Chronic diabetic wounds are often complicated by persistent infections and oxidative stress, which impede effective healing. In this study, we developed an injectable, dual-responsive nanocomposite hydrogel designed to simultaneously scavenge reactive oxygen species (ROS) and deliver photothermal and photodynamic therapy for wound treatment. The hydrogel was synthesized via dynamic boronate-ester crosslinking of poly(vinyl alcohol) (PVA) with a bis-boronic acid linker (TSPBA), enabling efficient ROS quenching. Incorporation of palygorskite/molybdenum disulfide (PGS/MoS₂) nanocomposites imparted photothermal and photodynamic properties, activated by NIR light. In vitro tests demonstrated that the PVA-TSPBA@PGS/MoS₂ hydrogel effectively scavenged ROS, induced mild hyperthermia (~ 48 °C) under NIR, and exhibited significant antibacterial activity against Staphylococcus aureus. In vivo, the hydrogel accelerated wound closure in a diabetic mouse model, with a 46.7% residual area at day 7, compared to higher residual areas in control groups (66.5–92.4%). By day 14, nearly complete wound healing was achieved, accompanied by reduced bacterial load and inflammatory markers, as well as enhanced re-epithelialization, angiogenesis, and collagen remodeling. These findings suggest that this multifunctional hydrogel represents a promising strategy for the treatment of infected diabetic wounds, offering a controlled, localized approach to wound healing. Chronic wound infections caused by methicillin-resistant Staphylococcus aureus (MRSA) often refractory to conventional therapies due to bacterial drug resistance and biofilm formation. To address this challenge, we developed a synergistic antibacterial nanoarchitectonics integrating bacteriophages and quantum dots (QDs-Phage). This system was constructed by bioconjugating MRSA-specific bacteriophage with highly photothermally efficient PbS quantum dots, creating an intelligent therapeutic platform that operates on a “target-first, activate-later” principle. Under near-infrared laser irradiation, the nanoarchitectonics exhibited significant synergistic bactericidal activity against both MRSA and its biofilms in vitro. In a murine model of full-thickness skin wounds infected with MRSA, topical application of QDs-Phage combined with light irradiation significantly accelerated wound closure and promoted tissue repair, without inducing obvious systemic toxicity. This study provides a novel strategy for developing localized antibacterial approaches that combine precise targeting with efficient physical bactericidal functions.

Staphylococcus aureus is a major pathogen responsible for hospital- and community-acquired infections, with the accessory gene regulator (agr) system playing a central role in virulence control. The ST9 lineage commonly associated with livestock exhibits multidrug resistance but remains understudied in terms of virulence. In this work, we examined nine clinical ST9 methicillin-resistant S. aureus isolates and identified two strains carrying spontaneous frameshift mutations in the agr locus, resulting in the loss of RNAIII and α-toxin expression. These agr-deficient strains exhibited reduced hemolytic activity, enhanced biofilm formation, and attenuated virulence in a Galleria mellonella infection model. These preliminary pieces of evidence suggested a critical role of the agr system in ST9 virulence. Comparative genomic analyses revealed distinct pathogenicity islands in ST9, differing from those in other common clinical lineages. Notably, we identified three novel prophage islands (φST9-A, φST9-B, and φST9-C), with φST9-B carrying the recently characterized tarP virulence gene, suggesting that recombination events within livestock-associated MRSA could contribute to the genetic diversification of ST9. These findings demonstrate the significant virulence potential of the ST9 lineage and the importance of agr-mediated regulation. Moreover, the presence of new prophages highlights the evolutionary adaptability of this lineage. Collectively, our results underscore the clinical threat posed by ST9 MRSA and the necessity for vigilant monitoring to control its spread.

Background Heterogeneous vancomycin-intermediate Staphylococcus aureus (hVISA) is associated with suboptimal glycopeptide treatment outcomes. However, conventional antimicrobial susceptibility testing fails to distinguish hVISA from vancomycin-susceptible Staphylococcus aureus (VSSA), potentially delaying effective therapy. The gold standard population analysis profiling with area under the curve (PAP-AUC) method is labor-intensive and time-consuming, necessitating rapid detection alternatives. Methods This study introduces a microfluidic platform based on fluorescence-activated droplet sorting (FADS) for the rapid detection and isolation of hVISA. Through systematic optimization of key parameters such as droplet generation conditions, bacterial suspension concentration, fluorescence probe selection, fluorescence duration, and vancomycin screening concentration, we established a rapid hVISA screening system. The efficacy of this system was evaluated using the hVISA standard strain Mu3, followed by validation with 15 clinical hVISA isolates. Genomic analysis further elucidated the genetic basis of drug resistance in hVISA strains. Results We developed a high-throughput detection platform for hVISA by integrating a microfluidic system with FADS. Based on a Poisson distribution theoretical model, the bacterial suspension concentration was optimized to 10 6 CFU/mL, achieving a single-bacterium droplet encapsulation rate of approximately 30%. The selected fluorescent probe exhibited detectable fluorescence signals as early as 6 h post-cultivation, with stability maintained for at least 1 week. Drug concentration validation demonstrated that 4 μg/mL vancomycin effectively inhibited susceptible bacteria without interfering with fluorescence detection. This platform successfully isolated resistant subpopulations from the hVISA reference strain Mu3 within 12 h, and its efficacy was further validated using 15 clinical hVISA isolates. Conclusions This study innovatively integrated a microfluidic detection platform and FADS to achieve rapid detection of hVISA. This technology reduced the detection time to just 12 h and significantly improved the enrichment efficiency of drug-resistant bacterial populations.

Staphylococcus (S.) aureus is an important microorganism that causes foodborne poisoning, which threatens food safety. However, few studies have investigated novel natural antibacterial peptides produced by the probiotic Lactobacillus sp., which effectively inhibits S. aureus . In mulberry orchard soil used as a screening resource, 28 LAB were isolated and found to have antibacterial effects on S. aureus . Among them, the antibacterial activity of Lacticaseibacillus (L.) paracasei GY1 reached 103 % when 40 g/L sucrose and 15 g/L yeast extract was used, the initial pH was 7, and the temperature was 37 °C for 48 h. This strain maintains cell viability in gastric and intestinal fluids as well as in 4 % NaCl, with its self-aggregation ability reaching 94 %. A novel antibacterial peptide (EKSQAHLDAGAK) was separated and purified by macroporous resin D101, DEAE Focusose 6FF anion exchange chromatography, Sephadex G-15 gel filtration chromatography, and preparative HPLC. Its EC 50 and MIC values were 117.57 and 176.64 μg/mL, respectively. Its inhibitory effect is achieved by destroying the cell membrane, enhancing cell permeability, and reducing cell division. The probiotic L. paracasei GY1 can be used to develop functional foods, and the antibacterial peptide serves as an important reference for the precise control of foodborne pathogens.

Lithium–sulfur (Li–S) batteries suffer from some serious issues, such as volume expansion, polysulfide migration, and so on, which lead to inferior rate properties, low Coulombic efficiency, and poor cycling stability. To a great extent, the significantly changing polarities, conductivities, and solubilities of sulfur species complicate these issues and make them hard to deal with. In this study, a biomimetic bipolar microcapsule is developed via bacteria fermentation and chemical modification. The biomimetic hierarchical microcapsule has a nonpolar core to load active materials and a polar shell to “selectively” control mass transport in and out. Each capsule acts as a microreactor, which adsorbs sulfur with the porous carbon core, retards polysulfide migration with the polar TiO 2 shell, and only facilitates lithium ion diffusion through the shell. The advantages of bipolar microcapsules are able to concurrently address S hosting, electron conducting, and polysulfide migration issues in particle scales instead of electrode scales. Because the resulting sulfur cathodes effectively interact with all sulfur species and confine them inside microcapsules, those intractable issues that lead to poor cycling properties are significantly managed. Thus, a high specific capacity and excellent cycling stability are achieved by using this novel structure design.

Bacterial infection can cause delay in wound healing, and reactive oxygen species (ROS) was accumulated in wounds, which threaten patients’ lives. Therefore, it needs to develop a multifunctional hydrogel for infected wound healing. Here, a multifunctional QAS-CS@GA-GM hydrogel with antibacterial and antioxidant property, established by the self-adaptive covalent bond, is prepared aiming to eradicate bacteria and eliminate ROS. It is observed that QAS-CS@GA-GM hydrogel displays remarkable ability of clearing ROS and killing bacteria in vitro. With its antibacterial and antioxidant activity, QAS-CS@GA-GM hydrogel significantly accelerates the MRSA-infected wound closure and reduces inflammatory, when QAS-CS@GA-GM hydrogel was applied to wound sites. Collectively, the research suggests that a self-adaptive QAS-CS@GA-GM hydrogel is a promising wound dressing as antibacterial and ROS-scavenging dressing for bacterial-infected wound.

Staphylococcus aureus, especially methicillin-resistant S. aureus (MRSA), poses a severe threat to human health due to the limited efficacy of traditional antibiotics and the rapid emergence of drug resistance. Quaternary ammonium compounds (QACs) are promising antibacterial agents with a membrane-disruptive mechanism that is less prone to inducing resistance. Herein, a series of novel phthalazine-derived QACs (2a–2v) were designed, synthesized, and evaluated for their antibacterial activities. The structure-activity relationship (SAR) analysis revealed that the length of N-alkyl substituent significantly affected antibacterial potency, with compound 2n emerging as the lead compound. Compound 2n exhibited broad-spectrum antibacterial activity against S. aureus, MRSA, Streptococcus pneumoniae, and Klebsiella pneumoniae, with MIC values of 2 μg/mL. It showed rapid bactericidal activity, low hemolysis (HC50 > 64 μg/mL), minimal resistance evolution, and excellent stability in physiological salts and against proteolytic enzymes, with certain concentration-dependent effects on biofilm. Mechanistic studies revealed that compound 2n exerts antibacterial activity via triggering bacterial membrane damage, which was verified by multiple phenotypic results, including Gram staining alteration, increased extracellular AKP activity, cell membrane depolarization, intracellular protein and nucleic acid leakage, as well as morphological observations from SEM and DAPI/PI staining. Molecular docking and competitive displacement assays indicated that compound 2n may bind to bacterial DNA, which is presumed to be a potential auxiliary antibacterial mechanism. These findings suggest phthalazine-derived QACs are promising and compound 2n has great potential for further development to combat drug-resistant bacterial infections.

Micrococcal nuclease (MNase) is a nonspecific endo-exonuclease that digests single-stranded/double-stranded DNA and RNA. The existence of MNase can serves as an important diagnostic biomarker of Staphylococcus aureus ( S. aureus ) infection. However, most of the substrates in MNase-based sensors are single-stranded DNA, which could also be digested by exonuclease I or S1 nuclease and interfere the MNase detection. In this work, we developed a highly selective fluorescent method for MNase detection using a specific dsDNA and nucleic acid dye SYBR Green I (SGI) as the indicator. After rational design, an AT-rich dsDNA with 3′ protruding termini was screened as the high-specific substrate of MNase assay and efficient enhancer of SGI. The AT-rich dsDNA substrate can resist the digestion of other exonuclease and greatly enhance the fluorescence of SGI. This high-specific substrate-based probe can detect MNase in buffer as well as biological sample with highly selectivity . Moreover, this method was also applied to monitor the MNase secreted by S. aureus . Thus, the proposed MNase-based assay has a strong potential to identify S. aureus in food safety and microbial infection due to its excellent analytical sensitivity and high selectivity.

The inhibitory mechanism of CUM on Staphylococcus aureus exotoxins and invasive enzyme, and the effect of CUM on sauced beef were investigated. Treatment with sub-minimum inhibitory concentrations of CUM observed very little decrease in cell density after 24 h. Additionally, CUM reduced hemolytic activity by inhibiting the formation of heptamer, and inhibited the content of enterotoxin in culture medium and sauced beef. The transcription of genes encoding α-hemolysin, staphylococcal enterotoxin A and Panton-Valentine leukocidin were down-regulated after treatment with CUM. Compared with the control group, the addition of CUM caused a significant decrease in S. aureus counts, total volatile base nitrogen, acid value, and thiobarbituric acid reactive substances of sauced beef after 9 d. Furthermore, CUM improved the redness (a*) of sauced beef. These results confirmed that CUM could effectively restrain the synthesis of S. aureus toxin, and the addition of CUM to sauced beef was the potential way to inhibit the growth of food pathogens and maintain quality profiles.

The objective of this study was to design a more effective antibacterial agent to overcome the problem of fast-growing bacterial resistance. The results showed that a Cu/H3BTC metal–organic framework (MOF) exhibited a notable antibacterial performance towards Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), and could significantly disrupt the cell membrane, resulting in the discharge of cell constituents. In addition, morphological changes were elucidated using scanning electron microscopy (SEM). SEM photographs demonstrated noticeable and apparent changes in the morphology of bacterial cells, signifying cell damage that was further confirmed via confocal laser scanning microscopy (CLSM). Furthermore, the result of agarose gel electrophoresis (AGE) showed that Cu/H3BTC MOF could enter the bacterial cells by impairing cell membranes and inhibiting the synthesis of DNA. These findings indicated that the Cu/H3BTC MOF is a potent antimicrobial material that can be applied to hinder the growth of pathogenic microbes.

Staphylococcus aureus is one of the most important food-borne bacterial pathogens and causes numerous illnesses. In this work, we report a sensitive and highly selective magnetic-aptamer biosensor based on a personal glucose meter (PGM) platform for the detection of Staphylococcus aureus . The aptamer for Staphylococcus aureus was immobilized on the magnetic bead by hybridization with the capture probe P. In the presence of Staphylococcus aureus , the aptamer was dissociated from the magnetic bead. Then the capture probe was exposed and could be hybridized with a biotinylated probe to trigger the DNA hybridization chain reaction (HCR), thus achieving the signal amplification. The concentration of streptavidin-labeled invertase can be read by PGM, thus can lead to the portable quantitative detection of Staphylococcus aureus . After optimization of various conditions, 5 μM probe P, the MB-P reaction time for 36 h, the competition time for 60 min, 0.5 μM H1 & H2, 0.5 M sucrose and the sucrose invertase catalytic reaction time for 50 min was chosen to achieve the better sensor performance. Under the optimal conditions, the fabricated sensor offers high sensitivity with the limit of detection about 2 CFU/mL. This sensitive PGM based sensor could successfully evaluate the Staphylococcus aureus concentration in real food samples, and the results are consistent with those obtained by using plate counting methods. Moreover, the PGM sensor can greatly reduce the required time compared to the plate counting methods. The fabricated sensor supplies an ideal solution for rapid portable detection of bacterial pathogens and holds its potential use in the quality control for agriculture and food enterprises, entry-exit inspection and quality testing for food.
