A drug delivery system based on magadiite for gemcitabine (namely GEM-Mag) has been constructed by protonation, intercalation with tetrabutylammonium ions (TBA + ), and ion exchange. Various measurements were used to characterize the samples. XRD results showed that the basal spacing of Na-magadiite (15.17 Å) decreased to 11.40 Å after protonation. Following intercalation with TBA + , the basal spacing increased to 25.79 Å. After further ion exchange with the positively charged GEM, the basal spacing reduced to 18.21 Å. FT-IR and TG-DTG indicated that the GEM was incorporated into the magadiite. Based on CHN elemental analysis and EDS results, the chemical formula of GEM-Mag was estimated to be H 1.59 (C 9 H 12 F 2 N 3 O 4 ) 0.41 Si 14 O 29 ·2.33H 2 O. According to the chemical formula, the loading capacity of GEM-Mag for GEM was 10.74%. Drug release tests were carried out either in phosphate buffered solution (PBS, pH = 7.40) or in HCl solution (pH = 1.35). Results revealed that the GEM-Mag exhibited sustained-release of GEM. While the drug released for 96 h in the PBS, the cumulative release percentage of GEM from drug delivery system was only 53.46%; whereas in the HCl solution (pH = 1.35), the cumulative release percentage was 52.15% after 700 min. Cytotoxicity assay confirmed that the magadiite-based drug delivery system for GEM exhibited stronger inhibitory effects on HepG2 cells compared with that of the free GEM. In addition, the drug-loading process and mechanisms have been discussed in detail.

The inherent molecular rigidity of alginate, which limits chain flexibility and prevents effective entanglement, has long hindered its direct electrospinning into uniform nanofibers from aqueous solutions. To overcome this limitation and expand the application of alginate in infection-prone wound repair, this study introduces a novel antibacterial alginate derivative (RA-OSA-APA) synthesized via oxidation-reductive amination using 6-aminopenicillanic acid (6-APA) as a hydrophobic modifier. Leveraging this functional polymer, RA-OSA-APA was blended with water-soluble polyvinyl alcohol (PVA) to successfully fabricate RA-OSA-APA/PVA composite nanofibers through electrospinning. Furthermore, a sequential multilayer electrospinning strategy was employed to sequentially co-spun chitosan (CS) and gelatin (GT) onto the surface of RA-OSA-APA/PVA composite nanofibers, resulting in RA-OSA-APA/PVA/CS/GT electrospun multilayer composite nanofibers. Comprehensive characterization results demonstrated that chemical modification disrupted alginate’s hydrogen bonding network, enhanced molecular flexibility, and facilitated chain entanglement with PVA, significantly improving electrospinnability. Although bead-free nanofibers could not be electrospun from pure RA-OSA-APA aqueous solution, its content was effectively increased within the uniform RA-OSA-APA/PVA composite nanofibers. Furthermore, the sequential layering of CS and GT onto the RA-OSA-APA/PVA composite nanofibers via multilayer electrospinning not only yielded uniform fibrous morphologies stabilized by intermolecular hydrogen bonding but also created a unique physical barrier effect. This architectural innovation effectively reduced water absorption, achieved the effective loading and controlled release of triclosan (TCA), and synergized with the inherent antibacterial functions and robust cytocompatibility of CS and GT to enhance biological performance. With their favorable physicochemical and controlled-release properties, the RA-OSA-APA/PVA/CS/GT composite nanofibers represent a promising multifunctional wound dressing platform for managing infected wounds.

Chronic wounds have persistent oxidative stress impairing tissue regeneration, necessitating antioxidant strategies that are both biologically effective and durable. While silk fibroin (SF) exhibits intrinsic antioxidant activity, how its molecular weight governs antioxidant performance across chemical and cellular contexts remains unclear. We systematically elucidate SF's molecular-weight-dependent antioxidant behavior and translate these insights into designing a sustained-release composite gel. SF fragments spanning ultra-low to high molecular weights were prepared and characterized. Ultra-low-molecular-weight silk fibroin (∼10.7 kDa) exhibited the strongest chemical antioxidant activity, achieving >60% DPPH scavenging and > 95% hydroxyl radical elimination, attributed to enriched tyrosine content and reduced β-sheet crystallinity. However, this superior radical-scavenging capacity did not translate directly to cellular repair. Instead, low-molecular-weight silk fibroin (L-SF, ∼27 kDa) provided the most robust and concentration-stable cellular protection, significantly restoring viability in oxidatively damaged fibroblasts and enhancing intracellular antioxidant defenses, including catalase (CAT) and superoxide dismutase activities ( p < 0.01). Building on this divergence, a composite high-molecular-weight silk fibroin/sodium carboxymethyl cellulose (H-SF/CMC-Na) hydrogel was engineered using H-SF for mechanical reinforcement and L-SF surface coating to impart sustained antioxidant activity. The optimized formulation (H-SF: CMC-Na = 2: 1, 40% solids) satisfied mechanical and conformability requirements for wound dressing applications and enabled sustained release of bioactive SF over 24 h. Gel extracts significantly improved cellular antioxidant indicators, increasing CAT activity 7.6-fold. This work establishes molecular-weight-guided design as a unifying principle for SF-based antioxidant biomaterials, demonstrating that effective wound dressing candidates must balance radical-scavenging capacity with cellular compatibility and sustained biological function.

Conventional pesticides are often plagued by poor controlled-release performance, hydrophobic nature, posing risks to nontarget organisms. The development of nano-delivery carriers with hydrophilicity on crop surfaces and sustained-release function within crops remains challenging. Inspired by host-guest recognition in supramolecular chemistry, we developed a molecular recognition-driven engineering strategy that exploits the pesticide structure to modulate its release performance and mechanism. The two structurally distinct fungicides azoxystrobin (AZO) and dimethomorph (DIM) as guests were encapsulated in methylated β -cyclodextrin cavity (M β CD) towards preparing nanofibers using electrospinning techniques to target crop diseases. SEM, Zeta potential, molecular simulations, DFT calculations, release kinetics and mechanism collectively confirmed that pesticide structure affected the host-guest inclusion behavior, nanofiber morphology, and ultimately the sustained-release feature. Both pesticide/M β CD inclusion complex nanofiber (pesticide/M β CD-IF) displayed highly water-soluble and good antifungal efficacy against Rhizoctonia solani and Haematonectria haematococca relative to the corresponding free pesticides. The LC 50 value of DIM/M β CD-IF was higher than that of DIM SC, indicating improved safety for soil nontarget organisms. This study presents a pesticide architecture-centered paradigm for nanofiber design, aiming to provide a feasible method for sustainable crop protection. Sustained release microspheres loaded with goserelin are regarded as a promising candidate for treating prostate cancer and other sex hormone diseases. However, their widespread adoption has been hindered by issues such as wide particle size distribution and unstable release characteristics. To address these challenges, we employed a combination of the solid-in-oil-in-water microspheres preparation approach (S/O/W) and innovative premix membrane emulsification technology and deeply investigated the effects of four key parameters on the loaded performance of microspheres and the microscopic mechanisms behind them. With this approach, we successfully produced goserelin-loaded sustained release microspheres of narrow particle size distribution (Span 0.642), remarkable encapsulation efficiency (DL = 4.23 %, EE = 93.98 %), low initial burst release (about 0.50 % within 2 h), and compatibility with small injection needles (23-G, inner diameter 0.33 mm, outer diameter 0.64 mm, maximal force 59 N). In the animal model(administered dose, 2.4 mg·Kg −1 ), goserelin long-acting sustained release microspheres sustained release for over 32 days, maintaining effective concentrations above 2 ng·mL −1 , and effectively reduced serum testosterone concentrations to castration levels (<1.0 ng·mL −1 ) by day 4, maintaining this inhibition for up to 21 days, exhibiting comparable efficacy to the positive control group. In vivo release kinetics analysis revealed that goserelin-loaded sustained release microspheres exhibited a release pattern dominated by diffusion with corrosion assistance in vivo. In summary, the systematic and comprehensive evaluation of uniform-sized goserelin-loaded sustained release microspheres has highlighted their excellent translational potential, and the study herein may provide new strategies and ideas for the development of microsphere dosage forms.

Acute myeloid leukemia (AML) is extremely difficult to cure due to the challenges in accurately targeting it, as it is characterized by rapid progression, high aggressiveness, and high drug resistance. In this study, biomimetic sustained release nanoparticles (PLGA-C-M) were designed and prepared to inhibit the survival and resistance pathways of AML. PLGA-C-M targeted AML cells by wrapping leukemia cell membranes, achieving sustained slow drug release in the blood, and then progressively affecting intracellular Ca2+ signaling by targeting TRPM2 ion channels that were highly expressed in AML in a step-by-step manner. PLGA-C-M can inhibit the growth of AML cells from three aspects: destroying mitochondrial function, reducing autophagy, and overcoming the drug resistance of cancer cells. Biomimetic nanoparticles achieved simultaneous regulation of intracellular ROS and Ca2+ signals to inhibit the growth of leukemia cells and provided ideas for the regulation of ion channel-related signal transduction in AML.

Hydroxypropyl chitin (HPCH) has been studied as a carrier for drug delivery due to its excellent biocompatibility, biodegradability and reversible thermosensitivity. However, the behaviors of the erosion/degradation of HPCH and release of drug for drug-loaded HPCH in vivo are unclear. Visualization of the drug-loaded HPCH could reveal the location and degradation of the carrier, and the release and distribution of the drug in vivo, which is more beneficial for promoting the clinical translation. Herein, we simultaneously tracked HPCH labeled by aggregation-induced emission fluorogens (AnDA-HPCH) and a model drug (indocyanine green, ICG) in vitro and in vivo without mutual interference. The erosion/degradation rate of AnDA-HPCH and the drug release of ICG from the ICG-loaded AnDA-HPCH sponge implanted in the buttock of mice were slower than those of the injected hydrogel containing the same amounts of drug and polymer. Further, the long-acting local anesthetic ropivacaine-loaded HPCH sponge implanted in the back of rats showed an effective analgesic time of approximately 30 h, which was 3-fold longer than that with the injected RPH@HPCH hydrogel (10 h) and much longer (6 times) than that of the xaracoll-like formulation (5 h). This thermosensitive HPCH sponge may be a promising visible biomedical implant for sustained-release drug carrier and tissue repair.

The KGM-based hydrogels exhibit great potential as a drug delivery material. However, they often fail to meet industrial requirements due to the weak property. In this paper, a simple method was proposed to prepare KGM composite hydrogel by cyclic freeze-thaw method using Fe 3+ ion as crosslinking agent and modified montmorillonite (CS@MMT) as nano-filler to improve the mechanical and slow release properties. The SEM images reveal that the incorporation of CS@MMT resulted in a compact network architecture within the hydrogel. The tensile strength of the KGM/PVA/Fe 3+ /CS@MMT hydrogel has been significantly enhanced by 250 %, attaining 0.30 MPa, in contrast to 0.12 MPa of the KGM/PVA hydrogel. Furthermore, the loading capacity has been significantly improved, with a loading rate of 52.56 ± 3.30 %, in contrast to 7.75 ± 1.39 % of the KGM/PVA hydrogel. In addition, multiple release kinetics models have proved that KGM/PVA/Fe 3+ /CS@MMT hydrogel has superior controlled slow-release property. In all, this study presents a direct way to enhance the mechanical property and slow-release capability of KGM/PVA hydrogels, which is anticipated to promote the practical applications of the KGM-based hydrogels, especially in the biomedical field, such as drug slow release and wound care.

A pesticide carrier system can offer advantages to active components, such as improving shelf life, extending the lasting period, and reducing toxicity, all of which could reduce the adverse effects of pesticides on the environment. Graphene oxide (GO) is a promising medical drug carrier material, and its research in the field of pesticides is still in the exploration stage. Herein, in order to load water-insoluble pesticides, a series of polylactic acid–graphene oxides (PLA–GOs) were produced incorporating small amounts (0.1–1 wt %) of GO using acetone–chloroform and N,N-dimethylformamide–dichloromethane, two dual-solvent systems. The prepared PLA–GO carrier materials were characterized by FT-IR, Raman spectroscopy, and differential scanning calorimetry analysis, and their effects on rice growth within 20 days were also evaluated. When the amount of GO added was 0.5% and the solvent system was acetone–chloroform, the amount of pyraclostrobin (Pyr) loaded on PLA–GO (A-0.5%) was very high (39.89%). The PLA–GO pesticide microsphere showed a significantly sustained release of Pyr (up to 30 d). Furthermore, the indoor bioassay results showed that the PLA–GO pesticide microsphere (EC50 = 1.2682) had the best fungicidal activity against Rhizoctonia solani, followed by Pyr technical (EC50 = 2.3137) and commercial Pyr microcapsule suspension (EC50 = 3.4570). In conclusion, it was found that PLA–GOs prepared using the acetone–chloroform system were more suitable as a pesticide carrier, which provided a preliminary basis for GO to realize the coating of water-insoluble pesticides.

Climate change has made on-site bloom control increasingly challenging. Therefore, it is necessary to study whether it is possible to control the outbreak of summer cyanobacterial blooms by early prevention of cyanobacterial recovery. This study used artemisinin sustained-release algaecides (ASAs), a typical allelochemical for in-situ cyanobacterial control, to evaluate recovery effects and microbial-ecological impacts through microcosm experiments. All doses inhibited the recovery and bloom of cyanobacteria (Chl-a < 66μg/L), prevented spore germination to varying degrees (especially at the low and high doses), and reduced the ecological niche of cyanobacteria (1.6–2.6). The results demonstrated that low-dose ASAs effectively prevented cyanobacterial recovery with minimal microbial-ecological impacts. Higher ASAs doses reduced bacterial diversity (shannon index dropped from 5 to around 2.8), enhanced environmental selection (the proportion increased from 15 % to 67 %), and loss of native species. The bacterial community underwent a pronounced shift: dominant cyanobacterial genera such as Limnothrix declined substantially, whereas Proteobacteria increased to 60–95 % phylum-level relative abundance. Conversely, eukaryotes enhanced heterogeneous selection (the proportion = 50 %–83 %), smaller diversity changes (Chao1 = 1000–1600), moderate community succession, and widespread microbial cooperation. The effects of ASAs on microbial networks were limited, but the effects of high doses were not negligible, especially the low occupancy module, which accounted for only 8.79 %, was more affected by the DOC/pH changes caused by high-dose. Moreover, ASAs prevented cyanobacterial recovery through bacterial-eukaryotic cooperation and functional microbes, provided the microbial community had not undergone severe environmental stress. Our findings emphasize the importance of considering the nonlinear microbial-ecological responses to dosing and the necessity of low-dose strategies during both in-situ control and prevention, as low-dose result in less changes in microbial community structure and lower risk of secondary cyanobacterial blooms.

Polygonatum sibiricumpolysaccharide (PsP), one of the main components ofPolygonatumsibiricumused in traditional Chinese food and medicine, has important bioactive functions, but it is difficult to fully utilize PsP because of the degradative effect of digestive gastric juices. This study aimed to innovatively synthesize a new food formulation for PsP, namely, a PsP–hydroxyapatite (HAP) sustained-release system, so as to reduce its degradation. The new food formulation was optimized and evaluated by the response surface method (RSM) and by in vitro experiments. The optimal stirrer temperature, reaction pH, etching pH, and loading time for synthesizing PsP-HAP were 85.62 °C, pH 11.12, pH 8.40, and 5.10 h, respectively, all of which were different from the findings of other similar research studies. The average encapsulation rate of PsP-HAP reached (40.16 ± 1.54)%, and the content of PsP was 8.98%. Additionally, PsP-HAP appeared to be pH-responsive, and its continuous antioxidative effect was first proven by the DPPH assay and then cytologically by a total antioxidative capacity assay. The CCK-8 assay indicated that PSP-HAP did not induce toxicity. This study successfully developed a new food formulation for PsP which appears to have the potential to reduce the degradative effect of digestive gastric juices. Thus, it is possible to achieve full utilization of PsP by using this new sustained-release food formulation.

By using dopamine (DA) as the monomer, the model drug avermectin (AVM) was loaded on polydopamine microspheres (AVM/PDAMS) and polydopamine microcapsules (AVM@PDAMC) by the method of impregnation and encapsulation, respectively. The materials’ structures were systematically characterized using Fourier transform infrared spectroscopy (FTIR), zeta potential analysis, scanning electron microscopy (SEM), thermogravimetric analysis (TGA), and differential scanning calorimetry (DSC). The comparison of antiultraviolet capability as well as release behaviors under different pH values of the materials were studied. The results showed that a spherical appearance was observed from both materials. The use of AVM/PDAMS and AVM@PDAMC made the decomposition temperature of AVM increase to 235°C and 245°C, respectively. After being exposed to ultraviolet light for 1400 min, the residual ratios of AVM of AVM/PDAMS and AVM@PDAMC were 42% and 54%, respectively. Both AVM/PDAMS and AVM@PDAMC showed acid sensitivity. AVM/PDAMS and AVM@PDAMC took about 13 h and 60 h to reach the release rate of 50% under pH 3. The release process of AVM/PDAMS could be explained by the Weibull model at pH 3, while the release behavior of AVM@PDAMC fitted the Baker–Lonsdale equation. At pH 7 and pH 9, both of the delivery materials followed the Korsmeyer–Peppas model and belonged to the Fick diffusion.
