UiO-67 decorated on porous carbon derived from Ce-MOF for the enrichment and fluorescence determination of glyphosate

Qiang Yu,Yang Weixia,Zhang Xiaoshuo,Luo Xueli,Tang Wenzhi,Yue Tianli,Li Zhonghong

A nanocomposite was prepared by loading UiO-67 nanoparticles onto porous carbon materials derived from Ce-MOF (Ce-PC) for fluorescence detection of glyphosate. The probe (UiO-67/Ce-PC) exhibits fluorescence emission at 414 nm as the response signal under excitation at 310 nm. The fluorescence enhancement mode of UiO-67 reduces the background interference, and the introduction of Ce-PC provide hierarchical nanostructure and large specific surface area that can increase the contact availability and improve the pre-enrichment effect, ensuring UiO-67/Ce-PC with superior sensitivity. The abundant metal hydroxyl group (M–O-H) of UiO-67/Ce-PC could recognize phosphoryl groups (-PO 3 H 2 ) of glyphosate through ligand exchange, which synergizes with H-bonding interaction and electrostatic attraction to exhibit specificity toward glyphosate. The competitive coordination effects weaken the ligand-to-metal charge transfer (LMCT) and consequently induce the fluorescence recovery. The calibration plot of the fluorescence enhancement response of UiO-67/Ce-PC towards glyphosate was recorded in the range 0.02–30 μg mL −1 with a low limit of detection (LOD) of 0.0062 μg mL −1 , which is superior to the pure UiO-67. In addition, the sensor exhibited high selectivity and satisfactory accuracy and precision with recoveries of 92.1–105.6% and RSDs below 3.4%. This work not only presents a feasible sensor for sensitive and selective determination of glyphosate from cereal samples, but also provides a promising strategy for the design of MOF-based nanocomposites to achieve trace detection of various pollutants. Ti4+ modified melamine foam in the pipette tip for effective solid-phase extraction of glyphosate in aqueous samples

Jinjie Xu,Yuehong Pang,Zhuying Yan,Xiaofang Shen

Glyphosate, a widely used herbicide, is prone to residue in the environment that may cause harm to humans. In this work, titanium ion-modified melamine foams (MF/PDA/Ti 4+ ) were prepared as sorbents to achieve rapid extraction of glyphosate by pipette-tip solid-phase extraction (PTSPE). The MF/PDA/Ti 4+ was characterized by SEM, EDS, XPS, and TGA, and its adsorption properties were evaluated by adsorption kinetics and isotherm experiments. The porous structure and good mechanical properties of MF made the MF/PDA/Ti 4+ fill into the pipette-tip easily and could achieve low back pressure. The developed PTSPE method showcased short extraction time (<2 min) for glyphosate and the limits of detection (LOD, 3σ/k) was 4.9 μg/L. It was applied in aqueous samples (tap water, lake water, tea drink, and orange juice) with recoveries ranging from 82.8 % to 108.2 % and the relative standard deviation (RSD, n = 3) was less than 7.7 %, demonstrating broad perspectives on the extraction of glyphosate from the aquatic and terrestrial environment.

A Turn-On Fluorescent Assay for Glyphosate Determination Based on Polydopamine-Polyethyleneimine Copolymer via the Inner Filter Effect

Pengjuan Ni,Siyuan Liu,Yizhong Lu

The threat of glyphosate to food safety has attracted widespread attention. Consequently, it is highly urgent to develop a sensitive and accurate method for glyphosate detection. Herein, a turn-on fluorescent method for glyphosate detection using polydopamine-polyethyleneimine (PDA-PEI) copolymer as a fluorescent probe andp-nitrophenylphosphate (PNPP)/alkaline phosphatase (ALP) as a fluorescence quenching system is developed. The PDA-PEI copolymer was prepared by a one-pot method under mild condition, and its fluorescence kept almost unchanged after storing in a refrigerator for one month. ALP catalyzed the hydrolysis of PNPP top-nitrophenol (PNP) that caused the fluorescence quenching of PDA-PEI copolymer via the inner filter effect. However, glyphosate inhibited ALP activity, thereby preventing the formation of PNP and restoring the fluorescence signal. Under the optimized conditions, the fluorescence of PDA-PEI copolymer depended on glyphosate concentrations ranging from 0.2 to 10 μg/mL with a detection limit of 0.06 μg/mL. Moreover, this assay was applied to detect glyphosate in real samples using the standard addition method. The recoveries were in the range from 88.8% to 107.0% with RSD less than 7.78%. This study provides a novel insight for glyphosate detection and expands the applications of fluorescent copolymer. Keywords:fluorescence;alkaline phosphatase;glyphosate;inner filter effect

Rational Design of a Molecularly Imprinted Sensor on a Biomass Carbon Platform for Glyphosate Monitoring in Traditional Chinese Medicines

Xin Wang,Delai Zhou,Xuxia Liu,Guodi Lu,Jia Hou,Jian Xu,Fude Yang

A molecularly imprinted electrochemical sensor was developed for the selective and sensitive detection of glyphosate in Traditional Chinese Medicine samples. An excellent conductive hierarchical porous carbon substrate made from sodium alginate and ammonium chloride co-carbonization was used to build the sensor. The molecularly imprinted polymer layer was systematically designed using Density Functional Theory calculations, which identified nicotinamide as the optimal functional monomer. A deep eutectic solvent was utilized as an effective green eluent for template removal. Under optimized conditions, the sensor demonstrated a wide linear detection range from 1.0 × 10−9to 1.0 × 10−6M with an exceptionally low detection limit of 8.8 × 10−10M. The sensor exhibited satisfactory reproducibility (RSD = 3.35%,n= 6), repeatability (RSD = 5.0% over 6 cycles), and robust stability (retaining >90% initial response after 10 days). The sensor displayed satisfactory recovery rates of 94.47–112.23% and RSD values ranging from 1.37–3.01% when applied to real traditional Chinese medicine samples, thereby confirming its accuracy and practical utility for glyphosate residue analysis in complex matrices. This study introduces an effective sensing platform that integrates rational design principles with environmentally friendly synthesis strategies for quality control in traditional medicine applications.

Co-exposure to polystyrene nanoplastics and glyphosate exacerbates NETs-mediated pyroptosis by activating the NLRP3 inflammasome in mouse liver

Huan Zeng,Jingyi Qi,Qiyi Chen,Ruowei Zhang,Chen Guo,Jinghua Zhao,Huimei Liang,Junlong Sun,Wei Wang,Houhui Song

Nanoplastics (NPs), highly prevalent due to large-scale plastic production, and glyphosate (Gly), the most utilized herbicides worldwide, are ubiquitous environmental contaminants. Growing concerns highlight that NPs can act as vectors for various pollutants like Gly, but their combined toxic effects in mammals and the underlying mechanisms remain poorly understood. In this study, the hepatotoxicity and potential mechanisms under the exposure of polystyrene nanoplastics (PSNPs) and/or Gly in vivo and in vitro were investigated. Mice were treated with PSNPs (25 mg/kg/day) and/or Gly (50 mg/kg/day) by oral gavage for 5 weeks. Results showed that exposure to PSNPs or Gly caused liver injury in mice, with co-exposure resulting exacerbated hepatotoxicity, evidenced by increased neutrophil infiltration and ultrastructural damages, elevated oxidative stress (LPO, H 2 O 2 , T-AOC and CAT), increased neutrophil chemokines (CCL2, CXCL12) and marker of neutrophil extracellular traps (NETs) formation (MPO), and upregulated pyroptosis-related factors (TLR4, NF-κB, NLRP3, ASC, Caspase-1, GSDMD, IL-1β, IL-18). In addition, a co-culture system of peripheral blood neutrophils (PBNs) and AML12 cells was established, revealing that co-exposure amplified ROS production, NETs formation (SYTOX Green staining) and pyroptosis. Notably, inhibition of the NLRP3 inflammasome significantly reduced NETs production, and degradation of NETs substantially decreased pyroptosis, demonstrating feedback between NETs and the NLRP3 inflammasome that drives inflammation induced by PSNPs and Gly. These results highlight that co-exposure exacerbated NETs-mediated pyroptosis through NLRP3 inflammasome activation. Our study equips new reference for understanding the mechanistic insights and health implications of the combined toxicity of PSNPs and Gly.

A novel hydrazide-based fluorescent chemosensor and its application to recognize Cu2+ and glyphosate in environmental and biological systems

Dong Zhang,Xiaohui Pang,Xinfu Zhu,Yi Wang,Meiqing Zhu

Background Although copper ions (Cu 2+ ) and glyphosate (GLY) tend to exist in the environment or living organisms at low doses, they gradually accumulate with exposure time and may eventually cause irreversible damage to living organisms. The existing detection methods are effective, but most of them lack the ability to detect multiple pollutants simultaneously or suffer from low sensitivity and poor selectivity. Therefore, it is important to develop a sensor that can efficiently and sensitively detect copper ions and glyphosate, but also has the potential for practical applications. Results In this study, a novel hydrazide-based DHBC capable of sequentially detecting Cu 2+ and glyphosate (GLY) was developed. The DHBC forms a complex with Cu 2+ , causing fluorescence quenching. Upon exposure to GLY, functional groups such as carboxyl, phosphate, and amino groups coordinate with Cu 2+ , displacing DHBC and restoring fluorescence intensity. The mechanism has been validated by various characterization methods such as HRMS and Job curves, DFT calculation, etc. The DHBC has excellent detection performance, including high selectivity, strong anti-interference, fast response (less than 1 min), and high sensitivity (Cu 2+ : 0.09 μM, GLY: 0.05 μM). The quantitative analysis of Cu 2+ /GLY in cycling experiments and actual samples demonstrated the great potential of DHBC for practical applications. The DHBC successfully imaged Cu 2+ /GLY in live models such as HeLa cells and zebrafish, demonstrating its broad applicability and stability in a wide range of biological models. Significance The high accuracy of DHBC in environmental pollution monitoring and its stable performance in biological modeling make it an important tool for assessing the impact of pollutants on ecosystems and human health. In addition, the wide applicability and real-time imaging capability of DHBC provide reliable technical support for environmental science, toxicology, and health risk assessment and have important prospects for scientific research and practical applications.

Co-exposure to polystyrene nanoplastics and glyphosate promotes intestinal apoptosis in mice via intestinal barrier impairment and immunoinflammatory dysregulation

Beining Wu,Han Liu,Ruoyu Dong,Haoyu Xu,Jingyi Qi,Huimei Liang,Chen Guo,Huan Zeng,Jinghua Zhao,Houhui Song,Yongchun Yang,Wei Wang

Environmental contaminants such as polystyrene nanoplastics (PSNPs, 1–1000 nm) and glyphosate pose significant environmental and public health risks. This study aimed to investigate the intestinal toxicity and molecular mechanisms induced by PSNPs and/or glyphosate. Mice were exposed to PSNPs (<100 nm), glyphosate, or a combination of both for 35 days via intragastric administration (PSNPs: 0.5 mg/d; glyphosate: 50 mg/kg-bw/day; PSNPs + glyphosate: 0.5 mg/d +50 mg/kg-bw/day). The control group received same volume of distilled water. Our findings revealed that exposure to PSNPs and/or glyphosate aggravated pathological alterations, including inflammatory cell infiltration, severe mitochondrial cristae fracture, and an approximately 50 % reduction in goblet cells in the intestine. Moreover, exposure to PSNPs and/or glyphosate caused a critical 75 % inhibition of FOXP3 and dissociation of tight junctions in the intestine (reflected by a 50 % decrease in Occludin, and a 20 %–50 % decrease in ZO-1). These changes were accompanied by significant alterations in beneficial gut microbiota, metabolic profiles, bile acid metabolism disorders, and a pronounced elevation in 3-β-deoxycholic acid, a metabolite tied to bile acid receptor signaling and barrier dysfunction. Although exposure to glyphosate led to the most significant upregulation of the pro-inflammatory factors TNF-α and the pro-apoptosis proteins Cleave-caspase-3, co-exposure did not exacerbate cell apoptosis in animal tissue experiments, which is contrasts with the cell-based findings. MODE-K (mouse intestinal epithelial) cells were treated with PSNPs (0.75 mg/mL) or glyphosate (0.5 mg/mL). In vitro experiments showed that PSNPs aggravated the disrupted Treg/Th17 immune-inflammatory balance, impaired intestinal barrier function (with a 50 % reduction in ZO-1 and Occludin), and increased cell apoptosis, caused by glyphosate. This study advances our understanding of the health risks posed by endocrine-disrupting chemical mixtures and provides critical insights into the molecular mechanisms of PSNP-glyphosate-induced intestinal toxicity. These findings lay the groundwork for future research aimed at mitigating the pathophysiological impacts of environmental pollutants.

An artificial neural network model based on CDs@MOF/COF composites for the ultra-sensitive fluorescence detection of glyphosate

Yingfei Hui,Yuli Wei,Hao Guo,Mingyue Wang,Liping Peng,Borong Ren,Yanyan Ma,Wu Yang

Glyphosate (Gly) overuse threatens public health and ecosystems, necessitating highly sensitive detection. This work developed a novel ratiometric fluorescent probe, CDs@Cu-MOF/TMTA-COF, by integrating carbon dots (CDs), metal organic frameworks (MOFs), and covalent organic frameworks (COFs). We encapsulated CDs within Cu-MOF in-situ grown onto sp 2 ‑carbon conjugated TMTA-COF. Gly binding to Cu 2+ causes MOF structural collapse, releasing CDs and enhancing its fluorescence at 425 nm. Simultaneously, Gly inhibits competitive absorption and acceptor-photoinduced electron transfer process between TMTA-COF and Cu-MOF, increasing COF fluorescence at 550 nm. This dual-signal change enables ratiometric Gly detection from 0.2 to 40 μM, with a detection limit of 0.07 μM. This probe was successfully used to measure Gly in soil, Yellow River water and soybean (recoveries: 95.3–104.1 %). Additionally, the back propagation neural network further enables fast detection of Gly. This work demonstrates the potential of MOF/COF hybrids for fluorescence sensing for pesticide analysis and paves the way for intelligent sensing platforms.

Adverse Effects of Glyphosate and Microcystin-LR on Fish Health: Evidence from Structural and Functional Impairments in Zebrafish Gills

Yidan Zhang,Han Hu,Linmei Song,Zhihui Liu,Junguo Ma,Xiaoyu Li

Simple SummaryThe coexistence of two environmental pollutants, glyphosate (GLY, a widely used herbicide) and microcystin-LR (MC-LR, a toxic byproduct from harmful algal blooms) has become an issue that cannot be ignored. The adverse effects of both individual and combined exposures to GLY and MC-LR on aquatic organisms warrant increasing attention. This study demonstrates that exposure to GLY and MC-LR, either individually or in combination, can induce toxic effects on fish gill structure, potentially impairing their physiological function and ultimately threatening fish survival. Overall, the findings of this study not only enhance our understanding of the detrimental impacts of these pollutants on aquatic animals but also provide valuable insights for ecological and health risk assessments.Glyphosate (GLY) and microcystin-LR (MC-LR) frequently co-occur in natural water bodies. In this study, a subacute exposure test was conducted on zebrafish treated with 3.5 mg/L GLY and 35 μg/L MC-LR, individually and in combination, for 21 d to determine their effects on the gills of zebrafish and their potential mechanisms. The hematoxylin and eosin staining and scanning electron microscopy examination results showed that GLY and MC-LR exposure caused structural damage to gills. Biochemical analysis revealed Na+-K+-ATPase activity decreased, and the levels of reactive oxygen species, 8-hydroxy-2′-deoxyguanosine, and malondialdehyde increased, inducing oxidative damage to DNA and lipids of gills. Meanwhile, the inflammatory and immune function of the gill was significantly influenced, as evidenced by the alteration of the expression of tumor necrosis factor-α, interleukin-1β, complement 3, and immunoglobulin M. RNA-seq results revealed that GLY and/or MC-LR treatment induced transcriptional changes in the fish gills, which may affect various biological functions, and the lipid metabolism disruption potentially involved in the aforementioned process. Integrating histopathological, biochemical, and transcriptomic analyses, this study revealed that both individual and combined exposures to GLY and MC-LR had adverse effects on zebrafish gills, with combined exposure appears to result in more pronounced adverse effects, potentially compromising fish health.

Preparation of a β-CD@Fe3O4/KHA/CMC/AM/MAH hydrogel and its slow-release herbicidal properties against glyphosate

Jie Song,Yingnan Zhou,Lijun Chen,Zhiwen Yuan,Yuhua Niu,Xi Li,Liangxian Huang

This study employed β-cyclodextrin (β-CD) for the hydrophilic modification of iron tetraoxide (Fe3O4) magnetic materials. Subsequently, potassium humate (KHA) and carboxymethyl cellulose (CMC) were used as raw materials. A β-CD@Fe3O4/KHA/CMC/AM/MAH hydrogel with magnetic properties which can be easily recycled was synthesized for the slow release of glyphosate (GLY). FTIR, XRD, TG and VSM analyses demonstrated that the hydrogel possesses good thermal stability and magnetic properties with a saturation magnetization strength of 29.77 emu g−1. The equilibrium solubility of the hydrogel in pH = 7.4 and pH = 8.5 buffer was 24.58 g g−1 and 25.83 g g−1, respectively. The maximum release of GLY was recorded at 63.31% and 67.78%, with the drug release behavior under different loading gels following the Fickian diffusion mechanism. A pot planting experiment indicated that the water content of plants treated with the β-CD@Fe3O4/KHA/CMC/AM/MAH drug-loaded gel decreased by 51.69%. Additionally, soluble protein levels decreased to 2.69 mg g−1, chlorophyll content decreased to 0.486 mg g−1, SOD levels decreased to 43.81 U g−1, and MDA increased to 4.02 μmol g−1, effectively inhibiting the growth of grass and providing a theoretical basis for achieving slow drug release.

An on–off–on fluorescent probe for the detection of glyphosate based on a Cu2+-assisted squaraine dye sensor

Shuhua Zhao,Lei Shi,Xiufeng Zhang,Xiaoran Sun,Wenxuan Zhu,Lijia Yu

The herbicide glyphosate, N-(phosphonomethyl)glycine, has been widely used in the past 40 years, and has had many adverse effects on human health. Here, we constructed a convenient “on–off–on” fluorescent platform for detection of glyphosate via Cu2+ modulated squaraine dye fluorescence quenching. The squaraine dye F-0 exhibited strong fluorescence, which could be quenched by the addition of Cu2+. However, the addition of glyphosate restored the fluorescence intensity of F-0 due to the formation of a Cu2+-glyphosate complex. F-0 was utilized as a fluorescent probe for the quantitative detection of glyphosate, with the lowest detection limit of 13.16 nmol L−1. Furthermore, this method demonstrated high selectivity and anti-interference capabilities. The successful monitoring of glyphosate in real samples was achieved using this detection strategy.

Attapulgite-based herbicide particles with smart wetting surface for environmental-safe glyphosate delivery induced by consecutive base-acid stimuli

Nana Jing,Chaowen Chen,Chuang Chen,Xue Yuan,Xueqi Ma,Guopeng Teng,Jia Zhang,Xingjiang Li,Zhengyan Wu

To minimize the pesticide loss and enhance the efficiency of active ingredient utilization, the pH-responsive controlled-release pesticide with easy operation has been widely applied despite for its potential on bothering the non-target organisms while using acid or alkaline alone. To overcome that obstacle, we developed an environmentally safe herbicide particle with a smart wetting surface that can be regulated through consecutive base-acid stimuli to maintain stable pH levels. Herein, attapulgite (ATP) is mixed with glyphosate (Gly) initially and the resultants are coated by suspensions of phenylamine-modified ATP (ph-ATP) and amino silicon oil (ASO), ultimately the outmost polyvinyl pyrrolidone (PVP) are constructed to acquire the smart herbicide (AG@ph-AA@PVP). The base may destroy the outmost PVP membrane, and the later acid treatment turns ph-AA layer gradually from hydrophobic to hydrophilic due to protonation of aniline groups, both of which allow water to invade further and promote Gly release. Therefore, the AG@ph-AA@PVP appears with excellent base-acid sensitivity and controlled-release performance, being demonstrating by the burst Gly release in water and significant weeds control efficiency in pot experiments. The interaction of the materials and the surface wettability were analyzed for explaining the controlled-release mechanism. Besides, AG@ph-AA@PVP possesses high environmental stability and good biosafety. This work develops a simple and feasible technology for environmental-safe and on-demand Gly delivery, assisting for advancement of sustainable and green agriculture.
