyl levulinate production from catalytic alcoholysis of Napier grass stem

Huai Liu,Yanping Kong,Weipeng Song,Rui Zhang,Junhua Zhang,Yong Sun,Lincai Peng

The inhibition effect of impurities in the raw lignocellulosic biomass greatly impedes its direct acid-catalyzed alcoholysis process to produce downstream chemicals. Previous literatures widely investigated the inhibition problems of lignin content and cellulose crystallite in the raw biomass for the production of alkyl levulinate (AL), while the effect of inorganic matters on this process only received cursory attention. In this contribution, the vital importance of ash removal in raw material for efficient catalytic alcoholysis of cellulose fraction been highlighted. Moreover, it has been found that the facile pretreatment of Napier grass stem greatly facilitates the acid-catalyzed alcoholysis of its cellulose fraction towards ethyl levulinate (EL), a versatile commodity chemical. EL yield as high as 72.1 % can be accomplished from the NaOH-pretreated Napier grass stem in ethanol/γ-valerolactone co-solvent medium over Al(OTf) 3 at 170 ℃ for 150 min, which is almost ten times that from the pristine one in ethanol medium. It is also interesting to note that a facile and green water prewashing treatment can efficiently remove the inorganic component of the raw biomass, which thus largely promotes the conversion of Napier grass stem to EL. The activation energies of cellulose degradation and glucopyranoside alcoholysis after pretreatment decreased to 62.4 kJ/mol and 68.6 kJ/mol, respectively. This work presents a highly-reliable strategy for valorizing grass lignocellulosic feedstock to produce bio-based ester compounds.

A Self-Oxygenating Wound-Adaptive Stem Cell Encapsulation Platform for Chronic Wound Repair

Wei Huang,Yi-Hui Zhang,Ming-Yu Chen,Jian-Hang Zhang,Lei Qiao,Zong-Huan Ba,Hong-Jie Gao,Jing Zang,Xuan Liu,Wen-Qian Liu,Ye-Zi You,Long-Hai Wang

Stem cell-based therapies hold substantial promise for the treatment of chronic wounds. However, limited clinical efficacy has frequently arisen not from inadequate cell potency, but from failure of current delivery paradigms to sustain cell survival and function within hostile host microenvironments. Here, we report a wound-adaptive, self-oxygenating stem cell encapsulation platform designed to overcome this delivery paradigm failure. The device embeds living stem cells within an immunoisolating hydrogel matrix conformally shaped to individual wound geometries and converts locally accumulated CO 2 into molecular O 2 , thereby providing an oxygen supply that preserves cellular metabolic activity in situ. Importantly, oxygenation in this system functions primarily to sustain cell viability rather than to directly oxygenate host tissue. Compared with conventional local or systemic delivery strategies, encapsulated cells exhibit markedly enhanced survival, persistence, and therapeutic function. Evaluation in diabetic rat and porcine wound models demonstrates accelerated resolution of inflammation, improved vascular maturation, and robust tissue regeneration. By reframing cell therapy as a problem of environment-cell compatibility, this platform establishes a clinically translatable delivery paradigm for precision stem cell therapy in chronic wound repair.

A hydrogel platform delivering mesenchymal stem cell lysate for potential myocardial tissue repair

Ziwei Du,Qi Zhang,Qiuhua Wang,Paul Santerre,Tianyi Ma,Peng Ma,Xiaoqing Zhang

Introduction Myocardial infarction is one of the most significant causes of death worldwide and the limited regenerative capacity of the myocardium often results in poor recovery after injury. Stem cell therapy has some promise for myocardial tissue repair, but clinical translation is hindered by poor cell survival and safety concerns, prompting the investigations of cell-free strategies such as stem cell-derived secretome or lysate. Methods In this study, a quaternized chitosan/tannic acid hydrogel was fabricated and characterized for its crosslinking stability, injectability and degradation kinetics. In addition, mouse adipose-derived mesenchymal stem cells (mADSCs) were isolated, characterized, and processed into cell-free lysate. The mADSCs lysate was then analyzed by TEM, NTA, Western blot and enzyme-linked immunosorbent assay. Results It was found that the mADSCs lysate contained abundant vesicles and a broad panel of cytokines/growth factors with anti-inflammatory, pro-angiogenic and antioxidant properties. Encapsulating the lysate within the hydrogel showed sustained release. The hydrogel-based delivery of mADSCs lysate could promote high metabolic activity and good cell number increases of H9c2-cardiomyocytes, L929-fibroblasts and human umbilical vein endothelial cells, demonstrating great potential for myocardial tissue repair applications. Discussion By integrating the regenerative potential of ADSCs lysate with this biocompatible, injectable hydrogel platform, this combined strategy circumvents the potential safety issues associated with live cell transplantation and offers a scalable, potential cell-free delivery strategy for myocardial repair-related applications, laying a foundation for future in vivo studies and further translational evaluation.

Preparation, purification, and antioxidant activities of low-molecular-weight polysaccharides from Dendrobium stem

Zhenzhu Yang,Qunqun Chen,Huiqin Chen,Junjie Sun,Jianting Yang

Dendrobium is famous for its edibility and medicinal uses, with polysaccharides as active ingredients. Optimal fermentation conditions were obtained through single factor experiments combined with response surface methodology. The results showed that the extraction of CDP (Crude Dendrobium polysaccharides) was up to 5.84% with the inoculum 1.0%, sucrose dosage 0.9%, fermentation broth pH 5.0 of fermentation condition. A homogeneous polysaccharides component, designated as DOP-1, was successfully isolated and purified from Dendrobium officinale stem. The structural characterization of DOP-1 was conducted utilizing ultraviolet (UV) spectroscopy, Fourier transform infrared spectroscopy (FT-IR), high-performance liquid chromatography (HPLC), nuclear magnetic resonance spectroscopy (NMR), and scanning electron microscopy (SEM). Structure analysis revealed DOP-1 is mainly made up of mannose and glucose, with a molecular weight of 5526 Da. The structure of DOP-1 is composed of alternating or segmented D-mannose and D-glucose connected by β - (1→4), and carries monosaccharide side chains at the C2 positions of the residues. DOP-1 showed antioxidant capacity by scavenging DPPH, ABTS and •OH radicals in vitro antioxidant experiments, with IC 50 values of 5.43, 8.62 and 5.88 mg/mL respectively. During in vitro digestion test, no significant differences ( P > 0.05) were observed in the contents of total sugars and reducing sugars of DOP-1 before and after digestion, whereas its antioxidant activity dynamically increased initially and then remained stable. After digestion, the scavenging rates of DOP-1 against DPPH, ABTS, and •OH radicals were 73.18%, 79.55%, and 78.41%, respectively. The results showed that DOP-1 exhibited good gastrointestinal tolerance and potential for prebiotic development.

Lipoic acid functions in Paneth cells to prevent human intestinal stem cell aging

Zhang Zehong,Wan Qianyi,Zhu Yuedan,Ye Jinbao,Fu Fang,Fan Xinxin,Chen Haiou,Ren Gang,Jiang Wei,Guo Xiaoxin,Zhou Juanyu,Yan La,Yan Jinhua,Liu Xingzhu,Yuan Yu,Chen Yi,Chen Haiyang

Intestinal stem cell (ISC) aging diminishes the regenerative capacity of the intestinal epithelia, but effective therapeutic strategies to counteract human ISC aging remain elusive. Here, we find that the synthesis of α-lipoic acid (ALA) is reduced in old human small intestine. Notably, ALA supplementation inhibits ISC aging and decreases the number of atypical Paneth cells in old human intestinal organoids and in old mouse small intestines. Importantly, we discern that the effect of ALA on mitigating ISC aging is contingent upon the presence of Paneth cells. Inhibiting the mTOR pathway in Paneth cells with ALA or rapamycin significantly increases cyclic ADP ribose (cADPR) secretion and decreases Notum secretion, which, in turn, enhances ISC functions. In this work, our findings substantiate the role of ALA in inhibiting human ISC aging and present a potential therapeutic approach for managing age-related human intestinal diseases.

Protocol to quantify protein phosphorylation in mouse extended pluripotent stem cell-derived blastoids

Tianxia Xiao,Yuxin Luo,Yang Yu

Summary Protein phosphorylation is an important post-transcription modification regulating essential cellular activities. Here, we present a proteomics protocol to quantify protein phosphorylation in the extended pluripotent stem cell (EPSC)-derived blastoids from Mus musculus with a standardized phosphoproteome analysis workflow. We describe steps for culturing blastoids, preparing samples, enriching phosphopeptides, and fractionating peptides. We then detail procedures for acquiring and analyzing high-resolution liquid chromatography-mass spectrometry (LC-MS) data. This protocol provides a framework for reproducible analysis of phospho-proteomes of mouse blastoids. For complete details on the use and execution of this protocol, please refer to Min et al. 1

Zwitterionic hydrogel microspheres for efficient expansion and stemness maintenance of mesenchymal stem cells

Qingyun Jiang,Shuai Shao,Zunhui Zhao,Na Li,Hangyu Zhang,Bo Liu

In this study, we engineered dual-network amphiphilic ionic hydrogel microspheres as microcarriers for culturing in mesenchymal stem cells (MSCs), enhancing stemness retention and proliferation. The microspheres were termed DCsa. We innovatively integrated the zwitterionic sulfobetaine methacrylate (SBMA) into the DCsa microsphere structure, and with its outstanding superhydrophilic properties, formed high-energy hydration barrier sites with strong electrical neutrality on the surface of the microspheres. These sites can efficiently block the adsorption of non-specific proteins, ensuring the maintenance of stemness in long-term cultured MSCs. In addition, the addition of AAm endows the surface of microspheres with abundant functional site groups, which exhibit significant promoting effects on cell adhesion and proliferation processes. Functional assessments confirmed that MSCs cultured on DCsa maintained high cells viability and stemness over extended periods. These findings underscore the potential of DCsa microspheres as an effective three-dimensional (3D) culture platform for the large-scale propagation of MSCs, offering significant implications for the advancement of regenerative medicine and cell therapy.

Triggered ROS cyclic responsive silicon nanowire arrays for gene transfection of stem cells

Yanyan Wang,Liping Zhang,Shengxuan Xu,Shuangling Liu,Wenxin Qiu,Hongwei Wang,Lin Yuan

The central challenge in gene delivery systems lies in achieving efficient intracellular delivery of plasmids and sustained release. In this study, a cyclic reactive oxygen species (ROS)-responsive gene delivery platform based on silicon nanowire arrays (SN) was developed. The high-density positive charged polyethylenimine (PEI) polymers were covalently grafted onto the SN surface, allowing for electrostatic adsorption of both plasmid DNA and trans-cinnamaldehyde precursor (TAC). When cells are co-cultured on this material, the needle-like nanostructures of SN would penetrate the cell membrane through physical effect, efficiently delivering the loaded plasmids into stem cells. The release mechanism of this system was based on an ROS-triggered cascade response, TAC oxidized by ROS to generate cinnamaldehyde, and then induced mitochondrial oxidative stress to promote cascade production of more ROS. This triggered the dissociation of SN-grafted PEI from the nanowires, enabling sustained plasmid release. In vitro release assays showed that the treatment of 1 mM H 2 O 2 (exogenous ROS stimulant) for only 10 min induced 79 % plasmid release from SN surfaces. For cells that are difficult to be transfected, such as mouse embryonic stem cells (mESC) and mesenchymal stem cells (MSC), this platform exhibited excellent transfection efficiencies of 94 % and 74 %, respectively, while maintaining good cytocompatibility. This study developed an SN-based ROS-responsive gene delivery platform, achieving efficient plasmid delivery and sustained release via synergistic physical penetration and biochemical-responsiveness. It provides a solution with important application value for gene transfection in hard-to-transfect stem cells and demonstrates promising translational potential in the fields of gene therapy and regenerative medicine.

Co-assembled Manganese-CpG Nanoliposomes for Enhanced Immunotherapy against Colon Cancer Stem Cells

Weijian Zhao,Siyu Sun,Xuesong He,Yiran Liu,Jing Meng,Qin Hu,Wang Sheng,Runqing Jia

Nanovaccines represent a promising strategy for colon cancer immunotherapy, with the potential to elicit potent, tumor-specific immune responses. However, the efficacy of these vaccines is often compromised by the presence of cancer stem cells (CSCs) in the tumor microenvironment (TME). Nanoliposome (NLP) is a widely used delivery system in nucleic acid and drug delivery research. In this study, we enriched MC38-derived CSCs (MCSCs) and developed a manganese-CpG-nanoliposome (Mn@CpG@NLP) nanocomplex using MC38 colon carcinoma tumor lysates as antigens to induce immune responses against MCSCs-derived tumors. Manganese NLPs (Mn@NLP) are initially engineered by incorporating manganese ions to enhance their positive surface charge, thereby optimizing their interaction with cellular membranes and activating the STING signaling pathway to promote bone marrow-derived dendritic cells (BMDCs) maturation. Subsequently, these liposomes are co-assembled with the CpG oligonucleotides (CpG ODNs) 1826 adjuvant, a Toll-like receptor 9 agonist, through electrostatic interactions to form the Mn@CpG@NLP nano-adjuvant complex. The Mn@CpG@NLP complex is efficiently taken up by dendritic cells (DCs), leading to their maturation and activation. This nanocomplex also effectively stimulated cytotoxic T lymphocytes and promoted the secretion of cytokines. In vivo treatment with Mn@CpG@NLP significantly inhibited tumor growth and prolonged survival in a mouse model. This study highlights the potential of nano-adjuvant platforms in immunotherapy targeting cancers driven by colon CSCs.
