D-α-tocopherol polyethylene glycol 1000 succinate-based microemulsion delivery system: Stability enhancement of physicochemical properties of luteolin

Yimei Zheng,Chengang Zhao,Boyu Chen,Hui Teng,Chao Ai,Lei Chen

In the present study, D-α-Tocopherol polyethylene glycol 1000 succinate-based self-microemulsifying drug delivery systems (TPGS-SMEDDS) were introduced to enhance the solubility and stability of luteolin. The ternary phase diagrams were constructed to obtain the maximum area of microemulsion and suitable formulations of TPGS-SMEDDS. The particle size distribution and polydispersity index of selected TPGS-SMEDDS were analyzed to be less than 100 nm and 0.4, respectively. The thermodynamic stability results suggested that the TPGS-SMEDDS was stable during the heat-cool and freeze–thaw cycle. Moreover, the TPGS-SMEDDS exhibited excellent encapsulation capacity (51.21 ± 4.39 to 85.71 ± 2.40%) and loading efficiency (61.46 ± 5.27 to 102.86 ± 2.88 mg/g) to luteolin. In addition, the TPGS-SMEDDS showed an admirable vitro release ability with a ratio of more than 88.40 ± 1.14% for luteolin in 24 h. Therefore, TPGS-based SMEDDS might provide an effective role for the oral administration of luteolin and holds promise as a potential delivery for poorly soluble bioactive compounds.

Synthesis of metal-organic framework hybrid nanocomposites based on MOFs@C3N4 with high selective separation ability for luteolin

Jingjing Zhen,Faguang Ma,Jing Yan,Rongxin Lin,Ming Yan,Yilin Wu

Complex substrates found in certain extracts-particularly those containing high phenolic compounds with significant medicinal value-pose considerable challenges to developing molecularly imprinted membranes (MIMs) with specific recognition abilities. In this study, UiO-66-NH 2 , a metal–organic structure (MOF), was chosen as an optimizer to enhance the basic support material C 3 N 4 . A composite imprinted membrane based on MOFs was then prepared for the targeted adsorption of luteolin (LTL). This not only increases the surface area and pore capacity of the composite membranes, but also ensures unprecedented control over the membranes structure and good stability. To maximize the proportion of imprinted sites, we utilize an advanced boron affinity sol–gel imprinting technique that capitalizes on the exceptional response of the bifunctional monomer. By finely manipulating the imprinting process, controlled distribution of imprinted sites was achieved. Furthermore, the incorporation of Ag nanoparticles into the composite matrix of MIMs enhances selectivity and efficiency due to their plasmonic properties and high affinity for phenolic compounds, providing increased adsorption surface area and additional binding sites. The results showed satisfactory rebinding ability (56.25 mg g −1 ) and superselective ( β AG/LTL  = 4.43 and β CT/LTL  = 8.49). It was worth noting that incorporating Ag nanoparticles into composite MIMs for the selective adsorption of LTL demonstrates significant potential. This includes enhanced selectivity, efficiency, and hydrophilicity of the membrane surface. Thus, enhanced hydrophilicity can establish stronger non-covalent interactions, namely hydrogen bonding and van der waals forces, with the target molecules, thereby significantly improving the adsorption capacity of the MIMs. The feasibility of this method was demonstrated via in-situ infrared analysis of the chemical structure changes before and after adsorption.

Fabrication of chitosan/phenylboronic acid/SiO2 hydrogel composite silk fabrics for enhanced adsorption and controllable release on luteolin

Xuemei He,Haiyan Mao,Shuzhen Wang,Zhongliang Tian,Tianchi Zhou,Lu Cai

Due to the growing demand for self-health and safety, eco-friendly health textile products with natural colors and pharmacological functionalities have gained considerable popularity. Rapid adsorption and controlled release of active molecules are important issues for functional health textiles. In this study, a functionalized chitosan-based hydrogel composite silk fabric was prepared using chitosan, 3-carboxyphenylboronic acid, and 3-(2, 3-epoxypropyl oxygen) propyl silane by dip-pad and vacuum freeze-drying techniques. The results showed that the incorporation of chitosan/phenylboronic/SiO 2 hydrogel into silk fibers improved the UV protection capacity, mechanical properties, and adsorption properties of silk fabrics. The effects of various parameters on the luteolin adsorption properties of silk fabrics were discussed, including metal salt types, salt dosage, pH value, dyeing temperature, initial luteolin concentration, and dyeing time. Under the dyeing temperature of 60 °C and pH of 6.8, the luteolin exhaustion of the composite silk was more than that of the untreated silk, and the adsorption process followed the quasi-second-order kinetic model and the Langmuir adsorption isotherm model. Furthermore, the luteolin-dyed composite silk materials exhibited strong antioxidant activity and controllable release behavior with various pH levels. The as-prepared chitosan-hydrogel composite silk could be a promising material for the sustained release of drugs in medical and healthcare textiles.

Investigating of zein-gum arabic-tea polyphenols ternary complex nanoparticles for luteolin encapsulation: Fabrication, characterization, and functional performance

Renjie Li,Zhuangwei Zhang,Jin Chen,Huihui Li,Hongjin Tang

Luteolin has extensive biological effects, but its low water-solubility and oral bioavailability have restricted its application. In this study, we successfully prepared new zein-gum arabic (GA)-tea polyphenols (TP) ternary complex nanoparticles (ZGTL) as a delivery system to encapsulate luteolin using an anti-solvent precipitation method. Consequently, ZGTL nanoparticles showed negatively charged smooth spherical structures with smaller particle size and higher encapsulation ability. X-ray diffraction revealed the amorphous state of luteolin in the nanoparticles. Hydrophobic, electrostatic, and hydrogen bonding interactions contributed to the formation and stability of ZGTL nanoparticles, as indicated by fluorescence and Fourier transform infrared spectra analyses. The inclusion of TP improved the physicochemical stability and luteolin retention rate of ZGTL nanoparticles by forming more compact nanostructures under different environmental conditions, including pH, salt ion concentration, temperature, and storage. Additionally, ZGTL nanoparticles exhibited stronger antioxidant activity and better sustainable release capacity under simulated gastrointestinal conditions due to TP incorporation. These findings demonstrate that ZGT complex nanoparticles have potential applications as an effective delivery system for encapsulating bioactive substances in food and medicine fields.

Surface Pretreatment with Luteolin to Achieve the Effective Flotation Separation of Sphalerite from Pyrite in a CuSO4-Xanthate System

Jiaxiu Chen,Lv Zhao,Haiyang He,Jinpeng Cai,Liuyang Dong,Dianwen Liu

The flotation separation of sphalerite (ZnS) and pyrite (FeS2) in a copper sulfate (CuSO4)–butyl xanthate (BX) system is challenging because the indiscriminate activation of Cu2+gives them similar floatability. To this end, the suitability of a novel depressant, luteolin (LUT), on the flotation separation of ZnS from FeS2in this system was studied. Micro-flotation tests and contact angle measurement results showed that ZnS could be selectively separated from FeS2using LUT as a depressant. At pH 7.0 ± 0.1, a flotation concentrate with a recovery of 93.90% ZnS and 2.84% FeS2was obtained after adding LUT in artificial mixed minerals tests. Adsorption differences and characteristics results showed that the affinity of LUT to FeS2was considerably higher than that of ZnS, which effectively hindered the subsequent adsorption of xanthate on FeS2but had a relatively weak effect on ZnS. Thus, a large floatability difference between ZnS and FeS2was achieved. Further analyses revealed that the adsorption behavior of LUT on FeS2occurred mainly through the interaction between 4–5 sites and Fe(II) at the solid/liquid interface. These findings shed light for the first time on the suitability of LUT for the successful flotation separation of ZnS from FeS2, which has important guiding significance for the development of a lime-free depressant of FeS2.

Multifunctional modification of bio–based polyamide 56 fabric via luteolin for durable colouristic, antioxidant, antibacterial and UV protective properties

Xue Zhang,Miaoyan Li,Xin Zhang,Xueni Hou,Xiangrong Wang

Natural extracts, characterized by their superior biocompatibility and multifunctionality, have demonstrated remarkable potential in fields such as antibacterial, antioxidant and ultraviolet (UV) protection. However, at present, natural extracts are facing a key bottleneck in functional finishing—poor washing durability. In this study, luteolin, a natural extract, was chosen to treat bio-based Polyamide 56 (PA56) fabrics. The findings indicated that the fabric dyed with luteolin not only achieved good coloration but also exhibited outstanding multi-functional properties: the ultraviolet protection factor (UPF) value reached 205, the ABTS radical cation (ABTS •+ ) scavenging rate was 99 %, and the antibacterial rates against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) were 70 % and 95 %, respectively. To further enhance the color fastness and functional durability, this study introduced metal ions. Through the coordination reaction between luteolin, metal ions and PA56, the color fastness was successfully raised to level 4 or above, and the durability of functionalities was significantly improved. After 10 washing cycles, the UPF value of the fabric remained at 155, the ABTS •+ scavenging rate exceeded 60 %, and the antibacterial rate against S. aureus remained at 75 %. This study proposed and verified a sustainable and coordinated strategy based on coordination chemistry, systematically addressing the durability issue of natural functional finishing agents.

Luteolin adsorption-oxidative desorption dual-effect regulation: an innovative strategy for inhibition and re-activation of copper-activated pyrite

Chen Jin,Jiaxiu Chen,Guohui Wang,Yiqiang Liang,Jinpeng Cai,Peilun Shen,Dianwen Liu

Luteolin effectively depresses copper-activated pyrite for selective Zn–S separation. • H2O2 reactivates LUT-depressed pyrite by inducing LUT desorption. • H2O2-induced LUT desorption occurs via direct molecular oxidation and indirect Fe2+/Fe3 + modulation. Organic depressants have been extensively investigated for pyrite depression under low-alkali conditions due to their abundance and environmental compatibility. However, studies on the de-depression and reactivation of depressed pyrite remain limited. This research proposes a solution wherein luteolin (LUT) depresses pyrite in a copper potassium butyl xanthate (PBX) system, and oxidative treatment with H 2 O 2 enables its desorption and reactivation. Flotation tests demonstrated a dramatic recovery of depressed pyrite from 1.05% to 96.95% post-H 2 O 2 treatment, with successful application in recovering pyrite from tailings during zinc separation in artificial mixed ore tests. Mechanistic investigations via contact angle measurements, Zeta potential analysis, UV–Vis, FTIR spectroscopy, ToF-SIMS and AFM revealed that H 2 O 2 induces LUT desorption from pyrite surface at natural pH, facilitating PBX adsorption to form a hydrophobic layer. Raman spectroscopy suggested oxidative modifications to LUT’s molecular structure. Additionally, XPS analysis confirmed increased Fe 3+ content on H 2 O 2 -treated pyrite, while LUT exhibits weaker interactions with Fe 3+ than with Fe 2+ revealed by microcalorimetry analysis. Consequently, H 2 O 2 reduces LUT adsorption, triggering its desorption and restoring pyrite’s flotation response. This work offers a practical strategy for reactivating pyrite inhibited by organic depressants, which is of great significance for promoting the efficient flotation separation of sulfide ore. A glucose-responsive hydrogel delivering gallium-luteolin nanocomplexes for infected diabetic wound repair via antibacterial and oxidative-inflammatory regulation

Nuo Li,Zhenjing Xing,Sheng Jiang,Shi Xu,Chenying Shen,Chengyue Jiang,Yuyang Li,Mingyue Cheng,Shuangshuang Ren,Can Xiao,Leiying Miao,Chao Liu

GL@PGB enabled glucose-responsive local delivery of gallium–luteolin nanocomplexes. • GL NCs acted as releasable coordination nanounits for antibacterial and oxidative-inflammatory regulation. • GL@PGB inhibited bacterial growth and disrupted established biofilms. • GL@PGB reduced intracellular ROS accumulation and favored macrophage transition toward a reparative phenotype. • GL@PGB reshaped the infection–ROS–inflammation microenvironment to improve infected diabetic wound repair. Diabetic wounds are frequently complicated by persistent bacterial infection, excessive reactive oxygen species (ROS) accumulation, and dysregulated inflammation, which collectively reinforce a hostile microenvironment that delays tissue repair. Herein, we report a glucose-responsive hydrogel platform (GL@PGB) for local delivery of gallium-luteolin nanocomplexes (GL NCs), designed to couple Ga-associated antibacterial metal-homeostasis perturbation with luteolin-associated oxidative-inflammatory regulation. In this system, the PVA/guar gum/borax (PGB) hydrogel functions as a hemostatic and wound-adaptive delivery matrix, enabling local retention and glucose-accelerated release of GL NCs under hyperglycemic conditions. The released GL NCs suppressed bacterial growth and disrupted established biofilms, and transcriptomic analysis suggested perturbation of metal-ion homeostasis and genetic information-processing pathways in S. aureus . Concurrently, GL@PGB reduced intracellular ROS accumulation and promoted macrophage polarization toward a reparative phenotype in vitro. In an S. aureus -infected diabetic full-thickness wound model, GL@PGB reduced bacterial burden, alleviated inflammatory responses, and promoted collagen deposition and angiogenesis, thereby improving wound repair. These results suggest that GL@PGB may serve as a glucose-responsive local delivery system for disrupting bacterial metal homeostasis while reshaping the infection-ROS-inflammation microenvironment. Luteolin Stabilized in Nanosheet and Cubic γ-Cyclodextrin-Based Metal Organic Framework for Enhanced Bioavailability and Anti-inflammatory Therapy

Mubarak G. Bello,Shiyu Huang,Zifan Qiao,Zhenhua Chen,Lihua Chen

Luteolin (LT), a biologically active flavonoid with potent anti-inflammatory properties, faces significant limitations due to its instability, poor absorption, and low bioavailability. To overcome these challenges, this study explores the encapsulation of LT within nanosheet and cubic γ-cyclodextrin-based metal-organic frameworks (N-MOF and CD-MOF). This approach achieved a two-fold improvement in LT loading efficiency compared to previously reported methods. A critical finding was the identification of pH as a key factor in optimizing the loading process, which significantly enhanced encapsulation efficiency. The resulting LT/N-MOF and LT/CD-MOF formulations demonstrated improved photo-stability and thermal stability, alongside significantly enhanced cellular permeation. In vitro studies revealed remarkable anti-inflammatory properties of these formulations. Moreover, the bioavailability of LT was increased by 6-fold compared to pure luteolin. In a rat paw edema model, the optimized LT/CD-MOF and LT/N-MOF formulations exhibited 4.86- and 5.4-fold improvements in anti-inflammatory activity, respectively, over pure luteolin. These results underscore the potential of CD-MOFs as effective delivery systems for enhancing the therapeutic efficacy of luteolin.

Luteolin attenuates experimental dry eye disease via dual modulation of macrophage JAK-STAT signaling and corneal epithelial MAPK pathways

Zhirui Du,Xueqiao Yan,Lin Cheng,Haiyu Shen,Guoguo Zhi,Baojie Li,Qian Yu

Background Dry eye disease (DED) is a multifactorial ocular condition marked by tear film instability, inflammation, and oxidative stress. Current therapies, including artificial tears and immunosuppressants, offer limited long-term benefit and potential side effects. Natural flavonoids such as luteolin possess anti-inflammatory and antioxidant properties, yet their therapeutic potential in DED remains underexplored. Purpose This study aimed to evaluate the therapeutic efficacy and mechanisms of luteolin in experimental DED. Methods Flavonoids were initially screened using lipopolysaccharide-stimulated RAW264.7 macrophages. A benzalkonium chloride-induced DED mouse model was treated with topical luteolin or cyclosporine A. Ocular surface evaluations included tear secretion, corneal fluorescein staining, and goblet cell density. Inflammatory and oxidative markers were examined via histology, immunofluorescence, and qPCR. RNA sequencing and network pharmacology were used to identify target pathways. In vitro studies with RAW264.7 cells and hyperosmolarity-stressed human corneal epithelial cells further validated mechanism-specific effects. Target engagement was further validated using pathway-specific pharmacological inhibitors. Ocular safety and pharmacokinetics were also assessed. Results Luteolin showed superior anti-inflammatory activity among tested flavonoids. It improved tear production, reduced epithelial damage, and preserved goblet cells. Mechanistically, it modulated macrophage-driven inflammation through JAK-STAT signaling and mitigated oxidative epithelial damage by suppressing the MAPK pathway. Transcriptomic and pharmacological analyses confirmed its dual immunomodulatory and cytoprotective roles. Combined STAT1/3 and MAPK inhibition mimicked its therapeutic effects. Topical luteolin was well-tolerated and achieved corneal bioavailability. Conclusions Luteolin provides a dual protective effect in DED by targeting inflammation and oxidative stress, supporting its development as a safe and effective disease-modifying therapy.

Coral-like cobalt/nitrogen-doped porous carbon synergized with MnO₂ for ultrasensitive electrochemical detection of luteolin in floral teas

Yafeng Jin,Fangming Han,Didi Tong,Xinyue Song,Di Wang,Tao Wang,Xiangquan Qin,Na Chen,Xiaobo Li,Guangri Xu

High-performance electrode materials are crucial for electrochemical sensing of bioactive food compounds. We report an eco-friendly synthesis of coral-like cobalt/nitrogen-doped porous carbon (Co/N-PC) via hydrothermal-pyrolysis, integrated with MnO₂ to construct a Co/N-PC@MnO₂/GCE sensor for ultrasensitive luteolin (LU) detection. Synergy between the conductive Co/N-PC framework (electron transport) and MnO₂ nanoparticles (Mn 2+ /Mn 3+ /Mn 4+ catalysis) delivers exceptional performance: dual linear ranges (0.01–1 μM; 1–100 μM) and a record-low 4 nM detection limit. Kinetic studies confirm the apparent electron transfer number (n app  = 4) arises from MnO₂ mediation. The sensor demonstrated high practicality in complex floral teas (honeysuckle, chrysanthemum), achieving 77.2–103.6 % recoveries. This work provides both a sensitive, green LU detection method and elucidates a MnO₂ catalytic cycle mechanism, revealing new insights into LU's electrochemical pathway.

Fabrication of the Rapid Self-Assembly Hydrogels Loaded with Luteolin: Their Structural Characteristics and Protection Effect on Ulcerative Colitis

Xin Bi,Han Peng,Hua Xiong,Lihua Xiao,Hua Zhang,Jiang Li,Yong Sun

Luteolin (LUT) is a fat-soluble flavonoid known for its strong antioxidant and anti-inflammatory properties. Nonetheless, its use in the food industry has been limited due to its low water solubility and bioavailability. In this study, hyaluronic acid, histidine, and luteolin were self-assembled to construct tubular network hydrogels (HHL) to improve the gastrointestinal stability, bioavailability, and stimulation response of LUT. As anticipated, the HHL hydrogel’s mechanical strength and adhesion allow it to withstand the challenging gastrointestinal environment and effectively extend the duration of drug presence in the body. In vivo anti-inflammatory experiments showed that HHL hydrogel could successfully alleviate colitis induced by dextran sulfate sodium (DSS) in mice by reducing intestinal inflammation and restoring the integrity of the intestinal barrier. Moreover, HHL hydrogel also regulated the intestinal microorganisms of mice and promoted the production of short-chain fatty acids. The HHL hydrogel group demonstrated a notably superior treatment effect compared to the LUT group alone. The hydrogel delivery system is a novel method to improve the absorption of LUT, increasing its bioavailability and enhancing its pharmaceutical effects.
