Development and characterization of electrospun nanofibers based on pullulan/chitin nanofibers containing curcumin and anthocyanins for active-intelligent food packaging

Mengxia Duan,Shan Yu,Jishuai Sun,Haixin Jiang,Jianbo Zhao,Cailing Tong,Yaqin Hu,Jie Pang,Chunhua Wu

An electrospun nanofiber based on pullulan/chitin nanofibers (PCN) containing curcumin (CR) and anthocyanins (ATH) was developed using an electrospinning technique for active-intelligent food packaging. The results of scanning electron microscopy and attenuated total reflection Fourier transform infrared spectroscopy indicated that CR and ATH were successfully immobilized on the film-forming substrate based on PCN. The physical and chemical properties of nanofibers with no colorant, a single colorant, and double colorants were compared. The nanofiber containing ATH and CR (PCN/CR/ATH) had stronger antioxidant and antimicrobial activities than those of nanofibers containing CR (PCN/CR) or ATH (PCN/ATH). With respect to pH sensitivity, the color of the PCN/CR nanofibers did not change obviously, but the color of the PCN/ATH and PCN/CR/ATH nanofibers changed significantly with the change in pH. Furthermore, the PCN/CR/ATH nanofibers clearly changed color with the progressive spoilage of Plectorhynchus cinctus at room temperature. Therefore, the electrospun PCN/CR/ATH nanofiber have great application potential in active–intelligent food packaging.

Synergistically Enhanced Inhibitory Effects of Pullulan Nanoparticle-Mediated Co-Delivery of Lovastatin and Doxorubicin to Triple-Negative Breast Cancer Cells

Wu Di,Chen Yao,Wen Shun,Wen Yi,Wang Rong,Zhang Qiuting,Qin Ge,Yi Huimei,Wu Mi,Lu Lu,Tao Xiaojun,Deng Xiyun

Triple-negative breast cancer (TNBC) is a subtype of breast cancer that is prone to drug resistance and difficult to treat. In this study, we grafted water-soluble pullulan with lovastatin (LV) to develop a novel amphiphilic conjugate, pullulan-encapsulated LV (PLV). The PLV conjugate was synthesized with three different ratios of pullulan to LV and characterized by Fourier transform infrared (FTIR). The degree of substitution (DS) of LV in terms of molar ratio was 7.87%, 3.58%, and 3.06% for PLV (1/2), PLV (1/3), and PLV (1/4), respectively, by proton NMR analysis. We selected the PLV (1/2) conjugate to prepare doxorubicin (DXR)-loaded PLV nanoparticles (PLV/DXR NPs) because of its superior properties. The average size and zeta potential for PLV (1/2) NPs were 177.6 nm and − 11.66 mV, respectively, determined by dynamic light scattering, and those for PLV/DXR NPs were 225.6 nm and − 10.51 mV, respectively. In vitro drug release profiling showed that PLV/DXR NPs sustainably released DXR within 72 h, which was more robust at pH 5.4 (97.90%) than pH 7.4 (76.15%). In the cytotoxicity study, PLV/DXR NPs showed greater inhibition of proliferation of TNBC MDA-MB-231 than non-TNBC MDA-MB-453 cells (IC 50 0.60 vs 11.05 μM). FITC-loaded PLV/DXR NPs were prepared to investigate cellular uptake: both cell lines showed a time-dependent uptake of NPs, but the number of NPs entering MDA-MB-231 cells was greater than that entering the MDA-MB-453 cells. Pullulan-based NP co-delivery of LV and DXR could efficiently inhibit TNBC cells, which may help in designing a powerful drug delivery system for treating TNBC.

Co-delivery of doxorubicin and shRNA of Beclin1 by folate receptor targeted pullulan-based multifunctional nanomicelles for combinational cancer therapy

Lili Chen,Ming Qian,Liuwei Zhang,Jing Xia,Yongming Bao,Jingyun Wang,Lianying Guo,Yachen Li

Doxorubicin (DOX) is a widely-used effective antitumor agent. However, its clinical application is limited due to its side effects including anti-apoptotic defense of cancer cells caused by DOX-induced autophagy and deleterious effects in normal tissues. Therefore, in this study, a new folate (FA)-decorated amphiphilic bifunctional pullulan-based copolymer (named as FPDP) was developed as an efficient nano-carrier for the co-delivery of DOX and short hairpin RNA of Beclin1, a pivotal autophage-related gene, to enhance the anticancer effect of DOX by the blockade of the Beclin1 protein mediated autophagy process. In FPDP molecules, pullulan was modified with lipophilic desoxycholic acid for the formation of micelles, the introduced low molecular weight (1 kDa) branched polyethylenimine (PEI) was for shBeclin1 delivery, and folate (FA) was employed as the tumor-targeting group. FPDP micelles demonstrated an average diameter of 161.9 nm, good biocompatibility, applicable storage stability, excellent loading capacities for both DOX and shBeclin1 and a sustained drug release profile. In vitro cell culture experiments demonstrated that the uptake amount of FPDP/DOX micelles in folate receptor positive (FR+) HeLa cells was more than that in folate receptor negative (FR−) HepG2 cells, leading to significantly higher cytotoxicity against FR+ HeLa cells. The simultaneous co-delivery of shBeclin1 and DOX to HeLa cells with FPDP micelles led to efficient reduction in the expression level of Beclin1 as well as synergistic cell apoptotic induction. Furthermore, in vivo studies revealed superior antitumor efficacy of tumor-targeted FPDP/DOX/shBeclin1 in comparison with non-FR-targeted PDP micelles and free DOX. These results highlighted that co-delivery of DOX and shRNA of Beclin1 with FPDP micelles has the potential to overcome the limitations of DOX in clinical cancer therapy.

Incorporation of dumbbell-shaped and Y-shaped cross-linkers in adjustable pullulan/polydopamine hydrogels for selective adsorption of cationic dyes

Ting Su,Lipeng Wu,Gancheng Zuo,Xihao Pan,Mingyang Shi,Cheng Zhang,Xiaoliang Qi,Wei Dong

Hydrogel adsorbents have attracted considerable attention due to their sludge minimization, good water permeability and renewable performance. Here, a promising strategy for the one-step preparation of pullulan/polydopamine hybird hydrogels (PPGels) was presented. Dumbbell-shaped cross-linker neopentyl glycol diglycidyl ether (NGDE, 2 arms) and Y-shaped cross-linker trimethylolpropane triglycidyl ether (TTE, 3 arms) were selected to study the relationship between cross-linker structure and hydrogel performances. The NGDE possessing less molecular repulsive force and higher reactivity demonstrated more effective cross-linking with the pullulan, which leaded to a decrease in pore size of the hydrogel. Meanwhile, the introduction of polydopamine significantly enhanced the adsorption ability and gave the resulting hybrid gel the specific selectivity toward cationic dyes (96 mg/g for crystal violet, 25.8 mg/g for methylene blue and barely not adsorption for azophloxine). Our data suggested that the electrostatic interaction played a vital role in the dye adsorption process, and the adsorption data could be explained by pseudo-second-order model and Langmuir isotherm model. Furthermore, the obtained PPGel could be easily separated after adsorption. This study describes the relationship between cross-linker structure and properties of pullulan/polydopamine hybrid gels, which provides a new strategy to create polysaccharide-based adsorbents for wastewater remediation.

Pullulan dialdehyde cross-linked dual-action adhesive with high adhesion to lung tissue and the capability of pH-responsive drug release

Ke Zhang,Hui Huang,Yilin Zhao,Qinghao Zhen,Dongjian Shi,Jinghua Chen,Xi Chen

To address the main challenges for thoracoscopic lung cancer surgery, including persistent pulmonary air leaks and cancer recurrence, this study developed an in-situ adhesive that can effectively adhere to the lung and release the anticancer drug in response to pH. The adhesive was formulated using hydrophobically modified cold-water fish skin gelatin (hm-CFG) and cross-linking agent pullulan dialdehyde (PDA), in which succinic dihydrazide-modified doxorubicin (SDH-DOX) can be incorporated. Utilizing PDA could improve both cohesion and interfacial adhesion, while also offering drug-loading sites through the aldehyde groups that were not involved in cross-linking. The optimal adhesive formulation was 9C10-CFG/PDA (30 w/v% 9 mol% decanal modified CFG/20 w/v% PDA). The 9C10-CFG/PDA adhesive exhibited suitable cohesive strength, good mechanical flexibility (tensile strain over 170 %), and strong interface adhesion. The burst strength of 9C10-CFG/PDA adhesive (131.5 ± 22.2 mm Hg) was almost 6-fold higher than that of commercial fibrin sealant. In a rat pneumothorax model, 9C10-CFG/PDA adhesive displayed favorable wound-sealing properties, as evidenced by CT imaging and restored rat behavior. When combined with the anticancer drug, SDH-DOX@Adhesive could release the drug in response to pH more gradually than DOX@Adhesive. This dual-action adhesive is anticipated to mitigate post-surgical occurrences of lung air leaks and cancer recurrence.

A pullulan-type α-D-glucan produced from lactose by Aureobasidium pullulans: structure and potential as a prebiotic agri-food ingredient

Tian Shu,Cheng Feng,Wu Pan,Jia Xuewei,Qu Lili,Xu Chunping

Background Microbial exopolysaccharides (EPS) are renewable biopolymers of increasing importance in the agri-food sector, and producing them from low-cost substrates offers a route to valorize agro-industrial resources. Aureobasidium pullulans is an industrially important fungus that secretes the α-glucan pullulan; however, the structure of the EPS it produces under lactose fermentation—a means of upgrading a low-value, dairy-derived carbon source—has not been clearly defined, which limits its rational use as an agri-food bioproduct. This study aimed to produce, structurally characterize, and preliminarily evaluate the physiological activity of this EPS. Results A homogeneous EPS was obtained by ethanol precipitation, Sevag deproteinization, and gel-filtration chromatography. Fourier-transform infrared spectroscopy indicated an α-configured, neutral polysaccharide, and gas chromatography identified glucose as the sole monosaccharide. Size-exclusion chromatography coupled with multi-angle laser light scattering showed a high-molecular-weight polymer. One- and two-dimensional NMR analyses established that the EPS is a linear pullulan-type α-D-glucan with the repeating unit [→6)-α-D-Glcp-(1→4)-α-D-Glcp-(1→4)-α-D-Glcp-(1→] n, in which two α-(1→4)-linked glucose residues alternate with an α-(1→6) linkage. In a high-fat-diet-induced obese rat model, dietary supplementation with the glucan attenuated body-weight gain, improved serum lipid profiles (TG, TC, HDL-C/TC), and lowered serum endotoxin and tumour necrosis factor-α, together with enhanced colonic short-chain fatty acid production, consistent with fermentation of the glucan by the gut microbiota. Conclusions Lactose fermentation by A. pullulans yields a structurally defined, linear pullulan-type α-D-glucan, demonstrating a route to convert a low-value dairy-derived substrate into a value-added biopolymer. Its prebiotic-associated metabolic effects indicate potential as a functional ingredient for the agri-food chain and warrant further mechanistic and safety studies. Graphical abstract

Characterization of a novel chickpea protein-pullulan hydrogels that efficiently load and release sodium salts: Microstructures, molecular dynamics, and rheological properties

Kexin Zhang,Yanhong Liang,Zhifeng Tan,Dayong Zhou,Deyang Li

This study investigated the gel properties, structural characteristics, and Na + loading capacity of hydrogels formed by chickpea protein isolate (CPI) and pullulan (PUL). The aim was to explore their potential as polymers for enhancing Na + distribution and facilitating Na + release in reduced-salt foods. The results showed that the CPI-PUL hydrogels added with 2 % PUL exhibited the highest water holding capacity (70.78 %) and gel strength (493.06 g·mm). Molecular docking analysis indicated that the interactions between CPI and PUL primarily involved hydrogen bonding, hydrophobic interactions, and van der Waals forces. The incorporation of PUL resulted in a denser and more stable network of the hydrogels, which enhanced their thermal stability and mechanical properties. Results from cold-field scanning electron microscopy demonstrated that the hydrogel network structure remained stable at a PUL content of 2 %. Furthermore, fluorescence inversion microscopy and energy dispersive spectroscopy confirmed that the CPI-PUL hydrogels were stably loaded with sodium salts.

pH-sensitive pullulan-doxorubicin nanoparticles loaded with 1,1,2-trichlorotrifluoroethane as a novel synergist for high intensity focused ultrasound mediated tumor ablation

Huanan Li,Chenhao Yu,Jingni Zhang,Qianyan Li,Hai Qiao,Zhigang Wang,Deping Zeng

Nanoemulsions as synergists of high intensity focused ultrasound (HIFU) have been widely investigated, since they possess huge potential for improving the efficiency to ablate tumor. However, their clinical applications are limited due to the unsatisfactory phase-transition effect of nanoemulsions during the process of generating nanobubbles . Herein, a novel synergist for HIFU therapy was designed by encapsulating 1,1,2-trichlorotrifluoroethane (CFC) into pH-sensitive pullulan-doxorubicin (Pu-DOX/CFC) nanoparticles . These Pu-DOX/CFC nanoemulsions, with moderate vaporization temperature threshold of 47 °C, could provide favorable phase-transition effect, thus facilitating HIFU energy deposition. Meanwhile, Pu-DOX/CFC nanoemulsions could effectively deliver DOX/CFC to the tumor site and carry out combined therapy. The in vitro and in vivo results confirmed that Pu-DOX/CFC nanoemulsions notably enhanced both ablation and therapeutic efficiency comparing with other synergists. In conclusion, Pu-DOX/CFC nanoemulsions might serve as a novel synergist for HIFU therapy, and possess great potential in clinical implication.

Preparation and characterization of carboxymethyl chitosan/pullulan composite film incorporated with eugenol and its application in the preservation of chilled meat

Zhen Zeng,Yu-Jing Yang,Qian Tu,Yu-Ying Jian,Da-Ming Xie,Ting Bai,Shan-Shan Li,Yun-Tao Liu,Cheng Li,Cai-Xia Wang,Ai-Ping Liu

To solve the problem of easy spoilage of chilled meat during storage, we fabricated a novel composite film using carboxymethyl chitosan (CMCS)/pullulan (Pul)/eugenol (E) by casting method. The results showed that the mechanical properties of the films were better when the CMCS/Pul ratio was 2.5/2.5. The Fourier transform infrared spectroscopy (FTIR) results showed that intermolecular hydrogen bonds were formed among E, CMCS, and Pul, which was consistent with the rheological test results. Scanning electron microscopic (SEM) images showed that eugenol was well dispersed in the CMCS/Pul matrix. The addition of eugenol significantly increased the antibacterial properties and antioxidant properties. Moreover, when 5% eugenol was added, the water vapor permeability (WVP) of the film reduced to 2.41 × 10 −11  g/m·s·Pa. Finally, the freshness of the chilled meat wrapped with the eugenol-containing composite film was prolonged, thereby offering a potential alternative to synthetic materials.

An injectable, self-healable, antibacterial, and pro-healing oxidized pullulan polysaccharide/carboxymethyl chitosan hydrogel for early protection of open abdominal wounds

Yeying Zhao,Ruojing Li,Yangyang Liu,Lei Song,Zhao Gao,Ze Li,Xingang Peng,Peige Wang

Open abdomen (OA) is an effective method for treating critical abdominal conditions such as severe abdominal infections. The temporary abdominal closure (TAC) technique is often used to temporarily restore the physiological environment of the abdominal cavity and maintain the homeostatic balance of the abdominal cavity. However, most of the common TAC materials available today lack bio-responsiveness, tend to abrade the intestinal canal, and lead to delayed tissue healing of the wound. Hydrogels could mimic the extracellular matrix and have shown significant potential in life science fields such as tissue regeneration, wound repair, and controlled drug release. In this study, a composite hydrogel scaffold was constructed by the Schiff base reaction of oxidized pullulan polysaccharide with carboxymethyl chitosan. The hydrogel exhibited excellent self-healing, cellular biocompatibility, and antibacterial and anti-inflammatory abilities, and in experiments it reduced secondary damage caused by friction between tissue and patch, thereby preventing serious complications such as intestinal fistula, promoted M1–M2 polarization of macrophages, reduced the inflammatory response, regulated the inflammatory microenvironment in vivo, promoted angiogenesis and granulation tissue regeneration, and accelerated wound healing. Therefore, our hydrogel provides a new strategy for material-assisted wound protection during OA and has potential clinical applications.

3D Electrospun Nanofiber Sponges of Insect Chitosan/Pullulan/Citric Acid Embedded with ZnMOF in situ for Enhanced Healing of MRSA-Infected Wounds

Fuchen Jiang,Qing Li,Yingxi Li,Xiaomin Lai,Yun Duan,Arlene McDowell,Zhen Huang,Shuang Liu,Ying Wang,Chen Zhang,Yan Qu,Xiaoli Pan

In biomedicine, one of the major challenges is effectively promoting wound healing of bacterial infections. Current wound dressings (e.g., gauze and bandages) are far from meeting the needs of the population as they lack various bioactivities to promote wound healing (including hemostasis, antimicrobial, anti-inflammatory, promotion of tissue regeneration, etc.) and are not biodegradable. In this study, we prepared an insect chitosan/pullulan/citric acid three-dimensional, layered electrospun nanofiber sponge embedded with ZnMOF in situ (3D-ZnMOF) and used it to promote MRSA-infected wound healing. The fabrication method and physicochemical characteristics of 2D nanofiber membranes and 3D nanofiber sponges were thoroughly assessed. SEM-EDS, XPS, XRD, and FT-IR were used to characterize the 3D-ZnMOF sponges, and the release behavior of zinc ions was examined. Experiments conducted both in vitro and in vivo demonstrated the hemostatic characteristics, biocompatibility, and antibacterial activity of 3D-ZnMOF sponges. 3D-ZnMOF sponges showed the ability to promote wound contraction, accelerate collagen deposition, inhibit inflammatory factors, and promote angiogenesis in an MRSA-infected rat whole-skin wound model. Consequently, 3D-ZnMOF nanofiber sponges offer a great deal of promise for accelerating the healing of wounds infected with drug-resistant bacteria.

Synergistic optimization of the water state and transport pathway in hydrogels via pullulan and directional Freezing-Assisted salting out for efficient evaporation

Huanwei Shen,Xiayue Yang,Wenhao Li,Zhihua Sha,Hongmei Luo,Zixuan Li,Sihan Dong,Wei Cui,Rong Ran

Improving the evaporation efficiency of solar-driven hydrogel evaporators is crucial for addressing water scarcity. Traditional efforts have been focused on increasing the intermediate water content or shortening the evaporation route through hydrogel structure design. However, rare attempts have been made to combine these two approaches. Here, we report the simultaneous achievement of high intermediate water content and short water transport paths in a hydrogel for efficient solar-driven evaporation. By introducing pullulan and MXene nanosheets into a poly(vinyl alcohol) (PVA) system that is subjected to directional freezing-assisted salting-out, we achieve a considerably high intermediate water to free water ratio of 2.11. Meanwhile, low- tortuosity yet compact channels within the hydrogel formed during directional freezing-assisted salting-out accelerate water transport during evaporation. Consequently, the hydrogel evaporator achieves an energy conversion efficiency of 93 % under one sun illumination, with a high vapor generation rate of 3.92kg m −2 h −1 . Even in highly saline water, an evaporation rate of 2.93 kg m −2 h −1 is preserved, accompanied by voltage generation through water-driven ion migration. Moreover, the hydrogel evaporator can be recycled by heating using the remoulade properties of PVA without deteriorating the evaporation rate, holding great potential in sustainable and recyclable water collection.
