Histidine-inspired polyacrylonitrile-based adsorbent with excellent hemocompatibility for the simultaneous removal of bacteria and endotoxin in septic blood

Ziyue Ling,Chunji Jiang,Xianda Liu,Yupei Li,Weifeng Zhao,Changsheng Zhao

Endotoxins (LPS) are components of the cell wall of Gram-negative bacteria that are released when bacterial cell membrane is destroyed, the bacteria and the released LPS have led to serious diseases such as sepsis and septic shock. Histidine functionalized adsorbents have been used to remove LPS, however, the present hemoperfusion adsorbents have drawbacks such as inadequate LPS adsorption capability, high cost of raw material, and so on. Inspired by the structure of histidine and incorporated antibacterial molecules, we synthesize allylamine-modified polyacrylonitrile (PAD)/poly (imidazolium bromide) (PMIBr) composite (PAI) microspheres as hemoperfusion adsorbents. The PAI microspheres show excellent antibacterial ability with high killing efficiency of 98.8% against S. aureus and 95.4% against E. coli . In addition, the adsorption capacity of the microspheres for LPS in PBS is 433.5 EU/g within 3 h, and the LPS concentration could also be reduced from 7.34 EU/mL to below 0.5 EU/mL in the plasma environment. The PAI microspheres exhibit outstanding blood compatibility, antibacterial property and endotoxin removal capability, which have great potential in the treatment of sepsis patients.

Sensitive and selective determination of aloin with highly stable histidine-capped silver nanoclusters based on the inner filter effect

Zhi-feng Cai,Chen-hua Deng,Jiao Wang,Yao Zuo,Jing-long Wu,Xiao-peng Wang,Tian-zhi Lv,Yuan-yuan Wang,Deng-yun Feng,Jie Zhao,Cai-feng Zhang,Ji-ming Zhang

Aloin is a natural medicine, which possesses the functions of enhancing immune function, anti-tumour, detoxification, anti-bacterial, anti-stomach damage, liver protection and skin protection. Excessive use will cause side effects. Thus, sensitive aloin determination is very necessary in daily life. As far as we know, there are rare reports about aloin sensing through fluorescent methods. In this work, a simple method for the synthesis of fluorescence silver nanoclusters (Ag NCs) was demonstrated by using histidine (His) as the templating agent and ascorbic acid as the reducer. The as-prepared silver nanoclusters (Ag NCs@His) possessed excellent water solubility and dispersion. Detailed research illustrated that the FL intensities of Ag NCs@His were linearly quenched in the presence of aloin (varying from 0.5 to 200 µM). The corresponding detection limit reached 0.052 µM. The static quenching and inner filter effect were the main fluorescence quenching mechanisms. This fluorescent sensor has been successfully employed for the detection of aloin in natural samples.

LAPONITE® nanoplatform functionalized with histidine modified oligomeric hyaluronic acid as an effective vehicle for the anticancer drug methotrexate

Jinyu Li,Yue Yang,Yibin Yu,Qi Li,Guoxin Tan,Yuanyuan Wang,Wei Liu,Weisan Pan

The synthetic clay material, LAPONITE® (LAP), having a nanodisk structure together with a negatively charged surface, has been used for effective drug encapsulation by virtue of its interlayer space. In this research effort, the LAP nanodisk was used for the first time to encapsulate the antifolic methotrexate (MTX); the MTX-loaded LAP nanodisks (LAP/MTX) demonstrated a high drug loading efficiency of 80.39%. An efficient and reliable tumor-targeting device that rests on the synthesized oligomeric hyaluronic acid-L-histidine (oHA–His) was then encapsulated in the MTX-loaded LAP disks (forming LAP/MTX/oHA–His nanohybrids). The drug released from the LAP/MTX/oHA–His nanohybrids was pH-dependent and matched the first-order kinetics that describes the diffusion mechanism. In vitro biological evaluation manifested that the MTX-loaded LAP nanocarriers, particularly the LAP/MTX/oHA–His nanohybrids that have targetability and lysosomal antineoplastic activity, can be effectively internalized by the MCF-7 cell line, and can exhibit a more prominent anticancer cytotoxicity than free MTX. In vivo studies with mice indicated that the LAP/MTX/oHA–His nanohybrids demonstrated much higher antitumor efficiency compared to the LAP/MTX nanohybrids and pure MTX. Taken together, the LAP/oHA–His, CD44 receptor targeting and pH-sensitive multifunctional nanohybrids conferred the MTX with excellent cytocompatibility, dispersion stability, sustained pH-responsive release properties, and improved anticancer activity, and may be further developed as a potential active nanoplatform for various anticancer drugs.

Sensitive detection of biothiols and histidine based on the recovered fluorescence of the carbon quantum dots–Hg(II) system

Juan Hou,Fengshuang Zhang,Xu Yan,Long Wang,Jin Yan,Hong Ding,Lan Ding

In this paper, we presented a novel, rapid and highly sensitive sensor for glutathione (GSH), cysteine (Cys) and histidine (His) based on the recovered fluorescence of the carbon quantum dots (CQDs)–Hg(II) system. The CQDs were synthesized by microwave-assisted approach in one pot according to our previous report. The fluorescence of CQDs could be quenched in the presence of Hg(II) due to the coordination occurring between Hg(II) and functional groups on the surface of CQDs. Subsequently, the fluorescence of the CQDs–Hg(II) system was recovered gradually with the addition of GSH, Cys or His due to their stronger affinity with Hg(II). A good linear relationship was obtained from 0.10 to 20   μmol   L −1 for GSH, from 0.20 to 45   μmol   L −1 for Cys and from 0.50 to 60   μmol   L −1 for His, respectively. This method has been successfully applied to the trace detection of GSH, Cys or His in human serum samples with satisfactory results. The proposed method was simple in design and fast in operation, which demonstrated great potential in bio-sensing fields.

l-histidine functionalized multi-walled carbon nanotubes for on-line affinity separation and purification of immunoglobulin G in serum

Zhuo Du,Suling Zhang,Chanyuan Zhou,Miao Liu,Gongke Li

In this work, the multi-walled carbon nanotubes were covalently functionalized with l -histidine (His–MWNTs) as online pseudospecific affinity adsorbent for immunoglobulin G (IgG) separation and purification with a simple surface modification method, using 1-ethyl-3-(3-dimethyaminopropyl) carbodiimide hydrochloride (EDC) and N-hydroxysuccinimde (NHS). The affinity of the His–MWNTs toward IgG was investigated in a microcolumn incorporated into a sequential injection system, which also involves an UV spectrometer with a flow cell for online real-time detection. The incorporation of histidine as affinity groups noticeably increased the selectivity and binding capacity of MWNTs for IgG and the His–MWNTs exhibited high retention and recovery rate of nearly 100% under optimized conditions. This separation and enrichment process made it possible to determine a lower concentration range of IgG in serum from 1.0–33   μg/mL with a detection limit of 0.3   μg/mL with a sampling volume of 4.0   mL. The static and dynamic adsorption capacities obtained were 267   mg of IgG/g His–MWNTs and 35   mg/g in aqueous solution, respectively, which are among the highest reported results in literatures employing affinity separation methods. Desorption of IgG from His–MWNTs could be accomplished by lowering the pH to 1.5 with glycine–HCl buffer. The practical application of His–MWNTs for separation of IgG in serum was evaluated with sodium dodecyl sulfate-polyacrylamide gel electrophoresis which confirmed that the purity of recovered IgG from human serum was over 85% and better than a commercial product.

A novel fluorescence and colorimetric dual sensing system for rapid and sensitive detection of histidine based on TSPP-CA

Dajie Zou,Chenyan Lu,Yanyan Jia,Huan Wang,Jiuli Wang

Histidine (His) plays an important role in human health and life activities. It will harm human health when His intake is insufficient or excessive. Its residue also will pollute the natural environment. Therefore, establishing a reliable and sensitive method for detecting histidine is particularly important. However, most of the existing detection methods of His rely on the single change of single signal, which are susceptible to interference from testing environmental factors and prone to generating false positive results. In contrast, the fluorescence and colorimetric dual-signal sensing system can not only effectively improve the reliability of detection, but also significantly reduce the risk of false positives. Therefore, the dual-signal sensing system has gradually become the research hotspot. Based on this, 5,10,15,20-tetra-(4-sulfophenyl) porphyrin (TSPP) was selected as the fluorescence and colorimetry dual-signal probe for the rapid detection of His in this study. Since TSPP could interact with many substances due to the specificity of molecular structure, it is the best choice to build an “ON-OFF-ON” sensing system in order to improve the specificity of the sensing system. Therefore, citric acid (CA) as an intermediate based on TSPP probe was successfully developed fluorescence and colorimetric dual-sensing system for the quantitative detection of histidine in real samples. The signal of the dual sensing system was “turned on” when the red TSPP solution obtained the fluorescence emission wavelength of 642 nm at the 514 nm optimal excitation wavelength and the UV–Vis absorption at 413 nm, respectively. The fluorescence intensity and absorbance of TSPP gradually decreased with the introduction of CA. At this time, the signal of the dual-sensing system became “turned off”, and the color of the solution changed from light pink to light green. The quenched fluorescence intensity and absorbance, however, was restored upon the introduction of histidine into the system. Simultaneously, the color of the solution changed from light green to light pink, and the dual-sensing system became “turned on”. Therefore, a novel fluorescence and colorimetric dual-signal sensing system based on TSPP was proposed for histidine detection. The results indicated that the linear ranges of the fluorescence sensing system were 8.34 × 10 −6  M − 1.51 × 10 −4  M and 1.85 × 10 −4  M − 1.4 × 10 −3  M with detection limits of 0.282 μM and 10.91 μM (LOD, S/N = 3), respectively. The linear range of the colorimetric sensing system was 2.04 × 10 −4  M–4.35 × 10 −4  M with a detection limit of 11.97 μM (LOD, S/N = 3). Meanwhile, the dual-sensing system provided a promising platform for practical samples sensing applications.

Dual-Shell-Solidified Gold Nanoclusters-Based Electrochemiluminescence Sensing Platform for Highly Selective and Sensitive Detection of Cu2+ and Histidine

Faisal Ahmad,Yelan Xiao,Tong Shu,Guangyao Zhang,Xueji Zhang

Electrochemiluminescence (ECL) is rapidly emerging as an excellent electrochemical analytical technique for the specific and sensitive detection of various biomarkers and hazardous trace metals. Among ECL emitters, gold nanoclusters (AuNCs) have proven to be excellent luminophores due to their remarkable luminescent properties, stability, and biocompatibility. However, the low ECL efficiency of AuNCs precludes their application in ultrasensitive biosensing. One of the key reasons for the low ECL efficiency of AuNCs is the nonradiative energy transitions arising from intramolecular vibrations and rotations of the ligands on the surface of AuNCs. Herein, we explore the ECL of Zn2+/Au22(SG)18/PDA nanoclusters (NCs), where Zn2+ and 2,6-pyridinedicarboxaldehyde (PDA) activate the restriction of intramolecular motion (RIM) through aggregation-induced emission (AIE) to aggregation-induced enhanced emission (AIEE). This effect is achieved via cross-linking of Zn2+ and PDA with glutathione on the surface of Au22(SG)18, resulting in a significant enhancement of ECL emission compared to Au22(SG)18 NCs alone. A sensitive ECL sensing system was devised with Zn2+/Au22(SG)18/PDA as the ECL emitter, with triethylamine (TEA) as the coreactant, for the detection of Cu2+ and histidine in the linear ranges of 0.1 to 40 μM and 1 to 85 μM, respectively, with detection limits of 0.705 and 1.928 μM, respectively.

Copper(II) Complex of Graphene Oxide for Highly Selective and Sensitive Fluorescence Sensing of Histidine in Water and Milk

Yijun Yu,Haibiao Chen,Chen Zhang,Jie He,Linling Li,Chao Teng

In this work, a good water-dispersible GO-Cu 2+ -1, 8-diaminonaphthalene complex (GCU) was synthesized from graphene oxide (GO), CuCl 2 , and 1, 8-diaminonaphthalene in mild condition. The GCU achieved histidine detection with high selectivity and high sensitivity. Upon addition of histidine, the fluorescence intensity of GCU at 410 nm increased significantly due to the blocked photoinduced electron transfer (PET) process. The linear concentrations range of histidine detection was from 0.1 to 4.0 µM, with a limit of detection (LOD) of 0.027 µM. Common amino acids and proteins do not affect the detection of histidine by GCU in water. Furthermore, the GCU has been successfully applied not only to detect histidine in water but also to measure histidine in milk. The fluorescence sensor-based assay using GCU represents a promising method for nutritional monitoring. Graphical A highly water-dispersible chemosensor GCU was synthesized from GO, CuCl 2 , and 1,8-diaminonaphthalene. The nanostructured ternary complex exhibits weak fluorescence at 410 nm due to the PET mechanism. It responds to histidine in aqueous solutions with a limit of detection (LOD) of 0.027 µM. Additionally, it can detect histidine in real milk samples, exhibiting promising application potential.

Synthesis of a histidine-magnesium hydroxide curing agent and improving the flame retardant, smoke suppression of epoxy resin composites

Qibin Yan,Yunfan Li,Jiaheng Zhao,Zhiwang Yang,Ziqiang Lei

Conventional curing agents typically possess a single property and the challenge of inadequate interfacial adhesion between inorganic flame-retardant additives and the epoxy resin (EP). This study presents a multifunctional flame retardant curing agent (HFP-MM) synthesized by functionalizing magnesium hydroxide (MH) with L-histidine (HIS), phosphite, and melamine (MEL). This curing agent was compounded with varying amounts (wt%) of Ammonium polyphosphate (APP) and added to EP to establish an intumescent flame retardant system. The results indicate that EP formulation containing 10 wt% HFP-MM and 10 wt% APP achieved UL-94 V-0 level and exhibited the limiting oxygen index (LOI) measurement reaching 29.6 %. Furthermore, compared to the flame retardant free of EP, the peak heat release rate, peak CO production rate and total smoke production were dropped by 88.9 %, 86.0 % and 74.8 %. Regarding flexural performance, the flexural modulus and flexural strength increased by 129.6 % and 137.4 %, respectively. The functionalized MH flame-retardant curing agent prepared in this study, when applied to EP, exhibits excellent flame-retardant and mechanical properties, proving effective in reducing the fire hazard of EP materials.

A highly sensitive luminescent probe based on Ru(II)-bipyridine complex for Cu2+, l-Histidine detection and cellular imaging

Shi-Ting Zhang,Panpan Li,Caiyun Liao,Tingting Luo,Xingming Kou,Dan Xiao

A ruthenium(II) bipyridyl complex conjugated with functionalized Schiff base (RuA) has been synthesized and functioned as a luminescent probe. The luminescence of RuA was greatly quenched by Cu 2+ due to its molecular coordination with paramagnetic Cu 2+ . Subsequently, the addition of l -Histidine can turn on the luminescence of the RuA-Cu(II) ensemble, which can be attributed to the replacement of RuA in RuA-Cu(II) ensemble by l -Histidine. On the basis of the quenching and recovery of the luminescence of RuA, we proposed a rapid and highly sensitive on-off-on luminescent assay for sensing Cu 2+ and l -Histidine in aqueous solution. Under the optimal conditions, Cu 2+ and l -Histidine can be detected in the concentration range of 5 nM–9.0 μM and 50 nM–30 μM, respectively, and the corresponding detection limits were calculated to be 0.35 and 0.44 nM (S/N=3), separately. The proposed luminescent probe has been successfully utilized for the analysis of Cu 2+ and l -Histidine in real samples (drinking water and biological fluids). Furthermore, the probe revealed good photostability, low cytotoxicity and excellent permeability, making it a suitable candidate for cell imaging and labeling in vitro.

Surface-Exposing Nickel Ion-Based Ni/NixPy Magnetic Microspheres for Stable and Efficient Separation and Purification of Histidine-Rich Proteins

Cong Xiang,Fan Xue,Shunzong Wu,Juan Wang,Chenyu Yang,Yike Huang,Xiangzi Li,Meifang Wang,Xiaomei Cheng

Histidine-rich proteins (His-proteins) are vital biomolecules with diverse physiological roles. However, their purification from complex biological samples remains challenging. Here, we successfully prepared Ni/NixPy composite nanospheres with different phosphating degrees, featuring surface-exposing Ni2+ properties for efficient separation and purification of His-proteins. The phosphated products exhibited a spherical morphology, reduced cytotoxicity, and good biocompatibility. The optimal Ni/NixPy demonstrated excellent adsorption and separation capacity for His-tagged bovine hemoglobin (BHb), which is attributed to their Ni2+ surface-exposing capability and magnetic properties, achieving a adsorption capacity of 2130.4 mg/g under the tested conditions. While the adsorption sites for His-proteins are enhanced, the separation process is simplified. SDS-PAGE analysis confirmed the selective adsorption of Ni/NixPy for His-proteins in a BSA/BHb mixed solution. The Ni/NixPy spheres demonstrated good stability and recyclability, maintaining an 85% adsorption efficiency after five cycles. In addition, His-proteins can be efficiently separated and purified from complex fetal bovine serum. Therefore, this work reports an effective surface-exposing Ni2+ approach to enhance the efficiency of protein separation and purification with the potential to expand applications in diagnostics and therapeutics.

Yolk-shell composite optical sensors with chiral L-histidine/Rhodamine 6G for high-sensitivity “turn-on” detection of L-proline

Yu-Han Geng,Yu Xin,Jing Du,Ming-Yi Cui,Yuan-Yuan Liu,Le-Xi Zhang,Bin Ding

Chirality is a ubiquitous phenomenon in nature and has attracted wide attention in the biomedicine, pharmaceutics and biosensing research fields. Enantiomeric recognition of chiral compounds, especially chiral drugs and chiral amino acids, is important for human health and nutrition. In this work, through the encapsulation of L-His&R6G (L-His = L-Histidine; R6G = Rhodamine 6G) into MOF@MOF framework ZIF-67@ZIF-8, composited material L-His&R6G@ZIF-67@ZIF-8 can be obtained. Additionally, through the etching process, a unique yolk-shell ZIF-8 chiral composite optical sensors L-His&R6G@ZIF-8 ( 1 ) can be successfully prepared. Photo-luminescent (PL) experiment also reveals that 1 can highly sensitively detect L-Proline (L-Pro) through the “turn-on” detection strategy (K BH = 1.22 × 10 4 M −1 and detection limit 1.9 μM). Further yolk-shell L-His&R6G@ZIF-8-based fabricate flexible mixed-matrix membranes has been prepared using doctor-blading technique, which show significant fluorescence enhancement effect under ultraviolet lamp. This work also provides the unique example of preparing chiral yolk-shell framework composite sensors, which have broad application in chiral sensing area.
